ABSTRACT The root-knot nematode Meloidogyne incognita is a major pathogen of a range of important crops. Currently, control is typically achieved by the use of nematicides. However, recent work suggests that manipulating the ability of roots to slough off border cells, which then act as a decoy to the nematode, can significantly decrease damage to the roots. We investigated the attractiveness of border cells to M. incognita and the response of the nematode to border cells in close proximity. We found very limited attraction, in that nematodes did not preferentially alter direction to move toward the border cells, but a large and significant increase in nematode speed was observed once they were in the immediate vicinity of border cells. We discuss the results in the context of physical and biological mechanisms in relation to the control of pathogenic nematodes.  相似文献

为了有效防治栝蒌根结线虫,调查了安徽省潜山县栝蒌根结线虫及其发生,并分析了栝蒌根围土壤线虫的组成。通过线虫形态学鉴定,认为潜山栝蒌上的根结线虫为Meloidogyne incognita。栝萎根围土壤中有口针线虫占调查总量的50.7%,根结线虫2龄幼虫(J2)占31.8%。对当年栝萎J2在土壤表、中、下层的分布和年发生调查结果表明,J2在各层的分布量因月份不同而不同,并分别于3月初到9月中旬问出现2次虫口高峰;防治试验结果表明,除有机肥外,各试验小区在施药后49天J2虫口相对减退率在89.6%以上;施药后124天,克线丹和1.8%阿维菌素处理的小区增果率为250%~480%,收到很好的防效。  相似文献

Aphanomyces root rot ( Aphanomyces euteiches ) has become a very destructive disease in French pea crops since 1993. The host specificity of the French pea-infecting populations of this pathogen was investigated by inoculating pea, common vetch, alfalfa, broad bean and green bean with 91 pea-infecting A. euteiches isolates, originating from the main areas of infestation in France. These isolates were compared to 13 isolates from various countries and hosts (pea, green bean, alfalfa). Virulence phenotypes were defined according to the pathogenicity data on the different hosts: all isolates from France infected two to five legume species, with most infecting pea, vetch, alfalfa and broad bean. Four pathotypes were characterized within the French isolates: one type corresponded to broad host range isolates, the second was composed of isolates preferentially agressive on pea/vetch/alfalfa and weakly aggressive on broad bean, and two others corresponding to more specialized isolates that preferentially infected pea/vetch or pea/vetch/alfalfa. Most isolates from France were preferentially pathogenic on pea, like the pea-infecting isolates from other countries, but were less specialized than the alfalfa- and green bean-infecting isolates from other countries. These results suggest that A. euteiches isolates may be maintained on wild or cultivated legumes other than pea in France.  相似文献

Two controlled environment experiments were conducted to explore the hypothesis that invasion and damage caused to potato roots by the potato cyst nematode Globodera rostochiensis might result in quantitative or qualitative changes in the release of root exudates to subsequently affect the growth of Rhizoctonia solani (AG3) in the potato rhizosphere. The growth of five R. solani isolates was compared on media amended either with root exudates from G. rostochiensis-infested or uninfested potato (cv. Désirée) plants at different time intervals after the introduction of the nematodes. In Experiment 1, the growth of R. solani was higher on medium amended with potato root exudates from G. rostochiensis-infested compared to uninfested plants, collected 4, 6, 8 and 12 days after the G. rostochiensis treatments were administered. Similarly, in Experiment 2, R. solani isolates grew faster on medium amended with potato root exudates from G. rostochiensis-infested than uninfested plants. This trend was particularly pronounced at the 12-day collection. At this time, 49% of the G. rostochiensis juveniles in roots were found to belong to the juvenile moults J2 and J3, indicating that root exudates were modified during the earlier stages of juvenile invasion. Carbohydrate analysis of root exudates indicated significantly higher levels of sucrose in root exudates from G. rostochiensis-infested than uninfested plants, whereas no significant differences were found in total nitrogen content. The results are discussed to help elucidate the mechanism behind the disease complex found between G. rostochiensis and R. solani in previous field research.  相似文献

ABSTRACT Root-knot nematodes (Meloidogyne spp.) are sedentary, obligate endoparasites in plants, where they induce specialized feeding sites. The feeding sites act as strong metabolic sinks to which photosynthates are mobilized. The histopathological modifications in the nematode-induced feeding sites of artificially inoculated chickpea cv. UC 27 were qualitatively and quantitatively compared using five isolates of M. artiellia and one isolate each of M. arenaria, M. incognita, and M. javanica. All Meloidogyne isolates infected chickpea plants, but root gall thickening was significantly less for M. artiellia isolates than for the other Meloidogyne species. Nevertheless, neither the number of giant cells in the feeding site (averaging four to six) nor the area of individual giant cells was influenced by nematode species or isolate. However, the number of nuclei per giant cell was significantly smaller, and the maximum diameters of nuclei and nucleoli were significantly greater, in giant cells induced by M. artiellia isolates than in those induced by M. arenaria, M. incognita, or M. javanica. In a second experiment, M. artiellia-induced giant cells in faba bean and rapeseed also contained a small number of large nuclei.  相似文献

ABSTRACT Pathogenic and genotypic variability among four populations of Aphanomyces euteiches from individual fields in Minnesota, Wisconsin, and Oregon were investigated using pathogenicity and randomly amplified polymorphic DNA (RAPD) analyses. About 50 strains were isolated from each of two pea fields in Minnesota, and 11 and 6 strains from pea fields in Wisconsin and Oregon, respectively, using pea (Pisum sativum) as a baiting host. Pathogenic variability and host range were evaluated in greenhouse studies with five pea lines or cultivars having different levels of resistance to Aphanomyces root rot and one cultivar each of alfalfa and snap bean. All strains were pathogenic on one or more pea cultivars, and 18 and 14% were pathogenic on alfalfa and bean, respectively. Disease severity incited by different strains varied significantly on individual pea cultivars and on all hosts combined. The percentage of strains pathogenic on different hosts varied among locations. Genotypic variation among all 114 strains was evaluated with RAPD analysis. Ten decanucleotide primers detected 92 polymorphic bands. Cluster and principal coordinates analysis revealed one large group containing 102 of the 114 strains from all locations. Two closely related minor groups of strains (12 strains) were genotypically distinct, with about 55% similarity to the main group of 102 strains. The strains in the minor groups were all isolated from the Minnesota locations and were pathogenic on two disease-resistant pea breeding lines (MN313 and MN314). Estimates of genetic diversity based on RAPD analysis ranged from 0.24 to 0.33 within populations to 0.35 among all strains from all populations. A. euteiches populations were genotypically and phenotypically variable, but no distinct genotypic differences were identified among populations from the four isolated locations.  相似文献

Abstract

In a survey of plant parasitic nematodes associated with or affecting rice throughout Nigeria, some important nematode pests, especially the white tip disease nematode, Aphelenchoides besseyi and the rice root nematodes Hirschmanniella spinicaudata and H. oryzae were identified from seed, soil and root samples from swamp rice fields respectively. The sugarcane cyst nematode, Heterodera sacchari occurred in swamp rice fields only around the major sugarcane estates in Nigeria. The root‐knot Meloidogyne incognita and the root lesion nematode, Pratylenchus brachyurus were also encountered in upland (rainfed) rice fields. The white tip disease nematode, A. besseyi occurred at low levels in soils and rice seeds throughout the country. High population levels of H. spinicaudata and H. oryzae were encountered especially in areas where monoculture of rice is practised. General chlorosis, poor tillering and significantly reduced yield have been observed due to H. spinicaudata. Rice plants attacked by H. sacchari also showed intense chlorosis, delayed and reduced tillering and reduced grain yield. The roots of attacked plants were twiggy, very necrotic and blackened. The root‐knot M. incognita and the root lesion nematodes P. brachyurus have both been observed to reduce rice yields. Rice cultivars screened for reactions to the nematodes showed varying degrees of susceptibilities. Some varieties were however resistant to the root‐knot nematode, M. incognita.  相似文献

ABSTRACT The role of asexual reproduction in the production of pathogenic and genotypic variation in Aphanomyces euteiches was investigated. Variation was studied among three groups of 18 single-zoospore progeny of A. euteiches derived from each of three single-zoospore parental strains. Pathogenicity was assessed by evaluating disease severity (DS) on roots of five pea lines possessing different levels of resistance to Aphanomyces root rot and of a susceptible cultivar of snap bean and alfalfa. None of the single-zoospore progeny incited significantly higher DS levels than their parental strain on any of the seven hosts; however, 3 or 4 of the 18 progeny in each group incited significantly lower DS than their parental strains. The host range of the progeny either decreased or remained the same as compared with parental strains. Genotypic variation was assessed with randomly amplified polymorphic DNA (RAPD) analysis. Polymorphic RAPD markers that distinguished parental and progeny strains were detected within two of the three groups of strains with two of four RAPD primers used. Of 76 total RAPD markers that were detected among all strains in all groups, four (5%) were polymorphic. The polymorphic markers were not associated with the pathogenic variation.  相似文献

Ficld bean (Vicia fab a L. cv. Maris Bead) was susceptible both to simazine and dimefuron applied to the root system in culture solution at concentrations of 0–5 and above. Pea (Pisum satirum L. cv. Kelvedon Wonder). however, exhibited significantly greater tolerance them field bean to dimefuron between concentrations of 0.5 and 1.0. The Hill activity of isolated chloroplasts from pea was more sensitive to dimefuron than that from field bean qualitative (autoradiography) and quantitative experiments dcmonstrated that root-applied 14C dimefuron was more readily transported to the shoot system in bean than in pea. A double-pot experiment revealed that the effective site of uptake by the field bean is the root system: foliar uptake of 14C-dimefuron was shown, but little movement of label from the treated leaf oceurred. Adsorption of dimefuron on a calcareous loam soil was relatively low. It is concluded that fieltolerance exhibited by field bean to pre-emergence applications of dimefuron is due principally to depth protection: in pea. retention in the root provides an additional basis iS for tolerance.  相似文献

Pathogenicity of Aphanomyces spp. from Different Leguminous Crops in Sweden   总被引:1,自引:0,他引:1  
Host range and pathogenicity of a range of Aphanomyces spp. isolates obtained from pea roots but also from a range of other field-grown leguminous crops in southern Sweden was investigated. The Aphanomyces euteiches isolates originating from pea and the few obtained isolates originating from alfalfa, green bean and yellow sweet-clover were highly pathogenic only to pea. The A. euteiches isolated from common vetch differed from these isolates by being weakly pathogenic to pea and other legumes, but highly pathogenic to common vetch. Vetch isolates also formed a well-defined separate cluster based on principal component analysis of pathogenicity pattern on tested crops. Oospores of A. euteiches were observed in root tissue of pea as well as common vetch, alfalfa, green bean, broad bean, red clover and yellow sweet-clover in the greenhouse pathogenicity tests. An Aphanomyces sp. that morphologically differed from A. euteiches, was frequently isolated from several leguminous plants, but was non-pathogenic to all tested crops in the pathogenicity tests. In isozyme analysis the banding pattern of these isolates resembled the pattern of A. cladogamus. Another, different and so far unidentified Aphanomyces sp. from roots of green bean and broad bean, was also non-pathogenic to the tested legume species. Based on the isolates tested, the results obtained suggest that the population of Aphanomyces spp. infecting legume roots in Sweden consists of a pea-specific and a vetch-specific group of A. euteiches. Two other groups comprised (i) Aphanomyces sp. isolates that resembled A. cladogamus, and (ii) isolates, which resembled neither A. euteiches nor A. cladogamus. In addition, the host range of Swedish A. euteiches isolates was not as broad as reported for A. euteiches isolates from other countries.  相似文献

近年来,随着我国荞麦面积的迅速增加,根结线虫病呈逐年加重趋势,严重影响了荞麦的产量与品质。本研究于2014年对西南10个地区17个乡镇秋播苦荞麦根结线虫的分布、发生种类以及危害程度进行了调查。结果表明,西南地区危害荞麦的根结线虫种类有南方根结线虫(Meloidogyne incognita)、爪哇根结线虫(M.javanica)和花生根结线虫(M.arenaria)3种,其中南方根结线虫为优势种群。田间根结线虫种群大多数为单一种群,23.5%的样品为南方根结线虫与爪哇根结线虫(或花生根结线虫)组成的混合种群。调查发现前作为烟草或马铃薯的地块,荞麦根结线虫发生危害严重,平均被害株率为6.0%~77.5%,病情指数为1.4~26.1,而前作为玉米的荞麦根结线虫则较轻,被害株率最高为16.0%,相应病情指数为2.3。  相似文献

Pathozone dynamics were derived for approx. 50 Meloidogyne incognita juveniles infecting a single root of tomato under controlled conditions from a point source of inoculum and described using simple, non-linear models. The pathozone decayed sigmoidally with distance, but increased over time as progressively more nematodes were able to infect the root. Despite the reported ability of M. incognita juveniles to travel up to 50 cm in some conditions, the maximum width of the pathozone for a single tomato root was estimated as 18·1 mm. It is conjectured that this was because of (i) diffusion from a point source of inoculum, (ii) a small infection court (a single root tip) and (iii) the limited life span of the nematode. A second experiment was used to assess the effect of the nematophagous fungus Pochonia chlamydosporia on the pathozone dynamics of the nematode. This fungus is known to produce nematicidal products in vitro , which may affect invasion of roots by the free-living nematode. To examine the possibility of a change in the position of the site of infection, changes in the probability of gall formation along the root length were also examined. In the absence of P. chlamydosporia , the pathozone dynamics of M. incognita were very similar to those of the first experiment. It was shown that P. chlamydosporia did not significantly affect the pathozone dynamics of M. incognita nor the site of gall formation, which appear to have little importance for the role of the fungus as a biological control agent.  相似文献

Expression of Tenod40, a tobacco (Nicotiana tabacum cv. Xanthi nc) homologue of Msenod40, a gene expressed early during alfalfa (Medicago sativa) nodule development, was significantly and consistently increased (about three-fold) in tobacco roots of plants grown in sand and colonized by the arbuscular mycorrhizal (AM) fungus Glomus intraradices, as compared to non-mycorrhizal controls. In alfalfa (Medicago sativa cv. Gilboa), a similar induction of Msenod40 expression was also detected in mycorrhizal vs. non-mycorrhizal roots. In the same experimental system, the application of the cytokinin 6-benzylaminopurine to non-mycorrhizal roots resulted in a similar increase in Msenod40 expression. A significantly higher incidence of AM fungal colonization (vesicles per cm of root) in M. sativa cv. Gilboa roots was observed following application of luteolin-induced Sinorhizobium meliloti exudates as compared to plants amended with non-induced exudates. Moreover, overexpression of the Mtenod40 gene in Medicago truncatula plants under the control of a 35S constitutive promoter resulted in significantly enhanced colonization as compared to non-transgenic plants. The fact that enod40 gene expression was increased both in a legume and in a non-legume during colonization by the AM fungus, and that colonization levels were higher in enod40 -overexpressing transgenic plants strongly suggests that this gene is involved in the establishment of the fungal partner in its host.  相似文献

Velvetbean (Mucuna spp.) is a summer annual that has been used as a cover crop to reduce erosion, fix nitrogen and suppress weeds and plant-parasitic nematodes. Crude aqueous extracts (1:15 dry weight plant/volume water) were made from velvetbean plant parts, and various concentrations of the extracts were evaluated in vitro for toxicities to different stages of Meloidogyne incognita (Kofoid and White) Chitwood and for suppression of hypocotyl and root growth and inhibition of germination of tomato (Lycopersicon esculentum L.) and lettuce (Lactuca sativa L.). Germination was only affected by the full-strength extract from leaf blades. Lettuce root growth was the most sensitive indicator of allelopathic activity of the plant part extracts. Lettuce and tomato root growth was more sensitive to the extract from main roots than to extracts of other plant parts, with lethal concentration (LC50) values of 1.2 and 1.1% respectively. Meloidogyne incognita egg hatch was less sensitive to extracts from velvetbean than the juvenile (J2) stage. There was no difference among LC50 values of the extracts from different plant parts against the egg stage. Based on LC50 values, the extract from fine roots was the least toxic to J2 (LC50 39.9%), and the extract from vines the most toxic (LC50 7.8%). The effects of the extracts were nematicidal because LC50 values did not change when the extracts were removed and replaced with water.  相似文献

ABSTRACT Digestive cysteine proteinases have been isolated from plant-parasitic nematodes as well as coleopteran and hemipteran insects. Phytocystatins, inhibitors of cysteine proteinases, are found in a number of plants where they may play a role in defense against pathogens and pests. The cDNAs of the phytocystatins from rice, oryzacystatin I (OC-I) and oryzacystatin II (OC-II), were expressed in alfalfa (Medicago sativa) plants under the control of the potato protease inhibitor II (PinII) promoter and the plants were evaluated for resistance to the root-lesion nematode (Pratylenchus penetrans). A PinII-beta-glucuronidase (GUS) gene was introduced into alfalfa to determine the pattern of gene expression from this promoter. Constitutive GUS expression was observed in leaf and root vascular tissue, and in some plants, expression was observed in leaf mesophyll cells. Mechanical wounding of leaves increased GUS expression approximately twofold over 24 h. Inoculation with root-lesion nematodes resulted in localized GUS expression. Populations of root-lesion nematodes in alfalfa roots from one line containing the PinII::OC-I transgene and one line containing the PinII::OC-II transgene were reduced 29 and 32%, respectively, compared with a transgenic control line. These results suggest that oryzacystatins have the potential to confer increased resistance to the root-lesion nematode in alfalfa.  相似文献

Barley and winter wheat were sampled over 3 years to characterize soil-borne organisms involved with cereal crown and root disease complexes. Winter wheat crowns were infected more often by Fusarium avenaceum and F. sambucinum than barley crowns. Bipolaris sorokiniana and F. graminearum were more common in crown tissue of barley underseeded with clover than in winter wheat crowns. In roots, the incidence of F. avenaceum was highest in winter wheat. In soil, populations of Rhizoctonia solani and F. sambucinum were higher in winter wheat than barley underseeded with ryegrass. Stunt nematodes (Tylenchorhynchus spp.) were greatest in winter wheat and barley underseeded with ryegrass. The incidence of F. avenaceum in roots of winter wheat correlated positively with the severity of crown and root rot symptoms. The severity of barley crown and root rot correlated positively with the incidence of R. solani in crowns, F. avenaceum in crowns and roots, B. sorokiniana in roots, and numbers of stunt nematodes in the soil. The incidence of soil-borne organisms was also recorded in annual ryegrass. soybean, potato, pea, and clover which were grown in rotation with barley and winter wheat. The incidence of R. solani and F. avenaceum was highest in clover crown tissue, and F. sambucinum was prevalent in soybean hypocotyls. Soil population levels of R. solani, F. avenaceum , and stunt nematodes were greatest in ryegrass and pea soil just prior to ploughing down or harvesting these crops, respectively.  相似文献
