AIM: To study the senescence of human umbilical vein endothelial cells (HUVECs) and Bcl-2, Bax gene expression associated with apoptosis induced by angiotensinⅡ (AngⅡ).METHODS: HUVECs were cultured in vitro and the cell viability was observed by methyl thiazolyl tetrazolium (MTT). HUVECs were intervened by AngⅡ and valsartan (AngⅡ type 1 receptor blocking) and divided into 3 groups: the control group, AngⅡ group (stimulated with AngⅡ10-6mol/L for 48 h), valsartan group (valsartan was added to cells 1 h before 10-6mol/L AngⅡ treatment). β-gal staining and cell cycle analysis were used to identify the cell aging status. Morphologic changes and percentage of apoptosis were assayed with Hoechst33258 under fluorescent microscope. The expressions of Bcl-2 and Bax, and the apoptosis-associated genes were detected by immunocytochemical staining, RT-PCR and Western blotting. RESULTS: The cell viability by AngⅡ-induced cells was (81.9%±4.1)%, the positive cell number of β-gal staining was significantly higher in AngⅡ-induced cells (80.10%±6.81)% than that in the control cells. The cell cycle was at G0-G1(91.36%±6.45)%, the apoptotic cells significantly increased (31.84±2.86)% under fluorescent microscope. In valsartan group, Bcl-2 mRNA and protein expression increased markedly (P<0.05), but Bax mRNA and protein expression decreased evidently (P<0.05) compared to those in the AngⅡ group.CONCLUSION: Cell apoptosis is possibly an important factor for endothelial cell senescence and vascular aging induced by AngⅡ. One of its molecular mechanisms might be associated with decreasing the expression level of Bcl-2 and increasing that of Bax, which regulate the imbalance between mRNA and protein expression of Bcl-2 and Bax. Valsartan improves endothelial cell aging.  相似文献

Our objective was to study the differentially expressed proteins (DEP) in various Malus spectabilis (crabapple) varieties (M. ‘Snowdrift’, M. ‘Hongling’ and M. ‘Hongjin’) in relation to Malus domestica (‘Gala’) and their role in pollination. Our method used a two-dimensional electrophoresis (2-DE) to analyse the differential proteins in the pollen of several crabapples. The 2-DE apples combined with the tandem mass spectrometry (MS-MS) and protein database retrieval helped us to identify the nature and function of DEPs in ‘Gala’ apples and crabapples. We identified 1195 proteins through 2-DE. Among these, six DEPs, namely chloroplast ferritin, Actin, Beta-fructofuranosidase, vacuolarH+-ATPase catalytic subunit, Full = Phosphoglucomutase, and Cytochrome b were identified by MS-MS. This study identified six DEPs among the pollen from the ‘Snowdrift’ crabapple, ‘Hongling’ crabapple, ‘Hongjin’ crabapple, and ‘Gala’ apples. The DEPs included metabolism related proteins, stress/regulatory proteins, and proteins involved in signal transduction.  相似文献

AIM: To investigate the gene polymorphisms of interferon-γ(IFN-γ) and interleukin-4(IL-4) and the association with asthmatic susceptibility and the levels of plasma IFN-γ, IL-4 and IgE of asthmatic children. METHODS: 100 asthmatic children and 122 control children were enrolled the study. The genotypes of IFN-γ gene-179G/T polymorphism, IL-4 gene-33C/T and-589C/T polymorphisms were tested by PCR-RFLP.The genotype of IFN-γ gene +874A/T polymorphism was tested by AS-PCR.The CA repeat polymorphism of IFN-γ gene was detected by capillary electrophoresis technique.The levels of serum IFN-γ, IL-4 and IgE were measured by ELISA. RESULTS: 100 asthmatic children and 122 control children were all GG homozygotes at -179 locus of IFN-γ gene.-179 locus of IFN-γ gene has no mutation. The genotypes and allele frequency of IFN-γ gene +874A/T and CA repeat polymorphisms showed no significant difference between asthmatic children and the control(P>0.05). An association was revealed between IFN-γ gene +874A/T polymorphism and the level of plasma IFN-γ.The level of IFN-γ was lower in AA genotype than in AT genotype(P<0.05). The genotypes and allele frequency of IL-4 gene -33C/T and -589C/T polymorphisms showed significant difference between asthmatic children and the control(P<0.05).The levels of plasma IL-4 and IgE were higher in TT genotype at -33 locus and -589 locus than those in CT genotype, but only -33C/T polymorphism was associated with the level of plasma IL-4(P<0.05). CONCLUSION: The IFN-γ gene +874A/T and CA repeat polymorphisms were not correlated with asthmatic susceptibility, but there is significant correlation between the level of IFN-γ and +874A/T polymorphism. TT genotype of IL-4 gene -33 locus and -589 locus maybe the susceptible genotype of asthma in children, and the -33 locus polymorphism is associated with the level of IL-4.  相似文献

第五节甘薯的贮藏。一、收获后甘薯生的理变化。(一)伤口愈合:在甘薯收获及运输过程中,要尽量减少薯块表皮的受伤,但总会有一部分薯块受伤。为减少病菌从伤口入侵,应当创造一个适于伤口愈合,即形成木栓层的环境条件。一般在温度30℃和相对湿度90%~92%的条件下,大约经过6天伤口便可愈合;而在温度20℃和相对湿度90%以上的条件下,则需要10天左右。但也绝不是把收获后的甘薯一直捂盖起来,如果这样会发生薯块腐烂。应当在田间晾晒半天,让薯块表面的潮气尽量散掉,然后再装筐或装袋并放在仓库或空屋子进行临时贮藏几天也可直接入窖。由于刚收下的薯块呼吸作用比较旺盛,会放出一定数量的热量和潮气,这恰好满足了甘薯伤口愈合所需的条件。  相似文献

According to the control, applications increased sapling height and this increasing was found important as statistically. Generally, bacteria applications more increase than Perlan on sapling height. Effect of bacteria and Perlan applications on sapling diameter showed different in cultivars and years. Generally, sapling diameter was increase by applications. Applications that on branching was important in 2009, but not important in 2010 as statistically. The most effective applications on branching were T8 and BA-8+T8 in ‘Granny Smith’, in ‘Red Delicious, Red Chief’ BA-8+T8 and BA-8, in ‘Fuji’ Perlan and BA-8 in 2009. Effect of applications on lateral branch degree was different in years and cultivars. According to the control, bacteria and Perlan applications raised length of lateral branch also bacteria was more effective than Perlan.Finally, plant growth promoting bacteria more increased branching and sapling quality than Perlan.  相似文献

Summary

The influence of exogenously applied indole-3-butyric acid (IBA) on root and shoot development of leafy cuttings was analysed in Prunus cerasus P. canescens ‘GiSelA 5’, a dwarfing cherry rootstock, in two successive years. Compared to control cuttings, IBA application (4 g l–1 in 2003; 2.5 g l–1 in 2004) caused higher indole-3-acetic acid (IAA) accumulation in the cutting bases, but that did not influence the percentage of rooted cuttings, nor their survival in either year. However, IBA inhibited callus formation and, consequently, influenced the quality of the developed cuttings. Callus formation impeded root development, reducing the number of main roots, and inhibited the growth of the cuttings, reducing the average total length of shoots formed by individual cuttings. Callus formation was most reduced in the cuttings in the second experimental year, with high initial IAA concentrations.  相似文献

AIM: To observe the effects of angiotensinⅡ (AngⅡ) and angiotensin-(1-7) on the expression of (pro)renin receptor in rat vascular smooth muscle cells.METHODS: The cultured VMSCs were randomly divided into control group, AngⅡ group, Ang-(1-7) group, AngⅡ+losartan (an AT1 receptor antagonist) group, AngⅡ+PD123319(an AT2 receptor antagonist) group and CGP42112A (an AT2 receptor agonist)group. The expression of (P)RR at protein and mRNA levels was detected by Western blotting and real-time PCR,respectively.RESULTS: Compared with control group, AngⅡ distinctly increased and Ang-(1-7) decreased the expression of (P)RR mRNA and protein in VMSCs in a dose-dependent manner (P<0.01). Compared with AngⅡ group, losartan did not inhibit the expression of (P)RR mRNA and protein in VMSCs induced by AngⅡ (P>0.05), but PD123319 did (P<0.01). CGP42112A also induced the expression of (P)RR protein and mRNA in VMSCs (P<0.01).CONCLUSION: AngⅡ induces the expression of (P)RR in VMSCs by AT2. However, Ang-(1-7) inhibits the expression of (P)RR in VMSCs.  相似文献

Summary

Calamondin (× Citrofortunella mitis J. Ingram & H. E. Moore) is known for its ability to develop floral organs to flower in the adult phase during the four seasons. To provide information on molecular events during floral differentiation from axillary buds, we constructed a cDNA library from floral differentiating axillary buds of adult phase calamondin. Ninety-six cDNA clones were randomly selected from this library and sequenced. Ninety-six nonredundant ESTs were identified. Two clones encoded S-adenosylmethione decarboxylase, a key enzymes in polyamine biosynthesis. CmSAMDC 1 (Citrofortunella mitis s-adenosylmethionine decarboxylase 1) contained an 1083 bp ORF that encoded a putative SAMDC precursor of 361 amino acids. Northern blot analysis revealed that CmSAMDC 1 was expressed in axillary buds prior to floral differentiation and in axillary buds immediately after floral differentiation, in immature flower (5 mm), in fully developed flowers (120 mm long), in floral parts (petals, pistils, and stamens) of trees in the adult phase of development, but not in leaves or axillary buds of juvenile phase nucellar seedlings or leaf tissue from trees in adult phase of development. In situ hybridization revealed expression of the CmSAMDC 1 gene in the floral apex of axillary buds after differentiation, and in the phloem of axillary buds and petioles. These results suggest that SAMDC could play a role in actively differentiating and developing reproductive and vegetative organs.  相似文献

SUMMARY

‘Sunrise’ papaya fruit harvested at two stages of maturity [colour break (< 10% yellow peel colour) and 25% yellow peel colour] were treated with 100 nl l–1 1-methylcyclopropene (1-MCP) to determine its effects on ripening, on the activities and levels of gene expression of polygalacturonase (PG), pectin methyl esterase (PME), and βgalactosidase ( βGal), and on the degradation of cell wall components. 1-MCP delayed ripening and the onset of the climacteric, although the peak in the respiration rate was almost the same as that in untreated control fruit. Colour-break fruit treated with 1-MCP exhibited a continuous increase in ethylene production, but at a lower rate than in control fruit. Consequently, 1-MCP-treated fruit ripened with a concomitant reduction in firmness, which was accompanied by an increase in PG and βGal enzyme activities and gene expression. On the other hand, fruit treated with 1-MCP at the 25% yellow stage exhibited lower levels of ethylene production and developed pulp with a rubbery texture at the ripe stage which was attributed to reduced PG, βGal, and PME enzyme activities and gene expression. This was consistent with the higher level of cell wall polysaccharides measured in 1-MCP-treated fruit. The above results indicated that ‘Sunrise’ papaya fruit can be treated with 1-MCP at the colour break stage since they have a greater capacity to recover from the effects of 1-MCP than fruit treated at the 25% yellow stage.  相似文献

The effects of two oxylipins – jasmonic acid (JA) and 9, 10-ketol-octadecadienoic acid (KODA) – on ethylene metabolism and ethylene receptor genes were examined in peach fruit (Prunus persica L.) harvested at 88 days after full bloom (DAFB), in the pre-climacteric stage, and at 102 DAFB, at the beginning of the climacteric stage. Immediate post-harvest applications of either n-propyl dihydrojasmonate (PDJ) (a JA analog) or KODA stimulated ethylene production and 1-aminocyclopropane-1-carboxylate (ACC) concentrations in fruit that were harvested at 88 and 102 DAFB. KODA application significantly increased levels of ACC oxidase (PpACO1) expression at 5 and 8 days after treatment (DAT) in 88-DAFB-harvested fruit, and at 1 DAT in 102-DAFB-harvested fruit. Expression levels of ethylene response factors (PpERS1) were not influenced by either PDJ or KODA applications at 88 or 102 DAFB. In contrast, levels of ethylene-receptor-coding genes (PpETR1) expression in 88-DAFB KODA-treated fruit increased at 8 and 14 DAT. Expression levels of constitutive triple response (PpCTR1) in the untreated control fruit decreased with DAT in both 88- and 102-DAFB-harvested fruit.

These results suggest that certain oxylipins may promote ethylene production in peach fruit, and that PpETR1 and PpCTR1 may be associated with the autocatalytic system of ethylene production.  相似文献

Plants of six strawberry cultivars were raised under controlled conditions and tested for flowering and yield potential. Short days (SD) at intermediate temperatures for 4 weeks in August induced profuse flowering in subsequent long days (LD) in all cultivars except the late-flowering ‘Malwina’. LD conditions induced flowering only in ‘Nobel’, which has an everbearing parent. ‘Nobel’ and ‘Saga’ exhibited broad temperature adaptation for SD floral induction, which was generally reduced or suppressed at 9 and 27°C. After autumn planting, all cultivars flowered most abundantly in plants raised in SD and intermediate temperatures. Flowering was earliest in ‘Nobel’ and ‘Rumba’. Plants that did not reach floral commitment after 4 weeks in SD continued and completed induction under subsequent natural SD conditions after planting in the field, demonstrating the capability of fractional induction. Berry yield varied in parallel with flowering in the field and was always higher in plants raised under SD conditions. The traditional cultivars ‘Florence’ and ‘Sonata’ out-yielded the more recent cultivars. Some cultivars lost more than two thirds of their initiated flowers during the winter with obvious consequences for their yields. With proper raising management, acceptable yields were obtained after autumn planting even in a cool Nordic climate.  相似文献

In the last few years, landscape researchers have sought to understand temporal and spatial patterns of landscape changes in order to develop comprehensive models of land cover dynamics. To do so, most studies have used similar methods to quantify structural patterns, usually by comparing various landscape structural indices through time. Whereas the necessity for complementary approaches which might provide insights into landscape dynamics at some finer scale relevant to local managers has been expressed, few studies have proposed alternative methodologies. Moreover, the important relationship between the physical constraints of the landscape and land use dynamics has been seldom emphasized. Here we propose a methodological outline which was applied to the study of a rural landscape of Southern Quebec, Canada, to detect spatial and temporal (1958 to 1993) patterns of land cover changes at field, patch and landscape level. We then relate these patterns to the underlying physical structure of landscape elements using GIS and canonical correspondence analyses. We use the different geomorphological deposit types as stable discriminant factors which may constrain land use.Canonical correspondence analyses showed relations of land use and land use changes to the physical attributes of the landscape elements, whereas spatial analyses revealed very dynamic patterns at finer spatial and temporal scales. They highlighted the fact that not only the physical attributes of the landscape elements but also their spatial configuration were important determinants of land use dynamics in this area. Thus more land use changes occurred at the boundary between geomorphological deposit types than in any other locations. This trend is apparent for specific small-size changes (e.g. forest to crop), but not for the large-size ones (e.g. abandoned land to forest). Although land use changes are triggered by socioeconomic forces in this area, these changes are nevertheless constrained by the underlying physical landscape structure. A thorough comprehension of historical changes will enhance our capability to predict future landscape dynamics and devise more effective landscape management strategies.  相似文献

Summary

Many people who are allergic to birch pollen are also allergic to apple fruit, due to cross- allergenicity. Since apples are the most extensively consumed fruit in Europe, it is highly relevant to develop a hypo-allergenic apple.Apples with significantly reduced levels of the allergen, Mal d 1, may allow many apple allergics to eat them without an allergic reaction. We are currently collaborating to develop a hypo-allergenic apple within the European Integrated Research Project, ISAFRUIT (www.isafruit.org). Hypo-allergenic apple plants (Malus × domestica Borkh., ‘Elstar’) with decreased levels of Mal d 1 mRNA were produced by RNA interference (RNAi) technology. Ten genetically modified (GM) apple lines were selected. In vitro plantlets were first transferred to a greenhouse, then grafted onto wild-type M.9 rootstock to promote the development of fruit-producing trees. Levels of Mal d 1 gene silencing were measured repeatedly by quantitative real-time PCR. Compared to leaf samples from wild-type ‘Elstar’, two GM lines showed modest levels of gene silencing (up to 250-fold), whereas the other eight GM lines were significantly silenced (up to10,000-fold) in Mal d 1 gene expression. These levels of silencing were unaffected by grafting, and have been stable over more than 3 years, and throughout all developmental stages.  相似文献

AIM:To determine the effects of adenovirus mediated hypoxia-inducible factor-1α(HIF-1α) gene on myocyte apoptosis and cardiac function after acute myocardial infarction (AMI) of rabbits.METHODS:Rabbits were made AMI model, divided into 4 groups at random and infected with Ad-HIF-1α, Ad-blank or Rz-HIF-1α, respectively. Sham group was served as control. The cardiac functions of rabbits at different time points were detected by Maclab/8s. Myocyte apoptosis was detected with TUNEL method at different time points.RESULTS:Apoptotic cells were the highest in Rz-HIF-1α treated group, less in Ad-HIF-1α treated group than that in Ad-blank group. Apoptotic cells decreased with extension of time and were found at the time of 56 d. The cardiac function parameters were the highest in sham, lowest in Rz-HIF-1α, and were higher in Ad-HIF-1α than those in Ad-blank.CONCLUSION:Myocyte apoptosis is inhibited by Ad-HIF-1α, increased by Rz-HIF-1α. The cardiac function is improved by Ad-HIF-1α, deteriorated by Rz-HIF-1α.  相似文献
