The field strain of Anopheles stephensi, the main malaria vector in south of Iran, was colonized in laboratory and selected with DDT and dieldrin in two separate lines for 3 generations to a level of 19.5- and 14-fold for DDT and dieldrin resistance, respectively. Synergist tests with chlorofenethol (DMC) and piperonyl butoxide (PBO) on the selected strains indicated that dehydrochlorination and oxidative detoxification might be the underlying mechanisms involved in the resistance to dieldrin and DDT in selected strains. DDT selection decreased susceptibility to DDT and pyrethroids including lambdacyhalothrin, permethrin deltamethrin and cyfluthrin. The result also showed that selection with dieldrin caused negative and positive cross-resistance to pyrethroid and fipronil, respectively. Based on these results, it can be concluded that besides metabolic resistance mechanisms, other factors such as mutation in γ aminobutyric acid (GABA) and voltage-gated sodium channels (Kdr) might be involved.  相似文献

BACKGROUND: Information on the insecticide susceptibility of striped stem borer, Chilo suppressalis (Walker), is essential for an effective pest management programme. An early detection of resistance development can prompt the modification of current control methods and increase the lifespan of insecticides through the rotation of chemicals with different modes of action. In this study, the susceptibility of this pest in Taiwan to four classes of insecticides has been examined. RESULTS: Over 1000‐fold resistance to carbofuran was detected in C. suppressalis collected from Chiayi and Changhua prefectures, with estimated LC50 values of > 3 mg cm?2. In addition, 61‐fold resistance to cartap was found in the Chiayi population. On the other hand, all tested populations of rice stem borer were still relatively susceptible to chlorpyrifos, fipronil and permethrin, with LC50 values ranging from 30 to 553 ng cm?2. Chilo suppressalis populations collected from the central parts of Taiwan have a higher degree of resistance to the tested insecticides than those from northern areas. CONCLUSION: The occurrence of high resistance to carbofuran in the Chiayi and Changhua areas suggests that this compound should be replaced with chemicals having a different mode of action, such as chlorpyrifos, fipronil and permethrin, to which low cross‐resistance has been detected. The frequency of resistance of eight strains of house flies, Musca domestica L., collected from caged‐layer poultry facilities across New York state, to nine insecticides (dimethoate, tetrachlorvinphos, permethrin, cyfluthrin, pyrethrins, methomyl, fipronil, spinosad and cyromazine) was measured relative to a laboratory susceptible strain. Percentage survival was evaluated at five diagnostic concentrations: susceptible strain LC99, 3 × LC99, 10 × LC99, 30 × LC99 and 100 × LC99. The highest levels of resistance were noted for tetrachlorvinphos, permethrin and cyfluthrin. There was substantial variation in the levels of resistance to the different insecticides from one facility to another, independent of their geographical location. There was very little cross‐resistance detected in these populations to either fipronil or spinosad. Overall, there was a good correlation between insecticide use histories and the levels of resistance. The apparent isolation of fly populations within poultry facilities suggests that there are good opportunities for the implementation of successful resistance management strategies at these facilities. BACKGROUND: The housefly, Musca domestica L., continues to be a major pest of confined livestock operations. Houseflies have developed resistance to most chemical classes, and new chemistries for use in animal agriculture are increasingly slow to emerge. Five adult housefly strains from four Florida dairy farms were evaluated for resistance to four insecticides (beta‐cyfluthrin, permethrin, imidacloprid and nithiazine). RESULTS: Significant levels of tolerance were found in most field strains to all insecticides, and in some cases substantial resistance was apparent (as deduced from comparison with prior published results). At the LC90 level, greater than 20‐fold resistance was found in two of the fly strains for permethrin and one fly strain for imidacloprid. Beta‐cyfluthrin LC90 resistance ratios exceeded tenfold resistance in three fly strains. The relatively underutilized insecticide nithiazine had the lowest resistance ratios; however, fourfold LC90 resistance was observed in one southern Florida fly strain. Farm insecticide use and its impact on resistance selection in Florida housefly populations are discussed. CONCLUSION: Housefly resistance to pyrethroids is widespread in Florida. Imidacloprid resistance is emerging, and tolerance was observed to both imidacloprid and nithiazine. If these insecticides are to retain efficacy, producer use must be restrained. Most of the studies on insecticide impregnated bednets efficacy in malaria control have been undertaken in areas where mosquitoes are pyrethroid susceptible. The efficacy of pyrethroid-treated bednets was not compromised even when mosquitoes were kdr resistant. Here, we evaluate a case in which mosquitoes have kdr-like pyrethroid resistance coupled with metabolic mechanisms. Metabolic and kdr-resistance mechanisms in Anopheles stephensi were characterised in our previous study and this easily colonised species was used as a model to examine the efficacy of pyrethroid-treated bednets in the laboratory. Bioassays performed on adults of susceptible (Beech) and resistant (DUB-S) strains using WHO 0.75% permethrin-impregnated papers showed a resistance ratio of 9.75. The recovery rate of the mosquitoes of the DUB-S strain was significantly higher than that of the susceptible strain Beech. The overall permethrin metabolism rates by DUB-S, measured by HPLC method, were 1.5-fold more than by Beech strain. Bioassays performed on DUB-S mosquitoes using different pyrethroid-treated bednets showed that only deltamethrin at 25 mg/m2 and α-cypermethrin at 40 mg/m2 produced adequate mortality rates. Four other pyrethroids, including permethrin, were ineffective. The deterrency test performed on susceptible and resistant An. stephensi showed that there are significant differences between the entry rates of susceptible and resistant mosquitoes into the exposure tube containing permethrin-treated bednet. These data show that when mosquitoes have both kdr-type and metabolic resistance mechanisms, the efficacy of pyrethroid-treated bednets is questionable.  相似文献

BACKGROUND: Application of insecticides in modern agriculture in order to enhance legume production has led to their accumulation in soils to levels that adversely affect soil microflora such as rhizobia and exert a negative impact on the physiological activities associated with them. This study was therefore designed to identify rhizobial strains expressing higher tolerance to insecticides fipronil and pyriproxyfen and synthesising plant growth regulators even amid insecticide stress. RESULTS: The fipronil‐ and pyriproxyfen‐tolerant Rhizobium sp. strain MRL3 produced plant‐growth‐promoting substances in substantial amounts, both in the presence and in the absence of the insecticides. In general, both insecticides at recommended and higher rates reduced plant dry biomass, symbiotic properties, nutrient uptake and seed yield of lentil plants. Interestingly, when applied with any concentration of the two insecticides, Rhizobium sp. strain MRL3 significantly increased the measured parameters compared with plants grown in soils treated solely with the same concentration of each insecticide but without inoculant. CONCLUSION: This study suggests that Rhizobium strain MRL3 may be exploited as a bioinoculant to augment the efficiency of lentil exposed to insecticide‐stressed soils. A German cockroach (Blatella germanica (L)) strain, Apyr‐R, was collected from Opelika, Alabama after control failures with pyrethroid insecticides. Levels of resistance to permethrin and deltamethrin in Apyr‐R (97‐ and 480‐fold, respectively, compared with a susceptible strain, ACY) were partially or mostly suppressed by piperonyl butoxide (PBO) and S,S,S,‐tributylphosphorotrithioate (DEF), suggesting that P450 monooxygenases and hydrolases are involved in resistance to these two pyrethroids in Apyr‐R. However, incomplete suppression of pyrethroid resistance with PBO and DEF implies that one or more additional mechanisms are involved in resistance. Injection, compared with topical application, resulted in 43‐ and 48‐fold increases in toxicity of permethrin in ACY and Apyr‐R, respectively. Similarly, injection increased the toxicity of deltamethrin 27‐fold in ACY and 28‐fold in Apyr‐R. These data indicate that cuticular penetration is one of the obstacles for the effectiveness of pyrethroids against German cockroaches. However, injection did not change the levels of resistance to either permethrin or deltamethrin, suggesting that a decrease in the rate of cuticular penetration may not play an important role in pyrethroid resistance in Apyr‐R. Apyr‐R showed cross‐resistance to imidacloprid, with a resistance ratio of 10. PBO treatment resulted in no significant change in the toxicity of imidacloprid, implying that P450 monooxygenase‐mediated detoxication is not the mechanism responsible for cross‐resistance. Apyr‐R showed no cross‐resistance to spinosad, although spinosad had relatively low toxicity to German cockroaches compared with other insecticides tested in this study. This result further confirmed that the mode of action of spinosad to insects is unique. Fipronil, a relatively new insecticide, was highly toxic to German cockroaches, and the multi‐resistance mechanisms in Apyr‐R did not confer significant cross‐resistance to this compound. BACKGROUND: Insecticide resistance is a likely cause of field control failures of Tuta absoluta, but the subject has been little studied. Therefore, resistance to ten insecticides was surveyed in seven representative field populations of this species. The likelihood of control failures was assessed, as well as weather influence and the spatial dependence of insecticide resistance. RESULTS: No resistance or only low resistance levels were observed for pyrethroids (bifenthrin and permethrin), abamectin, spinosad, Bacillus thuringiensis and the mixture deltamethrin + triazophos (<12.5‐fold). In contrast, indoxacarb exhibited moderate levels of resistance (up to 27.5‐fold), and chitin synthesis inhibitors exhibited moderate to high levels of resistance (up to 222.3‐fold). Evidence of control failures was obtained for bifenthrin, permethrin, diflubenzuron, teflubenzuron, triflumuron and B. thuringiensis. Weather conditions favour resistance to some insecticides, and spatial dependence was observed only for bifenthrin and permethrin. CONCLUSION: Insecticide resistance in field populations of the tomato pinworm prevails for the insecticides nowadays most frequently used against them—the chitin synthesis inhibitors (diflubenzuron, triflumuron and teflubenzuron). Local selection favoured by weather conditions and dispersal seem important for pyrethroid resistance evolution among Brazilian populations of T. absoluta and should be considered in designing pest management programmes. BACKGROUND: Glutathione S‐transferases (GSTs) have received considerable attention in insects for their roles in insecticide resistance. Laodelphax striatellus (Fallén) is a serious rice pest. L. striatellus outbreaks occur frequently throughout eastern Asia. A key problem in controlling this pest is its rapid adaptation to numerous insecticides. In this research, nine cDNAs encoding GSTs in L. striatellus were cloned and characterised. RESULTS: The cloned GSTs of L. striatellus belonged to six cytosolic classes and a microsomal subgroup. Exposure to sublethal concentrations of each of the six insecticides, DDT, chlorpyrifos, fipronil, imidacloprid, buprofezin and beta‐cypermethrin, quickly induced (6 h) up‐expression of LsGSTe1. The expression of LsGSTs2 was increased by chlorpyrifos, fipronil and beta‐cypermethrin. Furthermore, exposure of L. striatellus to fipronil, imidacloprid, buprofezin and beta‐cypermethrin increased the expression of the LsGSTm gene after 24 or 48 h. CONCLUSION: This work is the first identification of GST genes from different GST groups in Auchenorrhyncha species and their induction characteristics with insecticide types and time. The elevated expression of GST genes induced by insecticides might be related to the enhanced tolerance of this insect to insecticides and xenobiotics. Abstract

The leafminer Scrobipalpula absoluta Meyr. is a serious pest of tomatoes in the Los Valles region of the eastern foothills of the Bolivian Andes. An initial insecticide screening trial showed that the local population of S. absoluta had developed a high resistance to organophosphorous insecticides. Soil‐applied carbamates and Bacillus thuringiensis Berliner were also ineffective. Further trials were carried out to establish the optimum rates and timing of applications of two synthetic pyrethroids, fenvalerate and permethrin. The results showed that organophosphate‐resistant S. absoluta can be satisfactorily controlled under commercial growing conditions with tow doses of pyrethroid insecticides. Permethrin had greater toxicity and persistence than fenvalerate, and could be applied at longer intervals. It is recommended that permethrin, either as Ambush 50 e.c. at 100 ml/ha or as Pounce 30 e.c. at 170 ml/ha, be applied weekly or at 10–14‐day intervals at the beginning of the season or when infestations and damage are light.  相似文献

Fipronil is a relatively new insecticide with great potential for insect control, however widespread use of cyclodiene insecticides has selected for an A302S mutation in the Rdl (GABA gated chloride channel) allele. This mutation gives resistance to cyclodienes and limited cross-resistance to fipronil. Given the concern over the possible reduction in efficacy and/or lifetime that fipronil might be used for pest control (given the extensive use of cyclodienes in the past), it is important to know the frequency of the A302S Rdl mutation in field populations. To ascertain the relative frequency of the A302S Rdl mutation in house fly populations we used three experimental approaches. First, we attempted to select for fipronil resistance by initially treating 33,100 field collected flies and selecting 14 additional generations. We were unable to produce a highly resistant strain. Second, we directly sequenced field collected flies. Third, we tested field collected house flies with a diagnostic dose of dieldrin and then genotyped the survivors. Out of the 4750 flies tested, there were no Rdl resistance alleles detected. We conclude that the resistant Rdl allele is rare in house flies in the US due to decades without cyclodiene use and a fitness disadvantage (in the absence of cyclodienes) of the 302S Rdl allele. The limited cross-resistance provided by the cyclodiene resistant Rdl allele, combined with the very low frequency of this allele in field populations, suggests that fipronil could be a promising insecticide for house fly control.  相似文献

BACKGROUND: In recent years, outbreaks of the brown planthopper, Nilaparvata lugens (Stål), have occurred more frequently in China. The objective of this study was to determine the susceptibility of N. lugens to neonicotinoids and other insecticides in major rice production areas in China. RESULTS: Results indicated that substantial variations in the susceptibility to different insecticides existed in N. lugens. Field populations had developed variable resistance levels to neonicotinoids, with a high resistance level to imidacloprid (RR: 135.3–301.3‐fold), a medium resistance level to imidaclothiz (RR: 35–41.2‐fold), a low resistance level to thiamethoxam (up to 9.9‐fold) and no resistance to dinotefuran, nitenpyram and thiacloprid (RR < 3‐fold). Further examinations indicated that a field population had developed medium resistance level to fipronil (up to 10.5‐fold), and some field populations had evolved a low resistance level to buprofezin. In addition, N. lugens had been able to develop 1424‐fold resistance to imidacloprid in the laboratory after the insect was selected with imidacloprid for 26 generations. CONCLUSION: Long‐term use of imidacloprid in a wide range of rice‐growing areas might be associated with high levels of resistance in N. lugens. A residual bioassay was used to determine insecticide susceptibility in populations of Colarado potato beetle, Leptinotarsa decemlineata (Say), in eastern North Carolina. Mean bioassay mortalities were subjected to correlation analysis to assess patterns of resistance among tested insecticides. Within insecticide classes, cross-resistance patterns were observed between carbaryl and carbofuran (carbamates), fenvalerate and permethrin (pyrethroids), and among azinphosmethyl, methamidophos, and phosmet (organophosphates). Significant associations between insecticides from different insecticide classes were concluded to be due to multiple resistance. Knowledge of multiple and cross-resistance patterns are discussed in relation to the bioassay as a tool for monitoring insecticide resistance.  相似文献

Synergists were used to diagnose possible mechanisms of permethrin resistance in permethrin-selected strains of the tobacco budworm, Heliothis virescens (F.). In addition to permethrin, these strains of the tobacco budworm were resistant to α-cyano-pyrethroid insecticides, organophosphorus insecticides and DDT. The monooxygenase-inhibiting prop-2-ynyl aryl ethers were the only effective synergists of permethrin among 16 candidates tested. The most effective synergist was 1,2,4-trichloro-3-(2-propynyloxy)benzene. Piperonyl butoxide, a common monooxygenase-inhibiting synergist in other species and tobacco budworm strains, was inactive. These results suggested the presence and contribution of an unusual monooxygenase in the enzymatic detoxication of permethrin. DDT cross-resistance, which was not synergized, and broad pyrethroid cross-resistance supported previous evidence for target site insensitivity as a second pyrethroid-resistance mechanism in these strains. The actions of S,S,S-tributyl phosphorotrithioate (TBPT) and triphenyl phosphate (TPP) suggested that hydrolytic detoxication, important in methyl parathion-resistance tobacco budworm strains, had little or no role in conferring pyrethroid resistance in these strains.  相似文献

BACKGROUND: Imidacloprid has been a major neonicotinoid insecticide for controlling Aphis gossypii (Glover) (Homoptera: Aphididae) and other piercing–sucking pests. However, the resistance to imidacloprid has been recorded in many target insects. At the same time, cross‐resistance of imidacloprid and other insecticides, especially neonicotinoid insecticides, has been detected. RESULTS: Results showed that the level of cross‐resistance was different between imidacloprid and tested neonicotinoid insecticides (no cross‐resistance: dinotefuran, thiamethoxam and clothianidin; a 3.68–5.79‐fold cross‐resistance: acetamiprid, nitenpyram and thiacloprid). In the study of sublethal effects, imidacloprid at LC20 doses could suppress weight gain and honeydew excretion, but showed no significant effects on longevity and fecundity of the imidacloprid‐resistant cotton aphid, A. gossypii. However, other neonicotinoid insecticides showed significant adverse effects on biological characteristics (body weight, honeydew excretion, longevity and fecundity) in the order of dinotefuran > thiamethoxam and clothianidin > nitenpyram > thiacloprid and acetamiprid. CONCLUSION: The results indicated that dinotefuran is the most effective insecticide for use against imidacloprid‐resistant A. gossypii. To avoid further resistance development, the use of nitenpyram, acetamiprid and thiacloprid should be avoided on imidacloprid‐resistant populations of A. gossypii. BACKGROUND: In 2003 the development of insecticide resistance against neonicotinoids in the brown planthopper (BPH), Nilaparvata lugens (Stål) (Homoptera: Delphacidae), was first observed in Thailand and has since been found in other Asian countries such as Vietnam, China and Japan. However, the LD50 values of BPH and the whitebacked planthopper (WBPH), Sogatella furcifera (Horváth), against both neonicotinoid and phenylpyrazole insecticides have been poorly reported in many Asian countries. RESULTS: The topical LD50 values for imidacloprid in the BPH populations collected from East Asia (Japan, China, Taiwan) and Vietnam in 2006 were 4.3–24.2 µg g?1 and were significantly higher than those collected from the Philippines (0.18–0.35 µg g?1). The BPH populations indicated a positive cross‐resistance between imidacloprid and thiamethoxam. Almost all the WBPH populations from Japan, Taiwan, China, Vietnam and the Philippines had extremely large LD50 values (19.7–239 µg g?1 or more) for fipronil, except for several populations from the Philippines and China. CONCLUSION: Species‐specific changes in insecticide susceptibility were found in Asian rice planthoppers (i.e. BPH for imidacloprid and WBPH for fipronil). Insecticide resistance in BPH against imidacloprid occurred in East Asia and Indochina, but not in the Philippines. BACKGROUND: Fufenozide is a novel non‐steroidal ecdysone agonist with good efficacy against diamondback moth (DBM), Plutella xylostella (Lepidoptera: Plutellidae). At present, it is widely applied for the control of a range of lepidopterous pests in China. This study compared the activities of fufenozide and 12 other insecticides against unselected and fufenozide‐selected strains of DBM to examine potential patterns of cross‐resistance. The relative fitness of the fufenozide‐selected strain was assessed to provide information pertinent to insecticide resistance management. RESULTS: Compared with the susceptible strain (JSS), the fufenozide‐resistant strain (JSR) showed high cross‐resistance to dibenzoylhydrazines and benzoylphenylureas, low cross‐resistance to abamectin and no cross‐resistance to organophosphates, carbamates and pyrethroids. JSR had a lower reproductive ability and a relative fitness of 0.5 compared with JSS. CONCLUSION: P. xylostella has the potential to develop resistance to fufenozide, albeit at the expense of fitness. Cross‐resistance between the same and other classes of insecticides is of concern, and should be a key consideration when implementing fufenozide‐based control strategies for this species. In many insect species, resistance to cyclodiene insecticides is caused by amino acid substitutions at a single residue (A302) within the M2 transmembrane region of the gamma-aminobutyric acid (GABA) receptor sub-unit termed Rdl (resistance to dieldrin). These mutations (A302S and A302G) have also been shown to confer varying levels of cross-resistance to fipronil, a phenylpyrazole insecticide with a similar mode of action to cyclodienes. To investigate the possible occurrence of these mutations in the cat flea, Ctenocephalides felis (Bouché), a 176-bp fragment of the cat flea Rdl gene, encompassing the mutation site, was PCR amplified and sequenced from nine laboratory flea strains. The A302S mutation was found in eight of the nine strains analysed, although the relative frequency of the mutant allele varied between strains. Only one strain (R6) was found to be homozygous for the S302 allele in all the individuals tested, and this correlated with previous reports of low-level fipronil resistance in this strain. A PCR-based diagnostic assay, capable of screening individual fleas for this mutation, was developed and used to survey a range of fleas collected at random from veterinary clinics in the UK and USA. The A302S mutation was present at a high frequency in these domestic pet populations.  相似文献
