This study evaluated a novel stearic acid complexed high-amylose cornstarch (SAC) for the prevention of preneoplastic lesions in the colon of azoxymethane (AOM)-treated Fisher 344 rats fed resistant starches at 50-55% of the diet for 8 weeks. Uncooked SAC (r-SAC) diet was compared with raw normal-cornstarch diet (r-CS) or raw high-amylose cornstarch diet (r-HA), and water-boiled CS (w-CS) was compared with w-HA and w-SAC, respectively. w-SAC markedly reduced mucin-depleted foci (MDF) numbers compared with w-HA or w-CS. r-HA significantly decreased aberrant crypt foci (ACF) numbers compared with r-CS or r-SAC. Increased cecum weight and decreased cecum pH were observed in the SAC or HA groups. The highest amounts of total or individual short-chain fatty acids (SCFAs) in cecum and of butyrate or propionate in feces were observed in the AOM-treated w-SAC group. This study revealed the effectiveness of a novel resistant starch in inhibiting colonic preneoplastic lesions and the importance of high-moisture cooking on the suppression of colon carcinogenesis by this resistant starch. 相似文献

The inhibition of (R)-, (S)-, and (+/-)-6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acids (HTCCA) on mushroom tyrosinase was evaluated. All HTCCAs inhibited the tyrosinase activity. The ID(50) values were 1.88, 1.84, and 1.88 for the (R)-, (S)-, and (+/-)-HTCCAs, respectively. The inhibition kinetics analyzed by Hanes-Woolf plots indicated that both (R)- and (S)-HTCCAs are competitive inhibitors of the tyrosinase, with K(i) values of 0.83 and 0.61 mM, respectively. Dimethyl sulfoxide (DMSO) was also tested for its direct inhibitory activity against the tyrosinase and its potential influence on the tyrosinase inhibitory effects of (R)- and (S)-HTCCAs. DMSO, a widely used solvent for tyrosinase inhibitors, was found to dose-dependently inhibit the tyrosinase activity. Addition of DMSO in a tyrosinase digest containing either (R)- or (S)-HTCCA further dose-dependently reduced the tyrosinase activity. These data indicated a potential to use a HTCCA as a tyrosinase inhibitor in food, cosmetic, and medicinal products and a need to improve the solvent system for the studies of tyrosinase inhibitions. 相似文献

Inhibitory effects of nine dibenzylbutyrolactone lignans on a human matrix metalloproteinase, matrilysin, were examined. All of the lignans examined inhibited matrilysin with the IC(50) values ranging from 50 to >280 microM. Matairesinol, which has the basic structure of the other lignans, showed the weakest inhibition. Lignans with methylenedioxy ring(s) or a hydroxyl group at the C5-position inhibited matrilysin more strongly than matairesinol. 5-Hydroxypluviatolide, which has both a methylenedioxy ring and a hydroxyl group at the C5-position, was the most potent inhibitor (IC(50) = 50 microM), suggesting that the introduction of these two elements might enhance synergistically the inhibitory activity of lignans. 5-Hydroxypluviatolide inhibited matrilysin in a competitive manner, and its inhibitory effect was greatly suppressed by the presence of another competitive inhibitor, dimethyl sulfoxide. The precursors of matairesinol, coniferyl alcohol and secoisolariciresinol, had no inhibitory activity, indicating that the dibenzylbutyrolactone structure is essential for the inhibition. It has been shown that lignans have the potential to inhibit matrilysin, and the knowledge of their structure-activity relationship might be beneficial to developing selective inhibitors for matrix metalloproteinases. 相似文献

The inhibitory effects of tea catechins, the O-methylated derivatives of (-)-epigallocatechin-3-O-gallate (EGCG), and the polyphenol extracts from tea leaves (Camellia sinensis L.) on oxazolone-induced type IV allergy in male ICR mice were investigated. Four major tea catechins and two O-methylated derivatives, (-)-epigallocatechin-3-O-(3-O-methyl)gallate (EGCG3' 'Me) and (-)-epigallocatechin-3-O-(4-O-methyl)gallate (EGCG4' 'Me), showed significant inhibitory effects on mouse type IV allergy after a percutaneous administration at a dose of 0.13 mg/ear. Among tea catechins, the compounds including galloyl moieties, such as EGCG and (-)-epicatechin-3-O-gallate (ECG), showed the strongest inhibitory activities on mouse type IV allergy. The inhibitory activities of EGCG3' 'Me and EGCG4' 'Me were higher than that of EGCG at a dose of 0.05 mg/ear. Polyphenol extract from tea leaves of Benihomare cultivar, which includes EGCG3' 'Me, strongly inhibited mouse type IV allergy after percutaneous administration in comparison with that from Yabukita cultivar, which does not include EGCG3' 'Me, at doses of 0.05 and 0.13 mg/ear. EGCG3' 'Me is thought to contribute, at least in part, to the inhibitory ability of Benihomare tea leaves on mouse type IV allergy. EGCG and the polyphenol extracts from Benihomare and Yabukita tea leaves also inhibited mouse type IV allergy by oral administration at 1 h before the sensitization and at 1 h before the challenge with oxazolone. The effects of resveratrol and pterostilbene (two structurally related stilbene compounds) on three human colon cancer cells were systematically compared. Cell viability tests indicated that IC(50) values of pterostilbene were 2-5-fold lower than those of resveratrol in all three cancer cells. Pterostilbene was also more potent in inhibiting colony formation of all three cancer cells. Annexin V/propidium iodide costaining assay and Western blotting analysis showed pterostilbene had a stronger apoptosis-inducing effect, which was evidenced by the higher percentage of annexin V positive cells and higher levels of cleaved caspase-3 and poly(ADP-ribose) polymerase proteins in cancer cells treated with pterostilbene compared with resveratrol. High-performance liquid chromatography analysis demonstrated that intracellular levels of pterostilbene were 2-4-fold higher than those of resveratrol after treatments with individual compounds at the same concentration. Overall, the results demonstrated that pterostilbene had more potent inhibitory effects on colon cancer cells than resveratrol, which may be associated with the superior bioavailability of pterostilbene to resveratrol. 相似文献

Green tea is known to be a potential chemopreventive agent against cancer. In this study, we investigated the inhibitory activities of tea extracts, and in particular the polyphenolic component (-)-epigallocatechin gallate (EGCG), against heterocyclic amine-induced genotoxicity. The tea extracts displayed inhibition of 2-hydroxyamino-6-methyldipyrido[1,2-a,3',2'-d]imidazole (Glu-P-1(NHOH))-induced mutagenicity. This inhibition can be accounted for by the presence of EGCG in the extracts. The mutagenic effect of Glu-P-1(NHOH), which induces single-strand cleavage in supercoiled circular DNA under neutral conditions, was inhibited by EGCG. Using the Drosophila repair test, a test for gross DNA damage, and DNA adduct detection by (32)P-postlabeling, we showed that EGCG prevented 2-amino-3,8-dimethylimidazo[4,5-f]quinoline-induced DNA damage and adduct formation in insect DNA. EGCG was found to accelerate the degradation of Glu-P-1(NHOH) in vitro. This observation suggested that the inhibition by EGCG is associated with an accelerated degradation of metabolically activated heterocyclic amines. 相似文献

The fruit of Viburnum dilatatum Thunb. (gamazumi) was found in a previous study to have strong radical scavenging activity. The present study investigated the antioxidative functions of gamazumi crude extract (GCE) in rats having diabetes induced by the administration of streptozotocin. In rats given water (H(2)O group), plasma levels of glucose, total cholesterol, and lipid peroxide (TBARS) and erythrocyte levels of TBARS increased with time over the experimental period of 10 weeks. These increases were inhibited in rats given GCE (GCE group). After 10 weeks, hepatic, renal, and pancreatic TBARS in the GCE group were significantly lower than those in the H(2)O group. GCE contains a high concentration of polyphenols, and it is expected that they are the active components. These results demonstrate that GCE has an inhibitory effect on the oxidative stress induced by diabetes and suggest that GCE may be useful for the prevention of diabetic complications. Furthermore, as the increase of plasma glucose and total cholesterol was inhibited in the GCE group, GCE may also have anti-hyperglycemic activity in diabetes. 相似文献

Effects of naturally occurring compounds from plants on biotransformation of a mycotoxin, aflatoxin B(1), were evaluated. Among 77 naturally occurring compounds tested, anthraquinones, coumarins, and flavone-type flavonoids were shown to be potent inhibitors of aflatoxin B(1)-8,9-epoxide formation. Addition of the flavonoids galangin, rhamnetin, and flavone strongly inhibited mouse liver microsomal conversion of aflatoxin B(1) to aflatoxin B(1)-8,9-epoxide, a metabolically activated mutagenic product. In contrast to these results, addition of isoflavonoids, catechins, terpenes, alkaloids, and quinones to mouse liver microsomes did not inhibit formation of aflatoxin B(1)-8,9-epoxide. Formation of the aflatoxin B(1) reductase product, aflatoxicol, by chicken liver cytosols was strongly inhibited by curcumin, the diferuloylmethane present in turmeric and other Curcuma species. Curcumin analogues also showed inhibitory effects, and a structure-activity study established that beta-diketone groups linked with two benzyl moieties were essential for inhibition of aflatoxicol formation. An additional 37 naturally occurring compounds tested did not inhibit formation of aflatoxicol. These results demonstrate that dietary constituents in certain fruits, vegetables, and spices may have significant inhibitory effects on metabolic transformation of aflatoxins to their hepatotoxic or carcinogenic derivatives or, alternatively, may promote their transformation into nontoxic products. 相似文献

Summary
Oniscus asellus produced changes in the nutrients leached from Oie and Oa horizons of a hardwood forest soil. Soil with isopods lost more K+ (54%) from the Oie horizon and more Ca2+ (25%), Mg2+ (40%), and water-extractable S (23%) from the Oa horizon than soil without isopods. In contrast, soils with isopods lost less Ca2+ (39076) from the Oie horizon and less dissolved C-bonded S (33%) from the Oa horizon than soil without isopods. In addition, the Oia and Oa horizons exhibited different nutrient dynamics. When isopods were present, the Oa horizon leachates accumulated more Na+ K+, Ca2+, Mg2+, NO3
– , water-soluble SO4
2–, and dissolved C-bonded S, and the Oie horizon retained more of these nutrients. The type of leaching solution also had a major effect on nutrients. Leaching with a simulated soil solution resulted in smaller nutrient losses for K+ and Mg2+ in both horizons and for Na+, Ca2+, and NO3

The effect of carrot psyllid (Trioza apicalis F?rster) feeding and limonene and methyl jasmonate (MeJA) treatments on the essential oil composition and headspace volatiles of carrot (Daucus carota ssp. sativus), cvs. Parano and Splendid, leaves was studied. Carrot psyllid feeding significantly increased the concentrations of sabinene, beta-pinene, and limonene, whereas limonene treatment increased the concentration of (Z)-beta-ocimene in the leaves of both cultivars. The limonene treatment significantly increased the concentration of total phenolics in the leaves of both cultivars, and MeJA treatment increased phenolic concentration in the leaves of Parano. Exogenous limonene spray did not decrease the number of carrot psyllid eggs laid either 2 or 24 h after treatment. The results suggest that carrot psyllid feeding induces changes in the endogenous monoterpene pool in the carrot leaves. Limonene and MeJA treatments affect some induced defenses of the carrot, but the exogenous limonene spray is not an effective oviposition deterrent against carrot psyllid. 相似文献

A 24-well plate trial was conducted to determine the effects of conventional (oxamyl) and transgenic (avidin and aprotinin) insect control chemicals on the non-target bacterial-feeding nematode Bursilla sp. (Rhabditidae: Nematoda). Treatments were added to agar as pure oxamyl, avidin from egg whites or aprotinin from bovine lung, which was then inoculated with the bacteria Xanthamonas campestris as a food source for the nematodes. Oxamyl was toxic to the nematodes at a concentration of 400 ppm, significantly decreasing adult survival, egg laying and consequent progeny development and survival. The 400 ppm concentration of oxamyl also had a significant negative effect on bacterial growth in two of three experiments. Avidin is a biotin-binding protein and concentrations of 100 and 400 ppm significantly decreased the number of progeny produced by first generation nematodes through an effect on egg development. This effect was not significant (P>0.05) at either 10 or 25 ppm. This is the first report of avidin affecting nematode egg development and suggests a role for biotin in this process. Avidin had no effect on visual bacterial growth. Aprotinin is a protease inhibitor and concentrations up to 400 ppm had no significant effect on nematode development or bacterial growth. These lab-based findings need to be further investigated using plants producing avidin and aprotinin, growing in field soil in order to quantify their impact on environmental processes such as decomposition. 相似文献

Soil pH is one of the main factors influencing the solubility and availability of trace elements in arable soils. Thus pH can affect the trace element contents of agricultural crops and thereby indirectly influence human health. The aim of this study was to determine Cd, Ni, Zn, Cu, Mn, Cr, Al and Se contents in spring wheat, potatoes and carrots (Cd, Ni and Zn) and estimate their correlations with certain soil factors (surface and subsurface soil pH and organic matter content) governing the plant availability of these elements. Commercial fields were sampled in Sweden in order to cover a wide range of soil types with respect to pH, soil texture and organic matter content. Concentrations of Zn, Mn, Ni (grain) and Cd (straw) in spring wheat (n=43); Cd, Ni, Zn, Mn, Cu and Al in potatoes (n=69); and Cd, Ni and Zn in carrots (n=36) showed significant negative correlations with surface soil pH (0–25 cm). The Se content of potatoes and Cr content of spring wheat straw were positively correlated with soil pH. Stepwise multiple regressions including a combination of soil pHs (0–25 and 25–50 cm) and organic matter contents (0–25 cm) showed that the organic matter content as well as the surface and subsurface soil pH significantly influenced concentrations of several trace elements in one or more of the studied crops. It was concluded that, if acid deposition together with other acidifying processes (fertilisation, harvest of biomass, etc.) are not balanced by a sufficient amount of liming there might be a decrease in the pH of arable soils, which, in turn will lead to decreased levels of Se in edible crops but an overall increase concentrations of other trace elements. 相似文献

Selected enzymes (alpha-amylase, trypsin, and lysozyme) were allowed to react with some simple phenolic and related compounds (caffeic acid, chlorogenic acid, ferulic acid, gallic acid, m-, o-, and p-dihydroxybenzenes, quinic acid, and p-benzoquinone). The derivatized enzymes obtained were characterized in terms of their activity. In vitro experiments showed that the enzymatic activity of the derivatives was adversely affected. This enzyme inhibition depended on the reactivity of the phenolic and related substances tested as well as on the kind of substrate applied. The decrease in the activity was accompanied by a reduction in the amount of free amino and thiol groups, as well as tryptophan residues, which resulted from the covalent attachment of the phenolic and related compounds to these reactive nucleophilic sites in the enzymes. Kangaroo rats (Dipodomys) have been argued to exert keystone effects because they interact strongly with other species, and their removal results in major changes in community structure. We evaluated the nature and magnitude of such impacts by the endangered Stephens' kangaroo rat (Dipodomys stephensi), employing an exclosure experiment to determine how the removal of this species impacted relative cover by vegetation, plant species diversity, abundance of a dominant annual plant genus (Erodium), and predation of seeds from artificial trays. Fifteen months of D. stephensi removal resulted in significant increases in herb cover and Erodium abundance, and significant declines in bare ground and in seed predation. These results support the hypothesis that D. stephensi has major impacts on plant community structure and seems to function as a keystone species. It remains to be seen how the loss of this species from much of its range will influence local community composition and dynamics. 相似文献

In Korea, Orostachys japonicus has been used traditionally as a drug and health food. The aim of this study was to investigate possible inhibitory effects of O. japonicus extracts on the formation of N-nitrosodimethylamines (NDMA). Chloroform extraction was the most effective method for recovering the highest number of phenolic compounds and flavonoids; in these extracts the greatest nitrite-scavenging activity and inhibition of NDMA formation occurred at pH 2.5. The chloroform extract was separated into 10 fractions (J1-J10); fraction J4 inhibited NDMA formation by 90.1 +/- 0.4%. This fraction was then separated into five subfractions (J4-1-J4-5) using a silica gel column. Subfractions J4-2 [(+)-catechin] and J4-4 (3,4-dihdroxybenzoic acid) inhibited NDMA formation by 89.5 +/- 0.9 and 77.6 +/- 0.8%, respectively. 相似文献
