Blood collected from calves, sheep, goats, pigs, dogs, horses, ponies and donkeys, was allowed to clot under standard conditions. Thromboxane B2 generated during the clotting process was measured by radioimmunoassay in serum harvested from each sample. Highly significant differences were found between species and also between genera within a species. Highest concentrations of thromboxane B2 were detected in the dog samples (887.7 +/- 123.7 ng/ml) and lowest concentrations in samples from sheep (2.7 +/- 0.2 ng/ml). The amount of thromboxane produced per unit number of circulating platelets or per unit volume of platelets in each species was not the same and it would appear that platelets from each species have different inherent ability to produce thromboxane under the stimulus applied, or that some species generate thromboxane from other sources during the clotting process.  相似文献

Rotavirus C (RVC) causes enteric disease in multiple species, including humans, swine, bovines, and canines. To date, the evolutionary relationships of RVC populations circulating in different host species are poorly understood, owing to the low availability of genetic sequence data. To address this gap, we sequenced 45 RVC complete genomes from swine samples collected in the United States and Mexico. A phylogenetic analysis of each genome segment indicates that RVC populations have been evolving independently in human, swine, canine, and bovine hosts for at least the last century, with inter‐species transmission events occurring deep in the phylogenetic tree, and none in the last 100 years. Bovine and canine RVC populations clustered together nine of the 11 gene segments, indicating a shared common ancestor centuries ago. The evolutionary relationships of RVC in humans and swine were more complex, due to the extensive genetic diversity and multiple RVC clades identified in pigs, which were not structured geographically. Topological differences between trees inferred for different genome segments occurred frequently, including at nodes deep in the tree, indicating that RVC's evolutionary history includes multiple reassortment events that occurred a long time ago. Overall, we find that RVC is evolving within host‐defined lineages, but the evolutionary history of RVC is more complex than previously recognized due to the high genetic diversity of RVC in swine, with a common ancestor dating back centuries. Pigs may act as a reservoir host for RVC, and a source of the lineages identified in other species, including humans, but additional sequencing is needed to understand the full diversity of this understudied pathogen across multiple host species.  相似文献

Characterization of antigens from mycoplasmas of animal origin   总被引:4,自引:0,他引:4  
Alcholeplasma laidlawii, Mycoplasma gallisepticum, M mycoides subsp mycoides, M agalactiae, M bovirhinis, mycoplasmal strain ST-6, and culture medium were compared with M bovis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, enzyme-linked immunosorbent assay (ELISA), and gel electrophoresis-derived ELISA. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis indicated there were areas of homology and areas of heterology among the species tested. Sera from rabbits hyperimmunized with the mycoplasma organisms and noninoculated culture medium demonstrated ELISA reactivity with M bovis antigens immobilized on polystyrene. Absorption of the serum from a rabbit hyperimmunized with M bovis reduced 65.9% of its reactivity with culture medium, 29.7% to 32.7% of its reactivity with the heterologous species, and 21.1% of its reactivity with the homologous species. Gel electrophoresis-derived ELISA performed on immobilized M bovis antigens separated by molecular weight, using sera from rabbits hyperimmunized with the mycoplasmal species under study and noninoculated culture medium revealed antigenic components which are shared among species or with the culture medium and several components which may be unique to M bovis.  相似文献

This article is designed to assist the practitioner in the evaluation of bone marrow aspiration biopsies. The indications for marrow evaluation, methods of sample collection, sample preparation, and cytologic examination of bone marrow are discussed. High-performance liquid chromatography (HPLC) was used to determine the phospholipid (PL) composition of ovine, equine, bovine, porcine, and canine RBC membranes. Procedural modifications of established techniques provided for separation of 7 PL within a 15- to 20-minute sample run. Significant (P less than 0.05) differences were detected in RBC membrane PL composition among the various species. The concern for physiologic properties associated with hemolysis and/or sedimentation rate must include evaluation of differences in the PL bilayer structure.  相似文献

Culture of macrophages from bovine bone marrow   总被引:1,自引:0,他引:1  
Macrophages perform important immunoregulatory and host defense functions. Examination of this cell type in the bovine has been restricted because of lack of a means to obtain pure bovine macrophage populations reproducibly. We have developed a system for production of large numbers of macrophages from this species with greater than 99% purity. Stem cells were obtained from the bone marrow of neonatal calves and cultured in vitro in the presence of macrophage-colony-stimulating factor. Bovine bone marrow culture-derived macrophages were esterase-positive, expressed Fc receptors for aggregated IgG, and bovine macrophage differentiation markers. In addition, they displayed class I and class II major histocompatibility (MHC) antigens. The level of MHC antigen expressed could be further enhanced by treatment with recombinant bovine interferons. The macrophages exhibited expected functions, for example, Fc-mediated ingestion of opsonized sheep red blood cells. Augmentation of phagocytic capacity by either alpha or gamma interferon could also be demonstrated. The data reported here confirm that bone marrow culture is a convenient, reliable source of macrophages for investigations of this bovine cell type.  相似文献

Enriched methyl cellulose media together with either human urinary erythropoietin or serum collected from phlebotomized dogs exposed to hypoxia was used in the study of the erythroid colony forming (CFU-E) capacity of dog marrow. The dog serum erythropoietin was found to be more efficient in stimulating CFU-E than comparable concentrations of human urinary erythropoietin. Numbers of CFU-E were directly related to the culture concentration of the stimulating serum and to the number of cells per plate. Sheep plasma erythropoietin was also found to be effective in stimulating CFU-E growth. The system described is chemically better defined and produced more consistent results than has been reported for the plasma clot method.  相似文献

选择商品化的3种细胞裂解液(RNAiso Plus、TRNzol Plus和RNAOUT)和2种DNA/RNA共提试剂盒(RNA-DNA双提试剂盒、快速DNA/RNA共提试剂盒),分别提取猪繁殖与呼吸综合征病毒(PRRSV)JXA1-R株、猪瘟病毒(CSFV)HCLV株和乙型脑炎病毒(JEV)SA-14-14-2株的RNA,通过RNA直接定量和Real-time PCR间接评价了抽提的RNA的质量。RNA直接检测结果表明,RNAiso Plus、TRNzol Plus和RNAOUT 3种细胞裂解液抽提获得的RNA浓度相对较高;Real-time PCR间接检测结果表明,快速DNA/RNA共提试剂盒组RNA相对水平最高,RNAiso Plus和TRNzol Plus 2种细胞裂解液组其次。研究表明,RNAiso Plus、TRNzol Plus和RNAOUT 3种试剂可用于临床RNA病毒检测以及定量分析。  相似文献

The assessment of cutaneous microcirculation by laser-Doppler velocimetry (LDV) has been primarily limited to human studies. The purpose of this investigation was to establish normal values in various species and anatomic sites for blood flow, velocity, and volume as determined by LDV. Microcirculation was measured with a laser-Doppler velocimeter in 54 animals, 6 healthy animals from each of 9 species. The standard sites used were the buttocks, convex surface of the ear, metacarpal pad, humeroscapular junction, thoracolumbar junction, ventral portion of the abdomen, dorsal metacarpus (hooved animals), and ventral surface of the tail (horse). Significant differences in blood flow, velocity, and volume were measured between species and sites within species. The ventral portion of the abdomen consistently had the highest relative blood flow across all species except the monkey. Measurements in the canine metacarpal pad had a high SD, possibly indicating the stratum corneum and epidermis to be too thick for LDV. Our findings provide baseline data in several species, with application of LDV in comparative dermatologic research.  相似文献

Because the central nervous system (CNS) is encased almost entirely in bone, the means by which the clinician can evaluate it are limited. Additionally, the small size of many exotic companion animals further complicates diagnostic evaluation of the brain and spinal cord. Knowledge of the advantages and limitations of different imaging modalities, along with the neuroanatomical localization and assessment of likely causes of disease, will permit the clinician to choose the most appropriate imaging method for the patient. This article discusses the basic imaging principles of radiology, myelography, CT, and MRI of the nervous system of companion exotic animals to aid exotic animal clinicians in selecting imaging modalities and interpreting the results.  相似文献
