4-Nonylphenol (NP) toxicity in fish attracts much attention due to its ability in targeting several organs; however, the researches regarding its potential hepatotoxicity are conflicting and still require further investigation. Therefore, the objective of this study is to focus on this issue from the histophysiological point of view using NP intoxicated African catfish (Clarias gariepinus) as a model of hepatotoxicity. Twelve adult fish (6 per group) were divided into two groups; the first was considered as a control and the second was exposed to NP dissolved in water at a dose of 0.1 mg/kg BW for 3 weeks. A significant reduction in the hepatic alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase levels was observed in NP-exposed fish. Concerning the oxidant/antioxidant balance, a significant depletion in superoxide dismutase, catalase, and glutathione peroxidase was found along with a significant elevation in total peroxide and malondialdhyde. The histopathological examination of the hepatic tissues revealed that NP had marked hepatotoxic effects including hepatitis, centrilobular and focal hydropic and fatty degeneration, fatty change (steatosis), hepatic coagulative necrosis, and nuclear alterations in addition to apoptosis of hepatocytes and necrosis of endothelial cells. Depletion of the glycogen and increased in pigments (lipofuscin and hemosiderin) content in the hepatocytes were also recorded. Hemosiderosis and proliferation of the connective tissue around the blood vessels and branches of bile ducts and in the portal areas were also observed. In light of these findings, it was concluded that NP has a well-defined hepatotoxic impact paving the road towards other studies to investigate other detrimental cyto-physiological influences of this aquatic pollutant. 相似文献

Triploid African catfish (Clarias gariepinus) produced through cold shock 3 min post fertilization were compared to diploid offspring of the same parentage at 66 days
of age. Triploid fish were significantly shorter (11%) and weighed less (18%) than diploids but showed no significant difference
in mortality or cannibalism, which can be an important source of losses under aquaculture conditions. Erythrocytes of triploid
fish were more oval shaped than the normal spherical shape of diploid erythrocytes, were significantly larger (38%) and had
nuclei that were significantly larger (25%) than observed in diploid fish. Erythrocyte morphological characteristics show
potential as an indicator for the detection of triploidy in African catfish. 相似文献

In the current study, African catfish, Clarias gariepinus, was exposed to a sublethal concentration of carbofuran (CF) to investigate its negative effects on biochemical and oxidative stress biomarkers. Also, the putative role of lycopene (LYC) administration in alleviating these negative effects was evaluated. Fish were divided into six groups in triplicates as follows: group I was without treatment, group II was orally administered corn oil, group III was orally administered 18 mg LYC/kg body weight, group IV was exposed to 0.121 mg CF/L, group V was orally administered 9 mg LYC/kg body weight and exposed to 0.121 mg CF/L, and group VI was orally administered 18 mg LYC/kg body weight and exposed to 0.121 mg CF/L for 4 weeks. At the end of this period, blood and tissue (liver and kidney) samples were collected and biochemical and oxidative stress biomarkers were analysed. Also, histopathological changes were determined. Carbofuran caused significant increments of glucose, cortisol, aspartic amino transferase, alanine amino transferase, cholesterol, urea, and creatinine; meanwhile, serum acetylcholinesterase, total protein, albumin, and total lipids were significantly reduced. Significant increments in hepatic and renal malondialdehyde (MDA) and superoxide dismutase (SOD) levels and marked reduction in hepatic and renal catalase (CAT), glutathione (GSH), and total antioxidant capacity (TAC) levels were observed in CF-exposed fish comparing to the control group. Treatment with LYC attenuated the CF-induced oxidative stress, and this improvement was more pronounced in fish received the high LYC dose (18 mg/kg body weight). Further, congestion of the central vein with infiltration of mononuclear inflammatory cells, vacuolar necrosis, and haemorrhage was observed in the livers of CF-exposed fish. Oral administration of LYC reduced behavioural changes and histopathological alterations. All the altered biochemical parameters and antioxidant biomarkers were also restored to be near the normal levels. The obtained results evoked that LYC administration alleviated the destructive effects of carbofuran and reduced its toxicity effect on African catfish. 相似文献

The immediate-early gene (egr-1) expression was used to examine the neuron’s response in telencephalon of goldfish during spatial learning in small space.
Fishes were pre-exposed in the experimental apparatus and trained to pick food from the tray in a rectangular-shaped arena.
The apparatus was divided into identical compartments comprising three gates to provide different spatial tasks. After the
fish learned to pass through the gate one, two more gates were introduced one by one. Fish made more number of attempts and
took longer time (P < 0.05) to pass through the first gate than the gate two or three. This active learning induces the expression of egr-1 in telencephalon as established by western blot analysis. Subsequently, the fish learn quickly to cross the similar type
of second and third gate and make fewer errors with a corresponding decline in the level of egr-1 expression. As the fish learned to pass through all the three gates, third gate was replaced by modified gate three. Interestingly,
the level of egr-1 expression increased again, when the fish exhibit a high exploratory behavior to cross the modified gate three. The present
study shows that egr-1 expression is induced in the telencephalon of goldfish while intensively acquiring geometric spatial information to pass

This study was conducted to investigate the growth performance, biomarkers of oxidative stress, catalase (CAT), superoxide dismutase (SOD), and glutathione S-transferase (GST) as well as the haematological response of African catfish after being fed with fish feed containing different levels of cricket meal. The juvenile fish were assigned to three different treatments with isonitrogenous (35 %) and isoenergetic (19 kJ g?1) diets containing 100 % cricket meal (100 % CM), 75 % cricket meal (75 % CM), and 100 % fishmeal (100 % FM) as control groups for 7 weeks. The results indicated that a diet containing 100 % CM and 75 % CM improved growth performance in terms of body weight gain and specific growth rate, when compared to 100 % FM. The feed conversion ratio (FCR) and protein efficiency ratio (PER) did not differ significantly between all diets, but reduced FCR and increased PER were observed with a higher inclusion of cricket meal. A haematological examination of fish demonstrated no significant difference of red blood cells in all diets and white blood cells showed a significantly higher value in fishmeal-fed fish. On the other hand, haemoglobin and haematocrit significantly increased with increasing amounts of cricket meal in the diet. Antioxidant activity of CAT was higher in the 100 % CM group compared to fish fed other diets, whereas GST and SOD showed increasing trends with a higher incorporation of cricket, although insignificant differences were observed between all diets. These results suggest that cricket meal could be an alternative to fishmeal as a protein source in the African catfish diet. 相似文献

Heat stress is one of the major environmental concerns in global warming regime and rising temperature has resulted in mass mortalities of animals including fishes. Therefore, strategies for high temperature stress tolerance and ameliorating the effects of heat stress are being looked for. In an earlier study, we reported that Nrf-2 (nuclear factor E2-related factor 2) mediated upregulation of antioxidative enzymes and heat shock proteins (Hsps) provide survivability to fish under heat stress. In this study, we have evaluated the ameliorative potential of dietary curcumin, a potential Nrf-2 inducer in heat stressed cyprinid Puntius sophore. Fishes were fed with diet supplemented with 0.5, 1.0, and 1.5% curcumin at the rate 2% of body weight daily in three separate groups (n = 40 in each group) for 60 days. Fishes fed with basal diet (without curcumin) served as the control (n = 40). Critical thermal maxima (CTmax) was determined for all the groups (n = 10, in duplicates) after the feeding trial. Significant increase in the CTmax was observed in the group fed with 1.5% curcumin- supplemented fishes whereas it remained similar in groups fed with 0.5%, and 1% curcumin-supplemented diet, as compared to control. To understand the molecular mechanism of elevated thermotolerance in the 1.5% curcumin supplemented group, fishes were given a sub-lethal heat shock treatment (36 °C) for 6 h and expression analysis of nrf-2, keap-1, sod, catalase, gpx, and hsp27, hsp60, hsp70, hsp90, and hsp110 was carried out using RT-PCR. In the gill, expression of nrf-2, sod, catalase, gpx, and hsp60, hsp70, hsp90, and hsp110 was found to be elevated in the 1.5% curcumin-fed heat-shocked group compared to control and the basal diet-fed, heat-shocked fishes. Similarly, in the liver, upregulation in expression of nrf-2, sod, catalase, and hsp70 and hsp110 was observed in 1.5% curcumin supplemented and heat shocked group. Thus, this study showed that supplementation of curcumin augments tolerance to high temperature stress in P. sophore that could be attributed to nrf-2-induced upregulation of antioxidative enzymes sod, catalase, gpx, and the hsps. 相似文献

ABSTRACT: The present study was conducted to investigate the effects of the red clover extract on growth, body composition and survival of African catfish Clarias gariepinus . Three concentrations (25, 50 and 75 mg/kg) of red clover at dietary were used for 120 days. The final weights of red clover-treated groups were significantly different from each other and the control group. The best growth was observed in the group receiving the 75 mg/red clover kg diet. Fish fed with all levels of red clover had higher weight gains than the control. The highest values of the specific growth rate and the food conversion rate were 1.14 ± 0.01 and 2.26 ± 0.04, respectively, for the 75 mg/red clover kg diet group. During the red clover administration period, the survival ranged from 97.78% for the 50 p.p.m. group to 95.56% for the 25 and 75 p.p.m. groups. Protein contents of the dosage groups were significantly different from control ( P < 0.05). The highest value of protein content (19.97%) was observed for the 75 mg/ red clover kg diet. Lipid and ash contents were significantly affected by the red clover levels in the diets. The highest lipid contents were in the 75 mg/red clover kg diet (4.67%), and the highest ash content was 1.39% for the 25 mg/red clover kg diet. 相似文献

The influence of Tribulus terrestris on the activities of testicular enzyme in Poecilia latipinna was assessed in lieu of reproductive manipulation. Different concentrations of (100, 150, 200, 250, and 300 mg) Tribulus terrestris extract and of a control were tested for testicular activity of enzymes in Poecilia latipinna for 2 months. The testis and liver were homogenized separately in 0.1 mol/l potassium phosphate buffer (0.1 mol/l, pH 7.2).
The crude homogenate was centrifuged, and supernatant obtained was used as an enzyme extract for determination of activities.
The activities of testicular functional enzyme ALP, ACP, SDH, LDH, and G6PDH levels were changed to different extent in treated
groups compared with that of the control. The total body weight and testis weight were increased with the Tribulus terrestris-treated fish (Poecilia latipinna). These results suggest that Tribulus terrestris induced the testicular enzyme activity that may aid in the male reproductive functions. It is discernible from the present
study that Tribulus terrestris has the inducing effect on reproductive system of Poecilia latipinna. 相似文献

Francisella noatunensis orientalis is a bacterium that causes emerging bacteriosis in Nile tilapia (Oreochromis niloticus) in many parts of the world, including Brazil. It is a non-motile, Gram-negative, strictly aerobic, facultative intracellular coccobacillus. This species of bacteria is responsible for low to high mortality in fish farms, causing economic losses for fish farmers. This study aimed to detect the presence of F. noatunensis orientalis using qPCR (real-time polymerase chain reaction) and to describe lesions caused by the bacterium in O. niloticus in Brazilian aquaculture. For this purpose, 360 fish from six fish farms (30 per farm) were sampled at two time points (n = 180 per sampling). Necropsies and histopathology were performed for lesion observation, in addition to qPCR and sequencing for detection and identification of Francisella species. Environmental data were collected using a multiparameter sonde YSI EXO2. All measured limnological variables were within the optimum range for cultivation of Nile tilapia. The major lesions present were melanization of the skin, splenomegaly, granulomas, and inflammatory cell responses. The prevalence of francisellosis varied from 0 to 86.66% between time periods and fish farms analyzed, and an outbreak was observed during the second sampling period. This study describes the prevalence of francisellosis in O. niloticus and reports that the lesions found are not exclusively associated with this bacterial disease. 相似文献

Stress in response to anesthesia with benzocaine, MS-222, metomidate and isoeugenol was studied in Atlantic salmon (Salmo salar), Atlantic halibut (Hippoglossus hippoglossus), and Atlantic cod (Gadus morhua) with no concomitant stress from handling or confinement in association with anesthesia or sampling. All of the anesthetics
tested induced a stress response in all species, displayed by a release of cortisol to the water. MS-222 anesthesia elicited
the highest cortisol release rates, reaching maximum levels 0.5 h post-exposure and returning to basal levels after 3–4 h.
Benzocaine anesthesia caused a bimodal response where the initial peak in cortisol release rate was followed by a second increase
lasting towards the end of the trial (6 h). This bimodality was more profound in Atlantic salmon than in Atlantic halibut
and Atlantic cod. Metomidate anesthesia induced the lowest release of cortisol of the agents tested in both Atlantic halibut
and Atlantic cod, but resulted in a bimodal response in Atlantic salmon where the initial increase in cortisol release was
followed by a larger increase peaking at 2–2.5 h post exposure before returning to basal levels after 5 h. The stress induced
in Atlantic salmon by isoeugenol anesthesia resembled that of MS-222, but did not reach the same elevated level. Overall,
the cortisol release was most profound in Atlantic salmon followed by Atlantic halibut and Atlantic cod. 相似文献

The present study histologically examined the effects of glucan-containing and Lactobacillus rhamnosus GG (LGG)-containing diets on intestinal damages inflicted on Nile tilapia by Aeromonas challenges. Tilapia were fed control, glucan, and LGG diets for 2 weeks and were subsequently challenged with Aeromonas. The intestines were then histologically examined at 1, 7, 14, and 21 days post-infection. Mortality following the challenge
was lower for the fish fed the glucan and LGG diets. The intestines of these groups also showed increased inflammatory cell
infiltration and reduced intestinal damage from Aeromonas. Moreover, inflammatory cell infiltration occurred more rapidly in the glucan-fed than in the LGG-fed fish following the
challenge. Before the challenge, the dominant mucous cell was the acid type in all the tests. After the challenge, the main
mucus cell type in the proximal intestine of the glucan-fed fish shifted to AB-PAS double-staining cells, while in the LGG-fed
fish, it remained the acid type throughout the test period, and the number of double-staining cells was smaller than in the
control fish after the challenge. Thus, the different mucous cell and inflammatory cell responses show that glucan and LGG
might have different immunostimulative effects, although they both reduced the intestinal damage following Aeromonas challenges. 相似文献

Mortality of the short-neck clam Ruditapes philippinarum exposed to the toxic dinoflagellate Heterocapsa circularisquama was studied under controlled conditions to clarify the mechanisms of recurrent mass deaths of clams occurring in western
Japanese coastal areas. One-week mortality tests, involving three water temperatures, six H. circularisquama concentrations, and two clam body sizes, showed a significant increase in mortality with increasing temperature, H. circularisquama concentration, exposure duration, and body size (ANOVA, P < 0.01). Clam death was observed at concentrations as low as 50 cells/ml and temperatures as low as 15°C. Prior to death,
clams showed an extreme retraction of their mantle edge and siphon, along with recurrent vomiting behavior before initiating
a closure reaction followed by paralysis then death. Gills of paralyzed clams showed an important uptake of dye, implying
gill damage. This study is the first laboratory evidence of bivalve mortality induced by H. circularisquama at low concentrations and low temperature, and the first report of differential effects according to the body size of bivalves. 相似文献

To completely replace the fish meal by a mixture of earthworm and maggot meals, experimental diets were tested during 42 days on Clarias gariepinus fingerlings. Five isoproteic and isoenergetic diets (40 % crude protein and 17.9 ± 0.3 kJ g?1) including the control diet (D1) based on fish meal, were formulated. All these diets satisfied the essential amino acids requirements of C. gariepinus fingerlings. These diets were tested on triplicate groups of 50 fishes (initial body weight: 3 ± 0.1 g) bred in tank (0.5 m3). The approximate ratios 2:5; 1:4; 1:12 and 0:1 between the earthworm meal and the maggot meal were used, respectively, to formulate four diets D2, D3, D4 and D5 without fish meal. After the feeding period, significant differences (P < 0.05) were observed on growth, feed utilization between control diet (D1) and test diets (D2–D5). Fish fed earthworm- and maggot-based diets were grown better than those fed the control diet. Survival and feed utilization were not significantly affected by the ratio between earthworm meal and maggot meal in the test diets. Lipid content was higher in carcass and fillet of fishes fed earthworm- and maggot meals-based diets than that of those fed fish meal-based diet. This study indicates that when the ratio 2:5 between the earthworm meal and the maggot meal is used to entirely replace fish meal and the ratio lysine/arginine of the diet is inferior to 1, the growth performances and feed utilization of Clarias gariepinus fingerlings are improved. 相似文献

The effects of two alternative prey (crayfish and hake) were tested on growth and survival of both juveniles and adults of
Octopus vulgaris in two experiments. Octopuses fed the control (squid) were larger (3.0 ± 0.7 kg) than those fed crayfish (2.4 ± 0.6 kg) at
the end of experiment I. Similarly, overall growth rates were higher for octopuses fed squid (1.7 ± 0.3 and 1.2 ± 0.2 %BW day−1, respectively). Average feeding rates for the experiment were not different, being 6.5 ± 0.9 and 7.5 ± 0.9 %BW day−1, respectively, for octopuses fed either squid or crayfish. Nevertheless, food conversions for the experiment were higher
(42.4 ± 2.7%) for octopuses fed squid compared to the ones fed crayfish (23.9 ± 1.9 g). For experiment II, hake and crayfish
were compared to squid; the final weight of octopuses fed squid, hake or crayfish was 1,183.0 ± 242.7 g, 1,175.6 ± 240.1 g
and 922.3 ± 160.1 g, respectively. Overall growth rates for the experiment were 1.9 ± 0.2 %BW day−1, 1.9 ± 0.3 %BW day−1 and 1.1 ± 0.3 g, respectively. Final weight and growth rates were never different (P > 0.05) between octopuses fed squid and hake, but were always higher (P < 0.05) compared to the ones fed crayfish. Average feeding rates for experiment II were similar for the three diets, and
of 4.6 ± 1.5, 4.2 ± 1.3 %BW day−1 and 5.1 ± 0.9 %BW day−1, respectively, for octopuses fed squid, hake or crayfish. Food conversions for experiment II were of 41.0 ± 9.6%, 40.5 ± 9.9%
and 21.3 ± 7.4 g, respectively, for octopuses fed squid, hake or crayfish, and were always higher for octopuses fed squid
and hake compared to crayfish. The results indicate that crayfish is not an adequate replacement for the usual prey to fatten

Reproduction allows organisms to produce offspring. Animals shift from immature juveniles into mature adults and become capable of sexual reproduction during puberty, which culminates in the first spermiation and sperm hydration or ovulation. Reproduction is closely related to the precise control of the hypothalamic–pituitary–gonadal (HPG) axis. Kisspeptin peptides are considered as the important regulator of HPG axis in mammalian. However, the current understanding of kisspeptin in flatfish is not comprehensive. In this study, we cloned and analyzed the kiss2 and kissr2 genes in Cynoglossus semilaevis. Interesting alternative splicing in the 5′-untranslated regions (UTR) of the Cskissr2 gene was found. The expression profiles of Cskiss2 and Cskissr2 showed relative high messenger RNA (mRNA) levels at the late gastrula stage during embryonic development, at total length = 40 mm during early gonadal differentiation, and in the brains and gonads of all investigated tissues. These results suggested that the kisspeptin system participated in embryogenesis and in the regulation of gonadal differentiation and development. Considering that the control and regulatory mechanisms of kisspeptin in the central reproductive axis are still unclear, we documented that the intramuscular injection of kisspeptin caused different sGnRH and cGnRH mRNA levels in a dose- and tissue-dependent manner. The mRNA expressions of FSH and LH were stimulated in the ovary and were inhibited in the testis under the kisspeptin treatments. These results provided foundations for understanding the roles of kisspeptin in the neuroendocrine system in fish. The manipulation of the kisspeptin system may provide new opportunities to control the gonadal development and even reproduction in fish. 相似文献

Cadmium (Cd) compounds are widely distributed toxic environmental and industrial pollutants, and they may bring danger to
growth and development of aquatic organisms. In China, the Oujiang color common carp, Cyprinus carpio var. color, is a very important fish, from an economic point of view, and is well used for fish culture in paddy fields. The purpose
of this study was to show the low concentrations of cadmium-induced oxidative stress response and DNA damage in the livers
of Cyprinus carpio var. color. Superoxide dismutase (SOD), malondialdehyde (MDA), and glutathione (GSH) in liver were measured after exposure to Cd levels
(0.41, 0.52, 0.69, 1.03 and 2.06 mg/L, respectively) for 7 days and compared with the control groups. DNA damage, including
indicators of damage percentage, DNA tail length (TL) and DNA tail moment (TM) were also analyzed by comet assays. Results
showed that MDA and GSH levels in all treatment groups increased significantly relative to the controls (P < 0.01). Treatment with Cd at concentration of 0.41 mg/L increased SOD activity, while treatment with Cd at concentrations
>0.41 mg/L inhibited SOD activities. DNA damage percentage, TL and TM also significantly increased when the Cd level was >0.41 mg/L.
Positive correlations were also found between DNA damage levels and MDA levels (r = 0.74 for DNA damage percentage, r = 0.83 for TL, r = 0.84 for TM; P < 0.01 for all) as well as between GSH and MDA levels (r = 0.77, P < 0.01). These results strongly suggested that Cd-induced DNA damage in the livers of Cyprinus carpio var. color was due to lipid peroxidation and oxidative stress. 相似文献

The ability of an algicidal bacterium Marinobacter salsuginis strain BS2, isolated from shrimp pond water, to reduce shrimp mortality was investigated under laboratory conditions. When
two species of shrimp (Penaeus monodon and Litopenaeus vannamei) (body length 1.5–1.8 cm) were cultured together with the dinoflagellate Noctiluca scintillans, nearly 80 % of the shrimps died within 7 days. However, when bacterial strain BS2 was also added to the culture, N. scintillans was killed within 48 h, and shrimp survival rates on the 7th day improved from 23 to 87 % for both P. monodon and L. vannamei. The bacterium BS2 alone had no effect on shrimp condition. Under conditions of increased dissolved oxygen, the effect of
using BS2 was greater, and shrimp survival rates improved even more dramatically, from 26 to 98 %. These studies provide the
first evidence that the use of killing bacteria, isolated from shrimp culture water, can suppress harmful algal blooms (HABs)
and thus restore the efficiency of shrimp production. The control of HABs in this way in shrimp culture farms would be a major
benefit for shrimp production. 相似文献
