为获得犬脂肪间充质干细胞,本试验取犬腹股沟皮下脂肪组织,分别利用组织培养法和酶消化法分离犬脂肪来源间充质干细胞,对比观察不同来源细胞的形态和增殖特征,并通过诱导液促进细胞向成骨细胞和成脂细胞方向分化,检测其分化潜能。结果表明,通过组织培养法培养的青年犬脂肪组织,可获得大量脂肪间充质干细胞,该细胞生长旺盛,形态均一,可分化为碱性磷酸酶染色阳性的成骨细胞和油红O染色阳性成脂细胞。组织培养法分离培养犬脂肪间充质干细胞操作简单,可为细胞移植治疗等研究提供充足的细胞来源。  相似文献

ObjectiveTo develop and assess the efficacy of an ultrasound (US)-guided pericapsular hip desensitization (PHD) technique in dogs.Study designProspective, randomized, anatomical study and a case series.AnimalsA total of 30 healthy dogs, eight canine cadavers and seven dogs with hip osteoarthritis.MethodsAfter studying the US anatomy of the medial aspect of the coxofemoral joint and determining an acoustic window to perform an US-guided PHD in healthy dogs, the US-guided PHD was performed bilaterally in canine cadavers. A low [(LV) 0.1 mL kg–1] and high [(HV) 0.2 mL kg–1] volume of dye was injected per hip on each cadaver. The staining of the pericapsular nerves was assessed by anatomical dissection, and comparison between LV and HV was assessed using Fisher’s exact test. Then, the US-guided PHD was performed using a triamcinolone–bupivacaine solution in dogs with hip osteoarthritis. Dynamic pain response was assessed before and after injection. The canine brief pain inventory (CBPI) questionnaire was used to assess treatment efficacy and duration.ResultsThe US-guided PHD could be performed by inserting the needle between the iliopsoas muscle and the periosteum of the ilium. The articular branches of the femoral and obturator nerves were stained in all cadavers using both volumes. The main femoral nerve was never stained, but the main obturator nerve was stained in 37.5% and 100% of injections using LV and HV, respectively (p = 0.026). Treated animals showed decreased dynamic pain response after the injection. Compared with baseline, CBPI scores were reduced by ≥ 50% for ≥ 12 weeks in all but one dog.Conclusions and clinical significanceThe US-guided PHD with both 0.1 and 0.2 mL kg–1 volumes stained the articular branches of the femoral and obturator nerves in canine cadavers and was associated with clinical improvement in dogs with hip osteoarthritis.  相似文献

Adipose tissue‐derived stem cells (ASCs) can be obtained from different adipose tissue sources within the body. It is an abundant cell pool, easily accessible, suitable for cultivation and expansion in vitro and preparation for therapeutic approaches. Amongst these therapeutic approaches are tissue engineering and nervous system disorders such as spinal cord injuries. For such treatment, ASCs have to be reliably differentiated in to the neuronal direction. Therefore, we investigated the neural differentiation potential of ASCs using protocols with neurogenic inductors such as valproic acid and forskolin, while dog brain tissue served as control. Morphological changes could already be noticed 1 h after neuronal induction. Gene expression analysis revealed that the neuronal markers nestin and βIII‐tubulin as well as MAP2 were expressed after induction of neuronal differentiation. Additionally, the expression of the neurotrophic factors NGF, BDNF and GDNF was determined. Some of the neuronal markers and neurotrophic factors were already expressed in undifferentiated cells. Our findings point out that ASCs can reliably be differentiated into the neuronal lineage; therefore, these cells are a suitable cell source for cell transplantation in disorders of the central nervous system. Follow‐up studies would show the clinical benefit of these cells after transplantation.  相似文献

ObjectivesTo describe the technique of performing an ultrasound-guided distal parasternal intercostal block and to determine the distribution of two volumes of methylene blue dye solution injected in canine cadavers.Study designProspective cadaver study.AnimalsA group of seven canine cadavers weighing 12–34 kg.MethodsThe space between the transversus thoracic and the internal intercostal muscles is a virtual cavity. Ultrasound-guided injections in the distal (parasternal) intercostal space were performed using dye solution at 0.05 mL kg–1 in each intercostal space from the second to seventh (LV, low volume, six injections per dog) in one hemithorax, and 0.1 mL kg–1 in the third, fifth and seventh intercostal spaces (HV, high volume, three injections in each dog) on the contralateral side. Anatomical dissection was carried out to describe dye spread characteristics and staining of intercostal nerves.ResultsThe ultrasonographic landmarks for injection were identified in each cadaver. In the LV group the solution was found in every intercostal space (36/36), whereas the HV injection stained six intercostal spaces in two dogs, five in two, and in two dogs the solution was found in four and three spaces, respectively, demonstrating multisegmental distribution. Intrapleural staining was observed after two injections.Conclusions and clinical relevanceUltrasound-guided injection of 0.05 mL kg–1 at the distal intercostal space resulted in staining of the intercostal nerve in all dogs when performed in every space and may be an appropriate alternative to previously reported techniques. A single injection of 0.1 mL kg–1 may anaesthetize more than one intercostal nerve, but not consistently. Clinical investigations are warranted to better characterize and to refine this locoregional technique.  相似文献

BackgroundLaryngeal paralysis is a common idiopathic degenerative neurological disease in older medium-to-large breed dogs, with surgical correction of the obstruction being the treatment of choice.ObjectivesThis study evaluated the use of laryngeal silicone stents to treat canine laryngeal paralysis in dogs where classic surgical treatment was not accepted by the owners.MethodsDogs diagnosed with laryngeal paralysis, for which the owners refused arytenoid lateralization surgery as a first-line treatment, were treated with laryngeal silicone stents.ResultsSix dogs with bilateral laryngeal paralysis were included in the study. All dogs showed improvement in clinical signs immediately after the procedure. No clinical signs or radiographic changes were noted in four out of six dogs in the follow-up visit performed 1 wk later. One dog was suspected of aspirating water while drinking, but the signs disappeared after repositioning the stent. Another dog had a relapse of stridor due to caudal migration of the stent. This dog underwent arytenoid lateralization surgery because larger stents are not commercially available. At the time of writing, between seven and 13 mon after stent placement, no significant incidents have occurred in four dogs, and all owners report a satisfactory quality of life.ConclusionsLaryngeal silicone stenting is an interesting alternative for treating dogs with acquired laryngeal paralysis when the owners refuse classic arytenoid lateralization surgery. Furthermore, stent placement can be a temporary solution to stabilize these dogs until a permanent surgical treatment can be performed.  相似文献

Adult stem cells are of particular interest for therapeutic use in the field of regenerative medicine. Adipose-derived mesenchymal stem cells (ASCs) are an attractive stem cell source for all fields of regenerative medicine because adipose tissue - and therewith cells - can easily be harvested from each donor. However, common expansion using fetal bovine serum (FBS) can not be used for clinical applications as xenogenic proteins must be avoided. Adipose tissue from equine, canine and porcine donors was digested with collagenase to isolate ASCs. ASCs were either expanded in a cell culture medium supplemented with FBS or in a serum-free medium (UltraCulture; UC) supplemented with a serum substitute (UltroserG). From all three animal species, the adipogenic and osteogenic differentiation potential of ASCs cultured with different media was analyzed in vitro. Cell proliferation analysis showed a population doubling time of 48-68 h for canine cells, 54-65 h for porcine cells and 54-70 h for equine cells, expanded in different media. Except for porcine ASCs, cells cultured in media supplemented with FBS grew faster than cells expanded in UC medium with UltroserG. Yet, all cells maintained their potential to differentiate into adipocytes and osteoblasts. UltraCulture medium containing UltroserG can for all examined species be recommended if FBS needs to be avoided in the expansion of donor-derived (stem) cells.  相似文献

探讨同源异体骨髓间充质干细胞(BMMSCs)移植对犬急性肝功能损伤的治疗作用,为犬干细胞治疗相关疾病提供理论基础和技术支持。对患急性肝损伤的10只病犬经超声引导腹腔内肝门附近移植BMMSCs,移植后检测血常规、肝功能和肝脏B超,观察同期的症状体征和不良反应情况。结果显示,干细胞移植7天后可使患犬各项肝功能指标明显改善,10d后B超结果显示肝组织回声均匀,无明显异常,14d后患犬的精神、体力、食欲明显好转,干细胞移植21d后血常规各项指标恢复正常,患犬基本康复,移植干细胞的患犬均未发现有不良反应。说明相对于常规治疗,犬BMMSCs同源异体移植治疗急性肝损伤可明显缩短治疗时间,显著提高患犬生活质量。  相似文献

Abstract

CASE HISTORY: A dog that had received 8 months of cyclosporin and ketoconazole therapy for treatment of atopic dermatitis subsequently developed severe neurological disease, that failed to respond to treatment with trimethoprim-sulphadiazine and clindamycin.

HISTOPATHOLOGICAL FINDINGS: Histopathological examination of the pulmonary parenchyma and spinal cord revealed loose aggregates of Gram-positive, partially acid-fast, fine, beaded, filamentous bacteria, most consistent with Nocardia spp.

DIAGNOSIS: A presumptive diagnosis was made of disseminated nocardiosis of the spinal cord and lungs.

CLINICAL RELEVANCE: Nocardia spp. is an opportunistic actinomycete that may cause disseminated disease, particularly in immunocompromised animals. Cyclosporin is used in veterinary medicine to control immune-mediated and allergic disorders, with few reported adverse side effects. This case gives further evidence that involvement of the spinal cord in nocardiosis of the central nervous system (CNS) carries a poor prognosis, and opportunistic infection by Nocardia spp. may be a potential complication of immunosuppressive cyclosporin therapy in the dog.  相似文献

A 5.5‐year‐old, intact male Rottweiler dog was admitted with a history of multifocal nodular tongue lesions which progressively deteriorated during the previous year. Physical examination revealed several reddish nodules with central depression on the surface of the tongue in an otherwise healthy dog. Clinicopathologic abnormalities included eosinophilia and hyperproteinemia. Lingual nodule cytopathology, histopathology, and immunohistochemistry revealed Leishmania spp. amastigotes and a severe granulomatous glossitis. The dog was also seroreactive to L infantum antigens by an indirect immunofluorescence assay. Clinical reevaluation 3 months after the institution of treatment with allopurinol and miltefosine indicated that the nodular lesions had completely regressed. In endemic areas, lingual nodular lesions may rarely be the sole clinical sign of canine leishmaniosis. Standard medical treatment may provide an excellent prognosis.  相似文献

ObjectivesReport the long-term outcomes following transmembrane stent placement as a therapy for Cor Triatriatum Dexter (CTD).Materials and methodsRetrospective case series including six dogs with CTD treated with transmembrane stent placement. Follow-up information was obtained including the persistence of presenting clinical signs, additional therapies required, and survival.ResultsThe median follow-up time was 24 months (range 15–76 months). Long-term outcome was deemed excellent in four dogs (67%), good/fair in one dog (17%), and poor in one dog (17%). Three dogs had persistence of clinical signs of variable severity. These three dogs were Labrador Retrievers or their crosses with varying degrees of tricuspid valve dysplasia (TVD), two of which also had a right to left shunting patent foramen ovale (PFO). One of these three dogs died 23 months post-stent placement during attempted open-heart repair of the TVD and PFO. Another is alive 15 months post-operatively stable on medical therapy for right-sided congestive heart failure secondary to TVD. The final dog demonstrated improved but persistent mild exercise intolerance up to 76 months post-operatively associated with mild TVD and a concurrent PFO.ConclusionsTransmembrane stent placement for CTD is a viable long-term treatment option with improvement or resolution of clinical signs. In the presence of concurrent congenital heart disease, specifically Labradors with TVD, additional therapies may be necessary with a corresponding impact on prognosis.  相似文献

This longitudinal prospective exploratory study used serial measurements in five dogs to evaluate safety and retention of a tin‐117 m (117mSn) colloid after intra‐articular injection in normal elbow joints. Each dog was deemed healthy based on physical examination, laboratory results, and radiographic evaluation of both elbows. While anesthetized, each received an MRI of both elbows, followed by fluorine‐18 fluorodeoxyglucose positron emission tomography scans of both elbow joints and associated lymph nodes. Joint fluid (0.5‐1.0 mL) was withdrawn aseptically from the left elbow joint, followed by intra‐articular injection of 117mSn colloid (92.5 MBq; 1‐1.5 ml). Post‐injection assessments included blood counts, serum chemistry panels, urinalyses, radiographs, joint fluid analyses, MRI/positron emission tomography scans, scintigraphy, and biodistribution scans. On day 45‐47, each dog was euthanized and a complete postmortem examination was performed. Tissue samples were submitted for histopathology and radioisotope retention studies. Left elbow joints were decalcified and sectioned for future autoradiography. Scintigraphy, 1 day after injection, indicated slight radioisotope escape from the joint to regional lymph nodes. Serial blood, urine, feces, and organ counts indicated >99.1% of the 117mSn activity was retained in the joint for 45‐47 days. Radiation output levels were below patient release levels the day following injection. Maximum standard uptake value for the injected joint decreased. Joint fluid cytology was unchanged. No dog exhibited lameness during the study. Absence of joint damage and lack of systemic effects after injection of the 117mSn colloid in normal canine elbow joints indicate that this agent may be safely used for radiosynoviorthesis in dogs with osteoarthritis.  相似文献

Background – Filaggrin (FLG) is a key protein for skin barrier formation and hydration of the stratum corneum. In humans, a strong association between FLG gene mutations and atopic dermatitis has been reported. Although similar pathogenesis and clinical manifestation have been argued in canine atopic dermatitis, our understanding of canine FLG is limited. Hypothesis/Objectives – The aim of this study was to determine the structure of the canine FLG gene and to raise anti‐dog FLG antibodies, which will be useful to detect FLG protein in dog skin. Methods – The structure of the canine FLG gene was determined by analysing the publicly available canine genome DNA sequence. Polyclonal anti‐dog FLG antibodies were raised based on the canine FLG sequence analysis and used for defining the FLG expression pattern in dog skin by western blotting and immunohistochemistry. Results – Genomic DNA sequence analysis revealed that canine FLG contained four units of repeated sequences corresponding to FLG monomer protein. Western blots probed with anti‐dog FLG monomer detected two bands at 59 and 54 kDa, which were estimated sizes. The results of immunohistochemistry showed that canine FLG was expressed in the stratum granulosum of the epidermis as a granular staining pattern in the cytoplasmic region. Conclusions and clinical importance – This study revealed the unique gene structure of canine FLG that results in production of FLG monomers larger than those of humans or mice. The anti‐dog FLG antibodies raised in this study identified FLG in dog skin. These antibodies will enable us to screen FLG‐deficient dogs with canine atopic dermatitis or ichthyosis.  相似文献

ObjectiveTo describe the gross and microscopic anatomy of the sciatic nerve paraneural sheath and to report an ultrasound (US)-guided subparaneural approach to the sciatic nerve in dogs, comparing two different volumes of injectate.Study designProspective, randomized, anatomical study.AnimalsA group of nine middle-sized adult Mongrel canine cadavers (18 limbs).MethodsThe sciatic nerves of three pelvic limbs of two canine cadavers were identified, exposed and isolated between the greater trochanter and the popliteal fossa for gross anatomical and microscopic examination. An additional three pelvic limbs were surgically dissected on the lateral surface of the limb; the sciatic nerves were isolated, and a 26 gauge over-the-needle catheter was inserted through the paraneural sheath under direct visualization. A methylene blue solution was then slowly injected into the subparaneural compartment through the catheter under US visualization using an 8–13 MHz linear-array transducer. Subsequently, 12 pelvic limbs (six cadavers) were randomly allocated to one of two groups; using US-guided percutaneous subparaneural approach, either 0.1 or 0.05 mL kg–1of a 1:1 solution of methylene blue and 0.5% bupivacaine was injected. The spread of the dye solution and the amount of nerve staining were macroscopically scored. The stained sciatic nerves with their sheaths were then harvested for microscopic examination.ResultsThe paraneural sciatic nerve sheath was easily identified distinct from the nerve trunk both macroscopically and with US visualization, and microscopically. Complete staining was achieved in five of six (83.3%) sciatic nerves in each group; no difference was found in the amount of staining between the two groups. Microscopically, no signs of sciatic nerve intraneural injection were observed.Conclusions and clinical relevanceThe US-guided subparaneural injection of 0.05 mL kg–1 of a dye injectate resulted in satisfactory nerve staining without evidence of sciatic nerve intraneural injection.  相似文献

ObjectiveThe incidence of femoral traumatology and hip dysplasia shows the need to design canine specific femoral implants in veterinary surgery. A good knowledge of femoral morphology, and particularly of intra-species variability, is required to develop a well-adapted canine femoral intramedullary implant. The aim of this study is to evaluate the morphological variability of the canine femur and to propose a segmentation of this population.ProcedureThis study proposes different possibilities for the segmentation of a canine population of 103 dogs of various common breeds in relation to their femoral morphology. These segmentations were obtained with a statistical methodology, which takes into account 24 measured and calculated morphological parameters of 206 canine femurs.ResultsThe segmentation of this canine population into four or six homogeneous groups related to the femoral morphology were the two most relevant solutions. The total length of the femur and the femoral head diameter were the best discriminant parameters for this segmentation.ConclusionKnowledge of the variability of the femoral morphology in the canine species and the possibility of splitting the canine population into homogeneous morphological groups are useful for the design of specific canine femoral implants. The femoral morphological profiles of each group constitute an essential database for fitting the best orthopedic implant to the bone.  相似文献

ObjectiveTo evaluate an approach to the canine lumbar dorsal root ganglion (DRG), a significant contributor to the pain pathway, using new methylene blue staining.Study designProspective randomized study.AnimalsA total of three Beagle dog cadavers weighing 10.4 ± 0.7 kg (mean ± standard deviation).MethodsBilateral third to fifth lumbar DRG approaches were performed in three dog cadavers positioned in sternal recumbency. The mammillary process was palpated, and a 22 gauge spinal needle was inserted through the skin 1 cm lateral to the process and directed towards the median plane at a 45° angle to the dorsal plane. The needle was advanced along the transverse plane until touching bone, or a popping sensation was detected. Under fluoroscopic guidance, the position of the needle tip was adjusted to be in the cranioventral part of the intervertebral foramen. The location of the needle was confirmed by demarcation of the nerve roots after iohexol (0.1 mL) injection. For evaluation of the DRG approach, new methylene blue (0.1 mL) was injected. Subsequently, anatomical dissection of the area was performed. The DRG staining was scored as follows: 0, no staining; 1, partial (<50%); 2, partial (≥50%); and 3, complete staining. Comparisons among the staining scores of the third to fifth DRG were assessed with the Friedman test.ResultsStaining score 3 was achieved in 14 of 18 (77.8%) sites. Staining scores 2, 1 and 0 were identified at two, one and one of the 18 sites, respectively. No significant difference was noted in the staining scores among the third to fifth DRGs (p = 0.78).Conclusions and clinical relevanceThe technique used for DRG injections achieved adequate DRG staining, supporting use of the fluoroscopy-guided approach to the canine lumbar DRG.  相似文献

Summary

Histologically confirmed inadequate treatment resulted in a lower than expected recurrence percentage in dogs with haemangiopericytoma (38%) and mastocytoma (30%). Clinical suspicion of inadequate tumour treatment did not always correlate with the histologically assessed inadequacy, nor with the appearance of local recurrence. Local recurrence did not seem to be correlated with histological grade of malignancy and tumour size. Local injection of C. parvum vaccine did not result in a lower percentage of local recurrence or longer recurrence free intervals in any of the three tumour groups (canine haemangiopericytoma, canine mastocytoma, feline mammary carcinoma). Nor was palliative local adjuvant injection of Cp successful in dogs and cats with soft tissue sarcomas or in dogs with gingival melanoma. Re‐operation of locally recurrent tumour was successful in some dogs with haemangiopericytoma, in a few with mastocytoma, but not in cats with mammary carcinoma. A trend toward histological progression of recurrences and metastases, when compared with the primary tumours, was not evident. The possible reasons for the relatively low recurrence rate of some tumour types and for the failure of Cp‐treatment are discussed.  相似文献

ObservationsAnaphylactoid reactions were suspected in three dogs following the intravenous administration of the contrast agent gadobenate dimeglumine 0.05 mmol kg?1 (Multihance®).Case 1: A 14 kg 6–year–old atopic female dog was anaesthetized for brain magnetic resonance imaging (MRI). All monitored parameters remained stable during the procedure. Fifteen minutes following MR completion; facial, peri–orbital and sublingual oedema were noted. Resolution of the oedema was rapid and uneventful following treatment of clinical signs over 2 hours.Case 2: A 16 kg 10–month–old male dog was anaesthetized for brain and neck MRI. Ten minutes after MR contrast intravenous (IV) injection; heart rate (HR) increased (85–120 beats minute?1), mean arterial blood pressure (MAP) decreased (from 70 to 43 mmHg) and Pe′CO2 decreased (from 4.66 to 3.19 kPa). Labial, periorbital and lingual oedema were noted. Clinical signs responded to fluid bolus administration. The dog vomited in recovery but oedema resolved within one hour.Case 3: A 34 kg 2–year–old atopic male dog was anaesthetized for head MRI. Within 5 minutes of MR contrast IV injection; the dog suffered severe cardiovascular collapse. MRI procedure was aborted and administration of anaesthetics discontinued. Aggressive IV fluid resuscitation and IV epinephrine administration were necessary to re–establish cardiovascular stability. Some periorbital and labial oedema were noted. The dog vomited once and had soft faeces but made a complete recovery.ConclusionsThe administration of contrast medium may result in mild to severe anaphylactoid reactions.  相似文献

Allogeneic tooth transplantation was evaluated as a functional and aesthetic treatment for dental fracture in the dog. Of 7 dogs that received tooth transplants, 5 were research animals and 2 were clinical patients. Canine teeth were transplanted immediately after extraction in the research dogs. Endodontic therapy had been performed on the affected canine tooth of one clinical patient. The other clinical patient had bilateral maxillary canine fractures 2 months earlier. One of the research dog transplants failed at 3 weeks as a result of improper surgical technique. Four of the research dogs had a solid implant for 18 months, after which time the dogs were euthanatized serially. All transplanted teeth were anchored firmly into the alveoli, but were nonviable. Root resorption, with bone replacement, was first noticed at 24 months. The transplanted tooth in the first clinical patient remained functional for 3 months, after which time the tooth was fractured. The right canine transplant in the second clinical patient failed by 3 months, probably because of preexisting periapical inflammation. The left transplanted tooth remains stable at 38 months. It was concluded that allogeneic tooth transplantation may have merit as a rapid and inexpensive method for replacement of fractured teeth in the dog. Function is compromised gradually as a result of root resorption and ankylosis, with tooth fracture likely to occur after 2 years.  相似文献
