Drying and rewetting cycles are known to be important for the turnover of carbon (C) in soil, but less is known about the turnover of phosphorus (P) and its relation to C cycling. In this study the effects of repeated drying-rewetting (DRW) cycles on phosphorus (P) and carbon (C) pulses and microbial biomass were investigated. Soil (Chromic Luvisol) was amended with different C substrates (glucose, cellulose, starch; 2.5 g C kg−1) to manipulate the size and community composition of the microbial biomass, thereby altering P mineralisation and immobilisation and the forms and availability of P. Subsequently, soils were either subjected to three DRW cycles (1 week dry/1 week moist) or incubated at constant water content (70% water filled pore space). Rewetting dry soil always produced an immediate pulse in respiration, between 2 and 10 times the basal rates of the moist incubated controls, but respiration pulses decreased with consecutive DRW cycles. DRW increased total CO2 production in glucose and starch amended and non-amended soils, but decreased it in cellulose amended soil. Large differences between the soils persisted when respiration was expressed per unit of microbial biomass. In all soils, a large reduction in microbial biomass (C and P) occurred after the first DRW event, and microbial C and P remained lower than in the moist control. Pulses in extractable organic C (EOC) after rewetting were related to changes in microbial C only during the first DRW cycle; EOC concentrations were similar in all soils despite large differences in microbial C and respiration rates. Up to 7 mg kg−1 of resin extractable P (Presin) was released after rewetting, representing a 35-40% increase in P availability. However, the pulse in Presin had disappeared after 7 d of moist incubation. Unlike respiration and reductions in microbial P due to DRW, pulses in Presin increased during subsequent DRW cycles, indicating that the source of the P pulse was probably not the microbial biomass. Microbial community composition as indicated by fatty acid methyl ester (FAME) analysis showed that in amended soils, DRW resulted in a reduction in fungi and an increase in Gram-positive bacteria. In contrast, the microbial community in the non-amended soil was not altered by DRW. The non-selective reduction in the microbial community in the non-amended soil suggests that indigenous microbial communities may be more resilient to DRW. In conclusion, DRW cycles result in C and P pulses and alter the microbial community composition. Carbon pulses but not phosphorus pulses are related to changes in microbial biomass. The transient pulses in available P could be important for P availability in soils under Mediterranean climates.  相似文献

The aim of this study was to assess the feasibility of controlling the potato cyst nematode Globodera pallida through biofumigation with glucosinolate-rich Brassica juncea genotypes. The main glucosinolate of B. juncea is 2-propenyl glucosinolate which is the precursor of 2-propenyl isothiocyanate. Toxicity of 2-propenyl isothiocyanate to encysted G. pallida was tested in vitro. Fifty percent reduction in hatching was found within 2 h of exposure to 0.002% 2-propenyl isothiocyanate. Based on the in vitro results, we hypothesized that biofumigation with B. juncea would reduce hatching of G. pallida in vivo and higher 2-propenyl glucosinolate levels would have a stronger effect. Plant genotype, sulfur fertilization and insect herbivory affected 2-propenyl glucosinolate concentration of B. juncea. However, increasing 2-propenyl glucosinolate concentration of B. juncea did not affect G. pallida hatching after biofumigation. The absence of a biofumigation effect was most likely due to lower concentrations of 2-propenyl isothiocyanate in vivo compared to in vitro conditions. These results show that it is difficult to reach sufficiently high levels of toxicity to reduce hatching of G. pallida under realistic conditions.  相似文献

The side chains of the rhamnogalacturonan I fraction in sugar beet pectin are particularly rich in arabinan moieties, which may be substituted with feruloyl groups. In this work the arabinan-rich fraction resulting from sugar beet pulp based pectin production was separated by Amberlite XAD hydrophobic interaction and membrane separation into four fractions based on feruloyl substitution and arabino-oligosaccharide chain length: short-chain (DP 2-10) and long-chain (DP 7-14) feruloylated and nonferuloylated arabino-oligosaccharides, respectively. HPAEC, SEC, and MALDI-TOF/TOF analyses of the fractions confirmed the presence of singly and doubly substituted feruloylated arabino-oligosaccharides in the feruloyl-substituted fractions. In vitro microbial fermentation by human fecal samples (n = 6 healthy human volunteers) showed a selective stimulation of bifidobacteria by both the feruloylated and the nonferuloylated long-chain arabino-oligosaccharides to the same extent as the prebiotic fructo-oligosaccharides control. None of the fractions stimulated the growth of the potential pathogen Clostridium difficile in monocultures. This work provides a first report on the separation of potentially bioactive feruloylated arabino-oligosaccharides from sugar beet pulp and an initial indication of the potentially larger bifidogenic effect of relatively long-chain arabino-oligosaccharides as opposed to short-chain arabino-oligosaccharides.  相似文献

Fusarium head blight (FHB) of wheat (Triticum aestivum L.), caused by the fungus Fusarium graminearum, is a major concern worldwide. FHB grain is reduced in yield, may fail to germinate, and is often contaminated with deoxynivalenol, a trichothecene mycotoxin linked to a variety of animal diseases and feed refusals. Annual losses in the tens of millions of dollars due to FHB underscore the need to develop improved methods of disease control and prevention. Previous research has identified deoxynivalenol biosynthesis as a virulence factor on wheat. Recently, we found that the TRI14 gene of F. sporotrichioides, closely related to F. graminearum, was not required for synthesis of a related trichothecene, T-2 toxin. TRI14 does not share similarity with any previously described genes in the databases. In this study, we examined the role that F. graminearum TRI14 may play in both deoxynivalenol synthesis and in virulence on wheat. TRI14 deletion mutants synthesize deoxynivalenol on cracked maize kernel medium and exhibit wild-type colony morphology and growth rate on complex and minimal agar media. However, FHB assays on greenhouse-grown wheat indicate that FgDeltaTri14 mutants cause 50-80% less disease than wild type and do not produce a detectable quantity of deoxynivalenol on plants. We discuss a number of possible roles that TRI14 may play in the disease process.  相似文献

This study investigates the applicability of on-line coupling of capillary electrophoresis with electrospray ionization tandem mass spectrometry (CZE-ESI-MS) for the separation and characterization of alpha- and beta-acids and oxidized hop acids from crude extracts of different hop varieties. CZE-ESI-MS with negative-ion electrospray ionization proved to be a suitable technique for the determination of these types of natural compounds and their oxidized derivatives. The CZE parameters (pH, concentration, and buffer type) and ESI-MS parameters (nature and flow rate of the sheath liquid, nebulizer pressure, drying gas flow rate, temperature, and compound stability) were optimized. The optimized method provides the potential for a fast qualitative determination of hop acids and their oxidation compounds. The method was also applied to the determination of iso-alpha-acids in beer.  相似文献

Biochanin A and formononetin are abundant in legumes. These proestrogenic isoflavones can be converted by 4'-O-demethylation to the more potent phytoestrogens genistein and daidzein. Incubation of biochanin A or formononetin with human liver microsomes resulted in 4'-O-demethylation and the production of additional metabolites. Three new hydroxylated formononetin derivatives, 6,7-dihydroxy-4'-methoxyisoflavone, 7,8-dihydroxy-4'-methoxyisoflavone, and 7,3'-dihydroxy-4'-methoxyisoflavone, were isolated and characterized. We surveyed the O-demethylase competence of cytochrome P450 isoforms found in human liver. Human cytochrome P450 isoforms 1A2, 2E1, 2C9*1, 2C19, and 2D6*1 catalyzed biochanin A consumption and genistein production. Human cytochrome P450 isoforms 1A2, 2C9*1, 2A6, 2D6*1, and 2C19 catalyzed formononetin consumption and daidzein production. These isoforms also generated other hydroxylated metabolites. Although O-demethylation of isoflavones has been attributed to metabolism by gut microflora, our study demonstrates that human hepatic microsomal enzymes can perform the same transformation and may play a key role in the conversion of 4'-O-methylated isoflavones to more potent phytoestrogens.  相似文献

Organism succession during ecosystem development has been researched for aboveground plant communities, however, the associated patterns of change in below-ground microbial communities are less described. In 2008, a study was initiated along a developmental sand-dune soil chronosequence bordering northern Lake Michigan near Wilderness Park (WP). It was hypothesized that soil bacterial communities would follow a pattern of change that is associated with soil, plant, and ecosystem development. This study included 5 replicate sites along 9 soils (n = 45) ranging in age from ∼105 to 4010 years since deposition. Soil bacterial community composition and diversity were studied using bacterial tag-encoded FLX amplicon pyrosequencing of the 16S rRNA gene. Bray–Curtis ordination indicated that bacterial community assembly changed along the developmental soil and plant gradient. The changes were not affected by seasonal differences, despite likely differences in plant root C (e.g. exudates), temperature, and water availability in soil. Soil base cations (Ca, Mg) and pH declined, showing log-linear correlations with soil age (r ∼ 0.83, 0.84 and 0.81; P < 0.01). Bacterial diversity (Simpson's 1/D) declined rapidly during the initial stages of soil development (∼105–450 y) and thereafter (>450 y) did not change. Turnover of plant taxa was also more rapid early during ecosystem development and correlated with bacterial community structural change (P < 0.000001; r = 0.56). It is hypothesized that plants help to drive pedogenic change during early (<450 y) soil development (e.g. pH decline, cation leaching) which drive selection of soil bacterial communities. In mature soils (∼450–4000 y), resilient and stable soil bacterial community structures developed, mimicking steady-state climax communities that were observed during latter stages of primary plant succession. These relationships point to possible feedbacks between plant and bacterial communities during ecosystem development.  相似文献

Although numerous studies have demonstrated the health benefits of chlorophyll derivatives, information regarding the digestion, absorption, and metabolism of these phytochemicals is quite limited. To better understand the digestion of these pigments, green vegetables including fresh spinach puree (FSP), heat- and acid-treated spinach puree (HASP), and ZnCl(2)-treated spinach puree (ZnSP) were subjected to an in vitro digestion method which simulates both the gastric and small intestinal phases of the process. Native chlorophylls were converted to Mg-free pheophytin derivatives during digestion. Conversely, Zn-pheophytins were completely stable during the digestive process. Transfer of lipophilic chlorophyll derivatives, as well as the carotenoids lutein and beta-carotene, into the aqueous micellar fraction from the food matrix was quantified. Micellarization of total chlorophyll derivatives differed significantly (p < 0.05) for FSP (37.6%), HASP (17.2%), and ZnSP (8.7%). Micellarization of chlorophyll a derivatives was determined to be significantly more efficient than chlorophyll b derivatives in FSP and HASP (p < 0.01), but not in ZnSP (p > 0.05). Intestinal cell uptake of micellarized pigments was investigated using HTB-37 (parent) and clonal TC7 lines of human Caco-2 cells. Medium containing the pigment-enriched fraction generated during digestion was added to the apical surface of fully differentiated monolayers for 4 h. Pigments were then extracted from cells and analyzed by C18 HPLC with photodiode array detection. Both Caco-2 HTB-37 and TC7 clone cells accumulated 20-40% and 5-10% of micellarized carotenoid and chlorophyll derivatives, respectively. These results are the first to demonstrate uptake of chlorophyll derivatives by human intestinal cells and to support the potential importance of chlorophylls as health-promoting phytochemicals.  相似文献

Preparations of Echinacea are widely used as alternative remedies to prevent the common cold and infections in the upper respiratory tract. After extraction, fractionation, and isolation, the antioxidant activity of three extracts, one alkamide fraction, four polysaccharide-containing fractions, and three caffeic acid derivatives from Echinacea purpurea root was evaluated by measuring their inhibition of in vitro Cu(II)-catalyzed oxidation of human low-density lipoprotein (LDL). The antioxidant activities of the isolated caffeic acid derivatives were compared to those of echinacoside, caffeic acid, and rosmarinic acid for reference. The order of antioxidant activity of the tested substances was cichoric acid > echinacoside > or = derivative II > or = caffeic acid > or = rosmarinic acid > derivative I. Among the extracts the 80% aqueous ethanolic extract exhibited a 10 times longer lag phase prolongation (LPP) than the 50% ethanolic extract, which in turn exhibited a longer LPP than the water extract. Following ion-exchange chromatography of the water extract, the majority of its antioxidant activity was found in the latest eluted fraction (H2O-acidic 3). The antioxidant activity of the tested Echinacea extracts, fractions, and isolated compounds was dose dependent. Synergistic antioxidant effects of Echinacea constituents were found when cichoric acid (major caffeic acid derivative in E. purpurea) or echinacoside (major caffeic acid derivative in Echinacea pallida and Echinacea angustifolia) were combined with a natural mixture of alkamides and/or a water extract containing the high molecular weight compounds. This contributes to the hypothesis that the physiologically beneficial effects of Echinacea are exerted by the multitude of constituents present in the preparations.  相似文献

Sodium copper chlorophyllin (SCC), a mixture of water-soluble chlorophyll derivatives, is used as both a food colorant and a common dietary supplement. Although the potential antimutagenic and antioxidant properties of this commercial preparation have been demonstrated, limited information is available on its digestion and absorption by humans. Stability of SCC was examined during simulated gastric and small intestinal digestion. Three preparations were subjected to in vitro digestion: SCC in water, SCC in water + 10% corn oil, and SCC in applesauce. SCC components from raw material preparations and in digested samples were analyzed by C(18) HPLC with photodiode array detection. Cu(II)chlorin e(4), the major chlorin component of SCC, was relatively stable during simulated digestion. In contrast, greater than 90% of Cu(II)chlorin e(6) was degraded to undetermined products during digestion. Recovery of Cu(II)chlorin e(6) after digestion was increased by incorporation of SCC into applesauce, suggesting a protective role of the inclusion matrix for stabilization of labile SCC components. Accumulation of SCC derivatives was investigated by using differentiated cultures of the TC7 clone of the Caco-2 human intestinal cell line. Cellular accumulation from media containing 0.5 to 60 ppm SCC was linear with intracellular content ranging between 0.2 and 29.6 microg of total SCC per mg of cellular protein. Uptake of SCC by Caco-2 cells was significantly (p < 0.01) lower in cultures incubated at 4 degrees C than in those incubated at 37 degrees C. Although intracellular SCC was transported into both apical and basolateral compartments when Caco-2 cells were grown on inserts, apical efflux was significantly greater (p < 0.01) than basolateral efflux. Stability of Cu(II)chlorin e(4) during in vitro digestion and effective uptake by Caco-2 enterocyte-like cells support the likelihood that a portion of this SCC component or its metabolites is absorbed from the human intestine.  相似文献

Central Brazil is the region with the most dynamic agriculture expansion worldwide, where tropical forests and Cerrado (Brazilian savanna) are converted to pastures and crop fields. Following deforestation, agricultural practices, such as fertilization, tillage and crop rotations, alter soil parameters and affect microbial abundances and the C and N cycles. The objective of this study was to compare changes in soil fertility, stocks of soil C and N, microbial biomass, and abundance of bacteria, fungi and archaea in Cerrado soils following land use change to crops (soybean/corn/cotton) and pasture (the perennial forage grass Brachiaria brizantha A. Rich.). Agriculture increased soil fertility and conserved soil C and N since their absolute concentration values were highest in agriculture soils and the C and N stocks adjusted by soil density were similar to the native vegetation soils. At the same time, agriculture changed the microbial abundances (decrease of microbial biomass C and N, increase of archaea, and reduction of bacteria and fungi at the crop sites), and N dynamics (increase of soil ammonium and nitrate concentrations). Even if these changes can be beneficial for food and agricultural commodities production, all these soil alterations should be further investigated due to their possible unknown effects on biosphere–hydrosphere–atmosphere exchange processes such as greenhouse gases emissions and nitrate leaching.  相似文献

Hydrogenated soybean oil was obtained after 10 min of hydrogenation with 0.5% selective type Ni catalyst at 230 degrees C, a hydrogenation pressure of 0.049 MPa, and an agitation rate of 300 rpm. The conjugated linoleic acid isomers in the hydrogenated soybean oil were isolated by using a silver ion-impregnated HPLC. Gas chromatography-mass spectrometry of 4,4-dimethyloxazoline (DMOX) derivatives of the isolated conjugated linoleic acid isomers were carried out for the identification of their chemical structures. By interpreting the mass spectra of the DMOX derivatives of conjugated linoleic acid isomers isolated by silver ion-impregnated HPLC, 20 different conjugated linoleic acid isomers present in hydrogenated soybean oil were identified. This is the first report for the mass spectrometric identification of the conjugated linoleic acid isomers present in hydrogenated vegetable oil.  相似文献

As endocrine-disrupting chemicals, a few frequently used phthalate plasticizers were banned or restricted for use as additives in food in some countries. The interaction mechanisms between three phthalate plasticizers with human serum albumin (HSA) were studied by fluorescence (quenching, synchronous, and three-dimensional), UV-vis absorption, circular dichroism (CD), and Fourier transform infrared (FT-IR) spectroscopy, in combination with molecular modeling under simulative physiological conditions, respectively. The results obtained from fluorescence quenching data revealed that the plasticizers-HSA interaction altered the conformational strcture of HSA. Meanwhile, the alterations of HSA secondary structure in the presence of phthalate plasticizers were investigated. The binding distances for the plasticizers-HSA system were provided by the efficiency of fluorescence resonance energy transfer. Furthermore, the thermodynamic analysis implied that hydrophobic forces were the main interaction for the plasticizers-HSA system, which agreed well with the results from the molecular modeling study.  相似文献

While experimental addition of nitrogen (N) tends to enhance soil fluxes of carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O), it is not known if lower and agronomic-scale additions of urea-N applied also enhance trace gas fluxes, particularly for semi-arid agricultural lands in the northern plains. We aimed to test if this were true at agronomic rates [low (11 kg N ha−1), moderate (56 kg N ha−1), and high (112 kg N ha−1)] for central North Dakota arable and prairie soils using intact soil cores to minimize disturbance and simulate field conditions. Additions of urea to cores incubated at 21 °C and 57% water-filled pore space enhanced fluxes of CO2 but not CH4 and N2O. At low, moderate, and high urea-N, CO2 fluxes were significantly greater than control but not fluxes of CH4 and N2O. The increases in CO2 emission with rate of urea-N application indicate that agronomic-scale N inputs may stimulate microbial carbon cycling in these soils, and that the contribution of CO2 to net greenhouse gas source strength following fertilization of semi-arid agroecosystems may at times be greater than contributions by N2O and CH4.  相似文献

Arnica montana is a medicinally important plant due to its broad health effects, and it is used in Ayurvedic, Homeopathic, Unani, and folk medicines. We have used LC-MS(n) (n = 2-5) to detect and characterize in Arnica flowers 11 quantitatively minor fumaric and methoxyoxalic acid-containing chlorogenic acids, nine of them not previously reported in nature. These comprise 1,5-dicaffeoyl-3-methoxyoxaloylquinic acid, 1,3-dicaffeoyl-4-methoxyoxaloylquinic acid, 3,5-dicaffeoyl-4-methoxyoxaloylquinic acid, and 1-methoxyoxaloyl-4,5-dicaffeoylquinic acid (M(r) 602); 3-caffeoyl-4-feruloyl-5-methoxyoxaloylquinic acid and 3-feruloyl-4-methoxyoxaloyl-5-caffeoylquinic acid (M(r) 616); 1,5-dicaffeoyl-4-fumaroyl and 1,5-dicaffeoyl-3-fumaroylquinic acid (M(r) 614); 3,5-dicaffeoyl-1,4-dimethoxyoxaloylquinic acid (M(r) 688); and 1-methoxyoxaloyl-3,4,5-tricaffeoylquinic acid and 1,3,4-tricaffeoyl-5-methoxyoxaloylquinic acid (M(r) 764). All of the structures have been assigned on the basis of LC-MS(n) patterns of fragmentation, relative hydrophobicity, and analogy of fragmentation patterns if compared to caffeoylquinic acids. This is the first time when fumaric acid-containing chlorogenic acids are reported in nature.  相似文献

Abstract

We studied two unknown substances which were produced by incubation of p-coumaric and ferulic acids in a peat soil extract solution. The substances were identified as the cisforms of these acids, which were isomerized from their trans-forms during the incubation by exposure to fluorescent light. Such isomerization did not take place during incubation under dark conditions. After 5 days of incubation by exposure to fluorescent light, the maximal ratio of cis-form to trans-form p-coumaric and ferulic acids was 0.61 and 0.45, respectively.  相似文献
