Genetic diversity of populations stored ex situ or in situ can be altered due to the management practices they are subjected to. In this paper, we compare populations of two common bean (Phaseolus vulgaris L.) landraces grown on farms with material collected from the same farms and now kept in two ex situ collections (CIAT and REGEN) with the purpose to monitor any changes that have occurred due to ex situ conservation. The diversity was measured using seven bean microsatellite markers. Further phenotypic and developmental traits were registered in a field experiment. Compared with the in situ populations, the ex situ ones had a lower level of gene diversity and we suggest that this is due to the regeneration process. Most of the phenotypic traits did not differ significantly between ex situ and in situ populations, although for yield and 100-seed weight, the CIAT material showed significant lower values. We assume that these populations have gone through an adaptational change. Overall, the conservation ex situ has been successful in maintaining the majority of the adaptations found in the landraces studied, however, the probable loss of genetic diversity that we have observed, suggest that protocols for the regeneration process must be carefully worked out if the majority of alleles are to be preserved for the future. This study also highlights the complementarity of ex situ and in situ conservation methods in order to preserve landrace adaptations and to capture new, useful diversity generated in in situ populations.  相似文献

Diversity in morphological characters of 206 accessions of 14 wild Vigna species from India was assessed. Of these, 12 species belonged to Asian Vigna in the subgenus Ceratotropis and two were V. vexillata and V. pilosa belonging to subgenus Plectotropis and Dolichovigna, respectively. Data on 71 morphological traits, both qualitative and quantitative, were recorded. Data on 45 qualitative and quantitative traits exhibiting higher variation were subjected to multivariate analysis for establishing species relationships and assessing the pattern of intraspecific variation. Of the three easily distinguishable groups in the subgenus Ceratotropis, all the species in mungo-radiata group, except V. khandalensis, viz. V. radiata var. sublobata, V. radiata var. setulosa, V. mungo var. silvestris and V. hainiana showed greater homology in vegetative morphology and growth habit. The species, however, differed in other plant, flower, pod and seed characteristics. Within species variation was higher in V. mungo var. silvestris populations and three distinct clusters could be identified in multivariate analysis. V. umbellata showed more similarity to V. dalzelliana than V. bourneae and V. minima in the angularis-umbellata (azuki bean) group. Within species variations was higher in V. umbellata than other species in the group. In the aconitifolia-trilobata (mothbean), V. trilobata populations, were more diverse than V. aconitifolia. The cultigens of the conspecific wild species were more robust in growth, with large vegetative parts and often of erect growth with three- to five-fold increase in seed size and seed weight, except V. aconitifolia, which has still retained the wild type morphology to a greater extent. More intensive collection, characterisation and conservation of species diversity and intraspecific variations, particularly of the close wild relatives of Asian Vigna with valuable characters such as resistance to biotic/abiotic stresses, more number of pod bearing clusters per plant etc. assumes great priority in crop improvement programmes.  相似文献

Neglected and underutilized species often play a vital role in securing food and livestock feed, income generation and energy needs of rural populations. In spite of their great potential little attention has been given to these species. This increases the possibility of genetic erosion which would further restrict the survival strategies of people in rural areas. Ziziphus spina-christi is a plant species that has edible fruits and a number of other beneficial applications that include the use of leaves as fodder, branches for fencing, wood as fuel, for construction and furniture making, and the utilization of different parts e.g. Fruits, leaves, roots and bark in folk medicine. Moreover, the plant is adapted to dry and hot climates which make it suitable for cultivation in an environment characterized by increasing degradation of land and water resources. Lack of research in Z. spina-christi hinders its successful improvement and promotion. Therefore, studies are needed to fully exploit this species. This article aims at summarizing information on different aspects of Z. spina-christi to stimulate interest in this crop which is of importance in Sudan and other countries of the semi-arid tropics.

Gossypium mustelinum Miers ex Watt is a native from Northeastern Brazil and belongs to the primary gene pool of the cultivated cotton. The unique places where the species was known to occur were visited to plan ex situ and in situ preservation. Two populations at Caicó became extinct, and only eleven individuals were found. At Jaguarari, although one population became extinct, a new one was localized with approximately 500 adult plants, where a fence avoided cattle feeding. The population from Macururé consisted of 28 plants protected from animals by thorns of Bromelia laciniosa Mart. ex Schult. f. Thirteen SSR primer pairs amplified 40 alleles, among which 30% were exclusive of one of the populations. The genetic differences between populations represented 58.3% of the total genetic variation observed. The high genetic distances are likely to be caused by geographical isolation as well as by the small number of individuals which contribute to new generations, and consequential genetic drift.  相似文献

Random amplified polymorphic DNA markers were used to assess relatedness and genetic diversity for 15 lingonberry (Vaccinium vitis-idaea) populations. Seven primers yielding 59 polymorphic bands were used to analyse 13 populations, representing ssp. vitis-idaea from Sweden, Finland, Norway, Estonia and Russia, and two populations, representing ssp. minus from Japan and Canada. A cluster analysis and a multidimensional scaling analysis (MDS) showed similar phenetic patterns among populations, with a pronounced geographic grouping in most cases. Significant correlations were obtained between geographic and genetic distances for the entire set of populations as well as for the 13 ssp. vitis-idaea populations. Mean within-population diversity was 0.206 when estimated with Lynch and Milligan's index, and 0.431 when estimated with Shannon's index, which is in agreement with the mixed mating system reported for lingonberry. Within-population variability accounted for 68.6% of the total variance when all populations were included, and for 78.8% when only populations of ssp. vitis-idaea were analysed. Two different approaches were applied to the selection of plant material for a potential gene bank: (1) a hierarchical sampling strategy based on a cluster analysis and (2) the Maximum genetic diversity program, developed for the establishment of core collections. Random sampling was undertaken for comparisons with the selected data sets. The most diverse and representative set of lingonberry specimens was obtained when samples were selected with the Maximum diversity program.  相似文献

Population clonal diversity and fine-scale genetic structure of Oryza officinalis Wall. ex Watt, an endangered species in China recently experiencing habitat degradation, was estimated using inter simple sequence repeat markers. We analyzed the genetic variations of 440 samples exhaustively collected from nine O. officinalis populations. Relatively rich clonal diversity and poor genetic variation were found in the extant populations. We found that the number of genets, the percentage of polymorphic loci, and gene diversity decreased with population decline, suggesting that habitat degradation will lead to further genetic depletion of O. officinalis populations. A pronounced spatial genetic structure occurs at both the ramet and genet levels in several larger populations, which is the result of clonal growth and concomitant inbreeding. The in situ conserved population PS holds much more genotypes than other populations with the similar population size, which might have more seedling recruitments from the soil seed bank due to habitat disturbance, suggesting a moderate disturbance combined with habitat degradation-avoiding measures are effective for in situ conservation of this species.  相似文献

Perilla (Lamiaceae) contains one tetraploid species, P. frutescens (L.) Britt. and three diploid species, P. citriodora (Makino) Nakai, P. hirtella Nakai and P. setoyensis G. Honda. Tetraploid species have been traditionally cultivated in Asia for their seed oil and for their fragrant leaves that are used as medicine or as a garnish for fish. The center of diversity is still obscure. To conserve the genetic resources, it is important to know the diversity of the tetraploid species. The three diploid species, which are possible parents of the tetraploid species, are all believed to be indigenous to Japan. Their distribution in China and Korea was clarified on the basis of herbarium and field surveys. The tetraploid species is assumed to have originated somewhere around the mid-to downstream area of the Changjiang River. Though Perilla is not cultivated as often in these areas as in northern China, Korea, the Himalayan region, or Myanmar, these areas should also be important for the conservation of genetic resources of tetraploid Perilla crops because of the expected high genetic diversity.  相似文献

Diversity analysis was performed among 39 cultivated lentil (Lens culinaris Medik.) accessions of Central Asia and Caucasian origin using five highly polymorphic microsatellite markers. A total of 33 alleles determined ranging from 3 to 8 per locus. Estimated gene diversity value for 33 loci was 0.66. Genetic similarity indices among 39 accessions ranged from 0.24 to 1.0. Cluster analysis using the unweighted pair group method with arithmetic mean method classified accessions into six major groups at 0.5 similarity coefficient. More than half accessions from Tajikistan formed large cluster. On the other hand, a few accessions from each country showed unique genotypes. Overall, most of the accessions, except ones with closely related origin, were distinguished by the present high quality DNA fingerprinting. This molecular diversity information gives important basis for conservation strategy in gene bank and exotic germplasm introduction in breeding programs in Central Asia and Caucasian countries.  相似文献

Reliable characterization of the variation among wild and cultivated yams in Nigeria is essential for improved management and efficient utilization of yam genetic resources. RAPD and double stringency PCR (DS-PCR) analyses were used to investigate genetic relationships and the extent of redundancy among 30 accessions of two cultivated, and 35 accessions of four wild yam species collected from Nigeria. Twenty-five selected random decamer and two microsatellite primers were used individually and in combination to generate DNA profiles for each accession of the six Dioscorea species. The number of amplified fragments varied from 7 to 18 fragments per primer/primer combination. Different levels of intraspecific genetic diversity were found, with Dioscorea rotundata Poir. being the most variable. Based on identical profiles for the RAPD and DS-PCR primers, 12 duplication groups consisting of a total number of 37 accessions were observed in the present study. An UPGMA analysis grouped the majority of plants according to the species. Cultivated yams belonging to the D. cayenensis–rotundata species complex, which were classified into seven morphotypes/varietal groups, could be clearly separated into two major groups corresponding to D. rotundata Poir. and D. cayenensis Lam. D. cayenensis cultivars exhibited a low level of intraspecific variation and were genetically close to the wild species Dioscorea burkilliana J. Miège. D. rotundata cultivars classified into six varietal groups showed a high degree of DNA polymorphism and were separated into two major groups that appeared most closely related to Dioscorea praehensilis Benth. and Dioscorea liebrechtsiana de Wild. We propose, based on these results, that cultivars classified into D. cayenensis should be considered as a taxon separate from D. rotundata. The implications of intraspecific variability for the ex situ conservation of wild and cultivated yam germplasm in Nigeria are discussed.  相似文献

Amplified fragment length polymorphism markers were used to assess the genetic relatedness between Dioscorea alata and nine other edible Dioscorea. These species include D. abyssinica Hoch., D. bulbifera L., D. cayenensis-rotundata Lamk. et Poir., D. esculenta Burk., D. nummularia Lam., D. pentaphylla L., D. persimilis Prain. et Burk., D. transversa Br. and D. trifida L. Four successive studies were conducted with emphasis on the genetic relationship within D. alata and among species of the Enantiophyllum section from Vanuatu. Study 1 was carried out to select a set of polymorphic primer pairs using 11 combinations and eight species belonging to five distinct sections. The four most polymorphic primer pairs were used in study 2 among six species of the Enantiophyllum section. Study 3 focussed mainly on the genetic relationship among 83 accessions of D. alata, mostly from Vanuatu (78 acc.) but also from Benin, Guadeloupe, New Caledonia and Vietnam. The ploidy level of 53 accessions was determined and results indicated the presence of tetraploid, hexaploid and octoploid cultivars. Study 4, included 35 accessions of D. alata, D. nummularia and D. transversa and was conducted using two primer pairs to verify the taxonomical identity of the cultivars `langlang', `maro' and `netsar' from Vanuatu. The overall results indicated that each accession can be fingerprinted uniquely with AFLP. D. alata is an heterogeneous species which shares a common genetic background with D. nummularia and `langlang', `maro' and `netsar'. UPGMA cluster analysis revealed the existence of three major groups of genotypes within D. alata, each assembling accessions from distant geographical origins and different ploidy levels. The analysis also revealed that `langlang', `maro' and `netsar' clustered together with the cultivar `wael' (D. transversa) from New Caledonia. Results are discussed in the paper.  相似文献

Soil samples were collected from the upper soil horizon within 4 m of black locust ( Robinia pseudo-acacia) and tulip poplar trees ( Liriodendron tulipifera) from the same mixed forest in south-central Pennsylvania. The soil samples were analyzed for organic C levels, pH, NO3 -, NH4 +, catabolic diversity (Shannon diversity index; catabolic H), catabolic evenness (Simpson-Yule index; catabolic E), genetic H, and genetic E. The catabolic H and genetic H of microbes in these soils were found to correlate well with the levels of mineralized N, organic C, and pH. Significant variations in these parameters were found between the soils from near black locust and tulip poplar trees. Conditions in the soil near the black locust trees were more favorable to nitrification as indicated by the elevated pH, organic C, NO3 -, and total mineral N levels, along with lower NH4 + levels. The microbial genetic H and E were greater and the catabolic H and E were lower in the black locust soils than in the tulip poplar soils. This suggests that a more specialized environment exists in the soil near the black locust trees which selects for enhanced nitrification and the use of fewer, but preferred catabolic pathways by a more genetically diverse group of microbes that grow to a greater biomass. Conversely, the soils from near the tulip poplar trees are such that they do not select for some dominant catabolic pathways, rather they allow for the use of a greater variety of catabolic pathways by a less diverse microbial population, which appear to grow to a lower biomass. We believe that the combined application of the microbial genetic and catabolic diversity analyses, microbial biomass estimates, and traditional physico-chemical characteristics in soil studies provides information not easily available that can be useful during assessment of soil processes in different terrestrial habitats.  相似文献

Stylosanthes Sw. is considered one of the economically most important tropical forage legumes. The genus is essentially Neotropical, and Brazil, Venezuela and Mesoamerica are the main centres of species diversification. In view of its importance, an extensive review of Venezuelan Stylosanthes genetic resources was carried out. Firstly, the known natural distributions of all 11 species based on herbarium specimens were georeferenced and mapped and, by comparing them with information of germplasm (=seed) collection site distributions, for each species regions were identified which in terms of collecting and safeguarding genetic resources of Stylosanthes had not been explored yet. Secondly, the actual status of the Stylosanthes collection in Venezuela in terms of collected seed samples and available germplasm is discussed, as is the need for further explorations, including targeting ecologically important locations. Finally, the different Stylosanthes species in Venezuela are grouped according to their perceived agronomic value and potential as forage for animal production systems, and recommendations are given regarding research, collection and preservation needs. Overall, this study compiled baseline information for future considerations of an important genetic resource in Venezuela. The endangerment of the three endemic species, Stylosanthes falconensis Calles et Schultze-Kr., Stylosanthes sericeiceps S.F. Blake and Stylosanthes venezuelensis Calles et Schultze-Kr. should be a major concern and warrants priority considerations for preservation.  相似文献

A genetic fingerprinting technique (AFLP) was used to determine the relationships among Capparis spp. Genetic distances, based on AFLP data were estimated for 45 accessions of Capparis species, from Spain, Morocco and Syria. The results of this analysis support the differentiation of four of the five taxa involved. The group of plants recognised as C. spinosa on the basis of morphological characters, includes several cultivars and appears in an intermediate position between C. orientalis and C. sicula and overlaps with C. orientalis. The other two species C. aegyptia and C. ovata are separate from the rest. Capparis spinosa had a low number of unique bands in comparison with the other species. Although these results cannot confirm the hybrid origin of C. spinosa, the distribution of the bands supports this hypothesis, the most likely parental species being C. orientalis and C. sicula.  相似文献

Randomly amplified polymorphic DNA (RAPD) analysis was employed to assess genetic divergence among 29 neem accessions collected from two agro-ecological regions of India (11 agro-climatic sub-zones), which cover three states, Punjab, Haryana and Rajasthan. Out of 24, 10-mer random primers used for studying genetic divergence, 14 were polymorphic, generating a total of 73 amplification products with an average of 5.21 products per polymorphic primer and estimated gene diversity of 0.49. Genetic relationships among accessions were evaluated by generating a similarity matrix based on Jaccard’s coefficient, ranging from 0.70 to 0.96. The phenetic dendrogram generated by UPGMA analysis grouped accessions into five clusters. RAPD performed within accessions (individual seedlings collected from the same mother plant) showed no variation indicating homogeneous population within accessions. Primers OPA-18, OPC-08 and OPI-03 were found most informative based on their resolving power. The degree of genetic variation detected among the 29 accessions with RAPD analysis suggests that RAPD can be used for studying genetic diversity in neem. The study also demonstrated that neem germplasm collected from northwestern plains of India shows no eco-geographical isolation based on sub-zones because accessions collected from different sub-regions are grouping together in the genetic tree.  相似文献

The genetic diversity in genus Prunus is mainly confined to temperate regions of Himalaya and to a lesser extent in the sub-montane and hilly regions of peninsular India. The cultivated and wild species of Prunus have tremendous potential for improvement and utilization. This paper includes the genetic resources of cultivated and wild useful species of Prunus in India with emphasis on their distribution, potential traits/ useful characteristics and utilization. In this study, the genetic diversity and differentiation of 10 natural Prunus pseudocerasus Lindl. populations were investigated using inter-simple sequence repeat (ISSR) markers. Totally, 18 selected primers generated 150 loci, with an average of 8.33 bands per primer. The results showed that the percentage of polymorphic bands (PPB) was pretty low at the population level (PPB = 1.13–32%), but relatively high at the species level (PPB = 84%). Besides, a high level of genetic differentiation among populations was detected based on the gene differentiation coefficient (G ST = 0.7118) and the hierarchical analysis of molecular variance (AMOVA) (Φ ST = 64.53%, P < 0.001), in line with the low inter-population gene flow (N m = 0.2025). Moreover, Mantel test revealed a significant correlation between genetic and geographic distances among the populations (r = 0.5272, P < 0.005). The high level of intraspecific genetic diversity was probably related with its life history traits, while its small population size and the resultant high levels of genetic drift and inbreeding might explain the low genetic diversity within populations. The relatively high inter-population genetic differentiation was largely attributed to its small population size, habitat fragmentation, the mode of pollen and seed dispersal, and geographic isolation. Based on the present study, conservation strategies were proposed to preserve this valuable natural germplasm resource.  相似文献

Recent reports have shown a surprising diversity of agricultural crops in Omani oases which was attributed to the country’s traditional role at the crossroads of cultures and trade between the Middle East and Asia. Recent surveys have indicated a relatively small diversity of banana (Musa spp.) germplasm which was thought to reflect the prevailing hyperarid conditions of Oman where this drought-sensitive species can only be cultivated in well-watered oases in the North or in Dhofar, the monsoon-dominated south eastern tip of the Sultanate. Surveys near Umq Bi’r, a half-abandoned oasis in the hardly accessible spring area of Wadi Tiwi, famous with Arabic sailors for its abundant fresh water resources, led to the discovery of a so far undescribed triploid Musa acuminata cultivar surviving in a limestone rock niche. Its morphological traits are different from any of the currently grown banana cultivars. Since acuminata bananas are quite sensitive to drought spells, characteristic for the Arabian Peninsula, the occurrence of this specimen provides further evidence for Oman’s role as a refuge of relict crops imported from a more humid region such as coastal East Africa and nearby islands, or Indonesia.  相似文献
