This study was carried out in Arsin (Trabzon/Turkey) in 2011. The effects of different harvest time and altitudes on the quality of the nuts have been investigated. The study was performed on Fo?a hazelnut and the harvest process has been conducted at three terms, which are on normal harvest time and ten days before and after harvest time. The harvested nuts were dried in the shade on the concrete floor until their moisture content decreased to 5?%. Some properties of nuts including yield, fruit weight, internal weight, shell thickness, and protein, oleic, and linoleic acid amounts have been investigated. As evaluated all of the fruit properties it can be concluded that 11 August is the most suitable harvest date for coast zone. On the other hand, no significant differences were obtained in the point of protein, oleic, and linoleic acid amounts for different harvest time and altitudes. 相似文献

Poysean (Euphorbia × lomi Rauh) has been introduced in the Mediterranean countries as an ornamental plant for indoor uses. When used outdoor, few information is available about its ability to withstand the Mediterranean temperature extremes. An experiment in an open stand was performed to evaluate poysean tolerance to the winter and summer temperatures of the thermo Mediterranean climate and its ornamental value for urban greening. Two genotypes of poysean (Nguen Muang, NM; and Soi Budsanin, SB) with similar bracts and flower colours were grown at either 2 or 3 plants per pot during 23 months. Number of flower racemes and leaves per plant and plant height were recorded monthly. Overall ornamental value of each pot was scored by means of a panel test (PT) performed by independent examiners. Score assignment was homogeneous between examiners. On average, NM showed a 36% lower number of flower racemes, but a 30% higher ornamental value than SB. During fall and beginning of the winter, NM showed a higher number of flower racemes per plant than SB. Increasing plant density from 2 to 3 plants per pot did not influence flower racemes and leaves per plant. From March to November of the second year, number of flower racemes in NM was similar to the first year, whereas SB flower production was markedly higher, which can be due to a genotypic difference to Mediterranean temperature tolerance. Correlation between number of flower racemes per pot and ornamental value was higher in SB than NM, which suggest that in NM flower abundance contribute less to the pot ornamental value. The present data suggest that poysean is able to withstand the Mediterranean temperature extremes and still produces flower racemes during winter, which indicate it as a suitable ornamental plant in outdoor applications. 相似文献

Soil and leaf samples were analysed in an attempt to identify the causes of leaf-tip scorch commonly observed in cultivated Protea sussannae and Protea eximia hybrid ‘Sylvia’. Leaf-tip scorch was hypothesised to be caused by nutritional problems. Soil and leaf mineral contents were measured and compared between healthy plants and those suffering from leaf-tip scorch. Leaf-tip scorch was found to be associated with increase electrical conductivity, total cation concentration and the concentrations of Na, K, Ca, Mg, Mn and B in the soil. It was concluded that failed attempts of growers to remedy the problem by foliar spraying of micronutrients might have artificially increased the levels of Mn and B. Leaf-tip scorch was associated with decreased foliar concentrations of Fe and increased concentrations of Na and Mn. No association between leaf-tip scorch and either ∂13C or ∂15N was found. It was concluded that changes in water relations and N acquisition were not associated with the symptoms. The significant decrease in the Ca concentration in the tips of leaves showing advanced leaf-tip necrosis indicated that a deficiency of Ca in the leaf tips may have been the proximal cause of the pathology. AIM:To investigate Nrf2 that regulates the expression of γ-glutamylcysteine synthetase (γ-GCS) in the inflammatory cells in bronchial asthma guinea pig bronchoalveolar lavage fluid (BALF). METHODS:Adult male guinea pigs were randomly divided into control group (group A), asthmatic group (group B) and dexamethasone group (group C). Asthmatic model was established by the method of ovalbumin challenge. MDA concentration in the lung tissue homogenate was detected. The total cell count and the proportion of inflammatory cells in BALF were measured. The methods of immunohistochemistry and in situ hybridization were used for detection of the protein and mRNA expressions of Nrf2, Bach1 and γ-GCS. RESULTS:(1) The proportion of eosinophils (EOS) in BALF and the MDA concentration of the lung tissue in group B were higher than those in group A and group C. (2) The result of in situ hybridization indicated that the A value of γ-GCS was the highest in group A compared to group B and group C, but the A value of Nrf2 and Bach1 in 3 groups has no statistical significance. (3) Immunohistochemistry indicated that the A value of γ-GCS in group B was lower than that in group A. The positive rate in cell nucleus of Nrf2 in group B was lower than that in group A and group C. The positive rate in cell nucleus of Bach1 in group B was higher than that in group A and group C. (4) The mRNA expression of γ-GCS (A value) showed positive correlation with the positive rate in cell nucleus of Nrf2 and negative correlation with the positive rate in cell nucleus. A negative correlation between the proportion of EOS in BALF of group B and the γ-GCS mRNA was observed. CONCLUSION:There is disequilibrium between oxidation and anti-oxidation in bronchial asthma guinea pig. Inflammatory reaction decreases the expression of γ-GCS in the inflammatory cells in bronchial asthma guinea pig. Dexamethasone regulates the nuclear translocation of Nrf2/Bach1 and increases the expression of γ-GCS. 相似文献

SummaryIn order to confirm the hybrid origin of Phalaenopsis intermedia Lindl., internal transcribed spacers (ITSs) of ribosomal DNA (rDNA) and three fragments of chloroplast DNA (cpDNA) were studied. Nineteen clones of ITS sequences from three accessions of P. intermedia Lindl. were aligned with nine species of sections of Phalaenopsis and Stauroglottis that were candidate parents. A phylogenetic tree, derived from the ITS data, was constructed by the Neighbor-joining (NJ) method. Two major groups were shown for 19 clones of ITS sequences of P. intermedia based on the phylogenetic tree. The average genetic distance between the aforementioned two groups and the candidate parents was calculated based on the Kimura 2-parameter method. One group had the lowest genetic distance from the ITS repeat sequences of P. aphrodite Rchb.f., and another group had the lowest distance from that of P. equestris (Schauer) Rchb.f. The results indicated that both P. aphrodite and P. equestris are parents of P. intermedia based on the ITS data. In addition, analysis of three fragments of cpDNA, namely the trnL intron, the trnL-trnF intergenic spacer (IGS), and the atpB-rbcL IGS, showed that P. intermedia had the lowest genetic distance from P. aphrodite. Both the ITS and cpDNA data, as well as reference to the effects of maternal inheritance of cpDNA, suggest that P. aphrodite was the maternal parent and P. equestris was the paternal parent of P. intermedia. Therefore, molecular evidence supports P. intermedia being a natural hybrid derived from P. aphrodite and P. equestris. 相似文献

Trials in southern Tasmania examined the thinning effects of ethephon on ‘Gala’ and ‘Jonagold’ apples. Untreated controls were compared with ethephon thinning sprays applied at full bloom at 50,100,200,400,800 and 1 600 mg I“1 to both cultivars. Thinning of both cultivars was related to the concentration of the spray and in most cases logistic models were fitted. Thinning effectiveness was largely reflected in increased fruit weight and size. The ‘Gala’ fruit was still not large enough for Australian markets. Ethephon at 200 mg I“1 effectively thinned ‘Jonagold’ and produced the required increases in fruit size and weight. ‘Jonagold’ was overthinned by the high concentrations of ethephon, but this was not reflected in increased fruit weight and size. Ethephon also reduced vegetative growth at the higher concentrations which was considered an advantage. More work is required to establish specific recommendations for either cultivar. 相似文献

Lilium lancifolium Thunb. exhibits wide genetic diversity and numerous genetic traits within progeny populations. Parent morphology affects the distribution and assortment of progeny phenotypes. In this study, morphological analysis and fluorescence in situ hybridization (FISH) were conducted for cross progeny (F1 and BC1). Results showed that F1-1 hybrids had greater plant height than the parents; however, backcross progeny plant height (group 1) was low. Leaf number in both groups was considerably low in F1 hybrid, although high in backcrossed plants. Flower number and days to flower opening were intermediate to those of the parents for F1 hybrids, yet low in BC1 progeny. In group 1, backcross progeny showed small bulb length; however, group 2 progeny showed large bulb length. Positive and negative correlations between the phenotypic traits of parents and progeny confirmed significant variations. According to FISH results, F1-1 and F1-2 hybrids distinctly exhibited nine and eight 45S rDNA loci which were same in position with 45S loci of the parents. In backcross progeny, eight 45S rDNA signals were detected in four BC1 progeny of group 1, while 10 signals were observed in all group 2 progeny, same with the L. lancifolium karyotype. Morphological analysis and FISH helped in scrutinising progeny to obtain hybrids with desirable characteristics. 相似文献

A flavor-related enzyme from Toona sinensis (TS), ?-glutamyl-transpeptidase (GGT), was purified 40.67-fold with 13.40% recovery using ammonium sulfate precipitation, dialysis and gel filtration chromatography. Gel filtration chromatography and sodium dodecyl sulfate acryl amide gel electrophoresis (SDS-PAGE) revealed that GGT was a heterodimer with a molecular weight of 90 kDa, and the molecular size of its two subunits was 26 kDa and 64 kDa, respectively. The apparent Km and Vmax of GGT were 3.623 μmol/mL and 2.831 μmol/mL/min of protein, respectively. The optimum reaction temperature was 60°C, while the optimum pH was 7.0. GGT can tolerate temperatures of 70°C for a short time and can keep stable activity at 60°C for more than 2 hours. The effect of metal ions on GGT activity showed that Cu2+ and Zn2+ had strong inhibition, while Fe3+ had strong activation, and other metal ions had only a slight effect. LC-MS analysis indicated that GGT catalyzed (S, S)-γ-glutamyl-(cis-S-1-propenyl)-thioglycine, a flavor precursor found in TS, to release glutamic acid moiety. 相似文献

Urban areas are particularly vulnerable to climate change due to the Urban Heat Island (UHI) effect, which can be mitigated by urban vegetation through shading and evapotranspiration. Nevertheless, there is still a lack of spatially explicit information on the cooling capacity of green infrastructure for most Latin American cities. In this study, we employed Land Surface Temperature (LST) of the Neotropical Mexican city of Xalapa to (1) analyze its Surface UHI (SUHI) compared to its peri and extra-urban areas, (2) to assess the cooling capacity of urban green spaces larger than 1 ha, and (3) to evaluate the role of green spaces’ size, shape and their surrounding tree cover percentage (Tc) on green spaces cooling range. We evaluated the cooling range of green spaces and their relationships with green spaces metrics and Tc via a linear mixed-effect model and identified threshold values for the variables at 25 m, 50 m, 100 m, and 200 m from the borders of green spaces through Classification and Regression Trees. Xalapa exhibits a SUHI of 1.70 °C compared to its peri-urban area and 4.95 °C to the extra-urban area. Green spaces > 2 ha mitigated heat at ~2 °C and the cooling range was influenced by the size of green spaces ≥ 2.8 ha and Tc > 21% at 50 m and only by Tc surrounding the green spaces at 100 m and 200 m. This shows that the size threshold of urban green spaces should be complemented with the presence of Tc starting at least 50 m to maximize the cooling capacity provided by the green infrastructure. Planning agendas should account for the interaction between the size of green spaces and the cumulative cooling effect of scattered vegetation inside urban areas towards compact green cities to cope with urban warming. 相似文献

Process-based forest landscape models are valuable tools for testing basic ecological theory and for projecting how forest
landscapes may respond to climate change and other environmental shifts. However, the ability of these models to accurately
predict environmentally-induced shifts in species distributions as well as changes in forest composition and structure is
often contingent on the phenomenological representation of individual-level processes accurately scaling-up to landscape-level
community dynamics. We use a spatially explicit landscape forest model (LandClim) to examine how three alternative formulations
of individual tree growth (logistic, Gompertz, and von Bertalanffy) influence model results. Interactions between growth models
and landscape characteristics (landscape heterogeneity and disturbance intensity) were tested to determine in what type of
landscape simulation results were most sensitive to growth model structure. We found that simulation results were robust to
growth function formulation when the results were assessed at a large spatial extent (landscape) and when coarse response
variables, such as total forest biomass, were examined. However, results diverged when more detailed response variables, such
as species composition within elevation bands, were considered. These differences were particularly prevalent in regions that
included environmental transition zones where forest composition is strongly driven by growth-dependent competition. We found
that neither landscape heterogeneity nor the intensity of landscape disturbances accentuated simulation sensitivity to growth
model formulation. Our results indicate that at the landscape extent, simulation results are robust, but the reliability of
model results at a finer resolution depends critically on accurate tree growth functions. 相似文献

SummaryThe present work investigated the effects of different aqueous extracts of organic waste compounds on growth, proliferation and photosynthetic activity in ‘M9’ (Malus domestica Borkh.) shoot cultures, with the aim of determining the feasibility of using in vitro cultures as a tool for the rapid evaluation of organic amendments in agriculture. Aqueous extracts of the following organic waste compounds: cow manure (CM), sugarbeet industrial waste (SB), mixed grape, poultry and municipal solid waste (GPM), and citrus pruning and industrial waste (CPI) were prepared at a rate of 1:10 (w/v) compound:distilled water. The basal media used in the proliferation phase were: (i) PM1, modified Murashige and Skoog (MS) enriched with 4.4 µM 6-benzyladenine (BA); (ii) PM2, as PM1 but with a reduced cytokinin concentration (1 µM BA) to evaluate possible hormone effects; and (iii) PM3, 4.4 µM BA with reduced salt strength (0.33 MS) to induce nutrient deficiency. Hormone-free medium with half-strength MS salts was used for rooting. All media were enriched with each extract at 0, 0.2, 2, 20 or 200 ml l–1. Photosynthetic activity was measured on PM3 medium enriched with SB or CM. Standard culture conditions were 22° ± 2°C, with a 16 h photoperiod at 30 µmoles photosynthetically active radiation (PAR) m–2 s–1, but at 80 µmoles PAR m–2 s–1 to determine photosynthetic activity. Shoot weight increase in PM1 was not affected by the GPM and CPI extracts, while the growth trends of CM- and SB-treated shoots were described by a second-degree function with maxima at 2 ml l–1 and 0.2 ml l–1, respectively. Shoot proliferation for SB was represented by a quadratic curve (maximum at 2 ml l–1), was linearly reduced as GPM increased, but was not affected by CM or CPI. Treatments did not significantly affect rooting percentage and root length; however root number was increased by SB at 2 ml l–1.CO2 fixation increased linearly with both SB and CM, despite reduced growth at the highest levels of extract. 相似文献

AIM: To observe the role of zipper-interacting protein kinase (ZIPK) in the regulatory effects of protein kinase Cα (PKCα) and protein kinase Cε (PKCε) on calcium sensitivity during hemorrhagic shock(HS) in rats. METHODS: The skinned first class arborization of superior mesenteric artery (SMA) from HS rats were adopted to observe the influence of inhibitor of ZIPK on the effects of PKCα and PKCε agonists on calcium sensitivity after shock via measuring the contraction initiated by Ca2+ with isolated organ perfusion system, hypoxic vascualr smooth muscle cells (VSMCs) were adopted to measure the protein expression and activity of ZIPK after applying PKCα and PKCε agonists following hypoxia via Western blotting. RESULTS: (1) The calcium sensitivity of SMA was decreased after 2 h shock, and increased by agonists of PKCα and PKCε. Emax of Ca2+ was increased from 47.2%to 66.5% (P<0.01) and 66.3% (P<0.01) of normal control respectively as compared with 2 h shock group. The increasing effects of PKCα and PKCε agonists on calcium sensitivity of SMA after 2 h shock were weakened by the inhibitor of ZIPK. The cumulative dose-response curve of Ca2+ was shifted to the right, the Emax of Ca2+ was decreased to 42.6% and 47.5% of normal control (P<0.01), respectively. (2) The protein expression and activity of ZIPK in VSMCs were decreased after 2 h hypoxia, and were increased by the agonists of PKCα and PKCε following 2 h hypoxia. CONCLUSION: PKCα and PKCε regulate the calcium sensitization probably through changing the protein expression and activity of ZIPK following HS in rats. 相似文献

A single postharvest urea spray at 5 and 10% increased nitrogen (N) content in one year old bark and wood and in the flower buds and blossoms in 1987. In 1988, there was also an increase in N in one year old bark and wood but not in the N content of flower buds and blossoms. Fruit set was not significantly increased in either year. Control trees were much taller in N in 1988 than they were in 1987 since additional soil N was applied. Urea labelled with 15N was applied to branches and individual spurs immediately after harvest. When the l5N-urea was applied to branches, the next season’s flower buds had 12% of their N derived from the fertilizer (NFF), but when urea was applied to individual spurs only 8% NFF was detected. Remobilization of labelled N from treated to adjacent spurs resulted in less NFF than would occur if all spurs on the same branch received labelled N. Flower clusters distal and basal to the spurs treated with labelled l5N showed considerable amounts (>60% of sprayed spur) of N from the labelled spray. However, harvest analysis of adjacent and distant spur leaves and fruits from treated spurs and branches revealed that urea was only locally mobilized in the tree. Nitrogen status can be altered with postharvest urea sprays but the response varies with the N status of the tree. Postharvest soil application of labelled N showed N movement into blossoms but not in the flower buds. 相似文献

AIM: To investigate whether two-hit acute lung injury (ALI) model is better than one-hit, and to evaluate the inflammatory response in the lungs during these models by using [8F]FDG microPET. METHODS: Thirty three, adult, male Sprague-Dawley rats weighing 180-210 g were used and divided into 4 groups. Rats in LPS group (n=10) and LPS-HCl group (n=10) were challenged with intraperitoneal administration of LPS at the dose of 5 mg/kg, while rats in NS group (n=3) and HCl group (n=10) received normal saline solution intraperitoneally at the dose of 1 mL/kg, after 16 h, all animals were anesthetized with an intraperitoneal injection of sodium pentobarbital (40 mg/kg) and placed in a 60°inclined position, the femoral artery was cannulated and connected to a pressure transducer to record the arterial pressure on a polygraph recorder, the trachea was surgically exposed. Rats in HCl group and LPS-HCl group received direct intratracheal injection of HCl (pH=1.2) at the dose of 0.5 mL/kg while rats in NS group and LPS group received the same volume of normal saline solution. Blood gas samples (each 0.3 mL) were obtained at 30, 90 and 240 min after the instillation and replaced by the same volume of saline solution, the samples were analyzed using a blood gas analyzer. 240 min after HCl or NS administration, the rats underwent a microPET scanning, then, all the rats were sacrificed and the lungs were obtained for histological analysis. RESULTS: Blood gas analysis showed that rats in LPS-HCl group had higher PaO2 and lower PaCO2 than the other groups. MAP decreased markedly in LPS-HCl group, while MAP in other groups remained stable. The results of microPET showed that the ratio of ROI between the right lung and the muscle tissue of the right arm in LPS-HCl group was 9.00±1.41, and was significantly higher than that in LPS group (4.01±0.60) and HCl group (3.33±0.55). Histological examination showed that the mean lung injury score in LPS-HCl group was 12.70±0.95, while that was 8.40±1.26 in HCl group and 7.00±0.82 in LPS group, and there were significant differences (both P<0.01). CONCLUSION: LPS pretreatment significantly magnifies and prolongs the inflammatory response to the subsequent acid instillation in both lungs. When compared with “one-hit”, “two-hit” is easier to induce the ALI, and [8F]FDG microPET is a useful tool to evaluate the inflammatory reaction during ALI. 相似文献
