An experiment was conducted to evaluate the effects of dietary composition on apparent digestibility coefficients of protein, fat and carbohydrate of gilthead sea bream. In addition α‐amylase and proteolytic activities were measured in the digestive tract of fish, held at 20±1°C. Six experimental diets were formulated containing approximately 40%, 45% and 50% protein, 11% and 21% fat and a starch level which fluctuated from 14% to 36%. Fish having an average weight of 100–130 g were used, fed at 1.5% of their body weight daily, and digestibility was measured using an indirect method. Enzyme activities were measured in the digestive tract of fish, fed diets containing a combination of 40%, 50% protein with 11%, 21% fat at 0.5, 5, 10, 24 h after a single meal. Starch digestibility was reduced with its level in the diet. It also negatively affected fat digestibility as well as protein digestibility, the last only at the high fat level. Dietary fat level had a negative effect on starch and protein digestibility. Fat affected also strongly α‐amylase levels in the digestive tract, while its effect on protease activity was of smaller magnitude. These results indicate that digestive enzyme activities and nutrient digestibility values are affected by dietary composition, carbohydrates and fat indicating the strongest effect.  相似文献

The effect of dietary carbohydrate complexity on growth, feed utilization and activity of selected key liver enzymes of intermediary metabolism were studied in gilthead sea bream juveniles. Four isonitrogenous (50% crude protein) and isolipidic (16% crude lipids) diets were formulated to contain 20% of pregelatinized maize starch, dextrin, maltose or glucose. Triplicate groups of fish (117 g initial weight) were fed each diet to near satiation during 6 weeks. No effect of dietary carbohydrate on growth was noticed. Feed efficiency was lower in fish fed the glucose diet than the maltose and dextrin diets. The lowest protein efficiency ratio was observed in fish fed the glucose diet. Six hours after feeding, glycemia was higher in fish fed the glucose diet than the maltose and starch diets. Liver glycogen content was unaffected by dietary carbohydrate complexity. Hepatic glucokinase (GK) activity was higher in fish fed the glucose and the maltose diets, while higher pyruvate kinase (PK) activity was recorded in fish fed the glucose diet than in fish fed the starch diet. Fructose-1,6-bisphosphatase (FBPase) and glucose-6-phosphate dehydrogenase (G6PD) activities were higher in fish fed the starch diet compared to dextrin and glucose diets. Data suggest that dietary glucose and maltose are more effective than complex carbohydrates in enhancing liver glycolytic activity. Dietary glucose also seems to be more effective than starch in depressing liver gluconeogenic and lipogenic activities. Overall, dietary maltose, dextrin or starch was better utilized than glucose as energy source by gilthead sea bream juveniles.  相似文献

The effect of dietary starch source and level on growth performance, feed utilization, apparent digestibility coefficients and liver enzyme activities involved in intermediary metabolism of gilthead sea bream juveniles was studied. Five isonitrogenous (47% crude protein) and isolipidic (15% crude lipids) diets were formulated to contain 10% native (diet NS10) or waxy (diet WS10) maize starch; 20% native (diet NS20) or waxy (diet WS20) maize starch or no starch (control). Diets were adjusted with α-cellulose. Another diet was formulated without carbohydrates, and contained 70% crude protein and 15% crude lipids (diet HP). Each diet was fed to triplicate groups of 30 fish (initial weight: 20 g) for 12 weeks. The HP group was fed to near satiation and the other 5 groups were fed on a pair-feeding scheme according to the group that ingested less feed (control diet group). The reduction of dietary protein level from 70% to 47% by the incorporation of 20% starch did not significantly affect gilthead sea bream growth performance or feed efficiency. Compared to the control diet, neither the level nor the nature of starch had any measurable effect on growth performance and feed efficiency. Digestibility of starch was unaffected by source or dietary inclusion level. Diet had no effect on plasma glucose levels, but liver glycogen was higher in diet groups NS20, WS20 and HP. Dietary carbohydrates increased GK and G6PD enzyme activities and decreased ALAT and GDH enzyme activities while had only a minor effect on FBPase activity. The nature of dietary starch tested (native or waxy) had little influence on performance criteria.  相似文献

This study aimed to evaluate the effect of temperature and dietary starch level on growth performance, nutrient utilisation, whole-body composition and activity of selected hepatic intermediary metabolism enzymes in European sea bass juveniles. An experimental diet was formulated to contain 46% crude protein, 12% crude lipids and 30% pregelatinized starch (diet 30GS); two other diets were formulated to include the same levels of all ingredients as diet 30GS except for the pregelatinized starch, which was included at 20% (diet 20GS) or 10% (diet 10GS). Diets were assigned to triplicate groups of 25 fish (initial weight: 15 g) at two water temperatures (18 and 25 °C) for 10 weeks. Group receiving diet 30GS was fed to apparent satiation and groups fed diets 20GS and 10GS were pair-fed according to group fed diet 30GS, so that each group ate the same amount of all nutrients except for carbohydrates. At the end of the trial, growth rate, feed efficiency and protein efficiency ratio were higher at 25 than at 18 °C. Dietary starch level did not affect growth or N utilisation. Feed efficiency was inversely related to dietary starch level. Whole-body lipids and energy content were higher at 25 °C and increased with dietary starch levels. Whole-body protein and ash content were also higher at 25 °C but were not affected by dietary starch level. Liver glycogen, HSI and VI increased with dietary starch level. ADC of starch was higher at 25 °C and within each temperature decreased with increasing dietary starch levels. ADC of protein was neither affected by temperature nor dietary starch levels. Data on enzyme activities showed that hexokinase (HK) but not glucokinase (GK) was affected by temperature. Dietary starch level affected the activities of glucokinase and glucose-6-phosphate dehydrogenase (G6PD) but not of the others enzymes tested (HK, PK, FBPase and GDH).  相似文献

To assess the effect of feeding frequency on the utilization of dietary starch by white sea bream juveniles, triplicate groups of fish were fed an experimental diet (400 g kg?1 protein, 140 g kg?1 lipids and 350 g kg?1 pregelatinized maize starch) to apparent visual satiation 2, 3 or 4 times a day for 63 days. Growth performance, feed utilization efficiency, glycaemia, cholesterolaemia, plasma triacylglycerides, liver lipids and glycogen content were unaffected by feeding frequency. α‐Amylase activity increased from the pyloric caeca to the posterior intestine and was higher in fish fed twice a day than in fish fed 3 or 4 times a day. Hepatic glucokinase (GK) activity decreased with the increase in feeding frequencies, whereas fructose‐1,6‐bisphosphatase (FBPase) activity increased. Hepatic glucose‐6‐phosphate dehydrogenase and glutamate dehydrogenase activities were unaffected by feeding frequency. Overall, feeding frequency did not improve white sea bream dietary starch utilization. α‐Amylase, GK and FBPase activities responded to dietary starch consumed at each meal, denoting a good metabolic adaptation of the fish to the feeding conditions.  相似文献

An experiment was performed to evaluate the growth performance and feed utilization of white sea bream juveniles (initial weight, 14 g) fed diets of cornstarch of different origins (normal and waxy). Four experimental diets were formulated to be isolipidic and to contain normal and waxy starch (26% or 42%) at two protein (36% and 48%) levels. The growth trial lasted 15 weeks and, at the end of the trial, there were no differences in the growth rate among groups. At the highest dietary starch level – but not at the lowest level – the feed efficiency ratio and PER were significantly lower in fish fed the waxy starch diet. Protein efficiency ratio and N retention (% N intake) were not affected by starch source but were significantly higher in the diets with a lower protein content. No differences in energy retention (% energy intake) were observed among groups. Except for the protein content, which was significantly higher in fish fed diets with a high protein level, no other differences were observed among groups in whole‐body composition. Hepatosomatic Index (HSI) was significantly higher in fish fed high‐starch diets, but there were no differences in visceral indices among groups. The apparent digestibility coefficients of protein and energy were not affected by the dietary starch level, but were significantly lower in diets including waxy starch. Glutamate dehydrogenase activity was higher in fish fed high‐protein diets, but it was not affected by dietary starch source. Alanine aminotransferase and aspartate aminotransferase activities were not different among groups. Glucose 6‐phosphate dehydrogenase, malic enzyme and fatty acid synthetase activities were not affected by the dietary starch level, but were significantly lower in fish fed waxy starch. The results of this study indicate that diets for white sea bream juveniles may include up to 42% starch without negative effects on fish performance. Moreover, normal starch appears to be more efficiently used as an energy source than waxy starch.  相似文献

为探讨饲料淀粉水平对团头鲂成鱼生长、消化酶活性和肌肉成分的影响,选用初始均重为(161±2.7)g的团头鲂成鱼360尾,随机分成6组(每组3个重复),分别投喂含淀粉17.1%、21.8%、26.4%、32.0%、36.3%和41.9%的等氮等脂饲料9周。结果发现,团头鲂成鱼成活率和特定生长率不受饲料淀粉水平影响,但饲料添加适量淀粉能提高饲料和蛋白利用率,淀粉水平对肝脏和肠道中的总蛋白酶和纤维素酶活性不产生影响,却显著影响淀粉酶活性。肝体比、肝糖原和肌肉粗脂肪含量随饲料淀粉含量的增加而显著增加,血清血糖、胆固醇和甘油三酯含量不受饲料淀粉水平的显著影响,饲料中添加过量淀粉显著降低了血清补体3和补体4含量。以蛋白质效率和饲料效率为评价指标,经折线模型回归分析,得到团头鲂成鱼饲料中淀粉的适宜添加量分别为饲料干重的34.1%和31.4%,但考虑到血清补体的活性,团头鲂成鱼日粮淀粉水平不应超过36.3%。  相似文献

A 12‐week growth trial was conducted to evaluate corn gluten meal as an alternative protein source to fish meal in diets for gilthead sea bream juveniles. The experimental diets were formulated to be isonitrogenous and isoenergetic and to have 20%, 40%, 60% and 80% of fish meal protein, the only protein source in the control diet, replaced by corn gluten meal. At the end of the growth trial only the group fed the diet with 80% corn gluten protein exhibited significantly reduced growth and feed efficiency compared with the fish meal‐based diet. This was most likely due to a dietary amino acid deficiency in that diet. A trend was noticed for feed efficiency to improve with the replacement of fish meal protein in the diets up to 60%. There were no significant differences among groups in protein and energy retention (as percentage of intake). At the end of the trial whole body water content of the experimental groups was significantly lower and the lipid content of groups including 60% and 80% corn gluten protein was significantly higher than that in the control. No other differences were observed in whole body composition among groups. Apparent digestibility coefficients (ADC) of the diets were evaluated in a separate trial. The ADC of dry matter of the experimental diets was significantly higher than in the control diet; there were no significant differences among diets in the ADC of energy and protein, except for the ADC of protein of diet with 80% corn gluten protein, which was significantly lower than the control. The results of this study indicate that corn gluten meal can replace up to 60% fish meal protein in diets for gilthead sea bream juveniles with no negative effects on fish performance.  相似文献

Dietary mannanoligosaccharide (MOS) from commercial product, Bio‐Mos supplementation, has been examined for its effects on weight gain and feed conversion of domestic mammals and birds, but very few studies have evaluated the responses of aquacultural species to MOS. A feeding and digestibility trial was performed to asses the potential beneficial effect of two levels of Bio‐Mos on growth, feed utilization, survival rate and nutrients’ digestion of gilthead sea bream (Sparus aurata) with an initial average weight of 170 g. Bio‐Mos was added at 2 or 4 g kg?1 to a fish meal–based control diet, and each diet was fed to triplicate groups of 1‐year‐old gilthead sea bream. After 12 weeks, there were no differences in survival rate among fish fed experimental diets (P > 0.05). It was observed that a significant improvability existed for both growth and feed utilization in fish fed diets supplemented with Bio‐Mos (P < 0.05). Body proximate composition remained unaffected by Bio‐Mos supplementation in fish fed experimental diets (P > 0.05). Apparent digestibility values for protein, carbohydrate and energy were appreciably affected by the inclusion of two different levels of Bio‐Mos, only lipid digestibility was the exception. In conclusion, the results of this trial indicate that 2 g kg?1 dietary supplementation with BIO‐MOS seem to be most positive for gilthead sea bream production.  相似文献

Juvenile gilthead sea bream with a mean initial body weight of 5 g were fed for 12 weeks with experimental diets containing 10% and 20% fishmeal protein (sole protein source in the control diet) replaced by processed pea seed meals. The processed pea seed meals were dehulled, defibred, extruded and microground pea seed meal (PSM1) or whole pea treated by infrared radiation and ground (PSM2). Apparent digestibility coefficients of the experimental diets were determined in a separate trial. At the end of the growth trial there were no significant differences in growth performance, feed utilization or whole-body composition among experimental groups. There were no differences in apparent protein digestibility among experimental groups (except for fish fed PSM1 at the lowest inclusion level). Both dry matter and energy digestibility of the diets, including PSM2 and with the highest inclusion level of PSM1, were significantly lower than those of the control diet. The results of this study suggest that pea seed meal may replace up to 20% fishmeal protein in diets for gilthead sea bream juveniles without affecting fish performance. Further studies should focus on technological treatments to increase utilization of pea seed meal carbohydrate, as both apparent dry matter and energy digestibility were affected by dietary inclusion level and by pea seed meal processing method.  相似文献

A 12‐week growth trial was performed to evaluate the effect of lupin seed meal as a protein source in diets for gilthead sea bream (Sparus aurata) juveniles. Six experimental diets were formulated to be isonitrogenous and isoenergetic and to contain 10%, 20% and 30% of raw lupin (Lupinus angustifolius) seed meal protein or 20% and 30% lupin (L. angustifolius) seed meal processed by infrared radiation (micronized) in place of fish meal protein, the only protein source of the control diet. Fish accepted all diets well and no significant differences in feed utilization among groups were noticed during the trial. Final weight of fish fed the experimental diets was identical or higher than the control group. Final weight of fish fed diets including 20% micronized lupin protein was even significantly higher than that of fish fed the fish meal‐based control diet. Moreover, at the same dietary lupin seed meal protein inclusion levels, final weight of fish fed diets including micronized lupin was significantly higher than with raw lupin. A trend was also noticed for a decrease of final weight with the increase in lupin seed meal in the diets. At the end of the trial no significant differences in proximate whole‐body composition, hepatosomatic and visceral indices were observed among groups. It is concluded that lupin seed meal can replace up to 30% fish meal protein in diets for gilthead sea bream juveniles with no negative effects on growth performance. Furthermore, micronization of lupin seeds improves its dietary value for gilthead sea bream juveniles. At the same dietary lupin inclusion levels, diets including micronized lupin seeds promote significantly higher growth rates than raw lupin seeds.  相似文献

The purpose of this study was to determine whether gilthead sea bream and sea bass treated with combination of trimethoprim and sulfamethoxazole (TMP-SMX) differed in terms of physiological and innate immune biomarkers. Fish were exposed to TMP-SMX at 40 ppm concentration for 1 h as a prophylactic usage. Plasma cortisol, glucose, electrolytes (Ca, P, Na, K, Cl, and Mg) as well as plasma lysozyme activity, C-reactive protein (CRP), and ceruloplasmin (Cp) were measured soon after treatment and following 24 and 48 h in normal sea water for recovery. Treatment with TMP-SMX in both gilthead sea bream and sea bass led to an increase in plasma cortisol and glucose. Fluctuations in some electrolytes were found after treatment and during recovery period, however, the ratios of monovalent ions in treated sea bream were similar to control. Hematocrit values as well as plasma lysozyme activity in gilthead sea bream and sea bass were not affected by the treatment. CRP in gilthead sea bream and Cp in sea bass responded to the treatment with decreased levels. Both gilthead sea bream and sea bass displayed a rapid physiological stress response and sensitivity to TMP-SMX exposure, which requires more than 48-h period for regaining homeostasis.  相似文献

The effect of dietary protein reduction balanced by an increase in carbohydrate (starch) level on growth performance, feed utilization and intermediary metabolism of Senegalese sole juveniles was evaluated at two temperatures, 12 and 18°C. For that purpose two isolipidic (16% lipids) diets were formulated to contain 550gkg?1 protein and 90gkg?1 starch (diet HP:LC), and 450gkg?1 protein and 200gkg?1 starch (diet LP:HC). Each experimental diet was fed to triplicate groups of 20 fish (initial body weight: 15.9g) within each temperature. Diets had no effect on growth and feed utilization. Temperature affected growth but not feed efficiency, with fish growing more at 18°C. Fatty acid synthetase and glutamate dehydrogenase activities were higher at 12°C than at 18°C while activity of the other measured enzymes was not affected by water temperature. Glucokinase and malic enzyme activities were lower while fructose 1,6‐bisphosphatase activity was higher in fish fed the HP:LC diet. Our data suggest that protein can be reduced from 550 gkg?1 to 450 gkg?1 by increasing starch level in high lipid diets for Senegalese sole juveniles without affecting overall performance. A reduction in protein content through an increase in dietary starch decreases hepatic gluconeogenesis. Increasing temperature from 12°C to 18°C improves fish growth but does not affect feed efficiency.  相似文献

We evaluated the effects of different proportions of dietary protein (5%, 10%, 15%, 20%, 25% and 30% protein) on the activity of digestive enzymes of normal and albino Apostichopus japonicus. The experimental diets were fed for 60 days, the optimal conditions for digestive enzyme activity in sea cucumbers were studied. The optimal temperature for protease was 29.3°C and the optimal pH was 1.8. The optimal temperature for amylase was 34.3°C and the optimal pH was 6.7. The optimal temperature for cellulase was 56°C and the optimal pH was 5.9. The activity of intestinal protease increased at first and then decreased as the proportion of dietary protein increased, reaching the maximum when the proportion of protein was 19.7%. The activity of protease in the intestine of normal sea cucumber was significantly lower than that of albino sea cucumber, and the activity of amylase was significantly higher than that of albino sea cucumber. This study is expected to provide a basis for further explaining the ecological difference of albino and normal A. japonicus.  相似文献

After the development of the gastric function in juvenile fish, dietary proteins enter a two-phase digestive process comprising an acidic gastric phase followed by an alkaline intestinal phase. However, the main gastric protease, pepsin, is strictly dependent on the existence of a low-enough environmental pH. In 20-g gilthead sea bream, Sparus aurata, the mean minimal gastric pH is close to 4.5, while the mean pH in the duodenal portion of the intestine was nearly fixed at 6.5. The mean maximal gastric content of HCl was approximately 20 microEq for a low-buffering diet. Gastric proteases were more severely affected than intestinal proteases when assayed at actual sub-optimal pH values, 4.5 and 6.5, respectively. When the gastric proteases of juvenile fish were pre-incubated with a citric acid buffer at pH 6.0, the activity at pH 4.5 was very low, whereas when they were pre-incubated with the same buffer at pH 3.0, the activity at pH 4.5 was significantly increased; this fact suggests a deficient activation of zymogens during the gastric digestion and points to a potential approach to improve protein digestion in juvenile gilthead sea bream.  相似文献

The effects of steam pelleting or extrusion of a commercial feed on performance of 2 g (small) and 40 g (large) gilthead sea bream was studied. In addition the apparent digestibility of diets, gastric evacuation time (GET) and trypsin and amylase activities were measured in large fish. Fish size significantly increased protein and energy productive value, body protein and lipid, liver glycogen and liver lipid, however decreased daily growth index, feed intake and feed utilization, body moisture and hepatosomatic index. Extrusion processing decreased feed intake, more strongly for small fish, and increased daily growth index, feed utilization, protein and energy productive value, body protein and lipid. Digestibility of starch and energy increased with extrusion and GET of extruded feed was double that of steam-pelleted conditioned feed (i.e. 544 min versus 284 min). Digestive enzyme activities approached maximum values after 240 min for the pelleted diet, while those of the extruded continued to increase, at higher rates for amylase than trypsin. Pelleted diet indicated higher moisture for stomach digesta, while the moisture of the intestinal precipitate indicated a higher water-binding capacity for the extruded diet. The overall results indicate a better assimilation of the extruded diet, which could result from its prolonged gastric evacuation.  相似文献

A method for studying the efficacy of sulfadiazine as a marker to estimate feed intake is presented. Feeding studies were carried out with two species, rainbow trout as a freshwater fish model and gilthead sea bream as a marine fish model, using two temperatures and two sizes of juveniles. The study showed the different feeding behaviour observed in both species, depending on the temperature and the number of days feeding the particles with sulfadiazine, and confirmed a dominant‐subordinate behaviour especially in the case of gilthead sea bream juveniles.  相似文献

Early weaning of marine fish larvae with dry diets delays gut maturation and reduces growth rates. In juvenile and adult forms of several marine fish species, inclusion of dietary mannan oligosaccharides (MOS) improves gut integrity and functionality, but the effects of MOS inclusion in gilthead sea bream (Sparus aurata, L.) larval diets have not been addressed yet. Thus, this study assesses the effects of dietary MOS inclusion on survival, growth performance, gut morphology, feed acceptance and quality of gilthead sea bream larvae. For that purpose, 16 days post‐hatched gilthead sea bream larvae were fed four graded levels of MOS (Biomos®, Alltech, Nicholasville, KY, USA) in weaning diets as follows: 0 g kg?1 MOS, 0.5 g kg?1 MOS, 1.5 g kg?1 MOS and 2 g kg?1MOS. Dietary MOS did not affect feed acceptance in gilthead sea bream larvae (P > 0.05). MOS supplementation was correlated in a dose‐dependent way with higher larval survival (P = 0.026). After 15 days of feeding, dietary MOS increased whole larvae (P < 0.01) arachidonic acid, eicosapentaenoic acid and docosahexaenoic acid. Gilthead sea bream larvae fed 2 g kg?1 MOS presented higher gut occupation with goblet cells after feeding compared with larvae fed the other dietary treatments. Overall, the results suggest that inclusion of MOS in early weaning diets for gilthead sea bream improves essential fatty acid utilization and may promote growth and final survival.  相似文献

Four semi-purified diets, containing crystalline amino acids (CAAs), were fed to juvenile red sea bream, Pagrus major in order to ascertain the ideal dietary amino acid pattern for this species. A control diet containing 50% casein–gelatin as protein sources, but no CAAs were fed to the fish. The other diets contained 30% casein–gelatin and 20% CAAs. CAAs were added to diets to simulate with amino acid pattern of the red sea bream eggs protein (REP), red sea bream larvae whole body protein (RLP), red sea bream juvenile whole body protein (RJP), and brown fishmeal protein (BFP). The juveniles (average initial body weight, 1.58 ± 0.01 g) were maintained in triplicate tanks and fed twice daily for 30 days. The highest weight gain was observed in juveniles fed the RJP diet. No significant difference was observed in juveniles fed the RLP and BFP diet. Feed efficiency ratio, protein efficiency ratio and amino acid retention in the whole body were significantly (p < 0.05) affected by the simulated dietary amino acid patterns. The essential amino acid profile and A/E ratios of the whole body after the growth trial showed little difference among the dietary treatments. The results suggest that red sea bream juveniles are able to utilize high amounts of CAA in coated form. The amino acid pattern of RJP could be used as an appropriate of reference dietary amino acid for this species.  相似文献

Low temperatures, and the voluntary fasting that they induce, have been implicated in outbreaks of winter syndrome in farmed gilthead sea bream (Sparus aurata). Two experiments were performed with the objective of studying if a decrease in water temperature is sufficient to induce this disease or some of its associated signs. In the first experiment, water temperature was either acutely or gradually reduced to 8 °C. In the second experiment, fish were exposed to either 8 °C or 12 °C, or fasted at 14 °C. Although no mortalities due to winter syndrome were registered during any of the experiments, some of the signs described in affected sea bream were observed in 8 °C-exposed fish. Among the most relevant were the paleness and friability of the liver and the occurrence of fatty degeneration in the hepatocytes. During the experiments, the general state of health of fish was monitored by measuring hematic parameters and the plasma concentration of proteins, glucose and ions. Low temperatures (8 and 12 °C) and fasting at 14 °C resulted in significant decreases in the levels of the different plasma protein fractions. This drop was more important in all the 8 °C-exposed fish, due mainly to a higher descent of albumin, α1-globulins and fibrinogen. Moreover, 8 °C-exposed fish showed a fall in total white blood cells and a rise of plasma glycemia, as well as a significant drop of plasma potassium and calcium levels and a transient increase of plasma magnesium concentration. In 8 °C-exposed fish, the rate of water temperature descent did not modify the profiles of change of any of the studied parameters. These results indicate that at 8 °C gilthead sea bream are unable to maintain the levels of plasma protein fractions and ions present in control animals.
