Summary Conditions for a rapid, precise [100 g iodonitrotetrazolium chloride (INT)-formazan ml-1 assay mixture], and easily reproducible assay of potential soil dehydrogenase activity are described, using 2(p-iodophenyl)-3-(p-nitrophenyl)-5-phenyl tetrazolium chloride (iodonitrotetrazolium chloride, INT) as the substrate. Reduced iodonitrotetrazolium formazan (INTF) was measured by spectrophotometry (464 nm) after extraction with N,N-dimethylformamide and ethanol. With this method, the coloured complex formed is highly stable. The effects of pH, buffer concentration, temperature, substrate concentration, amount of soil weight, and reaction time on dehydrogenase activity were investigated. The rate of substrate hydrolysis was proportional to soil weight; the optimal INT reduction was achieved with 1 M TRIS buffer (pH 7.0) at 40 °C. It was possible to determine the biotic and abiotic substrate reduction by comparing assays of autoclaved and unsterile soil samples. Different investigations have confirmed that the intracellular enzyme is highly correlated with the microbial biomass, and indicate that this activity is suitable as an indirect parameter of microbial biomass, measurement.  相似文献

Summary The course of recovery in biological activity was assessed in the top 5 cm of undisturbed soil cores (29.7 cm diameter, 30 cm deep) that had been fumigated in the laboratory with methyl bromide. The cores were returned to their original pasture and forest sites, two with a moderate and two with a high rainfall, and untreated soils at all sites served as baselines. Sampling took place over 166 days (midsummer to midwinter). Microbial biomass (as measured by fumigation-extraction and substrate-induced respiration procedures) and dehydrogenase activity both recovered rapidly, but remained consistently lower in the fumigated than in untreated samples at both forest sites and at the moister of the two pasture sites. Bacterial numbers also recovered rapidly. Fungal hyphal lengths were, on average over 166 days, 25% lower in the fumigated soils. Levels of mineral N were initially highest in the fumigated soils, but declined with time. Fumigation generally had no detectable effects on the subsequent rates of net N mineralization and little effect on nitrification rates. Fumigation almost totally eliminated protozoa, with one to three species being recovered on day 0; the numbers recovered most rapidly in the moist forest soil and slowly in the dry pasture soil. The recoionization rate of protozoan species was similar in all soils, with species numbers on day 110 being 33 and 34 in the fumigated and untreated soils, respectively. Nematodes were eliminated by fumigation; recolonization was first detected on day 26 but by day 166, nematode numbers were still lower in fumigated than in untreated soils, the abundance being 10 and 62 g-1 soil and diversity 10 and 31 species, respectively. Overall, the results suggest that protozoan and nematode populations and diversities could provide a useful medium-term ecological index of the recovery in comprehensive soil biological activity following major soil pollution or disturbance.  相似文献

Summary Soil pH, total organic C, total N, exchangeable Al, available P, CO2 evolution, microbial biomass C and N, phosphatase and dehydrogenase activities were determined in acid soils sampled under spruce subjected to acid deposition, before and after liming. A slight decrease in pH values was observed from the edge of a tree canopy to the base of the trunk in acid soils. Liming drastically reduced exchangeable Al and increased CO2 evolution, microbial biomass, and the metabolic quotient. The microbial biomass C to total organic C ratio increased after liming but did not reach 2%, the average value considered valid in soils where the C content is in equilibrium, that is when C inputs are equal to C outputs. The microbial biomass C:N ratio decreased after liming, thus indicating that bacteria became predominant over fungi when soil acidity decreased. Dehydrogenase activity but not phosphatase activity was increased by liming. The decrease in phosphatase activity was not completely related to the increase in available P, but was also dependent on microbial growth and the decrease in acid phosphatase, the predominant component of acid soils.  相似文献

Summary We studied the build-up and turnover of microbial biomass following the addition of farmyard manure to an unmanured soil and to soils from a long-term experiment in which different levels of farmyard manure had been applied for the last 23 years. The application of farmyard manure at 15–90 t ha-1 to previously unmanured soil increased the microbial biomass during the first 3 months of incubation but a gradual decline occurred with further incubation for up to 12 months. Microbial biomass C was positively correlated with soil organic C and ranged from 1.8% to 2.2% of organic C after 12 months of farmyard manure applications. Biomass turnover increased with the application of farmyard manure, ranging from 0.81 to 0.87 year-1 with various levels of manure. Amendment of soils from the long-term manure experiment with various levels of farmyard manure led to a build-up and decline in biomass C as seen in the unmanured soils, but biomass C was higher in all treatments compared to the corresponding unmanured soil treatments. Biomass turnover was greater compared to the unmanured soil treatments and it decreased with increasing levels of farmyard manure. The average soil respiratory activity increased with increasing levels of farmyard manure, but respiratory activity per unit of biomass C decreased with increasing levels of manure. Enzyme activities were greater in long-term manured soils compared to unmanured soils amended with various levels of manure. There was a significant correlation between biomass C and enzyme activities.  相似文献

Modification of soil environment by different farming practices can significantly affect crop growth. Tillage causes soil disturbance, altering the vertical distribution of soil organic matter and plant nutrient supplies in the soil surface, and it may affect the enzyme activity and microbial biomass which are responsible for transformation and cycling of organic matter and plant nutrients. In this study, the influence of three conventional tillage systems (shallow plowing, deep plowing and scarification) at different depths on the distribution and activity of enzymes, microbial biomass and nucleic acids in a cropped soil was investigated. Analysis of variance for depth and tillage showed the influence of the different tillage practices on the activity of some enzymes and on the nucleic acids. Glucosidase, galactosidase, nitrate reductase and dehydrogenase activity were significantly affected by the three tillage modalities. Activity in the upper layer (0–20 cm) was higher in the plots tilled by shallow plowing and scarification than in those tilled by deep plowing. Positive relationships were observed between the soil enzymes themselves, with the exception of urease and pyrophosphatase activity. Moreover, significant correlations were found between DNA and β-galactosidase, and between RNA and β-glucosidase, β-galactosidase, alkaline phosphatase and phosphodiesterase. α-Glucosidase, β-galactosidase, alkaline phosphatase and phosphodiesterase were highly correlated with biomass C determined by the fumigation-extraction method. The effects of conventional and biological farming systems on soil P dynamics were studied by measuring some microbiological parameters after 13 years of different cropping systems. The treatments included control, biodynamic, bio-organic, and conventional plots and a mineral fertilizer treatment. The farming systems differed mainly in the form and quantity of nutrients applied and in the plant protection strategies. The results of a sequential fractionation procedure showed that irrespective of the form of P applied, neither 0.5 M NaHCO inf3 sup- nor 0.1 M NaOH-extractable organic P, but only the inorganic fractions, were affected. The residual organic P, not extracted by NaHCO3 or NaOH was increased in the biodynamic and bio-organic plots. The soil microbial biomass (ATP content) and the activity of acid phosphatase were also higher in both biologically managed systems. These results were attributed to the higher quantity of organic C and organic P applied in these systems, but also to the absence of or severe reduction in chemical plant protection. The relationship between acid soil phosphatase and residual organic P was interpreted as an indication that this fraction might be involved in short-term transformations. The measurement of the intensity, quantity, and capacity factors of available soil P using the 32P isotopic exchange kinetic method showed that P could not be the factor limiting crop yield in the biological farming systems. The kinetic parameters describing the ability of P ions to leave the soil solid phase, deduced from isotopic exchange, were significantly higher for the biodynamic treatment than for all other treatments. This result, showing a modification of chemical bonds between P ions and the soil matrix, was explained by the higher Ca and organic matter contents in this system.  相似文献

The effects of salinity and Mg2+ alkalinity on the size and activity of the soil microbial communities were investigated. The study was conducted along the border area of the alluvial fan of the Taolai River. Thirty soil samples were taken which had an electrical conductivity (EC) gradient of 0.93-29.60 mS cm−1. Soil pH ranged from 8.60 to 9.33 and correlated positively with Mg2+/Ca2+ ratio, exchangeable Mg2+ percentage and HCO3+CO32−. Mg2+/Ca2+ varied considerably from 3.04 to 61.31, with an average of 23.03. Exchangeable Mg2+ percentage generally exceeded 60% and had a positive correlation with Mg2+/Ca2+. HCO3+CO32− averaged 1.63 cmol kg−1 and usually did not exceed 2.0 cmol kg−1.Microbial biomass, indices of microbial activity and the activities of the hydrolases negatively correlated with Mg2+/Ca2+ or exchangeable Mg2+ percentage. Biomass C, biomass N, microbial quotient (the percentage of soil organic C present as biomass C), biomass N as a percentage of total N, potentially mineralizable N, FDA hydrolysis rate and arginine ammonification rate decreased exponentially with increasing EC. The biomass C/N tended to be lower in soils with higher salinity and Mg2+ alkalinity, probably reflecting the bacterial dominance in microbial biomass in alkalized magnesic soils. The metabolic quotient (qCO2) positively correlated with salinity and Mg2+ alkalinity, and showed a quadratic relationship with EC, indicating that increasing salinity and Mg2+ alkalinity resulted in a progressively smaller, more stressed microbial communities which was less metabolically efficient. Consequently, our data suggest that salinity and Mg2+ alkalinity are stressful environments for soil microorganisms.  相似文献

Summary Dehydrogenase activity was studied in typical soils of the Northwest German Lower Plains in order to test some criticisms raised by nannipieri et al. (1990) about methodology. We found that in the same soils with the sampe crop, dehydrogenase activity varies significantly. In the field dehydrogenase activity was dependent on the soil type and not on the cropping system. There is no adequate explanation of these findings. Dehydrogenase activity is a parameter of soil microbiological activity and is influenced by many factors. Either purely accidental the results are or dehydrogenase activity is affected by unknown ecological interactions and by the composition of soil microflora. We conclude that without simultaneous investigations of other microbiological parameters (microbial biomass, ATP levels, enzyme activities, etc.) the measurement of dehydrogenase activity creates confusion and may impede valid ecological comparisons of soils.Dedicated to Prof. H.-P. Summary Based on the reduction of dimethylusulfoxide (DMSO) to dimethylsulfide (DMS) by microorganisms, a simple, rapid, sensitive and inexpensive method for the determination of microbial activity in soil samples was developed. When DMSO was added to samples, DMS appeared immediately in the gas phase, which was quantitatively analyzed by gas chromatography. The DMS liberation rate was constant for several hours. The reaction immediately starts and its linearity indicate that neither the physiological state nor the number of organisms were changed by the assay. DMSO reduction is widespread among microorganisms; out of 144 strains tested (both fungi and bacteria) only 5 were unable to carry out this reaction. The reaction in soil samples was strongly inhibited by toluene, cyanide, azide, or by fumigation, but was considerably stimulated by glucose. These findings demonstrate that the reaction was due to the activity of microorganisms. The DMSO reduction in different soil samples was significantly correlated with arginine ammonification and heat output (r>0.9). A good correlation was observed with the organic-matter content (r = 0.74), but not with microbial numbers, clay content, or the pH of the soil. Standard deviations of less than 10% were routinely found. Furthermore, the method is sufficiently sensitive to allow measurements of activity in very small samples (< 0.1 g). For example, a microbial activity profile can be established for a single soil aggregate, revealing marked differences in activity on the outside and in the interior.  相似文献

A long-term experiment, which started in 1971 near Perugia, central Italy, was performed to investigate the effect of different crop residue management practices and rotation systems on some soil properties. Twenty years after the beginning of the experiment, chemical (organic C, total N, humified organic C, humic and fulvic acids), microbiological and biochemical parameters (microbial biomass, global hydrolase activity, dehydrogenase and catalase activities) were investigated. Two crop residue management practices were used in the experiment, i.e. removal (RCR soils) and burial (BCR soils). These treatments were factorially combined with eight rotation systems, i.e. five maize-wheat rotations of different lengths (M-1W, M-2W, M-3W, M-4W and M-5W) and three continuous wheat systems with different fertilization inputs, from 150 to 250 kg N ha–1. Soil samples were collected in the spring of 1991 for chemical determinations, and in the spring and autumn of 1992, 1993 and 1994, for microbiological and biochemical determinations. All soil chemical, microbiological and biochemical parameters investigated showed significant differences depending on the management of the crop residues. The BCR soils showed more favourable characteristics. In contrast, few significant effects were observed in relation to rotation and N-fertilization treatment. Significant correlations were found between organic-C content and all microbiological and biochemical parameters, as well as between the microbiological and biochemical parameters themselves, indicating that organic-C content plays an important role in determining the level of soil enzyme activity and, consequently, of soil fertility. This experiment showed that burying crop residues in soil can be considered good agronomic practice, which may help limit the gradual depletion of soil organic matter and improve the chemical properties of the soil.  The size and activity of the soil microbial biomass in grazed pastures was compared on the main grazing area and on stock camp areas where animals congregate. Two sites were on hill country and three on gently sloping border-dyke irrigated land. Due to the transfer of nutrients and organic matter to the camp areas via dung and urine there was an accumulation of soil organic C, organic and inorganic P and S and soluble salts in the camp areas. Soil pH also tended to be higher in camp areas due to transfer of alkalinity by the grazing animals. Water soluble organic C, microbial biomass C and basal respiration were all higher in soils from camp areas but the proportion of organic C present as microbial C and the microbial respiratory quotient were unaffected. Microbial activity as quantified by arginine ammonification rate and fluorescein diacetate (FDA) hydrolysis was higher in camp than non-camp soils but dehydrogenase activity remained unaffected. Activities of protease, histidase, urease, acid phosphatase and aryl-sulphatase were all higher in stock camp soils. The activities of both histidase and aryl-sulphatase were also higher when expressed per unit of microbial biomass C, indicating that the increased activity was the result of increased enzyme production by the microbial community. Prolonged regular applications of dairy shed effluent (diluted dung and urine from cattle) to a field had a similar effect to stock camping in increasing soil organic matter content, nutrient accumulation and soil biological activity. It was concluded that the stock camping activity of grazing animals results in an increase in both the fertility and biological activity in soils from camp areas at the expense of these properties on the main grazing areas. Soil amendment with manures from intensive animal industries is nowadays a common practice that may favorably or adversely affect several soil properties, including soil microbial activity. In this work, the effect of consecutive annual additions of pig slurry (PS) at rates of 30, 60, 90, 120 and 150 m3 ha−1 y−1 over a 4-year period on soil chemical properties and microbial activity was investigated and compared to that of an inorganic fertilization and a control (without amendment). Field plot experiment conducted under a continuous barley monoculture and semiarid conditions were used. Eight months after the fourth yearly PS and mineral fertilizer application (i.e. soon after the fourth barley harvest), surface soil samples (Ap horizon, 0-15 cm depth) from control and amended soils were collected and analysed for pH, electrical conductivity (EC), contents of total organic C, total N, available P and K, microbial biomass C, basal respiration and different enzymatic activities. The control soil had a slightly acidic pH (6.0), a small EC (0.07 dS m−1), adequate levels of total N (1.2 g kg−1) and available K (483 mg kg−1) for barley growth, and small contents of total organic C (13.2 g kg−1) and available P (52 mg kg−1). With respect to the control and mineral fertilized soils, the PS-amended soils had greater pH values (around neutrality or slightly alkaline), electrical conductivities (still low) and contents of available P and K, and slightly larger total N contents. A significant decrease of total organic C was observed in soils amended at high slurry rate (12.3 g kg−1). Compared with the control and mineral treatments, which produced almost similar results, the PS-amended soils were characterized by a higher microbial biomass C content (from 311 to 442 g kg−1), microbial biomass C/total organic C ratio (from 2.3 to 3.6%) and dehydrogenase (from 35 to 173 μg INTF g−1), catalase (from 5 to 24 μmol O2 g−1 min−1), BAA-protease (from 0.7 to 1.9 μmol  g−1 h−1) and β-glucosidase (from 117 to 269 μmol PNP g−1 h−1) activities, similar basal respirations (from 48 to 77 μg C-CO2 g−1 d−1) and urease activities (from 1.5 to 2.2 μmol  g−1 h−1), and smaller metabolic quotients (from 6.4 to 7.7 ng C-CO2 μg−1 biomass C h−1) and phosphatese activities (from 374 to 159 μmol PNP g−1 h−1). For example, statistical analysis of experimental data showed that, with the exception of metabolic quotient and total organic C content, these effects generally increased with increasing cumulative amount of PS. In conclusion, cumulative PS application to soil over time under semiarid conditions may produce not only beneficial effects but also adverse effects on soil properties, such us the partial mineralization of soil organic C through extended microbial oxidation. Thus, PS should not be considered as a mature organic amendment and should be treated appropriately before it is applied to soil, so as to enhance its potential as a soil organic fertilizer.  相似文献

Summary Dehydrogenase activity (a measure of microbial biomass), urease activity and CO2 evolution were measured in soils planted to rice (Oryza sativa L.) under three different agricultural practices prevalent in hill regions. The effects of hill slope, terrace and valley agriculture were investigated for two cropping seasons. The valleys and terraces were kept flooded during each cropping season while the hill slopes were cultivated with dryland practices. The type of agricultural practice and the date of observation had a significant influence on enzyme activity and CO2 evolution. A positive correlation was observed between dehydrogenase and urease activity and soil moisture content. Dehydrogenase and urease activity and soil respiration were positively correlated among themselves. The activity of both enzymes and CO2 evolution were highest in valley soils followed by terrace and hill-slope sites.  相似文献

Soil organic matter level, soil microbial biomass C, ninhydrin-N, C mineralization, and dehydrogenase and alkaline phosphatase activity were studied in soils under different crop rotations for 6 years. Inclusion of a green manure crop of Sesbania aculeata in the rotation improved soil organic matter status and led to an increase in soil microbial biomass, soil enzyme activity and soil respiratory activity. Microbial biomass C increased from 192 mg kg–1 soil in a pearl millet-wheat-fallow rotation to 256 mg kg–1 soil in a pearl millet-wheat-green manure rotation. Inclusion of an oilseed crop such as sunflower or mustard led to a decrease in soil microbial biomass, C mineralization and soil enzyme activity. There was a good correlation between microbial biomass C, ninhydrin-N and dehydrogenase activity. The alkaline phosphatase activity of the soil under different crop rotations was little affected. The results indicate the green manuring improved the organic matter status of the soil and soil microbial activity vital for the nutrient turnover and long-term productivity of the soil. Routine soil testing procedures that are rapid and accurate are needed to evaluate C and N mineralization in agricultural soils in order to determine soil quality and fertility. Laboratory methods were compared for their usefulness in determining soil microbial biomass and potential activity in a Weswood silty clay loam (fine, mixed, thermic Fluventic Ustochrept) subjected to long-term tillage, crop sequence, and N-fertilizer management practices. The methods included basal soil respiration, net N mineralization during a 10-day incubation, soil microbial biomass C with the chloroform fumigation-incubation technique with and without subtracting a control value, soil microbial biomass N with the chloroform fumigation-incubation technique, substrate-induced respiration, and arginine ammonification. All methods were highly correlated with each other and, therefore, appear to adequately reflect soil microbial biomass and potential activity under laboratory conditions. The longer incubation times used with the basal soil respiration, N mineralization, and microbial biomass C and N assays resulted in higher correlations and lower variation among replications compared to the shorter incubation times used with substrate-induced respiration and arginine ammonification. The relatively rapid procedural time (3 h) required for the latter two assays could make these methods more attractive for routine soil testing, although multiple assays on the same sample may be necessary because these methods are less precise than the incubation methods that require 10 days.  相似文献

Summary Fluoride-induced changes of chemical properties and microbial activities in humus soils were investigated in 12-week lysimeter experiments. The mull soil showed the highest F-adsorption capacity, in which 94% of the fluoride added was retained after addition of 4.5 mg F/cm2 as NaF. The moder and mor soils only adsorbed 52% and 41%, respectively. F-additions induced leaching of organic matter, Fe, Al and P and increases in soil pH in the moder and mor. In contrast no Al, Fe and P was leached from the mull and only minor amounts of organic matter dissolved after a single application of 4.5 mg F/cm2. Treatments with NaF up to 3700 mg F/kg did not significantly reduce respiration in any of the humus forms. Dehydrogenase, alkaline phosphatase and arylsulfatase activities as well as nitrification were inhibited at much lower F-additions in the moder and mor soils. A significant decrease in ammonification was observed in the moder. In contrast, microbial processes in the mull soil were not inhibited. This is due to its high adsorption capacity and the relatively low toxicity of F-ions. According to computations using GEOCHEM, the F-ion was the most abundant species in mull lysimeter leachates. Leachates of moder and mor soils contained mainly AIF-complexes (90%–99%). The lack of any effects from NaF treatment on soil respiration is attributed to the observed positive effects, e.g. dissolution of organic matter, desorption of P and increases in soil pH.  相似文献

Summary In pot experiments, lime and mineral salts were applied to forest soils taken from three different sites. Microbial biomass, basal respiration, N turnover, and the activities of protease, phosphatase, and CM-cellulase were determined monthly. The fertilized soils showed a great increase in biomass, respiration and N turnover, and some increase in protease activity, whereas phosphatase activity decreased in comparison with the untreated soils. Estimates of the number of colony-forming units of different classes of microorganisms showed an increase in treated soils in most cases. The untreated soils had the lowest biological activity but the maximum application of lime and mineral salts did not automatically lead to maximum activity.  相似文献

Summary A range of soil microbiological parameters were measured at intervals throughout the growing season of a potato crop. Treatments applied to the soil at sowing were zero N fertilisation of N fertilisation at 120 kg N ha–1, either alone or supplemented with straw or sucrose at 1200 kg C ha–1. C and N flushes determined by fumigation-incubation and fumigation-extraction, and substrate-induced respiration, were measured as indicators of microbial biomass. Microbial activity was measured as respiration (CO2 production) and dehydrogenase activity (formazan production). The greatest effects were obtained from the addition of N plus sucrose. Both biomass size and activity were significantly stimulated for up to 25 days after incorporation, with the magnitude of the effects consistently diminishing over time. By 125 days after planting, there was no detectable legacy from any of the treatmentson any of the biomass parameters that were measured, and all values had reverted to those prevalent at planting. There was no consistent effect from adding N, either alone or supplemented with straw, on any of the biomass parameters. There was no evidence for crop-induced stimulation of the biomass. The experiment demonstrates that biomass is only influenced where the quantity, quality, and rate of incorporation of C into the soil is appropriate, in this case, only by adding C as a pulse of sucrose.  相似文献

Near-infrared reflectance spectroscopy (NIRS) is known for its inexpensiveness, rapidity and accuracy and may become a useful tool for the assessment of soil quality. Objectives were (i) to evaluate the ability of NIRS to predict several chemical and biological properties of organically managed arable soils as well as the properties of grain yield from winter cereals for a closed population and (ii) to test whether the use of field-moist and pre-treated (quick-freezing followed by freeze-drying and grinding) samples will generate similar results. One hundred and sixteen soil samples from nine organically managed farms from Germany sampled in 2005 and 2006 were used for this investigation. Spectra of the near-infrared region (including the visible range, 400–2500 nm) from field-moist (<2 mm) or pre-treated soil samples were recorded. A modified partial least-square regression method and cross-validation were used to develop an equation over the whole spectrum (first–third derivation). For the pre-treated soils, good predictions were obtained for pH, contents of organic C, total N, plant-available P (Olsen) and exchangeable K (calcium-acetate-lactate (CAL)), contents of microbial biomass C and N (Cmic and Nmic) and ergosterol, basal respiration, metabolic quotient, the ratio of organic C/total N, the grain yield of winter cereals and grain nitrogen uptake. The RSC (the ratio of standard deviation of laboratory results to standard error of cross-validation) was greater than 2.0, the correlation coefficients (r) of a linear regression (measured against predicted values) were greater than or equal to 0.9 and the regression coefficients (a) ranged from 0.9 to 1.1. Similar good predictions were obtained if field-moist samples were used, with the exception of P (Olsen), K (CAL), metabolic quotient, grain yield of winter cereals and grain nitrogen uptake (satisfactory predictions) and ergosterol content (unsatisfactory prediction). Good predictions of the contents of Mg (CaCl2) and microbial biomass P (Pmic) were achieved for field-moist but not for pre-treated samples. Despite sample preparation, only satisfactory predictions were obtained for the ratios of Cmic/Nmic and ergosterol/Cmic and grain nitrogen content (1.4RSC2.0, r0.8 and 0.8a1.2). However, unsatisfactory predictions for field-moist and pre-treated samples were achieved for the content of P (CAL), the nitrogen mineralisation rate and the ratios of Cmic/Pmic and basal respiration/nitrogen mineralisation rate. Our results demonstrate that biological soil properties can be predicted with NIRS for closed populations in both sample states. The pre-treatment should be used if samples have to be stored prior to infrared measurements for periods longer than a month.  相似文献

The composition of microbial communities and the level of enzymatic activity in the soil are both important indicators of soil quality, but the mechanisms by which a soil bacterial community is generated and maintained are not yet fully understood. Two soil samples were collected from the same location, but each had been subjected to a different long-term fertilization treatment and was characterized by different microbial diversity, biomass and physicochemical properties. These samples were γ-sterilized and swap inoculated. Non-sterilized soil samples along with sterilized and inoculated soil samples were incubated for eight months before their nutrient content, microbial biomass, enzymatic activity and bacterial composition were analyzed. Total phosphorus, and potassium concentrations along with the overall organic matter content of the non-sterilized soil were all equal to those of the same soil that had been sterilized and self/swap inoculated. Additionally, the microbial biomass carbon concentration was not affected by the specific inoculum and varied only by soil type. The activities of catalase, invertase, urease, protease, acid phosphatase and phytase were smaller in the sterilized soils that had been inoculated with organisms from chemical fertilizer amended soil (NPK) when compared to sterilized soil inoculated with organisms from manure and chemical fertilizer amended soil (NPKM) and non-sterilized soil samples. Bacterial 16S rRNA examined by 454-pyrosequencing revealed that the composition of bacterial community reconstructed by immigrant microbial inoculum in the soil was mainly influenced by its physicochemical properties, although the microbial inoculum contained different abundances of bacterial taxa. For example, the pH of the soil was the dominant factor in reconstructing a new bacterial community. Taken together, these results demonstrated that both soil microbial composition and functionality were primarily determined by soil properties rather than the microbial inoculum, which contributed to our understanding of how soil microbial communities are generated and maintained.  相似文献
