运用组织学技术对36头处于发情周期的健康成年母牦牛卵巢卵泡的结构与发育状况进行了观察。结果表明,牦牛发情周期中卵巢卵泡发育的组织结构与其他牛基本相似。每对卵巢中原始卵泡数和生长卵泡数在发情前期、发情期、发情后期和发情间期之间差异不显著(P〉0.05);发情前期的囊状卵泡数与发情期、发情后期和发情间期之间差异不显著(P〉0.05),发情期和发情后期均与发情间期差异显著(P〈0.05)。闭锁生长卵泡数在4个时期之间差异不显著(P〉0.05);发情期的闭锁囊状卵泡数与其他3期之间差异极显著(P〈0.01),发情前期和发情间期均与发情后期差异显著(P〈0.05)。各级卵泡的闭锁形式各有特点。在生长卵泡、囊状卵泡及相应闭锁卵泡的卵泡膜中均见到了胶原纤维和网状纤维。 相似文献

选用48头成年母牦牛,采用光镜和电镜技术对发情周期中不同时期黄体的组织结构进行了观察。结果表明,牦牛黄体主要由2种细胞组成,即颗粒黄体细胞(granulasa lutein cell,GLC)和膜黄体细胞(theca lutein cell,TLC),其特征性变化主要表现在GLC和纤维的分布。成熟黄体中GLC及其胞核的平均直径分别为36.6μm和15.2μm,而TLC则分别为14.4μm和10.9μm。黄体细胞胞质中线粒体的比例随黄体的成熟而增高;脂滴在Ⅰ期黄体时较多,Ⅱ期时减少,Ⅲ期时显著增多,后又减少;滑面内质网也随黄体的成熟而增加,并随黄体的退化发生膨胀。黄体组织中有同心圆状或同心轮状的粗面内质网膜系统。黄体细胞间主要是缝隙连接,偶见中间连接。黄体组织中存在窗孔型毛细血管. 相似文献

为研究牦牛初乳期乳腺特异表达的基因,采用抑制消减杂交和斑点杂交法对麦洼牦牛初乳期(产犊后2 d)和常乳期(产犊后18 d)特异表达的基因进行了分离鉴定。结果发现2个在初乳期间特异表达的EST片段,并命名为LAO和COL1α1。测序结果LAO片段长698 bp,GenBank比对,其核苷酸序列与西藏黄牛(Bos taurus)、小家鼠(Mus musculus)、家犬(Canis familiaris)等的L-氨基酸氧化酶基因有较高的序列相似性,其序列一致性分别达到96%、73%、72%;COL1α1片段长337 bp,比对结果,其序列与Bos taurus、马鹿(Cervus elaphus)、家犬(Canis lupus fa-miliaris)等的Ⅰ型胶原蛋白α1有较高的序列相似性,其序列一致性分别为100%、100%、99%。2个序列与牛基因组的Blast分析表明,LAO片段定位于3号染色体LOC782545区,该区域编码1个L-氨基酸氧化酶基因;另一片段定位于19号染色体LOC615867区,该区域编码1个Ⅰ型胶原α1基因。研究表明,L-氨基酸氧化酶基因和Ⅰ型胶原蛋白α1基因是牦牛初乳期乳腺特异表达蛋白,这2个基因在牦牛初乳期乳腺组织中较高的表达水平可能与初乳期乳腺特殊的生理状态有关。 相似文献

为了解羊驼的生殖生理特性,采用大体解剖与组织学方法对羊驼副性腺的形态结构进行了研究。结果显示,羊驼副性腺有前列腺和尿道球腺,没有精囊腺。尿道球腺呈圆形,位于坐骨弓处,前列腺位于尿生殖道起始部的背侧,每一腺体都有腺管开口于尿生殖道;羊驼副性腺的基本组织结构和其他哺乳动物相似,但有较小的差异。尿道球腺为有许多弯曲分枝的复管泡状腺,分泌物较为黏稠,PAS染色强阳性,呈鲜红色;前列腺为复管泡状腺,前列腺的分泌物是一种稀薄的蛋白样液,PAS染色阳性,染色较浅。结果证实,羊驼前列腺和尿道球腺可分泌较多的糖物质,使精子生存较长时间;羊驼繁殖慢的特点可能与其生存环境差,易造成流产或妊娠期长且为单胎有关,而并不是精液本身的问题。 相似文献

This study was conducted to investigate structural transformations of the goat pineal gland during postnatal development. The pineals of newborn, 4-week-, 4-month-, 1-year- and 3-year-old male goats were prepared for qualitative and quantitative investigations at both light and transmission electron microscopy levels. In the first 4 months after birth, the pinealocytes developed very intensively, which was connected with enlargement of their volume as well as distinct qualitative and quantitative transformations of their cytoplasmic organelles. Parallel to these changes, the pineal parenchyma underwent deep reorganization and the mosaic pattern, characteristic for newborns, disappeared. At the same time the meshwork of blood vessels supporting the goat pineal gland developed conspicuously. From fourth months to one year of postnatal life continuation of the growth of some pinealocyte organelles and very intensive development of all components of the pineal connective tissue were observed. In the three-year-old goats some decrease in the relative volume of most cytoplasmic structures of pinealocytes as well as increase in collagen fibers in stroma were noticed. The investigations established intensive growth of the goat pineal to the age of 4 months, which included mainly some increase in the pinealocyte and their structures. The pineal gland of 4-month- and 1-year-old goats showed morphological features which are considered as symptoms of high secretory activity. The structure of the pineal gland in 3-year-old animals and the changes observed both in their pinealocytes and connective tissue in comparison with 1-year-old individuals point to some decrease in the goat pineal activity in this age. 相似文献

为研究水禽尾脂腺的结构、功能及其受环境的影响,将180只1日龄商品代三水白鸭随机分为水养和旱养2个处理,每个处理3个重复,每重复30只,公母各半.42日龄后每个重复各取2只鸭(公母各半)屠宰,取其尾脂腺做切片观察.结果表明:①肉鸭尾脂腺细胞分为明显的外围暗区和内部明区两个区域,结合透射电镜切片可以发现细胞器都位于外围暗区(胞器区),而内部(脂肪区)则充满了大小不一的脂肪滴;②42日龄水养组的三水白鸭尾脂腺细胞外围暗区具有类似于线粒体内嵴的向内突起,而旱养组的则无向内突起;③尾脂腺细胞内细胞器主要包括线粒体、内质网和细胞骨架组织,但没有观察到高尔基体;④旱养条件下,三水白鸭尾脂腺细胞内的线粒体和内质网都较水养条件下更为发达. 相似文献

In the present study, the authors describe a simple method to isolate chromosomes from eimerian oocysts and to submit them to sequential study by light and electron microscopy. This method includes a reliable and reproducible technique for transferring eimerian chromosomes from slides to grid that fulfills the essential requirements for generalized use in cytogenetics. In addition, this method overcomes the difficulty of the resistance of protozoan oocysts to disruption and permits the release of intact meiotic chromosomes. The observation by the authors of synaptonemal complexes in meiotic chromosomes of different Eimeria species by applying the above-mentioned method to oocysts revealed its importance to future applications. 相似文献
