Numerous subclinical diseases in sheep occur in the periparturient period and involve inflammatory processes; therefore, determining markers, such as acute-phase proteins (APPs), can allow an early diagnosis. Therefore, the objective of the present study was to assess changes in the plasma concentration of APPs and cortisol in clinically healthy ewes in the periparturient period for use in future studies. At the same time, haematological parameters were monitored. We showed that plasma APPs and cortisol concentrations were significantly higher in pregnant ewes than before insemination. A gradual increase in the SAA concentration was observed from the 14th day before to the day of parturition, while Hp was reduced from 2 weeks before up to 2 weeks after delivery. A significant increase in the Fb concentration was detected from the 14th day before to the 1st week after delivery. The cortisol concentration did not undergo significant changes in the periparturient period. We found an increase in the SAA and Fb concentrations and decrease in Hp in the periparturient period. The direction of the change in APPs of healthy ewes in the current study may be related to their distinct regulatory mechanisms during pregnancy. The APPs are usually altered during infection, inflammation, neoplasia, stress and trauma; therefore, knowing their reference values could help lead to an early diagnosis of subclinical forms of some diseases and pregnancy complications in ewes. The haematological analysis showed that ewes in late pregnancy and postpartum compared to dry period were under metabolic stress related to pregnancy and lactation.  相似文献

The morphological effects of phytooestrogen exposure were determined in 10 ewes exposed to subterranean clover for 60 days, compared with 10 controls. In a second experiment, the time course of the development of the changes was studied. Typically oestrogenic changes were observed in ovary, oviduct, uterus, cervix, vagina and mammary glands. There was an early increase in cervical mucus, followed by a decrease. The delta basophils of the pituitary became degranulated, and hyperactive in appearance. The adrenal and thyroid glands increased in weight, and thyroid epithelium increased in height. There appeared to be a temporary increase in neurophysin storage in the hypothalamus, and shrunken, hyperchromatic neurones were observed in the hypothalamus of some affected ewes. All changes were observed within three weeks of exposure.  相似文献

The incidence of oestrus was depressed in ewes with permanent clover infertility running on non-oestrogenic pasture, particularly in the first part of the breeding season. The depression was due to the failure of some ewes to display oestrus, and to a more erratic pattern of oestrous activity in the other ewes. The incidence of oestrus was restored to normal by treatment with progesterone. In contrast, the incidence of oestrus was reduced in affected ewes grazing in oestrogenic pasture.  相似文献

OBJECTIVE: To determine whether low dose xylazine with ketamine reduces the concentrations of cortisol and prolactin in sheep postoperatively and to characterise the effects of the drugs on behaviour during recovery. DESIGN: Analysis of variance was used to compare the effects of anaesthesia, surgery and combined ketamine/xylazine treatment on the plasma cortisol and prolactin concentrations and on behavioural variables in pregnant ewes subjected to abdominal surgery. PROCEDURE: Twelve ewes were randomly assigned to receive either ketamine/xylazine or placebo in association with anaesthesia and surgery. Both groups of ewes underwent anaesthesia alone followed a week later by anaesthesia with laparotomy and hysterotomy. Plasma cortisol and prolactin concentrations were assayed during these procedures and for 5 days afterwards. Behavioural observations were made remotely during recovery from anaesthesia and anaesthesia plus surgery. RESULTS: The concentrations of cortisol in the plasma of pregnant ewes undergoing surgery were increased by preoperative handling and the onset of thiopentone/halothane anaesthesia, with a further increase during surgery (P = 0.033). Cortisol concentrations decreased over the first four postoperative hours (P = 0.029) and were normal by 24 h. The drug treatment did not affect the immediate responses of ewes to anaesthesia or surgery, although treated ewes had lower cortisol concentrations than saline-treated controls over the first five postoperative days (P = 0.018). Prolactin concentrations increased in response to anaesthesia (P = 0.047), but were not affected by surgery or the drug treatment. Drug-treated ewes had prolonged sleeping time after surgery (P = 0.002), but they took no longer to stand than saline-treated controls and required fewer attempts to stand successfully (P = 0.025). CONCLUSION: At the doses used, ketamine and xylazine did not mitigate the immediate endocrine consequences of surgery but the behavioural data provide a basis for further investigations that may lead to improvements in analgesic treatments.  相似文献

Of 31 pregnant ewes with clinical signs of pregnancy toxaemia, 24 had hypoglycaemia and hyperketonaemia at the time that a single blood sample was obtained. Twenty-five of these had a plasma cortisol concentration in excess of 10 ng/ml and six had a value below this. All the seven animals which did not show both hypoglycaemia and hyperketonaemia had a plasma cortisol concentration in excess of 10 ng/ml. Taking all the sheep together, 80% had a high plasma cortisol concentration. This could be the consequence of increased adrenal output or reduced excretion by the liver.  相似文献

In Australia, more than 1 million ewes have permanently damaged reproductive tracts because they have been grazed on oestrogenic pasture. These effects occur in the absence of classical clinical "clover disease". The lesions result from an "organisational" action of oestrogen, causing a mild sexual transdifferentiation to occur in ewes during adult life, with the main lesion being found in the cervix. Diagnosis of the problem depends primarily on detection of the pathological changes, rather than history or clinical signs. On average, affected flocks have around a 10% increase in non-pregnant ewes, but most farmers can achieve an acceptable lambing rate by increasing their management and feed inputs to produce more twin lambs. Agronomic approaches have been used in the past to combat "clover disease", but further progress with the present problem may depend on the development of animal-based solutions.  相似文献

Morphometric methods were used to measure abnormalities in histological architecture of the endocervix in ewes with infertility after prolonged exposure to oestrogenic clover pastures. Similar measurements were made on the cervix of ewes exposed to oestradiol-17 beta for 6 months. In both cases the changes in histological appearance were essentially similar, and were most easily measured by counting the number of cervical crypts. Ewes treated with either phyto-oestrogens or oestradiol also had significantly wider cervical folds, greater areas of lamina propria, and more cervical glands. The results provide a basis for measuring the severity of permanent oestrogenic change in different flocks or after different experimental treatments.  相似文献

Previous results from this laboratory have demonstrated that in preterm fetal sheep (117-131 days gestation), stimulated ACTH secretion is highly sensitive and that in term fetal sheep (129-143 days), stimulated ACTH secretion is insensitive to the negative feedback effects of cortisol. The purpose of this study was to quantitate cortisol negative feedback inhibition of stimulated ACTH secretion in adult sheep. Adult, conscious, nonpregnant ewes, chronically prepared with carotid arterial loops, were infused intravenously with vehicle or cortisol at 4 different rates (denoted Groups I, II, III, and IV) for 5 hours. These infusions increased total and unbound plasma cortisol concentrations within the range observed after stimulation of the hypothalamus-pituitary-adrenal axis. One hour after termination of the cortisol or vehicle infusions, ACTH secretion was stimulated by intravenous infusion of sodium nitroprusside for 10 min at a rate of 20 micrograms/kg.min. Cortisol infusions suppressed ACTH responses to nitroprusside in a dose-related manner. After vehicle infusion, nitroprusside increased plasma ACTH to 735 +/- 229 pg/ml. After cortisol infusions, nitroprusside increased plasma ACTH to 292 +/- 63, 101 +/- 30, 73 +/- 12, and 67 +/- 24 in Groups I, II, III, and IV, respectively. Overall, there was a significant negative exponential relationship between plateau plasma cortisol concentration during the cortisol or vehicle infusion and the peak plasma ACTH concentration during the response to nitroprusside infusion (r = -0.81). The highest rate of cortisol infusion increased total and unbound plasma cortisol concentrations to 40.1 +/- 5.7 and 19.5 +/- 5.9 ng/ml and completely suppressed the subsequent ACTH response to nitroprusside.  相似文献

Macroscopic examination of 487 ewe reproductive tracts from 18 different sources was made at Perth abattoirs. Of these, 194 were examined microscopically and 217 bacteriologically. Observations were made during March to May, when ewes were not grazing oestrogenic pasture. Significant lesions in ovaries or oviducts were rare. Macroscopic cysts were found in the uterus or cervix of 159 ewes (32.7%). Microscopic lesions, including uterine or cervical cysts and squamous metaplasia or goblet cell hyperplasia of the epithelium, were observed in 76% of ewes studied. Bacteria were isolated from 59 ewes, the most common genera being Corynebacterium, Escherichia, Streptococcus and Bacillus. Escherichia coli and streptococci appeared to be the most commonly associated with endometritis. Presence of bacteria was closely associated with endometritis and with cysts in the cervix. Endometritis was observed microscopically in 48 ewes (24%) and was significantly associated with the presence of cysts in the uterus or cervix. These results suggest that there is a high incidence of clover disease lesions in West Australian ewes, and that under commercial conditions clover disease may cause functional damage to the cervix, allowing passage of bacteria and resulting in endometritis.  相似文献

This study was conducted to determine the effect of crutching, simulated shearing and conventional shearing in mid-pregnancy on maternal plasma cortisol levels and lamb live weight and body dimensions at birth. Twin-bearing Romney ewes (n = 96) were subjected to one of four treatments (control, crutched, sham shorn and shorn) on day 80 of pregnancy. Ewes were maintained as a single flock throughout pregnancy under commercial farming conditions. Within 12 h of birth the lambs were weighed and their body dimensions recorded. Lambs born to ewes shorn in mid-pregnancy were heavier (P < 0.05) at birth than lambs born to control or sham-shorn ewes by 300 and 400 g (7 and 10%) respectively. The birth weight of lambs born to crutched and control ewes did not differ. Shearing and crutching produced a greater integrated cortisol response than sham-shearing (P < 0.05). The failure of crutching to increase lamb birth weight suggests that the acute stress caused by shearing does not result in increased lamb birth weights.  相似文献

Nwannenna A.I., A. Madej, T.J-O. Lundh and G. Fredriksson: Effects of oestrogenic silage on some clinical and endocrinological parameters in ovariectomized heifers. Acta vet scand. 1994, 35, 173-183.–The influence of phytoestrogens was studied in 3 ovariectomized Swedish Friesian heifers fed 20 kg of 100% red clover silage per heifer/day for 14 days. Behaviour, reproductive organs and pituitary response to exogenous gonadotropin-releasing hormone (GnRH) injections were monitored. Clinical effects like oedema and mucous discharge in the vulva, presence of milky fluid in the mammae and increases in teat size and the cross-sectional distance of the uterus were observed in heifers fed red clover silage. Fluid accumulation in the uterus, visualized by means of ultrasonography, had still not disappeared 30 days after the red clover silage had been completely withdrawn. Red clover silage appeared to reduce the magnitude and duration of the pituitary response to GnRH injections.  相似文献

Glucocorticoids inhibit the plasma vasopressin responses to hemorrhage and hypoxia in dogs. Attempts to demonstrate glucocorticoid inhibition of vasopressin secretion in fetal sheep have been unsuccessful, suggesting the possibility that there is an influence of development on the expression of this interaction, or that the interaction cannot be demonstrated in all mammalian species. This study was designed to investigate these two possibilities. Adult ewes chronically prepared with carotid arterial loops, were subjected to 5 hr infusions of cortisol at a rate of 6 ug/kg min or vehicle (5% ethanol in saline). The infusion of cortisol increased plasma cortisol concentration from 26 +/- 3 to 46 +/- 8 ng/ml, while vehicle infusion was associated with a decrease in plasma cortisol concentration from 23 +/- 4 to 15 +/- 3 ng/ml. One hr after the end of the cortisol or vehicle infusions, vasopressin secretion was stimulated by arterial hypotension produced by 10 min infusions of sodium nitroprusside (20 ug/kg min). Nitroprusside decreased arterial blood pressure equally in both groups. Plasma vasopressin concentrations were increased to peak concentrations of 92 +/- 33 and 116 +/- 20 pg/ml in the vehicle- and cortisol-infused groups, responses which were not significantly different as tested by ANOVA. We conclude that increases in plasma cortisol concentration, equal to those observed during responses to stressors, do not inhibit vasopressin secretion in this species.  相似文献

Three cross-bred cows calved in March and April and were followed until day 62 after parturition. Each animal was suckled by 2 calves ad libitum. All calves were removed from the cows on day 55 after parturition. Blood was collected 3 times per day from the jugular vein by venipuncture. On 4 occasions after parturition--i.e. days 7-8, 21-22, 35-36 and 49-50, the cows were bled through a jugular venous catheter every 30 min during the 24 h. The plasma samples were analyzed for the content of 15-keto-13,14-dihydro-PGF2 alpha (main PGF2 alpha metabolite), LH, prolactin, cortisol and progesterone by radioimmunoassay methods. The concentration of PGF2 alpha increased from 280 to 730 pmol/l within the last 4 days before parturition. The highest geometric mean was 3106 pmol/l on the day of parturition. Thereafter a steady decrease of PGF2 alpha metabolite concentration was seen until day 21 when it reached plateau at 148 pmol/l. In all cows plasma LH concentrations increased significantly (P < 0.05) from about 1.6 micrograms/l on days 7-8 to 2.4 micrograms/l on days 21-22 post partum. The frequency of LH pulses showed no tendency to increase as the postpartum period progressed and averaged 6.5 pulses/24 h. Mean plasma LH concentrations increased from 2.1 micrograms/l 2 days before weaning to 3.2 micrograms/l 2 days after weaning (P < 0.05). LH peaks occurred less frequently in association with prolactin and cortisol peaks than in their absence. A partial positive correlation between PGF2 alpha metabolite and cortisol (r = 0.30) was found on days 7-8 post partum. Correlation between prolactin and cortisol on days 7-8 and 21-22 post partum was also positive (r = 0.20 and r = 0.27, respectively). There was a negative correlation between LH and cortisol on days 7-8 (r = -0.27) and days 49-50 (r = -0.21) post partum. The first and short progesterone increase observed after weaning was terminated in conjunction with PGF2 alpha metabolite peaks.  相似文献

The effects of single IV administered doses of dexamethasone on response to the adrenocorticotropic hormone (ACTH) stimulation test (baseline plasma ACTH, pre-ACTH cortisol, and post-ACTH cortisol concentrations) performed 1, 2, and 3 days (experiment 1) or 3, 7, 10, and 14 days (experiment 2) after dexamethasone treatment were evaluated in healthy Beagles. In experiment 1, ACTH stimulation tests were carried out after administration of 0, 0.01, 0.1, 1, and 5 mg of dexamethasone/kg of body weight. Dosages greater than or equal to 0.1 mg of dexamethasone/kg decreased pre-ACTH plasma cortisol concentration on subsequent days, whereas dosages greater than or equal to 1 mg/kg also decreased plasma ACTH concentration. Treatment with 1 or 5 mg of dexamethasone/kg suppressed (P less than 0.05) post-ACTH plasma cortisol concentration (on day 3 after 1 mg of dexamethasone/kg; on days 1, 2, and 3 after 5 mg of dexamethasone/kg). In experiment 2, IV administration of 1 mg of dexamethasone/kg was associated only with low (P less than 0.05) post-ACTH plasma cortisol concentration in dogs on day 3. In experiment 2, pre-ACTH plasma cortisol and ACTH concentrations in dogs on days 3, 7, 10, and 14 and post-ACTH plasma cortisol concentration on days 7, 10, and 14 were not affected by dexamethasone administration. The results suggest that, in dogs, a single IV administered dosage of greater than or equal to 0.1 mg of dexamethasone/kg can alter the results of the ACTH stimulation test for at least 3 days. AIM: To determine the plasma cortisol response to laparoscopy in ewes and investigate means of reducing it. METHODS: Ewes without lambs at foot (n=40) were subjected to one of three control or one laparoscopy treatments (n=10 ewes/treatment), being: no restraint or drugs; acepromazine maleate (ACP) control and no restraint; ACP and restraint in a cradle for 5 min; and laparoscopy following ACP. Additional ewes with lambs at foot (n=30) were subjected to: laparoscopy following ACP; laparoscopy following ACP and ketoprofen; and laparoscopy following detomidine. Drugs were injected 20 min before treatment, after a first blood sample had been taken. Blood samples were taken by jugular venepuncture from the ewes 20 min before treatment and at 20, 40, 60, 90, 120, 150 and 180 min after treatment, while all ewes were held in a pen. Plasma was harvested and assayed for its concentration of cortisol. RESULTS: Plasma cortisol concentrations (PCC) remained constant in ewes in the control restraint group for 80 min. In ewes given ACP, PCC increased for the first 20 min after treatment but then returned to pre-treatment concentrations. PCC of ewes given ACP and restrained in a cradle were elevated above pre-treatment concentrations for 90 min. PCC in ewes subjected to laparoscopy following sedation with ACP increased to a peak at 40 min and returned to pre-treatment concentrations after 60 (with lambs) or 120 (without lambs) min. When ACP and ketoprofen were given before laparoscopy, PCC peaked at 20 min and returned to pre-treatment concentrations by 40 min. PCC of ewes given detomidine before laparoscopy remained at pre-treatment concentrations throughout. PCC of ewes subjected to laparoscopy with ACP sedation only were greater than those of control restraint, ACP control, and ewes subjected to laparoscopy after being given ketoprofen or detomidine between 20 and 60 min after treatment. PCC of ewes subjected to laparoscopy were greater than those of control ewes placed in a cradle at 20 and 40 min. PCC of ewes given ketoprofen were lower than those of ewes subject to laparoscopy following ACP. CONCLUSIONS: Laparoscopy, even after sedation with ACP, caused some distress in ewes, as evidenced by increased plasma cortisol levels. Plasma cortisol response was alleviated by the administration of ketoprofen and eliminated by detomidine, probably because of both analgesic and sedative effects of the latter drug.  相似文献

Adrenal and/or thyroid gland function tests were evaluated in horses at various times during short-term therapy with phenylbutazone, stanozolol, and boldenone undecylenate. There were no significant treatment or time effects on mean basal plasma cortisol concentrations in horses during treatment with the following: phenylbutazone, given twice daily (4 to 5 mg/kg, IV) for 5 days; stanozolol, given twice weekly (0.55 mg/kg, IM) for 12 days; boldenone undecylenate, given twice weekly (1.1 mg/kg, IM) for 12 days; or nothing. There was no significant effect of phenylbutazone treatment on the changes in plasma cortisol concentration during the combined dexamethasone-suppression adrenocorticotropic hormone (ACTH)-stimulation test. Plasma cortisol concentration was significantly decreased from base line at 3 hours after dexamethasone administration and was significantly increased from base line at 2 hours after ACTH in all horses (P less than 0.05). Likewise, the stimulation of basal plasma cortisol concentrations at 2 hours after administration of ACTH (P less than 0.05) was not affected by treatment with stanozolol or boldenone undecylenate. There were no significant treatment effects on mean basal plasma concentrations of thyroxine (T4) or triiodothyronine (T3) among horses during the following treatments: stanozolol, given twice weekly (0.55 mg/kg, IM) for 12 days; boldenone undecylenate, given twice weekly (1.1 mg/kg, IM) for 12 days; or nothing. The involvement of prolactin in the periparturient rise in the faecal nematode egg count in sheep was investigated. Ostertagia circumcincta larvae (5000 third stage larvae three times weekly) were administered to adult immune ewes from three weeks before parturition to three weeks afterwards. Ten ewes were injected twice daily with 2-bromo-alpha-ergocryptine (bromocriptine), an antagonist of prolactin secretion, for two weeks starting two days after lambing while 10 ewes remained untreated. Bromocriptine treatment was initiated approximately two weeks pre partum in three other ewes. Plasma pepsinogen concentrations rose significantly by one week after the start of O circumcincta larval challenge in all the ewes but faecal egg counts remained negative until approximately one week post partum. Plasma prolactin concentration was reduced to a very low level in all bromocriptine treated ewes but this did not alter the dynamics of the periparturient rise in faecal egg counts. Neither cell-mediated nor humoral immunity of the ewes, as assessed by their sensitivity to BCG inoculation and by antibody titre raised against horse red blood cells, respectively, were impaired during the rise in faecal egg count, nor were these parameters altered by manipulation of plasma prolactin concentration. Lamb growth rate was not retarded by low plasma prolactin concentration in the bromocriptine treated ewes. These results are not consistent with the generally held hypothesis that elevated plasma prolactin concentration is directly associated with the periparturient rise.  相似文献

The relationships among hepatic corticosteroid-binding globulin (CBG) mRNA expression and plasma concentrations of cortisol and CBG was evaluated in fetal pigs (n=7-14 per age) on days 50, 70, 80, 90, and 104 of gestation and postnatal pigs (n=8 per age) on days 1, 3, 10, 20, 30, and 40 following birth. In fetal pigs, hepatic CBG mRNA expression was highest (P<0.01) on day 50 as compared to days 90 and 104, exhibiting an overall negative relationship (r=-0.63; P<0.01) with estimated gestation age. Plasma porcine CBG (pCBG) concentration was correlated (r=0.34; P<0.05) with hepatic CBG mRNA level. Plasma cortisol concentrations were not different over this same period. In postnatal pigs, hepatic CBG mRNA expression increased (P<0.01) from days 3 to 40. The pCBG concentration increased (P<0.01) from days 1 (6.1+/-3.4 microg/ml) to 10 (15.1+/-3.7 microg/ml), while plasma cortisol concentration remained constant. An understanding of the relation between hepatic CBG mRNA and circulating pCBG concentrations may provide insight into the mechanisms determining the bioavailability of cortisol necessary in prenatal development and the conservation of cortisol during postnatal development in the pig.  相似文献

The effect of various space allowances on the pituitary, adrenal and immune responses and on performance was investigated in 72 mature Holstein x Friesian beef bulls. The animals (weighing 403+/-3.5 kg) were blocked by weight and randomly assigned to two groups (familiar, F, and unfamiliar, UF) x three treatments (1.2, 2.7 and 4.2m(2) per bull; n=24 per space allowance), and housed for 83 days in 18 pens (n=4 per pen). Blood samples were collected on days -1, 0, 3, 14, 36 and 77 with respect to mixing and housing on day 0. The bulls were given exogenous adrenocorticotrophic hormone (ACTH) on day 3 and corticotrophin-releasing hormone (CRH) on days 14, 36 and 77. Basal plasma cortisol concentration was not affected (P>0.05) by mixing F and UF bulls. On day 3, basal cortisol was greater (P<0.05) in bulls housed at 1.2 than those at 2.7 and 4.2m(2) space allowances while no effect was observed in ACTH-induced plasma cortisol concentration among treatments. Following CRH administration, there was no effect (P>0.05) of treatment and treatment x time on plasma ACTH concentration. On day 14, interferon-gamma production was lower (P<0.05) in the bulls housed at 4.2 vs. 2.7 m(2) and was intermediate but not significantly different (P>0.05) for those housed at 1.2m(2). Animals housed at either space allowances had significant (P<0.05) neutrophilia, lymphopenia, eosinopenia and decreased haemoglobin on day 3 compared with day 0. The liveweight gain from days 0 to 83 was lower (P<0.05) in bulls housed at 1.2 compared with those at 2.7 and 4.2m(2). Housing bulls at 1.2m(2) space allowance had a detrimental effect on their growth and was associated with an acute rise in plasma cortisol concentration (on day 3) compared with those having space allowances of 2.7 and 4.2m(2)/bull.  相似文献

Objective To investigate whether events that may be stressful to young lambs, including simulated infection or social isolation, modulate pain experienced by lambs following castration and tail docking (C/D). Study design Randomised, controlled, prospective study. Animals Fifty male lambs born to 46 second‐parity Mule ewes. Methods Lambs were allocated randomly to one of four groups, experiencing either a potential stressor or handling on day 2 after birth, followed by C/D or handling only on day 3. Quantitative sensory testing (QST) data [mechanical nociceptive thresholds (MNT), Semmes Weinstein filaments (SW), response to cold] and serum cortisol concentration were measured at time points after application of treatments to lambs on days 2 and 3 after birth. The treatment groups were LPS, injection of bacterial lipopolysaccharide IV on day 2, C/D on day 3; ISOL, isolation from the dam for 10 minutes on day 2, C/D on day 3; CAST, handling only on day 2, C/D on day 3; CONT, handled only on days 2 and 3. Results Castration and tail docking caused transient hypoalgesia as measured by MNT and SW. Simulated infection and isolation caused hyperalgesia 3 hours after application, indicated by a reduction in MNT, however they did not alter the pain response to C/D compared to lambs in the CAST group. Injection of LPS and C/D caused increased serum cortisol concentration. The magnitude of the cortisol response to C/D was not altered by prior exposure to either LPS or isolation. Conclusions and clinical relevance LPS and isolation did not modulate the response to C/D but did cause hyperalgesia. This highlights the importance of flock health management and husbandry techniques to reduce the incidence of either systemic infection or psychological stressors in young lambs.  相似文献
