Twelve biotinylated lectins and an avidin-biotin-peroxidase method were used to detect and localize specific carbohydrate residues on formalin-fixed, paraffin-embedded female canine mammary gland sections. Histologic sections from 3 lactating and 7 nonlactating mixed-breed dogs (age 5.6 +/- 0.35 years) were incubated with Arachis hypogea agglutinin (peanut agglutinin; PNA), Concanavalia ensiformis agglutinin (conA), Dolichos biflorus agglutinin (DBA), Glycine max agglutinin (SBA), Griffonia simplicifolia agglutinin-I (GS-I), Lens culinaris agglutinin (LCA), Lycopersicon esculentum agglutinin (LEA), Phytolacca americana mitogen (pokeweed mitogen; PWM), Ricinus communis agglutinin-I and -II (RCA-I and -II), Triticum vulgaris (WGA), and Ulex europaeus agglutinin-I (UEA-I). Each lectin had a specific binding pattern, except SBA and DBA. In nonlactating glands, PNA, conA, LEA, and UEA-I stained duct cells in a linear-binding pattern, with a mean percentage of positive ducts per section of 28.7 (+/- 0.6), 65.7 (+/- 0.3), 100 (+/- 0), and 8.4 (+/- 0.2), respectively. Strong apical, lateral, basal, and cytoplasmic positivity on duct cells was seen after incubation of the sections with RCA-I, RCA-II, and WGA in all ducts. In acinar cells, the binding pattern and the staining distribution of all the lectins studied were similar to those in duct cells. However, for PNA, conA, and UEA-I, the mean percentage of positive lobules per section was 33.7 (+/- 0.9), 62 (+/- 0.5), and 10.5 (+/- 0.2), respectively. The function and/or morphological features of the vomeronasal olfactory system remain unclear in aquatic animals, although the system appeared first in urodeles based on phylogenic data. We examined the lectin binding patterns in the olfactory bulb of a semi-aquatic urodele, the Japanese red-bellied newt, Cynops pyrrhogaster, using 22 different lectins. Eleven of the lectins showed specific binding to the nerve fibres and glomeruli in the olfactory bulb. Among these, Wheat germ agglutinin, pokeweed and peanut agglutinin preferentially bound the main olfactory bulb, reflecting variation in the expression of glycoconjugates between the main and accessory olfactory bulbs. By contrast, the types of lectins bound to the Cynops olfactory bulb were considerably different from those reported in other urodele families. These results suggest a histochemical distinction between the main and accessory olfactory bulbs, and that glycoconjugate expression may differ significantly among urodele families.  相似文献

The present study was undertaken to characterise the carbohydrate profile of the porcine small intestine using lectin histochemistry during the period from 3 days prior to weaning to 9 days post-weaning. A total of 56 piglets weaned at 4 weeks of age were included in the experiment. The most prominent changes in the glycosylation pattern were observed in the goblet cells. The highest lectin reactivity of the goblet cells in the crypts was observed 7 days post-weaning which suggests that the protective effect of the mucus layer against pathogenic bacteria is increasing during the postweaning period. The staining pattern of the apical membrane remained unchanged during the experimental period. This indicates that the glycosylation process in the goblet cells is rapidly inducible whereas changes in the glycosylation pattern of the apical membrane requires more time. The glycosylation pattern of both goblet cells and apical membrane differed between the positions of the small intestine. As glycoconjugates can act as attachment sites for microorganisms, these differences in the distribution of sugar residues may be one explanation for the site-specificity of certain pathogens.  相似文献

BACKGROUND: Intoxication caused by Senecio sp is characterized by irreversible damage to liver cells and may be associated with oxidative stress. OBJECTIVES: The aim of this study was to evaluate the effects of intoxication by Senecio sp on lipoperoxidation, antioxidant defenses, and the osmotic resistance of erythrocytes in cattle. METHODS: Blood samples from 30 intoxicated animals (group 1) and 30 healthy animals (group 2) were analyzed. The diagnosis of poisoning by Senecio sp was based on histopathologic lesions verified through hepatic biopsy. The following biochemical parameters of oxidative stress in the erythrocytes were determined: thiobarbituric acid-reactive substances (TBARS), copper-zinc superoxide dismutase (CuZnSOD) activity, and nonprotein sulfhydryl (NPSH) groups. Erythrocyte osmotic fragility also was evaluated. RESULTS: TBARS concentration and CuZnSOD activity were significantly (P <.001) higher in group 1 when compared with group 2. The concentration of erythrocyte NPSH groups was significantly (P <.03) lower in group 1 when compared with group 2. Osmotic fragility was more pronounced in the erythrocytes of group 1 when compared with group 2 (P < .001). CONCLUSIONS: The results of this study indicate that poisoning by Senecio sp causes an increase in lipoperoxidation, oxidation of NPSH groups, and consequently, oxidative stress in bovine erythrocytes that may contribute to hemolysis. These findings may contribute to a better understanding of the mechanisms involved in cell damage in animals intoxicated by Senecio sp.  相似文献

The lectin histochemical pattern (LHP) was characterized and compared in normal and cystic ovaries of sows. Six biotinylated lectins (PNA, SBA, WGA, RCA‐1, DBA and UEA‐1) were used on tissue sections. In the normal ovaries, the reaction to UEA‐1 and SBA was mild to moderate in mesothelial and endothelial cells. RCA‐1 staining was mild to moderate in theca interna of growing follicles, corpora luteum and mesothelium. In addition, this lectin presented strong reaction in endothelial cells, granulosa cells of atretic follicles, zona pellucida of growing follicles and plasma. DBA showed strong intensity in mesothelial and endothelial cells. There was mild to moderate reactivity to WGA in granulosa cells, corpus luteum and theca interna of follicles in development, and moderate in zona pellucida, in granulosa cells of atretic follicles and mesothelium. PNA staining was mild to moderate in oocytes and in the adventitia and media of medullary arteries. Changes in the LHP of the cystic ovaries were noted; however, there were no differences in these findings between the follicular and luteinized cysts. UEA‐1 reactivity in the cystic ovaries was moderately reduced in the mesothelial and endothelial cells, whereas there was mild reduction in the DBA staining in the granulosa cells. Reaction to RCA‐1 and WGA in the cysts also was decreased in theca interna, zona pellucida and granulosa cells of atretic follicles. Furthermore, endothelium and theca interna in the cystic ovaries presented mild reduction of marcation to SBA, whereas there was decreased reactivity to PNA in the oocytes and adventitia and media layers of the medullary arteries. The results of the current study show that cysts modify the LHP in swine ovaries. These changes of glycoconjugates in many ovarian structures could modify diverse process and may be one of the reasons for decreased fertility in sows.  相似文献

OBJECTIVE: To test Solanum glaucophyllum calcinotic effects in adult New Zealand White rabbits in relation to cumulative dose and active principle concentration in plasma. DESIGN: An intoxication assay with controls. PROCEDURE: Rabbits were orally dosed with aqueous extracts of dry leaves of S glaucophyllum for 5, 7 or 9 days. During the experiment, body weight, calcaemia and phosphataemia were measured; retinal blood vessel calibre was observed by ophthalmoscopic examination of the ocular fundus. 1,25(OH)2 vitamin D plasma concentration was determined at the end of the experimental periods. Soft tissue calcium concentration and the presence of calcinotic lesions were studied after euthanasia. RESULTS: Toxic effects were evident in S glaucophyllum treated groups (loss of body weight, elevation of soft tissue calcium concentration, and presence of calcinotic lesions). Plasma 1,25(OH)2 vitamin D concentrations were negatively correlated with final body weight (r = -0.97; P < or = 0.001), and positively correlated with renal calcium concentration (r = 0.74; P = 0.02). There was also a significant regression of plasma 1,25 (OH)2 vitamin D concentration on the cumulative dose of S glaucophyllum (R2 = 0.87; P < or = 0.001). CONCLUSIONS: The procedure described here offers a sensitive and practical experimental model for the study of the pathogenesis of enteque seco.  相似文献

Intoxication by pods of Prosopis juliflora (mesquite beans) causes an impairment of cranial nerve function in cattle and goats. In goats, vacuolation of neurons in the trigeminal motor nuclei has been reported. To study the lesions in cattle caused by consumption of P. juliflora pods and dry ground pods, eight 6- to 12-month-old male cattle were divided into 4 groups: group 1 was fed a ration containing 50% of pods; groups 2 and 3 received a ration containing 50 and 75% of dry ground pods, respectively; group 4 was the control. After 200 days, all cattle were killed and sampled for histologic evaluation. Samples of the trigeminal motor nucleus were examined by electron microscopy. All cattle from groups 1, 2, and 3 showed clinical signs resulting from impaired function of cranial nerves V, IX, X, and XII, starting 45-75 days after consumption of the plant. The main histologic lesions were vacuolation and loss of neurons in trigeminal motor nuclei and other motor cranial nerve nuclei with Wallerian-like degeneration in the cranial nerves. Mild denervation atrophy was observed in the masseter and other masticatory muscles. On electron microscopy, neurons of the trigeminal nuclei had markedly swollen mitochondria, with the mitochondrial cristae displaced peripherally, disoriented and disintegrating. Intoxication by P. juliflora seems to have a novel pathogenesis, characterized by a selective, primary, chronic, and progressive injury to mitochondria of neurons of the trigeminal and other cranial nerve nuclei. Cranial nerve degeneration and denervation atrophy of the muscles occurs as a consequence of the neuronal lesion.  相似文献

A neurological disease of cattle (maldronksiekte), occurring in a localized area of the Northern Transvaal, was experimentally reproduced by feeding Solanum kwebense plants to cattle. The disease is characterized by temporary loss of balance and transient epileptiform seizures precipitated by a variety of stimuli, such as exercise, handling (dipping, loading, etc) and fright. When not disturbed, most affected animals appear to be completely normal. The most conspicuous histopathological lesion is a neuronopathy manifested by vacuolar degeneration and eventual necrosis of neurones, particularly of the Purkinje cells in the cerebellum. An atrophy of the cerebellar cortex is seen grossly. The history, clinical signs and experimental reproduction of the disease, as well as the pathology of 4 experimental and 18 natural cases, are described.  相似文献

Cattle in western Uruguay that were eating Solanum bonariense developed periodic episodes of ataxia, hypermetria, hyperesthesia, head and thoracic limb extension, opisthotonus, nystagmus, and falling to the side or backward. Similar clinical signs were experimentally reproduced in cattle by administration of S. bonariense via rumen cannula at a dose of 1,024 g/kg body mass. No significant gross lesions were observed in field cases or experimentally induced cases. Spontaneous and induced histologic lesions were similar and included vacuolation, degeneration, and loss of Purkinje cells. Axonal spheroids, microcavitations, and other changes of wallerian-type degeneration in cerebellar white matter were also observed. Ultrastructural changes included increased number of electron-dense residual storage bodies in membrane-bound vesicles in affected Purkinje cells, and similar vesicles and mitochondria in axonal spheroids. No histologic lesions were detected in the other examined tissues. The Purkinje-cell swelling and vacuolation with subsequent cerebellar degeneration are suggestive of Purkinje-cell specific toxin that produces abnormal lysosome function and cell specific axonal transport. This is the first report of S. bonariense toxicity.  相似文献

Brachiaria decumbens and B. brizantha (signal grass), which occupy millions of acres in Brazil, are an important source of fodder for ruminants. Sporadic outbreaks of photosensitization in ruminants grazing on signal grass have been reported. Intoxicated animals showed the presence of foamy cells in the liver, spleen, intestinal submucosa and lymph nodes. These foamy cells are macrophages. They are very difficult to distinguish with haematoxylin and eosin stain, especially in the case of isolated cells. The purpose of the present study was to detect specific carbohydrate residues of storage material in the foamy cells in tissues of cattle exposed to Brachiaria spp. The characterization of glycoconjugates provides clues to the pathogenesis of these cells. Besides, the lectin peanut agglutinin was found to be an excellent marker to differentiate and quantify the foam cells, and could be used as a specific marker.  相似文献

Carbohydrates on epithelial cell surfaces play an important role as attachment sites for different microorganisms like bacteria, viruses and protozoa. To obtain more information about the distribution of carbohydrates on the luminal surface along the intestine, lectin histochemical studies on different gut segments of chicks of different age groups were carried out using a panel of 13 lectins with specificities for Man, Glc, Gal, GalNAc, GlcNAc or GlcNAc oligosaccharides and Sia. Furthermore, we tried to find out whether previously reported specificities of certain lectins for M cells (membranous or multifold cells) in the bursa of Fabricius (BF) can be observed also on M cells of the intestine. As a result we were able to demonstrate binding of all lectins employed in these studies in all investigated gut segments. In some cases, the application of the same lectin led to varying staining intensities of the same histological structures in different age-groups (e.g. staining of the brush border with WGA, LEA, MAA or Conarva) or different gut segments (e.g. staining of goblet cells with CMA II, LEA and MPA). Hence, terminal carbohydrate residues of glycoconjugates on the intestinal epithelium vary depending on age and organ site. As glycoconjugates can act as attachment sites for microorganisms, these differences in the distribution of sugar residues may be one explanation for the site-specificity of certain pathogens. Furthermore, the binding of lectins to the follicle-associated epithelium (FAE) of the BF differs from that to the FAE of the intestine again stressing the site specificity of lectin binding. Thus, up to now no universal M-cell marker along the chicken intestine exists.  相似文献

BackgroundThe olfactory mucosa (OM) is crucial for odorant perception in the main olfactory system. The terminal carbohydrates of glycoconjugates influence chemoreception in the olfactory epithelium (OE).ObjectivesThe histological characteristics and glycoconjugate composition of the OM of Korean native cattle (Hanwoo, Bos taurus coreae) were examined to characterize their morphology and possible functions during postnatal development.MethodsThe OM of neonate and adult Korean native cattle was evaluated using histological, immunohistochemical, and lectin histochemical methods.ResultsHistologically, the OM in both neonates and adults consists of the olfactory epithelium and the lamina propria. Additionally, using periodic acid Schiff and Alcian blue (pH 2.5), the mucus specificity of the Bowman’s gland duct and acini in the lamina propria was determined. Immunohistochemistry demonstrated that mature and immature olfactory sensory neurons of OEs express the olfactory marker protein and growth associated protein-43, respectively. Lectin histochemistry indicated that numerous glycoconjugates, including as N-acetylglucosamine, mannose, galactose, N-acetylgalactosamine, complex type N-glycan, and fucose groups, were expressed at varied levels in the different cell types in the OMs of neonates and adults at varying levels. According to our observations, the cattle possessed a well-developed olfactory system, and the expression patterns of glycoconjugates in neonatal and adult OMs varied considerably.ConclusionsThis is the first study to describe the morphological assessment of the OM of Korean native cattle with a focus on lectin histochemistry. The findings suggest that glycoconjugates may play a role in olfactory chemoreception, and that their labeling properties may be closely related to OM development and maturity.  相似文献

The proportions of fibres of the collagenous and elastic systems were measured in the aortas of three normal heifers and in nine heifers given the calcinogenic plant Solanum glaucophyllum for 15, 30 or 60 days. There were decreases in the amount of collagen relative to reticulin, and in the proportion of elastic fibres which were related to the period of dosing. These changes may have an adverse influence on the animals' cardiovascular function.  相似文献

Dorsal lacrimal glands, superior glands of the third eyelid and Harderian glands (deep gland of the third eyelid) from 19 bison and 18 cattle free of apparent ocular disease were examined to compare the normal anatomical properties of these glands. All glands were characterized and measured (length and width). The gross anatomy of the dorsal lacrimal glands was similar, with the exception of a bipartite gland in cattle. The bison's superior gland of the third eyelid and Harderian gland was longer as compared with cattle. A subset of the bison and cattle samples (five bison and five cattle) was sectioned for histological and histochemical analysis. The histology of the dorsal lacrimal and superior gland of the third eyelid revealed tubuloalveolar cells with basophilic vacuolated cytoplasm in bison and eosinophilic granular cytoplasm in cattle. The Harderian glands consisted of a tubuloalveolar anterior part combined with large lumens acini lined with cuboidal epithelium in the posterior part; the posterior part of the bison Harderian gland was more predominant than in cattle samples. Mucosubstance histochemistry revealed acidic and neutral glycoproteins with similar staining patterns in all glands of both species.  相似文献

Ten brains of cows were, immediately after slaughter, embedded in celloidin, sliced, and stained to visualise neurocytes and nerve fibres, with the view to elucidating the structures of bovine diencephalon. Macroscopically, the diencephalon of cow is characterised by a comparatively strong epithalamic part, large laterally supported Corpora geniculata lateralis, pronounced ventricular bulgings, almost complete fusion of thalami with Massa intermedia, and very large pituitary gland. The five compartments of bovine interbrain are strongly developed and clearly delimitated by fibrous strings and platelets. By due consideration of the major fibre structures and cyto-architectonic characteristics, 62 nuclei may be differentiated in either half of the diencephalon, with 28 being situated in the thalamus and 23 in the hypothalamus. Two nuclear regions each were recordable from the epithalamus, metathalamus, and subthalamus.  相似文献
