In the present study, we examined whether mildly altering dietary cation-anion difference (DCAD) contributes to the prevention of milk fever in dairy cows. Thirty multiparous cows and ten primiparous cows (heifer group) were used in this study and the multiparous cows were randomly divided into three groups of ten animals each (anion, non-anion and control groups). The cows in the anion group were given supplemental salts that slightly lowered DCAD. These salts consisted of 115 g of CaCO3, 42 g of CaHPO4, 65 g of MgSO4 x 7 H2O and 80 g of CaCl2 x 2 H2O as a daily dose for each cow, using a catheter from 21 days before the expected date of parturition until parturition. The cows in the non-anion group were given only the same Ca, Mg and ip supplement but no sulfate and chloride salts as that in the anion group. The cows in the control and heifer groups were not given any additional supplement. The incidence of hypocalcemia in the anion group decreased to approximately half of those in the non-anion and control groups, while the heifer group did not develop hypocalcemia at all. In addition, the number of days spent for the treatment of hypocalcemia and the number of drug bottles (calcium borogluconate solution) used for the treatment decreased to less than half in the anion group compared with those in the non-anion and control groups. At parturition, the serum Ca concentration in the control (6.2 +/- 1.9 mg/dl, mean +/- standard deviation) and non-anion groups (6.4 +/- 1.7 mg/dl) were significantly lower than that in the heifer group (8.3 +/- 0.4 mg/dl), and the level in the anion group was intermediate (7.3 +/- 1.3 mg/dl). The change in ionized Ca concentration was almost the same as that in serum Ca concentration, but only the concentration in the anion group tended to increase slightly from a week before parturition and was significantly higher than that in all other groups three days before parturition. Urinary pH in the anion group was maintained at a mildly acidic level (6.8-7.0) for the last two weeks before parturition, compared with those in the control (7.3-7.5) and non-anion groups (7.9-8.1), and similar to that in the heifer group (6.3-7.3). The urinary Ca excretion was the highest in the anion group among all groups during the prepartum period. There were no specific changes in the excretion of parathyroid hormone and 1,25-dihydroxyvitamin D in all groups of multiparous cows while the levels of these hormones remained low in the heifer group throughout the experimental period. The data in the present study indicates that the administration of anion salts that slightly lowered DCAD in the preparum period was effective for preventing milk fever in multiparous cows. Safe and mild metabolic acidosis induced by the anion salts could be evaluated by urinary pH (6.8-7.0), and might increase the responsiveness to Ca requirement at parturition through some complex mechanisms unrelated to the excretion of Ca-related hormones. In addition, it was clarified that primiparous cows have a high potential to respond to sudden Ca demand unrelated to hormone excretion, and their Ca metabolism was in some respects similar to that in multiparous cows fed anion salts. Therefore, manipulating mildly DCAD is expected to be an effective, safe and natural method for preventing milk fever in dairy cows.  相似文献

In our previous study, it was demonstrated that the administration of anion salts, which slightly lower the dietary cation-anion difference (DCAD), in the prepartum period is safe and effective for preventing milk fever in multiparous cows. In the present study, several clinico-pathological constituents in serum and urine, which might be related to milk fever, were analyzed using stored samples from the previous study to identify clinico-pathological parameters for easily evaluating the efficacy of lowering DCAD and to further investigate the mechanism by which lowering DCAD prevents milk fever. Among the parameters analyzed in the present study, inorganic phosphorus (iP) was involved in milk fever because the serum concentration and urinary excretion of iP were significantly higher in the group of primiparous cows (heifer group), which did not develop hypocalcemia, than those in other groups of multiparous cows. Serum chloride concentrations in the heifer group and the group of multiparous cows fed anion salts (anion group) tended to remain higher than those in other control groups of multiparous cows suggesting that serum chloride concentration may be utilized for evaluating the status of metabolic acidosis and the efficacy of lowerng DCAD in dairy cows fed anion salts. In addition, plasma estradiol-17beta concentration in the heifer group tended to be lower at parturition compared with that in other multiparous groups suggesting that estrogen known as a potent inhibitor of bone resorption may be involved in developing milk fever.  相似文献

Background: Milk fever (MF) is a common calcium metabolism disorder in perinatal cows. Currently, information regarding the detailed metabolism in cows suffering from MF is scant.

Objective: The purpose was to study the metabolic profiling of serum samples from cows with MF in comparison to control cows, and thereby exploring other underlying pathological mechanisms of this disease.

Animals and methods: In the current study, we compared the serum metabolomic profile of dairy cows with MF (n = 8) to that of healthy dairy cows (n = 24) using a 500-MHz digital 1H-nuclear magnetic resonance (1H-NMR) spectrometer. Based on their clinical presentation and serum calcium concentration, cows were assigned either to the control group (no MF symptoms and serum calcium concentration >2.5 mmol/L) or to the MF group (MF symptoms and serum calcium concentration <1.4 mmol/L). For statistical analysis, a one-way analysis of variance was performed.

Results: We identified differences regarding nine metabolites between the two groups, among which glucose, alanine, glycerol, phosphocreatine, and gamma-aminobutyrate decreased, and β-hydroxybutyrate, acetone, pyruvate, and lysine increased in cows with MF. Most of these were carbohydrates and amino acids involved in various energy metabolism pathways.

Conclusion: The different metabolites in cows with MF reflected the pathological features of negative energy balance and fat mobilization, suggesting that MF is associated with altered energy metabolism.

Clinical importance: The 1H-NMR spectroscopy can be used to understand the pathogenesis of MF and identify biomarkers of the disease.  相似文献

Lecithin:cholesterol acyltransferase (LCAT) is the enzyme responsible for production of cholesteryl esters in plasma. The LCAT activity is reduced in cows with fatty liver developed during the nonlactating stage and those with the fatty liver-related postparturient diseases such as ketosis. The purpose of the present study was to examine whether reduced LCAT activity during the nonlactating stage could be detected before the occurrence of postparturient diseases. Sera from 24 cows were collected at approximately 10-day intervals from -48 to +14 days from parturition. Of the 24 cows, 14 were apparently healthy, whereas 7 had ketosis and 3 had milk fever at around parturition. Of the 14 healthy cows, 7 had unaltered LCAT activity during the observation period, whereas 7 showed reduced activity from -20 to +14 days. Ketosis and milk fever occurred at from -3 to +10 days, but reductions of LCAT activity in diseased cows had already been observed from days -20 to 0. These results suggest that LCAT activity is virtually unaffected during the peripartum period at least in some healthy cows and also that the reduction in LCAT activity can be detected before the occurrence of ketosis and milk fever.  相似文献

In a 6 x 6 Latin square arrangement, sheep of 41 kg body weight were fed myristic acid [C14:0; 50 g/kg dry matter (DM)] supplemented to two basal diets of forage : concentrate ratios of 1 : 1.5 and 1 : 0.5 and adjusted to dietary calcium (Ca) contents of either 4.2 or 9.0 g/kg DM (the latter only together with C14:0 supplementation). Various variables of energy, fatty acid and Ca metabolism were determined in combined digestibility and respiratory chamber measurements. With C14:0 addition the energy loss via the faeces increased (p < 0.05, post hoc test) without affecting energy digestibility of the complete diet. The apparent digestibility of supplemented C14:0 was higher (p < 0.01) with approximately 0.8 in the forage-based diet than in the concentrate-based diet (approximately 0.6). The elevated levels of plasma C14:0 were mainly accompanied by reduced C18:0 and C18:1 levels. The estimated apparent content of metabolizable energy (ME) of added C14:0 was either 24.5 MJ/kg (concentrate-based diet) or 32.1 MJ/kg (forage-based diet). Extra Ca equalized these differences between basal diets and ME contents amounted to 33.0 MJ/kg on average. As expected from corresponding slight shifts in energy metabolizability, the total efficiency of ME utilization increased (p < 0.1) with C14:0. The lower level of dietary Ca was still within the range recommended, but adding C14:0 to the concentrate-based diet reduced Ca retention in the body of the sheep from 0.9 to -0.1 g/day because of an impaired (p < 0.05, post hoc test) net Ca absorption, whereas no effect was found with the forage-based diet. With C14:0 addition, plasma total phosphorus (P) and serum calcitrol levels increased (p < 0.05, post hoc test) while Ca concentrations did not clearly reflect the reduced net Ca absorption. Increasing the dietary Ca content prevented adverse effects on Ca retention in the concentrate-based diet and improved Ca retention in the forage-based diet. In conclusion, the C14:0 supplementation reduced Ca availability in concentrate-based diets while an additional supply of Ca improved Ca and energy retention. Consequently, Ca supply should exceed recommended levels in diet types where dietary lipids are likely to reduce Ca availability and a compromise in basal diet type has to found to be able to profit best from the energetic value and the methane-suppressing properties of C14:0.  相似文献

The experiment was performed on 4 cows Black and White, 550-650 kg of body weight, before and during 100 days after parturition. Two cows were fed with control and the next two with urea supplemented diet (urea-N constituting about 20% of total N in ration, 13% c.p. in DM). In cows fed with urea supplemented diet the considerable but unequal increase of urinary orotic acid excretion (particularly evident in first 30 days of lactation) was observed. There was however no influence of urea ration on the level of orotic acid in milk. The average orotic acid level in blood plasma (1.14 +/- 0.71 mumol X l-1 and 0.94 +/- 0.22 mumol X l-1 for control and urea fed cow respectively) established 0.01 and 0.001 of the average orotic acid concentration in urine and milk, respectively. Feeding with urea ration did not affect the level of orotic and UMP but increased the uridine concentration in blood plasma. During the parturition the lowest level of orotic acid in blood plasma, milk and urine was observed.  相似文献

The aim of this study was to evaluate the effect of herbage allowance (HA) and type of silage supplemented (TS) on milk yield, dry matter intake (DMI) and metabolism of dairy cows in early lactation. Thirty‐six Holstein‐Friesian dairy cows were allocated to four treatments derived from an arrangement of two HA (LHA = 17 or HHA = 25 kg of DM/cow/day) and two TS (grass (GS) or maize (MS)). Herbage allowance had no effect on DMI or milk yield. Rumen pH and NH3‐N concentration were not affected by HA. The efficiency of microbial protein synthesis in the rumen (microbial protein (MP)) was affected by HA with 21.5 and 23.9 g microbial nitrogen per kg ruminal digestible organic matter for LHA and HHA, respectively (P < 0.05). Supplementation with MS showed higher values of milk yield by 2.4 kg/cow/day (P < 0.001), milk protein content by 0.10 % (P < 0.023) and herbage DMI by 2.2 kg/cow/day, and showed lower values for milk urea compared to GS (P < 0.001). The former results suggest that TS had a greater effect on milk yield, total feed intake and energy intake than increase in herbage allowance; however, increase in HA had greater effects on MP than TS.  相似文献

The effects of a strategic antiparasitic treatment against Hypoderma species and other parasites on the milk yield, composition, and somatic cell counts of 742 multiparous dairy cattle were investigated on 79 farms in a Hypoderma species-endemic area in western Switzerland; 357 of the cows were treated with eprinomectin, 252 with trichlorfon, and 133 were left untreated. The treatments were given between October and early December 1998, on average 53 days before calving. Milk yield, fat, protein and somatic cell counts were measured once a month until the end of lactation. Eprinomectin and trichlorfon significantly increased milk yield during the first month after treatment compared with the control cows by 2.14 (P < 0.001) and 2.50 kg per day (P < 0.001), respectively. Initially, the difference between the eprinomectin- and trichlorfon-treated animals was not significant, but the effect of eprinomectin on milk yield was persistent, whereas the effect of trichlorfon decreased significantly by 0.12 kg per day per month (P < 0.05). The somatic cell counts increased significantly slower in the treated cows than in the control cows (P < 0.05). Milk composition was not affected by the treatments.  相似文献

The objectives of this study were to determine the effects of parity on milk production, body condition change, periparturient health, and culling in Korean dairy herds. The data utilized included; milk yield, body condition score, cow parity, calving condition, periparturient disorders, culling, and reproductive status, which were recorded from 1290 calvings in eight dairy herds. The mean milk yield in cows over 305 days increased with increasing parity (p < 0.01). Cows with parities of 3, 4, and 5 or higher lost more body condition than those with a parity of 1 during month 1 of lactation (p < 0.01), and body condition recovery by cows with parities of 4 and 5 or higher was slower (p < 0.01) than recovery by cows with parities of 1, 2, or 3 until month 3 of lactation. The risk of retained placenta, metabolic disorder, and endometritis also increased with advancing parity (p < 0.05). Moreover, the incidence of ovarian cysts was lower in cows with a parity of one than in cows with greater parities (p < 0.01). Culling rate due to reproductive failure also increased with advancing parity (p < 0.01). These results suggest that parity increases milk yield, body condition loss during early lactation, the risk of periparturient disorders, and culling due to reproductive failure in dairy herds.  相似文献

Adequate blood calcium (Ca) concentrations are a prerequisite to maintain several physiological functions of mammals such as pig and ruminants. Thus, blood Ca levels have to be regulated within very close limits. This is basically ensured through the coordinated effects of the calcitropic hormones parathyroid hormone (PTH) and calcitriol (1,25-dihydroxyvitamin D3). If plasma Ca decreases, one main effect of subsequently secreted PTH is the stimulation of renal reabsorption of Ca from the glomerular filtrate to reduce urinary Ca loss. Another important action of PTH is the induction of the renal enzyme 1 alpha-hydroxylase, which is responsible for the production of calcitriol. In most monogastric species, so far investigated, one of the most important effects of calcitriol is to stimulate active absorption of Ca from the gastrointestinal tract, particularly the upper small intestines, via a vitamin D-receptor-mediated genomic action. Whether this concept can be transferred without any constrictions to ruminants is still under discussion. Marked interspecies differences have to be recognized with respect to ruminant or non-ruminant animals, particularly with respect to individual segments along the gastrointestinal axis and with respect to vitamin D sensitivity of Ca transport mechanisms. This review will elucidate some of the current concepts related to the mechanisms and sites of Ca absorption in pigs and ruminants with special emphasis on dairy cows where Ca homeostasis is occasionally compromised at the time of parturition.  相似文献

This study investigated the effects of low potassium diets with different levels of Ca compared to two diets low in dietary cation–anion difference (DCAD) fed prepartum as a strategy to prevent hypocalcemia on sorting behaviour, total tract digestibility, oxidative status and energy and protein metabolism of transition cows. Forty-eight pregnant dairy cows were assigned to 4 treatment groups: Low Ca, low K (LCLK), High Ca, low K (HCLK), Supplementation with anionic mineral mixture (AMS) supplementation with SoyChlor (CAS). After parturition, all animals were fed a standard postpartum diet. Data were collected until 21 DIM. Prepartum urinary pH was significantly reduced by the low DCAD diets, while postpartum Ca homeostasis was affected by the HCLK ration. Feeding AMS induced sorting against particles <1.18 mm in favour of particles >19 mm prepartum. In contrast, cows fed CAS showed an increase in selective consumption of fine particles and sorted against longer particles similar to the HCLK and LCLK groups. Postpartum sorting activity was not affected by the dietary treatments. After calving, apparent digestibility of NDF was significantly reduced in the HCLK group. Prepartum, we observed effects on serum concentrations of non-esterified fatty acids were higher and insulin sensitivity was lower in the AMS group. Blood urea nitrogen (BUN) was decreased in cows fed the CAS ration. Postpartum, we found serum protein to be decreased with the low DCAD diets while BUN was decreased in the CAS group. The low DCAD rations increased prepartum serum malondialdehyde concentrations, while postpartum total antioxidant capacity was lower in the HCLK and the AMS group. From these data, we conclude that AMS decreased prepartum intake due to compromised palatability. Intermediate protein metabolism was affected by the low DCAD diets, while parameters of oxidative stress were probably affected by acid–base balance and Ca homeostasis.  相似文献

The study postulated that differential nutritional management during the early lactation period would be reflected in endometrial expression of genes related to embryo growth at the end of the voluntary waiting period. Thus, the effect of the combined use of total mixed ration (TMR) and grazing under different herbage allowances during the first 75 days post‐partum (DPP) on endometrial gene expression was evaluated in primiparous dairy cows. Cows were blocked by body weight, age and body condition score and randomly assigned to three grazing treatments: high (HA, 30 kg DM per cow per day), medium (MA, 15 kg DM per cow per day) and low (LA, 7.5 kg DM per cow per day) herbage allowance (mixed pasture, 2,600 kg DM per ha) plus 8 kg DM of supplement or TMR (55% forage, 45% concentrate) fed ad libitum (TMR) from calving to 75 DPP. At 57 DPP, cows were synchronized for oestrus (day 0, 68 DPP) and at day 7, endometrial biopsies were obtained. The nutritional treatment did not affect insulin, IGF‐1 and leptin concentrations on days 0, 4 or 7. Expression of IGF1, IGFBP3, IGFBP4, ADIPOR1 and ADIPOR2 mRNA was significantly affected by the nutritional treatment. Endometrial IGF1 and IGFBP4 mRNA were twofold greater in TMR and HA than MA and LA cows. Expression of IGFBP3 and ADIPOR1 mRNAs was greater in TMR and HA than MA cows, but did not differ from LA cows. All groups had greater expression of ADIPOR2 mRNA than MA cows. This study provided solid evidence of the importance of nutritional management during early lactation on uterine environment at the end of the voluntary waiting period. The greater expression of genes related to embryo growth and uterine function (IGF system, progesterone and adiponectin receptors) in cows fed diets maximizing energy intake suggests a favourable environment for embryonic growth, which may explain the improved reproductive performance of cows in good energy balance.  相似文献

Feeding systems such as grazing affect the fatty acid profile of bovine milk fat. In addition, milk fat is formed as the product of fatty acid metabolism in cow bodies before being secreted into milk. However, how grazing influences milk fatty acid profile through the metabolism has not been completely characterized. When fatty acid concentrations in Holstein milk were compared between grazing and non‐grazing periods, α‐linolenic acid was significantly higher in the grazing period than in the non‐grazing period. This could be explained with an increase in α‐linolenic acid feeding with grazing. α‐linolenic acid had a linear positive correlation with conjugated linoleic acid (9c,11t‐18:2) (CLA) and vaccenic acid (VA) during the grazing period, whereas CLA had higher correlation with linoleic acid rather than with α‐linolenic acid during the non‐grazing period. These data indicate that the high content of dietary α‐linolenic acid affects CLA and VA formation in milk of grazing periods via α‐linolenic acid metabolism into VA.  相似文献
