In some areas of Japan, yellow spots with white pustules on leaves, stems, petioles, peduncles and calyces were found on Ipomoea nil, I. triloba, I. lacunosa and I. hederacea var. integriuscula. We demonstrated that the diseases on I. nil, I. triloba and I. lacunosa were caused by host-specific strains of Albugo ipomoeae-panduratae and defined three forma speciales of the fungus, respectively, for the three Ipomoea species: “f. sp. nile”, “f. sp. trilobae” and “f. sp. lacunosae”. Because the diseases were new to Japan, we coined the Japanese name “shirosabi-byo”, which means white rust. We also showed that the disease on I. hederacea var. integriuscula was caused by A. ipomoeae-hardwickii. We named this new disease “white rust (shirosabi-byo in Japanese)”.  相似文献

Indoor and field experiments were conducted to evaluate the efficacy of applying the mycoparasite Coniothyrium minitans to the aerial parts of rapeseed plants at the flowering stage to control sclerotinia diseases caused by Sclerotinia sclerotiorum. Under controlled conditions, a petal inoculation technique was used to determine the effect of conidial suspensions of C. minitans on suppression of sclerotinia leaf blight. Results showed that C. minitans was effective in inhibiting infection initiated by ascospores of S. sclerotiorum on flower petals by restricting mycelial growth of the pathogen. Suppression of lesion development was related to the conidial concentration of C. minitans, with larger lesions at low concentration (5×103conidia ml−1), but smaller lesions at high concentration (5×104 conidia ml−1 or higher). When C. minitans-treated rapeseed leaves were inoculated with mycelia of S. sclerotiorum, C. minitans failed to prevent infection of leaves, but caused a significant reduction in number of sclerotia produced on the diseased leaves. No significant difference in efficacy was detected between the two isolates of C. minitans, LRC 2137 and Chy-1, on the two rapeseed cultivars, Westar (spring type) and Zhongyou 821 (winter type). Results of field trials showed a significant reduction of stem rot of rapeseed in four (1997, 1999, 2003 and 2004) out of five years by aerial application of C. minitans, compared with controls. No significant difference in suppressive efficacy was observed between the treatments of C. minitans (106 conidia ml−1), C. minitans (106 conidia ml−1) + benomyl (50 μg ml−1) and benomyl (100 μg ml−1) in 2003, and between the treatments of C. minitans (106 conidia ml−1), C. minitans (106 conidia ml−1) + vinclozolin (100 μg ml−1) and vinclozolin (500 μg ml−1) in 2004. Sclerotia of S. sclerotiorum collected from diseased plants in plots treated with C. minitans in 1999, 2000 and 2003, or with C. minitans + benomyl in 2003 were infected by C. minitans at frequencies ranging from 21.3 to 54.5%. This study concludes that aerial spraying of C. minitans is an effective method for controlling sclerotinia diseases of rapeseed.  相似文献

 从河南省10个地市采集的小麦病根样品中分离得到82株病原分离株, 通过形态特征观察、回接致病性测定、以及分子生物学的方法对所分离菌株进行鉴定。其中47个菌株菌丝分支处都形成典型的“∧”状;子囊壳埋生或半埋生, 子囊棍棒状, 子囊孢子线形, 稍弯曲, 无色, 具有5~10个分隔, 大小为68~94 μm×2~4 μm;对82个分离株rDNA-ITS区进行PCR扩增和序列测定, BLAST序列比对结果表明, 47株菌株与小麦全蚀病菌的ITS序列同源性达97%~99%, 据此确定47株菌株为小麦全蚀病菌。应用小麦全蚀病菌4个变种的特异性引物进行PCR扩增都得到禾顶囊壳小麦变种(870 bp)的特异性片段, 鉴定47株菌株均为禾顶囊壳小麦变种 (Gaeumannomyces graminis var.tritici)。  相似文献

Surveys of 94 artichoke fields throughout the artichoke production areas of Comunidad Valenciana (eastern Spain) were conducted from 1999 to 2002 to determine the incidence and distribution of Verticillium wilt.Verticillium dahliae was isolated from 80.9% of the sampled fields, and detected in all artichoke-growing areas, with a mean disease incidence of 53.8% infected plants. The disease was found to cause severe damage to cv. ‘Blanca de Tudela’, which is the most important artichoke cultivar grown in Spain, and was also observed on the seed-propagated cv. ‘Imperial Star’. In field trials to study the role of infected planting material and soil inoculum on infection of artichoke plants during the cropping season,V. dahliae was transmitted from infected stumps to the plants, confirming that the use of infected stumps could have greatly contributed to the dissemination of the pathogen. Inoculum density ofV. dahliae in soil had an effect on crop infection, in that a higher number of microsclerotia per gram of soil resulted in a higher percentage of infected plants. In addition, yield of cv. A study was conducted to determine the feasibility of using sclerotia ofSclerotinia sclerotiorum for producing conidia ofConiothyrium minitans in liquid culture. The medium (SST) was made of water containing 2.0, 1.5, 1.0 or 0.5% (w/v) ground sclerotia ofS. sclerotiorum and 100 μgl −1 thiamine hydrochloride (HCl). One ml of conidial suspension (2 × 107 conidia ml−1) ofC. minitans LRC 2534 was inoculated into 100 ml of SST medium or control (thiamine HCl in water) and incubated at 20 ± 2°C on a shaker at 200 rpm. Subsamples were removed periodically and examined under a compound microscope. Conidia in the SST media germinated within 24 h, developed into branched hyphae within 48 h, produced pycnidia after 3–4 days, and the pycnidia released mature conidia after 7 days. Production of conidia varied with the concentration of sclerotia in the SST medium. It was lower (3.6 × 106 conidia ml−1) at 0.5% but higher (1.2 × 108 conidia ml−1) at 2%. The new conidia were viable and the colonies developing from them showed the original morphological characteristics. The mode of action of soluble silicon against strawberry powdery mildew (Sphaerotheca aphanis var. aphanis) was investigated in four experiments. First, silicon-treated leaves from plants grown with silicate (Si+) and control leaves were excised, inoculated with conidia, and subsequent germination and formation of appressoria in a petri dish was assessed after 24 h. The germination rate was 49.7% on Si+ leaves, and was 67.2% on control leaves (t-test, P < 0.01). Second, we soaked cellulose membranes in various solvents and then placed the membranes on 4% water agar, dusted the membranes with conidia, and examined after 12 h. No difference was apparent between any treatment and the control (distilled water). Third, strawberries growing hydroponically with additional silicon in the medium were inoculated with conidia, and leaves were observed with a scanning electron microscope 1–2 days after inoculation. Germ tubes and secondary hyphae were shorter and had fewer branches on Si+ leaves than on the control. Moreover, penetration appeared to be inhibited. Fourth, the cuticle was separated from leaves from plants grown as in the third experiment, placed on water agar, and dusted with conidia. Germination of conidia, observed with a light microscope, on Si+ leaves was suppressed markedly to 40%–60% of that of the control. These results suggested that soluble silicon induced physiological changes in the cuticle layer after absorption by the plant. In addition, soluble silicate reduced germination of conidia, formation of appressoria, and possibly the penetration of powdery mildew.  相似文献

In October 2004, Pythium ultimum var. ultimum was isolated from rotten stems of cabbage plug seedlings in a commercial nursery in Mie Prefecture (Japan). The isolated fungus was then used to inoculate seedlings and subsequently reisolated from the seedlings with the damping-off disease, showing that P. ultimum var. ultimum is a new pathogen causing cabbage seedling disease.  相似文献

The greater wax moth Galleria mellonella L. (Lepidoptera: Pyralidae) is occasionally found in beehives and is a major pest of stored wax. Entomopathogenic fungi have recently received attention as possible biocontrol elements for certain insect pests. In this study, 90 isolates of Beauveria bassiana and 15 isolates of Metarhizium anisopliae were screened for proteases and lipases production. The results showed significant variations in the enzymatic action between the isolates. In the bioassay, the selected isolates evinced high virulence against the 4th instar of the G. mellonella larvae. The isolates BbaAUMC3076, BbaAUMC3263 and ManAUMC3085 realized 100% mortality at concentrations of 5.5 × 106 conidia ml−1, 5.86 × 105 conidia ml−1, and 4.8 × 106 conidia ml−1, respectively. Strong enzymatic activities in vitro did not necessarily indicate high virulence against the tested insect pest. The cuticle of the infected larvae became dark and black-spotted, indicating direct attack of fungus on the defense system of the insects. The LC50 values were 1.43 × 103, 1.04 × 105 and 5.06 × 104 for Bba3263AUMC, Bba3076AUMC and Man3085AUMC, respectively, and their slopes were determined by computerized probit analysis program as 0.738 ± 0.008, 0.635 ± 0.007 and 1.120 ± 0.024, respectively.  相似文献

A total of 74 mass isolates of cucumber powdery mildew fungus (Podosphaera xanthii) were collected from commercial greenhouses with a history of boscalid use in Ibaraki Prefecture, Japan, and tested in a leaf disk assay for their sensitivity to boscalid. The mildew development of 40 of 74 isolates and five sensitive reference isolates on the disks was completely suppressed at 5 μg boscalid/ml. The minimum inhibitory concentrations (MIC) for the remaining 34 isolates were 50 μg/ml or higher, and 21 of these isolates also grew well at 500 μg/ml. Six single-spore isolates were resistant to boscalid with MIC values higher than 500 μg/ml; four of these were moderately resistant (MR), and two were very highly resistant (VHR) isolates. The growth of MR isolates was almost completely suppressed at 500 μg/ml, whereas two VHR isolates grew vigorously at 500 μg/ml. In foliar inoculation tests of potted cucumber plants, the efficacy of boscalid (500 μg/ml) against both MR and VHR isolates was very low. Partial DNA fragment of the iron–sulphur protein subunit (SdhB) gene of succinate dehydrogenase was PCR-amplified from five sensitive and five resistant isolates and directly sequenced, revealing that VHR isolates possess a substitution from a highly conserved histidine (CAT) to tyrosine (TAT) in a third cysteine-rich center of a putative SdhB, whereas MR isolates so far have not been found to have such substitution in SdhB.  相似文献

以生物学相容性较好的聚乙二醇辛基苯基醚为乳化剂,从五大类生化营养基质中筛选出海藻糖、葡萄糖、蛋白胨、酪氨酸和菜籽油作为孢子萌发促进剂的基本成份,采用5因子正交旋转组合设计并实施了36个组配试验以优化配方。将所获孢子萌发促进剂加入球孢白僵菌(Beauveria bassiana)孢子乳悬液中喷雾接种桃蚜(Myzus persicae)无翅成蚜,在20℃和12L:12D的条件下饲养并逐日观察发病死亡数。所获数据经时间-剂量-死亡率模型模拟分析,桃蚜接种后第4~6天为死亡高峰,在10~6~10~8个孢子/ml的浓度下喷雾处理6天后的累计死亡率为62.9%~88.7%,LT_(50)估计值为3.8~4.8天,接种后第4、5和6天的LD_(50)估计值分别为3.75×10~7、3.73×10~5和2.08×10~5个孢子/ml。在连续6天的观察中,接种成蚜日均产若蚜数比对照分别下降了40.7%~85.0%,下降幅度随孢子乳悬液浓度提高而增大。这些结果表明,以上孢子萌发促进剂具有作为球孢白僵菌孢子制剂添加剂的应用潜力。  相似文献

The antifungal activities of hyoscyamine and scopolamine, major alkaloids extracted from the desert plant Hyoscyamus muticus, against two rice pathogens, Magnaporthe oryzae and Rhizoctonia solani, were studied. The minimum inhibitory concentration of hyoscyamine that resulted in distinctive inhibition (MIC50) was 1 μg/ml for both fungi. Exposure to hyoscyamine caused the leakage of electrolytes from the mycelia of both fungi. Hyoscyamine (>1 μg/ml) irreversibly delayed or inhibited conidial germination and appressorium formation in M. oryzae grown on polystyrene plates. Hyoscyamine effectively inhibited the attachment of conidia to the surface of rice (Oryza sativa) leaves and inhibited appressorium formation on the leaves. A high concentration of scopolamine (1000 μg/ml) also delayed or inhibited conidial germination in M. oryzae, but conidial germination was restored after washing the conidia with water. Antifungal activity of hyoscyamine was reduced by scopolamine. Magnaporthe oryzae infection was significantly suppressed (by >95%) in leaves of intact rice plants treated with hyoscyamine (10 μg/ml). Moreover, 10 μg hyoscyamine/ml significantly reduced the disease severity index for sheath blight to ≤0.2, when compared with the disease index of control plants (>7.0). Hyoscyamine (>20 μg/ml) completely inhibited sclerotial germination and development of R. solani by delaying the initiation, maturation, and melanization of the sclerotia. These results suggest that tropane alkaloids may be useful for controlling blast and sheath blight diseases of rice and for studying the mechanisms that regulate conidial germination in M. oryzae and sclerotial germination and development in R. solani.  相似文献

A new bacterial disease of tall goldenrod (Solidago altissima L., “Seitaka-awadachiso” in Japanese), one of the most serious weeds in non-agricultural land, was discovered in Ibaraki Prefecture, Japan. Characterized by angular or round, dark brown necrotic spots on leaves, this disease resulted in defoliation and terminal dieback of the plants in severe cases. The disease was named “bacterial leaf spot”. The causal bacterium was identified as Pseudomonas syringae based on its bacteriological properties including those determined by LOPAT tests. The present bacterium was pathogenic to tall goldenrod alone but not to many other tested plants including weeds, flowers, trees and crops. In addition, P. syringae pv. syringae and other pathovars did not show any pathogenicity to tall goldenrod. Because no pathovars of P. syringae pathogenic to tall goldenrod have been reported, the present bacterium was concluded to be a new pathovar of P. syringae. We propose the name P. syringae pv. solidagae pv. nov. , and strain Sei 1 (MAFF 810063) is designated as the pathotype strain and has been deposited in the MAFF collection with two reference strains (MAFF 810064 and MAFF81066).  A single conidium of tomato powdery mildew was isolated from heavily infected leaves of tomato (cv. Moneymaker) grown in the greenhouse of Kinki University, Nara Prefecture, Japan. It was successively multiplied so the morphological and taxonomic characteristics of the pathogen and its host range under high humidity conditions could be analyzed. The isolate KTP-01 of the tomato powdery mildew optimally developed infection structures at 25°C under continuous illumination of 3500 lx. More than 90% of the conidia germinated and developed moderately lobed appressoria. After forming haustoria, the pathogen elongated secondary hyphae from both appressoria and conidia. The hyphae attached to leaf surfaces by several pairs of appressoria and produced conidiophores with noncatenated conidia. In addition to its morphological similarity to Oidium neolycopersici, the phylogenetic analysis (based on the sequence of internal transcribed spacer regions of rDNA) revealed that KTP-01 could be classified into the same cluster group as O. neolycopersici. In host range studies, KTP-01 produced abundant conidia on the foliage of all tomato cultivars tested and tobacco (Nicotiana tabacum), and it developed faint colonies accompanied by necrosis on leaves of potato (Solanum tuberosum), red pepper (Capsicum annuum), petunia (Petunia × hybrida), and eggplant (S. melongena). The pathogen did not infect other plant species including Cucurubitaceae plants, which have been reported to be susceptible to some foreign isolates. Thus, the present isolate of the tomato powdery mildew was assigned as O. neolycopersici, a pathotype different from foreign isolates of the pathogen. This study investigates the infection process of Phoma koolunga on field pea (Pisum sativum) stems and leaves using different susceptible and resistant pea genotypes for each tissue, viz. 05P778‐BSR‐701 (resistant) and 06P830‐(F5)‐BSR‐5 (susceptible) for stems and ATC 866 (resistant) and ATC 5347 (susceptible) for leaves. On both resistant and susceptible genotypes, light and scanning electron microscopy showed P. koolunga conidia infect stem and leaf tissues directly via appressoria or stomatal penetration, but with more infections involving formation of appressoria on stems than on leaves. On leaves of the resistant genotype, at 72 h post‐inoculation, P. koolunga penetrated more frequently via stomata (5.2 conidia per 36 893 μm2) than by formation of appressoria (1.8 conidia); yet no such difference was observed on stems of the resistant genotype. In contrast, at the same time point, the number of conidia infecting the susceptible genotype by formation of appressoria on either stems (135 conidia) or leaves (11.3 conidia) was significantly greater than via stomata (8 and 7.3 conidia, stems and leaves, respectively). Mean germ tube length of germinating P. koolunga conidia on both stems (29.8 μm) and leaves (32.9 μm) of the resistant genotype was less than on the susceptible genotype (40.5 and 63.7 μm, stem and leaves, respectively). In addition, there were differences in the number of germ tubes emerging from conidia on resistant and susceptible genotypes. These are the first insights into the nature of leaf and stem resistance mechanisms operating in field pea against P. koolunga.  相似文献

In March 2006, stored fruits of the medium-to-late-ripening citrus variety Shiranuhi ([Citrus unshiu × C. sinensis] × C. reticulata) were found to have a disease similar to blue mold. The fungus causing this disease differed distinctly from the well-known, blue mold agent, Penicillium italicum, because it formed whisker-like coremia measuring 1–8 mm. On the basis of the morphological characteristics and the phylogenetic analysis based on the nucleotide sequence of the β-tubulin gene, the fungus was identified as P. ulaiense. This is the first report of citrus whisker mold caused by P. ulaiense in Japan.  相似文献

Isolates of Diplocarpon mali, causal fungus of apple blotch, collected from four prefectures in Japan in 1997–1998 were tested for sensitivity to thiophanate-methyl. Results from mycelial growth tests showed that MIC values of the fungicide were 0.19 μg/ml against all isolates from Akita, Nagano and Saga prefectures but 100 or 200 μg/ml against all isolates from the Tokusa area in Yamaguchi prefecture. Detached apple leaves sprayed with the fungicide developed severe symptoms when inoculated with the isolate from Tokusa, but developed no symptoms with the isolate from Nagano. 研究了云南烟草黑胫病菌在不同培养基、不同温度的生长情况及不同菌龄与不同诱导时间孢子囊的产生数量。结果表明,菌株在8种供试培养基中,以燕麦培养基和选择性培养基上菌落生长最好,其次为胡萝卜培养基,在番茄汁和马铃薯培养基上生长最差。同一菌株在不同培养基上产孢量不同、产生的孢子囊大小亦有所不同。05P226菌株在玉米培养基上产孢量最高;05P55菌株在燕麦培养基上产孢量最高,在番茄汁培养基上产生的孢子囊最大,而05P62菌株在燕麦培养基上产孢量最高,在烟叶汁培养基上产生的孢子囊最大。在30℃菌丝生长速率最高,其次为28℃,以35℃生长最差。供试3个菌株间生长差异显著,以05P226生长最好,05P55菌株次之,05P62生长最差。05P226、05P55菌株皆以培养14d、诱导72h产孢量最高,最适宜试验中产孢培养。  相似文献

从褐带长卷叶蛾(Homona coffearia Nietner)幼虫虫尸中分离到一种颗粒体病毒。其颗粒体为椭圆形或卵形,大小约156~194×250~325毫微米。颗粒体在0.01M Na_2CO_3+0.05M NaCl 溶液中,能游离释放出杆状病毒粒子。其大小约62.5×275毫微米。实验室和田间试验结果证明,该病毒对褐带长卷叶蛾有很高的致病力;但对拟小黄卷叶蛾、拟后黄卷叶蛾及家蚕均不致病。  相似文献

Leaf spots on the lower leaf surface and sunken spots on the fruit of kiwifruit (Actinidia deliciosa and A. chinensis) were found in Fukuoka Prefecture, Japan. The observed fungus was identified as Pseudocercospora actinidiae Deighton. We propose the name sooty spot of kiwifruit (“susuhan-byo” in Japanese) as a new disease fullfilling Koch’s postulates.  相似文献
