Efficacy of ivermectin in a topical formulation was evaluated in cattle against adult gastrointestinal and pulmonary nematode infections (experiment 1, n = 24), the 2nd- and 3rd-stage larvae of Hypoderma spp (experiment 2, n = 12), and the biting louse Damalinia bovis (experiment 3, n = 12). Nematode infections were induced and grubs and lice were naturally acquired. Treatments consisted of a single dose of ivermectin in a topical formulation of 200, 500, or 1,000 micrograms/kg of body weight in experiment 1 or 500 micrograms/kg in experiments 2 and 3. At 1,000 micrograms/kg, ivermectin was 100% effective against Ostertagia ostertagi, Trichostrongylus colubriformis, Oesophagostomum radiatum, Nematodirus helvetianus, Haemonchus placei, and Dictyocaulus viviparus and was greater than 99% effective against Cooperia oncophora, C punctata, and T axei. At 500 micrograms/kg, the efficacy was 100% against C oncophora, C punctata, O ostertagi, T axei, Oes radiatum, N helvetianus, Haem placei, and Dict viviparus and greater than 99% against T colubriformis. At 200 micrograms/kg, the efficacy was 100% against Oes radiatum, Haem placei, and Dict viviparus, greater than 99% for O ostertagi, 96% for C oncophora, 86% for C punctata, 90% for T axei, 85% for T colubriformis, and 71% for N helvetianus. At 500 micrograms/kg, ivermectin was highly effective against the grubs Hypoderma bovis and H lineatum and eliminated the louse Damalinia bovis. 相似文献

An ivermectin tablet for o ral administration to sheep was developed for use in countries where it is customary to treat sheep with anthelmintic tablets. Tablets require no special administration equipment, and offer convenience for storage and transport. The ivermectin tablet, which delivers 10 mg of ivermectin (200 μg kg−1 in a 50 kg sheep), had similar bioavailability to a liquid formulation of ivermectin (IVOMEC® Liquid for Sheep) as determined by peak plasma ivermectin concentrations and area under the concentration curve in plasma (P>0.10). In dose confirmation trials in which nematode infections were induced in helminth-naive sheep, animals treated with the ivermectin tablet had significantly fewer adult and fourth-stage larval nematodes than untreated control sheep P<0.01 with efficacies >99% against all nematode species tested. In six field trials evaluating the efficacy of the ivermectin tablet in 240 Merino sheep, the reductions in faecal nematode egg counts ranged between 98 and 100%, as determined by comparison of pre- and post-treatment counts for the ivermectin-treated group. 相似文献

Twenty calves with naturally acquired gastrointestinal nematode infections were assigned to 10 replicates of 2 calves each and were treated with febantel paste (5 mg/kg of body weight, PO) or an equivalent volume of paste vehicle (control calves). Adverse reactions were not observed after febantel treatment. At necropsy, gastrointestinal nematode counts in febantel-treated calves, compared with control calves, were reduced by over 99% for all major species except Trichuris discolor, counts of which were reduced by only 69%. Counts of gastrointestinal nematode eggs in feces of febantel-treated calves were reduced by 100% after treatment. 相似文献

Anthelmintic efficacy of moxidectin, an experimental antiparasitic macrocyclic lactone, was evaluated in a group of 15 calves harboring naturally acquired gastrointestinal nematodes. Three groups of 5 calves each served as untreated controls (group 1) or principals that were given moxidectin PO at the rate of 0.2 mg/kg (group 2) or 0.4 mg/kg (group 3) of body weight. Equal numbers of control and treated calves were necropsied for parasite recovery on days 10 (3 control and 3 of each treatment group) and 11 (2 control and 2 of each treatment group) after treatment. Efficacies at both doses were greater than 99.8 and 99.9% against active and inhibited larvae and adults of Ostertagia spp, respectively. Twelve calves (mean weight, 175.5 kg) were used to confirm efficacy of ivermectin delivered from a prototype sustained-release bolus against naturally acquired gastrointestinal nematodes including early fourth-stage (inhibited) larvae of Ostertagia ostertagi. The calves were allocated by restricted randomization on weight to 1 of 2 groups: controls, to which a placebo bolus was given orally, and treated calves, to which a sustained-release bolus designed to deliver 8 mg of ivermectin/day at a steady rate was given orally. After treatment, the 2 groups were housed in separate pens with concrete flooring. Twenty-eight days after treatment, all calves were euthanatized and necropsied. The ivermectin-treated calves had no larval or adult Ostertagia spp and significantly (P less than 0.01) fewer adult Trichostrongylus axei and adult Cooperia (C oncophora, C punctata and C surnabada) than control calves. Efficacy of ivermectin was greater than 99% for Cooperia spp, and 100% for other parasites. Drug-related adverse reactions were not observed. 相似文献

The benzimidazole anthelmintic, oxfendazole, was used as an oral suspension at 4.5 and 2.8 mg/kg to treat naturally infested indigenous buffaloes in Egypt. Worms of the genera Haemonchus, Cooperia, Trichostrongylus, Ostertagia, Chabertia and Oesophagostomum were identified in the animals prior to treatment. A 100% reduction in faecal egg counts was obtained at the 4.5 mg/kg level when compared to untreated control animals. Reductions in faecal egg counts greater than 90% were recorded at 2.8 mg/kg and these exceeded reductions obtained by drenching with the older benzimidazole anthelmintic, thiabendazole, at 67 and 111 mg/kg. 相似文献

The efficacy of moxidectin, a new endectocide against natural nematode infections, was evaluated. Twenty-five calves were divided into two groups of eight calves each and a third group of nine calves. Moxidectin was administered s.c. to two groups (I, 0.2 mg kg-1 body weight (BW); II, 0.3 mg kg-1 BW) and the third group (III) received the vehicle (placebo) s.c. Two pre-treatment and one post-treatment faecal nematode egg count determinations were made from all calves, and they were necropsied 2 weeks after treatment for the identification and enumeration of nematode parasites. Group III calves, which received the drug-free vehicle, harboured eight species of nematodes (Ostertagia ostertagi, Trichostrongylus axei, Cooperia oncophora, Cooperia punctata, Nematodirus helvetianus, Trichuris discolor, Oesophagostomum radiatum and Dictyocaulus viviparus). The mean total worm burden for this group was 8935. There was a significant reduction in the numbers of many species of nematodes (Ostertagia, Trichostrongylus, Cooperia and Nematodirus) in both treated groups. Cooperia oncophora was reduced by 94% in Group I and by 96% in Group II, while all other nematode species were reduced by 99%. Immature stages of Ostertagia and Nematodirus were significantly reduced in the two treated groups. Two weeks after treatment, the mean faecal egg counts of both treated groups were reduced by more than 98%. The efficacy of ivermectin in oral paste formulation at a dosage of 200 micrograms/kg of body weight was tested against naturally acquired larval and adult stages of Parascaris equorum, in a controlled study. Twenty infected pony foals 18 to 27 weeks of age were randomly allocated to 2 groups of 10 each and were placed in dry lots. Foals in 1 group were given ivermectin on day 0. Necropsies and parasite recoveries from small intestines and lung tissues were performed on 5 foals in each group at 2 weeks after treatment (WAT) and on the remaining foals at 5 WAT. Ivermectin was 100% effective against adult P equorum. At 2 WAT, ivermectin was 100% effective against lung larval stages and 91% effective against intestinal larvae of P equorum. Efficacy of ivermectin against all intestinal stages was 93%. Intestinal and lung larval populations were similar in ivermectin-treated and nontreated foals at 5 WAT, indicating that the foals had become reinfected from the contaminated drylots, with larvae repopulating in the tissues 2 to 5 WAT. Apparent increases in severity of lymphoproliferative and inflammatory tissue reactions were observed histologically in lung and liver tissues of treated foals. Clear differences in clinical conditions between foals in treated and nontreated groups were not observed. However, weight gains in foals were greatest after treatment. 相似文献

The objective of this study was to evaluate the accuracy of the faecal egg count reduction test (FECRT) and the faecal egg count efficacy test (FECET) to assess the resistance status of ivermectin (630 μg/kg) and moxidectin (200 μg/kg), using the controlled efficacy test as a reference, and whether the results of the EPG are equivalent to the efficacy results from the parasitological necropsies. Two experiments were conducted. The results demonstrate that it was not possible to demonstrate that the EPG values were equivalent with the ivermectin and moxidectin efficacy obtained by parasitological necropsies, mainly if the phenomenon of parasites resistance is not advanced in a determined field population. Maybe the FECET technique would be possibly better than the FECRT. The high anthelmintic efficacy of 200 μg/kg moxidectin, in naturally infected cattle, against field population of nematodes that are resistant to 630 μg/kg ivermectin, was observed in this study. 相似文献

Two controlled studies involving 24 cattle were conducted in New Zealand to determine the efficacy of a topical, non-flammable formulation of eprinomectin against induced and naturally acquired nematode infections. In Trial 1, nematode infections were induced on Day -5 with third-stage larvae of Cooperia spp., Haemonchus contortus, Ostertagia ostertagi and Trichostrongvlus colubriformis so that the nematodes would be at the fourth larval stage when the cattle were treated. In Trial 2, cattle had naturally acquired nematode infections as determined by faecal nematode egg counts and larval cultures. The cattle were allocated on Day 0 (Trial 1) or Day 6 (Trial 2) on a stratified random basis according to bodyweight to one of two treatments: untreated control or eprinomectin (0.5% w/v) applied topically at 1 ml/10 kg bodyweight. Necropsies were undertaken on Days 14 and 15 and total nematode counts were done. In Trial 1, cattle treated with eprinomectin had significantly (p < 0.05) fewer Cooperia spp. and O. ostertagi than the controls. Larvae of H. contortus and T. colubriformis did not establish. In Trial 2, cattle treated with eprinomectin had significantly (p < 0.05) fewer of the following parasites than the controls: Haemonchus spp. (adult), Cooperia surnabada (adult), C. oncophora (adult), Cooperia spp. (L,), Ostertagia lyrata (adult), O. ostertagi (adult), Oesophagostomum spp. (adult), T. avei (adult and L1) and Trichuris spp. (adult). Reductions of 100% were observed for Capilfaria spp. (adult), D. viviparus (adult and L,), and Nematodirus helvetianus (adult), but these were not statistically significant (p > 0.05) because four or fewer control animals were infected with these parasites. In Trial 2, efficacies of greater than 99% were observed against all species for which moderate to high burdens occurred in the untreated controls. These findings indicate that eprinomectin in a topical formulation is a highly effective nematocide in cattle. 相似文献

Two trials involving a total of 36 Dorset horn lambs were conducted to assess the anthelmintic efficacy of ivermectin against experimental infections of benzimidazole-resistant strains of Haemonchus contortus and Ostertagia circumcincta. Two resistant strains of each of the two species were used and in each trial the lambs were allocated to three groups. One group was given 200 micrograms ivermectin/kg bodyweight orally, the second group was given 5 mg oxfendazole/kg bodyweight orally and the third group remained untreated as controls. Fourteen days after treatment the lambs were necropsied. Ivermectin was found to be more than 99 per cent to 100 per cent effective against all four benzimidazole-resistant strains, whereas oxfendazole was 78.6 per cent and 83.8 per cent effective against the H contortus strains, and 25.6 per cent and 39.8 per cent effective against the O circumcincta strains. 相似文献

The anthelmintic activity of ivermectin when administered orally and subcutaneously at a dosage of 0.2 mg/kg of body weight was evaluated in 4 trials involving 20 dromedary camels (Camelus dromedarius). Fecal egg counts were made on the day of treatment and at 2, 4, and 10 weeks after treatment. Trichostrongylid egg counts were reduced by a maximum of 100% when ivermectin was administered orally and by greater than 88% when given subcutaneously. Egg counts of Trichuris spp were reduced by greater than 85% with oral administration, but increased following subcutaneous treatment. No adverse local or systemic reactions were observed following treatment with ivermectin by either route. 相似文献

Dermatobia hominis, the tropical warble fly, is one of the most important ectoparasites of cattle in Latin America. The results of five trials conducted in Paraguay, Brazil and Colombia showed that ivermectin administered subcutaneously at a dose level of 200 mcg/kg body weight was highly effective against the three larval stages of D. hominis in cattle. 相似文献
