Sesame, Sesamum indicum L. is used worldwide for cooking oil and food. The United States Department of Agriculture, Agricultural Research Service, Plant Genetic Resources Conservation Unit (USDA, ARS, PGRCU) conserves 1,226 accessions originating from Europe, Africa, Asia, North America, and South America. Sesame accessions were directly seeded to the field in Griffin, GA between 15 May and 01 June 2003–2007. At 50% maturity, 192 accessions were characterized for morphological, phenological, and reproductive traits during the regeneration cycles. High quality plants regenerated from all accessions produced 131 to more than 80,000 seeds per accession. Sesame can be successfully grown and regenerated in Griffin, GA. Coefficients of variation and principal component analysis revealed considerable variability among accessions for phenological, morphological and reproductive traits. Assuming appropriate levels of heritability, sufficient variation among sesame accessions exists for days to maturity, plant height, and reproductive traits among these sesame accessions to warrant breeding programs for new sesame cultivar development.  相似文献

Guar, Cyamopsis tetragonoloba is used worldwide for food stabilization, fiber source, food, and industry. The United States Department of Agriculture, Agricultural Research Service, Plant Genetic Resources Conservation Unit (USDA, ARS, PGRCU) conserves 1,298 accessions originating from India, Pakistan, and breeding lines from the USA. My objectives were to determine: (1) if these guar accessions can successfully be regenerated in Georgia, USA, (2) which traits contribute the most variation, and (3) to provide estimates for genetic relatedness among these accessions. Guar accessions were directly seeded to the field in Griffin, GA between early May and mid June 2006–2008. At 50% maturity, 73 accessions were characterized for morphological, phenological, and reproductive traits during the regeneration cycles. High quality plants regenerated from most of the accessions and produced 80 to more than 9,300 seeds per accession. Guar can be successfully grown and regenerated in Griffin, GA. Coefficients of variation and principal component analysis revealed variability among accessions for these traits evaluated. Cluster analysis separated guar accessions into three groups (clusters) based on low, intermediate or high seed numbers. Guar accessions clustered in groups 2 and 3 appear to be more genetically related than those in group 1.  相似文献

Wild species of safflower, Carthamus oxyacanthus Bieb., is highly crossable with cultivated species, C. tinctorius L. and could be directly exploited in broadening safflower gene pool and improving the crop for biotic and abiotic stress environments. In this study, genetic diversity among accessions of C. oxyacanthus and their relationships with cultivated safflower were evaluated using agro-morphological traits and polymorphic inter-simple sequence repeats (ISSR) markers. Significant variation was observed among accessions particularly for seeds per capitulum, seed yield per plant, harvest index and capitula per plant. Cluster analysis based on agro-morphological traits classified the wild accessions in two groups according to their geographical regions, and separated them from the cultivated genotypes. ISSR marker also revealed a high genetic variation among the accessions, and cluster analysis based on this marker divided genotypes into four groups, with cultivated ones in a separate clade. Genetic variation observed among the wild safflower germplasm at the DNA level was higher than the agro-morphological traits, indicating that ISSR is an effective marker system for detecting diversity among safflower genotypes and their genetic relationships. Accessions of C. oxyacanthus with high genetic relationship to cultivated species could be used for interspecific hybridization in breeding programs of safflower.  相似文献

Calaverita an endangered Mexican orchid is one of the most elegant and showy orchids of the genus Laelia and essential symbol within the local traditional celebrations of the Day of the Dead, in the region of Chilapa-Guerrero. We investigated morphological variation in flowers of la calaverita (Laelia anceps Lind. subsp. dawsonii (J. Anderson) Rolfe f. chilapensis Soto-Arenas, Orchidaceae) using the labellum of the specimens cultivated in traditional home gardens in the region of Chilapa, Guerrero, Mexico. The use of morphometric traits of the labellum is useful in appraising infra-subspecific morphological variation within the chilapensis form. Floral morphological variation exists among the specimens examined. These specimens, found exclusively in traditional home gardens, were grouped into four morphotypes: NAT, Topiltepec, Xulchuchío, and Terrero. The study of variation of la calaverita revealed the existence of four local morphotypes that originated from a long and complex process of selection of traits which responds to traditional preferences and knowledge about specific biotic and agroecological conditions. The cultural and aesthetic influence of traditional homegarden owners in the Chilapa region acts directly on selection, use and conservation of the morphological traits of la calaverita.  相似文献

One hundred ninety four accessions of barnyard millet collected from different eco-geographical regions of India were evaluated for 14 quantitative traits during kharif 2003 and 2005. These accessions were grouped into 5 groups on the basis of place of collection. These groups differed significantly in their mean values for quantitative traits and magnitude of correlation among traits indicates origin based association. The group ‘C’ (source of origin is unknown) was found most diverse group (mean coefficient of variation 17.67%), while rest of the groups recorded mean coefficient of variation between 12 and 13%. Correlation studies indicate that flag leaf width, number of racemes along with internode length should be considered while performing selection in segregating generations. The plotting of first and second principal component axes scores suggests that presence of three distinct morphotypes in the present study. These morphotypes are quite similar to three botanical varieties viz., var. robusta, var. intermedia and var. stolonifera in their morphological traits, while botanical variety laxa (endemic to Sikkim area of India) was completely absent from the present study.  相似文献

The genus Arachis is divided into nine taxonomic sections. Section Arachis is composed of annual and perennial species, while section Heteranthae has only annual species. The objective of this study was to investigate the genetic relationships among 15 Brazilian annual accessions from Arachis and Heteranthae using RAPD markers. Twenty-seven primers were tested, of which nine produced unique fingerprintings for all the accessions studied. A total of 88 polymorphic fragments were scored and the number of fragments per primer varied from 6 to 17 with a mean of 9.8. Two specific markers were identified for species with 2n = 18 chromosomes. The phenogram derived from the RAPD data corroborated the morphological classification. The bootstrap analysis divided the genotypes into two significant clusters. The first cluster contained all the section Arachis species, and the accessions within it were grouped based upon the presence or absence of the ‘A’ pair and the number of chromosomes. The second cluster grouped all accessions belonging to section Heteranthae.  相似文献

Amorphophallus paeoniifolius (Dennst.) Nicolson is a tuberous herb occurring in the wild and cultivated state. Vegetative morphological characters were studied at full foliage stage in 17 accessions of A. paeoniifolius which include two cultivars, 15 wild accessions and a related species, A. dubius Blume. The first principal component (PC) accounted for 42.32% of phenotypic variance followed by second for another 18.38%. Major traits that accounted for more variability in both PC1 and PC2 include offset shape, cormel weight per corm, corm fresh weight, petiole surface pattern and canopy spread. The unweighted pair- group method with mathematical averaging (UPGMA) clustering method revealed three principal clusters which separated all the accessions between Euclidean distances of 0.3–1.6. Both cluster analysis and principal co-ordinate analysis revealed T10, a morphotype of A. paeoniifolius var. campanulatus (Decne.) Sivad. cultivated in Tamil Nadu and P19 accession of A. paeoniifolius var. paeoniifolius from Karnataka as morphologically distinct, which needs further validation on the basis of floral characters or molecular markers and G3 from Gujarat as an immediate ancestor of cultivated elephant foot yam. The genotypic (GCV) and phenotypic (PCV) coefficients of variation, broad sense heritability (h2B) and genetic advance (GA) as a percent of the mean assessed for 18 accessions revealed high heritability estimates. A highly significant (P < 0.01) correlation coefficient between circumference of petiole base and corm diameter, corm height, corm weight, east west spread and north south spread suggests that circumference of petiole and canopy spread are indicators to corm weight and size.  相似文献

The characterization of plant genetic resources is the first step towards improving their use. The Spanish Plant Genetic Resources Centre, which belongs to the National Institute for Agricultural and Food Technology Research (CRF-INIA), conserves accessions of wild underground vetches collected in Spain. In the present work, 26 underground vetch accessions were characterized in terms of their seed storage proteins (separated by SDS-PAGE) as a means of assessing the genetic variation of these plants and their agronomic value. Vicia sativa cv. Vereda was used as control. A total of 54 bands were detected, of which 49 were polymorphic. Fifty eight different electrophoretic patterns were observed in total. Protein bands were scored in terms of their presence (1) or absence (0) for all the seeds studied, and two matrices constructed, one with all the bands present in each accession, the other with the different patterns for each accession. Dendrograms based on the Jaccard similarity index and the UPGMA clustering method were produced from these matrices, and the degree of genetic variation between and within accessions was calculated. The groups obtained were compared with the chromosome number for each accession. The results reflect the great diversity of underground vetch seed storage proteins. The aerial and underground seeds of 16 accessions were then analysed separately. In some cases, the aerial and underground flowers of the same plant produced different proteins.  相似文献

The tomato wild relative species Solanum habrochaites (previously known as Lycopersicon hirsutum) is a potential source of novel genes for tomato breeding. It shows resistance to many diseases and pests, cold tolerance and fruit quality traits. This species inhabits the western Andean slopes at high elevations from central Ecuador to central Peru. In this study the genetic variation of S. habrochaites was studied using 91 accessions from the whole range of distribution of this species. To this end, we employed two kinds of markers: 9 SSRs and 6 AFLP combinations. The Principal Coordinate Analysis based on AFLP data showed the existence of clinal genetic variation from north to south. The accessions of the different geographic groups were sequentially arranged in the first axis from north to south and a clear separation between them was found. The groups from the centre of the area of distribution showed the highest variation and heterozygosis. The ones from the margins showed lower variability and presented higher homozygosis. The morphotypes typicum and glabratum considered by Müller constituted the extreme forms of the continuous variation in the pubescence grade. These differences in pubescence were not associated with the homozygosis grade.  相似文献

Genetic erosion in cultivated wheat provides a good reason for studying genetic diversity in crop wild relatives. In the present study, genetic diversity of 32 accessions belonging to T. boeoticum species collected from different parts of Iran were evaluated using 13 morphological traits as well as ten inter-simple sequence repeat primers. Statistical analysis for morphological traits showed significant differences among accessions (except number of fertile tillers and total tillers per plant). In principal component analysis, the first three PCA showed 82.65 % of the total morphological variation. Based on the morphological traits, accessions were separated into two main groups by cluster analysis. In molecular analysis, polymerase chain reactions amplified 105 DNA fragments, out of which, 95 (90.47 %) were polymorphic. From geographic perspective, the accessions sampled from western and southwestern of Iran showed the highest and lowest polymorphism, respectively. However, the maximum values of effective number of alleles (Ne), Nei’s gene diversity (He) and Shannon’s information index (I) was related to accessions collected from NW regions. Also, according to cluster analysis and PCoA plot genetic diversity was not related to geographical distribution. Overall, our results revealed a remarkable level of genetic diversity among studied Iranian T. boeoticum accessions; especially accessions collected from Kermanshah and Lorestan provinces, which can be of interest for future breeding programs. So, conservation of germplasm of these areas is recommended.  相似文献

Amplified fragment length polymorphism markers were used to assess the genetic relatedness between Dioscorea alata and nine other edible Dioscorea. These species include D. abyssinica Hoch., D. bulbifera L., D. cayenensis-rotundata Lamk. et Poir., D. esculenta Burk., D. nummularia Lam., D. pentaphylla L., D. persimilis Prain. et Burk., D. transversa Br. and D. trifida L. Four successive studies were conducted with emphasis on the genetic relationship within D. alata and among species of the Enantiophyllum section from Vanuatu. Study 1 was carried out to select a set of polymorphic primer pairs using 11 combinations and eight species belonging to five distinct sections. The four most polymorphic primer pairs were used in study 2 among six species of the Enantiophyllum section. Study 3 focussed mainly on the genetic relationship among 83 accessions of D. alata, mostly from Vanuatu (78 acc.) but also from Benin, Guadeloupe, New Caledonia and Vietnam. The ploidy level of 53 accessions was determined and results indicated the presence of tetraploid, hexaploid and octoploid cultivars. Study 4, included 35 accessions of D. alata, D. nummularia and D. transversa and was conducted using two primer pairs to verify the taxonomical identity of the cultivars `langlang', `maro' and `netsar' from Vanuatu. The overall results indicated that each accession can be fingerprinted uniquely with AFLP. D. alata is an heterogeneous species which shares a common genetic background with D. nummularia and `langlang', `maro' and `netsar'. UPGMA cluster analysis revealed the existence of three major groups of genotypes within D. alata, each assembling accessions from distant geographical origins and different ploidy levels. The analysis also revealed that `langlang', `maro' and `netsar' clustered together with the cultivar `wael' (D. transversa) from New Caledonia. Results are discussed in the paper.  相似文献

Variation and heritability of seed mass in the Mesoamerican crop chia, Salvia hispanica L. was studied to examine the feasibility of selection for the trait. Genotypic variation in seed mass in wild/cultivated and domesticated accessions of chia from different origins was assessed. Broadsense heritability of seed mass was estimated using variances associated with parental and F2 generations derived from two crosses and from the response to one cycle of selection. Significant (P . 0.0001) genotypic variation was observed among accessions. Mean seed mass for domesticated accessions (14.84 mg/100 seeds) was greater than that of the wild accessions (11.29 mg/100 seeds) by 31%. The heritability of seed mass was relatively high in chia (0.75), suggesting that this trait is under strong genetic control. This conclusion was supported by a single selection cycle from the F2 to F3 generation that produced a 16% increase in mean seed mass. The realized heritability estimated based on this one cycle of selection also was 0.75. These observations indicate that gains from selection in chia seed mass are possible when mass selection is conducted in early generations. Basic information is thus provided for future breeding efforts in a species for which little or no knowledge of inheritance currently exists.  相似文献
