A killed gI-negative vaccine combined with a gI enzyme-linked immunosorbent assay (ELISA) test was used for the first time in Sweden in an attempt to eradicate Aujeszky's disease from a weaner pig producing herd. The herd had experienced three severe outbreaks of the disease during a 10 year period and at the start of the programme 96 per cent of the herd's 104 breeding animals were seropositive to the Aujeszky's virus. In addition, there was serological evidence of active virus circulation among younger animals. During the programme, all breeding animals were vaccinated every sixth month and replacement animals were tested free of disease and vaccinated before entry into the herd. When the originally seropositive animals had been rotated out of the herd, all breeding animals and a sample of weaner pigs were tested twice at six weeks' interval. No seroconversions to gI had taken place and the herd was declared Aujeszky's disease-free, 22 months after the start of the programme.  相似文献

A study was conducted to examine the usefulness of a glycoprotein I (gI)-ELISA to monitor Aujeszky's disease virus infection in two vaccinated pig herds; the gI-ELISA can differentiate between pigs infected with Aujeszky's disease virus and pigs vaccinated against Aujeszky's disease with gI-negative vaccines. The two herds had been vaccinated with gI-negative vaccines for several years. The first survey, in September 1986, revealed that approximately 10 per cent of the breeding pigs in a large multiplier herd were seropositive for antibodies to gI of Aujeszky's disease virus, and it was decided to try to eliminate the virus from the herd by gI-ELISA testing and culling of gI-seropositive pigs. A one month quarantine period for incoming stock was established, and only gI-seronegative pigs were admitted to the herd. After two rounds of testing and culling the herd appeared to be free of wild-type Aujeszky's disease virus, and neither Aujeszky's disease virus nor antibodies could be detected either in 21 sentinel pigs placed on the farm or in 347 stillborn piglets or piglets that died shortly after birth. The herd probably remained free of Aujeszky's disease virus until the end of the 27-month period of monitoring except for two of 639 breeding pigs that were unexpectedly found to be positive in the gI-ELISA in November 1987. These sows were culled. A second breeding herd was monitored for antibodies to gI of Aujeszky's disease virus for two years. Bovine herpesvirus type 1 (BHV-1) is an important bovine pathogen, exacerbating poor health and the productivity of cattle. The aims of this study were to detect the efficacy of vaccination programmes in lowering the seroprevalence of BHV-1 gE within the dairy herd and to follow the dynamics of the infection in non-vaccinated herds with uninfected heifers. A two-year longitudinal study was carried out on seven herds that were vaccinated, and in five herds with uninfected heifers without applying a control programme. After the start of the vaccination programme, calves born remained free from the virus. However, in one herd, 7 per cent (95 per cent CI 2 to 18) of these animals showed antibodies to BHV-1 two years after the first vaccination. A decline in BHV-1 antibody prevalence was found in vaccinating herds. Among the five herds not under the control programme, one experienced active virus spread, although one herd experienced self-clearance of the virus. In the herds with high BHV-1 prevalence, vaccinating all cattle from three months of age twice a year with a commercial inactivated marker vaccine efficiently protected offspring from becoming infected, and lowered the prevalence of BHV-1 within the herd. The introduction of Aujeszky's disease virus into a herd of pigs usually results in a rapid spread of the virus and a high percentage of pigs become seropositive. However, herd monitoring for the virus occasionally reveals a single seropositive breeding pig, referred to as a single reactor. The seropositive status of single reactors may be due to previous vaccination against Aujeszky's disease, or to exposure to a field strain of the virus, or to a false positive reaction in the serological assay. During a monitoring programme in Minnesota, 30 pig herds with single serological reactors were detected. Twenty-seven of these single reactors from 19 herds were segregated from their herds immunosuppressed with dexamethasone. Aujeszky's disease virus was isolated from four of the 27 pigs. Three of the four herds subsequently had outbreaks of Aujeszky's disease, suggesting that some single reactors were infected with Aujeszky's disease virus and had the potential to spread the virus within and between herds.  相似文献

This study was carried out in nine unvaccinated Swedish weaner pig-producing herds, ranging in size from 20 to 134 breeding animals, which had experienced a previous outbreak of Aujeszky's disease (AD) and in which seropositive sows were still present. The objective was to quantify the seroconversion rate to Aujeszky's disease virus (ADV) in breeding animals and to find out whether unvaccinated herds could become free from ADV by using only seronegative animals for replacement. Serum samples were initially obtained from all adult pigs in the herds. Animals testing negative, as well as replacement animals, were subsequently tested every second or third month and the herds were followed for 10-28 months. The herd owners were advised to keep seropositive and seronegative animals separate whenever possible and to implement sanitary measures against the reintroduction of the virus into the herds. All herds experienced long consecutive periods (> or = 4 months, median 9 months) without seroconversion. Three herds became free from ADV during the serological study; in two of these herds no seroconversion was observed. In those seven herds where seroconversion occurred, between 9 and 86% of the susceptible pigs became infected. The reinfection was indicative of the reactivation of latent virus in all herds but two, where the reintroduction of the virus was most likely. The pattern of seroconversion was either sporadic, involving not more than three animals at a time, or epidemic, involving a large proportion of the susceptible animals in the herd. Severe clinical outbreaks hit the two largest herds and these only became ADV-free after the conclusion of the study when vaccine was used, which reduced the incidence of seroconversion to zero. The results from the present study show that it is possible for smaller herds to achieve freedom from ADV without any radical control methods, provided that only ADV-free replacement animals are used and the virus is not reintroduced. Moreover, the finding that long periods can elapse without transmission suggests that when infection is discovered in a herd, the risk of massive spread is not necessarily imminent.  相似文献

A 3-year study (1997-2000) was performed on 294 swine herds from Italy, where a National Programme of Control of Aujeszky's Disease (AD) based on compulsory vaccination has been operative since 1997. Aim of the study was to evaluate the progress of this control programme using a survival approach applied to gE-seropositive herds at the beginning of the programme. The cumulative proportion of herds still gE-seropositive at the end of the study was 0.57. No significant difference in the probability of becoming gE-seronegative during the study period was found between herds of different type (breeding versus farrow-to-finish) whereas significant differences were seen between herds from different areas. The Cox's proportional hazards regression, performed on data from 79 herds, showed that the only risk factor significantly associated with a higher probability of becoming gE-seronegative is again the geographical location. Other risk factors considered in the analysis were: type of enterprise, type of replacement of animals, herd size, pig and pig herds densities around the farm, distance from the nearest pig herd and year of beginning of the vaccination with a gE-deleted vaccine.  相似文献

The Pig Health Control Association launched a control scheme for Aujeszky's disease in Britain in January 1978. Participating herds must not show clinical signs suggestive of Aujeszky's disease and must be serum-sampled every six months for the absence of antibodies. Importations are allowed only from listed herds, or through hysterectomy or hysterotomy, embryo transfer, or artificial insemination (the donor animals in each case having been serologically screened beforehand). During the subsequent eight years, 103 herds have qualified, and at the end of 1985, 49 herds (average size 249 sows) were listed. Despite a high degree of inter-herd stock movements, no clinical or serological evidence of Aujeszky's disease has occurred, and the disease has not arisen in any herd established from the listed herds. During the same period, Aujeszky's disease was spreading in Britain and an eradication programme began in March 1983. By the end of 1985, 502 herds not on the Pig Health Control Association's scheme comprising 421,825 pigs had been slaughtered, together with 259 serologically positive animals in 69 other herds. A vaccination trial was carried out in 10 infected herds. The trial included 247 female kids, the number of animals in each herd varying from 15 to 38. About half of the animals in each herd were vaccinated twice at 3 to 4 week intervals, the first vaccination being carried out before the age of 4 months. A combination of a crude filtrate of C. pseudotuberculosis toxoid with whole organisms, was used. Overall, the prevalence of animals with superficial swellings was higher in the unvaccinated than in the vaccinated group during the first 1-2 years following immunization. However, in some herds superficial swellings were as common in vaccinated as in unvaccinated animals. An antibody response following vaccination was demonstrated in the hemolysis inhibition test, but not in the bacterial agglutination test. Superficial swellings were more common in vaccinated animals which were negative than in animals which were positive in the hemolysis inhibition test at 1 1/2 months after vaccination. The vaccine used in the present study, was not sufficiently efficacious to be recommended as the only protective measure against caseous lymphadenitis in Norwegian goat herds.  相似文献

Both in the Federal Republic of Germany and in some neighbouring countries the epizootic situation of Aujeszky's disease has been unsatisfactory for a long time, especially in areas with a high pig density. New findings on vaccines with certain protein deletions have recently indicated the possibility that the disease might be eradicated even in vaccinated herds. By using labelled vaccines it seems possible to distinguish the carriers of vaccine virus antibodies from the carriers of field virus antibodies and to eliminate the animals infected with field virus, while leaving the other animals in the herd and protecting them from infection by vaccination. This procedure would also be practicable in epizootically infected areas and would be less expensive than eradicating the disease on the basis of serological tests without using vaccines. A skeleton of an eradication programme, which was discussed and agreed with scientific experts and with the Federal Laender, as well as rough estimates of costs are being presented.  相似文献

Data from 42,224 cattle from 694 herds collected during the brucellosis eradication campaign were used to examine the effects of calfhood strain 19 vaccination. The prevalence of infection in vaccinated herds was 1.8% compared with 9.1% in non-vaccinated herds (p< 0.005). The mean titre in the former group was lower (p< 0.001). Vaccinated herds required 3.3 herd tests to achieve a provisionally free status compared with 4.8 in non-vaccinated herds (0.001 < p < 0.005). Vaccination did not significantly reduce the number of herd tests in herds with less than 100 breeding females. In tests after the initial herd test only 0.5% reactors were found in vaccinated herds compared with 6.9% in non-vaccinated herds (p< 0.005). There were 0.9% false positive to the Rose Bengal plate test in non-vaccinated and 2.1% in vaccinated animals (p< 0.005) in non-infected herds. In infected herds this percentage was 3.0% and 4.2% respectively by (p< 0.05). In the non-infected herds there were 0.04% false positives to the complement fixation test out of 10,506 non-vaccinated cattle tested and 0.2% out of 24,734 vaccinated cattle.  相似文献

In theory, pseudorabies virus (PRV) may be eliminated from any size of breeding herd by phased test and removal if replacement gilts are not infected with PRV, culling decisions are partially based on PRV status, and the cull rate is higher than the incidence rate of PRV. Annual cull rates are commonly at least 50%, but little information exists on the incidence of PRV within enzootically infected swine herds. The purpose of this study was to develop a method by which spread of PRV could be detected among breeding swine within enzootically infected herds and to determine the incidence of PRV infection in these herds. Data were collected from 17 herds that were quarantined for PRV and ranged in size from 120 to 1,100 sows. At each herd, within the first 5 days of introduction, a group of approximately 30 replacement gilts was identified, vaccinated with a glycoprotein X-deleted PRV vaccine, and blood sample was collected. The owner of 1 herd had a nonvaccinated breeding herd and elected to leave incoming gilts nonvaccinated. After vaccination, blood samples were collected every 1 to 2 months for an average of 13.6 months. Serum samples from vaccinated gilts were tested for antiglycoprotein X antibodies by a specific differential ELISA. Samples from nonvaccinated gilts were evaluated by serum neutralization test. Product-limit method was used to estimate the probability of not becoming infected with PRV. The experiments with sheep and young cattle were carried out to test the immunizing efficacy of inactivated adjuvant vaccine against Aujeszky's disease. The vaccine application at doses of 1 ml and 2 ml to lambs at the age of eight to ten months caused the neutralizing antibody production with a significant rise of titres after revaccination. A survival of infection induced with a dose of 10(5.5) TKID50 of virulent virus was recorded in 62.5% of once vaccinated animals and in 87.5% of twice vaccinated animals. When applying different doses of vaccines (from 1 to 10 ml) to young cattle, the antibody reaction level was directly dependent on the inoculum quantity. The double inoculation of animals with vaccines of 2 ml and 5 ml caused the neutralizing antibody production at titres of 1:35, or 1:46. The animals, immunized with the live or inactivated IBR-vaccine possessing high antibody titres against IBR-virus, reacted upon the vaccination with inactivated Aujeszky's vaccine anamnestically, by early production of antibodies in high titres. Metaphylactic vaccination (2 ml of vaccine) of cattle in herds with an acute course days, however earlier during five days from the revaccination when it was carried out in seven days following the first vaccination.  相似文献

Eradication of Aujeszky's disease from a large-scale pig farm by the 'test-and-cull' eradication procedure based on the use of gene-deleted vaccines and serological monitoring using gE ELISA is described. Aujeszky's disease free status was successfully achieved on a farrow-to-finish type farm characterised by three difficulties interfering with the success of eradication: (i) weaned (30-day-old) piglets remained in the farrowing house until 65 days of age; (ii) the farm did not use nursery rearing; (iii) fatteners were kept together with the breeding gilts. Thus, the immunisation programme had to be adjusted to the management technology used on the farm, and by regular serological monitoring it had to be periodically modified to make it suitable for attaining the goals. This meant that breeding animal candidates of a wide age range and kept in the same air space had to be provided with a continuously high-level immunity by regular vaccination sufficient even for rectifying potential human errors. The immunisation programme was designed by taking into consideration the disappearance of maternal immunity, and a strict vaccination programme was applied during the rearing of breeding gilts kept together with the fattening pigs. Frequent immunisation applied during the rearing of the own breeding animal replacements proved to be of decisive importance for the eradication of Aujeszky's disease from the entire herd. In addition to the accurate implementation of the vaccination programme, regular serological monitoring also made a major contribution to the success of eradication.  相似文献

Vaccination programs to eradicate pseudorabies virus (PRV) are being considered in several countries. Knowledge of factors that influence PRV transmission within vaccinated breeding herds may contribute to the success of these programs. A multivariate analysis of variance of the PRV-seroprevalence in sows in 209 herds (average seroprevalence 67.0% per herd) in the southern Netherlands revealed the following risk indicators: (1) presence of finishing pigs; (2) production type (producers of finishing piglets had a higher seroprevalence than producers of breeding stock); (3) vaccination of the sows during nursing (in comparison with vaccinating all sows simultaneously at 5 month intervals, or vaccination during the second half of gestation); (4) pig density in the municipality where the herd was located (seroprevalence increased with higher pig density); (5) herd size less than 100 sows; (6) average within-herd parity (seroprevalence increased with higher withinherd parity); (7) replacement pigs raised on the premises; (8) vaccine strain administered to the sows. Purchase policy (breeding pigs purchased between 10 weeks and 7 months of age, or use of home-bred gilts only) did not significantly contribute to the multivariate model.  相似文献

The use of an ELISA that can differentiate between swine infected with pseudorabies virus (PRV) and swine vaccinated with a specific PRV vaccine was evaluated on an individual and herd basis, and a system for interpreting ELISA results on a herd basis was developed. In 17 herds, recently introduced replacement gilts, seronegative for PRV, were vaccinated with a thymidine kinase- and glycoprotein X (gpX)-deleted vaccine. After vaccination, blood samples were collected from these gilts approximately every 1 to 2 months for up to 19 months. Serum samples were analyzed for antibodies to gpX antigen, using a commercially available ELISA kit according to the manufacturer's protocol. Herd status was determined as positive, suspect, or negative, according to the serum sample:negative control (S:N) values of the samples collected from the herd. From the 17 herds, 130 evaluations were performed. On 49 (38%) of the 130 herd evaluations, 1 or more gilts had suspect test results. Additional testing was required in 19 (39%) of these 49 herd evaluations to determine the PRV infection status of the herd. Status of herds having gilts with suspect results and no positive results was usually negative after retesting. Herds having gilts with positive results were unlikely to have negative status after retesting.  相似文献

ABSTRACT

Aims: To investigate the seroprevalence of infection with bovine viral diarrhoea (BVD) virus among 75 beef herds and seroconversion in cattle during early pregnancy, and to determine the practices and opinions of farmers towards BVD control and their association with real and perceived herd serological status.

Methods: Blood samples were collected before mating in 75 beef herds across New Zealand from 15 unvaccinated heifers that had delivered their first calf that season. Serum samples were tested for BVD antibodies using ELISA individually, and after pooling samples for each farm. Animals that were antibody-negative were retested at either pregnancy diagnosis or weaning. Farmers were asked to complete a detailed survey about herd demographics, BVD testing and vaccination practices, and opinions towards national BVD control.

Results: Based on the pooled serum antibody ELISA results, there were 28/75 (37%) negative herds, 15/75 (20%) suspect herds, and 32/75 (43%) positive herds. Of 1,117 animals sampled 729 (65.3%) tested negative for BVD virus antibodies; when retested, 47/589 (8.0%) animals from 13/55 (24%) herds had seroconverted. Among 71 famers providing survey responses 11 (15%) believed their herd was infected with BVD, 24 (34%) were unsure and 36 (51%) did not think their herd was infected. Only 19/71 (18%) farmers had performed any BVD testing within the past 5 years and 50/70 (71%) had not vaccinated any cattle for BVD. Support for national BVD eradication programme was strong in 51/71 (56%) respondents, but the biggest challenge to BVD control was considered to be famer compliance. Compared to farmers who did not think their herd was infected, more farmers who thought BVD was present in their herds had previously tested for BVD, would consider testing all replacement calves, and would support establishing a national BVD database; fewer would consider purchasing BVD tested or vaccinated cattle only.

Conclusions and clinical relevance: Only 15% of the beef farmers in this study believed their herds were infected with BVD virus and few of them had undertaken BVD screening. Nevertheless many were supportive of implementing a national BVD control programme. It is likely that the lack of farmer awareness around BVD and the failure of farmers to recognise the potential impacts in their herds are hindering progress in controlling the disease in New Zealand. There are opportunities for New Zealand veterinarians to be more proactive in helping beef farmers explore BVD management options.  相似文献

The present investigation comprised six integrated herds (100-150 sows each) and one specialized fattening herd. Serologic tests by ELISA were performed on blood samples from piglets and growing pigs during various rearing phases in order to determine the presence of Actinobacillus infection and to estimate the age of the animals at onset of the disease. Using this information, herd-specific vaccination programs were designed against Actinobacillus pleuropneumoniae serotype 2. The weaned piglets in the integrated herds were vaccinated twice before being moved to the fattening unit. In the specialized fattening herd, the pigs were vaccinated on arrival at the growing unit and just before transfer to the finishing unit. The frequencies of chronic pleuritis at slaughter demonstrated that a vaccination program adapted to the herd-specific problem, in combination with environmental improvements, reduces the prevalence of chronic pleuritis. On the other hand, only a moderate reduction of the prevalence of chronic pleuritis occurred as a result of vaccination if no environmental improvements were made. In two of the herds during the vaccination period, the number of feeding days and food consumption decreased, while growth per day increased. There was also an apparent correlation between higher serum titres and a decreased prevalence of pleuritis at slaughter in three of the herds.  相似文献

Summary The value of a skin test for the diagnosis of Aujeszky's disease (pseudorabies) in swine was examined. Semipurified and concentrated antigens, obtained by ether/tween 80 inactivation of Aujeszky's disease virus, were inoculated intradermally into seronegative, experimentally infected and vaccinated swine and into pigs with maternal antibodies. A specific skin reaction, characterized by a visible indurated swelling, was observed within 24 hours in animals with active immunity, No reaction was seen in pigs with maternal immunity or in seronegative uninfected and unvaccinated animals. In the infected and vaccinated groups, 89% and 58% respectively of the animals with seroneutralizing antibodies were positive by the skin test response. Positive reactions were observed as early as 8 days after the infection. All but one seronegative animals remained free of antibodies after 2 consecutive skin tests and the course of decline of maternal antibodies was not changed. An earlier skin test did not lead to sensitization for a later application. The present results, together with field experience on 8 farms, revealed that the skin test could be considered a reliable method for diagnosis of Aujeszky's disease in swine on a herd basis.  相似文献

The objective of this field study was to evaluate the protocol of test and removal (T&R) for the elimination of porcine reproductive and respiratory syndrome virus (PRRSV) from 5 chronically infected breeding herds. The T&R protocol involved sampling the entire breeding herd in one day, testing sera by polymerase chain reaction and ELISA to detect previously exposed and/or infected animals, and subsequently removing them from the herd. Following completion of T&R, breeding herds were monitored for 12 consecutive months, using ELISA, for the presence of antibodies to PRRSV. In order to be classified as a PRRSV-negative herd, all samples collected over the 12-month monitoring period were required to be negative by ELISA (s/p ratio < 0.4). At the conclusion of the monitoring period, all 5 farms were PRRSV-negative, according to the defined testing criteria. Approximately 2.2% (74/3408) ELISA false positive samples were detected across all 5 farms during the monitoring period. The diagnostic cost required during the T&R protocol was approximately US $10.66 per animal tested. Limitations of the study were a lack of herds with large (> 2000 sows) breeding herd inventories, and herds with a history of PRRSV vaccination.  相似文献

A study of the safety of a vaccine against lungworm was carried out with pregnant and lactating heifers from three dairy herds with a previous history of lungworm outbreaks in adult cows. Half of the heifers were vaccinated while the other half were not. A slight temporary cough following the vaccination was only observed in one herd. No adverse effects on pregnancy or milk production were seen. All heifers were serologically and coprologically examined before the first, before and after the second immunization, 3 months after introduction to pasture and at the end of the grazing season. Serological and faecal examination of the dairy cows before introduction into pasture confirmed the presence of at least one Dictyocaulus viviparus carrier in each herd. Lungworm infection occurred in all herds during the grazing season, most prominently in the herd with the highest number of heifers. In this herd, mild coughing associated with the lungworm infection was noticed, especially in the non vaccinated heifers. No other signs or symptoms were observed. It is concluded that a vaccine against D. viviparus can be used safely in heifers, before they are introduced into the adult herd, and that this vaccine can be used as a preventive measure against lungworm outbreaks in adult cattle.  相似文献
