The efficacy of Brucella Melitensis Rev 1 vaccine (Rev 1) for the prophylaxis of Brucella ovis ram epididymitis was evaluated. Twenty-nine 3-month-old rams were vaccinated with 2 X 10(9) Rev 1 and 14 were revaccinated with 5 X 10(8) at 14 months of age. Six rams remained unvaccinated as a control group. All rams were challenged with 5 X 10(8) B. ovis at 21 months of age. Before being slaughtered 8 weeks later, only one vaccinated ram developed epididymitis while four of the six control rams developed testicular alterations. Genital and selected extragenital organs and lymph nodes were removed at slaughter and inoculated on selective media. B. ovis was isolated from 26.6% of the vaccinated rams, 21.4% of the revaccinated rams and 100% of control rams. Portions of epididymis, testes and vesicular glands were also used for pathological studies. More severe lesions were observed in control rams than in vaccinated ones. In conclusion, these results show that vaccination of young lambs, followed or not by revaccination, is a suitable method for the prophylaxis of B. ovis infection of rams. 相似文献

Abstract Extract An infectious epididymitis of rams caused by Brucella ovis (Buddie, 1956) infection, first described in Australia (Simmons and Boyes, 1953) and New Zealand (Buddie and Boyes, 1953) was recognized subsequently in Czechoslovakia (Gdovin et al, 1955), the United States (McGowan and Shultz, 1956), South Africa (Van Rensburg et al, 1958), Rumania (Tudoriu, et al, 1958), and South America (Dr Justo Zomara B, 1961, pers. comm.). As the infection can affect ram fertility and, further, can be responsible for abortion in ewes and perinatal mortality in lambs, attention has been directed to the development, evaluation, and application of control measures in a number of important sheep-raising countries. 相似文献

Sera from rams vaccinated with antigens extracted chaotropically from Brucella ovis by potassium thiocyanate treatment were used to optimise a whole-cell, enzyme-linked immunosorbent assay (CELISA) and to monitor the temporal serological response of rams which had been challenged with infected semen by the intranasal or intrapreputial route. Three patterns of CELISA response were detected. Thirteen of 15 rams intranasally challenged did not respond serologically (pattern 1 or nil response). Only one of 15 rams in the intranasal group exhibited a rise and fall response with CELISA (pattern 2), while another showed a rise and surge response (pattern 3). The numbers of rams in the intrapreputial group which displayed a pattern 1 or 2 or 3 response were four, nine and two, respectively. No ram with a pattern 2 response excreted B ovis in the semen or showed any other evidence of infection, whereas rams with a pattern 3 response excreted B ovis in the semen and developed palpable lesions. Intrapreputially challenged rams that were CELISA-positive consistently mounted an antibody response against B ovis about two to four weeks earlier than intranasally challenged rams. 相似文献

A comparative study was made of the serological response of rams to an inactivated Brucella ovis saline in-oil vaccine administered either once or twice by either the subcutaneous or the intraperitoneal route. The serological response of rams to two spaced doses of vaccine was more consistent and more persistent than when a single dose of vaccine was used. The rise in titre was more rapid and the final titre of greater magnitude when the subcutaneous route of administration was used in comparison with the intraperitoneal route. On a serological basis, the most satisfactory vaccination technique investigated consisted of two subcutaneous injections of vaccine administered ten weeks apart. The least satisfactory technique was a single intraperitoneal injection. 相似文献

Lesions induced by inoculation of Brucella ovis into the epididymis were compared in rams previously vaccinated with B. ovis bacterin and unvaccinated rams. Inoculation of killed B. ovis did not produce significant lesions in either group whereas prior vaccination exacerbated epididymal lesions following inoculation of live B. ovis. Increased numbers of neutrophils, macrophages and lymphocytes were present in the interstitium and neutrophilic infiltration of the epididymal duct epithelium and intraepithelial cyst formation was more prominent. The inflammatory response surrounding extravasated spermatozoa was more severe in vaccinated rams but it was not determined if the response was directed at spermatozoa or intermixed brucellae, or both. 相似文献

Complement fixation tests using three B. ovis antigen preparations in warm fixation tests (WCFT) and cold fixation (CCFT) tests were done on 541 ram sera. Semen samples from the same rams were examined culturally to identify B. ovis excretors. The CCFT, using an antigen prepared by heat extraction of B.ovis cells, had a sensitivity of 97% in 124 rams which were shedding B.ovis. The specificity was 99% in 144 rams from non-infected flocks. Seventy-seven per cent of 156 rams which reacted to this test were shedding B. ovis in their semen. Tests with other antigens were inferior in sensitivity and/or specificity. The WCFT gave lower titres than CCFT. Vaccination caused large numbers of false positive reactions in 4 flocks. 相似文献

The sensitivity and specificity of the complement fixation, gel diffusion and ELISA tests for the diagnosis of Brucella ovis infection of rams have been compared using three different antigenic preparations. The antigens obtained by petroleum ether - chloroform - phenol, or cold saline extractions gave poorer diagnostic results than those obtained by hot saline extraction in all the tests. The best sensitivity was obtained with the ELISA (97.6 per cent) followed by the gel diffusion (96.4 per cent) and complement fixation tests (92.7 per cent). The gel diffusion test detected as positive the two rams negative in the ELISA, while the complement fixation test did not improve the sensitivity of the other tests. Under these conditions all the tests were 100 per cent specific when testing sera from rams free of B ovis. 相似文献

A study was conducted to estimate the seroprevalence of Brucella ovis infection in rams in the Estrie and Bas-Saint-Laurent regions (Quebec). Rams sera (n = 258) were serologically evaluated from 224 rams in 30 commercial flocks and from 34 rams at 2 slaughterhouses by using an enzyme linked immunosorbent assay. Epididymides and testes were examined by palpation on farms and microscopically for culled rams. No ram was seropositive to Brucella ovis or had lesions suggestive of brucellosis from the farm or slaughterhouse surveys. 相似文献

Rams shedding Brucella ovis in semen but without palpable abnormalities of the epididymides were treated with long-acting oxytetracycline for 15 days and dihydrostreptomycin for 7 days (n = 9) or conventional oxytetracycline and dihydrostreptomycin (n = 9) for 7 days. Nine rams were not treated. More treated rams were considered to have satisfactory breeding soundness examination results at posttreatment weeks 3, 7, 12, and 19. Nontreated rams continued to shed B ovis in semen. After treatment, B ovis was not recovered from 78% of rams given long-acting oxytetracycline and dihydrostreptomycin or from 89% of rams given conventional oxytetracycline and dihydrostreptomycin. At week 21, all rams were euthanatized, and specimens of the testes and epididymides were bacteriologically cultured for B ovis. Brucella ovis was not recovered from the testes of rams or from the epididymides from rams not shedding the organism in the semen. In one treated ram, B ovis was recovered from the semen but not from other tissues. All rams remained ELISA-positive, with the exception of 2 treated rams that ceased shedding B ovis in semen immediately after treatment was started; both these rams became ELISA-negative on the last examination at week 19. 相似文献

Serology and semen culture for the diagnosis of Brucella ovis infection in chronically infected rams
The serological response to Brucella ovis and the shedding of the organism in semen was followed for a period of 13-14 months in 42 naturally infected rams. Most rams remained chronically infected and excreted the organism in their semen throughout the investigation. B. ovis was isolated from 87.9% of the semen samples from the infected rams. The most common sites from which B. ovis could be isolated at necropsy were the epididymides and accessory sexual glands. In one ram the organism was isolated from lung, spleen, kidney and iliac lymph nodes. Three rams ceased to shed B. ovis in their semen during the course of the investigation. Seventy-five (11%) of 686 sera from infected rams were negative in the complement fixation test (CFT) although 76% and 77% of CFT-negative sera were positive in the gel diffusion precipitin test (GDT) and enzyme labelled immunosorbent assay (ELISA) respectively. The high incidence of CFT-negative infected rams was due to the selection for the investigation of many rams with histories of negative or vacillating CFT titres. Sera from five rams which never shed B. ovis in their semen reacted erratically in the three serological tests. AIM: To describe the performance characteristics (sensitivity and specificity) of an enzyme-linked immunosorbent assay (ELISA) for the diagnosis of Brucella ovis infection in rams. METHODS: Sera from a negative (n = 2535) and a positive (n = 589) reference population were tested in an ELISA for anti-B. ovis antibodies and cut-off values calculated from the raw, log10-transformed and fitted data. Statistical methods were used to fit curves to the frequency distribution of the data and receiver-operated characteristics analysis used to optimise the cut-off values. RESULTS: Analysis of the frequency distribution of the positive ELISA values suggested a normal distribution of the data, whereas, in the case of the negative population, a Pearson type IV curve appeared to best fit the data. The cut-off values calculated as the mean plus 1.96 standard deviations (s.d.) from the raw, log-transformed and fitted ELISA data did not differ markedly. The differences were much greater at the mean plus 3.09 s.d. cut-off, with the cut-off value calculated from the log-transformed data giving a much better estimate of specificity. Optimisation (minimisation of classification error) of the cut-off calculated from the fitted curves suggested varying cut-off values, depending on the prevalence of B. ovis infection. Discussion: Calculation of cut-off values from curves that were fitted from the observed data give more accurate estimations of the performance characteristics of an assay than traditional calculations from observed values. They also allow the calculation of optimal cut-off values taking into account the prevalence of B. ovis infection and give additional information about the performance of the assay at cut-off values varied according to the epidemiological situation. 相似文献

Primary hypoparathyroidism was diagnosed in three St. Bernard bitches. Anorexia, behavioural changes, muscle tremors, seizures, panting respiration, and cataracts were the clinical signs observed. The serum concentration of calcium was low, the phosphorus concentration elevated, and the immunoreactive parathyroid hormone level low in all dogs. The aetiology of the hypoparathyroidism was not determined in any of the dogs. Treatment with synthetic vitamin D (1,25-dihydroxycholecalciferol) and an oral calcium supplement was successful in restoring and then maintaining a normal concentration of serum calcium in two of the dogs. 相似文献
