Twenty-four barrows were used to investigate the effects of exogenous porcine somatotropin (pST) administration (0 and 100 micrograms.kg-1.d-1) between 30 and 60 kg on longissimus muscle morphology and meat tenderness of pigs grown to 90 kg. Administration of pST was by daily i.m. injection. Pigs were fed a fortified diet in restricted amounts between 30 and 60 kg and had ad libitum access to this diet from 60 to 90 kg. Excipient pigs had fewer alpha R fibers and more alpha W fibers than pST-treated pigs (P less than .05). Administration of pST increased (P less than .05) muscle fiber area for all three fiber types at both the 60 kg (34%) and 90 kg (29%) slaughter weights. Furthermore, pST administration increased shear-force (30% at 60 kg; 19% at 90 kg) of the longissimus muscle, indicating that pST administration reduced meat tenderness. All the pST-treated pigs exhibited the "giant fiber syndrome" and a high proportion (62%) of pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomaly resulted from increased muscle protein accretion rate or fiber degeneration is unknown. Results indicated that the stimulatory effects of pST on muscle growth are sustained following cessation of hormone treatment. However, the incidence of pale, soft, exudative muscle in 62% of the pST-treated pigs indicates that pST potentially has undesirable side effects.  相似文献

Thirty two barrows were used in a 2 x 3 factorial arrangement to evaluate the effects of porcine somatotropin (pST) administration (USDA-pGH-B1; 0 and 100 micrograms.kg BW-1.d-1) and feed intake (FI; ad libitum [A], 1.64 [R1:80% of A] and 1.38 [R2:60% of A] kg/d) on longissimus (LM) muscle fiber characteristics of pigs growing from 25 to 55 kg live weight. The administration of pST resulted in an increase (P less than .05) in muscle fiber size (all three fiber types) and shear-force (17.1%). The percentage of muscle fiber types was not affected by pST treatment. Feeding of the diet at restricted levels (R1 and R2) resulted in smaller (P less than .05) beta R and alpha W fibers compared with feeding ad libitum. Pigs with ad libitum access to their diet had fewer alpha R fibers and more alpha W fibers in the LM compared with those fed at R1 and R2 levels. Restricted feed intake (R1 and R2) resulted in lower (P less than .05) shear-force values compared with ad libitum feeding. The present study indicates that pST administration of young barrows results in an increase in muscle hypertrophy. Restricted FI reduces both transformation from alpha R to alpha W fibers and muscle fiber hypertrophy.  相似文献

Thirty-seven pigs with an initial live weight of 60 kg were used to investigate the effects of daily exogenous porcine somatotropin (pST) administration at two dose levels (0 and 100 micrograms.kg-1.d-1) for a 31-d period on muscle fiber characteristics and meat tenderness of boars, gilts and barrows. Excipient boars and gilts had more alpha W and fewer alpha R fibers than did those receiving pST. The percentage of muscle fiber type for barrows was not affected by pST treatment. The administration of pST resulted in an increase in muscle fiber size for all three fiber types in all three sexes, but these changes were of greater magnitude in barrows (31.8%) and gilts (27.8%) than in boars (9.3%). Somatotropin negated the intrinsic sex effect differences in fiber area of the pigs. There was no difference in tenderness among excipient boars, barrows and gilts; however, with the inclusion of pST, shear force decreased in boars and gilts and increased in barrows. A high proportion of the pST-treated pigs contained giant fibers in the longissimus muscle. Furthermore, a small proportion of the pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomalies occurred through increased muscle activity or from fibers undergoing degenerative changes was not determined.  相似文献

The dose-dependent effects of porcine somatotropin (pST) on cellular aspects of skeletal muscle growth, muscle composition and measures of pork quality were investigated in growing barrows and gilts. Eighty crossbred pigs weighing 46 kg were assigned randomly to receive daily subcutaneous injections of 0, 30, 60, 120 or 200 micrograms pST/kg BW until they weighed 100 kg. Semitendinosus muscle weights were increased with pST dose (linear, P less than .001) by 21%. Percentage of type I and type II muscle fibers was not changed with pST, but cross-sectional area of type I and type II fibers was increased in parallel with muscle weight. Percentage of moisture increased (P less than .01) and percentage of lipid decreased (P less than .01) as pST dose increased. The pH of the longissimus 24 h postmortem increased (P less than .01) .1 to .2 units with increasing pST dose, but subjective evaluation for color, firmness and wateriness of the longissimus indicated no discernible treatment effect. Gardner color difference meter "Rd" and "A" values decreased (P less than .01) with a pST dose of 60 micrograms/kg or more, signifying a slightly darker and less red color, respectively, of the longissimus muscle. Weight loss of loin chops 2.54 cm thick cooked to 71 degrees C (20.3% to 23.7%) and shear force of cores 1.27 cm in diameter (2.89 to 3.76 kg) were not related to pST treatment or dose.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Three litters of four pigs from each of four different groups were used to evaluate the effect of porcine somatotropin (pST) on growth performance, body gain composition, energy and N metabolism, and in vitro cytochrome oxydase (final enzyme of the respiratory chain) activity of tissues. The four groups included boars from a synthetic line (SG1) or the Large White breed (SG2) and barrows from the Large White breed (SG3) or crossbred between Large White and Meishan breeds (SG4). Inherent capacity for daily lean tissue growth (LTG) decreased from SG1 to SG4. Within a litter, one pig was slaughtered and dissected at the beginning of the experiment (55 kg BW) and the three others were fed the same daily supply of protein and amino acids (26 g of lysine/d) but relative daily energy levels were either 113 (without pST: E1/0), 100 (3 mg of pST/d: E2/pST) or 87 (3 mg of pST/d: E3/pST). The 100 energy level corresponded to the ad libitum intake of E2/pST pigs. Two energy and N balances were carried out in respiration chambers during the experimental period. Pigs were slaughtered and dissected at approximately 95 kg BW and composition of gain was estimated using the comparative slaughter technique. In E1/0 pigs, daily BW, lean, and N gain were affected (P less than .01) by group; 566, 471, 374, and 315 g/d of lean tissue gain in SG1, SG2, SG3, and SG4 pigs, respectively. At high ME intake (E2/pST vs E1/0), pST increased daily BW (+14%), lean (+27%), or N (+26%) gain and reduced adipose tissue (-50%) gain, but the pST effect was inversely related to LTG: for N, the improvement was 2.8, 7.1, 7.0, and 11.1 g/d in SG1, SG2, SG3, and SG4 groups, respectively. Energy restriction (E3/pST vs E2/pST) reduced (P less than .001) adipose tissue gain in all groups but did not affect lean tissue or N gains in SG1, SG2, and SG3 pigs. In the pST-treated pigs of the SG4 group, the lean tissue or N gains were reduced (P less than .01) by energy restriction. Energy restriction combined with pST treatment (E3/pST) led to negligible amounts of fat deposited (40 g/d for SG1 + SG2 + SG3 pigs) and a gain:feed ratio higher than 500 g/kg (580 in SG1 pigs). The dietary dose responsiveness of conjugated linoleic acid (CLA) addition relative to the fatty acid profile of edible lean tissue was examined in grower pigs treated with or without porcine somatotropin (pST). Gilts and barrows were fed CLA at 0, 0.25, 0.5, 1.0, or 2.0% of diet by weight from 20 to 55 kg BW. Additional pigs were administered (pST) at 0 or 100 microg x kg BW x d(-1) and fed either 0.5 or 2.0% CLA. Animals were fed diets containing 18% CP, 1.2% lysine, and 3.5 Mcal of DE/kg at 110% of ad libitum intake. The fatty acid profile in latissimus dorsi and dorsal s.c. adipose tissue samples was determined by gas chromatography. Dietary CLA replacement of corn oil increased the percentage of total fatty acids as stearic acid, whereas the percentages as oleic and linolenic acids were reduced in lattisimus muscle. Treatment with CLA + pST increased the percentages of linoleic and arachidonic acids while reducing the percentages of palmitic and oleic acids in lattisimus muscle. Dietary CLA increased the percentages of palmitic and stearic acids in s.c. adipose tissue while reducing the percentages of oleic, linoleic, linolenic, and arachidonic acids. The percentage of palmitic acid was reduced in s.c. adipose tissue, whereas linoleic acid was increased with CLA + pST. No synergistic effect was detected between CLA and pST for reducing carcass lipid content in grower pigs. However, pST increased the percentage of polyunsaturated fatty acids in lattisimus muscle and s.c. adipose tissue while reducing the percentages of saturated fatty acids in swine fed CLA.  相似文献

A possible management strategy to alter fetal development and enhance sow productivity and progeny performance was examined by maternal administration of porcine somatotropin during early gestation. Eighteen crossbred gilts were bred naturally to boars of similar genetics, and pregnancy was confirmed between Days 21 and 24 of gestation by ultrasound. All animals were allowed ad libitum consumption of a 16% CP gestation diet through Day 21 of gestation and 3.0 kg/d for the remainder of gestation. Gilts were injected twice daily with 0 (n = 10) or 15 μg/kg body weight (BW) (n = 10; total, 30 μg/kg BW per d) pituitary-derived porcine somatotropin (pST) during Days 28 to 40 of gestation. Data were collected postmortem during embryonic, neonatal, and market-weight phases. At 41 d of gestation, pST treatment increased embryonic survival (87.9 versus 77.0%; P < 0.05) and embryo crown rump lengths (77.96 versus 65.14 mm; P < 0.01), but embryo weight was not altered (10.15 and 9.03 g; P > 0.10). Pigs from pST-treated gilts had increased (P < 0.01) crown rump lengths at birth (31.5 versus 30.4 cm) and 21 d (50.9 versus 48.4 cm). However, no differences were observed in birth or 21-d weights as a result of pST treatment (P > 0.10). Neonatal carcasses of progeny (20 kg BW) from the pST-treated gilts had heavier semitendinosus muscles (76.1 versus 66.0 g; P < 0.10), larger longissimus muscle cross-sectional area (10.1 versus 8.2 cm2; P < 0.05), longer sides (51.2 versus 47.9 cm; P < 0.001), and decreased 10th rib backfat (6.67 versus 8.64 mm; P < 0.001) compared with those of controls. Carcasses of market-weight progeny (100 kg BW) from pST-treated gilts had larger longissimus muscle cross-sectional area (P < 0.10), heavier trimmed loins (P < 0.10), and longer carcass sides (P < 0.05). Data are supportive of a hypothesis that mechanisms during early embryonic development are sensitive to manipulation through selected management strategies of the sow and that modifications of this strategy may serve as a model for the examination of molecular and cellular events controlling early embryonic growth.  相似文献

Birth weight positively predicts postnatal growth and performance in pigs and can be increased by sustained maternal porcine ST (pST) treatment from d 25 to 100 of pregnancy (term ~115 d). The objective of this study was to test whether a shorter period of maternal pST treatment in late pregnancy (d 75 to 100) could also increase birth and weaning weights of progeny under commercial conditions. Gilts (parity 0) and sows (parities 2 and 3) were not injected (controls) or injected daily with pST (gilts: 2.5 mg?d(-1), sows: 4.0 mg?d(-1), both ~13 to 14 μg?kg(-1)?d(-1)) from d 75 to 100 of pregnancy. Litter size and BW were recorded at birth and weaning, and dams were followed through the subsequent mating and pregnancy. Maternal pST injections from d 75 to 100 increased litter average progeny weight at birth (+96 g, P = 0.034) and weaning (+430 g, P = 0.038) in sows, but had no effect on progeny weight in gilts (each P > 0.5). Maternal pST treatment did not affect numbers of live-born piglets and increased numbers of stillborn piglets in sows only (+0.4 pigs/litter, P = 0.034). Maternal pST treatment did not affect subsequent reproduction of dams. Together with our previous data, these results suggest that sustained increases in maternal pST are required to increase fetal and postnatal growth in gilt progeny, but that increasing maternal pST in late pregnancy may only be an effective strategy to increase fetal and possibly postnatal growth in sow progeny.  相似文献

Two experiments were conducted to assess the effects of porcine ST (pST) on the responses to a near-ideal blend of AA for pigs from 22 to 60 kg BW. Eighty Hampshire × Yorkshire gilts (40 gilts/experiment) were individually penned and assigned to a 4 × 2 factorial arrangement of treatments, consisting of 4 diets with and without pST injection. A fortified corn-soybean meal basal diet was formulated to contain 1.50% total Lys and Thr, Met, and Trp were added to obtain a near-ideal blend of these AA relative to Lys. In 3 additional diets, Lys was reduced to 1.25%, 1.00%, or 0.75% by diluting the basal diet with cornstarch, cellulose, and sand, such that the diets also contained the same ratios of AA. Pigs that received pST were administered a daily intramuscular injection of 2 mg of pST. Data from the 2 experiments were pooled. Administration of pST increased ADG (P < 0.01), G:F (P < 0.01), and LM area (P < 0.01), and decreased ADFI (P < 0.03), last rib backfat (P < 0.01), and 10th rib backfat (P < 0.01). Also, estimated carcass muscle and calculated lean gain increased (P < 0.01) in pST-treated pigs. Administration of pST also increased (P < 0.01) the percentage, total gain and accretion rate of water, protein, and ash in the carcass, and decreased (P < 0.01) the percentage, total gain, and accretion rate of carcass fat. Growth rate, G:F, and carcass traits improved (P < 0.01), percentage of carcass proteinand water increased (P < 0.01), and carcass fat percentage decreased (P < 0.01) with increasing dietary Lys. The percentage, total gain, and accretion rate of carcass protein increased to a greater extent in pST-treated pigs than in untreated pigs, resulting in a pST × Lys interaction (P < 0.05). The results indicated that pST improves performance, leanness, and protein accretion in pigs from 22 to 60 kg BW, and that these responses to dietary Lys and a near-ideal blend of AA is greater in growing pigs treated with pST than untreated pigs.  相似文献

The objective of this study was to evaluate the effects of coadministration of ractopamine-HCl (RAC) and trenbolone acetate plus estradiol (TBA) on LM fiber cross-sectional area (CSA), diameter, fiber-associated myonuclei, and satellite cell number. Culled crossbred beef cows (n = 98; 11 +/- 1.8 yr old; BCS 4.3 +/- 0.03) from a single ranch in south Florida were fed a concentrate diet for 92 d in a 2 x 2, randomized block design. Cows were blocked by BW on arrival into light (initial BW = 369.75 +/- 2.68 kg and end BW = 501.96 +/- 6.90 kg) and heavy (initial BW = 418.31 +/- 2.75 kg and end BW = 522.15 +/- 7.09 kg) groups before assignment to treatment. Factors included dietary treatment (0 or 15 ppm) and implant status (0 or 80 mg of trenbolone acetate + 16 mg of estradiol). Ractopamine was provided in the diet to 2 pens or half the treatments during the final 35 d of feeding. Cows were slaughtered on d 92. Forty-eight hours postmortem, the 6th-rib portions of the LM were obtained from 10 randomly selected carcasses from each treatment group (n = 40). Cryosections (12 mum) were immunostained for dystrophin and myosin heavy chain I or II for the measurement of fiber CSA and type, respectively. Fiber-associated nuclei and satellite cell numbers were measured in serial cryosections. There was a RAC x TBA interaction (P < 0.05). Type I fiber CSA and diameter were increased (P < 0.05) by TBA and RAC. Type I CSA and diameter were larger (P < 0.05) in TBA + RAC than RAC only. Type II fiber CSA and diameter were not affected by TBA (P = 0.48), RAC (P = 0.15), or TBA + RAC (P = 0.60). Satellite cell numbers and fiber-associated nuclei were not affected (P > 0.05) by implant status or ractopamine supplementation. These results indicate that TBA and RAC preferentially increase the size of type I fibers in cull cows.  相似文献

The dose-dependent effects of porcine somatotropin (pST) on growth performance and composition of carcass gain were investigated in 150 growing pigs. The experiment involved two genotypes (barrows from the Pig Improvement Company [PIC] and a University of Nebraska [NEB] gene pool line) and two sexes (PIC barrows and boars). At 30 kg, pigs were randomly assigned within each genotype and sex subclass to receive daily i.m. injections of 50, 100, 150, or 200 micrograms of pST/kg BW or an equivalent volume of an excipient. A diet (3.5 Mcal of DE/kg) supplemented with crystalline amino acids and containing 22.5% CP was available on an ad libitum basis until pigs were slaughtered at approximately 90 kg live weight. Excipient-treated PIC barrows exhibited faster and more efficient growth (P less than .001) and a higher capacity for carcass protein accretion (P less than .001) but similar rates of lipid deposition compared to excipient-treated NEB barrows. Within the PIC genotype, control boars grew at a rate similar to that of barrows, but they were more efficient (P less than .05) and deposited more carcass protein (P less than .05) and less lipid (P less than .001). Carcass protein accretion rate increased (P less than .001) up to approximately 150 micrograms of pST.kg BW-1.d-1, whereas lipid deposition decreased (P less than .001) with each incremental dose of pST. Although differences between PIC boars and barrows for all criteria were negated with increasing pST dose, they were maintained between the two genotypes. Polynomial regressions suggested that a slightly higher pST dose was required to optimize the feed:gain ratio compared with rate of gain and that the dose (micrograms per kilogram BW per day) was a function of the genotype and sex (feed:gain: 185, 170, and 155; rate of gain: 155, 155, and 125 for NEB barrows, PIC barrows, and PIC boars, respectively).  相似文献

A restriction/realimentation feeding strategy was applied to pigs to increase the age at market weight and final ADG, modify protein and lipid deposition rates at carcass and muscle levels, and thereby improve eating quality of the pork. A total of 126 Duroc x (Large White x Landrace) pigs (females and castrated males) were used. At the average BW of 30 kg, within litter and sex, pairs of littermates (blocked by BW) were randomly assigned to ad libitum (AL) feeding during growing (30 to 70 kg of BW) and finishing (70 to 110 kg of BW) periods (AL, n = 56), or restricted feeding at 65% of the ADFI of the AL pigs, on a BW basis, during the growing period and AL feeding during finishing (compensatory growth, CG; n = 56). In each feeding regimen, 15 pigs were slaughtered at 70 kg of BW, and 41 pigs were slaughtered at 110 kg of BW. Additionally, 14 pigs were slaughtered at 30 kg of BW to calculate tissue deposition rates. The CG pigs showed decreased ADG (-35%, P = 0.001) during growing but increased ADG (+13%, P = 0.001) during finishing (i.e., compensatory growth) due to greater (P = 0.001) ADFI and G:F. Hence, CG pigs were 19 d older at 110 kg of BW than AL pigs. The CG pigs were leaner at 70 kg of BW than AL (e.g., 11.7 vs. 13.5 mm of average backfat thickness for CG and AL pigs, respectively, P = 0.023), whereas the differences were reduced at 110 kg of BW (20.6 vs. 21.0 mm of average backfat thickness for CG and AL pigs, respectively, P = 0.536). At 70 kg of BW, intramuscular fat (IMF) content of LM did not differ between CG and AL pigs (1.25 vs. 1.49%, respectively, P = 0.118), whereas CG pigs had less IMF in LM at 110 kg of BW (2.19 vs. 2.53% for CG and AL pigs, respectively, P = 0.034). Feeding regimen influenced the composition of weight gain. From 30 to 70 kg of BW, feed restriction reduced (P = 0.001) lean and adipose tissue deposition at the carcass level and protein and lipid deposition at the muscle level. From 70 to 110 kg of BW, the CG feeding strategy increased (P = 0.016) deposition of adipose but not of lean tissue at the carcass level. However, lipid and protein deposition at the muscle level were not affected. Thus, realimentation promoted deposition of subcutaneous fat over IMF. Feeding regimen hardly affected technological meat quality at 110 kg of BW. The CG feeding strategy decreased (P = 0.014) the meat juiciness score in relation to the decreased IMF but did not influence other sensory traits. Elevated IMF content and improved pork quality might be achieved by modifying the onset or duration of the restriction and realimentation periods.  相似文献

The Western diet is characterized by a high intake of SFA relative to PUFA, and the consumption of n-3 PUFA is decreased relative to n-6 PUFA. Therefore, there has been much interest in recent years in ways to manipulate the fatty acid composition of meat. The objective of this work was to determine the effects of dietary extruded linseed, an n-3 PUFA source, on growth performance, carcass composition, meat quality, and oxidative stability of pigs slaughtered at 111.0 (+/-4.8) kg of BW and 160.0 (+/-9.2) kg of BW. The association of these factors with BW at slaughter was also examined. Forty barrows, 78.1 (+/-1.75) kg of initial BW, were fed a control diet (2.5% sunflower oil) or a linseed diet containing 5% of whole extruded linseed. Both diets contained 170 mg of vitamin E and 250 mug of selenium. Eight pigs from each dietary treatment were slaughtered at 110 kg of BW and the others at 160 kg of BW. There was no dietary effect (P > 0.05) on growth, carcass characteristics, meat quality, or the activity of malic enzyme in LM and backfat. Inclusion of linseed increased (P < 0.05) n-3 PUFA content in both LM and backfat and decreased the n-6:n-3 PUFA ratio from 12 to 4.5 in LM, and from 11 to 3 in backfat. Liveweight at slaughter significantly influenced carcass characteristics, meat quality, total lipid and oxidative stability of LM, malic enzyme activity in adipose tissue, and fatty acid content of LM and backfat. This study shows that the inclusion of linseed in swine diets may improve the fatty acid profile of pork without deleteriously affecting oxidation or color stability. Thus, such a feeding practice may improve human health based on the n-6:n-3 PUFA ratio recommended for the human diet.  相似文献

为了研究低聚木糖(XOS)对生长肥育猪生长性能、胴体性状和肉品质的影响,试验选取平均体重为30 kg左右的三元杂交猪80头,随机分为8组,每组10头,公、母各占1/2,单栏饲养。试验设对照组、抗生素组,3065 kg阶段100、250和500 g/t XOS组,以及30100 kg阶段100、250和500 g/t XOS组。分别于试验开始和结束时记录每头猪的空腹体重及采食量,计算平均日采食量(ADFI)、平均日增重(ADG)和料重比(F/G);于试猪平均体重达100 kg时屠宰采样,测定胴体性状、肉品质和肌肉化学成分。结果表明:与对照组或抗生素组相比,饲粮添加不同剂量的XOS对生长肥育猪的ADFI、ADG、F/G、胴体性状和肉品质均无显著影响(P0.05);3065 kg阶段添加250 g/t XOS可显著增加脾脏指数以及背最长肌粗蛋白质含量(P0.05);30100 kg阶段添加500 g/t XOS可显著增加脾脏指数及背最长肌粗蛋白质含量(P0.05)。综上所述,饲粮添加不同剂量的XOS虽对生长肥育猪的ADFI、ADG、F/G、胴体性状和肉品质等指标影响不显著,但可通过增加肌肉粗蛋白质含量而改善猪肉营养价值;以30100 kg阶段添加500 g/t XOS效果较佳。  相似文献

An experiment involving 32 individually housed, castrated male pigs was conducted to determine the effects of environmental temperature (12 degrees C = T12 or 28 degrees C = T28) on the performance, carcass characteristics, muscle and adipose tissue metabolism, and meat quality of pigs fed to achieve equal rates of weight gain between 8 and 92 kg live weight. The T12 pigs had less muscle (P less than .01) and more subcutaneous fat (P less than .01) in the ham than did T28 animals. Thyroids and adrenals were heavier (P less than .01) in T12 than in T28 pigs. Percentage of type I muscle fibers was higher (P less than .01) and fiber cross-sectional areas smaller (P less than .01) in semispinalis muscle (SS) of T12 pigs, whereas no effect of temperature was noticed in longissimus muscle (LD). Cold exposure induced a larger increase in oxidative metabolism in SS (+30 to 32%, P less than .001) than in LD (+14 to 17%, P less than .10) and an enhanced glycolytic metabolism in LD (P less than .05). Lipid concentration was higher in SS of T12 than in that of T28 pigs. Lipogenic enzyme activities and unsaturation of backfat were higher in T12 than in T28 pigs, whereas no effect of temperature was noticed in leaf fat. Cold treatment resulted in a faster postmortem pH decline (P less than .01), higher glycolytic potential (P less than .01), and lower ultimate pH (P less than .01) in LD. Paylean alters myosin heavy chain isoform content in pig muscle   总被引:1,自引:0,他引:1  
Feeding beta-adrenergic agonists promotes muscle growth. Early histological techniques failed to show precisely how feeding ractopamine-HCl (Paylean) alters muscle growth in pigs. To understand these effects, an indirect enzyme-linked immunosorbent assay (ELISA) was used to determine the abundance of each adult skeletal muscle myosin heavy chain isoform, one means of assigning muscle fiber type, in fast and slow muscles of pigs fed Paylean. Sixty growing pigs (-85 kg) were randomly assigned to three Paylean doses (0, 20, or 60 ppm). At 3, 7, 14, 28, and 42 d of treatment, four pigs per dose were harvested and white (WST) and red (RST) semitendinosus and longissimus (LM) muscles were removed and processed, and myosin heavy chain was quantified by ELISA. Feeding Paylean enhanced (P < 0.05) pigs' average daily gain. Muscle myosin heavy chain (slow, 2A, 2AX, and 2B) composition differed (P < 0.05) across muscles. Compared with LM, RST contained approximately five times more (P < 0.0001) slow and type 2A myosin heavy chain and three times more 2AX myosin heavy chain but nearly undetectable amounts of 2B myosin heavy chain. Myosin heavy chain composition of the WST closely resembled that of the LM (i.e., greater 2AX and 2B and less slow and 2A). After 42d of 60 ppm Paylean, the amount of slow, 2A, and 2AX myosin heavy chain decreased (P < 0.05) across the three muscles whereas the amount of 2B myosin heavy chain increased (P < 0.05). In contrast, relative amounts of 2A and 2AX myosin heavy chain increased (P < 0.05) in muscle of control pigs at 42d. Changes associated with the 20-ppm dose were intermediate to and different from (P < 0.05) control and 60 ppm treatments. Correlations (P < 0.05) among various myosin heavy chain within muscles suggest that slow, type 2A, and 2X decrease with increases in 2B myosin heavy chain. These data show that administration of Paylean affects myosin heavy chain isoform composition in a time- and dose-dependent manner and provides a mechanism of action for Paylean altering animal growth.  相似文献

选用荣昌(RC)猪和杜×长×大(DLY)杂交猪为试验对象,研究猪背最长肌肌纤维直径和密度的发育性变化及品种与营养影响特点。结果表明:2个品种猪的背最长肌肌纤维直径和密度的发育规律较为类似,肌纤维直径随体重增加呈线性增长,而肌纤维密度则随体重增加呈乘幂方程式降低。相关分析表明,肌纤维密度与直径均存在极显著负相关(P≤0.003)。背最长肌肌纤维直径和密度在20~50kg阶段品种间差异不显著(P≥0.099),但在80kg,RC猪的肌纤维直径显著低于DLY猪(P=0.0043),而RC猪的肌纤维密度则显著高于DLY猪(P=0.0049)。饲粮营养水平对2个品种猪背最长肌的肌纤维直径和密度均无显著影响。以上结果提示,2个品种猪的背最长肌肌纤维的生长规律较为类似,但不同品种的肌纤维直径存在差异,主要表现在80kg,RC猪显著低于DLY猪,可能与其优良肉质有关。  相似文献

Changes in muscle growth, calpastatin activity, and tenderness of three muscles were assessed in 20 callipyge and 20 normal wether lambs slaughtered at live weights (LW) of 7, 20, 36, 52, and 69 kg. At 24 h postmortem, the longissimus (LM), semimembranosus (SM), and supraspinatus (SS) muscles were removed and weighed and samples were obtained for calpastatin activity (CA; 24 h) and Warner-Bratzler shear force (WBS; aged 6 d). For muscle weights and calpastatin activity, the weight group x muscle x phenotype interaction was significant (P < 0.05). Muscle weights were similar (P > 0.05) between phenotypes for all three muscles at 7 kg LW. At 20 kg LW, the LM and SM muscles from the callipyge lambs were heavier (P < 0.05) than those from normal lambs; however, the SS did not differ (P > 0.05) between phenotypes at 7, 20, or 52 kg. From 20 to 69 kg LW, the LM and SM weights were 42 and 49% heavier (P < 0.05) for callipyge than for normal lambs. Calpastatin activity of the callipyge LM was greater (P < 0.05) than that of normal LM at 36, 52, and 69 kg. In the callipyge LM, CA was similar (P > 0.05) at 20, 36, and 52 kg LW and did not differ (P > 0.05) from 7-kg or 69-kg values. Calpastatin activity declined (P < 0.05) across the growth curve for the SM and SS, but values were higher (P < 0.05) in the SM in callipyge than in normal lambs. Shear force values of the LM were lower (P < 0.05) for normal lambs at 36, 52, and 69 kg LW than for callipyge lambs. In the SM and SS, WBS values decreased (P < 0.05) across the growth curve, but values were higher (P < 0.05) for callipyge lambs in the SM only. These data indicate that the selective muscular hypertrophy of the callipyge phenotype develops during the postnatal growth period between 7 and 20 kg LW (19 and 100 d of age). Longissimus and semimembranosus muscles in the callipyge lambs were over 40% heavier from 20 to 69 kg LW; however, they also had higher levels of calpastatin activity and Warner-Bratzler shear force during this time period, indicating the need for postmortem tenderization treatments to improve palatability.  相似文献

The effect of porcine somatotropin (pST) on the lipid profiles of adipose tissue and muscle was investigated. Sixteen crossbred barrows were injected daily with either 3 mg of pST or a placebo. After slaughter, total lipid and fatty acid composition of raw subcutaneous (SC) adipose and intermuscular (IM) adipose tissue and longissimus muscle were determined. The SC adipose tissue from pST-treated pigs had a 7.5% decrease in total lipid content; specific fatty acids 16:0, 18:0, and 18:1(n-9)c decreased most. The IM fat from pST-treated pigs had lower levels of 16:0 and 20:0. There was no effect of pST treatment on the lipid profile of the longissimus muscle. The data suggest that pST treatment produces small but significant changes in the saturated fatty acid content of adipose tissue in pigs.  相似文献
