ABSTRACT Spread of US-1 and US-8 isolates of Phytophthora infestans were observed in field plots of potato (cv. Russet Burbank) grown in Pullman, WA, in 1996 and 1997. Infected greenhouse-grown potato plants with similar lesion numbers for both strains were transplanted to field plots with four replications. Spread of the pathogen was favored by sprinkler irrigation during evening hours. Diseased leaves and stems were sampled over time to determine the spread of US-1 and US-8 isolates. In 1996, late blight developed in two of the four replications (105 and 87 total isolates recovered). From those two replications, two US-1 isolates were recovered, both from the same replication. Nine isolates from one replication and six isolates from another displayed a phenotype different from the initial isolates, as determined by compatibility type, allozyme genotype, and restriction fragment length polymorphism genotype. These putative recombinant isolates may have arisen from sexual recombination between the US-1 and US-8 isolates. The remaining isolates were of the US-8 strain. In 1997, late blight developed in all four replications (123, 122, 81, and 34 total isolates recovered). One US-1 isolate was recovered (out of 123) from one replication and three (out of 122) from another, and the remaining isolates were of the US-8 strain. Isolates with phenotypes differing from the initial isolates were not recovered in 1997. In both years, oospores were not observed in the plant tissue examined. The low number of putative recombinant isolates in 1996 and their absence in 1997 suggests that sexual reproduction between US-8 and US-1 isolates in a field setting is a rare event. The predominance of US-8 isolates recovered is a measure of the increased fitness and aggressiveness of the US-8 isolates relative to the US-1 isolate used in this study. This further substantiates the increased aggressiveness of the US-8 genotype observed on excised tissues and potted plants in previous laboratory and greenhouse studies.  相似文献

ABSTRACT The aggressiveness of 22 isolates of Phytophthora infestans collected from naturally infected potato plants in the Columbia Basin of Washington and Oregon was determined on detached potato leaflets at 18 degrees C in an incubator. Selected isolates were evaluated on whole plants in a greenhouse. Aggressiveness was measured by using the area under the lesion expansion curve (AULEC), incubation period, latent period, sporulation capacity, and lesion size on detached leaflets and the area under the disease progress curve and sporulation capacity on whole plants. The detached-leaflet assay was useful in that a large number of isolates were tested, several components of aggressiveness were studied, and significant differences were found among isolates. Significant variation for components of aggressiveness was found within and among isolates classified according to genotype. Significant interactions among isolates and cultivars were found for some components of aggressiveness, so results were pooled according to cultivar. On average, US-8 and US-11 isolates had higher AULEC scores, indicating aggressiveness higher than that of US-7, US-6, and US-1 genotypes. One US-8 genotype isolate had a higher standardized sporulation capacity than isolates of the other genotypes. US-6 genotype isolates were the least aggressive group, as indicated by low AULEC, sporulation capacity, and lesion size values. The replacement of the US-1 genotype by the US-8 genotype in the Columbia Basin may be partially explained by the increased aggressiveness of US-8 isolates. Additionally, potato growers may need to shorten intervals between fungicide applications and begin applications earlier.  相似文献

ABSTRACT Fungal, host, and environmental factors affecting sexual reproduction of Phytophthora infestans in planta were studied. Intact and detached leaves were coinoculated with sporangia of various combinations of A(1) and A(2) mating-type isolates; leaves were incubated under various conditions, and oospore production was estimated microscopically within whole, clarified leaflets. Some A(1) + A(2) isolate combinations were more reproductive than others, whereas some potato genotypes better supported oospore formation than others. Tomato usually supported more oospore formation than potato. To induce oospore formation, A(1) and A(2) sporangia were usually mixed at a 1:1 ratio. Ratios of 1:19 to 19:1, however, also allowed abundant production of oospores. Optimal temperatures for sexual sporulation ranged from 8 to 15 degrees C, but oospores also were produced at 23 degrees C. Oogonia developed 5 to 6 days after sporangial coinoculation, and oospores developed after 8 to 10 days. Light had little effect on oospore formation in both tomato and potato leaves provided that initial lesions were established under photoperiodic conditions. Although A1 and A(2) sporangia usually were mixed before inoculation on leaves to obtain oospores, we found that discrete A(1) and A(2) lesions produced on opposite sides of the midvein of tomato leaves also induced oospore formation in the midvein and adjacent tissues. Oospores also formed when the two halves of the leaves were cut and separated at 3 days after sporangial coinoculation, which corresponded with the appearance of late blight lesions. The continuous supply of moisture to infected leaves was essential to oospore production. No oospores or oogonia formed in severely diseased plants kept at 50 to 80% relative humidity. Such plants did allow some oospore formation when kept continuously wet for 2 weeks in plastic boxes or tents. Detached leaves floated on water supported the highest sexual sporulation. Under optimal conditions of wetness and temperature, as many as 100 oospores per mm(2) of tissue were observed.  相似文献

Thirty-six isolates of Phytophthora infestans originating from three different potato growing regions in the Netherlands were tested for their aggressiveness to leaves of potato cultivar Bintje under controlled conditions. Measurements of latent period, maximal growth rate, infection efficiency and sporulation intensity were made and a composite aggressiveness index was calculated. Large variation in aggressiveness was present among isolates for each regional P. infestans population studied. The three populations differed significantly in latent period, but not for maximal growth rate and infection efficiency. Phenotypic variation existed for all components of aggressiveness and the aggressiveness index in P. infestans from each regional source. No association was found between mating type and aggressiveness. It is concluded that high levels of variation for aggressiveness are being generated and maintained through sexual reproduction in P. infestans strains from regional potato growing practices.  相似文献

ABSTRACT Sixty Ecuadorian isolates of Phytophthora infestans from potato and 60 isolates from tomato were compared for dilocus allozyme genotype, mitochondrial DNA haplotype, mating type, and specific virulence on 11 potato R-gene differential plants and four tomato cultivars, two of which contained different Ph genes. Restriction fragment length polymorphism (RFLP) fingerprints of subsamples of isolates from each host were compared by using RG57 as the probe. All potato isolates had the allozyme genotype, haplotype, and mating type of the clonal lineage EC-1, which had been previously described in Ecuador. With the same markers, only one isolate from tomato was classified as EC-1; all others belonged to the globally distributed US-1 clonal lineage. RFLP fingerprints of isolate subsets corroborated this clonal lineage classification. Specific virulence on potato differentials was broadest among potato isolates, while specific virulence on tomato cultivars was broadest among tomato isolates. Some tomato isolates infected all tomato differentials but no potato differentials, indicating that specific virulence for the two hosts is probably controlled by different avirulence genes in P. infestans. In two separate experiments, the diameters of lesions caused by nine isolates from potato and 10 from tomato were compared on three tomato and three potato cultivars. All isolates produced larger lesions on the host from which they were isolated. No isolates were found that were highly aggressive on both tomato and potato. We conclude that there are two different populations of P. infestans in Ecuador and that they are separated by host.  相似文献

Isolates of Phytophthora infestans were obtained from late blighted plants from several potato-growing regions of Uruguay in 1998 and 1999. Of these, 25 representative isolates (4 from 1998, 21 from 1999) from the main potato-growing areas of the country, were characterised in terms of mating type, metalaxyl resistance, allozyme genotype, mitochondrial haplotype, RG57 fingerprint (1999 isolates only) and pathotype. All isolates proved to be A2 mating type, monomorphic and homozygous at the loci coding for glucose-6-phosphate isomerase and peptidase (Gpi 100/100, Pep 100/100) and to possess mitochondrial haplotype IIa. Metalaxyl-resistant isolates constituted 92% of the total. All the 1999 isolates possessed the same RG57 fingerprint, which was that previously reported as associated with the clonal lineage BR-1 from Brazil and Bolivia, which is also A2, Gpi 100/100, Pep 100/100. Most of the isolates displayed broad-spectrum virulence and five carried virulence to 10 of the 11 R genes tested despite the absence of R genes in commercially grown potato cultivars. It was concluded that the Uruguayan P. infestans isolates resembled isolates from neighbouring South American countries, notably Brazil, and belong to the new populations of the pathogen now predominant in many countries.  相似文献

ABSTRACT Phytophthora infestans causes a destructive disease on tomato and potato. In North Carolina (NC) potatoes are mostly grown in the east, whereas tomatoes are grown in the mountainous areas in the western part of the state. Five genotypes of P. infestans were identified from 93 and 157 isolates collected from tomato and potato over a 5 year period between 1993 and 1998. All isolates collected from potato in eastern NC were the US-8 genotype, whereas only a single isolate was the US-1 genotype. Tuber blight was found on immature daughter tubers in a single field in 1997, however infection on mature tubers was not observed. Within potato fields, a range of sensitivity to metalaxyl was observed among isolates but all were either intermediate or highly resistant to the fungicide. In contrast, isolates from tomatoes included previously reported US-7 and US-8 genotypes and two new genotypes called US-18 and US-19 (A2 mating type, allozyme genotype Gpi 100/100 and Pep 92/100). These genotypes had unique restriction fragment length polymorphism banding patterns, were sensitive to metalaxyl, and have not been reported elsewhere. All genotypes, with the exception of the US-1, were the Ia mitochondrial haplotype. Thus, isolates of P. infestans from tomato were more genetically diverse over time in NC than those from potato and include two new genotypes that are sensitive to metalaxyl.  相似文献

The population of Phytophthora infestans in Brazil consists of two clonal lineages, US-1 associated with tomatoes and BR-1 associated with potatoes. To assess whether host specificity in these lineages resulted from differences in aggressiveness to potato and tomato, six aggressiveness-related epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion area (LA), lesion expansion rate (LER) and sporulation at several lesion ages (SSLA) – were measured on detached leaflets of late blight-susceptible potato and tomato plants. Infection frequency of US-1 was similar on potato and tomato leaflets, but IF of BR-1 was somewhat reduced on tomato. Incubation period was longer on both hosts with US-1, although this apparent lineage affect was not significant. Overall there was no host effect on IP. On potato, BR-1 had a shorter LP (110·3 h) and a larger LA (6·5 cm2) than US-1 (LP = 162·0 h; LA = 2·8 cm2). The highest LER resulted when isolates of BR-1 (0·121 cm2 h−1) and US-1 (0·053 cm2 h−1) were inoculated on potato and tomato leaflets, respectively. The highest values of the area under the sporulation capacity curve (AUSC) were obtained for isolates of US-1 inoculated on tomato leaflets (6146) and for isolates of BR-1 on potato leaflets (3775). In general, higher values of LA, LER, SSLA and AUSC, and shorter values of LP were measured when isolates of a clonal lineage were inoculated on their original host than with the opposite combinations. There is evidence that there are quantitative differences in aggressiveness components between isolates of US-1 and BR-1 clonal lineages that probably contribute to host specificity of P. infestans populations in Brazil.  相似文献

ABSTRACT The temporal and spatial patterns of Phytophthora infestans population genetic structure were analyzed in the Del Fuerte Valley, Sinaloa, Mexico, during the crop seasons of 1994 to 1995, 1995 to 1996, and 1996 to 1997 by geographical information systems. Isolates of P. infestans were obtained from infected tissue of tomato and potato collected from two areas: (i) where both potatoes and tomatoes are grown, and (ii) where only tomatoes are grown. The isolates were characterized by mating type, allozymes at the glucose-6-phosphate isomerase and peptidase loci, restriction fragment length polymorphism (RFLP) fingerprint with probe RG57, metalaxyl sensitivity, and aggressiveness to tomato and potato. The results suggest presence of an asexual population with frequent immigrations from outside the valley. There was a shift of mating type in the population from predominantly A2 to completely A1 in this period. The co-occurrence of mating types was restricted to very few fields in the area around Los Mochis where tomato and potato crops are grown. Genotype variation based on allozyme analysis and mating type was low with only one genotype affecting both crops each year. The genotypes affecting both crops were the only genotypes highly aggressive to both tomato and potato in laboratory aggressiveness tests and the only genotypes widespread on both the tomato and potato crops in the valley each year. These predominant genotypes were highly resistant to the fungicide metalaxyl. Data on metalaxyl sensitivity indicate that allozyme analysis can discriminate between sensitive and resistant isolates in the Del Fuerte Valley. RFLP analysis with the probe RG57 gives further discrimination of genotypes within an allozyme genotype. In the 1995 to 1996 season, four different RFLP genotypes were found within an allozyme genotype. However, there were five other dilocus allozyme genotypes that could not be further split by RFLP analysis in 1995 to 1996 and 1996 to 1997 seasons. Spatial analysis of genotypes suggests that each season individual fields near Los Mochis became infected with one or more genotypes, but only a single genotype, aggressive on both potato and tomato, occurred south and east to the Guasave area.  相似文献

A total of 36 single-lesion isolates were collected from 9 crops of potato and 13 of tomato in different regions of Hungary in the past decade, particularly in 1998. These were analysed for mating type, sensitivity to metalaxyl, allozyme genotype at glucose-6-phosphate isomerase and peptidase loci and genotype at 24 loci detected using the multilocus RFLP probe RG57. The ratios of the mating types A1 to A2 were 8:9 and 4:15 among isolates recovered from potato and tomato, respectively. Resistance to metalaxyl was found more frequently among isolates from potato and in the A1 mating type. The populations were not clearly differentiated on the basis of host origin. All isolates were homozygous (100/100) at the locus for glucose-6-phosphate isomerase. Unlike in other European countries, the most common peptidase allele was 96. Genotypes at the peptidase locus were 96/96 (50%), 96/100 (27.7%) and 100/100 (16.6%). In addition, one isolate from 1993 and another from 1998, were defined as 83/96, a genotype that had not been described elsewhere. The 18 RG57 fingerprints that were observed among 36 isolates, with one exception, seem to be unique to Europe. On the basis of combined genotypic traits, 20 multilocus genotypes were designated. These data, which reveal a remarkable variability with unique genotypes of the late blight pathogen, suggest that migration and sexual and/or asexual recombination have a role in the recent evolution of the pathogen in Hungary.  相似文献

Potato fields in Denmark, Finland, Norway and Sweden were sampled for single-lesion isolates of Phytophthora infestans . The aggressiveness of the isolates was determined on detached leaflets of potato cvs Bintje (susceptible) and Matilda (moderately resistant). The aggressiveness tests were carried out in the respective home countries of the isolates, with the exception of the Danish isolates. Fifteen Danish isolates were studied in each of the other three countries, including five isolates tested in all three laboratories. Results obtained from the Danish isolates revealed substantial differences between the test laboratories for infection efficiency, lesion growth rate and sporulation capacity on detached leaflets. When the laboratory effect was taken into account, the differences in aggressiveness between the countries were generally small or inconsistent between the test cultivars and epidemiologically insignificant. By contrast, variation among isolates within countries was substantial. The magnitude of the variation depended on country and cultivar. Maximal variation for the means of the isolates was between 89 and 185 h for latent period, between 100 and 1297 sporangia mm−2 for sporulation capacity and between nearly zero and 6 mm day−1 for lesion growth rate. Typically less than 1% of sporangia were able to cause infections, except in Norway. These extraordinarily low values may be an artefact of the testing method. High variation in results between the test laboratories emphasizes the need for caution when comparing results obtained by different research groups.  相似文献

ABSTRACT The ability of Phytophthora infestans, the causal agent of potato and tomato late blight, to produce oospores in potato tuber tissue was studied in the field and under laboratory conditions. In 1998 and 2000 field experiments, the canopy of potato cvs. Alpha and Mondial, respectively, were coinoculated with A1 + A2 sporangia of the fungus, and the infected tubers collected at harvest were examined for the presence of oospores. In 1998, only 2 of 90 infected tubers had oospores, whereas none of the 90 tubers examined in 2000 had any oospores. In the latter experiment, infected tubers kept in storage up to 12 weeks after harvest had no oospores. Artificial co-inoculations of whole tubers with A1 + A2 sporangia resulted only rarely in the formation of oospores inside the tubers. Co-inoculations of potato tuber discs taken from dormant tubers 0 to 16 weeks after harvest failed to support any oospore production, whereas discs taken from sprouting tubers of >/=18 weeks after harvest allowed oospores to form. Tuber discs showed enhanced oospore formation when treated before inoculation with either sugars, amino acids, casein hydrolysate, beta-sitosterol, or chloroethylphosphonic acid. In contrast, reducing airflow into the petri dishes where potato tuber discs were incubated reduced the number of oospores produced. The number of oospores produced in tuber tissue was lower compared with that in leaf tissue regardless of the origin of isolates used. The data show that the ability of Phytophthora infestans to produce oospores in potato tuber tissue is very limited and increases with tuber aging.  相似文献

ABSTRACT Tomato fruits at the mature green stage coinoculated with A1 + A2 sporangia of Phytophthora infestans, the late blight causal fungus, showed abundant oospores in the vascular tissues, pericarp, columella, and placenta. Oospores were also formed on the surface of fruits kept in moisture-saturated atmosphere. Occasionally, oospores were enclosed between the epidermal hairs of the seed coat. In a few seeds, oospores were detected inside the embryo. The data suggest that blighted tomato fruits may carry a large number of oospores, thus making them a threatening source of blight inoculum. Such fruits may also release airborne oosporic inoculum that may introduce recombinant genotypes within a growing season. Although Phytophthora infestans is seedborne in tomato, to our knowledge, this is the first report on the occurrence of oospores in tomato seeds. Whether such tomato seeds produce blighted seedlings remains to be shown.  相似文献

A survey on the tomato late blight situation and current practices for disease management was carried out in Uganda using an informal structured questionnaire approach. Ten districts from different agroclimatic zones were selected for the survey. Phytophthora infestans isolates from tomatoes were obtained from the zones and only the A1 mating type was recovered. Tomato cultivation is practised year-round. The major commercial varieties grown, Moneymaker, Marglobe, Heinz and Roma, were susceptible to late blight. Late blight and bacterial wilt ( Pseudomonas solanacearum ) were the major diseases, with bollworm ( Helicoverpa armigera ) and thrips ( Thrips spp .) the major insect pests. Fungicides, mostly mancozeb, were the predominant control method used by farmers. Sanitation is practised by some farmers, but only in association with fungicides, to control late blight. Plant nutrition is not perceived as playing a major role in late blight management. Problems associated with pesticides and poor marketability were identified as major constraints to tomato production in Uganda.  相似文献

The aggressiveness of four Phytophthora infestans isolates collected from wild and cultivated potato species (sect. Petota ) and the level of resistance of nine Petota species were assessed in the highland tropics of Ecuador. For this, isolates of P. infestans were inoculated on whole plants of Petota species in the field and net house and six epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion size (LS), lesion growth rate (LGR), and relative area under the lesion expansion curve (RAULEC) – were measured during a single infection cycle. Additionally, host specificity was determined by testing for a significant host by pathogen interaction using the same components. The results showed significant differences among isolates of the EC-1 clonal lineage for IP, IF, and RAULEC. Significant differences among isolates were not found for the other components measured. There were significant differences in resistance among the accessions of Petota hosts tested. RAULEC, LGR, LP, and LS were in general more adequate in differentiating among the more resistant and more susceptible accessions but the importance of each component varied with host species. There was slight and inconsistent evidence for the existence of host specificity in some isolates of Petota hosts. IP was the only component for which a significant host by isolate interaction was observed and in most cases the isolates had the greatest aggressiveness on their hosts of origin.  相似文献

A collection of 241 isolates of Rhizoctonia solani obtained from potato plants grown in different areas in France was characterized for anastomosis grouping, symptomatology on tubers of different cultivars and sensitivity to three fungicides. Most isolates collected belonged to (anastomosis groups (AGs)) AG 3, but 2% and 4% of the isolates were AG 5 and AG 2-1. AG 3 and AG 2-1 isolates were mostly obtained from sclerotia on tubers, but all AG 5, some AG 3 and some AG 2-1 isolates were recovered from superficial tuber alterations, like deformations, corky or scabby lesions. Sclerotia were formed on tubers produced by healthy stem cuttings grown in soil artificially infested with AG 3, but not on tubers grown in soil infested with either AG 5 or AG 2-1. No variation in susceptibility to sclerotial formation was observed among five potato cultivars. In all cases, a large proportion of tubers showed superficial corky lesions, often associated with deformations. The proportion of tubers with lesions and deformations was highest in soil infested with AG 2-1 and significantly lower on cv. Samba in all treatments. All isolates were highly sensitive to flutolanil, iprodione and pencycuron, except the AG 5 isolates, moderately sensitive to pencycuron. These results show that, although AG 3 is the most common R. solani group on potato in France, AG 5 and AG 2-1 may be present. Isolates differed for pathogenicity. In vitro sensitivity to fungicides varied among AGs.  相似文献

ABSTRACT Sporangia germination of Phytophthora infestans isolates belonging to three clonal lineages (US-1, -7, and -8) was assessed at temperatures ranging from 10 to 25 degrees C. At 10 degrees C there were no significant differences in germination percents among US-1, -7, and -8. At 18 or 20 degrees C US-7 and -8 had significantly lower germination percents than US-1. At 21, 24, or 25 degrees C all clonal lineages had low germination percents. Sporangia of the US-7 and -8 lineages germinated more quickly at 15 degrees C (P = 0.001) during the first 2 h than did the US-1 lineage. The incubation period (IP), lesion area (LA), and sporulation per unit of lesion area (SPU) of the isolates were assessed on inoculated detached leaflets of susceptible potato cv. Norchip kept at 10, 15, 20, or 25 degrees C. In general, IP declined exponentially and LA increased exponentially with increasing temperatures. SPU had a quadratic shape, with the maximum at 15 degrees C. Averaged over all temperatures, the US-7 lineage had the shortest IP (59.3 h compared to 66.4 h for US-1 [P = 0.012] and 71.7 h for US-8 [P = 0.026]). Again, averaged over all temperatures, the US-8 lineage had a larger LA (P = 0.030) than US-1. There was no significant difference between US-7 and -1 for LA. There were no significant differences among lineages in terms of SPU. These results indicate that clonal lineages differ from each other in epidemiological attributes, but the differences can be complex.  相似文献

Isolates of the potato late-blight pathogen Phytophthora infestans , collected in several French regions from 1988 to 1992, were characterized in order to determine the structure and pathogenicity of French populations and to compare them with other European samples. All 254 French isolates tested were of the Al mating type, as were two Sicilian, five Portuguese and one Spanish isolate. Metalaxyl sensitivity of 157 isolates was investigated. The frequency of resistance to this chemical decreased in western France from 1989 to 1992. Two glucose phosphate isomerase phenotypes (90/100 and 90/90) were detected among the 46 isolates tested; the former largerly predominated (75–80%) in the samples investigated. Both phenotypes corresponded to the population recently introduced into Europe. A third genotype (86/100), typical of the former European population, was found in one isolate from Picardy, originating from an artificially infected field trial. Five isolates from Portugal and Italy were of the 100/100 phenotype. Race patterns among 26 isolates from western France were very similar, and could not be explained by local selection. The unusual composition of the Brittany population, consisting of 'new' genotypes but all belonging to the Al mating type, is possibly related to the metapopulation type of structure of P. infestans in Europe.  相似文献

Mating type, in vitro sensitivity to the phenylamide fungicide metalaxyl, and mitochondrial (mtDNA) haplotype were determined in some or all of 618 isolates of Phytophthora infestans from the years between 1978 and 1995. A2 mating type occurred infrequently in most but not all years and insensitivity to metalaxyl increased over time. After 1982, the mtDNA Ib haplotype was largely replaced (except for one isolate in 1986 and one in 1995) by two new haplotypes, Ia and IIa. Type Ia was much more common than type IIa.Approximately one quarter of these isolates (165) were compared using two components of fitness associated with aggressiveness (infection frequency × number of sporangia per lesion) on detached leaves of cultivars Maris Piper, Cara and Stirling, which were chosen as exhibiting increasing levels of race non-specific resistance. Isolates were compared with three standard isolates of low, intermediate or high aggressiveness, and the data standardised for comparison between experiments. On cvs. Cara and Stirling, but not on Maris Piper, mtDNA Ia and IIa haplotypes were more aggressive than type Ib in several separate experiments. Similarly, metalaxyl sensitive phenotypes were more aggressive than insensitive phenotypes on Cara and Stirling but not on Maris Piper. The displacement of mtDNA type Ib by types Ia and IIa over this period may have been a result of the lower aggressiveness and lack of complete insensitivity to metalaxyl in type Ib isolates.  相似文献
