The drinking rate of cod larvae 1–7 days post hatching was measured from the uptake of3H-labelled dextran (MW = 70000) admixed in the incubation seawater (34 ppt 5°C). The drinking rate increased gradually from 0.15% to 0.59% of the larval body weight on day 1 and day 7 respectively. This increase in drinking rate correlated with an observed decrease in the volume of the yolk sac and its water store. Autoradiographs showed the labelled dextran to be confined to the intestine. Electron micrographs showed an open mouth communicating with the oesophagus and the intestine in cod larvae at the time of hatching. Chloride cells were present on the opercular folds but not on the vestigial, developing gills. The data indicate that the water acquisition mechanism of larval cod is similar to that of adult marine fish. 相似文献

Changes in lipid class and fatty acid composition during development in pike (Esox lucius L) eggs and larvae 总被引:1,自引:0,他引:1
To establish the changes which occur during embryogenesis and early larval development, eggs, yolk sac larvae and swim-up
larvae of pike were examined for lipid class and fatty acid composition. At a water temperature of 15.5°C, the embryonic phase
was short (6 days) and characterized by a 41.3% decline in the lipid content of eggs, accompanied by large reductions in the
amount of phosphatidylcholine (41.4% decrease), sterol esters and triacylglycerols (respectively a 41.2% decrease and a 58.1%
decrease), but not phosphatidylethanolamine which increased markedly (35.6%). By the time of yolk sac absorption (7 to 11
days after fertilization) the larvae remained inactive and a limited utilization of lipids was observed. Yolk sac phosphatidylcholine
was selectively incorporated into larval bodies while the levels of other lipid classes remained unchanged in the yolk. When
the swim bladder was filled and the swimming stage was reached (11 days to 13 days af), the yolk was completely depleted and
yolk phosphatidylcholine together with yolk triacylglycerols were catabolised. Yolk phosphatidylethanolamine and yolk sterol
esters were partly incorporated into the body lipids. In the subsequent swim-up larval stage (13 to 15 days af), a steady
decrease in lipids was observed (41.6%). Fluctuations in the levels of polyunsaturated fatty acids, monounsaturated fatty
acids or saturated fatty acids examined from eggs or larvae were consistent with changes in lipid classes during pike development.
During yolk sac absorption, pike incorporated yolk PUFA released on hydrolysis of phosphatidylcholine into the larval body.
The results are discussed with reference to water temperature and in relation to the ontogenic and ecological context of pike
development.

The present work reports a characterization of mean wet weight and moisture, the lipid class and fatty acid (FA) composition from the total lipids (TL), of both culture and wild eggs of the cuttlefish, Sepia officinalis , throughout the embryonic development. Additionally, reproductive data, such as the number of spawnings, number and mean weight of eggs and duration of spawning period of cultured cuttlefish is provided. Both types of eggs were similar in mean wet weight, moisture content, TL content and lipid composition throughout embryonic development. Females from the cultured group spawned 13 times and laid 8654 eggs in 64 days, with a mean weight of 0.607 ± 0.179 g. A sex ratio of 1.57 (11♀ for 7♂) promoted an individual fecundity of 787 eggs/♀ (the biggest until now on our culture facilities), which might be related to increased bottom areas. The TL increased with day/stage of embryonic development ( P < 0.05) only in the cultured egg group. However, no differences were found on TL between culture and wild eggs at the same day/stage ( P > 0.05). Eggs displayed predominant levels of phosphatidylcholine, phosphatidylethanolamine (PE), cholesterol and triacylglycerol at the end of embryonic development. Polar and neutral lipids of both eggs groups remained consistently proportional (∼50% for each lipid fraction) and a significant increase ( P < 0.05) was observed in phosphatidylserine, PE and free FA throughout the embryonic development. In either egg type and day, 16:0, 18:0, 20:5n-3 and 22:6n-3 accounted for approximately 70 g Kg−1 of all FA and saturated and n-3 totals seemed to have the same proportion in the cuttlefish eggs. We carried out an experiment to determine how rapidly the early incubation temperature of Atlantic cod eggs can be increased without affecting normal embryonic development and hatching. Atlantic cod eggs were incubated at a constant low temperature (4.5 ± 0.5°C; T5 – control) and four temperature increment treatments where the temperatures were increased stepwise from 4.5°C at zygote stage to 9.5 ± 05°C (T1‐8 h, T2‐32 h, T3‐64 h and T4‐96 h). Embryonic cell symmetry, embryonic mortality, hatching success and larval skeletal abnormalities, length and yolk sac volume were recorded. Larval samples were also taken at hatch for histological analysis. Except for higher egg mortality and lower hatching success in the T1, the differences among experimental groups were minor. Cell asymmetries and embryo mortalities were not significantly different between the control and T2–T4 treatment groups. Control larvae were significantly longer and had smaller yolk reserves at hatch than T1–T4 larvae and larvae from T2 had the largest yolk reserves. Tissue and organ histology of hatched larvae were similar. Considering embryonic cleavage pattern, hatching success and larval morphology and histology, a gradual increment of temperature in 32 h seems to be the better choice for future developmental programming studies in Atlantic cod. 相似文献

Embryonic development of common dentex (Dentex dentex) was investigated at nine different constant water temperatures (8°C, 10°C, 12°C, 14°C, 16°C, 18°C, 20°C, 22°C and 24°C). The observed effects were compared using of regression analysis. Constant water temperatures between 12°C and 18°C were found to support successful embryonic development. A negative relationship between the rate of embryonic development and incubation temperature was observed. While embryonic development was completed within this range (12°C–18°C), there was no cell division at water temperatures of 8°C and 24°C. Total mortalities were observed at the 128 cleavage stage at a temperature of 10°C, and after the beginning of gastrulation at 20°C and 22°C. 相似文献

The effect of two disinfectants on eggs and larvae of Baltic cod, Gadus morhua, was investigated. The eggs were disinfected for 10 min using various concentrations of either glutaraldehyde (100, 200, 400, 600 and 800 mg L?1) or iodophor (10, 50, 100 and 150 mg L?1), 1–4‐days post‐fertilization. Bactericidal effect of disinfection, survival to hatching, hatching success and larval abnormalities were assessed. Larval survival was recorded at 5‐, 10‐ and 15‐days post‐hatch (dph). Although Baltic cod eggs have an unusually thin chorion, they could tolerate surface disinfection. A reduction in bacterial growth was observed with increased concentrations of disinfectant (3.0 × 107–1.6 × 101 CFU mL?1). Abnormalities in newly hatched larvae were not related to disinfection. Survival of the yolk sac larvae was significantly better for eggs treated with 400 mg L?1 glutaraldehyde for 10 min at 10 and 15 dph. Effective disinfection was also recorded using 100 mg L?1 Actomar K30. Egg batch effect rather than initial bacterial concentration, disinfectant type or incubation method determined the survival of the eggs to hatching and survival of larvae. Because of the carcinogenic effect of glutaraldehyde, iodophor is recommended for routine disinfection of cod eggs. 相似文献

Lipids and essential fatty acids, particularly the highly unsaturated fatty acids, 20:5n‐3 (eicosapentaenoic acid; EPA), 22:6n‐3 (docosahexaenoic acid; DHA) and 20:4n‐6 (arachidonic acid, AA) have been shown to be crucial determinants of marine fish reproduction directly affecting fecundity, egg quality, hatching success, larval malformation and pigmentation. In Atlantic cod (Gadus morhua L.) culture, eggs from farmed broodstock can have much lower fertilization and hatching rates than eggs from wild broodstock. The present study aimed to test the hypothesis that potential quality and performance differences between eggs from different cod broodstock would be reflected in differences in lipid and fatty acid composition. Thus eggs were obtained from three broodstock, farmed, wild/fed and wild/unfed, and lipid content, lipid class composition, fatty acid composition and pigment content were determined and related to performance parameters including fertilization rate, symmetry of cell division and survival to hatching. Eggs from farmed broodstock showed significantly lower fertilization rates, cell symmetry and survival to hatching rates than eggs from wild broodstock. There were no differences in total lipid content or the proportions of the major lipid classes between eggs from the different broodstock. However, eggs from farmed broodstock were characterized by having significantly lower levels of some quantitatively minor phospholipid classes, particularly phosphatidylinositol. There were no differences between eggs from farmed and wild broodstock in the proportions of saturated, monounsaturated and total polyunsaturated fatty acids. The DHA content was also similar. However, eggs from farmed broodstock had significantly lower levels of AA, and consequently significantly higher EPA/AA ratios than eggs from wild broodstock. Total pigment and astaxanthin levels were significantly higher in eggs from wild broodstock. Therefore, the levels of AA and phosphatidylinositol, the predominant AA‐containing lipid class, and egg pigment content were positively related to egg quality or performance parameters such as fertilization and hatching success rates, and cell symmetry. 相似文献

Live and dead Atlantic salmon eyed eggs were challenged with eight different Saprolegnia isolates, selected because of their varied origins, known morphological characteristics and growth/germination pattern. Some isolates were also tested for pathogenicity to Atlantic salmon parr. Challenge of eggs was performed by exposure to spores in suspension or by co-incubation of live eggs with infected dead eggs. The phenotypic characteristics of the isolates were evaluated in relation to their observed pathogenicity from the challenge experiment, to identify possible virulence factors leading to egg-infection by Saprolegnia. The results from the experiments confirm that live eggs are refractory to infection with Saprolegnia spores in suspension and that an infection of live eggs can only occur from an infection nucleus represented by dead eggs or debris. It was observed that strains pathogenic to salmon parr were not particularly infective towards eggs, and the isolates that gave the highest infection rates to eggs were species considered to be saprotrophs. 相似文献

We use a simple model of temperature-dependent egg development and mortality to develop several hypotheses concerning the effect of temperature on the occurrence of eggs of Atlantic cod, Gadus morhua , in Placentia Bay, Newfoundland. Predictions from this exploratory model were tested through a series of synoptic ichthyoplankton surveys throughout spawning and postspawning periods during 1997 and 1998. Although several egg mortality relationships were explored through the simulation, a constant mortality rate best represented the pattern observed in the two years of data. Peaks in late stage egg densities occurred in August of both 1997 and 1998 and were apparently decoupled from egg production peaks in April. We observed a decrease in mortality and the distance dispersed during egg development with increases in water temperature. We suggest that the effects of predation are small relative to the advective effects within this system, and that the interaction between advection and temperature-dependent vital rates of eggs may have dramatic consequences for coastal retention of propagules produced by inshore spawning events. 相似文献

The autonomic innervation of the ovary of the Atlantic cod was investigated using histochemical and physiological/pharmacological methods. The paired ovary receives autonomic innervation via branches of the posterior splanchnic nerve (vesicular nerve).Histochemical studies demonstrated vasoactive intestinal polypeptide (VIP)-immunoreactive, 5-hydroxytryptamine-immunoreactive and adrenergic nerve fibers, but a number of antisera raised against other peptides failed to reveal any specific reaction in the tissue preparations. It is concluded that the cod ovary receives a double antagonistic autonomic innervation of excitatory cholinergic fibers and non-adrenergic inhibitory fibers. The nature of the inhibitory neurotransmitter is not known. 相似文献

Cultivated Atlantic cod (Gadus morhua) entering their first year of gamete maturation were fed diets with different levels of arachidonic acid (ARA) and eicosapentaenoic acid (EPA) for 6.5 months prior to commencement of spawning. Gravid females were stripped three times: at the beginning, peak and end of spawning. Lipid composition and egg and larval quality of 34 family crosses were investigated. Results indicated that ARA uptake into eggs from broodstock diet was highly efficient achieving proportions of ARA up to 84% higher in eggs than in the diet. EPA was 42–76% higher, and DHA was 155–173% higher in eggs than in diets. Cod fed the diet with the lowest EPA/ARA ratio had the greatest egg production. Eggs from fish on a diet with high ARA level had significantly higher fertilization and hatching success than those fed low levels of ARA. This diet produced on average 71 viable eggs g?1 female compared with 32.5 and 4 eggs in diet B and C, respectively. Furthermore, larval survival until 8 days posthatch was higher in diets with lower ARA levels. The combined results showed that ARA dietary supplementation and low EPA/ARA ratio yielded a greater number of viable larvae kg?1 female. 相似文献

Folate mobilization from the yolk compartment during larval development was studied by analysing the folate concentration in whole body, embryo and yolk in a single batch of Atlantic halibut, Hippoglossus hippoglossus L., eggs and larvae that showed successful fertilization and development. There was a net loss of approx. 50% of folate from yolk during endogenous feeding. Further, only 23% of the decrease in yolk folate was retained in the larval body. The data suggest a need for folate for metabolic and growth purposes during embryogenesis of approximately 2 μg g?1 weight gain. Relative to these data and published folate requirement for cold‐water species, batches of egg from 16 Atlantic halibut brood fish contained variable and, for some batches, critically low levels of folate. This study describes changes in cathepsin L activity during early development in Eurasian perch (Perca fluviatilis) and determines to which extend a precocious proteolysis of the yolk reserves by activated cathepsin L could be involved in the seasonal decline of egg and larval quality. During embryogenesis and early larval stages, samples were collected for determination of fertilization and hatching rates, assays of cathepsin L activity, and evaluation of larval resistance to stress (osmotic shock and fasting). Cathepsin L activity increased significantly during the embryonic development, reaching a maximum of 177.1±12.3 nmol min–1mg fresh weight–1 on day 1 post-hatching and significantly decreasing on day 3. Activity increased significantly during the spawning season and individual variations between egg strands became very high during the second half of the spawning period. An inverse relationship was established between the cathepsin L activity in 7-day old eggs and the decrease of hatching rate. A negative exponential regression was calculated between the cathepsin L activity of eggs and the resistance of corresponding newly-hatched larvae to osmotic shock. Resistance to fasting was significantly reduced in larvae from eggs with cathepsin L activity higher or lower than 20 nmol min–1 mg–1 (lethal time50=4.7±0.7 and 3.5±0.4 days, respectively). The involvement of cathepsin L in the degradation of yolk reserves and its potential consequences on the quality of perch eggs and larvae are discussed in relation to results reported in salmonid fishes. 相似文献

Tawny puffer Takifugu flavidus is a species found in China considered to have potential for aquaculture. Experiments were conducted to determine the optimal temperature for its incubation and larval culture. Fertilized eggs collected from cultured broodstocks that were induced to ovulate with a [d ‐Ala6‐Pro9‐Net]‐luteinizing hormone‐releasing hormone analogue were inseminated. The effect of temperature (19, 20, 23, 26 and 29 °C) on the hatch rate, incubation period, viability of 24 h post‐hatch larvae and total mortality rate was assessed. The effect of temperature (20, 23, 26 and 29 °C) on the growth and survival of larvae from 3 to 19 days after hatching (dah) was also assessed. The results showed that the optimal temperature for successful development of fertilized eggs ranged from 23 to 26 °C, and the highest hatch rate, the optimal viability of 24 h post‐hatch larvae and the lowest total mortality rate were all predicted using quadratic equations. The relationship between temperature and the incubation period of tawny puffer eggs was determined using the effective degree‐day model. The temperature at developmental zero (t0) was 11.34 °C, and the sum of effective degree‐days (k) was 52.356. The survival rate of tawny puffer larvae at 20 °C was significantly lower than among 23, 26 and 29 °C, whereas the survival rate was not significantly different from that at 23, 26 and 29 °C. The larval growth rate increased rapidly as the temperature increased, showing a linear relationship in the range of temperatures investigated. Tidepool sculpins live in a variable environment where water temperature, salinity, gas tensions, and pH can change considerably with the daily tide cycle. Tidepool sculpins are primarily ammoniotelic, with 8–17% of nitrogen wastes excreted as urea. The majority of net ammonia (Jnet
amm; 85%) and urea (Jnet
urea; 74%) excretion occurred across the gill, with the remainder excreted across the skin, the kidney, and/or gut. Acute (2h) exposure to 50% seawater significantly increased Jnet
urea (2.8-fold), but reduced Jnet
amm (3.5-fold). In fish exposed to 50% seawater for 1 week, Jnet
urea returned to control values, but Jnet
amm remained slightly depressed. Unidirectional urea influx (Jin
urea) and efflux (Jout
urea) were measured using14C-urea to determine if urea was excreted across the gills by simple diffusion or by a carrier-mediated mechanism. Jin
urea increased in a linear manner with increasing urea water levels (0–11 mmol N l–1), while Jout
urea was independent of external urea concentrations. As well, Jnet
urea and Jout inurea were not significantly different from one another, indicating the absence of back transport. Urea analogs and transport inhibitors added to the water did not have any consistent effect on unidirectional urea flux. These results demonstrate that ammonia and urea excretion rates and sites of excretion in tidepool sculpins are very similar to those found in other marine and freshwater teleosts. Urea and ammonia may play a role in osmoregulation as excretion rates and tissue levels were influenced by changes in water salinity. Finally, we found no evidence for a specific urea carrier; branchial urea excretion is likely dependent on simple diffusion. 相似文献

Thyroid hormone concentrations in unfertilized eggs of 26 species of various freshwater, marine and diadromous teleosts were
examined, together with changes in their concentrations during egg development in some species. Significant quantities of
both thyroxine (T4) and triiodothyronine (T3) were found in eggs of all species examined. Mean T4 and T3 concentrations in eggs varied from 0.04 (marbled sole) to 15.00 ng/g (chum salmon), and from 0.07 (goldfish) to 9.95 ng/g
(Pacific herring), respectively. T4 concentrations were significantly greater than T3 concentrations in eggs of most freshwater fishes, whereas T3 concentrations were greater in seawater fishes. During the course of development, thyroid hormones in eggs decreased markedly
before hatching. These findings suggest that thyroid hormones are consistently present in teleost eggs, and thus may play

Developmental profiles of thyroxin (T4), triiodothyronine (T3) and radioactive iodide uptake were established for eggs and T4 and T3 profiles were established for larvae (whole-body, yolk-only and body-only) of coho and chinook salmon. T4 and T3 were consistently present in all samples. In eggs, hormone levels remained fairly constant in all cohorst for at least the first three weeks of incubation, but then fluctuated in both directions in some sample groups. Large increases in T4 (from 9 ng/g to 245 ng/g) were seen in 1985 chinook eggs 28 days after fertilization. Radioactive iodide uptake (which was used as a possible indicator of thyroxinogenesis) increased at least 10-fold in both 1986 coho and chinook eggs from 23–30 days after fertilization. T4 (62 ng/g) and T3 (393 ng/g) were found in the bodies of 28-day-old 1986 chinook embryos. In whole larvae, hormone levels varied depending upon the cohort studied. In general, initial body-only concentrations of both T4 and T3 decreased as body weight increased, but before yolksac resorption was completed, both thyroid hormone content and concentration increased (except for chinook T3). T4 and T3 content in larval yolk stayed constant as yolksac size decreased, resulting in increased thyroid hormone concentration in the yolksac. All of these data suggest that the initial source of thyroid hormones in coho and chinook salmon eggs is maternal, but that by approximately 3–4 weeks after fertilization, the developing embryos begin to produce their own thyroid hormones. After hatching, increases in tissue T4 and T3 concentration coupled with constant T4 and T3 content in diminishing yolksacs suggest that larvae also produce their own thyroid hormones; yolksac content then may reflect both the original maternal hormones and the larva-producted hormones. 相似文献

Radioimmunoassay measurement of pituitary AVT content and plasma AVT concentration indicated comparable levels in fully adapted sea water (SW) and fresh water (FW) flounders. Circulating AVT represented less than 0.1% of the pituitary AVT reserve. The urophysis contained AVT but the total content was only 2 or 3 fold that of circulating AVT. In fish adapted to hypertonic media, there was a close correlation between plasma AVT concentration and plasma Na+ concentration or osmolality. The present study examined the effects of acute osmotic challenge, associated with FW to SW transfer, and the influence of extracellular fluid volume status on AVT secretion. Short-term transfer of fish from FW to SW (up to 3 days) did not evoke a clear change in plasma AVT levels, though pituitary content was reduced at 24 h. During the first 3 days after transfer to SW, only small increments in plasma tonicity were apparent. The sensitivity of AVT secretion to osmotic stimuli may only be expressed when plasma osmolality has exceeded a specific threshold, which was probably not reached in these transfer studies. Fish in hypotonic media showed no relationship between plasma osmolality/tonicity and plasma AVT concentration. Acute extracellular fluid volume expansion of SW adapted fish also abolished the normally observed relationship between plasma osmolality and AVT concentration in these hypertonic media fish. This trend indicates that volume status may modulate the sensitivity of AVT secretion to osmotic stimuli as occurs in tetrapods. 相似文献

对养殖施氏鲟( Acipenser schrenckii)、西伯利亚鲟( A.baerii)和小体鲟( A.ruthenus)卵中常规营养成分、氨基酸(AA)和脂肪酸(FA)的组成和含量进行了测定。结果表明,三种鲟鱼中,西伯利亚鲟卵内粗蛋白(22.6%)和粗脂肪(17.9%)含量最高,且与施氏鲟的粗蛋白和粗脂肪含量都显著高于小体鲟( P<0.05);三种鲟鱼卵内均含有18种氨基酸,其中必需氨基酸总量( TEAA)占总氨基酸( TAA)均高于49%,且施氏鲟卵内TEAA占TAA的50.06%,显著高于西伯利亚鲟和小体鲟( P<0.05)。三种鲟鱼卵内含量最多的必需氨基酸( EAA )均为亮氨酸,种类间的色氨酸含量差异显著(P<0.05),其他EAA含量无显著差异(P>0.05);各种非必需氨基酸(NAA)含量种类间差异不显著(P>0.05),以谷氨酸含量最高。三种鲟鱼卵内,含量最高的饱和脂肪酸(SFA)均为C16∶0,种间无显著性差异(P>0.05);单不饱和脂肪酸(MUFA)中均以C18∶1n-9含量最高,且施氏鲟卵内含量显著高于西伯利亚鲟和小体鲟( P<0.05);多不饱和脂肪酸( PUFA)中均以C22∶6 n-3含量最高,其中西伯利亚鲟卵内含量显著高于施氏鲟和小体鲟( P<0.05);西伯利亚鲟卵内n-3/n-6值最高,小体鲟卵内最低,且三种鲟鱼间n-3/n-6值均有显著差异( P<0.05)。西伯利亚鲟鱼卵内, DHA、 EPA、 PUFA、 n-3 PUFA总含量及n-3/n-6值均显著高于另外两种鲟鱼卵( P<0.05)。因此,在配置鲟亲鱼配合饲料时,要重视各种脂肪酸之间的添加比例。 相似文献
