A killed gI-negative vaccine combined with a gI enzyme-linked immunosorbent assay (ELISA) test was used for the first time in Sweden in an attempt to eradicate Aujeszky's disease from a weaner pig producing herd. The herd had experienced three severe outbreaks of the disease during a 10 year period and at the start of the programme 96 per cent of the herd's 104 breeding animals were seropositive to the Aujeszky's virus. In addition, there was serological evidence of active virus circulation among younger animals. During the programme, all breeding animals were vaccinated every sixth month and replacement animals were tested free of disease and vaccinated before entry into the herd. When the originally seropositive animals had been rotated out of the herd, all breeding animals and a sample of weaner pigs were tested twice at six weeks' interval. No seroconversions to gI had taken place and the herd was declared Aujeszky's disease-free, 22 months after the start of the programme.  相似文献

The production of antibodies in pigs to 11 herpesviruses was investigated in relation to their ability to cross-react with Aujeszky's disease virus (suid herpesvirus 1--SHV1). Of the herpesviruses tested only two, sheep herpesvirus (caprine herpesvirus 1) and dog herpesvirus (canid herpesvirus 1), failed to produce homologous virus antibodies. Only the antibodies to bovine herpesvirus 1 (BHV1) produced a cross-reaction by SHV1 enzyme-linked immunosorbent assay (ELISA). No SHV1 neutralizing antibodies were detected in any of the herpesvirus antisera. A cross-reaction with SHV1 by a serum from a pig naturally infected with BHV1 or with any of the other herpesviruses tested was considered unlikely.  相似文献

The Aujeszky's disease virus (ADV) eradication campaign in The Netherlands is based on compulsory vaccination and certification of ADV-free herds and consists of three stages: reduction of transmission of ADV, tracing and elimination of remaining sources of ADV, and, in the last stage prohibition of vaccination. This paper summarizes the progress made in the first two stages of the eradication. Between 1994-1999, ADV seroprevalence decreased sharply in all four regions in The Netherlands, both in the sow and the finishing pig population. In the sow population, the mean ADV prevalence in the northern, eastern and western regions decreased to less than 0.5% in the first quarter of 1998, and to approximately 1% in the southern region. For the finishing pig population, the mean ADV prevalence decreased to less than 0.6% in all four regions in the first quarter of 1998. The proportion of sow herds that were certified ADV-free increased steadily from approximately 40% in the last quarter of 1996 to 96% in the first quarter of 1999. The proportion of finishing pig herds that were certified ADV-free showed the same spectacular increase as in the sow population: from approximately 14% in the last quarter of 1996 to 90% in the first quarter of 1999. It is anticipated that the last stage of the ADV eradication campaign, stopping vaccination, will be achieved very soon. Monitoring wild boars for ADV between 1994 and 1999 indicated that ADV is uncommon within the wild boar population. Therefore, it seems that ADV infection in the wild boar population is not an important reservoir in The Netherlands.  相似文献

Rapid diagnosis of Aujeszky's disease in pigs by immunofluorescence   总被引:2,自引:0,他引:2  
Direct immunofluorescence on impression smears of brain and pharynx was compared with virus isolation in cell culture for the diagnosis of Aujeszky's disease in experimentally and naturally infected pigs. Pharyngeal impression smears were more sensitive than virus isolation in two pigs killed 10 and 12 days after experimental infection. Both methods were of similar sensitivity in the detection of virus from field cases of disease. Smears of brain and pharynx were more sensitive than virus isolation for tissue which had been stored at room temperature (approximately 20 degrees C) for up to 48 hours. Some reduction in the amounts of virus recovered from tissues and the intensity of fluorescent staining occurred in these samples.  相似文献

This work describes the evaluation of two commercial ELISA kits for the detection of gI antibodies against Aujeszky's disease. A collection of experimental sera from infected pigs, field sample sera, and sera from pigs vaccinated with seven different modified gI-negative commercial vaccines were used to evaluate each test. Both ELISA kits showed good reproducibility, and specificity, but differences could be appreciated in sensitivity when sera obtained at early stages of infection was analysed. These results also indicated that both kits could be used in conjunction with the seven vaccines evaluated in this study.  相似文献

Samples of blood and blood serums of pigs were examined for the presence of antibodies to the Aujeszky's disease virus. The virus-neutralizing (VN) test and the enzymoimmunologic (ELISA) method were used for this examination. As indicated by comparison of the average titres of antiviral antibodies determined by both methods, the ELISA method is 60 to 600 times more sensitive than the VN test. The high sensitivity of the ELISA method enabled to detect antiviral antibodies even in samples considered as negative after VN-testing. The method has been used with success for the sanitation of three swine stocks where the Aujeszky's disease was eradicated without interruption of operation.  相似文献

The serum neutralisation test (SNT) and the indirect enzyme-linked immunosorbent assay (ELISA) for Aujeszky's disease were compared, utilising 3202 sera from Aujeszky's disease free pig herds, and 304 SNT reactor and 245 non-reactor sera from Aujeszky's disease infected piggeries. ELISA was found to give good discrimination between positive and negative sera, results showing 96.9% and 99.7% agreement with the SNT in classifying positive and negative reactor pigs respectively. The ELISA appeared to detect a slightly higher proportion of reactors than did the SNT. Absorbance values obtained with ELISA showed a high degree of overall correlation with SW titres (r = 0.916), though correlations were lower when applied to individual sera. The ELISA was considered to be a rapid and convenient procedure, offering many advantages over the SNT for routine use.  相似文献

In the present report, the extent of the reduction in Aujeszky's disease virus (ADV) dissemination achieved when pigs were intensively vaccinated with gI-deleted vaccines under field circumstances, was examined. On widely dispersed breeding-fattening farms, a gI-negative status was most rapidly obtained and the rate of new waves of infections was lowest when the attenuated Bartha strain was administered to both the sows and the fatteners. It was more difficult not only to reach but also to keep a gI-negative status on farms on which the sows were vaccinated with an inactivated vaccine and the fatteners with the attenuated Bartha strain or when the fattening pigs were not vaccinated at all. In a densely populated area, 9 of the 17 farms had gI-positive fatteners at the start of the intensive vaccination programme in which the attenuated Bartha strain was given to both the sows and the fatteners. Antibodies were not detected in the sera of the fatteners of each farm at some time during the experiments, but the fatteners on 7 of the 18 farms still showed antibodies against gI after 20 months of vaccination. At the end of the experiment, the percentage of fatteners with antibodies on these farms was markedly reduced compared with the percentage at the start of the experiment. Therefore, elimination of field virus may be feasible if intensive vaccination is carried out over a sufficiently long period of time. However, the high rate of reinfections experienced either due to reintroduction of the virus or to recrudescence should be a warning against too much optimism, particularly in regions with a dense swine population.  相似文献

A non-suppurative encephalitis accompanied by intraneuronal intranuclear inclusions were observed in the brain from a cow that died within 10 hours of developing nervous signs. Immunogold-silver staining located Aujeszky's disease virus antigen in neuronal cytoplasm and the virus was isolated from large volumes of suspensions of nervous tissues and tonsils. Fattening pigs in adjacent buildings had high antibody titres to Aujeszky's disease virus. The methods by which the cow could have acquired infection are considered, and the significance of transient low titres of antibodies to Aujeszky's disease virus in in-contact cows is discussed.  相似文献

Five of eight bears died during an outbreak of Aujeszky's disease in a travelling circus in the north of Spain. The bears had been fed on a diet which included raw pigs' heads. One of three Himalayan bears and a Kodiak bear died acutely with signs of the disease. One of four polar bears died acutely without signs, another died with signs of Aujeszky's disease while it was being treated, and a third died with enteritis and disseminated intestinal coagulation some time later without showing signs of the disease. A fourth polar bear recovered from the same gastrointestinal problem without showing signs of the disease. Although one of the two surviving Himalayan bears showed some signs referrable to Aujeszky's disease, the results of tests for neutralising antibodies were negative. Two of the polar bears, the Himalayan bear and the Kodiak bear were examined postmortem and three of them were examined histologically. No lesions referable to Aujeszky's disease were found. The tissues from one female polar bear were examined and shown to be positive for Aujeszky's disease virus by virus isolation, polymerase chain reaction, electron microscopy and fluorescent antibody tests. The DNA of the isolate was shown to be similar to that of the strains of the virus circulating in pigs in northern Spain some years earlier.  相似文献
