1. The molecular weight of delta‐globulin was estimated chromato‐graphically to be about 10,700 daltons.

2. Ultracentrifuge experiments at 2.7 × 105 g gave values of s 20, W = (1.49 + 0.16C) × 10?13 s for the sedimentation coefficient and D20 w = (1.12 + 0.19c) × 10?6 cm2 s?1 for the diffusion coefficient, c being the protein concentration (g/100 ml).

3. According to the Svedberg equation, these imply a molecular weight of 12,470 daltons, assuming [v_bar] = 0.74 ml/g.

In an experiment, the possible influence of tannic acid (TA) and polyethylene glycol (PEG) on the absorption capacity of intestine for d-xylose and β-carotene in broiler chicken was investigated. Four groups of nine 28-day-old broiler cockerels received d-xylose (500 mg) and β-carotene (52 μg) solutions (Group 1 to 4) with TA (1 g, Group 2 to 4) and PEG (500 mg Group 3 and 1 g Group 4), orally. One blood sample prior to, and four others after the administration of test materials, were collected from wing vein on 40 min basis, for 160 min and the concentration of plasma d-xylose was determined. The concentration of β-carotene was also measured in plasma of blood samples taken prior to and 160 min post-administration of the test materials. Plasma d-xylose concentration of all groups showed quadratic correlations with time (p < 0.001, r(2) = 0.84, 0.60, 0.70 and 0.74 for Group 1 to 4, respectively). Administration of TA reduced the plasma concentration of d-xylose in Group 2. However, feeding PEG after TA raised the concentration of d-xylose in Group 4 to the level that there was no difference in that variable between this group and Group 1. Although the plasma concentration of β-carotene was increased in 160 min post-ingestion of the test material, no difference was found in that variable among the experimental groups. In conclusion, TA and its interaction with PEG have impacts on the absorption capacity of intestine for d-xylose and highly likely other simple sugars, but TA or PEG have no influence on the absorption of β-carotene and most probably other fat soluble vitamins.  相似文献

Summary

The acid‐base status of 58 calves (all in normal anterior presentation) was determined from venous blood samples before the onset of traction and immediately after vaginal delivery.

Calves were assigned to one of three groups according to their blood pH value: group 1 ‐ normal, pH >7.2; group 2 ‐ acidotic, pH 7.2 to 7.0; group 3 ‐ severely acidotic, pH < 7.0. Before the onset of traction (i.e. during the 30 minutes following the appearance and rupture of the membranes), 43 calves (74.1%) had normal acid‐base values, 14 (24.1%) had slight acidosis, and only 1 (1.7%) had severe acidosis. At birth the three groups of calves showed the following distribution: 23 (39.7%) were normal, 29 (50%) had slight acidosis, and 6 (10.3%) had severe acidosis.

Seven calves (12. 1%) died during birth or within 48 hours after birth; 2 were normal shortly before birth, 4 were acidotic shortly before birth, and 1 was severely acidotic even before the onset of traction. Traction was significantly longer for cows that delivered severely acidotic calves compared to cows in the other two groups.  相似文献

1. Three experiments performed to study the effects of amino acid imbalances on the growth of genetically lean (LL) and fat (FL) male chickens from 28 to 42 d of age were described by Leclercq et al. (1994). The plasma amino acid concentrations of birds on selected treatments from that paper are reported here. In experiment 1, three dietary concentrations of digestible lysine were compared (4.75, 6.75 and 7.75 g/kg). In experiment 2, two dietary concentrations of digestible arginine were compared (6.53 and 10.00 g/kg). In experiment 3, three diets were compared: a high‐protein diet (189 g CP/kg), a low‐protein diet containing added essential amino acids (144 g CP/kg), and this low‐protein diet supplemented with 40 g/kg of non‐essential amino acids (NEAA; glutamic and aspartic acids).

2. The present results are compared with two earlier reports on the same genotypes. The LL consistently had lower plasma concentrations of me‐thionine, cystine, phenlyalanine, isoleucine and valine, and higher concentrations of histidine, than the FL chickens. In 4 of 5 experiments, LL leucine concentrations were lower, and glutamic acid, tyrosine, glutamine and alanine were higher, than in the FL. The other amino acids measured; arginine, lysine, aspartic acid, glycine and serine, exhibited variable responses among the experiments.

3. When the limiting essential amino acids, lysine and arginine, were added to a deficient diet, the plasma concentration of the supplemented amino acid increased while the others remained constant or decreased.

4. When glutamic and aspartic acids were added to the low protein diet, plasma amino acid responses were similar to those of adding a limiting amino acid to a deficient diet, except that alanine exhibited a dramatic increase.

5. Although there were genotype by diet interactions for several amino acids, the interactions were caused by differences in the degree of the responses, not in their direction.

6. These results suggest that the FL and LL genotypes do not utilise various amino acids with the same efficiency and, as a consequence, the ideal profile of dietary amino acids should not be the same for both lines. The results support the hypothesis that selection for fatness and leanness changed the amino acid requirements independently of the: effects of food intake.  相似文献

  1. The present study was conducted to determine the effects of α-lipoic acid supplementation on post-mortem changes in the fatty acid profile and concentrations of nucleotide-related substances, especially those of a taste-active compound, inosine 5?-monophosphate, in chicken meat.

  2. Mixed-sex broiler chicks aged 14 d were divided into three groups of 16 birds each and were fed on diets supplemented with α-lipoic acid at levels of 0, 100 or 200 mg/kg for 4 weeks. Blood and breast muscle samples were taken at 42 d of age under the fed condition and then after fasting for 18 h. The breast muscle obtained from fasted chickens was subsequently refrigerated at 2°C for one and 3 d.

  3. α-Lipoic acid supplementation did not affect any plasma metabolite concentration independently of feeding condition, while a slight increase in plasma glucose concentration was shown with both administration levels of α-lipoic acid. In early post-mortem breast muscle under the fed condition, α-lipoic acid had no effect on concentrations of fatty acids or nucleotides of ATP, ADP, and AMP.

  4. In post-mortem breast tissues obtained from fasted chickens, total fatty acid concentrations were markedly increased by α-lipoic acid feeding at 200 mg/kg irrespective of length of refrigeration. This effect was dependent on stearic acid, oleic acid, linoleic acid and linolenic acid. However, among fatty acids, the only predominantly increased unsaturated fatty acid was oleic acid.

  5. Dietary supplementation with α-lipoic acid at 200 mg/kg increased the inosine 5?-monophosphate concentration in breast meat and, in contrast, reduced the subsequent catabolites, inosine and xanthine, regardless of the length of refrigeration.

  6. Therefore, the present study suggests that α-lipoic acid administration altered the fatty acid profile and improved meat quality by increasing taste-active substances in the post-mortem meat obtained from fasted chickens.

1. The effects of graded amounts of dietary ascorbic acid with or without 10 μg/kg dietary 1,25‐dihydroxycholecalciferol on performance, blood and bone variables were measured in broiler chicks.

2. 1,25‐Dihydroxycholecalciferol prevented the tibial dyschondroplasia and rickets caused by feeding a low calcium diet. Gain:food was decreased, but body weight was not affected by feeding 10 μg/kg 1,25‐dihydroxycholecalciferol. Phytate phosphorus retention was increased by dietary 1,25‐dihydroxycholecalciferol.

3. Dietary ascorbic acid did not influence the incidence of tibial dyschondroplasia, but did reduce the incidence of rickets at a dietary concentration of 250 mg/kg. Gakv.food was increased when 250 or 500 mg ascorbic acid/kg diet were added along with 10 μg/kg 1,25‐dihydroxycholecalciferol in one of two experiments.

4. Alkaline phosphatase activity and plasma 1,25‐dihydroxycholecalciferol were not affected by the dietary treatments. Plasma 25‐hydroxycholecalcif‐erol concentration was decreased by dietary 1,25‐dihydroxycholecalciferol.

5. Ascorbic acid had no synergistic effects with 1,25‐dihydroxycholecalciferol on the blood and bone variables investigated in broiler chickens at the dietary concentrations of 1,25‐dihydroxycholecalciferol used in this work.  相似文献

Pork is traditionally low in docosahexanoic acid(DHA, C22:6n-3) and deficient in omega-3 fats for a balanced human diet. DHA as triglycerides was commercially prepared from the microalgae Schizochytrium and injected into fresh pork loins. Treatments of a mixed brine control(CON), 3.1% sunflower oil in mixed brine(SF) and a 3.1% DHA oil in mixed brine(DHA) were injected into pork loins at 10 mL/100 g and grilled at 205°C. After cooking, the CON and SF pork loins contained 0.03 to 0.05 mg DHA/g of pork and the DHA injected loins contained approximately1.46 mg DHA/g. This also changed the fatty acid profile of omega-6: omega-3 from, 5 to 1 in the CON pork, to a ratio of 1.7 to 1 in DHA pork. The appearance, odor, oxidation rates and sensory taste, as judged by a trained panel,determined the DHA injected meat to be, ‘slightly desirable' and gave lower ‘off flavour' scores, relative to the CON and SF injected pork. Pork can be fortified with DHA oil to 146 mg/100 g serving, which would meet half the recommended daily omega 3 fatty acid requirements for adult humans and would be desirable in taste.  相似文献

The incorporation of oral rehydration solutions (ORS) into ‘milk meals’ is potentially an effective, time-saving method of treating diarrhoeic calves. Although milk-based ORS are effective in improving the hydration and acid–base status of healthy calves, this effect remains to be confirmed in dehydrated/diarrhoeic animals. In this study, six experimentally-dehydrated calves were fed with either milk replacer (MR) or an ORS prepared in either water (WORS) or MR (MORS). In one experiment, calves were not treated and blood samples were taken before and after feeding. Parameters of hydration status were determined and blood gas analysis was performed.Plasma volumes increased significantly following the intake of a ‘fluid meal’ whereas they remained constant in the absence of treatment. The rate of plasma volume expansion was reduced by the feeding of MR relative to WORS or MORS. In dehydrated calves, the expansion of plasma volume was more pronounced following the intake of WORS but the increase was less and plasma osmolality increased significantly following the ingestion of MORS. The acid–base status of animals improved as a result of fluid absorption, but this effect was less obvious as the experimental protocol resulted in severe dehydration and moderate acidosis. Feeding hypertonic MORS raised the plasma osmolality in dehydrated calves, and may increase the risk of hypernatraemia in diarrhoeic calves, which should therefore have ad libitum access to water when undergoing treatment with hypertonic ORS. Further research is planned to assess whether feeding ORS reconstituted in milk or MR combined with ad libitum access to water offers a practical treatment for diarrhoeic calves.  相似文献

An experiment was conducted to evaluate the effects of laying hen diets supplemented with δ-aminolevulinic acid (ALA) and vitamin C (VC) on productive performance, iron status and egg quality. A total of 252 Hy-line brown commercial laying hens were fed two levels of VC (0 and 500 mg/kg) and three levels of ALA (0, 5 and 10 mg/kg) in a 2 × 3 factorial arrangement from 57 to 63 weeks of age. Each treatment contained seven replicates with six hens in each replicate. Supplementation of the diet with both ALA and VC resulted in a significant increase in egg production during 4-6 weeks (p < 0.05). The haematocrit (HCT), total iron-binding capacity (TIBC), white blood cell (WBC), total protein and albumin concentrations were not affected by the dietary treatments. However, ALA and interactive effect of ALA and VC were observed to increase the RBC, haemoglobin and serum iron concentrations (p < 0.05). In addition, inclusion of both ALA and VC increased blood lymphocyte percentage in relation to hens not supplemented with ALA (p < 0.05). Moreover, single effect of ALA or VC improved (p < 0.05) the yolk colour unit and albumin height from 3 to 6 weeks. The egg shell colour was increased (p < 0.05) by the effect of ALA. The addition of VC also led to an increase (p < 0.05) of the Haugh unit at the end of the experiment. However, neither ALA, nor VC exerted an effect on egg weight, egg shell breaking strength or egg shell thickness. The results demonstrate that dietary ALA and VC supplementation can improve the iron status and egg quality of laying hens.  相似文献

1. The study aimed to establish the optimum time after ingestion and optimum sampling site for the development of an ileal amino acid digestibility assay for broiler chickens.

2. To establish the optimal sampling time, 4‐week‐old broiler chickens were given one of 6 protein sources (meat‐and‐bone, soyabean, cottonseed, fish, maize and wheat meals) as the sole source of protein in a test diet. The diets contained chromic oxide as an indigestible marker. The birds were starved for 24 h, fed and subsequently killed for sampling of ileal digesta (terminal 15 cm) at 2, 3, 4, 5 and 6 h after the start of feeding.

3. For the soyabean, fish, wheat and maize meal diets, sampling time had no significant effect on apparent ileal nitrogen digestibility, whereas for the meat‐and‐bone and cottonseed meal diets there was a significant quadratic effect of sampling time. The amount of digesta collected was maximised and the mean apparent ileal nitrogen digestibility had the lowest variation around the 4 h sampling time.

4. To establish the optimum sampling site, 4‐week‐old chickens were given either a meat‐and‐bone, a soyabean or a wheat bran meal‐based diet. The birds were killed 4 h after the start of feeding, and digesta were sampled from 0.10, 0.15, 0.20 or 0.25 cm of terminal ileum.

5. There was no significant effect of sampling site on the apparent ileal digestibility of dietary nitrogen. The terminal 15 cm of ileum was considered a preferred site for sampling ileal digesta from broiler chickens.  相似文献

Summary

The experiments demonstrate that the acid‐base status of porcine blood does not change significantly during a six hours’ storage period at 0° C. They further show that the sampling of blood via a vena cava puncture in comparison with sampling via an indwelling ear vein catheter is associated with severe stress that affects the acid‐base statuts of the blood.  相似文献

1. Diets high in total lipids, saturated fatty acids, trans fatty acids, and having high ω-6:ω-3 fatty acid ratios, have been shown to be related to increased instances of coronary heart disease, while diets high in ω-3 fatty acids have been shown to decrease the risk.

2. Feeding ω-3 fatty acid diets to laying hens has been shown to improve the quality of eggs produced in terms of saturation and ω-3 content.

3. A study was undertaken to determine if the ω-3 fatty acid source, when fed to hens, influences the amount transferred to eggs.

4. Flaxseed and flaxseed oil, along with chia seed and chia seed oil, were the two main sources of ω-3 fatty acid examined during the 84?d trial.

5. All α-linolenic enriched treatments yielded significantly higher ω-3 fatty acid contents per g of yolk and per yolk, than the non-α-linolenic enriched diets. Chia oil and chia seed yielded 54·5 and 63·5% more mg of ω-3 fatty acid per g of yolk for the 56?d test period, and 13·4 and 66·2% more for the 84?d test period, than flaxseed oil and flaxseed, respectively.

6. The differences in omega-3 content were significant, except for the chia oil compared with the flax oil, at the end of the trial.

7. This trial has shown that differences in conversion exist among ω-3 fatty acid sources, at least when fed to hens, and indicates that chia may hold a significant potential as a source of ω-3 fatty acid for enriching foods, thereby making these foods a healthier choice for consumers.  相似文献

The effects of freeze-thaw conditions on the fermentation quality and aerobic stability of oat silage were investigated. Four strains of freeze-thaw-resistant lactic acid bacteria(LAB)with rapid growth and efficient acid production were screened from 437 strains,which had been isolated from the Qinghai-Tibetan Plateau. Based on physiological and biochemical characteristics and 16S rRNA gene sequence alignment,the four strains were identified as Lactobacillus plantarum 157(LP157),L. plantarum 160(LP160),Lactobacillus brevis 248(LB248),and Lactobacillus pentosus 260(LPE260). In order to investigate their effects on the fermentation quality,nutrient composition,microbial counts and aerobic stability of oat silage under freeze-thaw conditions,seven treatments were tested as follows:1)LP157;2)LP160;3)LB248;4)LPE260;5)LP160+Lactobacillus rhamnosus 753 mixture (MIX);6)A commercial bacterial silage additive(COM);7)The same amount of sterilized distilled water(CK). All treatments were ensiled for 60 days at a constant temperature of 20 ℃ and under a 20 ℃/-5 ℃ regime alternating every 12 h(freeze-thaw). LP inoculation improved silage quality:The pH and dry matter loss in silages inoculated with LP157,LP160,LB248 and LPE260 were significantly lower than CK(P<0. 05)and lactic acid and acetic acid contents were significantly higher than CK(P<0. 05),and the growth of spoilage micro-organisms such as coliform bacteria,yeast and mold were inhibited,both with fermentation at constant 20 ℃ and under freeze-thaw conditions. After aerobic exposure for 5 days,the pH and NH3-N of silages treated with LP157,LP160,LB248 and LPE260 were significantly(P<0. 05) lower than CK,MIX and COM. The aerobic stability of oat silage was improved,especially when inoculated with LP160(70 h vs. CK 11 h). 1. Growth and food conversion efficiency of broilers were reduced at low dietary choline concentrations. These effects were accentuated by low dietary total sulphur amino acids (TSAA).

2. Increased mortality, perosis and other pathological changes involving a number of tissues were observed in birds fed on a low‐choline diet.

3. Methionine replaced part of the dietary requirement for choline at low dietary choline concentrations.

4. High dietary concentrations of choline (> 1 750 mg/kg) increased the requirement for dietary TSAA.

The accuracy of the Precision Xceed® hand-held meter as an on-site method for measuring blood β-hydroxybutyric acid (BHBA) and glucose concentrations, for the diagnosis of pregnancy toxemia and ketosis in dry and lactating dairy sheep, was assessed. Five to eight hours after the start of the morning feed, blood was collected once from 193 clinically healthy sheep (143 dry and 50 lactating). BHBA and glucose analyses were performed with serum in the laboratory, and with whole blood with the Precision Xceed®. Overall, BHBA and glucose determinations by the two methods were not statistically different (P > 0.05). Strongly significant positive correlations were found for glucose and BHBA concentrations between the Precision Xceed® and laboratory results (r = 0.76, n = 150, P < 0.01 and r = 0.99, n = 193, P < 0.01, respectively). The Precision Xceed® was highly sensitive (98.6%) and specific (98.2%), and had excellent test agreement for the detection of pregnancy toxemia and ketosis.  相似文献

1. The influence of age, sex and injection of Escherichia coli lipopolysaccharide (LPS) on the plasma concentration of alpha‐1 acid glycoprotein (AGP) was determined in broilers using the single radial immunodiffusion method.

2. Plasma AGP concentration increased in the 3 d after hatching, and then stabilised at 240 ± 33 μg/ml up to 14 d of age.

3. No sex‐related differences in plasma AGP concentration were observed up to 6 weeks of age.

4. A single injection of 900 μg LPS per chick resulted in a 5‐fold increase in AGP concentration compared with that in saline‐injected chicks. Multiple injections of LPS (200 μg/chick every 2d for 14d) caused only a 50% increase in AGP concentration.  相似文献

Antioxidant vitamins are commonly used as colorant, preventive, sweetener, nutritive and antioxidant materials in food industry. Thus, many studies have been conducted in recent years. We have performed the present study to understand the effects of α-lipoic acid (ALA), vitamin C and E, added into rations of Japanese quails, on chronic heat stress. This study was carried out in five groups as: control, stress, ALA, vitamin E and vitamin C groups. Heat stress is applied to all groups except the control group. Superficial pectoral muscles tissues were used for biochemical determinations. During the research, it is found that heat stress exerted undesired effects such as increasing lipid peroxidation (LPO) level indicating oxidative stress in Japanese quails. In addition, superoxide dismutase (SOD) and catalase (CAT) enzyme activities and glutathione (GSH) amounts were studied to understand the behaviour of defence mechanism. While stress increased LPO ratio, it was determined that all antioxidant added into the ration decreases LPO significantly (p < 0.05). In addition, it was determined that all of antioxidants added into ration effectively restored SOD activity and LPO ratio which heat stress affected negatively. Interestingly, vitamin C did not adjust GSH ratio in contrast to vitamin E and ALA, where vitamin E and C did not perform any positive effect on heat stress decreased CAT activity. Finally, it can be thought that antioxidant vitamins relatively ameliorated these undesired affects caused by stress factors given.  相似文献
