During 1983 and 1984, ileocecal lymph node specimens were obtained from clinically normal cattle at 76 US Department of Agriculture-inspected cull cattle slaughterhouses in 32 states and Puerto Rico. Mycobacterium paratuberculosis was isolated from the lymph nodes of 119 of 7,540 cattle. The prevalence of bovine paratuberculosis was 1.6% overall, with 2.9% in dairy culls and 0.8% in beef culls. The prevalence for females and males did not appear to differ significantly.  相似文献

A serologic study was undertaken to determine the prevalence of bovine leukemia virus (BLV) infection in dairy and beef cattle in Florida. Using the agar gel immunodiffusion test with a glycoprotein antigen 47.8% of 7,768 dairy cattle and 6.7% of 4,911 beef cattle were found to have antibodies to BLV. The prevalence of BLV antibodies increased significantly (P less than 0.0001) with increasing age. After data were adjusted for age, prevalence of BLV antibodies was significantly associated with dairy breed (P less than 0.05) but not with species (Bos taurus and B indicus) or sex.  相似文献

Mass screening ELISA methods were developed for testing cattle serum for antibodies against 14 common livestock diseases simultaneously. The absorbance values were transformed to a %ELISA (spectrophotometric antibody end point) by a computer interfaced with a microplate reader. A histogram indicating a cutoff point and a report for the veterinarian also was generated. The computer program produced a print-out of the antibody profile for each animal tested, the antibody concentration against each disease, and a histogram (antibody profile) showing the prevalence of each disease in the herd. Serum samples were obtained from 1,953 cattle, including 880 dairy cattle from 10 herds and 1,073 beef cattle from 20 herds. These samples were obtained from June 1988 through June 1989. The highest antibody prevalence was against bluetongue virus. Of the 1,953 cattle tested, 1,223 (63%) were seropositive for bluetongue virus, including 502 (57%) of the dairy cattle and 721 (67%) beef cattle. Other antibody prevalences, in descending order, were: rotavirus (44%), Pasteurella spp (25%), Leptospira spp and Haemophilus spp (22%), Mycoplasma spp (18%), parainfluenza virus (17%), Campylobacter spp (16%), Anaplasma marginale (15%), bovine leukosis virus (13%), Brucella spp (8%), Mycobacterium paratuberculosis (8%), bovine viral diarrhea virus (3%), and infectious bovine rhinotracheitis virus (3%). Major differences in antibody prevalence between dairy and beef cattle were that only 4% of the dairy cattle were seropositive for A marginale, compared with 25% of the beef cattle, and conversely, 29% of the dairy cattle were seropositive for bovine leukosis virus, compared with 1% of the beef cattle.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

OBJECTIVE: To estimate the specificity of an absorbed enzyme-linked immunosorbent assay kit for Johne's disease (JD) when used in mature cattle populations resident in northern Australia. DESIGN: Blood samples were collected from beef cattle in northern Queensland, the Northern Territory and northern Western Australia, and from dairy cattle in northern Queensland. The specificity of a serological test for JD was estimated by testing the blood samples with an absorbed ELISA kit. Further samples were collected from cattle with positive ELISA results to determine the presence or absence of infection with Mycobacterium avium subsp paratuberculosis. PROCEDURE: During 1995 and 1996, blood, tissue and gut contents were collected from beef cattle at abattoirs in Queensland and the Northern Territory; and blood and faecal samples were collected from dairy cattle in herds assessed to be most at risk for JD in northern Queensland. The blood samples were tested using an absorbed ELISA kit. Tissues and gut contents from beef cattle that had positive ELISA results were cultured for M. avium subsp paratuberculosis, and tissues were examined histologically. Faecal samples from dairy cattle with positive ELISA results were cultured for M. avium subsp paratuberculosis. RESULTS: Estimates of specificity for this absorbed ELISA in mature northern Australian cattle were 98.0% (97.0 to 98.8%, 95% CI) in beef cattle, and 98.3% (96.7 to 99.3%, 95% CI) in dairy cattle. CONCLUSION: Estimates of specificity in this study were lower for beef cattle from the Northern Territory and northern Western Australia and for dairy cattle from northern Queensland than those quoted from studies on cattle in southern Western Australia. This should be considered when serological testing using the JD ELISA is carried out on northern Australian cattle.  相似文献

Conclusion Estimates of specificity in this study were lower for beef cattle from the Northern Territory and northern Western Australia and for dairy cattle from northern Queensland than those quoted from studies on cattle in southern Western Australia. Beef and dairy cattle serum samples, collected during 2000 at sale barns throughout Georgia, were obtained from the Georgia State Brucellosis Laboratory and were used to conduct a retrospective epidemiological study. Statistical samplings of 5,307 sera, from over 200,000 sera, were tested for antibodies to Mycobacterium avium ssp. paratuberculosis, (Johne's disease) using a commercial enzyme-linked immunosorbent assay test kit. An overall period seroprevalence in all classes of cattle tested was 4.73%. The period seroprevalence in dairy cattle was 9.58%, in beef cattle it was 3.95%, and in cattle of unknown breed it was 4.72%. It was concluded that the seroprevalence of Johne's disease in cull beef and dairy cattle in Georgia is economically significant.  相似文献

A seroepidemiological survey of bovine leukemia virus (BLV) infection was conducted in Japan in 2007 using an enzyme-linked immunosorbent assay (ELISA) and an agar gel immunodiffusion (AGID) test. A total of 5420 cattle (dairy, 3966; breeding beef, 797; fattening beef, 657) from 209 farms in seven prefectures in Japan were tested. The overall prevalence of BLV infection was 28.6%. The prevalence of BLV infection in dairy cattle (34.7%) was higher than for both fattening beef cattle (7.9%) and breeding beef cattle (16.3%). Age-specific prevalence showed that BLV prevalence increased with age in all types of cattle and was notably different between dairy and beef cattle under 1 year of age. Among 207 farms, 141 herds (68.1%) had one or more positive animals. The proportion of these positive farms was significantly higher among dairy farms (79.1%) than among beef breeding farms (39.5%) and beef fattening farms (51.9%) (P < 0.001). Dairy farms (40.5%) also showed a significantly higher within-herd prevalence than beef breeding (27.4%) and fattening (14.9%) farms (P = 0.001). This study indicated that BLV is more widely spread in dairy cattle than in beef breeding cattle in Japan. Given the prevalence of BLV infection in dairy and beef cattle was 8- and 1.7-fold higher, respectively, than rates previously found in 1980–1982, BLV appears to be spreading particularly among the dairy cattle population during the last two decades. Further investigation is required to determine the risk factors necessary to control BLV infection that take into account the different farming practices that exist between dairy and beef sectors.  相似文献

A simple random survey was conducted in Ireland during 2005 to estimate the ELISA-prevalence of paratuberculosis, commonly called Johne's disease (JD), in the cattle population. Serum samples were collected from all 20,322 females/breeding bulls over 12 months-of-age in 639 herds. All samples were tested using a commercially available absorbed ELISA. The overall prevalence of infected herds, based on the presence of at least one ELISA-positive animal, was 21.4% (95% CI 18.4%-24.9%). Herd prevalence levels amongst dairy herds (mean 31.5%; 95% CI: 24.6%, 39.3%) was higher than among beef herds (mean 17.9%; 95% CI: 14.6%-21.8%). However, the animal level prevalence was similar. The true prevalence among all animals tested, was calculated to be 2.86% (95%CI: 2.76, 2.97) and for animals >= 2 yrs, it was 3.30% (95%CI: 3.17, 3.43). For animals in beef herds, true prevalence was 3.09% (95%CI: 2.93, 3.24), and for those in dairy herds, 2.74% (95%CI: 2.59, 2.90). The majority of herds had only one ELISA-positive infected animal. Only 6.4% (95% CI 4.7%-8.7%) of all herds had more than one ELISA-positive infected animal; 13.3% (CI 8.7%-19.7%) of dairy herds ranging from two to eight ELISA-positive infected animals; and, 3.9% beef herds (CI 2.4%-6.2%) ranging from two to five ELISA-positive infected animals. The true prevalence of herds infected and shedding Mycobacterium avium subspecies paratuberculosis is estimated to be 9.5% for all herd types; 20.6% for dairy herds; and 7.6% for beef herds. If ELISA positive animals <2-years-of-age are excluded, the true herd prevalene reduces to: 9.3% for all herd types; 19.6% for dairy herds; and 6.3% for beef herds based on a test specificity (Sp) of 99.8% and test sensitivity (Se) (i.e., ability to detect culture-positive, infected animals shedding at any level) of 27.8-28.9%.  相似文献

OBJECTIVE: To determine prevalence of paratuberculosis among dairy cattle herds and to identify associated soil-related risk factors. SAMPLE POPULATION: Serum and soil samples for 121 Michigan dairy herds. PROCEDURE: Blood samples were collected from cows at each farm and tested for Mycobacterium paratuberculosis, using an antibody ELISA. Soil samples were collected from pastures and exercise lots; pH and available iron content were determined. A questionnaire was administered to collect data regarding farm management practices and productivity. RESULTS: 55% of the herds tested had > or = 2 M paratuberculosis-positive cattle. Adjusting sample prevalence for distribution of herd size strata yielded a statewide herd prevalence of 54%. Of 3,886 cattle tested, 267 had positive results. Prevalence of test-positive cattle was 6.9%. For every part per million (ppm) increase in soil iron content, there was a 1.4% increase in the risk of a herd being test-positive. An increase in soil pH of 0.1 was associated with a 5% decrease and an increase in soil iron content of 10 ppm was associated with a 4% increase in the number of test-positive cattle. Application of lime to pasture areas was associated with a herd being only 10% as likely to be paratuberculosis positive and with a 72% reduction in number of test-positive cattle. CONCLUSIONS AND CLINICAL RELEVANCE: Prevalence of paratuberculosis-positive dairy herds in Michigan (54%) was greater than expected, but prevalence of paratuberculosis-positive cattle (6.9%) was within anticipated values. These prevalences were associated positively with acidic soil and increased soil iron content. Application of lime to pasture areas was associated with reduced risk of paratuberculosis.  相似文献

Identifying spatial patterns of risk is important in the study of diseases with ecologic causes. Furthermore, relatively complex hierarchical modeling is required to determine how factors that are organized across levels interact, such as how an ecologic cause interacts with farm management and with animal characteristics. The objective of this study was to map the risk for Mycobacterium avium subspecies paratuberculosis (MAP - the causative agent of Johne's disease) and Neospora caninum (NC - the cause of neosporosis) infections in Alberta beef and dairy cattle. This objective utilized Bayesian generalized linear kriging to partition herd effects into a portion attributable to location and a portion that was independent of location. Seropositivity to NC in beef cattle showed strong support for spatial covariance, suggesting that ecologic causes were important for beef cattle but not dairy cattle. There was little evidence of spatial covariance for MAP seropositivity in either beef or dairy cattle.  相似文献

OBJECTIVE: To estimate the prevalence of paratuberculosis in purebred beef cattle in Texas and identify risk factors for seropositivity. DESIGN: Epidemiologic survey. ANIMALS: 4,579 purebred cattle from 115 beef ranches in Texas. PROCEDURE: Blood was collected, and serum was analyzed for antibodies with a commercial ELISA. Fecal samples were collected and frozen at -80 degrees C until results of the ELISA were obtained, and feces from seropositive cattle were submitted for mycobacterial culture. Herd owners completed a survey form on management factors. RESULTS: Results of the ELISA were positive for 137 of the 4,579 (3.0%) cattle, and 50 of the 115 (43.8%) herds had at least 1 seropositive animal. Results of mycobacterial culture were positive for 10 of the 137 (7.3%) seropositive cattle, and 9 of the 50 (18%) seropositive herds had at least 1 animal for which results of mycobacterial culture were positive. Risk factors for seropositivity included water source, use of dairy-type nurse cows, previous clinical signs of paratuberculosis, species of cattle (Bos taurus vs Bos indicus), and location. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that seroprevalence of paratuberculosis among purebred beef cattle in Texas may be greater than seroprevalence among beef cattle in the United States as a whole; however, this difference could be attributable to breed or regional differences in infection rates or interference by cross-reacting organisms. Veterinarians should be aware of risk factors for paratuberculosis as well as the possibility that unexpected serologic results may be found in some herds.  相似文献

A cross-sectional study was performed to determine the odds of having a positive paratuberculosis ELISA result if the dam was ELISA positive in Texas beef cattle, adjusted for individual and herd-level risk factors for seropositivity. Texas beef cattle (n = 2,621) were tested for paratuberculosis by using a commercial ELISA and microbiologic culture of feces for Mycobacterium avium subsp. paratuberculosis (MAP). Pedigree data were collected to identify dam-and sire-offspring pairs. Bayesian mixed-effects logistic regression was used to estimate the odds of seropositivity associated with age, dam ELISA status, sire ELISA status, herd size, herd history of clinical paratuberculosis, within-herd seroprevalence, within-herd fecal MAP prevalence, and within-herd fecal non-MAP Mycobacterium spp. prevalence. Herd of residence was included as a random effect to account for the correlation of observations within the same herd. Statistically probable associations were observed between ELISA status and herd fecal MAP prevalence [OR (odds ratio) 1.28 per 1% increase; P < 0.001] and herd seroprevalence (OR 1.21 per 1% increase; P < 0.001). The association with dam ELISA status was small (OR 1.35) and not highly probable (P = 0.69). Results indicate that use of dam ELISA status to make culling decisions in beef cattle may not improve the success of paratuberculosis control programs. Alternative strategies may be more effective for reducing the odds of seropositivity.  相似文献

OBJECTIVE: To determine prevalence of papillomatous digital dermatitis (PDD) among culled adult dairy and beef cattle in the southeastern United States. ANIMALS: 815 cattle examined during 4 visits to a slaughterhouse. PROCEDURE: The left hind foot of each animal was examined for gross lesions of PDD. Breed and sex of the animals were recorded. Lesions were examined histologically for pathologic changes and bacteria, especially spirochetes. RESULTS: 22 of 76 (29%) dairy cattle and 29 of 739 (4%) beef cattle had gross lesions of PDD. Detection of lesions was not associated with sex of dairy cattle, but male beef cattle were more likely to have lesions of PDD than were female beef cattle. Histologically, acute and chronic lesions were seen; the most severe changes were localized to the stratum corneum. Spirochetes were seen in lesions from 31 of 51 (61%) cattle. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that PDD is common among culled adult cattle. Prevalence was higher in culled adult dairy cattle than in culled adult beef cattle.  相似文献

A serological survey for Neospora caninum and Besnoitia besnoiti was carried out in beef and dairy cattle in South Australia. Serum samples of dairy cattle (n=133) from 9 properties and tank milk samples from a further 122 dairy herds were tested. An additional 810 sera from beef cattle from 51 properties were also tested. Testing at the individual animal level by IDEXX NEOSPORA X2 Ab test ELISA revealed a low prevalence of N. caninum antibodies of only 2.7% (95% CI; 1.6-3.7%) sera positive, as did the milk testing that showed 2.5% (95% CI; 1.4-3.6%) of tank milks being positive. At the herd level, 29.4% (95% CI; 16.9-41.9%) of beef, and 44.4% (95% CI; 12.0-76.9%) of dairy cattle herds showed serum antibodies. The highest within-herd prevalence in beef was 20% and 25%in dairy, which explains the low herd prevalence in dairy detected by bulk milk testing. Testing for B. besnoiti antibodies by PrioCHECK(?) Besnoitia Ab 2.0 ELISA initially identified 18.4% (95% CI: 15.8-21.0%) of 869 individual cattle sera as positive by ELISA at the manufacturer's suggested cut-off threshold (15 PP). Additional tests by immunoblot and IFAT, however, could not confirm any of the ELISA results. The use of a higher (40 PP) threshold in the ELISA is suggested to improve specificity. There is thus no evidence of B. besnoiti infection in South Australian cattle.  相似文献

For evaluation of the prevalence of anti-Neospora caninum antibodies and its associated risk factors, serum samples from 2109 cattle (11 beef, 50 dairy and 25 mixed farms) and 174 dogs were examined in the State of Rond?nia, Western Amazon, Brazil. An inquiry was applied in each farm. Sera were examined by the Indirect Fluorescence Antibody Test (IFAT) using cut off dilution of 1:25 for cattle and 1:50 for dogs. Statistical association between the serologic status and several variables were analyzed by linear and logistic regression. The overall herd prevalence of anti-N. caninum antibodies for 86 farms was 72% (61.3-81.2%). Prevalence values were 100, 70 and 64% in beef, dairy and mixed herds, respectively. Herd prevalence in beef herds was significantly different (P<0.05) from dairy and mixed herds. The overall animal prevalence of N. caninum in cattle was 8.8%. Prevalence values by animal were similar in different production types (P>0.05), with values of 9.5, 11.2 and 9.7% for beef, dairy or mixed cattle, respectively. Antibodies were found in 12.6% of the 174 examined dogs. Sixteen (22.8%) out of 70 farms with dogs had at least one dog with anti-N. caninum antibodies. The occurrence of antibodies in cattle was statistically associated with farms having more than 25 cows (OR 9.7, 95% IC 2.9-32.2; P=0.0002). There was no significant association between the presence of the dogs, jungle contact or reproductive variables with the occurrence of antibodies in cattle.  相似文献

AIM: To estimate the prevalence of Neospora infection in a sample of New Zealand beef cattle. METHODS: The prevalence of Neospora caninum infection in New Zealand beef cattle was estimated by collecting blood at slaughter from 499 beef cattle from 40 different farms at 2 slaughter plants in the North Island and 1 in the lower South Island . Sera were tested using an ELISA against Neospora tachyzoite antigen. RESULTS: The prevalence of seropositive cattle was 2.5% (n=120), 3.6% (n=166) and 2.3% (n=213) at the plants surveyed, the overall prevalence being 2.8%. The serologically positive cattle came from 9 farms, 3 of which had more than 1 positive animal. The highest prevalence recorded amongst animals from 1 farm was 4/13 (31%), in a group of young steers. CONCLUSION: Neosporosis appears to be present at a lower level in the New Zealand beef cattle population than in the New Zealand dairy cattle population. Nevertheless, from the high seroprevalence evident amongst young cattle on 1 farm, we suggest that Neospora may be a cause of infertility in beef cattle in this country.  相似文献

OBJECTIVE: To evaluate the epidemiologic efficacy and economic efficiency of current and potential future control programs for paratuberculosis (Johne's disease) on midsize dairy herds in the United States. DESIGN: Stochastic dynamic computer simulation model. SAMPLE POPULATION: Data on prices and other input variables collected from various sources were used to represent a population of midsize US dairy herds infected with paratuberculosis. PROCEDURE: The simulation model was modified to reflect management and production characteristics of midsize dairy herds in the United States. The model was validated by use of field data and expert opinion. Various control strategies then were simulated and compared on an epidemiologic basis and on the basis of economic efficiency. RESULTS: Test-and-cull strategies and vaccination against paratuberculosis were not able to decrease the mean prevalence of disease in the United States. Typically, only vaccination was economically attractive. Improved management strategies decreased the prevalence of paratuberculosis considerably and had high economic benefits. CONCLUSIONS AND CLINICAL RELEVANCE: Analysis of results of this study suggests that test-and-cull strategies alone do not reduce the prevalence of paratuberculosis in cattle and are costly for producers to pursue. Vaccination did not reduce the prevalence but was economically attractive. Finally, improved calf-hygiene strategies were found to be critically important in every paratuberculosis control program and most were economically attractive programs for midsize US dairy farms with the disease.  相似文献

Serums from 4,394 dairy cattle in 100 herds and from 2,794 beef cattle in 50 herds were tested for antibody to the bovine (C-type) leukemia virus (BLV), using the agar gel immunodiffusion test. Reactors were found in 66% of the dairy herds (10.2% of the cattle) and in 14% of the beef herds (1.2% of the cattle). The prevalence of reactors was examined with respect to age, herd size, and sex. Few of the reactors were less than 2 years old. There was a high percentage of reactors in small dairy herds (less than 50 cattle). In 22 dairy herds (1,354 cows and 96 bulls), the rate of infection in cows was compared with that in bulls. In those herds, 13.5% of the cows and 10.4% of the bulls were reactors.  相似文献

Prior to establishing a control and prevention program for Johne's disease in cattle in Galicia (northwest Spain), a survey was conducted to estimate the prevalence of the disease. For this survey, 61,069 animals of at least 1-year of age from 2735 randomly selected herds were bled and samples analyzed with a commercial ELISA. The estimated true individual-level prevalences – assuming the manufacturer's reported test sensitivity of 48.5% and specificity of 98.9% – were 3.02% in dairy cattle, 1.03% in beef cattle and 2.83% in animals from farms with both dairy and beef cattle. True herd prevalences (with herds declared positive if one or more animals tested positive) were 10.69% for dairy herds, 0% for beef herds and 2.71% for mixed herds. When herds were declared positive if at least two animals tested positive, true herd prevalences were 14.75% for dairy herds, 1.47% for beef herds and 12.01% for mixed herds. Assuming a higher specificity of 99.4%, true individual-level prevalences increased to 4.03% in dairy herds, 2.07% in beef herds and 3.84% in mixed herds. Herd prevalences were 27.77%/18.79%, 2.78%/2.40% and 5.70%/12.24% (using the one/two-animal cut-offs) in dairy, beef and mixed herds, respectively. In conclusion, these results seem to indicate that a small percentage of cows and a rather high percentage of dairy herds in this region are MAP-seropositive.  相似文献
