Summary In vitro tuberization responses of eleven clones of potato were evaluated under different photoperiods and light intensities. No growth regulators were added to the medium to avoid the potential modification of the response to the environmental stimuli. Plantlets were grown on MS medium containing 3% sucrose for four weeks under long days (16 h). Tuberization was induced by short days (8 h). Five photoperiodic combinations and three light intensity treatments were applied. Light applied after the tuber induction phase delayed or inhibited tuber initiation (at proper photoperiods both at 111 and 55 μmol m−2 s−1 light intensities at 24/15 °C day/night temperature). Darkness following the induction stage accelerated and synchronized tuber initiation after high light intensity. However, these effects of dark treatment depended on light intensity applied in short days before dark treatment. There was an interaction between the duration of darkness applied in photoperiodic treatments and the light intensity applied in short days on tuber initiation. No relationship was observed between the maturity groups of clones tested and their tuber initiation responses. Tight interaction (P≤0.01) of treatments and clones was proved by statistical analysis. Reliable tuberization and high rate of tuberization (1.20–1.52) can be reached using these hormon-free systems.  相似文献

This experiment was done on the three potato cultivars [Agria (susceptible), Satina (semi-tolerant) and Ceaser (tolerant to water deficit)] and three irrigation treatments (after 30 mm evaporation from basin class A, after 60 mm evaporation + spraying by Potassium Humate, and after 60 mm evaporation from basin class A) for two locations in 2007. Experimental design was Split Plot with three replications. Potassium Humate spraying (250 mL ha(-1)) were done in three stages of emergence, before tuberization and during tuberization period. Combined analysis of variance showed that there were significant differences between locations, cultivars, irrigation treatments and location x cultivars interaction as effect on tuber yield. Comparison of means for irrigation treatments showed that spraying by Potassium Humate in stress condition induced increasing of tuber yield. Spraying by Potassium Humate in water deficit condition increased tuber yield up to 11.01 ton ha(-1). Ceaser had the highest tuber yield. It had higher tolerance to water deficit as well. Ceaser had a high potential in control and severe stress. Decrease in yield of Ceaser after 60 mm evaporation + spraying by Potassium Humate and after 60 mm evaporation from basin class A, relative to control (after 30 mm evaporation from basin class A) was 1.03 and 13.08 ton ha(-1) but for Satina was 7.83 and 16.61 ton ha(-1), respectively. Satina had the lowest Environmental Variance, Environmental Variance Coefficient, Finlay and Wilkinson's and Eberhart and Russell's model and was the most stable cultivar. Lin and Binns parameter showed that Ceaser and Satina were the most stable cultivars. Results of GMP, STI and MSTI were very considerable and Ceaser and Satina had a high yield in water stress and control conditions.  相似文献

A two-year study was conducted to determine the effects of (1) jasmonic acid (JA) pre-treatment, (2) JA supplement in culture media, (3) cultivar (Amisk, Atlantic, Russet Burbank, Shepody, and Umatilla Russet), (4) light (0 h, 8 h), and (5) dormancy breaking treatment (Rindite, gibberellic acid) on greenhouse production of minitubers from microtubers andin vitro plantlets. The microtubers were produced under short day (8 h) light conditions and in darkness, from stock plantlets pre-treated with JA and untreated, and on tuberization media with or without JA.In vitro plantlets (the industry choice in nuclear seed potato production) of all five cultivars performed well, meeting the standard criteria for greenhouse production of minitubers. Production of minitubers from microtuber-derived plants of cvs Amisk, Russet Burbank, and Umatilla Russet was similar to that of plantlet-derived plants with regard to number of minitubers. Yields (weight), however, were lower than those from plantlets. Microtuber responses to JA varied with cultivar. Amisk produced the highest number of minitubers per plot from microtubers derived from JA pre-treated plantlets. Jasmonic acid-pretreated microtubers also gave significantly more minitubers in Russet Burbank and Umatilla Russet than the microtubers from other treatments. Shepody did not benefit from JA treatments and JA pre-treated Atlantic microtubers performed poorly, producing significantly lower yields of minitubers than other cultivars. Independently of cultivar, microtubers produced under 8-h photoperiod gave significantly higher yields of minitubers than microtubers produced in the dark. Dormancy release was the key factor influencing microtuber performance. Rindite proved to be a much more effective dormancy breaking treatment than gibberellin. JA conditioning of stock plants prior to tuberization is being proposed as a treatment in production of microtubers for greenhouse production of minitubers.  相似文献

To gain a better understanding of the relative importance of the problems facing expansion of potatoes in Asian countries, a questionnaire was sent to heads of National Potato Programs in which factors limiting the production and use of this crop were evaluated. This paper briefly describes the method employed to tabulate the survey results and then analyzes the findings themselves. Constraints that were consistently singled out include the high cost of seed potatoes, price instability, and viral diseases such as potato leaf roll. These results should serve to facilitate further refinement of regional research strategies for countries facing common problems.  相似文献

Summary Boron (B) requirements differ widely among plant species and the concentration range between toxicity and deficiency is less for B than for any other nutrient. Excess B can adversely affect calcium (Ca) uptake and plant growth. Potato cvs Bintje and Norland plantlets were micropropagated on Murashige & Skoog (MS) nutrient medium, supplemented with 3 (MS control level) or 9 mM Ca, and a range of H3BO3 levels (0.025 to 0.300 mM B). Medium B levels of 0.100 and 0.300 mM decreased Ca content in leaves and shoots of cv. Norland, but not Bintje. Medium B level of 0.025 mM, which is 25% of the control MS level, enhanced Ca uptake in cv. Norland and did not compromise normal plantlet growth in either cultivar. This lower H3BO3 level (0.025 mM B), and a B-free gelling agent such as Gelrite, could be used for micropropagation of potato and possibly other species susceptible to Ca defifiency disorders.  相似文献

Summary A reverse of the delaying effect of gibberellic acid (GA3) was observed in in vitro tuberization of potato plantlets when the medium contained 0.8×10−5 M jasmonic acid (JA). The promoting effect of JA seemed to be correlated with the initial absence of growing roots, probably through direct activity of JA in microtuber-producing buds in the stoloniferous shoots. This is based on the fact that JA did not inhibit root growth a posteriori. Endogenous gibberellins (GAs) synthesized by roots did not interfere with the previous activity of JA in tuberization. The absence of a JA promoting-effect on tuberization of previously-rooted plantlets could be related to the capacity of potato roots to synthesize endogenous GAs which might antagonize the JA effect on buds of stoloniferous shoots.  相似文献

Summary Potato clones and cultivars differ in their ability to tolerate damage by the potato cyst nematodes:G. rostochiensis andG. pallida. This paper reports a simplified field assessment whereby single, spaced plants are grown in heavily infested soils. These trials identified those clones which are either very sensitive to damage or those that grow and yield well in the presence of nematodes. The use of such a screening procedure in a breeding programme is discussed.  相似文献

The latex agglutination test (LAT) and enzyme-linked immunosorbent assay (ELISA) were evaluated in separate studies for their ability to detect potato virus X (PVX) and potato virus S (PVS) in tissue culture plantlets. Healthy and infected clonal lines of several potato cultivars were used. LAT was unsatisfactory because only low levels of agglutination were obtained with infected samples, and because variable and inconsistent results were obtained with both healthy and infected clones. ELISA, however, consistently gave high spectrophotometric readings and intense visual reactions for infected but not for healthy clones. The results indicate that ELISA can be used to detect PVX and PVS in tissue culture plantlets, and in programs where tissue culture is employed, early detection and elimination of infected plantlets is possible.  相似文献

There has been a marked increase in the use of herbicides in potato production. In many cases growers have simply added herbicides to their existing weed control program with little or no reduction in tillage. On Long Island six or more separate mechanical operations may be used by growers to control weeds. The current paper summarizes 3 years’ results with various tillage — herbicide combinations carried out under widely varying levels of weed infestation. Under conditions of low to moderate infestation good results were obtained with one to two tillage passes plus the use of an herbicide. The use of an incorporated herbicide such as EPTC plus a single hilling operation as early as ground-brack was satisfactory. Greening of tubers was not appreciably increased by this or other early hilling practices. Fields with severe weed infestations required two to three tillage treatments plus an herbicide application. No yield reduction occurred from these treatments, with some indication of a trend in the opposite direction.  相似文献

Micropropagated potato plantlets (Solanum tuberosum cvs Acadia Russet, Red Gold, Red Pontiac, and Russet Burbank) were used to test the effects of exposure to ultra violet rays (UV) or inclusion of vitamin D3 (cholecalciferol) in the test solution on plantlet calcium (Ca) uptake. Ca uptake was determined by measuring shoot tissue45Ca2+ concentration inex vitro plantlets placed into test solutions containing radiolabelled calcium (45CaCl2). Shoot tissue45Ca2+ concentration was very significantly increased in all cultivars exposed for 24 h to UV treatment compared with control plantlets. This increase in shoot45Ca2+ concentration was similar (three cultivars) or greater (one cultivar) than that caused by 10 ing L?1 vitamin D1 in the test solution. When plantlets were exposed to UV, then placed for 24 h in the dark, significantly greater shoot45Ca2+ concentrations occurred compared with plantlets tested immediately following treatment. This suggests that a change in the UV-elicited cellular product occurred during the dark interval that promoted more Ca uptake into plantlets.  相似文献

Summary The seaweed concentrate ‘Kelpak’ is used commercially in the greenhouse and field to improve plant quality. ‘Kelpak’ was added to the in vitro culture medium of potato cv. ‘BP1’ and also applied as a leaf/soil drench immediately after transplanting. The addition of 0.25% seaweed concentrate to the medium improved plantlet quality and led to better establishment in the greenhouse. No beneficial effect of seaweed concentrate in the tissue culture medium was observed if a second cutting was part of the micropropagation process. Additional application of 0.5% seaweed concentrate in the greenhouse to plantlet cuttings derived from tissue culture was not beneficial.  相似文献

Two days of chilling (exposure to 1 C) had no detectable effect on chip color or sugar content of potato (Solanum tuberosum L.) tubers. Four days of chilling led to increases in sugars if potatoes were stored at 10 C or 19 C for 3 to 4 days after chilling ended. Longer chilling gave increases in sugars by the end of the chilling period, and returning tubers to 10 C or 19 C magnified the response. Glucose and fructose gave similar increases from chilling treatments, except that glucose levels were consistently greater. Sucrose was increased by chilling, but the response pattern did not always resemble that of the reducing sugars. Twenty-seven days of storage at 19 C after chilling lowered levels of reducing sugars late in the storage season but not in December. Cultivars differed in response to chilling. Kennebec tubers accumulated far more reducing sugars but much less sucrose than did Norchip tubers. Storage at 19 C after chilling lowered the sucrose content of Kennebec and Monona while raising that of Norchip.  相似文献

Field experiments were designed to subject Superior potato plants to various levels of defoliation by the Colorado potato beetle (CPB)Leptinotarsa decemlineata (Say), Coleoptera: Chrysomelidae. Defoliation occurred during each of five consecutive plant growth periods and CPB population, plant leaf area, and tuber weight data were recorded for each period. A visual defoliation rating scheme provided an accurate estimate of actual potato plant leaf area of defoliated plants. Data generated from regression analysis demonstrated a significant dependence of leaf area on CPB numbers per plant during some plant growth periods, but numbers of CPB accounted for very little of the total variation in tuber weight. Plant leaf area was the most important independent variable in the tuber weight regression model.  相似文献
