This report described the histopathological and immunohistochemical features of cutaneous mast cell tumor (MCT) in six hedgehogs. The hedgehogs presented single cutaneous mass with ulcer and crusting. Histologically, the neoplastic lesions were characterized by the proliferation of well-differentiated mast cells (3 cases), and atypical mast cells (3 cases) with one atypical histiocytic morphology. Immunohistochemically, tumor cells were positive for KIT and mast cell tryptase, and were negative for Iba-1. In well-differentiated MCT, all patients were clinically improved and survived more than 365 days after surgical excision, whereas an atypical histiocytic MCT showed aggressive behavior with re-recurrence, and the animal died 115 days after surgery. These findings suggest that, compatible with other animals, well-differentiated MCT has a better prognosis in hedgehogs. 相似文献

Previously, we reported that ovarian hormones affect the immune response against
E. coli isolated from the dogs affected with pyometra. In order to
investigate mechanisms underlying the immune modulation, we examined the effects of
ovarian hormones on the generation of dendritic cells (DCs), the most potent antigen
presenting cell. DCs were differentiated from peripheral blood monocytes (PBMOs) using a
cytokine cocktail. Both estrogen receptor and progesterone receptors were expressed by the
PBMOs and immature DCs. When various ovarian hormones were added to the culture for the DC
differentiation, progesterone significantly decreased the expression of DC maturation
markers, such as CD1a, CD80 and CD86, on mature DCs. Conversely, the addition of estrogen
to the cultures increased the expression of CD86, but not other maturation makers.
Furthermore, DCs differentiated in the presence of progesterone did not stimulate
allogeneic mononuclear cells in PB. Taken together, these results indicate that
progesterone diminishes the maturation of DCs, leading to decreased immune responses

The present study involved the isolation and characterization of canine tumor endothelial
cells (TECs) from 2 malignancies. TECs were isolated using magnetic cell sorting following
FITC labeling with UEA1 lectin, and they were characterized by measuring genetic and
histopathological endothelial markers. Isolated TECs exhibited a cobblestone-like
morphology and expressed both vascular endothelial growth factor receptor 2 (VEGFR2) and
Von Willebrand factor (vWF). Further, both TECs and tumor cells derived from a seminoma
exhibited increased C-X-C chemokine receptor type 7 (CXCR7) expression. However, CXCR7
expression was not detected in TECs and tumor cells derived from a hepatocellular
carcinoma. Understanding TEC specific traits may be important in the development of more
efficacious anti-angiogenic therapies that do not induce adverse effects. 相似文献

We evaluated the postsurgical outcomes of cutaneous or subcutaneous mast cell tumors (MCTs, n=25) in 23 dogs, resected with lateral surgical margins proportional to the widest tumor diameter, including at least one facial plane. The tumor diameter range was 0.3–2.6 cm (median: 0.9 cm), and all were histologically diagnosed as Kiupel’s low-grade MCT. Resection was histologically complete in 20, close (deep margin) in three, and incomplete (deep margin) in two. No dogs developed local recurrence at the site of initial surgery during follow-up of 161–2,219 days (median: 976 days). These results suggest that resection of low-grade, relatively small MCTs with surgical margins proportional to the tumor diameter is a practical procedure with high success rates. 相似文献

Dogs with ovarian papillary adenocarcinoma occasionally present with ascites and/or pleural effusion. These aspirated fluids often contain a large number of cells, and distinction between neoplastic cells and activated mesothelial cells can be difficult. In this study, 7 cases of canine ovarian papillary adenocarcinoma, including 3 with ascites and pleural effusion, were immunohistochemically examined. Ovarian tumor cells were positive for cytokeratin CAM5.2 (CAM5.2), Wilms’ tumor 1 (WT-1) and progesterone receptor (PR) in all 7 cases. A metastatic lesion of the mediastinum in one case was also positive for CAM5.2, WT-1 and PR. Immunohistochemistry on cell blocks obtained from ascites and/or pleural effusion of 2 cases revealed the presence of PR-positive epithelial cells. Whereas, activated mesothelial cells in ascites or pleural effusion collected from dogs without neoplastic lesions were negative for PR. In addition, surface epithelium and subsurface epithelial structures (SES) of normal canine ovaries, that are considered to be the cell of origin for ovarian papillary adenocarcinoma, were also positive for CAM5.2, WT-1 and PR. These results indicate that, together with CAM5.2, WT-1 and PR is a useful diagnostic marker for canine ovarian papillary adenocarcinoma. Expression of PR may be associated with progesterone-dependent nature of canine ovarian papillary adenocarcinoma. 相似文献

The efficiency of spermatogonial stem cell (SSC) isolation and culture from pubertal
donors is currently poor primarily, because of contamination with other testicular cells.
This study aimed to purify SSC-like cells using different extracellular matrixes and a
discontinuous gradient density. In experiment 1, testes (n=6) were analyzed for histology
and SSC-related protein expressions (laminin, SSEA-4, DDX-4 and GFRα-1). After enzymatic
digestion, the cell suspension was plated onto either a laminin- or gelatin-coated dish.
The number of SSC-like cells was determined at 15, 30 and 60 min of culture (experiment
2). Experiment 3 was performed to test whether or not the additional step of Percoll
gradient density centrifugation could really improve purification of SSC-like cells.
Testicular histology revealed complete spermatogenesis with laminin expression essentially
at the basal lamina of the seminiferous tubules. SSEA-4 and GFRα-1 co-localized with DDX-4
in the spermatogonia. The relative percentage of SSC-like cells, as determined by cells
expressing SSEA-4 (59.42 ± 2.18%) and GFRα-1 (42.70 ± 1.28%), revealed that the highest
SSC-like cell purity was obtained with the 15-min laminin-coated dish compared with other
incubation times and gelatin treatment (P<0.05). Percoll treatment
prior to laminin selection (15 min) significantly improved SSC-like cell recovery (91.33 ±
0.14%, P<0.001) and purity (83.82 ± 2.05% for SSEA-4 and 64.39 ± 1.51%
for GFRα-1, P<0.05). These attached cells demonstrated a typical
SSC-like cell morphology and also expressed POU5F1, RET
and ZBTB16 mRNA. In conclusion, double enrichment with Percoll gradient
density centrifugation and laminin plating highly enriched the SSC-like cells

Ischemic acute kidney injury (AKI) is the most key pathological event for accelerating
progression to chronic kidney disease through vascular endothelial injury or dysfunction.
Thus, it is critical to elucidate the molecular mechanism of endothelial protection and
regeneration. Emerging evidence indicates that bone marrow-derived cells (BMCs) contribute
to tissue reconstitution in several types of organs post-injury, but little is known
whether and how BMCs contribute to renal endothelial reconstitution, especially in an
early-stage of AKI. Using a mouse model of ischemic AKI, we provide evidence that
incorporation of BMCs in vascular components (such as endothelial and smooth muscle cells)
becomes evident within four days after renal ischemia and reperfusion, associated with an
increase in stromal cell-derived factor-1 (SDF1) in endothelium and that in
CXCR4/SDF1-receptor in BMCs. Notably, anti-CXCR4 antibody decreased the numbers of
infiltrated BMCs and BMC-derived endothelium-like cells, but not of BMC-derived smooth
muscle cell-like cells. These results suggest that reconstitution of renal endothelium
post-ischemia partially depends on a paracrine loop of SDF1-CXCR4 between resident
endothelium and BMCs. Such a chemokine ligand-receptor system may be attributable for
selecting a cellular lineage (s), required for renal vascular protection, repair and
homeostasis, even in an earlier phase of AKI. 相似文献

We investigated the
concentration of the bacterial endotoxin lipopolysaccharide (LPS) in the blood, ovarian
follicular fluid and uterine fluid of a clinical case of bovine metritis. A 2-year-old
lactating Holstein cow exhibited continuous fever >39.5°C for more than 2 weeks after
normal calving. The cow produced a fetid, watery, red-brown uterine discharge from the
vagina and was diagnosed with metritis. The LPS concentrations in plasma and uterine fluid
were 0.94 and 6.34 endotoxin units (EU)/ml, respectively. One of seven
follicles showed an extremely high level of LPS (12.40 EU/ml) compared to
the other follicles (0.62–0.97 EU/ml). These results might suggest the
presence of high concentration of LPS in follicles in cows with postpartum metritis. 相似文献

This study established the precision and accuracy of a modified latex agglutination turbidimetric immunoassay (LATIA) reagent, and evaluated the ability of the measurement of serum amyloid A (SAA) compared to haptoglobin and α1-acid glycoprotein, which are acute phase proteins (APPs), for diagnosis of clinical mastitis. Concentrations of APPs in cows with mastitis were significantly higher than those in healthy cow. Only the plasma SAA concentration in cows with clinical mastitis (44.90 mg/l; n=15) was significantly higher than that in those with subclinical mastitis (10.70 mg/l; n=16), enabling their diagnosis in contrast to other APPs. Thus, the SAA assay using a LATIA reagent is useful in assessing mastitis severity due to its higher sensitivity and specificity than other APP assays. 相似文献
