Platelets in blood collected from pigs identified as normal (21) or stress susceptible (25) on the basis of their response to halothane challenge, were subjected to electron microscopic examination in order to test whether the ultrastructural features of stress susceptible pigs exhibit any deviation from those of normal. The most striking feature of the platelets from stress susceptible pigs was the extent of dilatation of the open canalicular system (OCS). The difference in platelet morphology between normal and stress susceptible pigs was consistent regardless of the anticoagulants used for blood collection indicating that the platelet alteration may be an inherent component of the porcine stress syndrome.  相似文献

A study was undertaken to test whether a previously reported alteration in platelet morphology could be of predictive value for the detection of stress-susceptibility in pigs. Platelets from 20 normal pigs, nine pigs classified as stress-susceptible on the basis of their response to halothane challenge, and 11 siblings of halothane reactors belonging to two different breeds were subjected to electron microscopic examination. A quantitative analysis of electron micrographs, based on the extent of dilatation of the open canalicular system in platelets and the percentage of affected platelets, revealed that halothane reactor pigs could be distinguished from normal animals on the basis of their open canalicular system score. The discrete nature of the score categories in siblings indicates that platelet alteration may be an inherent component of the porcine stress syndrome and suggests that some of the false negatives in the halothane test may be identified as stress-susceptible on this criterion. Further studies involving a larger number of halothane reactors and siblings are needed to ascertain the consistency of the open canalicular system features and eventually, to develop a simple test system based on platelet alterations for the detection of stress-susceptibility in pigs.  相似文献

The concentrations of phenytoin (DPH) in the CNS of adult dogs given the drug by IV injection, continuous IV infusion, or repeated daily oral administration and of newborn pups given DPH by IV injection were consistently proportional to serum concentrations of the drug at the time of blood sample collections. Adult dogs injected IV with [14C]DPH failed to show predilection of the drug for the 13 anatomic brain sections sampled. The pharmacokinetics of DPH were studied in adult and neonatal dogs given 15 mg of the drug/kg of body weight as a single IV injection. The mean half-life of the drug in adult dogs injected IV was 4.5 hours. The serum half-life of injected DPH was lowest in pups 30 days of age when compared with that in other age groups. The serum half-life of injected DPH was increased in adult dogs under pentobarbital anesthesia.  相似文献

The relationships of various iron-related analytes were evaluated in 95 dogs. Liver and spleen nonheme iron content was determined coulometrically on acid-digested tissue specimens. Serum iron concentration and total iron-binding capacity also were measured coulometrically, whereas serum ferritin concentration was measured by ELISA. Significant (P less than 0.0002) correlation was found between serum ferritin concentration and nonheme iron stores. Significant correlation was not found between nonheme iron stores and serum iron concentration or total iron-binding capacity. Serum ferritin concentration should provide a convenient and relatively noninvasive means of estimating iron stores in dogs.  相似文献

The measurement of serum pepsinogen concentrations by enzymatic method and immunoassay provides diagnostic values and should be helpful in the detection of gastric diseases related to a rise of blood pepsinogen. In the present study, the correlation between a conventional enzymatic method and a recently developed radioimmunoassay (RIA) for serum pepsinogen A was investigated. A total of 123 sera samples of porcine foetuses (n = 28), adult healthy pigs (n = 56), pigs with parakeratosis (n = 25) and pigs with ulceration of the pars oesophagea (n = 14) were tested. Overall, there was a slight correlation between the two methods (r = 0.60). In relation to individual animal groups, the correlations (r) were 0.39 (P>0.05), 0.74 (P<0.001), 0.19 (P>0.05) and 0.34 (P>0.05) in foetuses, healthy pigs, pigs with parakeratosis and pigs with ulcers, respectively. In both methods, pepsinogen concentrations (means+/-SE) were significantly higher (P<0.05) in pigs with parakeratosis (1778 +/- 86.00 mUTyr/L; 690 +/- 53.00 ng/mL) and in pigs with ulcers (2026 +/- 153.00 mUTyr/L; 1747 +/- 94.00 ng/mL) when compared to healthy pigs (935 +/- 58.00 mUTyr/L; 275 +/- 35.00 ng/mL). The proteolytic method gave a significant increased activity (P<0.05) in foetuses (1150 +/- 82.00 mUTyr/L) vs. (935 +/- 58.00 mUTyr/L) in healthy adult pigs, indicating an additional proteolytic activity in the sera of foetuses or neonates.  相似文献

Mannan-containing products are capable of modulating immune responses in animals. However, different products may have diverse immunomodulation. The experiment was conducted to examine effects of mannan oligosaccharide (Actigen; ACT) on growth performance and serum concentrations of antibodies and inflammatory mediators in weanling pigs (Sus scrofa) experimentally infected with porcine reproductive and respiratory syndrome virus (PRRSV). A total of 32 PRRSV-negative pigs (3 wk old) were randomly assigned from within blocks to 1 of 4 treatments in a 2 by 2 factorial arrangement [2 types of diet: control (0%) and ACT addition (0.04%); and with and without PRRSV] in a randomized complete block design. Pigs were blocked by initial BW within sex. Ancestry was equalized across treatments. Pigs (8/treatment) were kept individually in each pen. After 2 wk of an 8-wk period of feeding the treatments, pigs received an intranasal inoculation of PRRSV or sham medium at 5 wk of age. Infection by PRRSV decreased ADG, ADFI, and G:F throughout the experiment (P < 0.01). Actigen did not affect ADG (P = 0.450), but decreased (P = 0.047) ADFI from 28 to 42 days postinoculation (DPI). During that time, ACT improved G:F in infected pigs but not in sham controls (interaction, P = 0.009). Dietary ACT did not affect viremia in infected pigs (P > 0.05), but increased PRRSV-specific antibody titer at 35 DPI (P = 0.042). Infection with PRRSV induced the febrile responses of pigs from 3 to 10 DPI (P < 0.001) with return to normal at 14 DPI. During the experimental period, the rectal temperature of pigs was found slightly elevated by ACT (P = 0.045). Infected pigs had greater serum concentrations of IL-1β, tumor necrosis factor (TNF)-α, IL-12, interferon (IFN)-γ, IL-10, and haptoglobin (Hp) than sham controls (P < 0.001). These results indicate that PRRSV stimulated secretion of cytokines involved in innate, T-helper 1, and T-regulatory immune responses. Actigen tended to decrease the serum TNF-α concentration regardless of PRRSV (P = 0.058). The ACT × PRRSV interaction was significant for IL-1β (P = 0.016), IL-12 (P = 0.026), and Hp (P = 0.047), suggesting that infected pigs fed ACT had greater serum concentrations of these mediators than those fed the control. The increases in IL-1β and IL-12 may favorably promote innate and T-cell immune functions in infected pigs fed ACT. Feeding ACT may be useful as ACT is related to increased PRRSV antibody titers and G:F in infected pigs at certain times during infection.  相似文献

The serum concentrations of serotonin and lipids--triglycerides, total cholesterol, low density lipoprotein, high density lipoprotein and very low density lipoprotein--were determined in 18 normal dogs and 23 dogs diagnosed as aggressive on the basis of interviews with their owners and an assessment of their behaviour with reference to a canine overt aggression chart. The serum serotonin levels in the aggressive dogs were significantly lower than in the normal dogs (P<0.01), but the differences in serum lipids between the two groups were not statistically significant.  相似文献

The activity of digestive enzymes was determined in the gastrointestinal tract of newborn, 1-, 3- and 5-week-old piglets and in adult pigs. The pancreas of newborn piglets contained considerable ribonuclease (RNase) activity, which continued to increase with age. After the initial values an opposite tendency was found in the intestinal contents, probably due to the increase of proteolytic degradation with advancing age. Serum RNase showed little age dependence. The time-course of development of pancreatic RNase resembled more that of proteolytic enzymes than tha of amylase. The data indicate that a high pancreatic secretion of RNase commences much before the appearance of RNA in the diet.  相似文献

The objective of this study was to investigate the relationship between marbling and serum leptin concentrations. First, we compared serum leptin concentrations between Japanese Black cattle and Holstein calves, and found that the mean serum concentration was significantly higher (P < 0.05) in Japanese Black cattle (2.67 ± 0.41 ng/mL, n = 13) than in Holstein calves (1.79 ± 0.43 ng/mL, n = 5). We then examined the effects of roughage intake or vitamin A on the serum leptin concentrations in Japanese Black cattle. Although the leptin concentrations for the high roughage group were significantly higher than those for the low roughage group at 4, 7 and 20 months old, there was no significant difference in the carcass characteristics of the two groups. In addition, although there was a significant difference in the marbling score between the high vitamin A group and low vitamin A group, there was no significant difference in serum leptin concentrations between the groups. These results suggest that although serum leptin concentrations were not significantly correlated to the marbling scores they may be related to the genetic advantage for marbling in Japanese Black cattle.  相似文献

In connection with transfer of sheep from the lowland near Oslo to mountain pastures at an altitude of 1,200 m above sea level, investigations were carried out in 37 animals to study the effect of physical stress on serum enzymes and other blood constituents. The sheep were adult ewes and lambs. About half of the animals had been accustomed to outdoor life on pasture for more than one month, while the others were moved directly from indoor feeding. Blood was collected before departure, after six hrs. of long-distance transportation by lorry, and after three hrs. of subsequent continuous herding on foot. The following blood components were determined: Aspartate aminotransferase (AspAT = GOT), alanine aminotransferase (A1AT = GPT), α-hydroxybutyrate dehydrogenase (HBD), total lactate dehydrogenase (LDH), LDH isoenzymes, alkaline phosphatase, calcium, inorganic phosphorus, magnesium, blood sugar, total serum proteins, and haemoglobin.In summary, it may be said that the lambs reacted with greater changes of the blood components than adult animals, and that untrained, indoor fed lambs were distinctly more sensitive than those taken from pasture. The “indoor” lambs showed a statistical significant increase from the initial values in AspAT, HBD, total LDH, the isoenzymes LDH3 and LDH4, and blood sugar. Significantly decreased values were recorded in Ga, P, Mg, and total serum protein. Some of these changes, as in Mg and P, were most pronounced after transportation, while elevations of serum enzyme levels continued to increase during the subsequent herding.Based upon the shift in LDH isoenzyme distribution towards a more cathodically dominated pattern it is supposed that the main origin of increased serum enzyme activity was skeletal muscle.  相似文献

We found previously that porcine growth hormone (pGH) causes an increase in growth rate with a concurrent improvement in carcass composition in pigs. The somatomedin, insulin-like growth factor 1 (IGF-1), is though to play a major role in mediating some of the anabolic actions of GH, while the glucocorticoid hormones are potential counter-regulators of these effects. The present study was conducted to determine the temporal and dose-response relationship between GH administration and serum IGF-1 and cortisol concentrations in pigs. Twelve Yorkshire barrows, fitted with femoral artery catheters, were injected (im) with either 0, 10, 100 or 1,000 micrograms/kg pGH. Blood sampling began 40 min prior to pGH injection and was continued for 37 h. Serum GH, IGF-1 and cortisol concentrations were determined by radioimmunoassay. In control animals, serum GH concentrations ranged from 1.6 to 5.7 ng/ml over 37 h. In the animals treated with increasing doses of pGH, peak serum GH concentrations reached 28, 112 and 286 ng/ml and levels remained elevated for 4, 12 and 24 h, respectively. Serum IGF-1 concentrations were elevated by pGH after a lag time of 4 to 6 h. When the IGF-1 concentrations were integrated over time, the response appeared to be dose-dependent, with an ED50 of 710 micrograms/kg body weight (BW). Data for serum cortisol concentrations showed a great deal of individual variation. A transient increase in cortisol was observed, but only in the group treated with 1,000 micrograms pGH/kg BW. Vitamin E (alpha-tocopherol) is an antioxidant vitamin important in protecting unsaturated fatty acids in lipid membranes from peroxidation. Variation in collection, storage, and shipping conditions of samples can potentially lead to breakdown of vitamin E prior to analysis. Therefore, the purposes of this project were 1) to determine the stability of vitamin E in refrigerated and frozen porcine liver and serum and 2) to evaluate the effects of red blood cell (RBC) hemolysis on porcine serum vitamin E concentrations. Porcine liver and nonhemolyzed serum were collected and stored refrigerated or frozen. Samples were analyzed for vitamin E immediately or on days 2, 3, 7, or 14. In addition, porcine RBCs were added to normal serum at concentrations from 1 x 10(6) to 1 X 10(9) RBC/ml and hemolyzed by freeze-thaw prior to analysis for vitamin E or products of lipid peroxidation.  相似文献

The level of P4 at the time of embryo transfer (ET) is important. P4 concentrations and numbers of corpora lutea for 126 recipients were evaluated. Nuclear transfer embryos were transferred into 126 surrogates. 11 maintained their pregnancy until full-term delivery, 17 miscarried, and implantation failed in 98 animals. P4 levels in the full-term group were significantly different from those of the pigs that aborted or in which implantation failed (p < 0.05). However, the numbers of corpora lutea were not significantly different. These findings indicate that the concentration of progesterone can be an important factor for successful ET in pigs.  相似文献

To establish the relationship between serum levels of three acute phase proteins, haptoglobin (Hp), C-reactive protein (CRP) and serum amyloid A (SAA), and the occurrence and severity of lesions at slaughter, a study was carried out using 70 fattening pigs from a finishing unit. Pigs were divided into three groups: Group 1 (25 pigs with clinical signs of disease), Group 2 (25 apparently healthy pigs with lesions at slaughter) and Group 3 (20 apparently healthy pigs with no lesions at slaughter). Serum levels of CRP, SAA and Hp were significantly higher in pigs with clinical signs of disease than in apparently healthy animals. Additionally, in apparently healthy pigs, serum levels of Hp and CRP were significantly higher in animals with lesions than those without lesions. The extent and severity of lung lesions were related to serum levels of Hp.  相似文献

This study examined the effect of stress classification (stress-positive, stress-carrier, stress-negative) of pigs on selected properties of postmortem muscle, including protein solubility and degradation of proteins such as titin. Longissimus muscle samples were removed 45 min postslaughter, divided into samples, and stored at 0 to 2 degrees C for analysis at 0, 1, 3, 5, and 7 d postmortem. Whole-muscle samples (homogenates) and purified myofibrils were prepared from each sample for analysis by SDS-PAGE. A portion of each muscle sample also was extracted 1) with a low-ionic-strength solution to obtain a sarcoplasmic protein fraction and 2) with two different high-ionic-strength solutions to obtain a myofibrillar/cytoskeletal protein fraction for measurement of protein solubility and for analysis of extracts by SDS-PAGE. No significant differences were observed between muscle from stress-negative and stress-carrier animals in this study. Sarcoplasmic (P less than .05) and myofibrillar/cytoskeletal (P less than .01) protein solubility was lower in muscle samples from stress-positive animals than in muscle samples from stress-carrier and stress-negative animals at all postmortem times studied. The high molecular weight protein titin was degraded more slowly postmortem in muscle from stress-positive than in muscle from stress-negative animals, as observed by SDS-PAGE analysis of whole-muscle samples (homogenates) an myofibrils. The combination of lowered protein solubility and reduced rate of postmortem degradation of structural proteins such as titin may explain, at least in part, the reduced quality and protein functionality of muscle from stress-positive pigs.  相似文献
