Of eight mongrel bitches, the antimesometrial side of the nonpregnant left horn of the uterus at the pregnant or nonpregnant luteal phase was scratched with a Kirschner's wire. Cystic endometrial hyperplasia (CEH) was induced in seven of the eight bitches (87.5%). No difference in the incidence of CEH in the left horn was seen between the pregnant and the nonpregnant groups. Histological examinations showed CEH with a dilatation of the basal glands, resembling "Swiss cheese endometrium".  相似文献

Mucosal biopsies from canine jejunum and ileum were established in organ culture, and remained structurally intact for up to 72 hours. Infection of such mucosal biopsies with an enteropathogenic Escherichia coli (EPEC) 0111 results in the characteristic loss of brush border observed during natural and induced infection in other animal species. Within four hours of infection, bacteria had become attached to the mucous overlying the microvilli and by eight hours had penetrated between the microvilli resulting in partial loss of the brush border. After 24 hours incubation there was almost complete effacement of the brush border with cup and pedestal formation at sites of bacterial adherence. We conclude that EPEC should be considered as a potential cause of diarrhoea in the dog and that organ culture of canine mucosal biopsies provides a useful model for studying infection with EPECs and other enteric pathogens.  相似文献

Our objectives were to develop an ovine model for Escherichia coli-induced preterm delivery, and monitor E. coli (lux modified for photonic detection) invasion of the fetal environment--ewes (124+/-18d of gestation) received intrauterine inoculations using E. coli-lux as follows: control (n = 5), 1.2 x 10(6) CFU/ml (n = 5), 5.6 x 10(6) CFU/ml (n = 5) E. coli-lux. Preterm delivery occurred between 48 and 120 h post-inoculation in 60%, 60% of ewes infected with 1.2, and 5.6 x 10(6) CFU/ml E. coli-lux, respectively, with presence of emitting bacteria confirmed by real-time imaging of lamb tissues. In summary, preterm delivery and/or fetal distress were observed in a majority of inoculated ewes. Finally, the use of photonic bacteria with imaging was a feasible means to monitor bacterial presence ex vivo.  相似文献

The objective of the study reported here was to characterise the microscopic appearance of peptic-injured equine gastric squamous epithelium in relation to the duration of peptic injury. Erosions and ulcers were induced in equine gastric squamous epithelium using a feed deprivation protocol that results in prolonged increased gastric acidity. Specimens of normal gastric mucosa and mucosa with lesions created after 48 and 96 h of feed deprivation were compared for characteristics associated with angiogenesis and mucosal proliferation. Fifteen mature horses, 9 geldings and 6 mares, age 3-20 years, were divided into 3 groups. Group 1 (n = 5) had normal-appearing gastric squamous mucosal epithelium and had been killed due to problems unrelated to the gastrointestinal tract. Groups 2 (n = 5) and 3 (n = 5) had lesions induced in the gastric squamous epithelium by alternating 24 h periods of feed deprivation and ad libitum access to hay, for totals of 48 and 96 h feed deprivation, respectively. Following lethal injection of barbiturate, stomachs were removed and fixed by filling with 4-6 l 10% buffered formalin. Sections were made from lesions in the gastric squamous epithelium adjacent to the margo plicatus along the right side of the stomach/greater curvature and the lesser curvature. Measurements of total epithelial thickness, keratinised epithelium, nonkeratinised epithelium, epithelial projections, capillary extension into the epithelium and lamina propria thickness were made. The cross-sectional areas of arterial and venous vascular structures in the lamina propria at the lesions and their margins were measured using image analysis software. All horses, except one, in Group 2 developed erosions or ulcers in the gastric squamous epithelium after feed deprivation. There were several changes in the epithelium adjacent to erosions and ulcers, compared to normal epithelium, from horses in Groups 2 and 3: total epithelial thickness was significantly (P<0.05) greater, including both keratinised and nonkeratinised layers in most specimens; the length of epithelial projections and extent to which capillaries from the lamina propria extended toward the luminal surface, and the cross-sectional area of vascular structures (arterioles, capillaries, venules) in the lamina propria were significantly greater. Epithelial thickness of erosion beds was not significantly less than normal epithelium, although a greater proportion of the epithelium in erosions consisted of epithelial projections (Group 1, 23%; Group 2, 76%; Group 3, 72%). The cross-sectional area of vascular structures in the lamina propria beneath erosions was significantly greater than in normal mucosa only in Group 2 tissues, whereas in the lamina propria of ulcers it was significantly greater than in normal mucosa only in Group 3 tissues. The epithelial proliferation and increased vascular cross-sectional area in the lamina propria associated with peptic-induced gastric lesions are consistent with processes associated with the initiation of ulcer healing, and these changes temporally coincided with the initiation of peptic insult to the gastric squamous epithelium. These findings demonstrate that processes that promote ulcer healing begin soon after peptic injury and that they progress even with repeated peptic injury. Furthermore, our findings support observations that gastric ulcers often heal without medical intervention, and the theory that medications that reduce gastric acidity do not initiate healing, but rather facilitate ulcer healing by providing a microenvironment that is optimal for healing to proceed.  相似文献

为表达和鉴定犬瘟热病毒(CDV)F1蛋白,本研究通过RT-PCR扩增了CDV HLJ2-07株F1基因,并将其克隆至pET-30a(+)载体中,转化大肠杆菌BL21(DE3),在IPTG诱导下获得大小约为47.5ku以包涵体形式表达的F1重组蛋白,重组蛋白占菌体总蛋白31.5%。Western blot分析表明,表达产物能够被兔抗CDV阳性血清所识别,具有良好的抗原性。间接ELISA检测表明,重组蛋白与犬细小病毒、犬副流感病毒、犬传染性肝炎病毒及犬波特氏杆菌等疾病的阳性血清无交叉反应。本研究为进一步建立检测CDV抗体间接ELISA方法及研制F1蛋白亚单位疫苗奠定了基础。  相似文献

仔猪大肠杆菌性腹泻是由猪致病性大肠杆菌引起的一种常见传染病 ,具有发病急、传播快、死亡率高等特点 ,严重危害养猪业。产肠毒素大肠杆菌 (EnterotoxigenicEscherichiacoli,ETEC)是引起新生和断奶仔猪腹泻的主要致病菌之一 (Yokoya ma等 ,1 992 )。长期以来 ,该病的防治主要依赖于抗菌药物、大肠杆菌多价苗和基因工程疫苗。然而 ,抗生素在动物体内会导致药物残留和产生抗药性 ;用疫苗免疫母猪 ,虽可提高母乳中抗体含量 ,但母源抗体持续时间短 ,不足以保护仔猪整个生长阶段不致病 ;口服高免动物血清虽能有效防治该病 ,但成本较高很难在…  相似文献

河南省部分地区鸡源致病性大肠杆菌生物学特性的研究   总被引:8,自引:0,他引:8  
从河南省不同发病鸡群中采取典型鸡大肠杆菌病痛料,分离鉴定出8株鸡大肠杆茵。8株分离茵对雏鸡具有不同程度的致病作用,其中以兰考株和南阳株致病力最强,可使接种雏鸡100%发病死亡,其次是西华株和登封株,致死率均达75%。药敏试验结果表明:8个茵株均对氟苯尼考、头孢噻呋、头孢曲松、头孢唑啉、阿米卡星、卡那霉素、氯霉素高度敏感;大多数茵株对氧氟沙星、环丙沙星、氨苄西林、链霉素严重耐药。O因子血清凝集试验结果表明:8个茵株的血清型,大多数为O78,占75%,少数为O119占25%。  相似文献

The pregnancies of 13 ewes which were inoculated intrauterine with one of two strains of ovine ureaplasmas resulted in 9 normal and 3 abnormal births, and one ewe was found to be no longer pregnant on postmortem examination. Vaginal ureaplasma infection was detected in the majority of ewes only after lambing. Of the 12 ewes examined at postmortem, ureaplasmas were isolated from the uterus of 5 out of the 6 necropsied up to 21 days post-partum. The vulvar/preputial areas of the majority of lambs that survived were infected with ureaplasmas for the duration of the experiment, but infections of the nasal cavity and eye areas, detected at birth in 4 lambs, were resolved within 8 days post-partum.The only pathological effects detected that could possibly be attributed to ureaplasma infection were a placentitis in an ewe that aborted, and the resorption of the foetus in another.  相似文献

To determine the influence of Ureaplasma diversum on bovine fertility 11 uninfected virgin heifers with normal ovarian cyclic activity were randomly allocated to test or control groups. At a synchronized estrus, five test heifers were given an intrauterine broth inoculum containing 1.09 x 10(8) to 1.4 x 10(9) colony forming units of U. diversum and six control animals were infused with sterile ureaplasma broth medium. All animals were artificially inseminated within one hour of infusion. Pregnancy was diagnosed in one of five test heifers and all of six controls by serum progesterone concentrations measured to 25 days postinsemination. The difference in pregnancy rates between the two groups was statistically significant (p = 0.0152). It was concluded that under the conditions of this experiment U. diversum is capable of causing infertility in cattle.  相似文献

Chronic superficial keratitis (CSK) is an inflammatory disease of the canine cornea, characterised by infiltration of leucocytes into the anterior corneal stroma. The present study describes a quantitative histomorphometric analysis of the cell types infiltrating the corneal stroma in this disease. Infiltrating cells were quantified in samples taken at superficial keratectomy and processed for routine histology. Further characterisation of lymphocyte phenotypes was achieved by immunohistochemistry performed using a panel of monoclonal antibodies recognising canine lymphocyte CD antigens. Lymphocytes expressing the CD4 antigen were found to be the predominant infiltrating cell types in the CSK lesion. A significantly smaller number of lymphocytes expressed the CD8 antigen. The CD4/CD8 ratio was consistently above 2 and rose to above 4 at the advancing border of the lesion. A proportion of lymphocytes were shown by immunohistochemistry to contain gamma interferon. This study forms a basis for work further evaluating the cytological events central to the development of this spontaneous potentially auto-immune corneal disease.  相似文献

To evaluate the possibility that Streptococcus equi subsp. zooepidemicus (S.z) the causative bacterial agent of equine shipping fever pneumonia (ESFP), as well as to investigate its pathogenesis, 10 horses (seven Thoroughbreds and three Anglo-Arab species, ranging from 2-4 years in age) were experimentally inoculated, via an endoscope, into bronchus of the lung lobe with a dose of 30 ml of 1-7 x 10(8) CFU/ml of S.z. After inoculation, autopsy and pathological examinations were sequentially conducted 30 min, 1, 2, 3, 4, 17, 20 hr and 2 weeks later. Pneumonia induced by the intrapulmonary inoculation of S.z was characterized by small purulent pneumonic foci in the inoculated areas. With the lapse of time, these foci developed into serous hemorrhagic pneumonia, hemorrhagic purulent pneumonia, and then purulent, coagulation necrotic pneumonia. These pathomorphological characteristics of experimental pneumonia closely resemble those naturally occurring ESFP. There is strong evidence that S.z. is implicated as a causal factor in ESFP. S.z. grew in the mucus, exudate, and pulmonary effusions. Further, the bacteria showed resistance against phagocytosis by pulmonary alveolar macrophages (PAM) and neutrophils. Inhibition of PAM and neutrophil function is considered to be important in the development of pneumonia. With the progression of the disease, the neutrophils often adhered to the endothelial surface of the alveolar capillary lumen and played a role in generating coagulation necrosis of lung tissues.  相似文献
