Cytoplasmic male sterility (CMS) is an indispensable trait for F1 hybrid seed production in bunching onion (Allium fistulosum L.). Expansion of the cytoplasmic diversity of F1 hybrid cultivars by introduction of various CMS resources has great potential to eliminate vulnerability to cytoplasm type-specific diseases. This study aimed to evaluate appearance frequency of male sterile plants in several bunching onion accessions and to identify CMS resources. In eight (‘Nogiwa Aigara’, ‘Bansei Hanegi’, ‘Amarume’, ‘Kimnung’, ‘Zhangqiu’, ‘INT/CHN/1990/GOTOU’, ‘Natsunegi’ and ‘Guangzhou’) of 135 accessions collected from Japan, China, Mongolia, Korea and Taiwan, male sterile plants appeared with varied frequencies from 1.7% (‘Nogiwa Aigara’ and ‘Bansei Hanegi’) to 24.5% (‘Zhangqiu’). The inheritance mode of Zhangqiu- and Guangzhou-derived male sterility was confirmed to be CMS by sib-crossings and interbreed crossings. Microscopic examination of microsporogenesis in the CMS plants revealed that microspore protoplasm rapidly degenerated without mitotic division after the release of microspores from tetrads. The CMS germplasm described here would be useful for the development of “A” lines to be used in F1 hybrid seed production of bunching onion. Male fertility in ‘Nogiwa Aigara’, ‘Bansei Hanegi’, ‘Kimnung’, ‘INT/CHN/1990/GOTOU’ and ‘Natsunegi’ was verified to be controlled by a single fertility restoration locus.  相似文献

Differentiation of cultivars with simple sequence repeat (SSR) markers is a very useful technique for the true-to-type characterization of cultivars and clarification of parent-offspring relationships. We developed an SSR marker set for cultivar identification comprising 15 markers that were screened from 46 previously published SSRs. This marker set could be used for apple varieties including Malus × domestica and/or other Malus species. These SSRs successfully characterized 95 apples, including the leading and major founding cultivars used worldwide for modern apple breeding. Therefore, this marker set could be applied to almost all apple cultivars. We also analyzed the parent-offspring relationships of 69 cultivars by considering allele transmissions. This analysis revealed the true parentage of the following seven cultivars: ‘Kizashi’, ‘Chinatsu’, ‘Honey Queen’, ‘Haruka’, ‘Seirin’, ‘Ozenokurenai’, and Morioka #48. This analysis also revealed a parentage discrepancy for ‘Hacnine’. From the parent-offspring analysis, two microsatellite mutation events at alleles inherited from pollen parents were observed.  相似文献

Farmers’ seeds are most often lumped together in one broad category called ‘landraces’. But such a category covers variety types that reflect different levels of farmer involvement. Those differences matters when we discuss such issues as genetic erosion, on-farm conservation and seed related policies. The term landrace can be traced to the time when ‘modern’ varieties of cereals were introduced to European farmers in the late nineteenth century. The farmers’ varieties of the time were called ‘landraces’ and understood as seeds adapted to local growing conditions through natural adaptation usually with no intentional selection. But the term was quickly adopted as generic for all farmers’ varieties including those that are bred and maintained by active seed selection on-farm. Such farmer-bred varieties are better termed ‘folk varieties’. The article discusses how interaction of crop characteristics and developing technologies resulted in the evolution of crop varieties as either landraces or folk varieties. It is argued that vulnerability to different agents of genetic erosion and feasibility of on-farm conservation are clearly different for the two categories of farmers’ varieties. Likewise seed policies, particularly the issue of Farmers’ Rights would benefit from clarity of type of farmers’ varieties.  相似文献

Polyploidy is widespread in plants and has played a major role in the evolution and diversification of the plant kingdom. Unreduced (2n) gametes are an interesting tool for polyploidisation and the creation of genetic variation in plant breeding. Especially in ornamentals, polyploidisation can broaden attractive features within a species. A Begonia collection was screened on the occurrence of 2n pollen with the aid of four different techniques: pollen size measurement, flow cytometric analysis of nuclei isolated from germinated and non germinated pollen, investigation of the microsporogenesis and analysis of progeny. In ten of the 70 screened genotypes (B. dregei, B. pearcei, B. ‘Anna Christina’, B. ‘Bubbles’, B. ‘Florence Rita’, B. ‘Orococo’, B. ‘Rubaiyat’, B. ‘Spatflacier’, B. ‘Tamo’ and B276), large pollen were observed with a rather spherical than normal ellipsoidal shape. Flow cytometric data proved that these aberrantly shaped pollen were associated with 2n ploidy levels, although they were not always viable. Meiotic aberrations in these large pollen producers resulted mainly in dyads although also monads, triads and polyads were observed. Successful crosses were obtained with B. dregei, B. ‘Orococo’, and B276 as pollinators; DNA content had increased in all or a part of the progeny. The results show that the occurrence of 2n pollen is not a rare phenomenon in Begonia.  相似文献

The elite and popular cultivars of Cymbopogon martinii were examined for genomic and expressed molecular diversity through RAPD, enzyme and SDS-PAGE protein polymorphisms. The allelic score at each locus of the enzymes as well as presence and absence profiling in RAPDs, overall occurrence of band types etc. were subjected to computation of gene diversity, expected heterozygosity, allele number per locus, and similarity matrix. These, in turn, provide inputs to derive primary account of allelic variability, genetic bases of the cultivated germplasm, putative need for gene/trait introgression from the wild or geographically diverse habitat etc. in elite selections. ‘PRC1’ possessed highest number of unique bands based on RAPD polymorphism. In variety ‘IW31245E’, diaphorase and glutamate oxaloacetate transaminase isozymes generated two unique bands as dia-III 2 and got-II 4. ‘RRL(B)77’ exhibited three unique bands; one produced by esterase as allele est-II 1 and two by malic enzyme (me-III 1,3). Only one unique band was generated by malic enzyme in variety ‘Trishna’. But sofia had three unique bands, two contributed by diaphorase (dia-II 3 and dia-II 4 and one by glutamate oxaloacetate transaminase (got-II 2). SDS-PAGE analysis revealed presence of unique polypeptide fragments (97.7 kDa to 31.6 kDa) in varieties ‘IW31245E’, ‘RRL(B)77’, ‘Tripta’, ‘Trishna’, ‘PRC1’ and var. sofia, generated as a diagnostic marker. In general, molecular distinctions associated with var. motia and var. sofia have been clearly noticed in C. martinii. Genetic capacity for green plant regeneration in anther culture were mapped in a population comprising 50 doubled haploid lines from a cross between two wheat varieties ‘Ciano’ and ‘Walter’ with widely different capacity for green plant regeneration. Bulked segregant analysis with AFLP markers and composite interval mapping detected four QTLs for green plant percentage on chromosomes 2AL (QGpp.kvl-2A), 2BL (QGpp.kvl-2B.1 and QGpp.kvl-2B.2) and 5BL (QGpp.kvl-5B).The three QTLs detected on chromosome 2AL and 2BL all derived their alleles favouring green plant formation from the responsive parent ‘Ciano’.The remaining QTL on chromosome 5BL had the allele favouring green plants from the low responding parent ‘Walter’. In a multiple regression analysis the four QTLs could explain a total of 80% of the genotypic variation for green plant percentage. None of the chromosomal regions with QTLs for green plant percentage showed significant influence on either embryo formation or regeneration frequencies from the anther culture. The three major QTLs located on group two chromosomes were fixed in a second DH population derived from two parents ‘Ciano’ and ‘Benoist’,both with high capacity to produce green plants. A QTL explaining31.5% of the genetic variation for green plant formation were detected on chromosome 5BL in this cross as well. Fourteen developed microsatellite markers were characterized for their use in genotyping and diversity studies of olive varieties. After optimisation of microsatellite assay and allele sizing, ninety-six alleles were found in nineteen varieties, with an average of 6.8 alleles per locus. The characteristics of the microsatellite markers were used to identify markers that can be reliably applied for variety genotyping. Such features were the generation of complex banding patterns supported by underlying allele sequences, `short allele dominance', an unstable repeat structure and a low number of alleles. AFLP analysis was performed on the same set of olive varieties using eight primer pair combinations. The genetic relationships among nineteen olive varieties were compared on the basis of microsatellite and AFLP polymorphisms. Genetic distances between all pairwise combinations of the varieties were calculated using Jaccard's coefficient of similarity and dendrograms were constructed by the UPGMA method. The results of clustering analysis with both molecular systems showed the common genetic background of Tuscan varieties, and genetic divergence within Slovene olive germplasm. Slovenian varieties ‘Buga’, ‘Štorta’ and ‘Samo’ might represent regionally selected olives, while ‘Zelenjak’ and ‘Črnica’ are probably derived from the Central Italian region. The predominant local ‘Istrska belica’ was introduced to Slovenia independently from the other regional varieties and showed the lowest genetic similarity with the other regional varieties. Summary Inheritance of fruit acidity in pomegranate (Punica granatum L.) was studied in 3 sweet or low acid (‘Ganesh’, ‘Ruby’ and ‘Kabul Yellow’) and 3 sour or high acid (‘Nana’, ‘Daru’ and ‘Double Flower’) varieties and their progenies. The F1 and F2 data of ‘Ganesh’ × ‘Nana’ showed that fruit acidity is monogenically controlled and the sour nature is dominant over sweet. Further, whether a genotype produces sweet or sour fruit is determined by a major gene (SS) while a few modifiers with small effects cause fluctuations in the acidity levels within sour and sweet types. All the trees of 3 crosses involving ‘Daru’ produced acidic fruits but those of (‘Ganesh’ × ‘Nana’) × ‘Daru’ reached acidity as high as 71.2 g/l which could be because of cumulative influence of modifying genes derived from the two acidic varieties ‘Nana’ and ‘Daru’. Pollination of functionally sterile ‘Double Flower’ variety with single (normal) flower types revealed that ‘Double Flower’ is a dominant mutant from an acidic fruited genotype (Ss). The segregation pattern in F1 indicated the possible linkage between genes governing total acidity and flower type. All the F1 hybrids between ‘Kabul Yellow’ and ‘Ganesh’ (sweet × sweet) were sour fruited with almost 8-fold jump in fruit acidity over the mid-parental value. The steep increase in acidity cannot be convincingly attributed to overdominance which is certainly rare at major gene level. Alternatively, linked dominant alleles or epistatic effect of neighboring loci which readily simulate overdominance (pseudo-overdominance) could have caused a major shift in F1 fruit acidity.  相似文献

Genotypes for the glume colour character have been studied in 27 cultivars of common wheat (Triticum aestivum L.) originated from old landraces, and 1 specimen of T. petropavlovskyi Udacz. et Migusch. by means of analysis of the F2 populations. The following tester lines have been used: white-glumed ‘Novosibirskaya 67’ ‘Diamant I’, and ‘Federation’, carrying the Rg1 gene alone; lines RL5405 and near-isogenic ‘Saratovskaya 29’ *5 (T. timopheevii Zhuk./T. tauschii (Coss.) Schmal.), carrying Rg2; line (1A ‘CS’ × ‘Strela’) with Rg3. The red glume colour in 21 cultivars of Triticum aestivum and in the accession of T. petropavlovskyi has been shown to be determined by the single gene Rg1, located on chromosome 1B. Five cultivars carrying the gene Rg3 for red glumes on chromosome 1A have been revealed. The cultivars ‘Zhnitsa’ and ‘Iskra’ carry the gene Rg3 alone. The red glume colour in the cultivars ‘Milturum 321’, ‘Milturum 2078’, ‘Sredneural'skaya’ is controlled by two genes, Rg1 and Rg3. In two common wheat cultivars, ‘Sarrubra’ and ‘Krasnoyarskaya 1103’ the red glume colour is determined by Rg1, inherited from local populations (‘Turka’ and ‘Kubanka’ respectively) of tetraploid wheat T. durum Desf. var. hordeiforme Host. Wide occurrence of the Rg1 gene in common wheat has been confirmed. On the contrary, none of the investigated varieties carries the gene Rg2. Summary Extensive linkage maps, consisting primarily of molecular markers, are being developed for apple, pear, and grape varieties. The intrinsically high heterozygosity of outcrossing perennial species is utilized to produce segregating populations directly from a cross between varieties. Nearly complete linkage maps have been generated for the apple varieties ‘Rome Beauty’ and ‘White Angel’. The map for ‘Rome Beauty’ contains 161 molecular markers, while that for ‘White Angel’ has 251 markers. Maps for the pear varieties, ‘Bartlett’ and NY10353, also are being developed. Linkages conserved between the pear and apple genomes have been identified. In grapes, maps for four varieties are available, the most extensive being those for ‘Cayuga White’ and ‘Aurore’. The apple maps have been used to investigate the genetic basis of morphological and physiological characters. A gene controlling the presence of anthocyanins in the skin of the fruit is located on linkage group 3. Genes controlling early bud break, branching habit, and production of root suckers have also been identified and mapped.  相似文献

While the roles of sugars and acids in fruit flavour are well known and described, the roles of volatile compounds that contribute to flavour and odour are more difficult to define. The determination of volatiles that make a significant contribution to the flavour of kiwifruit (Actinidia chinensis) is essential for efficient and cost effective flavour breeding. The aims of this study were to explore associations between volatile compounds, sensory and fruit attributes of A. chinensis fruit and to identify potential key flavour impact volatiles, superior parents and selection methods, using multivariate analysis. We investigated the volatiles produced by ripe fruit of 24 genotypes selected by principal component analysis (PCA) to represent the diversity of taste and fruit characteristics in a breeding population. Seventy-two volatiles were detected. Extended use of multivariate analysis proved powerful for gaining maximum information from the limited plant material. The volatiles were successfully grouped into four clusters, using hierarchical clustering of variables based on phenotypic correlations between volatiles to avoid a singular correlation matrix in PCA. Based on these clusters, associations between volatiles, sensory and fruit attributes were explored using PCA and multiple linear regressions. Principal components provided a measure of the balance of complex volatiles that is likely to affect consumer responses. Thirteen potential key impact volatiles that made a substantial contribution to the flavour of A. chinensis fruit were identified. Five esters were strongly associated with the flavours and odours characteristic of ‘Hort16A’. Volatiles associated with ‘sour taste’, ‘fruit ripeness’, ‘atypical ‘Hort16A’-like odour’ and ‘atypical kiwifruit flavour’ were also recognized. Parents with potentially desirable volatile profiles and a possible selection method for flavour breeding were identified.  相似文献

In vitro shoots of six pear (Pyrus communis L.) cultivars, ‘Conference’, ‘Doyennéd'Hiver’, ‘Passe Crassane’, ‘Bartlett’, ‘AbbéFetel’ and ‘Butirra Precoce Morettini’ were irradiated with gamma rays (3.5 Gy). After three subcultures, microcuttings from the irradiated shoots and from additional non-irradiated microcuttings were rooted to establish plants for survey orchards. All trees were individually observed for variation in fruit traits and for productivity. Trees were selected for improved characters related to production such as early bearing and consistent productivity. Variations observed in fruit appearance concerned degree of russeting, fruit shape and size. The frequencies of the observed variations in fruit traits depended on the cultivar, ranging from 0.81% in ‘Doyennéd'Hiver’ to 3.64% in ‘Passe Crassane’. Fusarium wilt incited by Fusarium oxysporum f. sp. melonis (F.o.m) is one of the most widespread and devastating melon diseases. While resistance to physiological races 0, 1, and 2 is relatively frequent in different botanical varieties, sources of resistance to race 1,2 are restricted to a few Far-Eastern accessions. In this work, the results of a screening for resistance to F.o.m. race 1,2 among 32 accessions are presented. Three Japanese accessions (‘Kogane Nashi Makuwa’, ‘C-211’, and ‘C-40’) showed the highest resistance levels, but useful levels of resistance were also detected in one Russian ‘C-160’ and two Spanish (‘C-300’ and ‘Mollerusa-7’) accessions. These resistant materials, together with other accessions previously described as resistant to F.o.m. races 0, 1, and/or 2 have been morphologically and molecularly characterized. Based on cluster analysis, these accessions have been grouped according to the botanical subspecies they belong to. Assessment of genetic diversity indicated that the resistant accessions to races 0, 1 and 2, are scattered along the established clusters. On the other hand, high levels of resistance to the race 1,2 could be found only among accessions belonging to Cucumis melo subsp. agrestis, nevertheless, a certain degree of resistance to this race could also be found within some accessions belonging to subsp. melo. As far as we know, this is the first report of resistance to F.o.m race 1,2 found out from the Far-Eastern melon material. Based on fruits characteristics, it appears that several inodurus and cantalupensis accessions could be exploited in breeding programs as resistance sources to F.o.m races 0, 1 and/or 2 for the improvement of these melon types. The accessions with the highest levels of resistance to the race 1,2 appeared to be very distant both molecularly and morphologically from the commercial types. Nevertheless ‘C-160’, ‘C-300’, and ‘Mollerusa-7’ classified as var. inodorus are morphologically very similar to the Spanish commercial types and might be used as resistant sources in breeding these melon types.  相似文献

The quantity and quality of pollen produced by a genotype are important attributes to be considered in the selection of male parents for breeding. The objective of this research was to determine the quantity and quality of pollen in 13 selected East African Highland bananas (EAHB) in relation to six diploids commonly used as male parents. Pollen quantity was scored on a scale of 0–6 while the variation of pollen stainability along the rachis was determined using acetocarmine glycerol jelly. Actual male fertility was determined by using pollen from the EAHB varieties on M. acuminata spp. burmannicoides, ‘Calcutta 4’, as the female. The diameter of 200 viable pollen grains was measured under a microscope with the aid of a graduated eyepiece. Significant differences were obtained for mean pollen stainability and pollen diameter. The pollen stainability for diploids was higher than that for the EAHB. Node numbers contributed significantly to variation of pollen stainability in EAHB (P ≤ 0.01) and ‘Pisang lilin’ (P ≤ 0.001). ‘Pisang lilin’ and the EAHB varieties had about 100 nodes as opposed to about 200 in other diploids. The EAHB varieties produced less pollen than most of the diploids. The actual male fertility of EAHB varieties was low (1 seed/bunch), compared to that of ‘Calcutta 4’ (223.3 seeds/bunch) and ‘Pisang lilin’ (13.7 seeds/bunch). Although the production of 2n pollen in EAHB varieties was low (0–7%), the potential exists for using them in new breeding approaches.  相似文献

Optimal carbamate treatment conditions were studied for flower induction in seedlings of an asparagus cultivar ‘Mary Washington 500 W’. Flower induction was most accelerated by soaking seeds in 50 mg l−1 carbamate solution for 12 days at 25 °C under the fluorescent light. Longer exposure to carbamate over a 12 day period induced a higher percentage of seedlings to flower. A higher percentage of flowering seedlings were male. Flower induction frequency among seven cultivars through carbamate treatment widely ranged from 13 to 67%. ‘Geynlim’, ‘Mary Washington 500 W’ and ‘Welcome’ exhibited a high percentage of flowering seedlings, while ‘Larac’ and ‘Vulkan’ showed low values. Only male flowers were induced in all-male cultivars. Variation in pollen germination was found within all cultivars. ‘Geynlim’, ‘Cito’ and ‘Mary Washington 500 W’ showed high values of average pollen germination. Application of carbamate compound to induce flower production can rapidly produce homogenic cultivars which include both sexes. This is necessary for genetic studies and breeding purposes. The nutrient mining abilities of six Musa genotypes: ‘Agbagba’, ‘PITA 22’, ‘Nsukka Local’, ‘FHIA 17’, ‘Fougamou’, and ‘BITA 7’, grown in organic medium formulated by decomposing rice husks with poultry manure in volume proportions of 3:2, were determined at the Teaching and Research Farm of the Department of Crop Science, University of Nigeria, Nsukka, Nigeria. Results indicated significant variation in nutrient mining with respect to Musa genotype and duration of growth. The medium previously cropped with ‘Agbagba’ recorded highest residual mean values for P and Ca, while the medium cropped with ‘Fougamou’ had the highest residual concentration of Mg and S. Residual amount of K was highest in the medium where ‘PITA 22’ and ‘Nsukka Local’ were previously grown. Peak residual concentration of P and K occurred in the medium in which Musa plants were grown only for 4 months, decreasing thereafter, in contrast to Ca, Mg, and S. Residual N tended to increase with longer growth duration of the Musa crops. Follower crop of maize grown in the medium previously cropped with Musa genotypes showed variable performance. While medium previously cropped with ‘PITA 22’ and ‘BITA 7’ favored growth attributes and leaf greenness of the follower maize plant, ‘Fougamou’ favored maize fresh weight attributes. Ectomycorrhiza association was observed in medium previously cropped to ‘BITA 7’ and ‘PITA 22’. Generally, ‘Agbagba’ and ‘Fougamou’ seemed to be the most reticent nutrient miners while uptake of P and K appeared to be low 4 months after planting (MAP) in contrast to Ca, Mg, and S uptake. However, substrate previously cropped with ‘PITA 22’, ‘BITA 7’ and ‘Fougamou’ gave rise to superior growth and fresh weight attributes, respectively, of the maize, the follower crop. A prospect of utilizing the ectomycorrhiza association observed with some genotypes to upgrade Musa yields and those of associated or follower crops exist.  相似文献

Summary ‘Wijcik’, a sport of ‘McIntosh’ with columnar growth habit, was crossed with ‘Wellington Bloomless’, which has apetalous flowers and bears parthenocarpic seedless fruit if not pollinated. The seedlings segregated for columnar versus normal habit but all had normal flowers and fruit. Four columnar seedlings were crossed with ‘Spencer Seedless’, another apetalous cultivar, and the resulting seedlings segregated not only for plant habit but also for apetalous versus normal flowers, approximately 1 : 1. Thus apetaly is controlled by a recessive gene, for which the symbolape is proposed. Apetalous columnar apples may be useful for planting in very high density orchards, cropping without pollination and thus not dependent on bees, pollinator varieties and warm weather at flowering time; moreover, being seedless they may avoid biennial bearing tendencies that are attributable to developing seeds inhibiting fruit bud formation.  相似文献

Twenty-eight Egyptian cotton (Gossypium barbadense L.) genotypes (varieties and hybrids) were used for analysis of genetic diversity using DNA based markers (ISSR, SSR, and EST) and to study varietal development of cotton. The ISSR markers gave the highest percentage of polymorphic bands as well as polymorphic information content compared with the other molecular markers (i.e. EST and SSR markers). Using clustering analysis, no general clustering according to the pedigree history of the genotypes was observed. Using principal coordinate analysis (PCOORDA), cotton genotypes were separated by the first three principal coordinates (PC1, PC2, and PC3) accounting for 11.5, 8.6, and 7.2% of the total genetic variance, respectively. The cotton genotypes were distributed into three parts based on the first PC, each part containing a group of varieties having a common ancestor. ‘Giza 12’ variety was the common ancestor for the varieties included in the first part and ‘Ashmouni’ variety was the common ancestor for the varieties included in the second part, while both ‘Sakha 3’ and ‘Sakha 4’ varieties were common ancestors for the varieties included in the third part. The results of the PCOORDA also showed better resolution of the genetic diversity than cluster analysis especially in the illustration of the varietal development of cotton. That means that principal coordinate analysis can be strongly used either alone or in combination with cluster analysis to discuss both genetic diversity and varietal development in the cotton genotypes.  相似文献

The pear cultivar ‘Osa-Nijisseiki’ (S2Ssm 4; sm = stylar-part mutant) has been used as a parent to breed self-compatible cultivars that produce excellent fruits. However, determination of the self-compatibility of ‘Osa-Nijisseiki’ offspring requires a lot of time, 6 years or more, by conventional cross breeding. We have designed a rapid reliable method for the identification of self-compatible varieties of ‘Osa-Nijisseiki’ offspring based on the polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) with S-allele specific restriction endonucleases. By using this method, 8 self-compatible varieties were selected among 16 selections resulting from a cross between the self-compatible cultivar ‘Osa-Nijisseiki’ (S2Ssm 4) and the self-incompatible cultivars ‘Niitaka’ (S3S9), ‘Whasan’ (S3S5), ‘Chuwhangbae’ (S4S6). The S-genotypes of 16 ‘Osa-Nijisseiki’ offsprings were also determined. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献
