One IBV isolate, SC021202, was isolated from the kidneys of the infected young chickens by inoculating embryonated eggs, and its morphology, physiochemical and haemagglutonating properties were detected. Virulence of the isolate SC021202 was determined with specific pathogen-free (SPF) chicken inoculation. Nucleotide acid sequence of S1 gene of the isolate SC021202 was further sequenced and analysed. The physiochemical and morphological properties of the isolate SC021202 were in accordance to that of typical infectious bronchitis virus (IBV). In a pathogenicity experiment, the clinical signs and related gross lesions resembling those of field outbreak were reproduced and the virus isolate SC021202 was re-isolated from the kidneys of the infected chicken. Sequence data demonstrated that the full length of the amplified S1 gene of the isolate SC021202 was composed of 1931 nucleotides, coding a polypeptide of 543 amino acid residues. Compared with IBV strains from GenBank, the nucleotide and deduced amino acid sequence of S1 gene of the isolate SC021202 shared 60.0-91.4% and 49.1-88.9% identities, respectively. A nucleotide fragment of 'CTTTTTAATTATACTAACGGA' was inserted at nucleotide site 208 in the S1 gene of the isolate. These results indicated that IBV isolate SC021202 was a new variant IBV isolate and responsible for field outbreak of nephritis.  相似文献

Infectious bronchitis virus (IBV) Cal99 variant was isolated from the kidneys of seven 2-5-mo-old game chickens with nephritis and respiratory disease. IBV Cal99 variant is usually associated with respiratory disease in broiler chickens in California. Macroscopically, the majority of the birds had moderately to severely enlarged and mottled pale kidneys, with increased urates in the ureters. Microscopically, most of the birds had acute nephrosis and interstitial nephritis. The birds also had sinusitis, tracheitis, bronchopneumonia, airsacculitis, salivary gland adenitis, and lymphoid depletion in the thymus and bursa of Fabricius. Immunohistochemistry was strongly positive for IBV antigen in the cytoplasm of tubular epithelial cells in the kidneys and also in the epithelium of the respiratory tract, salivary glands, proventriculus, intestine, and bursa of Fabricius. Infectious bronchitis virus was isolated from the trachea, lungs, kidneys, and cecal tonsils. Sequencing of the hypervariable region of the S1 gene of the kidney IBV isolate, designated IBV/CA99variant/07, revealed that the virus was 98% homologous to the Cal99 serotype of IBV.  相似文献

本研究对分离自上海某养鸡场的疑似鸡传染性支气管炎病毒(Infectious bronchitis virus,IBV)进行了分离和鉴定.所采用的方法有:鸡胚致病性试验、对鸡新城疫病毒(Newcastle disease virus,NDV)的干扰试验、动物回归试验以及RT-PCR鉴定等.试验结果表明,该病原接种鸡胚后,48 h可引起鸡胚死亡,胚体呈侏儒样变化,对NDV感染有明显干扰作用;用分离的病毒接种SPF雏鸡可产生明显呼吸道症状,肾脏表现肿大和大量尿酸盐沉积等病理变化;用IBV特异性引物可以从分离物中扩增出IBV的N基因序列,测序结果表明,该毒株属于肾型IBV.综上所述,本研究在上海流行区域成功分离到一株IBV,将其命名为SH11株.  相似文献

鸡传染性支气管炎病毒CQ/01/2004株的分离与鉴定   总被引:2,自引:0,他引:2  
2004年从重庆某肉用鸡场疑似鸡传染性支气管炎的病鸡中采集病料,按常规处理后接种9~10日龄鸡胚,通过鸡胚连续传代培养3代,并对该分离毒株的鸡胚致病性、血凝性和NDV的干扰特性进行检测.同时进行了动物回归试验。结果表明,该分离株具有IBV感染特征,可使鸡胚胚体出血、蜷缩、矮化;该分离毒株无直接血凝性,对NDV有明显的干扰作用;动物回归试验中有75%的感染鸡在10d内发病或死亡。剖检病死鸡可见肾苍白、肿胀,肾小管内充塞大量尿酸盐,支气管有出血点、有大量粘液。采用反转录.聚合酶链式反应(RT-PCR)技术对CQ/01/2004的纤突蛋白S1基因进行扩增、克隆和序列测定,结果表明该基因具有IBVSl基因的共有分子特征,将测序结果提交GenBank进行同源性检索,发现分离株CQ/01/2004和J株S1的同源性最高,核苷酸同源性为94%,氨基酸同源性为89.4%,与M41的核苷酸同源性为80.6%,氨基酸同源性为78.0%。试验结果表明,分离的病毒株CQ/01/2004为鸡传染性支气管炎病毒。  相似文献

鸡传染性支气管炎病毒(IBV)具有不同致病特性,将IBV XDC-2株接种9日龄SPF鸡胚培养,可引起鸡胚死亡和出现侏儒胚,病毒EID50达5×10-5.33/mL。将IBV XDC-2株接种18日龄SPF鸡,饲养观察14 d,病鸡临床症状表现为:精神沉郁,羽毛凌乱,双翅下垂,轻微腹泻,多数拉白色水样稀粪。病死鸡出现肾肿大、呈花斑状、大量尿酸盐沉积。鸡发病率为100%,死亡率为25%。死亡鸡肺脏、肾组织制作组织切片,发现病理变化明显,主要为:肾小管扩张,上皮细胞呈玻璃样变性,部分管腔内可见坏死脱落之上皮细胞,于肾间质可见大量单核细胞浸润,肾间质有充血、出血现象;肺内动脉、毛细血管充血,淋巴细胞浸润。死亡鸡肺脏、肾组织接种鸡胚分离病毒,RT-PCR检测结果为阳性,表明该分离株为鸡传染性支气管炎病毒,具有很强的嗜肾性。  相似文献

从黑龙江省绥化市某养鸡场疑似鸡传染性支气管炎的鸡群中采集病料,经鸡胚传代、电镜观察、生物学特性和分子生物学鉴定以及动物回归试验,表明该分离株具有明显的鸡传染性支气管炎病毒特征,具有鸡胚出现卷曲、出血、发育延迟等作用;对新城疫病毒有明显的干扰作用;动物回归试验导致未免疫鸡出现明显的感染症状,剖检可见明显的肾脏肿胀、尿酸盐沉积现象,同时可见呼吸道有出血现象。对该毒株的N基因进行扩增,并将其序列与NCBI的参考毒株的N基因进行序列对比,发现其与国内的分离株SC和N毒株的N基因具有较高的同源性,为97.6%;与国外参考毒株和国内的疫苗株同源性较低,为84.8%。表明分离的病毒为鸡传染性支气管炎病毒。  相似文献

The emergence of new infectious bronchitis virus (IBV) genotypes or serotypes along with the poor cross-protection observed among IBV serotypes have complicated the avian infectious bronchitis (IB) control programs in different geographic regions. In Cuba, the lack of genetic information regarding IBV and the increasing epidemiological importance of this virus in Cuban chicken flocks demand further characterization of IBV isolates. In the present work, studies of genetic diversity and phylogenetic relationships among recent IBV isolates from Cuban chicken flocks showing respiratory disorders were performed. Two putative genotypes genetically different to the Massachusetts genotype H120 strain used in the Cuban vaccination program were found in the flocks assessed. In addition, a potential nephropathogenic IBV isolate was found by first time in Cuba.  相似文献

鸡传染性支气管炎是由鸡传染性支气管炎病毒引起的一种急性、高度接触性传染性疾病。以张口呼吸、气管啰音、咳嗽、肾脏病变为主要临床症状,可引起鸡的饲料报酬率降低,蛋鸡产蛋数量和蛋的品质下降。IBV的特点是高频变异,血清型众多,导致疾病的临床和病理变化复杂多样,由于其变异株出现比较快,毒株之间交叉保护力差,给IB的诊断和防治带来了较大困难。从IBV的分型、检测技术、综合防治方面对该病的研究进展进行了综述。  相似文献

根据基因库中收录的鸡传染性支气管炎病毒 (IBV)S1基因的序列 ,设计了一对引物并采用RT -PCR扩增了鸡传染性支气管炎病毒HN99株的S1基因 ,扩增产物进行了克隆、测序 ,获得了IBVHN99株S1基因片段 ,其大小为1 739bp(含前导序列 ) ,其核苷酸序列与H1 2 0、H52、M41、Gray、Holte的S1基因核苷酸序列同源性较低 ,分别为 79.1 %,79.2 %,77.3%,77.8%,79 .4%,有大量的点突变并伴有基因插入和缺失 ;IBVHN99株的S1基因推导的氨基酸与H1 2 0、H52、M41、Gray、Holte株氨基酸的同源性分别为80 .1 %,79.9%,79.5%,78.5%,78.5%,经S1基因系统进化分析 ,提示IBVHN99株与其它各毒株的亲缘关系较远 ,初步证实IBVHN99株为一新的IBV毒株  相似文献

Nephropathogenic infectious bronchitis (NIB) was diagnosed in 28 infectious bronchitis virus (IBV)-vaccinated commercial chicken flocks in Pennsylvania from December 1997 to July 2000. Early dinical signs were increased flock mortality and urinary water loss (polyuria and pollakiuria) leading to wet litter. Daily mortality ranged from 0.01% in layers to 2.45% in broilers, with total broiler mortality as high as 23%. Severe renal swelling and accumulation of urates in the tubules were commonly seen. Visceral gout and urolithiasis were less frequently observed. Histopathologic changes included characteristic tubular epithelial degeneration and sloughing with lymphoplasmacytic interstitial nephritis. Minimal respiratory disease signs were noted in broilers. Egg production and shell quality declined in layers. Confirmatory diagnosis of NIB was made by IBV antigen-specific immunohistochemical staining of the renal tubular epithelium and virus isolation. Sequencing of the S1 subunit gene of 21 IBV isolates showed the NIB outbreak to be associated with two unique genotypes, PA/Wolgemuth/98 and PA/171/99. The cases from which the genotypes were isolated were clinically indistinguishable. The NIB viruses were unrelated to previously recognized endemic strains in Pennsylvania and were also dissimilar to each other. Genotype PA/Wolgemuth/98 was isolated almost exclusively during the first 14 mo of the outbreak, whereas PA/171/99 was recovered during the final 18 mo. The reason for the apparent replacement of PA/Wolgemuth/98 by PA/171/99 is not known.  相似文献

从发病鸡群中分离到一株病毒,通过鸡胚接种、电镜观察、动物回归试验、气管环交叉中和试验等,初步确定为传染性支气管炎病毒,暂命名为A2。外源病原检查结果为阴性。攻毒试验:在气管、肾以及肌肉等组织均有明显的病变;病理组织观察;在气管、肾脏和肌肉均发生了一定程度的病理变化。利用电泳对传统的病毒株和分离株进行了病毒结构蛋白分析,A2株出现了31kD的特殊蛋白条带。气管环交叉中和试验表明A2株与4/91病毒为同一血清型;临床的动物免疫试验结果表明,4/91疫苗有较好的保护作用。  相似文献

Despite the existence of an active vaccination program, recently emerged strains of nephropathogenic infectious bronchitis virus (IBV) in Korea have caused significant economic losses in the poultry industry. In this study, we assessed the pathogenic and antigenic characteristics of a K-IIb type field strain of IBV that emerged in Korea since 2003, such as Kr/Q43/06. Specific pathogen free 1-week-old chickens exhibited severe respiratory symptoms (dyspnea) and nephropathogenic lesions (swollen kidneys with nephritis and urate deposits) following challenge with the recent IBV field strain. The antigenic relatedness (R value), based on a calculated virus neutralization index, of the K-IIb type field strain and K-IIa type strain KM91 (isolated in 1991) was 30%, which indicated that the recent strain, Kr/Q43/06, is a new variant that is antigenically distinct from strain KM91. This report is the first to document the emergence of a new antigenic variant of nephropathogenic IBV in chicken from Korea.  相似文献

为调查辽宁地区鸡传染性支气管炎病毒(IBV)的流行及遗传演化情况,从辽宁某鸡场疑似鸡传染性支气管炎病料中分离得到1株病毒,经分子生物学检测、鸡胚矮小化试验、新城疫病毒血凝特性干扰试验和动物回归试验,确定该毒株为IBV,并命名为CH/LN/2019。扩增该毒株S1基因并进行序列分析发现,分离株S1基因全长1 620 nt,编码540个氨基酸,S1蛋白裂解位点为HRRRR,属于基因Ⅰ型(QX型)毒株;同源性比对发现,分离株与基因Ⅰ型代表毒株QXIBV株的核苷酸和氨基酸序列同源性最高,分别为95.6%和94.8%;与国内常规型疫苗H52、H120和Ma5毒株的同源性较低,核苷酸和氨基酸序列同源性仅为77.2%~76.9%和75.4%~76.2%。本试验为辽宁地区鸡传染性支气管炎的流行病学调查及免疫防控提供了参考。  相似文献

To investigate genetic variation of avian infectious bronchitis virus (IBV) in Guangxi province,one strain of IBV was isolated from chicken.Two pairs of primers for amplifying the N and M genes of IBV were designed according to the sequences in GenBank.The N and M genes of the strain were amplified by RT-PCR,and they were proved to be the N and M genes of IBV by cloning,sequencing and compared with reference IBV strains published in GenBank.The results showed that the N gene from the IBV isolate consisted of 1 230 bp,coding 409 amino acids.The M gene from the IBV isolate consisted of 678 bp,coding 225 amino acids.The sequence analysis of N gene showed that it shared 87.2% to 93.3% nucleotide homologies and 90.0% to 94.4% deduced amino acid sequence homologies with IBV strains from GenBank.The M gene sequence analysis showed that it shared 83.6% to 91.0% nucleotide homologies and 82.7% to 92.9% deduced amino acid sequence homologies.The phylogenetic tree analysis showed that it was closely related to BJ and LX4 strains,and were clustered into one group;But with the distant relatives from other strains of IBV.These results suggested that the isolate was a new variant of IBV.  相似文献

OBJECTIVE: To identify and partially characterize a coronaviruslike virus isolated from naturally infected pigeons. ANIMALS: 50 specific pathogen-free (SPF) embryonated chicken eggs, 30 White Leghorn SPF chickens, and 12 clinically normal pigeons. PROCEDURES: Pancreatic tissue specimens from sick pigeons were inoculated into SPF embryonated chicken eggs for viral isolation and investigation of morphologic and hemagglutinating properties of the isolate, called PSH050513. Furthermore, virulence studies in SPF chickens and experimental pigeons were performed. The spike (S) glycoprotein gene of PSH050513 was further sequenced and analyzed. RESULTS: PSH050513 was isolated and identified from the experimentally infected pigeons by a routine method, which was in accordance with Koch's postulates. The complete S protein (1,167 amino acids) was compared with published S protein sequences of other avian and mammalian coronaviruses. A high degree of sequence identity (79.3% to 99.6%) was observed between the S protein sequence of PSH050513 and published sequences of avian infectious bronchitis virus (IBV); only limited identity (< 37.8%) was observed with turkey coronavirus and mammalian coronaviruses. Furthermore, when the virus was inoculated into SPF chickens, pancreatitis developed. CONCLUSIONS AND CLINICAL RELEVANCE: PSH050513 has been tentatively identified as a novel member of group 3 coronaviruses that have close genetic relationships with IBV strains.  相似文献

A virus isolated from a respiratory disease in chickens was identified as infectious bronchitis virus (IBV). By serum neutralisation tests the virus was shown to be serologically distinct from the Massachusetts type but to possess a one-way relationship with the Connecticut type. Experimentally, it caused a relatively mild respiratory disease in baby chicks. Vaccination of young chicks with H-120 Massachusetts IBV vaccine afforded a high degree of protection against challenge with the isolate. Some problems relating to the isolation and classification of field strains of IBV are briefly discussed.  相似文献
