sRNA伴侣蛋白Hfq敲除条件下沙门菌的转录组分析

为探明沙门菌(Salmonella)在sRNA伴侣蛋白Hfq敲除条件下转录组变化情况。本试验采用HiSeq测序平台对沙门菌LT2菌株及其Hfq敲除株进行高通量测序,通过DESeq2差异分析方法筛选敲除sRNA伴侣蛋白Hfq条件下的差异表达基因,对其进行生物信息学GO功能显著性富集分析和Pathway显著性富集分析。结果显示,对照组和试验组分别得到12 753 534条、8 254 308条Clean Reads。通过DESeq2差异分析获得差异基因1 055个,其中表达上调基因516个,表达下调基因539个。GO功能显著性富集分析表明显著差异表达基因主要富集在钴胺素代谢过程、钴胺素生物合成过程、碳水化合物代谢过程等生物过程中。Pathway显著性富集分析表明显著差异表达基因富集到细菌趋化性、丙酸代谢和碳代谢等11个信号通路中。结果表明,本试验应用RNA-seq技术丰富了sRNA伴侣蛋白Hfq调控基因数量,筛选出20个受伴侣蛋白Hfq调控的显著差异表达基因,其中4个基因功能及编码蛋白未知,注释了差异表达基因的生物学功能及信号通路,推断Hfq在细菌对数期主要通过影响营养提供和趋化性等途径,继而影响细菌的正常生理活动,为Hfq协同sRNA的调控机理研究及sRNA靶基因的筛选奠定了基础。

杂交鲟维氏气单胞菌的分离鉴定及其耐药性分析

试验旨在对贵州省某养鱼场发病杂交鲟体内分离的优势菌株进行鉴定、致病力探究及耐药性分析。本试验通过革兰染色、生化鉴定、细菌16S rDNA基因分析、动物回归试验、药敏试验、部分毒力基因检测与耐药基因检测。结果显示:分离菌为革兰阴性短杆菌,细菌16S rDNA扩增测序与比对,得大小为1 477 bp的片段,其片段基因序列与GenBank中的维氏气单胞菌序列相似度达99.00%,药敏试验显示:分离菌对环丙沙星、氟苯尼考、头孢哌酮等药物敏感,对四环素、磺胺异唑、苯唑西林药物出现耐药,耐药基因检测显示:分离菌含有耐磺胺基因(Sul1,sul2)、Ⅰ类整合子(Intl1)3种耐药基因,毒力基因检测显示:分离菌含有气溶素(Aer)和粘附素(Aha)两种毒力基因。综合本试验结果,分离菌为维氏气单胞菌(A.veronii),含有毒力基因,并对环丙沙星、氟苯尼考、头孢哌酮等药物敏感。本试验对贵州地区鲟发病流行的病因做出准确诊断,为该地区细菌性鱼病预防与合理用药提供依据。

一株H9N2亚型猪流感病毒的遗传进化和致病性分析

本研究从有流感症状的病猪中分离到一株H9N2亚型猪流感病毒(SIV),命名为A/swine/Jiangsu/1/2015(SW/JS/1/15)。为探究其遗传特征和生物学特性,本研究采用RT-PCR技术扩增其全部基因节段后测序并进行遗传分析,并研究了其对鸡和豚鼠的致病特性。遗传进化分析显示,分离病毒SW/JS/1/15株是由BJ/94系、DK1系、G1系和F/98系4个分支病毒重组而成,8个基因节段均属于G57基因型。分离株HA蛋白裂解位点为PSRSSR*GL,符合低致病性流感病毒的特征。HA蛋白有9个潜在糖基化位点,其中218位糖基化位点缺失,145位与313位各新增一个糖基化位点。与疫苗株SH/F/98、SD/6/96、GD/SS/94相比,分离病毒HA抗原位点发生了G^90E、S^127R、S^145N、D^153G、N^167S、A^168N、A^198T、T^200R、N^201D、和Q^235M(H9numbering)突变;NA蛋白发生6个氨基酸突变:K^367R、K/E^368N、D^369N、D^401E、K^143N和T^434P。同时NA蛋白颈部缺失aa63~aa65。分离病毒的8个基因节段与2株禽源H9N2病毒的相应基因高度同源,其6个内部基因与两株人源H7N9病毒的内部基因高度同源。致病性试验结果显示分离病毒可以感染鸡和豚鼠,但不能在豚鼠群内水平传播,且可能作为H7N9等新型流感病毒内部基因供体,同时表明猪可以感染禽流感病毒(AIV),且可能是AIV获得感染哺乳动物能力的过渡宿主。本研究为H9N2亚型SIV的致病性以及遗传特征的研究提供科学依据。
