Fibril reconstruction process, that is, the nucleation and growth of mixed type I collagen fibril of shark and pig, progressed faster than that of the individual collagen species of shark or pig. The reconstructed mixed collagen fibril had a greater resistance to return to the solution or to melt into gelatin in comparison with the counterpart consisting solely of shark collagen. The denaturation temperature of the mixed collagen gel was about 10 degrees C higher than that of shark, and about 5 degrees C lower than that of pig. By scanning electron microscopy, the diameter of mixed collagen fibril showed an intermediate range between shark and pig collagen fibril. The breaking strength of the mixed collagen gel was tougher than that of pig, but weaker than that of shark. Other physicochemical properties of the mixed type I collagen gel were observed to be at intermediate positions between those of shark and pig type I collagen gels.  相似文献

Alkali-solubilized collagens, prepared by alkali-acid extraction and alkali direct extraction (abbreviated AASC and ALSC, respectively), were characterized by dynamic viscoelastic measurement of collagen solution (10 mg/mL). The optimum preparative conditions in terms of yield and polypeptide size are as follows: for the alkali-acid extraction, a pretreatment with 0.5 or 1 M NaOH containing 15% Na(2)SO(4) within 5 days at 20 degrees C followed by the subsequent acid extraction, and for the alkaline direct extraction, a treatment with 0.5 M NaOH containing 10% NaCl at 4 degrees C for 20-30 days. A major portion of the polypeptide sizes of AASC and ALSC is composed of alpha chains (alpha1 and alpha2). Dynamic viscoelasticity of collagen solution was measured as a function of temperature. AASC showed a greater contribution of elastic behavior rather than viscous behavior. On the contrary, ALSC exhibits a stronger viscous behavior than elastic behavior.  相似文献

为了探讨不同花生品种的感官品质、理化与营养品质、加工品质与其蛋白质凝胶性之间的关系,提出适宜肉制品加工凝胶型蛋白质专用花生品种的主要特性和评价方法,构建适宜加工凝胶型蛋白质花生品质评价模型。以62个花生品种制得的分离蛋白粉为原料,采用物性仪测定其分离蛋白凝胶性,分析各品种花生的品质特性与凝胶性之间的关系,采用有监督主成分分析对41个品种的品质数据建立凝胶型蛋白质花生品质评价模型,并用另外21个品种品质数据进行验证。结果显示,不同品种花生蛋白质在相同条件下制得的凝胶特性差异显著。果形、粗蛋白、粗纤维、半胱氨酸、甘氨酸、精氨酸、亮氨酸、伴花生球蛋白Ⅰ、花生球蛋白/伴花生球蛋白、23.5kDa等10个指标与凝胶性在0.05水平上关系显著,采用有监督主成分分析建立凝胶型蛋白质花生品质评价模型,验证模型的相关系数为0.937。研究结果表明,花生的品质特性显著影响花生蛋白质的凝胶性。花生粗蛋白含量、胱氨酸含量、精氨酸及伴花生球蛋白Ⅰ含量高的品种具有更好的凝胶性。通过凝胶型蛋白质花生品质评价模型可以预测该品种花生蛋白质的凝胶性,为不同花生品种的加工利用和专用品种的选择提供依据。  相似文献

采用长春市具有代表性的阶地农田的黏壤土、河漫滩的细砂土和黄土台地的粉壤土为基质,以提取的土壤胶体为对象,采用实验室模拟方法研究了土壤胶体理化性质对其稳定性和可移动性的影响。研究结果表明3种土壤胶体的临界絮凝浓度(CFC),与伊利石/蒙脱石矿物含量成反比并随着矿物含量的增大而降低,可能是因为伊利石/蒙脱石矿物是易絮凝矿物,水化驱动力及水化后的不稳定性较大;去除游离氧化铁后的农土去铁胶体提高了稳定性,而黄土去铁胶体和河土去铁胶体降低了稳定性,主要是由不同的土壤胶体去除游离氧化铁后性质改变程度不一样引起的;去除有机质后的农土去有机质胶体、黄土去有机质胶体和河土去有机质胶体降低了CFC,并且降低程度是与土壤胶体的有机质含量成反比。  相似文献

Pollenosis is a disease that affects 1 in 10 of the Japanese population. During the season of cedar pollen dispersal, many patients suffer from symptoms such as sniffling, sternutation, and itching of the eyes. Japanese butterbur is a popular vegetable and is one of the few domestic vegetables in Japan. The anti type I allergic effects of an aqueous ethanol extract from aerial parts of Japanese butterbur (JBE) were evaluated in rats and RBL-2H3 mast cells. In the passive cutaneous anaphylaxis reaction in rats, a single oral treatment of JBE (1000 mg/kg) was found to suppress the reaction. In IgE-sensitized RBL-2H3 cells, JBE (10-100 microg/mL) inhibited beta-hexosaminidase release, leukotriene C(4)/D(4)/E(4) synthesis, and TNF-alpha production. Moreover, a high concentration of JBE (1000 microg/mL) suppressed smooth muscle constriction induced by histamine (10 microM) and leukotriene D(4) (10 nM) in a guinea pig trachea strip. The search for components in JBE with an inhibitory activity on mast cell degranulation was guided by inhibition of beta-hexsosaminidase release. Two eremophilane-type sesquiterpenes, six polyphenolic compounds, and two triterpene glycosides were isolated. Of these compounds, fukinolic acid, a principal polyphenol constituent, showed potent inhibitory activity (IC(50) value = 2.1 microg/mL). Consequently, On the basis of its inhibition of mast cell activation and direct smooth muscle reaction induced by released mediators, JBE was found to suppress the type I allergic reaction.  相似文献

通过在植烟土壤中添加玉米秸秆、猪粪、未腐熟有机肥和腐熟有机肥4种有机物料,研究了其对烤烟不同生长时期土壤理化性质的影响。结果表明:(1)4种有机物料均能在一定程度上改善土壤环境,提高植烟土壤肥力;(2)不同有机肥对土壤理化性质的影响存在差别。与其它3种有机物料相比,施用腐熟有机肥在提高土壤肥力、改良酸性土壤、增强土壤保肥、供肥性能和缓冲能力方面具有较好的效果;秸秆对于提高土壤钾含量以及控制土壤碱解氮水平具有重要作用;猪粪在提高土壤磷含量方面效果较好。  相似文献

The breakdown of collagenous networks with aging results in hypoactive changes in the skin. Accordingly, reviving stagnant collagen synthesis can help protect dermal homeostasis against aging. We searched for type I collagen biosynthesis-inducing substances in various foods using human dermal fibroblasts and found that cinnamon extract facilitates collagen biosynthesis. Cinnamon extract potently up-regulated both mRNA and protein expression levels of type I collagen without cytotoxicity. We identified cinnamaldehyde as a major active component promoting the expression of collagen by HPLC and NMR analysis. Since insulin-like growth factor-I (IGF-I) is the most potent stimulator of collagen biosynthesis in fibroblasts, we examined the effect of cinnamaldehyde on IGF-I signaling. Treatment with cinnamaldehyde significantly increased the phosphorylation levels of the IGF-I receptor and its downstream signaling molecules such as insulin receptor substrate-1 and Erk1/2 in an IGF-I-independent manner. These results suggested that cinnamon extract is useful in antiaging treatment of skin.  相似文献

该文为了得到生产微酸性电生功能水最好的操作条件,采用4因素Central-Composite中心组合设计,研究了不同生产条件(电压、盐酸溶液的流量、盐酸溶液的质量分数和原水的流量)对微酸性电生功能水的有效氯浓度、pH值、氧化还原电位和电导率的影响。试验结果表明,电压、盐酸溶液的质量分数和原水流量对有效氯浓度影响极显著(p<0.01);盐酸溶液的流量、盐酸溶液的质量分数和原水流量对微酸性电生功能水的pH值影响极显著(p<0.01);盐酸溶液的流量、盐酸溶液的质量分数和原水流量对微酸性电生功能水的氧化还原电位和电导率影响极显著(p<0.01)。这些差异通过线性和二次性模型来描述。有效氯浓度、pH值、氧化还原电位和电导率的决定系数都大于0.95。生产微酸性电生功能水最佳的工作参数是电压4.4V、盐酸溶液的流量1.04L/h、盐酸溶液的质量分数2.8%、原水的流量210L/h。生产的微酸性电生功能水的指标为有效氯浓度(32±2)mg/L、pH值6.40±0.1、氧化还原电位(920±10)mV和电导率(1130±30)μS/cm。结果表明此方法可以得到生产微酸性电生功能水的最优的操作条件。  相似文献

封育是退化草原生态系统恢复的有效措施之一。在梳理现有封育研究成果的基础上,归纳封育对植物多样性、生物量、植被演替、土壤物理与化学性状的影响效应,发现:受地理区域、气候条件及封育前退化程度等本底条件的影响,封育条件下植被多样性呈现出显著增加、基本稳定和明显下降3种结果,生物量则表现为围封初期增加,围封时间过长不利于维持较高生物量的变化趋势;封育使得种群拓殖能力与群落资源冗余,土壤种子库结构改善、密度增长,退化草原呈正向演替,并可能出现3种演替模式。封育消除了土壤紧实层,改善了土壤结构与性状,但对土壤养分的影响效应不一致。基于研究现状,提出今后应深入研究的6个内容与方向,对丰富封育条件下退化草原恢复效应具有指导意义。  相似文献

为了进一步改善高凝胶蛋白粉的品质,探讨了利用酶交联反应制取高凝胶蛋白粉的新方法,在进行了琥珀酰化酪蛋白添加量、反应pH值、转谷氨酰胺酶添加量对蛋白粉凝胶强度影响的单因素试验基础上,该文通过响应面分析法对制取高凝胶性蛋白粉的酶交联反应条件进行了优化,并通过SDS-PAGE电泳、扫描电镜等手段探讨了酶交联蛋白粉凝胶性能增加的机理。结果表明,最佳工艺条件为:琥珀酰化酪蛋白添加量2.29%、反应pH值5.87、转谷氨酰胺酶添加量1.06%、反应温度40℃、反应时间40min。理论最大凝胶值为1072.8g/cm2,验证值为1054.5g/cm2,说明预测模型可靠性高,可用于反应条件的优化。  相似文献

针对速溶豆腐花粉的制备工艺中需要对大豆进行热处理,热处理过程中大豆蛋白的热变性程度对豆腐花粉凝结的凝胶强度与凝结所需的时间具有显著影响,而现在速溶豆腐花粉的工业生产中还没有对豆浆热处理程度较为合适的标准。该文以大豆、大豆分离蛋白(soybean protein isolate,SPI)、大豆球蛋白(glycinin,11S)、β-伴大豆球蛋白(beta-conglycinin,7S)为原料,研究大豆蛋白热变性程度对速溶豆腐花粉凝胶成型的影响。研究表明11S比7S更难发生完全变性,SPI中的7S和11S比单独存在的7S、11S更难发生完全变性。传统制备方式、前热处理后喷雾干燥或冷冻干燥制备方式、先喷雾干燥或冷冻干燥后热处理制备方式对豆腐花凝结成型影响不同,其中传统方式制备的豆腐花凝胶效果最好,先干燥后热处理制备的豆粉凝胶效果比前热处理后干燥的豆粉好,引起豆腐花凝胶强度差异的主要原因是大豆蛋白中7S和11S热变性程度不同。制备同一凝胶强度的豆腐花,热处理温度越低,所需的热处理时间越长;制备高凝胶强度的豆腐花比制备低凝胶强度的豆腐花所能进行的热处理温度与时间范围小。大豆蛋白的7S处于完全变性而11S处于未完全变性的状态时,适合制备速溶豆腐花粉的大豆蛋白变性程度应控制热处理温度与时间范围为80℃时热处理20~65 min,85℃时热处理15~50 min,90℃时热处理10~35 min,95℃时热处理5~20 min。该研究结果为调控速溶豆腐花粉的凝胶特性提供理论依据。  相似文献

摘要:根据中国北方地区花生垄作覆膜播种的特点,为提高花生播种机工作效率,改善作业质量,研制了2BFD-4型花生覆膜播种机。该播种机突破了一垄两行的播种模式,增加了排种单体和仿形机构,解决了单体运动干涉与仿形问题,实现了宽幅花生播种。对排种器与开沟器结构进行了优化,提高播种和开沟性能。整机结构紧凑,性能可靠,作业效率高,覆膜平整。经田间试验表明:该播种机播种质量较好,空穴率为0.58%,重播率为1.75%,种子破碎率为0.37%,穴粒数合格率为95.03%,种肥垂直间距为41.52 mm,出苗率为88.33%,各项指标均达到了相关标准,可以适应中国目前北方花生机械化的发展要求。  相似文献

A procedure developed for soybean protein precipitation which was based on electrodialysis was tested for the production of acid casein from reconstituted skim milk. In a previous paper, the performance of bipolar membrane electroacidification (BMEA) was evaluated under different conditions of ionic strength (micro(added) = 0, 0.25, 0.5, or 1.0 M) and added salt (CaCl(2), NaCl, or KCl) (1). The aim of this study, which is the complement of the work on evaluation of BMEA performance, was to evaluate the functionality of the protein isolates produced by BMEA and to compare the BMEA isolates to commercial isolates and an isolate produced by chemical acidification. It was not possible to show differences between the functional properties of isolates produced by BMEA, except at 1 M CaCl(2) micro(added), due to the variability of the isolates. However, the results showed that it is possible to obtain isolates similar to commercial isolates and that the addition of salt during the process does not induce variations in functional properties. From results on mineral concentrations, it appeared that the addition of monovalent cations did not influence the retention of monovalent or divalent cations in the BMEA isolates, while addition of divalent cations (CaCl(2)) influenced the retention of magnesium. According to previous results on evaluation of BMEA performances under different conditions of ionic strength and added salt, the difference observed for the BMEA isolate produced at 1.0 M CaCl(2) was confirmed.  相似文献

The heat-induced denaturation curve of ovalbumin followed by the ellipticity at 222 nm in circular dichroism spectra was consistent with that monitored by fluorescence with thioflavin T, which is an indication of amyloid fibril formation, while other proteins such as lysozyme and ovotransferrin did not fluoresce with thioflavin T during heat denaturation. The amount of soluble aggregate formed during heat denaturation was proportional to the increase in fluorescence with thioflavin T. The binding of soluble aggregates with thioflavin T was greatly suppressed in heat-denatured ovalbumin in the presence of thioflavin T. The similar inhibition effect of thioflavin T on the gel formation of heat-induced ovalbumin was observed. These results suggest that the amyloidogenic intermolecular beta-structure is involved in the formation of soluble aggregate and gel of heat-induced ovalbumin.  相似文献

To prepare composite films from biopolymers with anti-listerial activity and moisture barrier properties, the antimicrobial efficiency of chitosan-hydroxy propyl methyl cellulose (HPMC) films, chitosan-HPMC films associated with lipid, and chitosan-HPMC films chemically modified by cross-linking were evaluated. In addition, the physicochemical properties of composite films were evaluated to determine their potential for food applications. The incorporation of stearic acid into the composite chitosan-HPMC film formulation decreased water sensitivity such as initial solubility in water and water drop angle. Thus, cross-linking of composite chitosan-HPMC, using citric acid as the cross-linking agent, led to a 40% reduction in solubility in water. The water vapor transfer rate of HPMC film, approximately 270 g x m(-2) x day(-1) x atm(-1), was improved by incorporating chitosan and was further reduced 40% by the addition of stearic acid and/or cross-linking. Anti-listerial activity of films was determined on solid medium by a numeration technique. Chitosan-HPMC-based films, with and without stearic acid, inhibited the growth of Listeria monocytogenes completely. On the other hand, a loss of antimicrobial activity after chemical cross-linking modification was observed. FTIR and 13C NMR analyses were then conducted in order to study a potential chemical modification of biopolymers such as a chemical reaction with the amino group of chitosan. To complete the study, the mechanical properties of composite films were determined from tensile strength assays.  相似文献

为研究牛鲨咬合力大的原因,并以牛鲨牙齿为原型设计仿生刀具,以牛鲨上颌齿和下颌齿为研究对象,利用扫描电子显微镜观察牛鲨牙齿的微观结构,利用能谱仪测试牙齿的元素组成。采用微观摩擦磨损试验机进行摩擦磨损试验,考察了牛鲨上颌齿、下颌齿的摩擦学性能,通过激光共聚焦扫描显微镜测量磨损深度,利用扫描电子显微镜观察牛鲨牙齿的磨损形貌。试验表明:釉质层由条状纤维束形成致密的结构,本质层为多孔结构,孔径范围在3~20μm,这些孔洞呈不规则的排布;能谱仪测试结果中,牛鲨牙釉质比牙本质中Ca、F含量高,氟和钙的组合效应可以帮助其在生存环境下有更好的耐酸性和硬度。牛鲨上下颌齿牙釉质磨斑表面有片状磨屑剥落,且有大量的细小磨粒,上颌齿磨损体积约为1.72×10~7μm~3,主要是磨粒磨损;下颌齿出现明显裂纹,磨损体积约为2.11×10~7μm~3,比上颌齿的磨损体积大。上述结果为新型刀具的仿生设计提供理论依据。  相似文献

豆浆的钙含量相对较低,且具有令人不愉快的豆腥味,限制了豆浆产品的消费需求。该研究以羟基磷灰石(hydroxyapatite, HA)为钙强化剂,研究了钙添加量和均质压力对豆浆粉理化性质、感官评价和挥发性风味物质组成的影响。结果表明:钙的添加会降低豆浆粉粉体的流动性,润湿和分散时间显著(P<0.05)增加;豆浆粉冲调后,剪切黏度增加,口腔摩擦系数明显提高,口感变得稠厚。在1.5%的钙添加量基础上,采用均质处理进一步改善豆浆粉的品质。低压均质(≤60 MPa)改善了豆浆粉的流动性、润湿和分散性。随着均质压力的升高,豆浆粉颗粒的表面变得光滑,坍塌减少,呈现接近球体的形态。当均质压力继续增大至80 MPa时,豆浆粉颗粒形貌略有破损。均质处理能使冲调后豆浆的黏度减小,口腔摩擦系数明显降低,口感变得顺滑。相比未加钙豆浆粉的风味物质种类(26种),钙强化豆浆粉的风味物质种类达35种。相比于未均质钙强化豆浆粉,20 MPa均质处理显著(P<0.05)降低关键性豆腥味物质(正己醛等)检测的含量,且关键性非豆腥味物质(正壬醛、反-2-辛烯醛、反-2-壬烯醛等)得到较好的保留,豆浆粉的色泽、风味、顺滑度和滋味显著(P<0.05)改善,感官评分最高。然而,过高的压力处理(80 MPa)使检测的正己醛含量略有上升,而正壬醛、反-2-壬烯醛的含量显著(P<0.05)下降,劣化了豆浆粉的感官品质,感官评分降低。该研究结果可为钙强化豆浆粉的加工及感官品质改善提供科学依据。  相似文献

Vescalagin and castalagin are two diastereoisomers. The variability of their principal physicochemical properties, compared with their small structural differences, suggests important conformational variations. This study shows, experimentally, that vescalagin has a greater effect on polarity, oxidizability in solution, and thermodegradability than castalagin. Conformational analysis by molecular mechanics demonstrated that vescalagin was more hydrophilic and was more reactive to electrophilic reagents than castalagin. Experimental results were thus explained and demonstrated the distinct behaviors of vescalagin and castalagin. These results were attributed to the C1 position of the two compounds because vescalin and castalin have comparable characteristics. Experimental data were confirmed and interpreted by molecular mechanics. This work represents one of the first attempts to correlate conformation and the properties of phenolic compounds. This step constitutes a predictive method for the pharmacology or chemistry of new compounds.  相似文献

Purpose

The aim of this article was to explore the hyperaccumulative property from cell membrane permeability, subcellular distribution, and chemical form of Solanum nigrum L. to Cd compared to the nonhyperaccumulator Solanum melongena L. (cultivar name Liaoqie 3) in the same plant family.

Materials and methods

Soil pot culture experiment was conducted, and the cell membrane permeability, subcellular distribution, and chemical forms of Cd in S. nigrum and S. melongena were determined.

Results and discussion

The results showed that the tolerance of S. nigrum to Cd was higher and owned all Cd hyperaccumulator characteristics. The relative electric conductivities in root and leaf were higher of S. nigrum than S. melongena, but the malondialdehyde concentrations in former were roughly lower than that in latter. Cd concentration ratio in cell wall, NaCl extractable form, and HAc (acetic acid) extractive form were higher in root, stem, and leaf of S. nigrum.

Conclusions

Compared to S. melongena, stronger Cd tolerance, higher cell membrane permeability with intact cell membrane, weak Cd forms in subcellular distribution, and relative lower bioactivity speciation may be part reasons of S. nigrum hyperaccumulating Cd.  相似文献

Oxazolidine E, an aldehydic cross-linking agent, is used to impart hydrothermal stability to collagen. The purpose of this study was to investigate the exact nature of oxazolidine E induced cross-links with collagen by using synthetic peptides having sequence homology with collagen type I. Tandem mass spectrometry revealed the formation of methylol and Schiff-base adducts upon reaction of oxazolidine E with the peptides. This was confirmed by allowing the reaction to proceed under reducing conditions using cyanoborohydride. Mass spectrometry (MS)-MS analysis clearly showed interaction of tryptophan and lysine residues with oxazolidine E and demonstrated that arginine could be cross-linked with glycine in the presence of oxazolidine E through the formation of a methylene bridge. Collagen fibrils regenerated from monomers in the presence and absence of oxazolidine E were studied using atomic force microscopy to investigate morphological alterations. Regenerated fibrils showing the typical 65 nm D-banding pattern were obtained from those formed both in the presence and absence of oxazolidine E, and there was no evidence of a change in the D-periodicity of these fibrils. This indicated that oxazolidine E does not hinder collagen molecules from correctly aligning to form the quarter-stagger structure.  相似文献
