For two successive years, cell walls were isolated from the internal part of skin cells of Vitis vinifera L. cv. Cabernet Sauvignon grape berries grown in a vineyard. Procyanidin localization and composition were determined over the course of development. Tannins were mainly localized in the inner cell fraction, due to their biosynthesis and storage. Cell wall tannins always exhibited a higher mean degree of polymerization as compared to the internal cell fraction, which had a constant mDP. The mDP of cell wall tannins also tended to increase at the end of maturation. Our results suggest tannin polymerization near the cell wall but an aggregation in the vacuole during growth. The tannin composition was typical of skins, and small differences were noted between the two cell parts. Surprisingly, epigallocatechin-3-gallate was also detected, although in a very small amount. Epicatechin was present in significant proportions in both fractions, especially as an extension subunit, while epigallocatechin was likewise abundant, also as a terminal subunit. Last, procyanidin composition and organization seemed to be characteristic of the Cabernet Sauvignon variety.  相似文献

Proanthocyanidins are supposed to have some therapeutical properties as antioxidants and antineoplasics. Most of the proanthocyanidins, however, are not commercialized since their separation from natural sources is either very expensive or not well-known. In this work, the feasibility of application of mixtures of carbon dioxide and alcohol under supercritical conditions for selective extraction of some phenolic compounds from grape seeds has been studied, among them some low polymerized proanthocyanidins, their main monomer units, (+)-catechin and (-)-epicatechin, and some low molecular weight phenolic compounds, like gallic acid. An analytical-scale supercritical fluid extractor, whose operation was previously optimized, was used to carry out the experiments. A commercial concentrate of complex phenols and tannins from grape seeds was subjected to supercritical extraction in order to find the best operation conditions before directly extracting defatted milled grape seeds. The solvent capacity was found to increase with pressure and with the amount of alcohol used as cosolvent as expected. Such variation in solvent capacity could be used for design of a selective separation process where individual phenolic compounds or groups of them could be obtained. HPLC coupled with two types of detectors, diode array and mass spectrometry, was used for tentative identification and quantification of complex phenols and tannins in the extracts and in the raw materials used for extraction.  相似文献

To evaluate the amount and type of condensed tannins binding salivary proteins, which are supposed to be involved in astringent sensation, model systems allowing further analyses of proteins and condensed tannins were developed. The precipitates formed after addition of grape seed tannins to salivary proteins indicate that a binding interaction occurs. Dissociation of insoluble complexes was achieved by sodium dodecyl sulfate treatment. Thiolysis reaction allowed the quantification and characterization of proanthocyanidins on both the resulting pellet and the supernatant. Binding proteins were investigated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The higher polymerized tannins predominantly precipitated together with the salivary proteins. The condensed tannins remaining in solution were low molecular weight polymers.  相似文献

Quantitative and qualitative modifications of tannins and anthocyanins in grape skin were investigated at different dates of harvest, from berries sorted on the basis of their density. Free anthocyanins accumulated until 170 g/L of sugars in pulp before undergoing a slight decrease. Changes in anthocyanin composition were observed with increasing sugar levels in the pulp that reflected structural differences between classes of anthocyanins. The proportion of methoxylated anthocyanins continued to increase in the skin as sugar accumulated while the proportion of coumaroylated anthocyanins initially increased (up to 200 g/L of sugars in the pulp) and then rapidly decreased. In comparison, no major quantitative nor qualitative change was observed for tannins, except for a slight increase of the mean degree of polymerization. Whatever the physiological stage of the pulp, the extraction yield of skin phenolics into hydroalcoholic solution for 5 h was lower than 77% for anthocyanins and 38% for proanthocyanidins. For both classes of compounds, no clear evolution in these extraction yields could be observed as sugars accumulated in pulp (from 162.6 to 275.0 g/L). Nevertheless, some structural features within each family of compounds significantly influenced extractability, for example, a lower extraction yield for coumaroylated anthocyanins and for tannins with a high degree of polymerization. Finally, no direct relationship could be found in extraction media between the amounts of all red pigments (measured in acidic conditions) and the color intensity at 520 nm (measured in wine-like model solutions).  相似文献

The composition of seed storage proteins (SSPs) in olive endosperm and cotyledon has been analyzed. Precursor forms of these proteins are made up of individual proteins, which have been purified to homogeneity and further named p1-p5 (20.5, 21.5, 25.5, 27.5, and 30 kDa, respectively). N-terminal sequences of p1 and p2 proteins displayed relevant homology to the basic subunit of the 11S family of plant SSPs (legumins). Two-dimensional polyacrylamide gel electrophoresis experiments allowed us to verify the basic character of p1 and p2 and the acidic character of p3, p4, and p5 proteins. In addition, the putative presence of highly similar isoforms or posttranslational modifications of these polypeptides was detected. As a result, a model describing the putative association of p1-p5 proteins into subunits of alpha(acidic)/beta(basic) type has been proposed. Solubility experiments have shown that the majority of these olive seed proteins from the 11S storage protein family are extracted with aqueous alcohol and only partially with water and diluted saline solutions, therefore suggesting their similarity to prolamines. Moreover, no visible differences were found in either subunit composition or 11S proteins mass among six olive cultivars examined. This result suggests that the synthesis of storage proteins is highly conserved in this plant species. By using a rabbit antiserum raised to p1 protein, the proteins have also been immunolocalized in olive seed tissues, showing that they accumulate in conspicuous protein bodies present in both the endosperm and the cotyledon.  相似文献

Previous work has shown that red wines, grape juices, and other grape products cause endothelium-dependent relaxation (EDR) of blood vessels in vitro by increasing nitric oxide production. In this paper we describe the isolation and characterization of some of the compounds responsible for EDR activity. Concord grape seeds were extracted with methanol and the compounds were separated by Toyopearl TSK HW-40S chromatography. Resulting fractions (primarily phenolic acids, catechins, and proanthocyanidins) were further separated semipreparatively by reversed-phase HPLC, and peaks were collected and bioassayed for EDR activity using the rat aorta preparation. EDR-active compounds were subsequently characterized by HPLC retention times and electrospray-ion-trap mass spectrometry. The compounds exhibiting the most EDR activity were proanthocyanidin trimers, tetramers, pentamers, and polymers and their gallates, as well as a dimer gallate (EC50 values in the range of 0.6-2.5 microg catechin equivalents/mL). These compounds should be useful for in vitro and in vivo studies, particularly as they relate to improvement of cardiovascular function.  相似文献

We compared the concentrations and contents of protein and oil in mature seeds from nodulated and non-nodulated soybean plants grown on soils with four different N levels during maturation. We observed a positive correlation between the contents of protein and oil in seeds from nodulated plants. Seeds from nodulated plants grown on urea-treated soil showed higher protein and lower oil contents than those from plants grown on soil treated with coated slow release N fertilizer (LP-100). Contents of these compounds in seeds from nodulated plants grown on LP-100 soil were almost the same as those from non-nodulated plants on the same soil. These observations indicated that N economy in roots during seed maturation affects the contents of storage compounds. We suggested that the control of the N2 fixation activity of soybean plants and management of soil N level during seed maturation are important to determine the contents of protein and oil in seeds.  相似文献

A simple, fast, and more selective approach is presented in this study for the identification of haze-active proteins. Grape seed proteins were unfolded by 1% SDS and then interacted with different amounts of tannin at 4 degrees C, followed by gel electrophoresis. It was found that the intensity of the band at 45 kDa was decreased as tannins increased. The amino acid composition of this isolated 45-kDa protein was higher in proline (9.49%) than the average proline content of total grape seed proteins (4.85%). To verify the selectivity of the proposed method, a globular protein (bovine serum albumin, BSA) and a proline-rich protein (gelatin) were selected and used in the model system. As expected, gelatin was removed as it reacted with the increasing added tannins, whereas BSA did not. These results showed that it is possible to identify haze-active proteins by modulating the accessibility of protein to tannins, suggesting this new method can be used by the beverage industry to trouble-shoot haze problems and for quality control.  相似文献

Samples (n = 620) of homogenized red grape berries were analyzed using a visible and near-infrared (NIR) spectrophotometer (400-2500 nm) in reflectance. The spectra and the analytical data were used to develop partial least-squares calibrations to predict dry matter (DM) content and condensed tannins (CT) concentrations. The coefficient of determination in cross-validation and the standard error of cross-validation were 0.92 and 0.83% w/w for DM and 0.86 and 0.46 mg/g epicatechin equivalents for CT, respectively. The standard error in prediction was 1.34% w/w for DM and 0.89 mg/g epicatechin equivalents for CT, respectively. By implementing a NIR spectroscopy method to measure DM and CT in red grape homogenates, we have developed an approach that is suited to large-scale compositional analysis in commercial wine production facilities, as it enables the analysis of large numbers of samples needed to stream batches of fruit. From an economical point of view, the calibration models could be achieved with relatively small data sets. Thus, NIR offers a suitable and efficient tool for the simultaneous measurement of DM and CT in addition to other important parameters in red grape homogenates such as total anthocyanins, total soluble solids, and pH, with minimal sample preparation and low cost.  相似文献

Cabernet franc berries were sampled at five stages from berry set to harvest from an experimental vineyard in mid Loire Valley. Seeds were collected from representative berries in term of stage of development. The evolution of seed was followed both macro- and microscopically. For microscopy analysis, seeds were cut, put in a fixation solution, and cut into thin sections with a microtome. Five staining solutions were used for each seed sample: toluidine blue O, phloroglucinol, periodic acid-Schiff's reagent and naphtol blue black, vanillin, and p-dimethylaminocinnamaldehyde. Toluidine blue O staining revealed the evolution of tissue structures during grape seed development. We studied the changes in chemical compounds (lignin, polysaccharides, proteins, and tannins) with the other reagents. Seed lignification was achieved at veraison. Proanthocyanidins were localized in epidermis, inner cells of the soft seed coat, and inner cell layer of the inner integument. Finally, the localization of flavan-3-ols was linked with changes in cell walls of the outer integument.  相似文献

Wine producers may attempt to increase vine productivity, which usually decreases the quality. The quality of Cava (Spanish sparkling wine; Certified Brand of Origin in Spain) mainly depends on the composition of its colloids, which also determine foam quality. The yield (kg of grape/ha) and maturation index (ratio between soluble solids and titratable acidity) are two external factors that may alter grape characteristics, especially its composition. Twenty white grapes of the same variety (Parellada), but different yields (between 6000 and 17,000 kg/ha) were studied. By PCA, two grape groups were separated according to yield (<10,500 and >10,500 kg/ha). Yield and maturation index were not significantly correlated. Grapes obtained with yield >10,500 kg/ha had significantly higher content of soluble proteins and lower content of total polysaccharides and some polysaccharides fractions (M(r) 62,000-48,000, M(r) 28,000-24,000, and M(r) 7000-5000) than the grapes of the low-yield group (<10 500 kg/ha). Thus, it may be interesting to monitor yield, below 10 500 kg/ha, in vines used to produce Cava.  相似文献

为了开发植物源葡萄籽补钙制剂,该研究通过亚细胞定位试验表明,葡萄籽的胚乳中含有大量的钙元素。通过电泳分析发现,葡萄籽的水溶性蛋白包括2种主要成分,其中一种是11 S球蛋白(蛋白质B),也是最主要的钙结合蛋白,另一种是表观分子量为670 k Da的蛋白质A。在蛋白质组成相同的情况下,用传统的碱溶酸沉法来分离葡萄籽蛋白会导致大量的钙流失。但用30%~50%硫酸铵沉淀法得到的蛋白质得率是(22.5±0.02)g/kg,蛋白质中钙质量分数(3.47%)是碱溶酸沉法(1.11%)的3倍。该研究结果为食品工业中矿物结合蛋白质的分离纯化提供参考。  相似文献

The effect of addition of grape seed tannins on the phenolic composition, chromatic characteristics, and antioxidant activity of red wine was studied. Two highly pure commercial grape seed tannins (GSE100 and GSE300) were selected, and their phenolic compositions were determined. Two types of red wines were made with Castela?o/Tinta Miu?da (3/2, w/w) grapevine varieties by fermentation on skin using two different maceration times, which correspond to the wines rich and poor in polyphenols, respectively. Each of these wines was used for experimentation with the addition of GSE100 and GSE300 before and immediately after alcoholic fermentation. Phenolic composition, chromatic characteristics, and antioxidant activity of the finished red wines were analyzed by HPLC-DAD, CIElab 76 convention, and DPPH radical test, respectively. The results showed that the addition of grape seed tannins had obvious effects of increasing color intensity and antioxidant activity only in the wines poor in polyphenols. Although GSE300 contained much higher amounts of di- and trimer procyanidins and a lower amount of polymeric proanthocyanidins, it provided effects of increasing the color intensity and antioxidant activity of the wines poor in polyphenols similar to those of GSE100. Furthermore, GSE100 released more gallic acid to wines than GSE300, although no gallic acid was detected in GSE100. Tannins added after alcoholic fermentation had a better effect on phenolic composition of red wine than tannins added before alcoholic fermentation.  相似文献

Grape seeds and skins are good sources of phytochemicals such as gallic acid, catechin, and epicatechin and are suitable raw materials for the production of antioxidative dietary supplements. The differences in levels of the major monomeric flavanols and phenolic acids in seeds and skins from grapes of Vitis vinifera varieties Merlot and Chardonnay and in seeds from grapes of Vitis rotundifolia variety Muscadine were determined, and the antioxidant activities of these components were assessed. The contribution of the major monomeric flavonols and phenolic acid to the total antioxidant capacity of grape seeds and skins was also determined. Gallic acid, monomeric catechin, and epicatechin concentrations were 99, 12, and 96 mg/100 g of dry matter (dm) in Muscadine seeds, 15, 358, and 421 mg/100 g of dm in Chardonnay seeds, and 10, 127, and 115 mg/100 g of dm in Merlot seeds, respectively. Concentrations of these three compounds were lower in winery byproduct grape skins than in seeds. These three major phenolic constituents of grape seeds contributed <26% to the antioxidant capacity measured as ORAC on the basis of the corrected concentrations of gallic acid, catechin, and epicatechin in grape byproducts. Peroxyl radical scavenging activities of phenolics present in grape seeds or skins in decreasing order were resveratrol > catechin > epicatechin = gallocatechin > gallic acid = ellagic acid. The results indicated that dimeric, trimeric, oligomeric, or polymeric procyanidins account for most of the superior antioxidant capacity of grape seeds.  相似文献

The phenolic composition of Malbec (Vitis vinifera L.) grape skins and seeds during ripening and the effect of cluster thinning (CT) in two consecutive seasons (2008-2009) were evaluated by high-performance liquid chromatography-diode array detection/electrospray ionization-mass spectrometry (HPLC-DAD/ESI-MS). Removal of 50% of clusters was performed at 40 days (T1), 80 days (T2), and 100 days after flowering (T3) in a vineyard located in southern Mendoza (Argentina). Yield components, with the exception of cluster weight, were significantly affected by CT in both seasons, but no statistically significant differences were found among treatments. Cluster thinning and its timing had little or no influence on physical parameters and fruit chemical composition, and the differences with respect to the control were mainly due to the season. At harvest in 2008, T1 encouraged the biosynthesis of individual anthocyanins in skins, generating 44.0, 39.6, and 41.2% more glucosylated, acetylated, and total anthocyanins, respectively, as compared to the control, whereas in seeds, T1 and T2 mainly changed the concentrations of (+)-catechin, epicatechin-3-gallate, procyanidin B4, dimer gallate 1, trimer gallate 2, and tetramer. Conversely in 2009, T1 significantly affected the content of flavanols and flavonols in skins, whereas in seeds, T1 and T2 modified the level of (+)-catechin, procyanidins B4 and B6, and trimer gallate 2. Moreover, in 2008 the grapes had a higher concentration of most phenolic compounds, indicating a greater potential for more complex wines. Finally, dihydroquercetin-3-glucoside was the major compound among all nonanthocyanin phenolics detected in Malbec skins and represented 25.7% (2008) and 39.9% (2009) of the total content of those compounds at harvest. This finding could represent a distinctive feature of this grape variety.  相似文献

试图揭示板栗种子褐变与脱水敏感性和可溶性糖含量之间的关系。板栗种子成熟脱落时,脱水敏感性相对较弱,子叶和胚轴中棉籽糖和水苏糖含量达最高峰,电导率和紫外吸收值最低,总酚含量与POD活性较低。种子脱落后,脱水敏感性上升,子叶和胚轴中棉籽糖和水苏糖含量下降,膜渗漏物质增多,总酚含量及POD活性急剧上升,从而导致褐变发生。胚轴中总酚含量与POD活性存在显著的正相关(r=0.999),而棉籽糖含量分别与POD活性和总酚含量存在显著的负相关(r分别为-0.998和-0.995)。结果表明,可溶性糖的组分及其含量可能与板栗种子脱水敏感性大小有关,板栗种子脱落后还原性糖麦芽糖含量的上升与褐变的发生趋势一致,而非还原性糖特别是棉籽糖可能在抑制褐变方面具有一定作用。  相似文献

The composition of grape (Vitis vinifera L. cv. Shiraz) skin proanthocyanidins has been determined at different stages of berry development. Beginning approximately 3 weeks after fruit set and concluding at commercial ripeness, the composition of isolated skin proanthocyanidins was determined using the following analytical techniques: elemental analysis, UV-Vis absorption spectroscopy, reversed-phase HPLC after acid-catalysis in the presence of excess phloroglucinol, gel permeation chromatography, electrospray ionization mass spectrometry (ESI-MS), and (13)C NMR. On the basis of these analyses, berry development was correlated with an increase in proanthocyanidin mean degree of polymerization, an increase in the proportion of (-)-epigallocatechin extension subunits, and increases in the level of anthocyanins associated with the proanthocyanidin fraction. Additionally, data acquired from ESI-MS of the isolates following acid-catalysis in the presence of excess phloroglucinol is consistent with pectin-bound proanthocyanidins.  相似文献

Soybean seeds are an important source of dietary tocopherols, but like seeds of other dicotyledonous plants, they contain relatively little alpha-tocopherol, the form with the greatest vitamin E activity. To evaluate potential effects of environmental stress during seed maturation on tocopherols, soybeans were raised in greenhouses at nominal average temperatures of 23 degrees C or 28 degrees C during seed fill, with or without simultaneous drought (soil moisture at 10-25% of capacity), during normal growing seasons in 1999 (cvs. Essex and Forrest) and 2000 (cvs. Essex, Forrest, and Williams). Total free (nonesterified) tocopherols increased slightly in response to drought in Essex and Forrest. All three lines responded to elevated temperature and, to a lesser extent, drought with large (2-3-fold) increases in alpha-tocopherol and corresponding decreases in delta-tocopherol and gamma-tocopherol. The results suggest that weather or climate can significantly affect seed tocopherols. It may be possible to breed for elevated alpha-tocopherols by selecting for altered plant response to temperature.  相似文献

Condensed tannins (CT) can strongly affect litter decomposition, but their fate during the decomposition process, in particular as influenced by detritivore consumption, is not well understood. We tested the hypothesis that litter CT are reduced by the gut passage of two functionally distinct detritivores of Mediterranean forests, the millipede Glomeris marginata, and the land snail Pomatias elegans, as a fixed proportion of initial litter CT, but more so in Pomatias since snails are known to have a more efficient enzymatic capacity. Contrary to our hypothesis, both detritivore species reduced litter CT to near zero in their faecal pellets irrespective of the wide range in initial leaf litter CT concentrations of 9-188 mg g−1 d m among three Mediterranean tree species (Pistacia terebinthus, Quercus ilex, Alnus glutinosa) and different decomposition stages of their litter. The almost complete disappearance of CT even from some litter types highly concentrated in CT, due to either degradation by gut microorganism or complexation of CT into insoluble high molecular weight structures, suggests a high “de-tanning” efficiency across functionally distinct detritivore species. The transformation of CT-rich litter into virtually CT-free faecal pellets by detritivores might be highly relevant for the subsequent decomposition process in ecosystems with a high macrofauna abundance and CT-rich plant species such as Mediterranean forests.  相似文献
