为研究鲤热休克蛋白90(HSP90)的免疫效果,根据鲤HSP90 mRNA的基因序列(GenBank),构建真核表达质粒pEGFP-C1-HSP90,并将pEGFP-C1-HSP90转染至鲤脑细胞(CCB)中,荧光倒置显微镜下观察显示,重组质粒pEGFP-C1-HSP90在CCB中成功表达;以不同免疫剂量的重组表达质粒与pIRESORF81联合免疫鲤,通过间接酶联免疫吸附试验(ELISA)检测免疫前后抗体水平。结果表明:注射重组质粒的血清中能够检测到KHV特异性抗体,其抗体水平随着免疫次数的增加而增加,第三次免疫后两周,抗体水平达到最高;联合免疫与单独免疫核酸疫苗pIRES-ORF81相比,抗体水平明显增加,但无显著性差异(P0.05)。研究表明,鲤HSP90作为锦鲤疱疹病毒的免疫佐剂具有一定的目标效果。  相似文献

TLR4能识别革兰氏阴性菌脂多糖,并将信号向下游传递,启动天然免疫反应并诱发获得性免疫反应,在机体的防御反应中发挥重要作用。研究利用PCR-RFLP方法对TLR4基因的3个SNPs位点(c.611 T>A、c.962 G>A和c.1027 C>A)进行基因型分析,同时利用荧光定量PCR法检测每个个体TLR4基因的表达量,统计学方法比较不同基因型个体间表达量的差别。发现SNPs c.611 T>A和c.962 G>A显著影响TLR4的转录(P<0.05)。结果表明,SNPs c.611 T>A和c.962 G>A引起的蛋白质合成量变化,影响机体免疫反应。  相似文献

Toll-like receptor 4 (TLR4) is essential for initiating the innate response to lipopolysaccharide (LPS) from Gram-negative bacteria by acting as a signal transducting receptor. In order to help in investigating TLR4 as a candidate disease-resistance gene in cows, we isolated the cDNA (GenBank accession no. DQ839566) by RT-PCR and rapid amplification of cDNA ends (RACE) experiments and analyzed the sequence characters by bioinformatics. The results showed that cattle TLR4 gene about 3 739 bp contains an open reading frame of 2 526 bp encoded 841 amino acids (aa), 470 bp 5′ untranslated region (UTR), and 743 bp 3′ UTR. Tissue expression profile by RT-PCR indicated that TLR4 gene expresses in mammary glands, liver, muscle, duodenum, fats, uterus, kidneys, hearts, lungs, pancreas, and ovary. TLR4 protein domain predicted by bioinformatics consists of signal peptide, transmembrane helices domain, 3 sorts of leucine-rich repeat domains (LRR, LRR-TYP, and LRRCT), and a toll-interleukinl-resistance domain (TIR). Leucine-rich repeat domains were related with recognizing a broad of pathogen-associated molecular patterns (PAMP) from pathogen, and TIR domain for downstream signaling transduction was most conservative (98% identify) than other domains after alignment of protein from ovine, porcine, human, and mouse. In addition, a 470 bp 5′-flanking region sequence was amplified by PCR, and 15 putative DNA binding sites were predicted, but this sequence lacks TATA box, CCAAT character, and GC-rich regions.  相似文献

为筛选猪丹毒丝菌灭活疫苗免疫佐剂,将5种佐剂(聚合物佐剂、矿物油佐剂ISA 201 VG、蜂胶佐剂、弗氏佐剂、铝胶佐剂)分别与3株猪丹毒丝菌株(AEr21、AEr31和AEr32)制备成15种灭活疫苗,经物理性状检验、无菌检验、安全检验合格后分别免疫小鼠,应用ELISA方法检测小鼠血清中IgG抗体水平和细胞因子含量(IL-4、IL-10、TNF-β、IFN-γ、MCP-1)。通过腹腔注射和灌胃2种方式对免疫后小鼠进行攻毒,计算免疫保护率。结果显示,5种佐剂与3株猪丹毒丝菌株制备的灭活疫苗均可刺激小鼠产生良好的体液免疫和细胞免疫,矿物油佐剂ISA 201 VG疫苗免疫组诱导小鼠产生的IgG抗体水平和细胞因子含量最高,与铝胶佐剂、弗氏佐剂疫苗免疫组差异显著(P<0.05),与聚合物佐剂、蜂胶佐剂疫苗免疫组差异不显著(P>0.05);矿物油佐剂ISA 201 VG疫苗免疫组在攻毒后的免疫保护率最高,对腹腔注射和灌胃攻毒小鼠的免疫保护率分别为80%、80%、60%和100%、100%、80%。试验结果表明,矿物油佐剂ISA 201 VG可作为猪丹毒丝菌灭活疫苗制备的候选免疫佐剂。  相似文献

【目的】研究苏钟猪TLR4的多态性及其编码区第1 027 bp处突变对TLR4蛋白功能的影响。【方法】采用PCR-SSCP的方法检测TLR4在苏钟猪中的多态性并利用real-time PCR方法检测不同基因型(CC型和AC型)的猪肺泡巨噬细胞经脂多糖(lipopolysaccharide,LPS)诱导后,TLR4及促炎症因子TNF-α、IL-1β表达量的变化情况,探讨该处突变对TLR4识别LPS的影响。【结果】①共检测到3个突变:G962A、C1027A、G960A,其中前两个为有义突变,且C1027A突变可引起编码氨基酸性质的改变;②LPS能快速诱导猪肺泡巨噬细胞中TLR4及促炎症因子TNF-α和IL-1β表达水平上调且TLR4编码区C1027A不同基因型(CC型和AC型)的猪肺泡巨噬细胞(pulmonary alveolar macrophages,PAMs)对LPS的敏感性及反应强度存在显著性差异。【结论】苏钟猪TLR4编码区的序列相对保守、多态含量较低,群体遗传变异程度较小。TLR4编码区C1027A突变能影响TLR4识别LPS的能力,等位基因C为苏钟猪抗革兰阴性菌感染的优势基因。  相似文献

将沙门氏菌鞭毛素和弓形虫免疫优势表面抗原1(SAG1)融合表达,研究其在小鼠上激发的体液免疫应答.首先,采用PCR扩增SAG1片段.其次,将SAG1连接到原核表达载体pET-28a中构建表达质粒pET-28a-SAG1;连接到实验室已有的pET-28a鞭毛素质粒中构建表达质粒pET-28a-F-SAG1;质粒转化至大肠杆菌BL21(DE3)菌株中并进行表达.将纯化后的蛋白免疫小鼠,每隔2周免疫一次,共免疫3次,第3次免疫后15 d取小鼠血清.最后,采用蛋白印迹检测蛋白的大小;用酶联免疫吸附方法检测血清中多克隆抗体的效价.结果表明,诱导出的SAG1蛋白大小为30 ku,F-SAG1大小为70 ku.酶联免疫吸附方法检测的D450 nm均为阴性对照的两倍,表明该多克隆抗体具有较高效价.可见,鞭毛素可以作为分子佐剂促进弓形虫亚单位疫苗的体液免疫应答.  相似文献

研究了佐剂蛋白大肠杆菌热敏性肠毒素LTRG与鸡新城疫疫苗(NDV)联合冻干制品的免疫效果。选择不同剂量的佐剂蛋白与鸡新城疫病毒(Newcastle disease virus,NDV)LaSota抗原配比,按生产程序制备冻干疫苗。经滴鼻途径免疫雏鸡,检测血清抗体、粘膜抗体水平,分析不同配伍疫苗的免疫效果。结果表明:含4μg LTRG佐剂蛋白的NDV冻干疫苗,免疫雏鸡血清IgG抗体水平明显高于其他添加组及NDV疫苗组,差异显著;粘膜IgA抗体与NDV疫苗组比较,差异极显著。LTRG佐剂蛋白与鸡新城疫疫苗配伍制备冻干疫苗,能增强免疫雏鸡的免疫应答能力,尤其是可产生较好的粘膜抗体水平。  相似文献

目的 研究Toll样受体4(TLR4)896A>G位点基因多态性与溃疡性结肠炎(UC)发病风险的关系。方法 检索相关英文及中文数据库筛选文献,以OR值及95%CI为效应指标,运用RevMan 5.2和Stata 11.0进行Meta分析和敏感性分析,并采用Egger′s test评价发表偏倚。结果 研究共纳入14篇文献,包括2 174例UC患者和3 134例对照者。Meta分析结果表明携带等位基因G的人群较携带等位基因A的人群患UC的风险性增加,在各个基因模型下均差异有统计学意义[等位基因模型G/A:OR=1.41,95%CI(1.21~1.66),P<0.000 1;显性模型AG+GG/AA:OR=1.37,95%CI(1.16~1.62),P=0.000 2;隐性模型GG/AA+AG:OR=3.74,95%CI(1.78~7.86),P=0.000 5;共显性模型AG/AA:OR=1.42,95%CI(1.07~1.87),P=0.01;共显性模型GG/AA:OR=3.85,95%CI(1.82~8.12),P=0.000 4]。亚组分析结果表明,在等位基因模型G/A、显性模型AG+GG/AA、共显性模型AG/AA下,差异仅在白种人中存在。结论 TLR4 896A>G基因多态性与UC易感性相关,携带等位基因G会增加白种人患UC的发病风险。由于该研究纳入有关亚洲人群及非洲人群的文献数量较少,相关结果需要更多研究予以验证。  相似文献

In order to investigate the functional significance of membrane lipid unsaturation, we have isolated a series of mutants of Arabidopsis thaliana which are deficient in particular membrane fatty acids. The first of these mutants completely lacks Delta3-trans-hexadecenoate, an acyl group that until now has been thought to play an important role in the structure or function of thylakoid membranes in photosynthetic eukaryotes. The apparent absence of any marked physiological effect of the mutation illustrates the potential of this approach to the analysis of membrane structure and function.  相似文献

Toll-like receptor (TLR) 4 plays an important role in the innate immune system and has been involved in resistance/ susceptibility to a number of diseases as revealed by studies in human and other domestic animals.Wild boar survives in natural environment without artificial interference and may be different from domestic pig in innate immune system.Here,the complete coding sequence of TLR4 and TLR4A was cloned in wild boar,and two other alternative splicing variants,TLR4B and TLR4C,were obtained.Compared to the counterpart from domestic pig (GenBank No.AJ628065),there were five SNPs,c.510T>C,c.960A>G,c.962A>G,c.1605T>G and c.1824A>G,in the coding sequence of wild boar TLR4A gene.TLR4 gene was expressed in all the tissues from wild boar studied with the most abundance in spleen tissue,and mRNA level was significantly lower in spleen from wild boar than that from Min pig.The allele distribution was significantly different at polymorphic loci c.962G>A and c.1027C>A (p<0.01) between wild boar and Min pig.The results would contribute to understand the innate immune system in wild boar.  相似文献

为研究囊素肽对禽流感(H9N2)油乳剂灭活疫苗免疫效果的影响,将200只鸡分为疫苗组、疫苗+囊素组、囊素组和对照组,每组50只。从13日龄起每7 d测定其法氏囊指数、禽流感(H9)HI抗体水平。结果表明:囊素肽能提高禽流感(H9N2)油乳剂灭活疫苗免疫后的抗体水平,并延长高水平抗体的维持期,能显著地促进法氏囊的生长发育。  相似文献
