进境玉米种子携带玉米褪绿斑驳病毒的检测与鉴定   总被引:3,自引:0,他引:3  
 Imported maize seeds were planted in a quarantine greenhouse and two seedlings with chlorotic mottle symptoms were observed. The ELISA results showed that the two seedlings reacted positively with antibody against Maize chlorotic mottle virus (MCMV). The positive samples were further identified by RT-PCR method and the 711 bp size target bands were amplified specifically. The similarity range of nucleotide sequence of both RT-PCR product and six MCMV coat protein genes was 97%-99%. The amino acid sequence homology was 97.88%-99.89%. Based on the above results, the virus was identified as MCMV. It was the first time that MCMV was intercepted from imported maize seeds.  相似文献

侵染葫芦的黄瓜绿斑驳花叶病毒广西分离物分子鉴定   总被引:1,自引:0,他引:1  
从广西南宁市郊温室大棚中的葫芦[Lagenaria siceraria(Molina)Stand.]上采集到一个表现脉绿、花叶症状的病毒样品,ELISA检测表明,该样品与黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)有密切的血清学关系,利用RT-PCR方法从样品中扩增获得约500bp的DNA片段,序列分析表明,该片段是CGMMV的外壳蛋白基因,暂将该病毒分离物定名为GX-BG。外壳蛋白基因核苷酸序列系统进化树分析表明,已报道的CGMMV主要分为3大群体,GX-BG与中国辽宁分离物(CGMMV-LN)分别属于不同的群体。  相似文献

甘薯羽状斑驳病毒外壳蛋白基因的分子变异   总被引:4,自引:0,他引:4  
应用单链构象多态性(single-strand conformation polymorphism,SSCP)技术结合核苷酸序列测定的方法,对我国甘薯主产区11个省份的甘薯羽状斑驳病毒(Sweet potato feathery mottle virus,SPFMV)外壳蛋白(CP)基因的分子变异情况进行了研究.结果表明,SPFMV CP基因的RT-PCR产物表现了较丰富的图谱类型,50个分离物共产生9种主要的SSCP带型;对显示不同带型的20个样品的CP基因进行了序列测定和进化树分析,CP基因核苷酸序列一致性为77.2%~99.9%.说明这些样品的SPFMV的CP基因存在较大的分子变异,可划分为EA、RC、O和C4个株系.  相似文献

A luteovirus isolate infecting chickpea in Morocco was experimentally transmitted by Myzus persicae to Physalis floridana, on which it produced mild symptoms. When tested in western blots against antisera to known legume luteoviruses, this isolate reacted strongly to beet western yellows virus (BWYV) antiserum, moderately to bean leafroll virus antiserum, while no reaction was recorded with the antiserum against subterranean clover red leaf virus. In PCR, a fragment of ca. 950 bp was amplified, comprising the 3' end of the open reading frame (ORF) 3, the complete coat protein gene (ORF 4), and the non-translated region in between these ORFs. The nucleotide sequence of the amplified fragment showed high similarity with BWYV (approximately 96%), and lower (50–60%) with other luteoviruses reported to infect legumes. On the basis of these data, the Moroccan isolate was identified as BWYV. This is the first molecular evidence for the occurrence of BWYV on chickpea in Morocco, and on food legumes in general in North Africa.  相似文献

The relationship between red clover mottle virus (RCMV) isolated in the Ukraine (designated RCMV-Uk) and well-characterised strains from Sweden has been investigated. Nucleic acid hybridisation indicate that both RNAs from RCMV-Uk are highly homologous to their counterparts from RCMV strain S, a conclusion supported by protein sequence analysis of the two viral capsid proteins. Nucleic acid sequence analysis of a portion of RCMV-Uk RNA2 confirmed the high degree of similarity between RCMV-Uk and RCMV strain S. This information suggests that RCMV-Uk should be considered an isolate of RCMV strain S.  相似文献

 Peanut mottle virus(PeMoV) was detected via RT-PCR from two peanut samples(QD5 and QD6) with mottle symptom collected from Qingdao, Shandong Province.The 3'-terminal 892 bp fragments of their genome were cloned and sequenced.The cp genes of QD5 and QD6 were 837 bp in length and encoded 278 amino acids(aa), with DAA at aa sites regulating aphid transmission.QD5 and QD6 shared nucleotide identities of 95.3%-99.4% and aa identities of 93.5%-99.6% in cp genes with other PeMoV isolates available in the GenBank.The phylogenetic results showed that PeMoV were clustered to three groups, America, Asia and Australia, which were consistent with their geographical origins.This is the first molecular evidence on the incidence of PeMoV in China.  相似文献

Two viruses that frequently occur in many Lilium species are Lily mottle virus (LMoV) and Cucumber mosaic virus (CMV), which usually co-infect lilies causing severe disease symptoms. Recent reports have revealed that the viral coat protein (CP) affects chloroplast ultrastructure and symptom development. This study used western blot analysis to confirm that in leaves infected by mixed virus infections of LMoV and CMV, CPs of both viruses were accumulated in lily chloroplasts. Immunogold labelling further demonstrated that both the LMoV CP and CMV CP were localized in the stroma and the thylakoid membranes of the chloroplasts. In addition, it was found that CPs of both viruses were rapidly transported into isolated, intact chloroplasts (in vitro), and their transport efficiencies were positively related to CP concentrations. The lowest transmembrane concentration of CMV CP decreased from 38 μg mL−1 recorded in the single CMV CP import system to 10 μg mL−1 in the mixed import system of LMoV CP and CMV CP. CPs of both viruses exhibited species selection in their transmembrane transport into chloroplasts. This is the first report that the CPs from two viruses (LMoV and CMV) are simultaneously present in lily chloroplasts. Accumulation of high levels of LMoV CP and CMV CP inside the chloroplast appears to contribute to a synergistic interaction inducing the development of mosaic symptoms.  相似文献

 马铃薯Y病毒(potato virus Y,PVY)主要侵染马铃薯和烟草等茄科作物,给世界农业造成巨大经济损失。本文对测定的23个及GenBank中注册的52个中国PVY分离物ORF序列进行了系统发育、重组和选择压等分析。系统发育分析表明,根据ORF序列可把我国75个PVY分离物和国外30个参比分离物分成O、C、E、NTN-NW(SYR-I型)、NTN-NW(SYR-II型)、NTN(NTN-a型)、NTN(NTN-b型)、NA-N/NTN、Eu-N、N-Wi(N:O型)和N-Wi(N-Wi型)等11个分子株系,其中中国PVY分离物属于除E和C株系外的9个分子株系。除ME162、guiyang、PVYzu、SD-G、WA-13和CN:JL-1:17等 6个分离物基因组中未检测到重组,其余69个分离物均存在明显重组。根据重组位点的不同,中国PVY可分为11种重组类型,其中5种为新的重组类型。选择压分析表明,中国PVY分离物的11个基因均处于负选择,其中核内含体b基因受到的选择压最大,PIPO受到的选择压最小。基因流分析表明,黑龙江、河南和山东PVY分离物间基因交流频繁,马铃薯与烟草PVY分离物之间基因交流频繁。本研究的结果明确了中国PVY分离物的分子株系组成,对指导PVY的检测和防控具有积极作用。  相似文献

 多花黑麦草斑驳病毒(Ryegrass mottle virus,RGMoV)属南方菜豆花叶病毒属,是牧草病毒病的重要病原。利用反转录技术合成和扩增了RGMoV RNA的cDNA。将cDNA克隆在载体pUC18上,对RGMoV的基因组RNA作序列分析。RGMoV RNA是一个正义单链RNA分子,全长4 210个核苷酸,包含4个开放阅读框架5'非翻译区包含99个核苷酸,3'非翻译区有198个核苷酸。RGMoV ORF的结构和南方菜豆花叶病毒以及水稻黄斑病毒的结构一样。ORF1编码1个14.6 kD的蛋白质,这个蛋白质的功能还不清楚;ORF2的产物由947个氨基酸组成,分子量为103.6 kD;ORF3编码1个19.8 kD的蛋白质;ORF4编码1个25.6 kD的蛋白质,通过N末端氨基酸分析,确认ORF4为病毒外壳蛋白。RGMoV RNA的全序列以及染色体结构已被GenBank登录,登录号分别为AB040446和NC_003747。  相似文献

通过病毒dsRNA分离技术、非序列依赖性PCR扩增(sequence-independent amplification,SIA)技术和RTPCR等手段对采自山西太谷的西瓜病样进行了检测与鉴定,确定其为黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)所侵染。为明确CGMMV西瓜分离物(CGMMV-SXTG)的分类地位,本研究进一步克隆了CGMMV-SXTG的外壳蛋白基因(coat protein,CP)全序列并进行序列分析,系统进化树分析显示该分离物与亚洲分离物亲缘关系较近,而与欧洲分离物亲缘关系较远,表明该病毒的不同分离物在分组上可能与地域有一定的关系;接着对不同分离物的CP基因核苷酸序列和氨基酸序列进行同源性比对分析,结果表明该分离物与中国山东分离物(TANG)同源性最高,分别达到99.8%和99.6%。  相似文献

 通过高通量测序和RT-PCR扩增,克隆并分析蟹爪兰X病毒(Zygocactus virus X,ZyVX)贵州分离物ZyVX-GZ基因组序列。对表现病毒病症状的红肉火龙果植株进行高通量测序,获得4条ZyVX相关contig。RT-PCR扩增并拼接获得ZyVX-GZ基因组,全长为 6 567 nt,5′-UTR和 3′-UTR分别为60 nt和70 nt。含有5个ORF,分别编码复制酶蛋白、TGB1、TGB2、TGB3蛋白和外壳蛋白。ZyVX-GZ与P39和B1分离物基因组序列一致性均为91%。TGB1-3、外壳蛋白基因与大多数分离物核苷酸和推导的氨基酸序列一致性分别为95%~99%和96%~100%;复制酶基因的核苷酸和推导的氨基酸序列一致性分别为80%~89%和92%~97%。系统进化分析表明,ZyVX群体的遗传分化与寄主种类、地理分布不存在相关性。本研究结果丰富了ZyVX群体的基因组序列信息,为ZyVX的监测和防控提供了基础。  相似文献

 首次用PCR方法,自大麦条纹花叶病毒中国株(BSMV-CH)的RNA2(GenBank登录号为:AY789694,未报道)中扩增出外壳蛋白(coat protein,CP)基因,并亚克隆到pMD18-T载体上进行测序分析。结果表明BSMV-CH的CP全长597bp,它与BSMV其它株系CP的核苷酸同源性和氨基酸同源性分别为93.1%~93.8%和91.4%~92.4%;与同属的早熟禾半潜病毒(PSLV)和剪秋罗环斑病毒(LRSV)的核苷酸同源性分别为53.0%和41.8%,氨基酸同源性分别为48.1%和40.0%。根据大麦病毒属病毒已经报道的CP氨基酸序列绘制系统发育树,分析表明分离的各毒株在进化上存在明显地理位置的相关性。把CP基因亚克隆到GST融合表达载体pEGX-6p-1上,优化IPTG诱导终浓度后,在37℃ IPTG终浓度为1.0mmol/L时,融合蛋白GST-CP在大肠杆菌BL21中得到了特异表达。12% SDS-PAGE表明,表达产物大小为48.5kDa,主要以包含体形式存在。对蛋白进行定量后,用冰冷的KCl显色,分离表达的蛋白质特异条带制备抗原,免疫家兔得到抗血清,酶联法(enzyme-linked immunosorbant assay,ELISA)测定的抗血清效价为1/6 400。用抗血清对感病大麦和健康大麦进行检测,结果表明抗血清有很高特异性。  相似文献

Cowpea mild mottle virus (CMMV) has physicochemical properties typical of carlaviruses, but has remained unclassified due to a number of unusual properties, including no serological cross-reaction with 18 carlaviruses; production of brush-like inclusion bodiesin vivo; and the ability to be transmitted by whiteflies (Bermisia tabaci). In this paper we report the use of a carlavirus specific PCR primer to identify CMMV as a member of the carlavirus group. This is confirmed by nucleotide sequence (958 nucleotides) from the 3 terminal region of CMMV RNA which contains a partial open reading frame (ORF) having high similarity with the coat proteins of other carlaviruses. The sequence also contains an 11.7K ORF at the 3 terminus, containing a zinc-finger motif which is unique to carlaviruses.  相似文献

The cause of a streak disease of pearl millet (Pennisetum glaucum), originating from Nigeria, has been attributed to a geminivirus belonging to the African streak virus cluster. A full-length, infectious clone of the virus was obtained which was transmissible by the vectorCicadulina mbila (Naudé). Analysis of the complete nucleotide sequence of the coat protein gene of this virus shows it to be most closely related to sugarcane streak virus. The possible evolutionary implications of this finding are discussed.  相似文献

 从河北、北京、山东泰安和潍坊等地区的萝卜、大白菜、甘蓝、油菜上得到8个芜菁花叶病毒(Turnip mosaic virus,TuMV)分离物,测定了它们3'-末端cDNA片段的序列。根据衣壳蛋白基因(cp)核苷酸序列可以将这8个分离物与另外2个TuMV分离物分为3组:泰安旧镇大白菜(JZBC)、甘蓝(JZGL)和萝卜(JZLB)分离物为一组;北京(BJILB)、潍坊萝卜分离物(WFLB)与引起萝卜红心病的TuMV分离物(WFLB99)为一组;泰安范镇、河北、潍坊(WFLB04)萝卜和泰安旧镇油菜(JZYC)上的TuMV分离物为一组。农壳蛋白氨基酸序列比较结果与此基本一致,所不同的是分离物WFLB99与所有分离物的差异均较大,形成单独一个分支。有必要对TuMV的变异情况和致病机理进行深入研究。  相似文献
