The major acute phase proteins (APPs) in cattle are haptoglobin (Hp) and serum amyloid A (SAA), and in swine, are Hp, SAA, C-reactive protein (CRP), and Pig major acute phase protein (Pig-MAP). Many methodologic assays are presently available to measure these parameters, which are still being improved to increase their specificity, sensitivity, user-friendliness, and economic availability. In cattle, the main applications are the diagnosis and monitoring of frequent diseases such as mastitis and metritis in dairy cows and respiratory problems in young calves. In pigs, APPs are useful in the control of bacterial and viral infections, and they may be used at the slaughterhouse to monitor subclinical pathologies and improve food safety. The utility of APP in animal production must not be forgotten; optimization of protocols to improve performance, welfare, and nutrition may benefit from the use of APPs. Other sample types besides serum or plasma have potential uses; APP determination in milk is a powerful tool in the control of mastitis, saliva is a non-invasive sample type, and meat juice is easily obtained at the slaughterhouse. Increasing our knowledge of reference intervals and the influence of variables such as age, breed, sex, and the season is important. Finally, worldwide harmonization and standardization of analytical procedures will help to expand the use of APPs.  相似文献

Stressors such as weaning, mixing and transportation have been shown to lead to increased blood concentrations of acute phase proteins (APP), including serum amyloid A (SAA) and haptoglobin, in calves. This study was therefore undertaken to assess whether SAA and haptoglobin levels in blood mirror stress in adult cattle. Six clinically healthy Holstein cows and two Holstein heifers were transported for four to six hours to a research facility, where each animal was housed in solitary tie stalls. Blood samples for evaluation of leukocyte counts and serum SAA and haptoglobin concentrations were obtained before (0-sample) and at 8, 24 and 48 hours after the start of transportation. Upon arrival the animals gave the impression of being anxious, and they appeared to have difficulty coping with isolation and with being tied on the slippery floors of the research stable. Serum concentrations of SAA and haptoglobin increased significantly in response to the stressors (P < 0.01 and 0.05 at 48 hours, respectively). Additionally, the animals had transient neutrophilia at 8 and 24 hours (P < 0.05). In conclusion, the results of the study suggest that SAA and haptoglobin may serve as markers of stress in adult cattle.  相似文献

The aim of the study was to describe the acute phase protein and leukocyte responses in dairy heifers during acute, oligofructose-induced ruminal acidosis. The study included 2 trials involving oral oligofructose overload (17 g/kg BW) to nonpregnant Danish Holstein heifers. Trial 1 included 12 heifers all receiving oligofructose, and the experiment consisted of a 3 day control period prior to overload and 9 days surveillance afterwards. Eight heifers were fed grass hay and 4 were fed barley silage. Trial 2 included 10 heifers receiving oligofructose, 4 were euthanized 24 h after overload and 6 were euthanized 72 h after overload. Six heifers received tap water as control treatment and were euthanized 72 or 96 h later. Sampling of blood was performed at 6–48 hour intervals. Samples were analyzed for serum amyloid A (SAA), haptoglobin, and fibrinogen, and total white blood cell counts (WBC) were performed.  相似文献

The aim of this study was to evaluate three acute phase proteins (APP) [haptoglobin (HPT), lipopolysaccharide binding protein (LBP) and transferrin (Tf)] in feedlot cattle with naturally occurring respiratory disease diagnosed by a calf health scoring chart (CHSC). Seventy-seven beef calves were observed for signs of Bovine Respiratory Disease (BRD) during the first 28 days after arrival at the feedlot. Fourteen cases and pen matched controls were selected based on the CHSC. BRD cases were defined as a score of ≥5, while controls were defined as a score ≤4. The mean CHSC score in cases was 6.9 which was significantly greater than the controls 2.8 (P < 0.01). Mean plasma LBP and HPT concentrations were significantly greater in cases than controls (P < 0.01). Our study results show that measurement of HPT and LBP could be useful in detecting respiratory disease in feedlot conditions. Transferrin concentrations between the two groups were not statistically different.  相似文献

Treating inflammation in the equine gastrointestinal tract remains a challenge. Our most potent anti-inflammatory drugs, COX inhibitors and glucocorticoids, have unwanted effects on the gastrointestinal tract and host defense that often limit their use. Newer strategies targeting specific cells and molecules that regulate a subset of the events occurring during inflammation are rapidly becoming available and should allow clinicians to reduce the detrimental effects of inflammation without inhibiting the beneficial aspects.  相似文献

Bovine babesiosis due to Babesia bigemina infection induces systemic inflammation, evidenced by increased sialic acid (SA) levels and declined cholinesterase activity. The current study was undertaken to assess further indicators of the systemic inflammation in the naturally infected cattle. To this end, serum levels of some selected acute phase-proteins (APPs) including serum amyloid A (SAA), haptoglobin (Hp), ceruloplasmin (Cp), and fibrinogen were measured. Additionally, sensitivity and specificity of the APPs were calculated by receiver operating characteristic curve. The correlation among APPs, SA and cholinesterase activity were also assessed. Our previous blood specimens were used to measure APPs. Briefly, the diseased animals were divided into two groups according to the parasitemia: 12 mildly (20 % <) and 8 severely (20 %>) infected animals. Moreover, 10 healthy animals as the control were included. The levels of all APPs were measured to be significantly elevated in a parasitemia burden-dependent fashion as compared to the control. Furthermore, all the APPs showed 100 % specificity, but only SAA and Cp had 100 % sensitivity. A strong and positive correlation was calculated between the APPs and SA; however, cholinesterase activity was inversely correlated with AAPs and SA. In conclusion, inflammatory reactions play a pivotal role in the pathogenesis of bovine babesiosis and APPs can be considered as the potential indicators of inflammation.  相似文献

Serum acute phase protein concentrations are used as diagnostic, therapeutic and prognostic markers in human and, less frequently, in animal medicine. The aim of this study was to determine how the health status and signalment of the cat are associated with concentrations of acute phase proteins. Generally, medians of the positive acute phase proteins appeared to be higher in sick cats compared to healthy cats. In multivariable regression models, log-transformed serum amyloid A concentration was higher in older cats, in sick and in female cats, while log-transformed α1-acid glycoprotein and haptoglobin concentrations were higher in older cats and were associated with interactions of health status (sick/healthy) and gender (male/female). The data from healthy cats in this study contribute to the limited knowledge of normal reference ranges for this species. This study highlights the potential of acute phase proteins as diagnostic markers in sick cats, but also emphasises that the signalment of the cat needs to be taken into consideration.  相似文献

This study was undertaken to evaluate the acute phase responses via the assessment of the concentration of serum sialic acids (total, lipid bound and protein bound), inflammatory mediators (IFN-γ and TNF-α) and acute phase proteins (Hp and SAA) in 20 adult crossbred cattle naturally infected by Anaplasma marginale. The infected animals were divided into 2 subgroups on the basis of parasitemia rate (<20% and >20%). Also, as a control group, 10 clinically healthy cattle from the same farms were sampled. Our data revealed significant decreases in red blood cell count (RBC), hematocrite (PCV) and hemoglobine (Hb) in infected cattle compared to healthy ones. Conversely, the concentrations of Hp, SAA, ceruloplasmin, fibrinogen, serum sialic acids and the circulatory IFN-γ and TNF-α were increased in the diseased cattle (P<0.05). In addition, it was evident that the progression of parasitemia in infected cattle did not induce any significant alterations in the hematological indices (RBCs, PCV and Hb) and the concentrations of Hp, SAA, ceruloplasmin and fibrinogen. SAA was the most sensitive factor to change in the diseased cattle. Therefore, increase in SAA concentration may be a good indicator of inflammatory process in cattle naturally infected with Anaplasma marginale.  相似文献

As part of a study of the development of hoof horn haemorrhages in first-lactation heifers, measurements were made of acute phase reactants to investigate the link between the acute phase response and the development of the haemorrhages. Over a period of two years, blood samples were taken from two separate groups of heifers, weekly in the three weeks before they calved and then twice weekly until eight weeks after calving. Plasma total protein, albumin, fibrinogen, haptoglobin, seromucoid and serum iron and caeruloplasmin were measured and the relationships between the peak concentration (or activity) or the area under the curve of each acute phase reactant and the peak scores for sole or white line haemorrhages were assessed by linear regression. The results suggested that the development of the hoof horn haemorrhages observed in the study was not accompanied by an acute phase response, and the haemorrhages were therefore not primarily caused by endotoxicosis. The diets and husbandry systems used were typical of dairy farms in the UK and the results therefore suggest that a significant proportion of hoof horn haemorrhages observed in UK dairy cows may not be caused by endotoxicosis.  相似文献

Acute phase proteins are serum proteins which increase in concentration during the acute phase response to inflammation or infection. The response occurs in all animals, but in different species the response of individual proteins can be significantly different. Of the numerous acute phase proteins which have been identified in humans, a number have been examined in cattle and dogs but usually on an individual basis with little reference to their part in the acute phase response. Biochemical, physiological and clinical investigations into haptoglobin, fibrinogen, 1-proteinase inhibitor, ceruloplasmin, seromucoid and C-reactive protein of cattle and dogs have therefore been reviewed with the emphasis on their role in this response to tissue damage.  相似文献

The acute phase response is a nonspecific inflammatory reaction of the host that occurs shortly after any tissue injury. The response includes changes in the concentration of plasma proteins called acute phase proteins (APPs), some of which decrease in concentration (negative APPs), such as albumin or transferrin, and others of which increase in concentration (positive APPs), such as C-reactive protein, serum amyloid A, haptoglobin, alpha-1-acid glycoprotein, and ceruloplasmin. Most positive APPs are glycoproteins synthesized mainly by hepatocytes upon stimulation by proinflammatory cytokines and released into the bloodstream. The acute phase response and clinical application of monitoring APPs in dogs and cats are reviewed in this article, including biochemical characteristics, assays developed for each individual APP, and preanalytic and analytic factors influencing APP results that should be taken into account for proper and adequate clinical interpretation. In addition, the diagnostic use of APPs and their possible application in monitoring treatment, which can be considered one of the most interesting and promising practical applications of these proteins, will be discussed. Finally, challenges and future developments of APPs in dogs and cats will be considered, because it is expected that new and cheaper automated assays for determination of the main APPs in small animals will contribute to a wider use of these proteins as biomarkers of infection and inflammatory lesions.  相似文献

In this review, an overview is provided on the current achievements regarding the interplay between rumen digestive disorders and diet-induced inflammation in dairy cattle. It starts with a review of factors favoring the disturbances in the rumen metabolism, which culminate with development of sub-acute rumen acidosis (SARA). The latter digestive disorder is often linked to greater metabolic stress of gastrointestinal (GI) microbiota and lowered fiber digestion, as well as with disruption of the barrier functions of the GI epithelia, which open the route of deleterious molecules to translocate from the GI lumen into the portal system. A model is suggested to illustrate the mechanisms of the involvement of digestive disorders in the disruption of the host's inner homeostasis leading to activation of acute phase response (APR). The latter is part of multifaceted innate immune and metabolic responses of the host. According to this model, endotoxin, its toxicity, and other metabolic compounds of microbial origin are regarded as important immunogenic components of GI tract, which when favored by disruption of host barriers triggers a systemic APR. Although the activation of an APR is viewed as a protective reaction aiming to reestablish the disturbed homeostasis, the presence of inflammatory state over long periods might be associated with negative consequences for the host. The review concludes that prolonged systemic inflammation can: (1) cause significant changes in the energy and lipid metabolism in different body tissues, (2) lead to the development of refractory states associated with immune suppression and increased susceptibility to various diseases, and (3) artificially increase host's requirements in energy and nutrients, lowering the efficiency of energy and feed use by the animal. The paper emphasizes the critical role that formulation of healthy diets plays for curbing down inflammation and enhancing metabolic health of dairy cows.  相似文献

The term "fog fever" was originally used identify a disease of adult cattle grazing lush pastures in the autumn. Unfortunately, the name has subsequently been applied to other respiratory disorders which occurred under different epidemiological circumstances, so that the name "fog fever" has lost much of its original specificity and become almost synonymous with "acute respiratory distress". The pulmonary lesions in 151 cattle, of all ages, with acute respiratory distress are described in this report. While most of the animals were referred as examples of "fog fever", in only 43 of the 151 cases were the clinical signs, epidemiology and post mortem findings consistent with that disease. Twelve other pulmonary disturbances were encountered in the other animals and the pathology of these conditions has bee described.  相似文献

Acute phase proteins (APPs) such as haptoglobin, serum amyloid A and pig major acute phase protein are plasma proteins that increase in concentration following infection, inflammation, or trauma. The circulating concentrations of these proteins in pigs and cattle can provide an objective measure of the health status of an animal and are increasingly being used as markers of animal health and welfare. Plasma concentrations of APPs are related to the severity of the underlying condition, and provide a ready means of evaluating both the presence and extent of disease. Haptoglobin, for example, has been used to identify both clinical and subclinical disease in animals, and for objectively monitoring antibiotic therapy in experimentally infected animals. Interpretative benefit can be further enhanced by the "acute phase index", derived from a mathematical formula that uses both positive- and negative-reacting APPs. Research suggests that in the future, assays for APPs will be used routinely to assess animal health, optimize production rates, monitor antibiotic therapy, detect diseases such as mastitis in dairy cows, and assess the health of animals at slaughter. These applications have considerable benefit for human food safety. Before APP assays can be applied in animal production on a worldwide basis, however, the calibration of assay methods must be harmonized to ensure that results obtained in the laboratory or on the farm are universally comparable and of consistent quality. In February 2000 the European Commission Directorate General Research Concerted Action was established to fulfill the task of international standardization of APPs. The Concerted Action Group consists of a network of 14 institutions representing 9 European countries. In this report, the background and goals of the Concerted Action Group, and scientific presentations from the group's first colloquium are described. In addition, the progress of the group to date and the standardization plan for the full 30-month duration of the Concerted Action are summarized.  相似文献

The hepatic lipidosis (HL) in fattening turkeys is a disease has been known for a long time, but the cause and pathogenesis is still not clarified. A recent study reported unexplained high levels of iron in liver tissue of fattening turkeys suffering from HL. In this study, the iron status, possible infectious or inflammatory influences in form of an acute phase reaction and the analysis of fatty acid pattern in liver tissue of turkeys affected by HL were examined. Three cases of HL on three different fattening turkey farms were investigated during the outbreak of the disease. Clinically affected and non-affected animals were subjected to a pathological examination, where the diagnosis HL or non-affected was made. In total, 70 birds were examined (40 with HL, 30 without HL) and blood and liver samples were taken. Additionally, samples from 15 slaughtered birds were taken as a further control group. In liver tissue, the iron content and the content of long-chain fatty acids were determined; in blood samples, ferritin and transferrin were measured. The iron content in liver tissue was more than three times higher for animals with HL than among non-affected animals and the control group. The transferrin levels were lowest for animals with HL, highest in the control group and in between for non-affected animals. The fatty acid pattern in liver tissue of affected animals indicated a shift from polyunsaturated fatty acids to saturated fatty acids and monounsaturated fatty acids compared to the control group and the non-affected animals. Overall, the non-affected animals of a flock affected by HL were similar to the healthy animals of the abattoir. The low acute phase protein levels for animals with HL together with high iron contents could indicate a previous malnutrition/starvation period and/or severe liver damage for those animals suffering from HL.  相似文献

The purpose of this study was to investigate the relationship between swine health status and the concentration of the serum acute phase proteins, haptoglobin (HP), and C-reactive protein (CRP). A total of 378 clinically healthy pigs from farms A and B, plus 20 pigs culled from farm A due to poor growth, were used in this experiment. Each pig was examined and blood samples were collected during slaughter. The HP concentration was measured by using an HP-hemoglobin binding assay. The CRP concentration was measured by using a CRP enzyme immunoassay. Gross and histopathological lesions were examined and recorded at slaughter. Representative samples were then collected in order to isolate pathogens. Swine enzootic pneumonia, found in 47.7% of the pigs, was the most common lesion. Other lesions included pleuropneumonia (32.7%), suppurative pneumonia (10.3%), fibrinous pericardititis (4.3%), Ascaris migration in the liver (33.9%), and intestinal serositis (3.0%).  相似文献
