研究赤霉素(GA3),冷湿和温度 对五个种源的印度冷杉(Abies pindrow)和长叶云杉(Picea smithiana)种子萌发的影响.种子被浸泡在GA3 (10 mg(L-1)中24小时,然后在3(5(C温度的条件下冷藏15天.设计4个温度(10(C, 15(C, 20(C 和 25(C)条件来促进种子的萌发.结果表明,浸泡和冷湿处理明显增加种子的萌发率.在10 (C时种子的萌发率最高.总体结果表明,种子被浸泡在GA3 (10 mg(L-1)中24小时,冷湿藏15天,可以有效地促进印度冷杉和长叶云杉的种子萌发.统计数据表明,浸泡处理、温度和种源以及与温度的相互作用都对种子的萌发有明显的影响.  相似文献

对喜马拉雅长叶松种子进行24小时GA3和H2O2浸泡预处理和15d的2-3℃低温预处理后,研究了在20℃,25℃和30℃萌发条件下,21个不同种源的喜马拉雅长叶松种子的萌发情况。结果表明,H2O2(1%v/v)和GA3(10mg/L)浸泡预处理,种子萌发率分别是82.39%和78.19%,而未经预处理的种子平均萌发率为70.79%。GA3和H2O2浸泡预处理分别使种子萌发时间缩短了8d和10d。在超过21天的20℃萌发条件下,湿冷处理提高了种子萌发率和缩短了萌发时间 而在25℃和30℃萌发条件下,总的萌发率未受到影响。喜马拉雅长叶松种子因缺少休眠而表现出很好的萌发,但是因为越来越多的造林项目需要大量的种子,播种预处理有利于提高种子萌发率和萌发速率,有助于满足种子需求。  相似文献

Following dispersal from the parent tree, seeds of yellow-cedar (Chamaecyparis nootkatensis[D. Don] Spach) exhibit low germination, primarily as a result of coat-imposed dormancy. Dormancy of the mature (intact) seed is effectively terminated by traditional warm/cold treatments. A chemical treatment using the anaesthetic 1-propanol combined with a three day warm water soak (30 °C), a two day GA3 treatment and 60 d of moist chilling not only promotes high germinability of yellow-cedar seeds, but also elicits vigorous post-germinative growth following seedling emergence under nursery greenhouse conditions. Here we compare the effectiveness of the more traditional warm/cold treatments with the chemical treatment in terms of their capacity to elicit vigorous growth and establishment in natural stands following transplant of seedlings from a nursery greenhouse environment. Two seed lots (42313 and 43697) and open-pollinated seed from parent trees 13-6 and 19-8 showed equivalent seedling growth in natural stands following the chemical treatment and two traditional warm/cold treatments typically used for dormancy breakage by the forest industry and by the Ministry of Forests in British Columbia. The chemical protocol offers the advantage of reducing the time required to break seed dormancy. We have now demonstrated that it yields seedlings that exhibit vigorous growth and are capable of withstanding the vagaries of the environment.  相似文献

Seeds of Strychnos nux-vomica have slow and erratic germination; thus different presowing treatments were applied to enhance the germination of its seeds collected from Tamaraikulam, Tamil Nadu, India. In addition, the effects of desiccation and different storage conditions on the germination of S. nux-vomica seeds were investigated. The results show that soaking in 500 ppm gibberellic acid (GA3) for 24 h, incubation of seeds at 40 °C for 3 days and alternate water soaking (16 h) and drying (8 h) for 14 days significantly increased the percentage germination compared to the control. Desiccation of seeds down to 10% moisture content resulted in better germination. Germination of S. nux-vomica seeds differed significantly between different storage periods, moisture contents of the seed and for first and second order interactions (p<0.001). The highest germination (92%) was achieved when seeds with 10% moisture content were stored at ambient temperature for 30 weeks. Evidence from the present study indicates that S. nux-vomica seeds possess physiological dormancy that can be broken effectively by after-ripening. As seeds of S. nux-vomica are found to be desiccation tolerant, dry seed (10% moisture content) can be hermitically stored at ambient temperature for 30 weeks without losing their viability.  相似文献

Laurel (Laurus nobilis L.) seeds were collected from the west part of Turkey in the fall of 2002. Seeds with pericarp (+) or without pericarp (−) were treated with 1000 mg/l, 2000 mg/l and 3000 mg/l GA3 concentrations, cold stratified at +4 ± 1°C for 25 and 50 days, punctured and seed coat removed to overcome and assess the mechanism of laurel seed dormancy. A period of 50 days cold stratification and removing seed coat significantly increased germination rate to 55% ± 1.91 (Mean ± SE) and 85% ± 3.00, respectively. None of the seeds with pericarp germinated regardless of treatment they underwent. Results suggested that seed dormancy was mainly due to pericarp and perhaps inhibitors linked to seed coat.  相似文献

Stratification by moist chilling is often used to break seed dormancy in loblolly pine (Pinus taeda L.). The role that moist chilling plays at the cellular level in preparing the embryo for germination and early seedling growth was investigated. Moist chilling did not affect the embryo's ability to mobilize seed storage proteins, which is a biochemical marker of early seedling growth. Analysis of in vivo protein synthesis profiles indicated that moist chilling has only a subtle effect on gene expression in the embryo or germinant. The means by which moist chilling affects interactions between the seed coat and the living tissues of the seed also was studied. Although the seed coat is mainly a mechanical barrier to germination, seed coat replacement studies showed that moist chilling alleviates some factor(s) in the seed coat that significantly inhibit germination.  相似文献

We measured physiological parameters including water uptake, in-vitro embryo germination ratio, and seed coat structure observed by scanning electron microscopy (SEM) to explore the influence of seed coat on the germination of seeds of tallow tree (Sapium sebiferum (Linn) Roxb.). Tallow tree seeds had good water permeability. We found that germination of cabbage seeds was inhibited when cabbage seeds were soaked in extracted solutions from tallow tree seed coat. Seed coat structure at the side of the radicle appeared to be a barrier to seed germination. We tested methods to break tallow tree seed dormancy. Dormancy of tallow tree seeds was overcome by soaking the seeds in 500 mg·L-1 or 1000 mg·L-1 GA3, followed by 100 days of cold stratification.  相似文献

Five chemical and four non-chemical presowing treatments were tested on seeds of three conifers (Larix principis-rupprechtii Mayr, Pinus sylvestris L. var. mongolica Litv, and Pinus tabulaeformis Carr.). By itself, cold stratification with snow for 60 days did not significantly improve the germination percentage over that of soaking the seed in warm water (45 °C) for 12 hours. However, including either one or two dehydration-rehydration cycles after the cold stratification did improve both the percentage and speed of germination of seed from all three species. Soaking seed for 12 hours in various chemicals (KNO3, CaCl2, CuSO4, KMnO4 and GA3) before germination was less effective than the snow stratification followed by double dehydration-rehydration treatments (Chang and Lin, 1989).  相似文献

Leucaena leucocephala and Acacia farnesiana are tree species used for several agricultural purposes in the Mediterranean region. The seeds of these species exhibit dormancy, causing delayed germination. Two experiments were conducted to investigate the effect of pre-sowing treatments (scarification, hot water, or soaking) on seed germination of L. leucocephala and A. farnesiana. In one experiment, seeds were exposed to three pre-sowing treatments: control, sandpaper scarification, or soaking in 70°C water for 4, 8, 12, 16, 20, or 24 min. In another experiment, seeds were soaked in 70°C water for 20 min, and then soaked in water at room temperature for an additional 24, 48, or 72 h or blade scarified. In general, soaking the seeds of the two species in hot water was more effective in breaking seed dormancy than scarification. Sandpaper scarification was not effective for either species. Blade scarification increased A. farnesiana seed germination to 56%, indicating that seed dormancy was mainly a consequence of hardseededness. L. leucocephala seeds collected from Jordan University of Science and Technology (JUST) site and soaked in 70°C water for 20 min and then soaked for 24, 48, or 72 h had germination rates above 97%. Our results suggest that blade scarification of A. farnesiana seeds and soaking of L. leucocephala seeds in 70°C water for 20 min are effective treatments to break seed dormancy and enhance seed germination of these vital species.  相似文献

[目的]通过研究滇重楼种子层积过程中不同阶段内源激素含量与种胚长度的动态变化情况来探讨种子休眠机理.[方法]应用酶联免疫吸附法测定不同阶段种子内激素含量.[结果]种子萌发过程中,脱落酸(ABA)含量由54.34 ng·g-1·FW降至32.16 ng·g-1·FW,赤霉素(GA)、玉米素核苷(ZR)显著升高,吲哚乙酸(IAA)含量波动较大,但整体呈逐渐升高趋势,随层积时间延长,未萌发种子中ABA含量持续降低,GA含量与GA/ABA值持续升高,仍显著低于已萌动种子中激素含量(p<0.05);种子在层积42天后,种胚开始膨胀,胚率快速增加;相关性分析结果,内源激素含量及比值与种胚生长呈显著相关(p<0.05),其中,GA/ABA值对种胚生长的影响最大.[结论]种子萌发过程中仅ABA含量降低不足以解除滇重楼种子休眠,同期GA含量升高和GA/ABA值达到某一阈值时能有效的解除滇重楼种子的休眠和促进胚的发育;种子在层积40天左右后,由于各项激素的综合作用,滇重楼种子由休眠状态逐步转变为破除休眠状态和萌发状态.  相似文献

目的]筛选云南甜龙竹和黄竹种子最佳表面灭菌条件,研究其萌发特性。[方法]筛选NaClO(2%、3%、4%)与HgCl_2(0.01%、0.1%)最佳消毒浓度组合;采用流水冲洗时间(6、12、18 h)、2%NaClO浸泡时间(5、10、15 min)和0.1%Hg Cl_2浸泡时间(5、10、15 min),设计正交试验实验,探讨2种竹种最佳表面灭菌处理的时间组合,并对竹种分别在滤纸、MS培养基以及经3 mg·L~(-1)GA_3浸泡后在MS培养基上的发芽率进行了探讨。[结果]NaClO、Hg Cl_2最佳消毒浓度组合为2%NaClO+0.1%HgCl_2;2种竹种最佳表面灭菌时间组合均为:流水冲洗6 h、2%Na Cl O浸泡10min、0.1%HgCl_2浸泡15 min,经此处理后云南甜龙竹和黄竹的发芽率分别为84.4%、72.2%,污染率分别为18.7%、29.6%。不同处理种子萌发率差异t检验结果显示:2种竹子种子是否经过3 mg·L~(-1)GA_3浸泡,对其在MS培养基上的发芽率无显著影响,但种子在MS培养基上的发芽率显著高于滤纸上的发芽率。经相同处理黄竹的发芽率均低于云南甜龙竹。[结论]云南甜龙竹、黄竹最佳表面灭菌组合均为:流水冲洗6 h、2%NaClO浸泡10 min、0.1%HgCl_2浸泡15 min;3 mg·L~(-1)GA_3浸泡处理对云南甜龙竹和黄竹种子在MS培养基上的发芽率无显著影响,但MS培养基上种子的发芽率显著高于其在滤纸上的发芽率。  相似文献

Morphological and biochemical changes of Chinese horse chestnut (Aesculus chinensis Bge.) seeds were studied during the process of maturation. Fruit and seed diameter increased linearly from DPA (Days Post Anthesis) 60 to DPA 150 and were stable thereafter. The weight of fruit and seed increased sigmoidally between DPA 100 and DPA 160. The water contents of whole seeds, axes and cotyledons had a sharp reduction from DPA 110 to DPA 130 but changed little after DPA 130. The water contents of whole seeds, axes and cotyledons were 1.42, 1.93 and 1.30?g?g?1DW, respectively at the end of maturation. The starch content of cotyledons from mature seeds was high on a dry weight basis (31.4%) while the total soluble sugar content varied between 12 and 20%. In both axes and cotyledons, the total soluble sugar content decreased during the process of maturation, as did all five soluble sugar components measured (glucose, sucrose, trehalose, raffinose and stachyose). Sucrose and stachyose contents remained relatively high at the time of seed maturation, followed by glucose and a trace level of trehalose. The oligosaccharide/disaccharide (O/D) ratio in the axes was 3.1 at DPA 140. The high level of sucrose and stachyose might contribute to the greater desiccation tolerance of Aesculus chinensis seeds relative to typical recalcitrant tropical seeds. High levels of ABA content in seed coats and a low GA3/ABA ratio in both seed coats and axes might also influence the desiccation tolerance and shallow dormancy of mature Aesculus chinensis seeds.  相似文献

The effects of exogenous gibberellic acid (GA3) and abscisic acid (ABA) on the total respiratory rate, percentages of total respiratory rate contributed by respiratory pathways [Embden-Meyerhof- Parnas Pathway (EMP), Pentose Phosphate Pathway (PPP), and Tricarboxylic Acid Cycle (TCA)], and conversion of starch to soluble sugars in the buds of black currants during secondary bud burst were investigated to determine the relationship between respiratory rates and secondary bud burst. ‘Adelinia’, a black currant cultivar that is prone to secondary bud burst after the first harvest, was used in this study. Mature bushes of Adelinia were sprayed with 30 mg/L GA3 and 50 mg/L ABA to manipulate bud burst. The results showed that exogenous applications of GA3 and ABA had opposite effects on bud respiratory rate. Generally, GA3 treatment increased the total respiratory rate and respiratory rate of the TCA and PPP, and the respiratory rates after GA3 treatment were higher than those of control. While ABA treatment mostly decreased the total respiratory rate and the respiratory rate of TCA and PPP in buds in comparison to control. In terms of the percentage of the three respiratory rates in comparison to the total respiratory rate, GA3 treatment significantly increased the percentage of TCA and PPP respiratory rate in comparison to the control (P < 0.01), whereas ABA decreased the rates. GA3 significantly increased the content of soluble sugars and decreased the starch content, while the starch content in buds after ABA treatment was significantly higher than that of the control. All results showed that PPP is a critical process for the second bud burst in black currants. While the EMP–TCA pathway is still dominant in bud respiration, provides a series of basic materials and energy (ATP). The conversion of starch to soluble sugars is essential for bud burst. Thus, we conclude that an energy shortage is a main contributor in ABA inhibition of the secondary bud burst of black currants.  相似文献

An unusually severe snow disaster in southern China in 2008 exposed variation among different species of bamboo: some species were highly tolerant to low-temperature damage, but some suffered extensive damage or death. Therefore, this study was conducted to determine the effects of cold stress on endogenous hormones and the expression of homologs of CBF 1 in four bamboo species differing in their tolerance to cold. All four bamboo species were subjected to cold stress with a temperature gradient (10, 5, 0, −5, and −10°C), and each low temperature treatment was maintained for 48 h. Results showed that ABA, GA1, and GA4 were significantly affected by cold stress: ABA increased with cold stress in all species, while GA1 and GA4 reduced with cold stress. ABA levels and the ratio of ABA/(GA1 + GA4) in leaves of Neosinocalamus affinis and Bambusa rigida were both higher than those of hybrid bamboo (Bambusa pervariabilis × Dendrocalamopsis grandis) and Dendrocalamus latiflorus for all treatments, while GA1 and GA4 were conversely lower as compared to the hybrid variety and D. latiflorus. Moreover, CBF 1 homolog was up-regulated during cold stress, with higher expression levels in the leaves of cold-tolerant species than cold-sensitive ones. Results revealed that high levels of ABA and CBF 1 , as well as the ABA/GAs balance, are important in increased plant resistance to cold stress.  相似文献

The effects of seed pretreatment and harvest date on the germination of European rowan (Sorbus aucuparia L) seeds were examined. In one experiment, seeds were subjected to drying, storage, soaking, warm and chilling treatments after harvesting in mid- and late August. In another experiment, fully imbibed (FI) seeds were given warm treatment for six weeks and then various durations of chilling (4?±?1°C) for up to 24 weeks. Thereafter, the seeds were adjusted to target moisture content of 35% and 30% or remained in the FI state and were then subjected to either subsequent chilling or freezing (?3°C) for up to 32 weeks. The treated seeds were allowed to germinate at a constant 15°C with 8 hours of lighting per day. Treatment effects were generally consistent for each harvest date. Seeds did not germinate unless they were chilled and germination rates were low unless the seeds were soaked. Warm treatment applied before chilling appeared to induce dormancy. The effect of storage varied with harvest date and storage treatment, with germination being highest for seeds harvested in late August. Mild freezing of ≥16 weeks duration following 16–24 weeks initial chilling resulted in >80% germination, more than could be achieved using chilling alone.  相似文献

We used pre-sowing treatments v/z., soaking seeds in concen- trated sulphudc acid (CSA), gibberellic acid (GA), combined treatment of CSA + GA and mechanical scarification to overcome seed dormancy and enhance synchronous germination of Macaranga peltata seeds. We analysed percent seed germination data by logistic regression and con- finned that except in GA treatment, time and acid concentration together were crucial for enhancing germination. The combination treatment of CSA and GA resulted in higher percent germination (up to 74%) than either treatment used separately, but produced defective seedlings (26%). Mechanical scarification of seed coat had the greatest impact in enhanc- ing germination (78%) and reducing imbibition time (6 days) against the control (0%). Germination studies and SEM analysis confirmed that seed germination in M. peltata was inhibited by seed coat dormancy.  相似文献

The ability of solid matrix priming (SMP) to overcome seed dormancy ofseeds of four fir species (Pacific silver fir, subalpine fir, grand fir andnoble fir), was examined. Seeds were subjected to treatments that combinedmoistchilling and SMP (SMP-chilling) and their germination compared to that elicitedby traditional moist chilling. For the SMP-chilling, six solid matrices withdifferent water holding capacities were tested (sphagnum, cat litter, peatmoss,Agro-Lig Greens Grade, Agro-Lig Micro Fine and sand). For each of the speciestested, germination performance was significantly improved by SMP-chilling; theresults were dependent on the type of solid matrix, the matrix water contentandthe treatment duration. SMP-chilling, using the best solid matrices for each ofthe species tested, resulted in germination percentages and rates that were24–35% and 8–21% higher, respectively, as compared with thatyieldedfrom control moist-chilling. High germination percentages (79% or greater) ofthree of the four species (Pacific silver fir, subalpine fir, and grand fir)were promoted after just four weeks (grand fir in sand) or eight weeks oftreatment (Pacific silver fir in peat moss and subalpine fir in Agro-Lig GreensGrade). The germination rate of grand fir seeds benefited from SMP-chillingperiods longer than four weeks, but this resulted in germination during thedormancy-breaking treatment. The water contents of the solid matrices that wereoptimal in terms of eliciting the best germination performance were 320% (w/w)for peat moss, 40% for Agro-Lig Greens Grade and 5–15% for sand.Conducting SMP at chilling temperatures (4 °C) is a practicalapproach to shorten the time required for effective dormancy termination ofAbies seeds; treatment times were shortened by 27 days forPacific silver fir and subalpine fir and more vigorous seedling growth wasexhibited by grand fir and noble fir.  相似文献

Mature seeds of H. salicifolia, collected from five provenances (i.e. Hanuman Chatti, Helang, Lata, Rambara and Janggal Chatti) in west Himalaya, India, were treated with stratification (at 4°C for 15, 30 and 60 days) and in different concentrations of GA3 (5, 10, 20 mM), KNO3 (50, 100, 200 mM) and Thiourea (50, 100, 200 mM) solution to determine the variations in seed germination. Results reveal that the germination rates of seeds from different provenances under different pre-sowing treatments are significantly increased compared to those in control (24%–30%). The seeds treated with Thiourea (100 mM) have highest germination rate (76%–83% for different seed sources), followed by those (63%–71% for different seed sources) pretreated with stratification (4°C, 30 days). GA3 treatment significantly shortens the mean germination time (MGT) and improves seed germination percentage. Considering the practical applicability and cost effectiveness, thiourea (100 mM) and stratification (at 4°C) treatments for seed germination are recommended for mass multiplication through seeds of H. salicifolia in village/forest nurseries of the west Himalaya, India.  相似文献

对八棱海棠种子分别进行赤霉素溶液浸泡处理、低温裸层积、低温层积以及赤霉素与低温层积结合等处理,而后进行发芽试验,统计分析发芽率、发芽势和发芽指数等指标.结果表明:赤霉素溶液浸泡不能完全打破种子休眠;低温层积是解除种子休眠的有效方法,层积80 d后,即可解除种子休眠;赤霉素溶液浸泡后,再低温层积可显著缩短层积时间.  相似文献
