为探究马铃薯在应答马铃薯块茎蛾Phthorimaea operculella取食时相关抗虫指标的动态变化, 本研究以‘青薯9号’‘合作88’‘米拉’3个马铃薯品种为材料进行室内盆栽试验, 在马铃薯块茎蛾取食0、3、6、12、24、48、72、96 h时取样测定各品种生理指标。结果表明, 被马铃薯块茎蛾取食的叶片中过氧化氢酶、多酚氧化酶、萜烯合成酶活性, 单宁和类黄酮含量与对照相比都有不同程度的显著升高, 总氨基酸含量显著下降, 且各指标在马铃薯品种之间存在差异。从各指标平均增降率来看, ‘合作88’对马铃薯块茎蛾取食的响应程度要强于‘青薯9号’和‘米拉’‘米拉’的响应程度最小。本研究为进一步明确马铃薯抗虫生理防御机制和害虫综合防治提供理论依据。  相似文献

Potato tuber worm, Phothorimae operculella Zeller (Lepidoptera: Gelechiidae) is a common pest of potato which causes a heavy loss in yield either in fields or storage. The effect of six potato cultivars (Agria, Florida, Impala, Picasso, Satina, and Sprint) were evaluated on the life history, life table and demographic parameters of P.operculella under laboratory conditions, which could be appropriate indices in resistance and susceptibility evaluation of potato cultivars. The longest development time was on Picasso (28.76 ± 0.36d) cultivar. The male longevity ranged from 4.83 ± 0.44d on Sprint to 7.45 ± 0.60d on Picasso. Impala and Satina with 53.22 ± 7.78 and 28.74 ± 3.54 eggs/individual had the highest and the lowest net reproductive rates (R0). The highest values for the intrinsic rate of increase (rm) and finite rate of increase (λ) were on Impala (rm: 0.119 day?1, λ: 1.75 day?1), and the lowest value was on Sprint (rm: 0.090 day?1, λ: 1.01 day?1). The results indicated that Sprint was the most resistant cultivar for the feeding of P.operculella, which could be useful in the development of integrated pest management programs for this pest.  相似文献

为了筛选非寄主植物滇杨的挥发物中驱避马铃薯块茎蛾产卵的活性成分,在室内利用选择性产卵试验方法分别测定了8种挥发物:丁香酚、苯甲酸、苯甲醇、苯乙醇、2-羟基苯甲醛、β-紫罗兰酮、水杨酸甲酯、苯甲醛不同浓度(0.75~12mg/L)的溶液对马铃薯块茎蛾产卵选择的影响。结果表明:水杨酸甲酯在6~12mg/L浓度范围内对马铃薯块茎蛾产卵有极显著的引诱效果。丁香酚在3~12mg/L浓度范围内对产卵有驱避效果,12mg/L时的产卵驱避率为62.1%。苯甲酸在1.5~12mg/L浓度范围内对产卵有驱避效果,但各浓度下的产卵驱避率不存在显著差异。12mg/L的苯甲醇和苯乙醇的产卵驱避率分别为12.1%和20.2%。6mg/L和12mg/L的2-羟基苯甲醛的产卵驱避率分别为44.1%和33.2%。β-紫罗兰酮在1.5mg/L浓度时产卵驱避率为30.9%。12mg/L的苯甲醛产卵驱避率为38.5%。本文测试的8种挥发物中有7种在不同浓度范围内表现出显著的产卵驱避效果。  相似文献

庚醛与桉叶油醇组合对马铃薯块茎蛾产卵选择的影响   总被引:3,自引:0,他引:3  
马铃薯块茎蛾(Phthorimaea operculella)是世界上为害马铃薯最严重的害虫之一,本研究利用桉叶油醇与庚醛驱避-引诱组合对其产卵行为进行调控,以探究最佳驱避-引诱组合规律及影响因素。在室内条件(27±1℃,L∥D=14h∥10h,RH:50%~70%)测定了桉叶油醇与庚醛单组分及两种组分驱避-引诱组合对马铃薯块茎蛾产卵选择的影响。桉叶油醇在6~12mg/L浓度范围内对马铃薯块茎蛾表现为产卵引诱作用,在18~30mg/L浓度范围内则表现为产卵驱避作用;庚醛在6~30mg/L浓度范围内对马铃薯块茎蛾表现为产卵驱避效果。将对马铃薯块茎蛾产卵行为引诱效果较好的桉叶油醇(3,6,12 mg/L)与驱避效果较好的庚醛(12,18,24mg/L)进行引诱-驱避组合,12mg/L庚醛与3mg/L桉叶油醇push-pull组合效果最好,push-pull效应达36.6%。对马铃薯块茎蛾产卵行为调控最佳的驱避-引诱组合为最低驱避浓度(12mg/L庚醛)与最低引诱(3mg/L桉叶油醇)浓度组合。产卵选择指数和push-pull效应与交配率、单雌产卵量不具有相关性,但是浓度变化可以导致交配率和单雌产卵量的变化。  相似文献

BACKGROUND: Sex pheromones of the potato tuber moths Phthorimaea operculella (Zeller) and Symmetrischema tangolias (Gyen) are ideal tools to monitor pest flight activity but are not used as means of control. The aim of the present study was to test the suitability of an attract‐and‐kill strategy consisting of pure pheromones and the contact insecticide cyfluthrin as the active ingredient, formulated with plant oils and ultraviolet absorbers, and applied in droplet sizes of 100 µL. RESULTS: Cyfluthrin at a concentration of 5 g L?1 resulted in the highest and fastest killing of males after 48 h, with a 100% mortality after 3–4 days. In contrast, control males survived for 13 days. In olfactometer experiments, the pheromone concentration of 0.5 g L?1 was significantly most attractive against eight virgin females. At controlled conditions (20 °C), no reduction in efficacy of the attract‐and‐kill formulation was observed for a minimum period of 36 days, whereas under natural environmental conditions the efficacy reduced gradually after day 6 of exposure. The longer the droplet was exposed, the longer was the time to reach 100% mortality of males. CONCLUSIONS: Compared with attract‐and‐kill studies for other pest species, the results are promising as a means of achieving highly effective control of potato tuber moths under field conditions. The potato tuberworm, Phthorimaea operculella (Zeller), is one of the most important pests of potato, Solanum tuberosum L., worldwide. We evaluated six Beauveria bassiana strains with potential for control of larvae. Strain SD was the most virulent, causing up to 98% mortality 10 days after inoculation of first instars. The other tested strains (HL, XJ, SC, JL and GZ) were only marginally virulent compared to the controls. In cage trials, SD strain applied to potato plants significantly reduced the survival of tuberworm larvae. Our results suggest that strain SD may be recommended for effective biological control of P. operculella.  相似文献

室内测定了从云南分离的7株球孢白僵菌对马铃薯块茎蛾幼虫的毒力,结果表明,在接菌孢子浓度为105个/mL和107个/mL下,接种后7d马铃薯块茎蛾幼虫累积死亡率分别为23.3%~70.0%和33.3%~86.7%,添加吐温80的清水对照死亡率仅为3.3%。筛选出毒力较高的Bb7001、Bb7004、Bb8001 3个菌株,其中菌株Bb8001对马铃薯块茎蛾幼虫的毒力最高,在孢子浓度1×105、1×107个/mL下,第7天的累计死亡率分别为70.0%和86.7%,其毒力回归方程Y=0.366X+3.409(R=0.956),剂量效应LC50=2.24×104个/mL,孢子浓度107个/mL下的LT50为4.3d。测定了毒力较高的菌株与12种农药的生物相容性,结果表明在供试农药推荐用量及其101、00倍稀释液下,球孢白僵菌Bb7001、Bb7004、Bb8001的3个菌株孢子在含所选农药的营养液中生物相容性良好的农药有:苏云金芽孢杆菌可湿性粉剂(Bt)、10%吡虫啉可湿性粉剂、2.5%菜喜悬浮剂及松毛虫病毒可湿粉剂,可配伍使用;而阿维菌素、农安、卡死克、抑太保对球孢白僵菌有很强的抑制作用,应避免配伍使用。  相似文献

Detailed characterization of a number of isolates of PVYSYR, a novel recombinant strain of Potato virus Y (PVY) from Syria, was conducted to elucidate their origin, assess their significance and achieve a final classification of PVYSYR. Recombination analysis grouped isolates of PVYSYR into three recombination patterns, SYR-I, SYR-II and SYR-III, which varied in the first 700 nucleotides of their genomes, with the second recombination pattern, SYR-II, the most frequent. PVYSYR isolates shared highest genomic identity and close phylogenetic relationships with PVYNTN and PVYNW isolates from Syria, suggesting a common origin and local emergence of these isolates in Syria. All PVYSYR isolates (total of 20) induced tobacco veinal necrosis, but reacted to a PVYO monoclonal antibody, typical characteristics of the previously reported PVYNW (or PVYN:O). In potato, however, four isolates tested (one of SYR-I and three of SYR-II) induced potato tuber necrotic ringspot disease (PTNRD), which is the characteristic phenotype of PVYNTN. Given the shared properties of SYR-I and SYR-II isolates with PVYNTN and PVYNW, it was decided that they represent a new recombinant strain of the PVYN strain group, with the proposed name PVYNTN-NW. The classification of SYR-III will be possible only after testing the phenotype in potato. The high prevalence of PVYNTN-NW in potatoes and weeds, as well as its ability to induce PTNRD, demonstrates its importance and the necessity for its control.  相似文献

为实现田间草地贪夜蛾成虫种群动态的智能化监测,本研究对本文作者前期研发的基于性诱和物联网的害虫智能监测设备进行了田间应用和评估。研究发现,该智能监测设备的实时采集数据和人工计数数据所反映的草地贪夜蛾高峰期均为9月20日-9月22日、10月14日-10月22日和11月4日-11月6日,自动计数的平均准确率达75.03%;桶形诱捕器仅监测到9月20日-9月22日和10月14日-10月22日这2个高峰期,但整个监测期间桶形诱捕器诱集到的草地贪夜蛾种群数量与智能监测设备监测到的种群数量没有差异(Z=-0.784,P=0.433)。此外,该智能监测设备的实时监测数据表明:草地贪夜蛾成虫具有明显的昼夜活动节律,而且受环境温度影响较大。当日平均温度≥16℃时,大多在18:00后开始活动,晚上12点左右达到活动高峰期;当日平均温度<16℃时,草地贪夜蛾成虫整体活动性不强,但在16:00-16:59和23:00-00:59出现两个活动小高峰期。以上结果表明,基于性诱和物联网的智能化监测可以用于草地贪夜蛾的远程实时测报。  相似文献

To investigate the ability of black dot symptoms to develop on infected potato tubers during storage, the growth of Colletotrichum coccodes was followed in vitro on malt agar at temperatures ranging from 5–27°C, and in vivo on artificially infected potato tubers kept at 5, 10 and 15°C. In vitro , 13 isolates from different geographical origins grew at all temperatures tested; growth started with a delay of 10 days at 5°C and of 4 days at 10°C, and was fastest at 27°C. All isolates had similar growth patterns and produced conidia and sclerotia at all temperatures. Minitubers were successfully infected at 5, 10 and 15°C by depositing either a mycelial plug or a drop of conidial suspension on the tuber surface. Sclerotia were observed after 7 days at the point of inoculation. Symptoms extended in all cases, although more slowly at 5 and 10 than at 15°C. Latent infections were detected in up to 21% of tubers without black dot symptoms at harvest. These results show that latent infections by C. coccodes are probably quite frequent, and that the pathogen is able to develop at low temperatures in controlled conditions. This suggests that black dot symptoms can increase during storage if stores are not adequately managed.  相似文献

Reactions of three Polish potato cultivars to potato virus S (PVS) were investigated at 22°C. Cultivars Tajfun and Tonacja exhibited partial resistance with systemic infection detected in some inoculated plants; cultivar Bryza was susceptible to PVS with systemic infection detected in all inoculated plants. The virus was not detectable by ELISA at 23 days postinoculation (dpi) but was detected after 40 dpi. Infection rate and viral accumulation were significantly lower in Tonacja and Tajfun than in Bryza, but no statistically significant difference between Tajfun and Tonacja was detected. Both susceptible and resistant genotypes displayed various, either common or cultivar-specific, symptoms. Delayed systemic infection at 56 dpi was observed in some cases in Tonacja and Tajfun. Resistance-related alteration of a set of miRNAs and mRNA targets in the tested cultivars in response to PVS at 22°C exhibited inter- and intracultivar variability. The majority of tested genes were altered only in the partially resistant Tajfun and Tonacja but not in the susceptible Bryza. Enhanced expression of AGO1-2, DCL1, stu-miR482 and its target Gpa2 was observed in Tonacja and plants of Tajfun in which PVS was detected, with the highest induction of Gpa2 in Tajfun (30.2-fold). However, their expression remained unchanged or decreased in plants of Tajfun in which PVS was undetected. Increased expression of stu-miR168a and stu-miR172e was observed in Tonacja and the PVS-undetected plants of Tajfun, respectively, but not in the PVS-detected plants of Tajfun. This is the first report on cultivar-specific alteration of miRNA in a potato–PVS resistance interaction.  相似文献

Pectobacterium brasiliense (Pbr) infects a wide range of crops worldwide, causing potato blackleg and soft rot and vegetable soft rots. This study aimed to characterize the genetic diversity and virulence variability among 68 Pbr strains isolated from either symptomless potato progeny tubers, diseased potato plants, ware potatoes wash water, or vegetables grown in Israel, as well as strains isolated from symptomless seed tubers grown in Europe, or diseased potato plants grown in France. The collection was typed using PCR and TaqMan real-time PCR analyses, dnaX sequence analysis, pulsed-field gel electrophoresis (PFGE), and pectolytic activity. dnaX phylogeny grouped almost all strains in a common genetic clade related to Pbr, which was distinct from the other Pectobacterium species. PFGE analysis identified two main clusters, including one major group of 47 strains with 95%–100% similarity. Maceration assays on two potato cultivars showed significant differences between strains but with no correlations with the source of the strains nor the status of the host (with/without symptoms). Molecular (dnaX sequences and PFGE profiles) and phenotypic analyses (tuber maceration tests) showed that the tested Pbr strains are not a homogeneous group. Analysis of the tested Pbr strains isolated from potato and vegetables grown in fields with a history of potato cultivation suggests that seed tubers imported from Europe may be the main source for Pbr in Israel. To the best of our knowledge, this is the first study that describes biodiversity and population structure of P. brasiliense isolated from potato and vegetables under hot climate conditions.  相似文献

A PCR-based diagnostic method was developed for direct detection from tuber lesions of pathogenic Streptomyces causing common scab of potato. Primers were designed to amplify a fragment of the txtAB ( txtA and txtB ) genes, which are pathogenicity determinants in the main pathogenic Streptomyces species. The method was evaluated on 84 naturally infected potato samples, comprising 19 potato cultivars, harvested in the years from 2000 to 2004 in the Netherlands, the UK, France, Germany and Spain. Pathogenic Streptomyces in tuber lesions were detected by PCR in 70 samples and were also successfully isolated from these 70 samples. All pathogenic isolates showed the basic general phenotypic traits of the S. scabiei phenetic cluster. RFLP analysis of amplified rRNA sequences, together with carbon source utilization and repetitive BOX profiles, allowed most isolates to be assigned to S. europaeiscabiei , which emerged as the main cause of potato common scab in Western Europe.  相似文献

首次在南昌应用高空测报灯对蝼蛄全年田间发生种群动态进行诱集监测。通过逐日统计灯下蝼蛄诱捕成虫结果表明:南昌地区灯下诱捕到的蝼蛄100%全部为东方蝼蛄。2014年5月15日至12月1日,期间共计诱捕东方蝼蛄4967头,全年灯下未见华北蝼蛄。东方蝼蛄成虫在全年内有多个高峰时期,灯下成虫高峰时期主要分布在6月上旬、6月底至7月上旬和7月下旬,8月5日后至监测结束,成虫零星发生。  相似文献

Over a 5‐year period (2006–2010), 277 certified, visually healthy potato seed lots, imported from Europe to Israel for commercial use, were tested for Dickeya spp. latent infection by PCR analysis (277 seed lots) and ELISA (154 seed lots). Seeds from these lots were grown in commercial potato fields which were inspected twice a season by Plant Protection and Inspection Services (PPIS). Stem samples were tested for the presence of Dickeya spp. by PCR analysis. PCR and ELISA results from seed lot testing correlated with disease expression in 74 and 83·8% of the cases, respectively. Positive laboratory results with no disease symptoms in the field (‘+lab/?field’ results) comprised 24·7 and 9·7% of the PCR and ELISA analyses, respectively, whereas negative laboratory results with disease symptoms in the field results (‘?lab/+field’) were obtained in 1·3 and 6·5%, of cases respectively. Maximum disease incidence, as well as the number of cultivars expressing disease symptoms, increased over the years of this study, indicating an increase in the prevalence of the disease. Severe disease incidence was observed on cvs Dita, Rodeo, Desiree, Mondial, Tomensa and Jelly. Of the 55 imported seed lots from which disease was recorded in the field, 49 originated from the Netherlands, four from Germany and two from France. None originated from Scotland.  相似文献

Changes in the incidence and onset of potato late-blight epidemics in Finland were investigated and compared with possible changes in climate, presence of soil-borne inoculum, and aggressiveness of Phytophthora infestans populations. Datasets were constructed from leaf blight assessments in cultivar trials or fungicide tests carried out at eight experimental sites during the periods 1933–1962 and 1983–2002. Additional data were obtained from late-blight monitoring projects carried out from 1991 to 2002. From 1998 to 2002, the risk of blight outbreak was 17-fold higher compared with the periods 1933–62 and 1983–1997. Simultaneously, the outbreaks of the epidemics began 2–4 weeks earlier. The changes observed were associated with a climate more conducive to blight in the late 1990s. Lack of rotation also advanced blight epidemics by an average of 9 days in 1998–2002, but it did not have this effect in 1992–1997, suggesting that soil borne inoculum may not have been a significant threat to potato until the late 1990s. The aggressiveness of the P. infestans isolates seemed to have only minor effect on the onset of the epidemics after 1991, as the apparent infection rate remained unchanged despite weather conditions more favourable to late blight in the late 1990s. As a consequence of the more frequent and earlier epidemics, the sales of fungicides used against late blight in Finland increased 4-fold from the 1980s to 2002.  相似文献
