In a survey which lasted one year and included data of 73 dairy cows with their calves, colostrum immunoglobulin G (IgG) of 22 primiparous cows and serum IgG of their calves were lower than the corresponding IgG levels of 51 multiparous cows and their calves. Serum IgG concentration was not correlated with diarrhoea incidence. Although there were no seasonal differences in the IgG concentration of colostrum and calf serum, neonatal diarrhoea incidence was higher in calves which were born in winter than in calves which were born in summer (P < 0.01). Thus the high diarrhoea incidence in winter was not a consequence of an insufficient IgG transfer to the calves. The 60 calves of the second study were fed colostrum on the first day of life. From the second to the tenth day 28 experimental calves received milk and 0.5 l of surplus colostrum of the first and second milking twice a day, whereas 32 control calves received milk only twice a day. Two of the 28 experimental and 11 of the 32 control calves suffered from diarrhoea during the first ten days of life (P < 0.05). These results show that the ingestion of surplus colostrum in addition to milk after the first day of life protects the new-born calf against infectious diarrhoea. 相似文献

To study whether or not passive immunity of neonatal calves against Shiga toxin-producing Escherichia coli (STEC) O26, O111, and O157 was obtained by colostrum administration, serum antibodies in calves after the feeding were determined by enzyme-linked immunosorbent assay (ELISA) in comparison with antibodies in colostrum and sera from donor dams. The highest antibody titers to STEC in colostrum from dams were detected soon after parturition. The antibody titers were found to be elevated in sera of neonatal calves (4-9 hr after birth) orally administered with colostrum with high antibody titers, suggesting that passive immunity of neonatal calves to STEC infection may be obtained by feeding colostrum. These results suggest that colostrum administration to neonatal calves may play an important role in elevating serum antibodies against STEC in neonatal calves. 相似文献

An increased susceptibility to disease in neonatal calves may be attributable to high glucocorticoid levels that influence immune reactions. We tested whether dexamethasone (DEXA) administration influences the proliferation, apoptosis, and number of B- and T-lymphocytes in Peyer's patches (PP) and thymus in calves fed colostrum (C) or a milk-derived formula. All calves were subcutaneously administered bovine colostrum-derived immunoglobulin G and fed chicken-egg derived immunoglobulins that protected against rotavirus and pathogenic Escherichia coli. The DEXA (30 microg/kg of BW daily) was injected for 4 d into groups fed colostrum on the first 3 d (CD+) and those fed the formula that contained nutrients in amounts as in colostrum but no immunoglubulin G (FD+). Groups CD- and FD were fed the same as the other two groups, but did not receive DEXA. Immunohistochemical methods were used to evaluate cell proliferation rates (by labeling of 5-bromo-2'-deoxyuridine), apoptosis rates (by terminal deoxynucleotidyl transferase-mediated X-dUTP nick end labeling). Numbers of T- and B-lymphocytes were determined with antibodies specific for CD3 and CD79 surface proteins. There were significant effects (P < 0.05) of DEXA treatment (decrease of cell proliferation rates in follicles of PP and thymus, increase of apoptotic rate in follicles of PP and thymus, decrease of B-lymphocyte numbers in follicles of PP, increase of B-lymphocyte numbers in domes of PP, increase of T-lymphocyte numbers in follicles of PP, and a decrease of intraepithelial T-lymphocyte numbers). There were significant effects (P < 0.05) of C feeding (decrease of cell proliferation rates in follicles of PP and of B-lymphocyte numbers in interfollicular areas, domes, and follicular-associated epithelium of PP, and an increase of cell proliferation rate in the thymus). A DEXA x feeding interaction (P < 0.001) was found for cell proliferation rate in the thymus. In conclusion, DEXA treatment decreased cell proliferation rates in follicles of PP and thymus and enhanced apoptotic rates in follicles of PP. Colostrum feeding decreased cell proliferation rates, likely of B-lymphocytes, in follicles of PP and numbers of B-lymphocytes in domes, follicular-associated epithelium, and interfollicular areas of PP and enhanced cell proliferation rates and selectively modified DEXA effects in the thymus. 相似文献

Serum immunoglobulins of the IgG isotype recognizing common gram-negative cell core epitopes were serially measured, using a direct ELISA, on samples obtained from 20 neonatal Holstein calves. An R-mutant Escherichia coli (strain J5) was used as a plate antigen in this assay. Total serum IgG concentration was measured using radial immunodiffusion. Half-lives of core antigen-specific IgG (7.56 days) and total serum IgG (22.66 days) were dramatically different (P less than 0.0005). This may be an indication of cross-reactive consumption of core antigen-specific immunoglobulins. 相似文献

OBJECTIVE: To determine the amount of colostral IgG required for adequate passive transfer in calves administered colostrum by use of oroesophageal intubation and evaluate the impact of other factors on passive transfer of colostral immunoglobulins in calves. ANIMALS: 120 Holstein bull calves. PROCEDURES: Calves were randomly assigned to specific treatment groups on the basis of volume of colostrum administered and age of calf at administration of colostrum. Colostrum was administered once by oroesophageal intubation. Equal numbers of calves received 1, 2, 3, or 4 L of colostrum, and equal numbers of calves received colostrum at 2, 6, 10, 14, 18, or 22 hours after birth. Serum samples were obtained from calves 48 hours after birth for IgG determination by radial immunodiffusion assay. Effects of factors affecting transfer of colostral immunoglobulins were determined by use of a stepwise multiple regression model and logistic regression models. RESULTS: A minimum of 153 g of colostral IgG was required for optimum colostral transfer of immunoglobulins when calves were fed 3L of colostrum at 2 hours after birth. Substantially larger IgG intakes were required by calves fed colostrum > 2 hours after birth. CONCLUSIONS AND CLINICAL RELEVANCE: Feeding 100 g of colostral IgG by oroesophageal intubation was insufficient for adequate passive transfer of colostral immunoglobulins. At least 150 to 200 g of colostral IgG was required for adequate passive transfer of colostral immunoglobulins. Use of an oroesophageal tube for administration of 3 L of colostrum to calves within 2 hours after birth is recommended. 相似文献

Five hypernatremic, diarrheic, neonatal calves were treated mainly by the intravenous administration of 5% dextrose alone or with isotonic sodium bicarbonate. All calves recovered without complications. The average reduction rate of serum sodium concentration was about 4 times that recommended and has not been tried successfully before in hypernatremic scouring calves. 相似文献

A simple immunoassay system has been evaluated for use on farm to examine the level of maternal immunoglobin absorbed by newborn calves. Whole blood can be used in the test rather than serum. The test proved easy and reliable when used in studies on 10 farms with 650 calves. More than 50 per cent of the animals were found to be colostrum deficient and, as with other studies, such calves were subject to health breakdown and poor performance although this could be adequately offset by good management. 相似文献

Colostrum intake is important for health and postnatal development of neonatal calves. We studied the effects of enhanced first colostrum feeding on growth, health status, and metabolic and endocrine traits in calves during their 1st wk of life. Calves of group CL (GrCL; n = 7) were fed colostrum of milkings 1 to 6 twice daily during the first 3 d of life, followed by milk replacer (MR) up to d 7. Calves of group CH (GrCH; n = 7) were fed colostrum of the first milking during the first 3 d and then colostrum (of the first milking) twice daily, which on d 4, 5, 6, and 7 was diluted with 25, 50, 75, and 75 parts of MR, respectively. Pre- and postprandial blood samples were taken on d 1, 2, 3, and 7 for the determination of various metabolic and endocrine traits, and on d 5 intestinal absorption capacity was measured using the xylose absorption test. Rectal temperatures and fecal scores were higher (P < .05) in GrCH than in GrCL. Plasma concentrations of total protein and albumin were higher (P < .05) on d 7, IgG on d 2 and 3, and urea on d 2, 3, and 7 in GrCH than in GrCL. Plasma concentrations of triglycerides were higher (P < .05) on d 2 and of phospholipids and cholesterol were higher (P < .01) on d 7 in GrCH than in GrCL. Plasma insulin and glucagon concentrations were higher (P < .05) in GrCH than in GrCL, whereas prolactin and growth hormone concentrations were higher (P < .05) in GrCL than in GrCH. Enhanced colostrum intake had no effects on xylose absorption on d 5. Higher plasma protein, urea, and lipid concentrations in GrCH partly mirrored higher protein and fat intake but additionally pointed to higher protein synthesis and lipid turnover. 相似文献

Effect of colostrum intake on hepatic gluconeogenesis and fatty acid oxidation in the neonatal pig 总被引:1,自引:0,他引:1
A total of 24 newborn pigs were used to determine 1) the relationship between the quantity of colostrum consumed and the capacity for gluconeogenesis and fatty acid oxidation and 2) whether fatty acid oxidation limits gluconeogenesis in isolated hepatocytes. Neonatal pigs were obtained prior to nursing and allotted to one of three treatment groups; fed (ad libitum), limit-fed (25% of fed group), or fasted. Hepatocytes were isolated when pigs were 24 h old. Colostrum intake altered the metabolic status of neonates such that the capacity for glucose synthesis and oxidation of octanoate increased with intake. Glucose synthesis with lactate as the substrate was greater (P less than .01) for fed pigs (10.79 mumol glucose.h-1.mg DNA-1) than for either limit-fed (6.56) or fasted counterparts (4.78), which were similar (P greater than .10). Colostrum intake failed to stimulate synthesis from alanine. The oxidation rate for octanoate was similar for fed and limit-fed pigs (.62 and .61 nmol CO2.h-1.mg DNA-1, respectively) but greater (P less than .05) than that observed for fasted counterparts (.36). Oxidation of octanoate (2 mM) was approximately 30-fold greater than for oleate (1 mM); oxidation of the latter was not affected by either colostrum intake or the addition of carnitine (1 mM). The increase in octanoate oxidation, however, did not elicit an increase in glucose synthesis by fasting pigs with either lactate or alanine as precursors. Thus, we conclude that gluconeogenesis is a function of colostrum intake and that reducing equivalents and(or) ATP may not be primary factors limiting glucose synthesis in pigs fasted from birth. 相似文献

Pairs of newborn calves were exposed to bovine leukemia virus (BLV) when they were given their 1st colostrum feeding. Calves that were given 10(6) BLV-infected lymphocytes in colostrum free of BLV-specific antibody became infected. Calves that were fed 10(7), 10(8), or 10(9) infected lymphocytes in colostrum that contained BLV-specific antibody did not become infected. One of 2 calves inoculated intradermally with 250,000 infected lymphocytes was protected by colostral antibody, but the other was not. Colostral antibody titers in the unprotected calf decreased normally until the calf was 4 months old and then increased markedly; this pattern indicates that the presence of colostral antibody may have prolonged the latent period of the BLV infection. 相似文献

Gnotobiotic calves (24 hours old) were monoinfected with calf rotavirus (CRV) strain NCDV, an enterotoxigenic Escherichia coli (ETEC) strain B44 (K99+), or a nonenterotoxigenic E coli (NETEC) strain 123 (K99-). Calves also were dually infected with CRV and either ETEC or NETEC. Eighteen calves equally allotted between 6 treatment groups were used in these studies: Noninfected controls--group A; CRV--group B; ETEC--group C; NETEC--group D; CRV + ETEC--group E; and CRV + NETEC--group F. Severe diarrhea and villous atrophy were observed in calves of treatment groups B, C, E, and F. Mortality was present only in treatment groups C and E as result of ETEC infection. There were no significant differences in the clinical responses or enteric lesions between treatments B and F, although a significant increase in the concentrations of NETEC was demonstrated in calves dually infected with CRV + NETEC (group F) as compared with calves monoinfected with NETEC (group D). Calves inoculated with ETEC (group C) had severe villous atrophy, neutrophilic infiltration of intestinal lumen, and moderate enterocyte necrosis. Calves dually inoculated with CRV + ETEC (group E) had the most extensive and severe lesions, similar to those in group C, plus a pronounced necrotic fibrino-hemorrhagic enteritis. Infection of enterocytes by CRV did not affect in any way the adherence of ETEC to the intestinal mucosa. Dual viral and bacterial infections of the same enterocytes were evident. 相似文献

During examination of lysozyme concentration we have found out that 90.3% of calf sera had the lysozyme concentration to 0.5 microgram . ml-1, 5.7% of sera showed zero values and only 4.0% of sera values from 0.5 to 1.4 microgram . ml-1. A higher lysozyme content was recorded in sera of calves up to the age of ten days in comparison with sera of calves after the second week post natum. The lysozyme concentration showed in calves a two-phase increase in the age dynamics up to the seventh to eighth week of life with a peak in second and fifth to eighth week of age. The increase rate in the first phase and the time onset of increase in the second phase were in negative, and/or positive relation to the level of immunity obtained through colostrum. The lysozyme concentration in serum of dairy cows was on fourth day post partum as much as ten times higher than in serum of their progenies 48 hours after parturition. In first colostrum the lysozyme concentration fluctuated within the range of 0.15 to 0.65 microgram X ml-1, with an average of 0.30 microgram X ml-1. The concentration of lysozyme and immunoglobulins of the IgG and IgM class in colostrum showed a contrary trend in first and second milk yield, with a tendency towards increase for lysozyme and towards decrease for immunoglobulins. 相似文献

Nine of seventeen neonatal Holstein-Friesian calves were thymectomized, treated with antilymphocyte globulin, and monitored for immunologic functional ability for 4 to 6 months. The thymus weights for 4 to 10-day-old calves and 4 to 6-month-old calves indicated a continued increase in total weight. This indicated significant thymic involution had not occurred at 4 to 6 months. Following thymectomy a wasting syndrome was not observed although an increased incidence of a lowly virulent virus infection did occur. A significant decrease in circulating lymphocytes was observed.Peripheral blood lymphocytes were stimulated in vitro by non-specific mitogens, phytohemagglutinin, bacterial lipopolysaccharide and pokeweed mitogen using the whole blood culture method. Observations included a greater response to phytohemagglutinin and pokeweed mitogen in summer months and variable age related response to all mitogens. Lipopolysaccharide stimulation results were inconclusive. It was concluded that neonatal thymectomy was not a satisfactory experimental procedure for the production of selective immunosuppression in the bovine species. 相似文献

The study objective was to investigate the physiological effects of weaning on beef calves and identify a panel of blood-based welfare biomarkers. On the day (d) of weaning (d 0), 16 spring-born, single-suckled, beef bull calves that previously grazed with their dams at pasture, were assigned to one of two treatments: (1) control (n = 8), calves were loose-housed with their dam, (2) weaned (n = 8), calves were abruptly separated from their dam and loose-housed. Jugular blood was collected on d −4, 0, 1, 2, 3, 7, and 14 relative to weaning (d 0) and assayed for inflammatory and steroid variables. Total leukocyte counts were measured in whole blood. It is concluded that neutrophil number is a robust biomarker of stress and that plasma CXCL8 is a sensitive indicator of stress in weaned and control calves. In future studies, these two biomarkers should be central to the characterisation of stress responses. 相似文献
