Japanese encephalitis was recently reported from individuals in the mountain districts of Nepal without travel history to Japanese encephalitis virus (JEV) endemic areas. We performed a cross-sectional study to estimate the seroprevalence of JEV in pigs and subsequently conducted a survey of farmers to identify risk factors associated with seropositivity. In July and August, 2010, 454 pig serum samples were collected and tested by competitive ELISA. Data from a 35-question survey of 109 pig owners were analysed using multivariate logistic regression. Seventy-six (16.7, 95% CI 13.6-20.4) pigs tested positive for anti-JEV antibodies, none of which had been vaccinated against JEV or sourced from JEV endemic areas. Risk factors associated with JEV seropositivity were 'summer abortion', 'wells as a water source', 'urban location', 'reported presence of mosquitoes' and 'lower elevation'. Our results suggest that JEV is likely circulating in the mountain districts of Nepal, and that locally acquired JEV should be considered a risk for residents and travellers in these areas.  相似文献

A study was conducted during 1985 and 1986 to evaluate the roles of mosquito species as possible vectors of Japanese encephalitis (JE) virus in Hokkaido. The number of Culex tritaeniorhynchus was very low among the four pig farms where outbreaks of abortion caused by JE virus were observed in swine populations. At one farm near Sapporo, only one Cx. tritaeniorhynchus was found among a total of 510 mosquitoes collected during the survey period from July to October 1985, even when JE virus activity among sentinel pigs was revealed by seroconversion. At another farm in the south, no individuals of this mosquito species were found among 987 mosquitoes collected at the time of the outbreaks of abortion. Cx. pipiens pallens, Anopheles species, Aedes vexans nipponii, and Ae. japonicus were predominant over Cx. tritaeniorhynchus. Cx. tritaeniorhynchus, almost a solve vector species of JE virus in the southern part of Japan, is probably not a vector of the virus in Hokkaido. The collected mosquitoes (2,332 from 1985 and 1,403 from 1986) were processed for virus isolation but no JE virus was isolated. More extensive field studies are necessary to provide further information on the role of mosquito species other than Cx. tritaeniorhynchus in the transmission of JE virus in the northern limits of its range including Hokkaido.  相似文献

A regional survey was conducted in Nepal for antibody to Japanese encephalitis virus (JEV) in domestic animals. Sera from pigs, and limited numbers of ducks and horses were collected from 16 districts in 2002-2003 and subjected to three serological tests. Of 270 porcine sera tested by C-ELISA, 55% were found positive for the presence of antibodies against Japanese encephalitis virus. Additional testing for IgM antibody to JEV revealed less than 2% of C-ELISA positive sera had evidence of recent JEV infection. Plaque reduction neutralisation tests (PRNT) using JEV, Murray Valley encephalitis (MVEV) and Kunjin (KUNV) viruses implicated JEV as the flavivirus associated with the observed antibody response in most sero-positive pigs. However, eight porcine sera with predominant neutralising antibody for KUNV (an Australasian subtype of West Nile Virus) provided evidence for the circulation of West Nile virus in Nepal.  相似文献

Veterinarians specializing in pig health care responded to a questionnaire regarding their experiences with vaccinations. The goal of the questionnaire was to gain insight into a) the reasons for vaccination and b) the factors involved in the efficacy of vaccination. The results indicated that vaccinations were typically initiated because of health problems at the farm. Veterinarians worked together with farmers and other concerned parties to initiate vaccinations. Respondents predicted that the number of vaccinations would decrease substantially with optimal farm management, but would increase if farmers were allowed (under strict conditions) to vaccinate their own pigs. The results further indicated that the nature of financial compensation predicted the rate of vaccination. Veterinarians paid according to a fixed 'fee for service' system vaccinated less frequently than did veterinarians who were paid according to an ongoing farm management contract. In conclusion, veterinarians appear to be restricted in their capacity to disseminate their experience and knowledge due to the competing needs of the farming network (animals, farmers, retailers).  相似文献

South African pig sector is a contributor to the agricultural industry. A study was conducted to identify the production constraints and compare the management practices in smallholder pig farms in Mpumalanga, South Africa. A total of 220 selected smallholder pig farmers were interviewed. Smallholder pig farming was predominated by male (64 %), age above 50 years (54 %), black Africans (98.6 %), and three quarters of the smallholder farmers were poor to just below average. Majority (80 %) have no pig husbandry training, while only 33 % received assistance from government’s Agricultural Department. In terms of stock, mixed breeds (89 %) from exotic pigs were mostly kept and majority (87 %) of the farmers kept ≤10 sows in their herds. Many farmers (75 %) engaged in risky behavior of buying auctioned-sourced boars, free-range boars, and untested boars from neighbors and relatives. Few (17 %) farmers practiced vaccination and only 10 % kept farm records. Majority of the responses on pre-weaning mortality (50 %) and post-weaning mortality (90 %) were within acceptable range of 1–10 and 1–5 % mortality rates, respectively. The lead causes of mortality were weak piglets and crushing (46 %), diarrhea (27 %), poor management knowledge (19 %), and malnutrition (16 %). Agricultural training and government incentives will facilitate improved productivity in smallholder pig farming.  相似文献

Classical swine fever (CSF) causes major losses in pig farming, with various degrees of disease severity. Efficient live attenuated vaccines against classical swine fever virus (CSFV) are used routinely in endemic countries. However, despite intensive vaccination programs in these areas for more than 20 years, CSF has not been eradicated. Molecular epidemiology studies in these regions suggests that the virus circulating in the field has evolved under the positive selection pressure exerted by the immune response to the vaccine, leading to new attenuated viral variants. Recent work by our group demonstrated that a high proportion of persistently infected piglets can be generated by early postnatal infection with low and moderately virulent CSFV strains. Here, we studied the immune response to a hog cholera lapinised virus vaccine (HCLV), C-strain, in six-week-old persistently infected pigs following post-natal infection. CSFV-negative pigs were vaccinated as controls. The humoral and interferon gamma responses as well as the CSFV RNA loads were monitored for 21 days post-vaccination. No vaccine viral RNA was detected in the serum samples and tonsils from CSFV postnatally persistently infected pigs for 21 days post-vaccination. Furthermore, no E2-specific antibody response or neutralising antibody titres were shown in CSFV persistently infected vaccinated animals. Likewise, no of IFN-gamma producing cell response against CSFV or PHA was observed. To our knowledge, this is the first report demonstrating the absence of a response to vaccination in CSFV persistently infected pigs.  相似文献

OBJECTIVE: To evaluate the safety and efficacy of a human adenovirus-5 vaccine for protecting weaned pigs against swine influenza virus subtype H3N2 infection when administered via 2 injection methods. ANIMALS: 76 pigs. PROCEDURE: 6 groups of weaned pigs received a 10-fold serial dilution of recombinant adenovirus expressing H3 hemagglutinin and a constant amount of recombinant adenovirus expressing nucleoprotein, either via a needle-free injection device or by traditional IM injection. In each group of 10 pigs, 1 served as a nonvaccinated contact pig to monitor whether there was spread of vaccinial virus from pig to pig. Vaccinated pigs and nonvaccinated controls were challenged or sham-inoculated 5 weeks later. After challenge, pigs were observed for clinical signs and nasal secretions were tested for virus. On day 5 after challenge, pigs were euthanatized; lungs were examined for gross lesions, and bronchoalveolar lavage specimens were tested for virus replication. RESULTS: A hemagglutination inhibition (HI) antibody response was elicited in a dose-dependent manner. Traditional IM administered vaccination induced consistently higher HI antibody responses than vaccination via needle-free injection, but the differences were not significant. Likewise, traditional IM administration was superior at reducing nasal virus shedding except at the highest dose, at which both methods blocked virus replication. The severity of lung lesions was reduced in a dose-dependent manner by both vaccination methods. Sentinel pigs did not seroconvert. CONCLUSIONS AND CLINICAL RELEVANCE: The human adenovirus-5 vaccine at high doses prevented nasal virus shedding after challenge exposure with both methods of administration. The replication-defective vaccine virus was not transmitted to sentinel pigs.  相似文献

Purpose and methods

In situ conservation of pig genetic resources requires understanding of the farming systems under which the pigs are kept. The objective of the study was to characterise smallholder pig production systems where indigenous pigs are kept in order to assess opportunities for in situ conservation of these pigs. Factors influencing pig herd sizes, ranking of pigs in smallholder production systems, breeds and breed preferences, pig ownership patterns, traits preferred by farmers and constraints to production were investigated in a survey involving 199 farmers from Chirimhanzu District of Zimbabwe, Vhembe, Alfred Nzo and O. R. Tambo Districts of South Africa.

Results

Income was the major factor influencing most pig production related parameters. Most pigs (69.67 %) were owned by women, with most of the women falling into the very low income group. Farmers kept pigs for several reasons that were common across geographical zones and income groups. The odds of a farmer ranking pigs first in the production system depended on the absence of other livestock (P?<?0.05). Farmers in the higher income group tended to mention poor feed resources and access to information as constraints (P?<?0.05) compared to the very low income group. There was preference heterogeneity in the breeds kept and the reasons for breed choice across the income groups and geographical zones. Fewer farmers in the very low income group (42.62 %) recognised the need to conserve local pigs compared to the low income group (80.49 %), the medium income group (50.00 %) and the high income group (100.00 %; χ 2?=?19.14; P?<?0.001).

Conclusions

It was concluded that farmers see value in local pigs and are willing to conserve them. In situ conservation programmes are possible and these should recognise the role of poor women in conserving and enhancing indigenous pig genetic resources.  相似文献

Kathmandu, Nepal has been classified as a high‐risk area for highly pathogenic avian influenza (HPAI) by the Nepali Government. While ducks have an important role in the transmission of avian influenza viruses (AIV), including HPAI, seroprevalence of antibodies to AIV in domestic ducks of Kathmandu has never been assessed. The objectives of this study were (i) to estimate the prevalence of seroconversion to AIV in domestic ducks in major duck‐raising areas of Kathmandu and (ii) to assess the effect of age, sex, presence of swine and the number of ducks on the farm on the carriage of antibodies to AIV in these ducks. From April through July of 2011, a cross‐sectional study was conducted and a total of 310 ducks in the major duck‐raising areas of Kathmandu were sampled. The estimated prevalence of AIV antibodies was 27.2% [95% confidence interval (CI): 24.6–29.5]. Of 62 enrolled farms, 42% had at least one seropositive duck. Half of the enrolled farms also kept pigs of which 52% had at least one seropositive duck. Bivariate analysis indicated association between ducks' seroconversion to AIV and their age, sex and farm size. However, the final multivariable model, after controlling for clustering of ducks within farms, identified age as the only significant risk factor. Based on this model, ducks older than 1 year of age were more likely to be seropositive compared to ducks <6 months of age [odds ratio = 2.17 (1.07–4.39)]. These results provide baseline information about the AIV seroprevalence in domestic ducks in the major duck‐raising areas of Kathmandu and identify a high‐risk group that can be targeted in surveillance activities. Future studies should be conducted to differentiate the subtypes of AIV present among domestic ducks in Kathmandu, with particular interest in the presence of HPAI viruses.  相似文献

The present study was conducted to know the smallholder pig production system in tribal areas of Sikkim State, India. Two hundred tribal farmers were selected randomly from the North and East District of the state. Information on socio-economic characteristics of farmers (gender, occupation, educational status, and farming experience), management practices, disease prevalence, and economics in pig production was collected. The study recorded the mean land holding as 1.2?±?0.8 ha, and the number of pigs per farm was 5.0?±?0.28. Pigs were mainly kept as a source of income, and 70 % of farmers reared crossbreed pigs. Ninety percent (90 %) of respondents practiced the intensive system of management whereby kitchen wastes along with cooked mixture comprising maize bhusa, mustard oil cake, pseudostem of banana, tuber, stem, and plant leaves were used to feed their animals. About 40.5 % of farmers procured their breeding stock from government farms that had good records and utilized veterinary services like timely vaccination and deworming. The diseases prevalent in the study area were swine fever, diarrhea, helminthoses, sarcoptic mange, pneumonia, etc. The litter sizes at birth (local, 4.3?±?0.45; crossbreed, 7.2?±?0.33), at weaning (local, 2.79?±?0.24; crossbreed, 6.1?±?0.21), and age at first farrowing (local, 365.39?±?7.96 days; crossbreed, 337.24?±?8.79 days) were recorded. Production costs of meat extracted from local and crossbred pigs were 1.08 $/kg and 0.86 $/kg, respectively.  相似文献

A three-phase study was conducted in high-potential farming and peri-urban area in Kikuyu Division central Kenya to obtain farm and management data and to monitor health and productivity of pigs in smallholder farms. The first phase was a cross-sectional study in which 87 farms (that had been selected from a total of 179 farms using a simple random selection) were visited once and data on important farm and management factors were gathered using semi-structured questionnaires. The second phase was a pilot study that was conducted in the 87 study farms for a period of 3 months to pretest the data-collection tools and to evaluate the general research methodology for the longitudinal study. The third phase was a prospective 12-month observational study in which health and productivity of pigs were monitored monthly in 76 herds that were still active and had participated in the previous studies.

The initial voluntary enrolment among the eligible farms was 99%. The median farm size was 1 acre. All the farms kept crossbreed pigs of Large White or Landrace (median nine pigs per farm). The median number of sows per herd was one. Most farmers engaged in farrow-to-finish pig-production system and most (60%) did not keep a breeding boar. The pigs were stall-fed the year round. Guard rails/piglet devices were present in 22% of the herds. Few (8%) farmers disinfected pig pens (especially the farrowing area). None of the farmers reported the use of vaccination against pig diseases. Most farmers (84 and 96%) indicated that they controlled for mange and worm infestations, respectively. To control mange, 50% of the farmers used acaricides, 34% used engine oil and 12% used both. Anthelmintics were used to control worms. No farmer had a particular control programme in place for both worms and mange. Artificial heating for piglets was not used in any of the farms. High costs of feeds (which were of variable qualities) lack of credit and genetically high-quality breeding boars and diseases were ranked highly by the farmers as the main production constraints. Thirteen percent (11/87) of farmers withdrew during the pilot study; 10 farmers had sold their pigs and one had died. Thirty-three percent (25/76) of the farms withdrew during the longitudinal study for various reasons that included death of pigs (3%; 2/76) and sale of the pigs (30%; 23/76)—mainly because of financial need.  相似文献

猪乙型脑炎(Japanese encephalitis,JE)是由乙型脑炎病毒(JEV)引起的一种经蚊虫传播的人畜共患病,且是严重威胁人畜健康的一种中枢神经系统的急性传染病。猪是JEV的重要储存宿主和扩增宿主,同时也是乙型脑炎的主要传染源,可引起母猪繁殖障碍和公猪睾丸炎。人是JEV的终末宿主,特别是儿童,感染发病后,死亡率可达25%。目前,仍无有效药物治疗乙型脑炎感染,而其流行区域的扩大及其优势基因型的改变为JEV防控策略带来了新的挑战。近年来,随着基因工程和蛋白质工程的发展,以及对病毒分子结构和功能的深入研究,一些新技术新手段被应用到JEV疫苗研发中,基因工程亚单位疫苗、病毒样颗粒疫苗、嵌合病毒减毒活疫苗、多表位疫苗、DNA疫苗等应运而生。在疫苗制备过程中,培养工艺的优化、病毒抗原的纯化、新型佐剂和耐热保护剂的应用等生产工艺的提升策略,为保障JEV疫苗的质量提供了新方向。文章简述了猪JEV疫苗的使用现状及研发进展,旨在为研制更安全、有效的猪JEV疫苗提供参考依据。  相似文献

为了解东莞市猪乙型脑炎的流行情况,于2009年-2011年对东莞市内猪养殖场采集猪血清4 282份,进行血清学与病原学检测。其中,应用ELISA检测方法对3 918份血清进行血清学检测,检测抗体阳性数2217份,抗体阳性率56.58%;应用实时定量RT-PCR检测方法对364份血清进行病原学检测,检测病毒阳性10份,病毒阳性检出率2.75%。结果表明,东莞市内饲养的猪存在乙型脑炎带毒的情况,免疫接种猪群的免疫效果比较理想,未免猪群存在乙型脑炎感染情况。  相似文献

Japanese encephalitis (JE) virus is a mosquito-borne flavivirus that can cause encephalitis and death in horses and humans. It is an emerging disease of international concern because it has been spreading into previously nonendemic areas. Major epidemics may occur where the virus moves into new areas, but many infections are subclinical. This article presents information on the virus, its epidemiology, and what little information has been published on the disease in horses. The methods available for the diagnosis and control of JE are described and issues raised, particularly for those countries faced with the threat of imminent incursions as the virus extends its range.  相似文献

In the Mekong delta, backyard pig rearing plays an integral role in recycling nutrients in farming systems and generating valuable cash income. However, development has been hampered by fatal epizootics of piglets and reproductive failure of sows. Diseases are named by symptoms and blindly treated with antibiotics. As antibiotics are often ineffectual, involvement of viral diseases are suspected. To identify the causative agent, we first sero-surveyed porcine reproductive and respiratory syndrome (PRRS) and pseudorabies with 478 sera from non-vaccinated pigs collected from backyard farms, state farms and slaughterhouses in Can Tho province between 1999 and 2002. Antibodies for PRRS were first detected in 2002 in backyard farms and at high prevalence in state farms with increased piglet mortality. A few backyard breeder pigs had antibodies for pseudorabies in 2000 and 2002. With compulsory classical swine fever (CSF) vaccination, we examined the relationship between vaccination and antibodies in 70 serum samples. Seventy-nine percent of vaccinated breeders had CSF antibodies-higher than expected with irregular vaccination. Since circulation of CSF virus was suspected, isolation was attempted at 10 farms with fatal epizootics between 2002 and 2003. The viruses were detected at all farms and clustered within genogroup 2, despite vaccines corresponding to genogroup 1. This study demonstrated virologically/serologically the existence of PRRS, pseudorabies and CSF viruses in the Mekong delta of Vietnam. We also identified CSF as a cause of piglet mortality that disastrously affected backyard farming. Vaccine standardization and proper instructions are needed to simplify diagnosis and complement established simultaneous vaccination of sows with piglets.  相似文献
