ε-Polylysine (ε-PL) is a food-grade cationic antimicrobial that is highly effective against a wide range of food pathogens and spoilage organisms. However, its application within foods and beverages is currently limited because of its tendency to associate with anionic substances, thereby increasing product turbidity or forming sediments. In this study, we examined a potential means of overcoming these problems by forming electrostatic complexes between cationic ε-PL and anionic pectin. The nature of the complexes formed depended on the mass ratio of pectin to ε-PL (R(P-PL)), since this determined their electrical characteristics, aggregation stability, and antimicrobial efficacy. The electrical charge on the complexes went from positive to negative with increasing R(P-PL), with the point of zero charge being around R(P-PL) ~ 8. Soluble complexes or stable colloidal dispersions were formed at low and high R(P-PL) levels, but insoluble complexes were formed at intermediate levels (i.e., 4 ≤ R(P-PL) ≤ 16). The complexes maintained good antimicrobial activity (minimum inhibitory concentration of ε-PL < 10 μg/mL) at R(P-PL) ≤ 20 against two acid resistant spoilage yeasts: Zygosaccharomyces bailli and Saccharomyces cerevisiae. Finally, we showed that certain ε-PL-pectin complexes (10 μg/mL ε-PL; R(P-PL) ≥ 2) could be incorporated into green tea beverages without adversely affecting their appearance or physical stability. This work has shown that the function of a cationic antimicrobial agent (ε-polylysine) can be improved by incorporating it within electrostatic complexes using a food-grade anionic biopolymer (pectin).  相似文献

In compositionally complex environments, like those found in most foods and beverages, the antimicrobial activity of cationic ε-PL is likely to be impacted by its interactions with anionic components. The purpose of this study was to characterize the interactions between cationic ε-polylysine and an anionic biopolymer (high methoxyl pectin, HMP) using isothermal titration calorimetry (ITC), microelectrophoresis (ME), and turbidity measurements. ITC and ME measurements indicated that ε-PL bound to pectin, while turbidity measurements indicated that the complexes formed could be either soluble or insoluble depending on solution composition. Ionic strength and pH were also shown to affect the interactions significantly, highlighting their electrostatic origin. This study demonstrates that ε-PL can form either soluble or insoluble complexes with anionic biopolymers depending on the composition of the system. Our study provides basic knowledge that will facilitate the more rational application of ε-PL in complex food systems.  相似文献

The possible adverse effects on health of diet-derived advanced glycation endproducts (AGEs) and advanced lipoxidation endproducts (ALEs) is of current interest. This study had the objective of determining the effects of the addition of AGE/ALE inhibitors and different types of sugar and cooking oil on Nε-(carboxymethyl)lysine (CML) and Nε-(carboxyethyl)lysine (CEL) formation in model foods (sponge cakes). The cake baked using glucose produced the highest level of CML (2.07±0.24 mmol/mol lysine), whereas the cake baked using fructose produced the highest concentration of CEL (25.1±0.15 mmol/mol lysine). There were no significant differences between CML concentrations formed in the cakes prepared using different types of cooking oil, but significant differences (P<0.001) were observed between the cakes prepared using different proportions of cooking oil. The cakes containing oil generated greater concentrations of CML than sucrose. α-Tocopherol and rutin did not inhibit CML and CEL formation. In contrast, ferulic acid and thiamin, thiamin monophosphate, and thiamin pyrophosphate reduced CML and CEL formation.  相似文献

In this study, water-in-oil emulsions were prepared from water containing different salt concentrations dispersed in an oil phase containing a mixture of β-sitosterol and γ-oryzanol. In pure oil, the β-sitosterol and γ-oryzanol molecules self-assemble into tubular microstructures to produce a firm organogel. However, in the emulsion, the water molecules bind to the β-sitosterol molecules, forming monohydrate crystals that hinder the formation of the tubules and resulting in a weaker emulsion-gel. Addition of salt to the water phase decreases the water activity, thereby suppressing the formation of sitosterol monohydrate crystals even after prolonged storage times (~1 year). When the emulsions were prepared with less polar oils, the tubular microstructure was promoted, which significantly increased the firmness of the emulsion-gel. The main conclusion of this study is that the formation of oryzanol and sitosterol tubular microstructure in the emulsion can be promoted by reducing the water activity and/or by using oils of low polarity.  相似文献

Distribution of chemical forms of manganese(Ⅱ)(Mn(Ⅱ))on plant roots may affect Mn(Ⅱ)absorption by plants and toxicity of Mn(Ⅱ)to plants at its high level.The chemical forms of Mn(Ⅱ)on soybean roots were investigated to determine the main factors that affect their distribution and relationship with Mn(Ⅱ)plant toxicity.Fresh soybean roots were reacted with Mn(Ⅱ)in solutions,and Mn(Ⅱ)adsorbed on the roots was differentiated into exchangeable,complexed,and precipitated forms through sequential extraction with KNO_3,EDTA,and HCl.The exchangeable Mn(Ⅱ)content on the roots was the highest,followed by the complexed and precipitated Mn(Ⅱ)contents.Mn(Ⅱ)toxicity to the roots was greater at pH 5.5 than at pH 4.2 due to the larger amount of exchangeable Mn(Ⅱ)at higher pH.The cations Al~(3+),La~(3+),Ca~(2+),Mg~(2+),and NH_4~+competed with Mn(Ⅱ)for cation exchange sites on the root surfaces and thus reduced exchangeable Mn(Ⅱ)on the roots,in the order Al~(3+),La~(3+)Ca~(2+),Mg~(2+)NH_4~+.Al~(3+) and La~(3+) at 100μmol L~(-1) decreased exchangeable Mn(Ⅱ)by 80%and 79%,respectively,and Ca~(2+) and Mg2+at 1 mmol L~(-1) decreased exchangeable Mn(Ⅱ)by 51%and 73%,respectively.Organic anions oxalate,citrate,and malate reduced free Mn(Ⅱ)concentration in solution through formation of complexes with Mn(Ⅱ),efficiently decreasing exchangeable Mn(Ⅱ)on the roots;the decreases in exchangeable Mn(Ⅱ)on the roots were 30.9%,19.7%,and 10.9%,respectively,which was consistent with the complexing ability of these organic anions with Mn(Ⅱ).Thus,exchangeable Mn(Ⅱ)was the dominant form of Mn(Ⅱ)on the roots and responsible for Mn(Ⅱ)toxicity to plants.The coexisting cations and organic anions reduced the exchangeable Mn(Ⅱ)content,and thus they could alleviate Mn(Ⅱ)toxicity to plants on acid soils.  相似文献

The area of the Jacupiranga Alkaline Complex and its surroundings is characterized by a variety of slope forms. Their spatial differentiation and the differences in relative relief and hilltop heights are a function of lithology. Significant contrasts exist, particularly between the ultramafic rocks of the Alkaline Complex and the quartz-rich rocks of the surrounding Precambrian basement. Although climatic conditions are uniform, the physical and chemical properties of the weathering products formed on the various rock types are very different. Through their properties, they affect the hillslope processes and thereby influence the hillslope development. Slope-forming processes on deeply weathered ultramafic rocks are slow mass movements and chemical denudation. On the more resistant ultramafic rocks, wash processes appear to be more important. Wash and rapid mass movements predominate on gneisses, granites and mica schists.The geomorphological development of the Jacupiranga Alkaline Complex can be explained in terms of a long, continuous downwearing. The large differences in elevation within the Alkaline Complex suggest that the development towards a more differentiated relief may extend well back into the Tertiary. The mutual adjustment of slope forms, processes and lithological controls favoured the development of rock-dependent summit heights, where heights are determined by the steepness of the slopes and the spacing of the drainage lines. Climatic fluctuations in the Pleistocene failed to produce any fundamental change in the long-term development of the hillslopes as their duration was too short with respect to the relaxation time of the hillslopes or their influence was not great enough to alter the overall trend of slope development.  相似文献

The effect of 2(N‐morpholino)ethane)sulfonic acid (MES) on the growth of cucumber (Cucumis sativa L. var. Marketer) in hydroponic culture was determined at 0, 1, 5, and 10 mM concentrations. The effect of adding the MES to the solution at the time of transfer to hydroponic culture or waiting one week was also determined. MES was observed to strongly affect plant growth with increasing concentration in nutrient solution. Tissue and nutrient solution analysis determined that MES affects manganese (Mn) uptake. MES appears to be reduced by Mn, precipitating the Mn out of solution. The suitability of MES as a pH buffer in hydroponic culture is discussed in terms of this effect.  相似文献

The present study was designed to compare surface plasmon resonance (SPR) biosensor, enzyme-linked immunosorbent assay (ELISA), and ultraperformance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) methods for the analysis of Nε-(carboxymethyl)lysine (CML) in glucose-bovine serum albumin (BSA) model systems and to investigate the possible inhibitory effect of selected compounds (α-tocopherol, ferulic acid, rutin, thiamin, thiamin monophosphate, and thiamin pyrophosphate) on CML formation. The reported levels of CML detected were dependent upon the method of analysis employed. The highest reported concentrations were obtained with the SPR biosensor, whereas the lowest were found by ELISA. However, a high correlation was observed between these two immunochemical procedures. CML concentrations were dependent upon the type and concentration of the candidate CML inhibitor. All inhibitory compounds investigated, with the exception of α-tocopherol, decreased the level of CML formation in the glucose-BSA system.  相似文献

A stable isotope dilution assay for quantification of pantothenic acid in sea buckthorn berries, juice, and concentrate using a four-fold labeled isotopologue of vitamin B5 as the internal standard was adopted using reversed phase liquid chromatography-mass spectrometry with electrospray ionization. Because of a rapid sample clean up procedure without the necessity of external calibration, this methodology permits the accurate analysis of a high number of samples within a short time. Sea buckthorn juice was stored at 25 and 40 degrees C for up to 7 days to determine the effects of storage temperature on the stability of pantothenic acid. Analysis of kinetic data suggested that the degradation follows a first-order model. The results of the experiments showed that storage of sea buckthorn juice for 7 days at ambient temperature (25 degrees C) already resulted in a significant degradation of pantothenic acid of about 18%. The processing effects of juice production and subsequent concentration revealed a decrease of about 6-7% in the juice and of 23% in the juice concentrate.  相似文献

Enchytraeids are saprophagous soil organisms, appearing in high abundances and contributing to ecological processes within the soil. For decades they have been used as model species for biological research. In the framework of research on genetically modified plants, however, they have not been considered to date. Following the ISO/DIS guideline, survival and reproduction of Enchytraeus albidus, fed with diets containing Bt-maize (N4640Bt Cry1Ab, DKC5143Bt Cry3Bb1) leaf material were analysed. For comparison, diets with the corresponding untransformed near-isolines (N4640, DKC5143) were examined. Additionally a high quality control diet (oat flakes) was included. Survival and reproduction showed no significant differences between the Cry3Bb1 treatment and the treatment with the untransformed counterpart. For the Cry1Ab treatment survival was significantly higher than for the treatment with the corresponding near-isoline. In contrast, reproduction was significantly lower for the Cry1Ab treatment compared to that for the isoline. For the Cry3Bb1 treatment, no effect was shown on survival or reproduction. For the Cry1Ab variety and its untransformed counterpart, a contrasting result was detected, which is unlikely to be caused by the Bt-protein but rather by differences in other plant components. Overall survival and reproduction were highest for the control.  相似文献

In order to improve the bias of using the Web of Science databases for bibliometric studies, the “front page” has been proposed by Ho’s group in 2012. Using the “front page” as the filter resulted in a huge difference compared to the method from the original paper of Mao et al. Furthermore, a cited reference plays an important role in a published paper. Readers are interested in the paper’s statements and might find useful information from the following researches. The reference section is an important part in a paper; however, it was easily ignored by the authors. This comment also offered information for citing the original idea of publication indicators used for the comparison of countries’ and institutions’ publication performances.

To evaluate adverse impacts of metal pollution originating from smelting activities on the aquatic ecosystem of Deûle river in northern France, water samples were collected from five selected stations along a contaminated region of this river (during two surveys: April–June 2005 and April–May 2007). All samples were analysed using inductively coupled plasma (ICP) atomic emission spectroscopy and/or ICP-mass spectrometry. Both the concentrations of dissolved and particulate elements were determined, and analytical data were compared with national and international water/particle quality guidelines as well as with some values reported in the literature for polluted rivers. For all the metals studied (i.e. Cd, Cr, Cu, Mn, Ni, Pb and Zn), our investigations showed that the effects of the dissolved phase on this aquatic medium were weak, according to water quality status established by US Environmental Protection Agency, USEPA (1994, 1999). Conversely, the levels of metals in suspended particulate matter were found to be much higher than local background contents and natural reference levels in French catchments. These levels were further quantified as “serious” contamination, i.e. above the “red” range that was previously elaborated by most existing metal-contamination scales in French basins of similar geology. The affinity of these metals for the particulate phase in Deûle waters follows the order: Cd >Cr > Pb > Zn = Mn > Cu > Ni. The trace metals released from anthropogenic activities were found to be partly bound to the reactive particulate phase, calcite, which is sensitive to physico-chemical variations occurring in the river ecosystem. To appraise the risk of ecotoxicity by metals, predictions on the ability to release metallic pollutants from calcite into waters were made successfully by testing three equilibrium geochemical speciation models (JCHESS, VISUAL MINTEQ and WINHUMIC) in which soluble organic matter was taken into account. Calculations showed that metal–water–calcite systems in Deûle River are close to thermodynamic equilibrium with generation of solid solutions, MeαCa1-αCO3, by (co)precipitation and/or adsorption reactions. On the basis of results mentioned here, more measurements of river chemistry and assessments of predictive capabilities of chosen water-quality guidelines with time would be developed in aquatic and calcareous areas for controlled dredging operations or other treatment engineering works.  相似文献

A study of Lestes dryas was made in 1978. None was found in ten localities in south east England, which had held populations in the period 1940–1947. Major changes had occurred in eight of the localities, and these could readily account for the disappearance of the insect from them. A search for L. dryas was made in apparently suitable habitat in the Cambridgeshire Fens but none was found. L. dryas was found on three out of eighty waterbodies visited in the west and south of Ireland. Neutral or slightly acid waters with extensive emergent vegetation, notably Equisetum fluviatile, Scirpus lacustris and Typha sp., and supporting the dragonflies Sympertrum sanguineum and/or Coenagrion pulchellum, appear to indicate a suitable habitat for the species. The decline of L. dryas appears to have been caused by a combination of factors: loss of habitat, periods of drought and small population numbers.  相似文献

A pot experiment was conducted to investigate the effect of epigeic earthworm (Eisenia fetida) and arbuscular mycorrhizal (AM) fungi (Glomus intraradices) on soil enzyme activities and nutrient uptake by maize, which was grown on a mixture of sterilized soil and sand. Maize plants were grown in pots inoculated or not inoculated with AMF, treated or not treated with earthworms. Wheat straw was added as a feed source for earthworms. Mycorrhizal colonization of maize was markedly increased in AM fungi inoculated pots and further increased by addition of epigeic earthworms. AM fungi and epigeic earthworms increased maize shoot and root biomass, respectively. Soil acid phosphatase activity was increased by both earthworms and mycorrhiza, while urease and cellulase activities were only affected by earthworms. Inoculation with AM fungi significantly (p?<?0.001) increased the activity of soil acid phosphatase but decreased soil available phosphorus (P) and potassium (K) concentrations at harvest. Addition of earthworms alone significantly (p?<?0.05) increased soil ammonium-N content, but decreased soil available P and K contents. AM fungi increased maize shoot weight and root P content, while earthworms improved N, P, and K contents in shoots. AM fungi and earthworm interactively increased maize shoot and root biomass through their regulation of soil enzyme activities and on the content of available soil N, P, and K.  相似文献

Long-term effects of chemical fertilizers and farmyard manure (FYM) in rice (Oryza sativa)–wheat (Triticum aestivum) cropping system were monitored for two consecutive years after 38 and 39 years on productivity and soil biological properties of Mollisols. The study encompasses varying chemical fertilizer levels of optimum fertilizer rate (120, 26 and 37 kg ha?1 N, P and K, respectively) for both the crops. The treatments were application of 50% NPK, 100% NPK, 150% NPK, 100% NPK + hand weeding (HW), 100% NPK + Zn, 100% NP, 100% N, 100% NPK + 15 t FYM ha?1, 100% NPK(-S) and unfertilized control. The rice and wheat yields were highest with 100% NPK + 15 t FYM ha?1. This treatment also gave maximum and significantly more counts of bacteria, fungi and actinomycetes in soil than all the other treatments after crop harvest. The soil microbial biomass C (410.0 and 407.5 µg g?1) and N (44.53 and 48.30 µg g?1) after rice and wheat, respectively, were highest with 100% NPK + 15 t FYM ha?1, which were significantly higher over all the other treatments. The activities of soil enzymes like dehydrogenase, acid and alkaline phosphatase, arylsulphatase and urease and CO2 evolution rate with 100% NPK + 15 t FYM ha?1 were also found significantly higher over the other treatments. Fertilizer treatments with 100% NPK and 150% NPK were comparable and significantly better than application of 50% NPK, 100% N, 100% NP and 100% NPK(-S) in various studied soil biological properties. Integrated use of 100% NPK with FYM sustained the higher yields and soil biological properties under ricewheat cropping system in Mollisols. Application of Zn and hand weeding with 100% NPK were found better over 100% NPK alone in rice and wheat productivity. Imbalanced use of chemical fertilizers had the harmful effect on soil biological health.  相似文献

Abstract

Effects of fertilizer timing and placement on the grain yield and leaf nutrients of uninoculated sole‐crop cowpeas in two experiments during the minor cropping season on Njala upland soil of Sierra Leone were studied. Timing and placement and their interaction had no significant effects on grain yield. Placement affected highly significantly lamina Mg, Cu and Co and significantly lamina N and Ca. Placement and timing x placement interaction affected highly significantly petiole N and petiole P respectively. Timing affected highly significantly petiole N, which decreased as fertilizer application was delayed. The spectrum of nutrient distribution in both lamina and petiole was decidedly different. Multiple regression studies indicated that variability in only petiole Mg, Cu and Co significantly contributed to grain yield variability. By the introduction of 15 nutrients (6 lamina and 9 petiole) after examining the linear regression coefficients into a composite multiple regression study, lamina Ca, petiole Fe and petiole Co were selected as significant contributors to grain yield variability with marked improvement in R2. Removal of the effects of the correlated variables produced significances in the effects of fertilizer placement and timing x placement interaction on grain yields.  相似文献

Silicon (Si) is considered as a beneficial element to higher plants especially under stress conditions. A factorial experiment, in a completely randomized design with four replications, was used to investigate the effects of two application methods (spraying and soil drenching) and eight levels of Si compounds including 0, 5, 10 and 15 mM of potassium silicate (K2SiO3) and 0, 5, 10 and 15 mM of nanosilica (SiO2). The results indicated that Si application (all or some treatments) decreased transpiration, specific leaf area, petiole length, and promoted the flowering, fruit firmness, leaf/crown number, fresh and dry weight of shoot and root, water use efficiency. They also showed it did not affect the fruit set percentage, yield, chlorophyll index, total soluble solid, leaf area, fresh and dry weight of crown, photosynthesis, stomatal conductance, internal carbon dioxide (CO2) concentration and mesophyll efficiency of strawberry. The results suggest the beneficial effects of Si on growth and development of strawberry.  相似文献
