β-Galactosidases (β-Gal) of lactic acid bacteria produce oligosaccharides from lactose when suitable acceptor carbohydrates are present. This study aimed to elucidate the structure of oligosaccharides formed by galactosylation of N-acetylglucosamine (GlcNAc) and fucose. Crude cellular extract of Lactobacillus bulgaricus and LacLM of Lactobacillus plantarum were used as sources of β-Gal activity. Disaccharides obtained by galactosylation of GlcNAc were identified as Gal-β-(1→4)-GlcNAc or Gal-β-(1→6)-GlcNAc by liquid chromatography-tandem mass spectrometry (LC-MS/MS) and comparison with external standards. Trisaccharides were identified as Gal-β-(1→6)-Gal-β-(1→[4 or 6])-GlcNAc by LC-MS, analysis of the MS/MS spectra of selected in-source fragment ions, and their relative retention times. LC-MS analysis revealed the presence of five galactosylated fucosides, but their linkage type could not be identified, partly due to the lack of reference compounds. β-Gal of lactic acid bacteria may serve as suitable tools for the chemoenzymatic synthesis of therapeutic oligosaccharides. 相似文献

beta-Glucan extracted from oats was subjected to reductive amination, producing a cationic beta-glucan derivative of which physiological properties were characterized. The degree of substitution was obtained from elemental analysis, which was 0.48. In addition, the distribution of amino groups in the beta-glucan derivative was investigated by FT-IR analysis. In vitro bile acid binding capacity of the aminated beta-glucan was examined, showing significantly higher bile acid binding activity than native beta-glucan. Moreover, the beta-glucan derivative showed pronounced antimicrobial effects against Escherichia coli and Bacillus subtilis, and ACE (angiotensin-converting enzyme) inhibition activities which were dependent on its concentration. Furthermore, bronchoalveolar lavage (BAL) experiments demonstrated that the beta-glucan derivative stimulated the synthesis of nitric oxide. The improved functionalities of the derivative could be explained by its polycationic characteristics. 相似文献

The structural, physicochemical, and biological properties of sulfated oat beta-glucan were characterized. The degree of substitution of the sulfated oat-beta glucan was obtained by elemental analysis, which was 0.68. Compared to native oat beta-glucan, the FT-IR spectra of the derivative showed two new absorption bands at 1250 and 810 cm(-)(1), which would be attributed to (S=O) and (C-O-S) groups, respectively. The molecular weight of the sulfated beta-glucan was determined to be 68 kDa and its viscosity decreased by almost 2 orders of magnitude while its solubility increased by more than 100% compared to that of the native beta-glucan. In addition, the sulfation caused the reduction of in vitro bile acid binding capacity of oat beta-glucan due to the new anionic character and decreased molecular weight. The sulfated derivative exhibited, however, anticoagulant activity which showed a concentration-dependent increase. 相似文献

Eight apple pomace liquefaction juices were produced to characterize soluble cell wall material released by the action of pectolytic and cellulolytic enzyme preparations. Very high colloid values from 9.7 to 19.6 g/L were recovered from the juices by ethanol precipitation. The crude polysaccharides consisted mainly of galacturonic acid (49-64 mol %), arabinose (14-23 mol %), galactose (6-15 mol %), and minor amounts of rhamnose, xylose, and glucose. Separation of the polysaccharides by anion-exchange chromatography yielded one neutral, one slightly acidic, and one acidic polymer accounting for 60% of total colloids. Preparative size exclusion chromatography of the acidic fractions resulted in four polymers of different molecular weights and different sugar compositions. Among them, high molecular weight arabinans and rhamnogalacturonans as well as oligomeric fractions consisting of only galacturonic acid could be found. Linkage studies were performed on neutral fractions from anion-exchange chromatography and size exclusion chromatography. They revealed highly branched arabinans, xyloglucans, and mainly type I arabinogalactans. 相似文献

and the relationship between carbon and microorganisms to soil carbon) were shown by arable soils. Among post-agrogenic soils,
the soils of neglected fields, which formed after the termination of plowing, are more biogenic than soils of perennial dry
meadows; this is due to the combination of a high level of fertilizer elements and the annual receipt of a soft vegetative
substrate. The carbon dioxide emissions of a soil as a multifactor and strongly varying indicator cannot be the only criterion

The establishment of a symbiotic interaction involves a signal exchange between the host legume (flavonoids) and the nitrogen-fixing rhizobia (nodulation factors (NFs)). Likewise, abiotic stress conditions, such as salinity and drought, strongly reduce the nodulation process, possibly affecting also the signal exchange. In this work we characterized the structure and biological activity of NFs produced by Bradyrhizobium japonicum USDA 138 under control, salt, and osmotic stress conditions. This strain is the most widely used in Argentine soybean culture; under control conditions, it produces a mixture of four types of NFs (V(C16:0,MeFuc), V(C18:1,MeFuc), IV(C18:1), and V(C18:1,Ac,MeFuc)). Interestingly, under stress conditions, this strain produces new types of NFs, one common for both stress conditions (V(C16:1,MeFuc)) and another one only present under salt stress (IV(C18:1,MeFuc)). All mixtures of NFs, extracted from control, salt, and osmotic stress conditions, showed biological activity in soybean plants, such as root hair deformation, and the radical application of purified NFs induced systemic differences in dry matter accumulation. The inoculation of soybean with genistein-induced bacteria cultured under both control and stress conditions had a positive effect on the number of nodules formed and in some cases on dry matter accumulation. Bread spoilage is mainly due to the growth of filamentous fungi, and metabolites produced during sourdough fermentation by lactobacilli can inhibit fungal growth. One of these metabolites is phenyllactic acid (PLA), which is a catabolite from phenylalanine. In this work, the influence of peptide supply and cosubstrates was determined on PLA formation from phenylalanine by Lactobacillus plantarum TMW1.468 and Lactobacillus sanfranciscensis DSM20451(T). Transport of single amino acids is not efficient in lactobacilli, and only 1% of the offered phenylalanine was converted to PLA. PLA yields were increased 2-4-fold when peptides instead of single amino acids were used as a substrate. The accumulation of phenylalanine after peptide addition indicated that, after transport, transamination was the second limiting factor. In L. plantarum TMW1.468, PLA yields were increased from 5 to >30% upon the addition of alpha-ketoglutarate. In L. sanfranciscensis DSM20451, a combination of both citric acid and alpha-ketoglutarate increased PLA formation. The combined effect of citric acid and alpha-ketoglutarate can be attributed to changes in the NAD/NADH ratio. 相似文献

It was previously reported that (-)-epigallocatechin-3-O-gallate (EGCG) suppresses the expression of the high-affinity IgE receptor FcepsilonRI in human basophilic cells and that this suppressive effect is associated with EGCG binding to the cell surface. This study examined the effects of five methylated derivatives of EGCG, (-)-epigallocatechin-3-O-(3-O-methyl)gallate (EGCG 3' 'Me), (-)-epigallocatechin-3-O-(4-O-methyl)gallate (EGCG 4' 'Me), (-)-4'-O-methyl-epigallocatechin-3-O-gallate (EGCG 4'Me), (-)-epigallocatechin-3-O-(3,4-O-methyl)gallate (EGCG 3' '4' 'diMe), and (-)-4'-O-methyl-epigallocatechin-3-O-(4-O-methyl)gallate (EGCG 4'4' 'diMe) on FcepsilonRI expression and ERK1/2 phosphorylation, and each of their cell surface binding activities was measured. Of these five methylated derivatives, three that are methylated at the 3' '- and/or 4' '-position, EGCG 3' 'Me, EGCG 4' 'Me, and EGCG 3' '4' 'diMe, suppressed FcepsilonRI expression and ERK1/2 phosphorylation, although the suppressive effects were lower than that of EGCG. EGCG 4'Me and EGCG 4'4' 'diMe, both of which are methylated at the 4'-position, did not demonstrate a suppressive effect. Furthermore, it was found that EGCG 3' 'Me, EGCG 4' 'Me, EGCG 3' '4' 'diMe, and EGCG 4'Me, which are methylated at the 3' '- and/or 4' '-positions or the 4'-position, could bind to the cell surface even though their binding activities were lower than that of EGCG. Only EGCG 4'4' 'diMe, which is methylated at both the 4'- and 4' '-positions, could not bind. These results suggest that the trihydroxyl structure of the B ring is essential for EGCG to exert the suppressive effects and that the hydroxyl groups on both the 4'-position in the B ring and the 4' '-position in the gallate are crucial for the cell surface binding activity of EGCG. 相似文献

Concerns that raisins may be contaminated by fumonisins stem from the persistent occurrence of Aspergillus niger spores on raisins and the recent discovery of fumonisin production by A. niger on grapes, which leads to the widespread occurrence of fumonisin B(2) in wine. This study presents an LC-MS/MS survey of fumonisins in retail raisins. In 10 of 21 brands collected in Denmark, Germany, and The Netherlands, fumonisins B(2) and B(4) were detected at levels up to 13 and 1.3 μg/kg, respectively. Only fumonisin B(2) has been detected in wine, so the presence of fumonisin B(4) in raisins suggests that the fumonisins are produced mainly during the drying process concomitant with the decreasing water activity. Analysis of multiple packages from one manufacturer showed a 3-fold package-to-package variation, suggesting that a few raisins per package are contaminated. 相似文献

It was previously reported that cell cultures from Lactobacillus plantarum CECT 748 (T) were able to decarboxylate phenolic acids, such as p-coumaric, m-coumaric, caffeic, ferulic, gallic, and protocatechuic acid. The p-coumaric acid decarboxylase (PDC) from this strain has been overexpressed and purified. This PDC differs at its C-terminal end when compared to the previously reported PDC from L. plantarum LPCHL2. Because the C-terminal region of PDC is involved in enzymatic activity, especially in substrate activity, it was decided to biochemically characterize the PDC from L. plantarum CECT 748 (T). Contrarily to L. plantarum LPCHL2 PDC, the recombinant PDC from L. plantarum CECT 748 (T) is a heat-labile enzyme, showing optimal activity at 22 degrees C. This PDC is able to decarboxylate exclusively the hydroxycinnamic acids p-coumaric, caffeic, and ferulic acids. Kinetic analysis showed that the enzyme has a 14-fold higher K(M) value for p-coumaric and caffeic acids than for ferulic acid. PDC catalyzes the formation of the corresponding 4-vinyl derivatives (vinylphenol and vinylguaiacol) from p-coumaric and ferulic acids, respectively, which are valuable food additives that have been approved as flavoring agents. The biochemical characteristics showed by L. plantarum PDC should be taken into account for its potential use in the food-processing industry. 相似文献

Soil contaminated by chromium (Cr) is a major concern for sustainable agriculture. Considering this as a basis, the present study was designed to isolate Cr(VI)-reducing and plant growth-promoting bacterial strain from contaminated sampling sources. In this study, Rhizobium strain ND2 was isolated from the root nodules of Phaseolus vulgaris grown in leather industrial effluent contaminated soil. The strain ND2 exhibited strong resistance to different heavy metals and reduced 30 and 50 µg ml?1 concentrations of Cr(VI) completely after 80 and 120 h of incubation, respectively, as well as chromium adsorption and immobilization were confirmed by scanning electron microscopic equipped with energy X-ray spectroscopy. In addition, the strain produced 21.73 and 36.86 µg ml?1 of indole-3-acetic acid at 50 and 100 µg ml?1 of L-tryptophan supplimentations, respectively. Strain ND2 positively affected the exo-polysaccharide, ammonia, protease and catalase production and stimulated root length of various test crops under Cr(VI) stress. Moreover, Rhizobium strain ND2 has the potential to colonize the diverse agricultural crops. Thus, the present findings strongly suggested that the multipotential properties of ND2 could be exploited for bioremediation of contaminated sites with Cr(VI) as well as potential bio fertilizer for enhancing the agricultural productivity. 相似文献

As part of an environmental control program aimed to assess possible future effects of air contaminants from a new destruction plant for hazardous chemical residuals in Central Sweden, a study was undertaken the year before the plant went into operation to collect base-line information on biological activities in the soils of the surrounding forests. Measurements were made of N-mineralization, respiration, phosphatase-, dehydrogenase-, and nitrogenase activity on sieved samples from the forest floor (0 to 3 cm). The results were evaluated in relation to a previous contamination of the area by air pollutants from a plant for oil production. This plant produced oil from the local alun shale between 1942 and 1966. During these years somewhere between 6 × 105 and 1.2 × 106 tonne of S, mainly in the form of SO2, were released into the atmosphere. The results showed that the forest floor was not especially acid when compared to similar sites in locally unpolluted areas. The concentration of Cu, however, was five times higher than expected. No disturbances could be detected as regards N-mineralization, respiration or phosphatase activity. Dehydrogenase- and nitrogenase activity were much more difficult to evaluate. A suppression of dehydrogenase activity was indicated on the study plots closest to the previous plant. In the case of nitrogenase activity no appropriate reference material was available to judge whether disturbances existed. 相似文献

Structural characterization of guaiacyl-rich lignins in flax (Linum usitatissimum) fibers and shives

The structural characteristics of the lignins from flax (Linum usitatissimum) fibers and shives were studied. Significant differences in the content and composition of the lignin from both parts were observed. The lignin contents were 3.8% in the fibers and 29.0% in the shives. Analysis by Py-GC/MS indicated a H:G:S molar ratio of 13:72:15 in the milled wood lignin (MWL) isolated from flax fibers and a molar ratio of 5:87:8 in the MWL isolated from flax shives. In addition, 2D-NMR showed a predominance of β-O-4' aryl ether linkages, followed by β-5' phenylcoumaran and β-β' resinol-type linkages in both MWLs, with a higher content of condensed linkages in flax shives. Thioacidolysis (followed by Raney nickel desulfurization) gave further information on the lignin units involved in the different linkages and confirmed the enrichment of G units. The thioacidolysis dimers released were similar from both lignins, with a predominance of the β-5' followed by β-1' and 5-5' structures. 相似文献

An H(2)O(2)/NaOH/DMSO system has been developed for the formation of three free radicals, and the application of the system was examined with the antioxidants ascorbic acid and tocopherol. Superoxide anion, hydroxyl radical,and methyl radical are simultaneously generated in this system. The scavenging activity of ascorbic acid and tocopherol for these radicals was estimated by 5, 5'-dimethyl-1-pyrroline-N-oxide spin trapping electron spin resonance. Both water-soluble and oil-soluble antioxidants could be evaluated by using this system. Ascorbic acid specifically inhibited the superoxide anion and hydroxyl radical, whereas tocopherol suppressed the methyl radical. 相似文献

Phenyllactic acid (PLA) is a novel antimicrobial compound synthesized by lactic acid bacteria (LAB), and its production from phenylpyruvic acid (PPA) is an effective approach. In this work, a lactate dehydrogenase (LDH), which catalyzes the reduction of PPA to PLA, has been purified to homogeneity from a cell-free extract of Lactobacillus sp. SK007 by precipitation with ammonium sulfate, ion exchange, and gel filtration chromatography. The purified enzyme had a dimeric form with a molecular mass of 78 kDa (size exclusion chromatography) or 39 kDa (SDS-PAGE). The ratio of enzyme activity with PPA to that with pyruvate being almost invariable at every purification step indicated that, in Lactobacillus sp. SK007, LDH is responsible for the conversion of PPA into PLA. HPLC profiles of PPA transformation into PLA by growing cells, cell-free extract, and purified LDH of Lactobacillus sp. SK007 were also investigated. Results showed that the presence of NADH was found to be necessary for the enzymatic production of PLA from PPA. The purified LDH displayed optimal activity for PPA at pH 6.0 and 40 degrees C. The Km values of the enzyme for PPA and pyruvate were 1.69 and 0.32 mM, respectively. Moreover, because other screened LAB strains exhibiting relatively high LDH activity toward PPA produced also considerable amounts of PLA, LDH activity for PPA could be therefore used as a screening marker for PLA-producing LAB. 相似文献

Two lots of human foodstuffs from conventional and organic brand foods were purchased from supermarkets and analyzed for three Fusarium toxins, deoxynivalenol, by GC-ECD, and fumonisins B(1) and B(2) (FB(1)-FB(2)), by LC-MS. The occurrence of deoxynivalenol contamination was higher than 80% in both organic and conventional foods; fumonisin B(1) was found in 20% of organic foods and in 31% of conventional ones and fumonisin B(2) in more than the 32% of the food samples from both the agricultural practices. The highest median concentration of deoxynivalenol occurred in conventional rice-based foodstuffs (207 microg/kg): that of fumonisin B(1) in conventional maize-based foods (345 microg/kg) and that of fumonisin B(2) in organic wheat-based foods (210 microg/kg). 相似文献

Samples of maize grown in various districts of Taiwan were collected and analyzed for the presence of fumonisin B(1) (FB(1)) and fumonisin B(2) (FB(2)) using high-performance liquid chromatography. Forty-nine (44.5%) and 2 (1.8%) of 110 samples were found to contain FB(1) (109-1148 ng/g) and FB(2) (222-255 ng/g), respectively. The frequency of detection and also the maximum FB(1) concentration were found in samples from Penton (2/2, 262 ng/g), followed by Chiayi (18/26, 264 ng/g), Tainan (8/16, 160 ng/g), Hualinen (5/14, 1148 ng/g), Taitung (7/20, 109 ng/g), and Yunlin (9/26, 361 ng/g). Of the 110 samples examined, only 2 samples from Hualinen had been detected containing FB(2). During an analysis of the distribution pattern of FB(1), it became apparent that >79% of tested samples had FB(1) concentrations <100 ng/g, whereas 2.7% (or 3 samples) contained FB(1) >300 ng/g. These results clearly illustrated that domestically produced maize for human consumption is frequently contaminated with FB(1). 相似文献

Bovine beta-lactoglobulin, genetic variant B, has been labeled with 2-(4'-maleimidylanilino)naphthalene-6-sulfonic acid through covalent attachment through the Cys-121 thiol group for the study of stepwise pressure denaturation of this whey protein by fluorescence spectroscopy. The labeling was performed under nondenaturing conditions with a factor of 5 excess of the fluorophore in dimethylformamide/water (1:10) to yield the whey protein highly labeled after chromatographic separation. MALDI-TOF mass spectroscopy confirmed labeling. The emission from the fluorophore, which is sensitive to the microenvironment, has been characterized for the labeled protein (aqueous pH 7.4 solution, 25 degrees C) and has a lambda(em,max) = 410 nm (lambda(ex,max) = 318 nm) with a fluorescence lifetime of 6.1 +/- 0.2 ns. Fluorescence anisotropy increases and fluorescence quantum yield (Phi(f) = 0.103 at 320 nm) decreases with increasing excitation wavelength. For increasing hydrostatic pressure, fluorescence quantum yield showed a minimum at approximately 50 MPa, corresponding to the pre-denatured "pressure-melted" state in which thiol reactivity previously was found to increase prior to reversible protein unfolding. 相似文献
