Establishment of vesicular-arbuscular mycorrhizal fungi in plant roots involves a pre-infection phase of propagule germination, hyphal growth and appressorium formation, followed by growth of the fungus within the root. The effect of soil temperature on the pre-infection stage was examined by counting the numbers of fungal “entry-points” on the main roots of Medicago truncatula and Trifolium subterraneum, grown at soil temperatures of 12°, 16°, 20° and 25°C for periods up to 12 days. Increased root temperature was positively associated with increased numbers of “entry-points”. This effect was more marked between 12° and 16°C than at higher temperatures, as shown by comparing plants at the same stage of development (emergence of spade leaf) and by calculating the results as entry points per cm root.The first root nodules appeared sooner at higher temperatures (20° and 25°), but subsequent development of nodules (measured as nodule number and aggregate volume of nodules per plant, up to 21 days) was best at 16°C for both host Rhizobium combinations in non-sterile and autoclaved soil. There was no evidence that competition between mycorrhizal fungi and Rhizobium for infection sites occurred.A method of obtaining numbers of infective propagules of vesicular-arbuscular mycorrhizal fungi in soil is described.  相似文献

This study is the first report assessing the effect of soil inoculation on the signalling interaction of Bradyrhizobium japonicum, arbuscular mycorrhizal fungi (AMF) and soybean plants throughout the early stages of colonisation that lead to the tripartite symbiosis. In a study using soil disturbance to produce contrasting indigenous AMF treatments, the flavonoids daidzein, genistein and coumestrol were identified as possible signals for regulating the establishment of the tripartite symbiosis. However, it was unclear whether soil disturbance induced changes in flavonoid root accumulation other than through changing the potential for AMF colonization. In this study, soil treatments comprising all possible combinations of AMF and B. japonicum were established to test whether (1) modifications in root flavonoid accumulation depend on the potential for AMF colonization, and (2) synthesis and accumulation of flavonoids in the roots change over time as a function of the early plant-microbial interactions that lead to the tripartite symbiosis. The study was comprised of two phases. First, maize was grown over 3-week periods to promote the development of the AM fungus Glomus clarum. Second, the interaction between soybean, G. clarum and B. japonicum was evaluated at 6, 10, 14 and 40 days after plant emergence. Root colonization by G. clarum had a positive effect on nodulation 14 days after emergence, producing, 30% more nodules which were 40% heavier than those on roots solely inoculated with B. japonicum. The tripartite symbiosis resulted in 23% more N2 being fixed than did the simpler symbiosis between soybean and B. japonicum. The presence of both symbionts changed accumulation of flavonoids in roots. Daidzein and coumestrol increased with plant growth. However, development of the tripartite symbiosis caused a decrease in coumestrol; accumulation of daidzein, the most abundant flavonoid, was reduced in the presence of AMF.  相似文献

Uptake of zinc from solution by uninfected roots, ectomycorrhizas of Pinus radiata, and endomycorrhizas of Araucaria cunninyhamii occurred by a non-metabolic phase and by metabolically mediated absorption. The metabolically mediated absorption of Zn was 1.4–4.5 times greater for ectomycorrhizas than for uninfected roots and 2.6 times greater for endomycorrhizas than for uninfected roots. Part of the increase with ectomycorrhizas was due to their larger size than uninfected roots and when corrected for this, the uptake/cm2 was 1.4–3.0 times greater. The roles of mycorrhizas in uptake of Zn and other trace elements from soil are discussed.  相似文献

Pea plants were grown in γ-irradiated soil in pots with and without addition of the AM fungus Glomus intraradices at sufficient N and limiting P. Depending on the growth phase of the plant presence of AM had negative or positive effect on rhizosphere activity. Before flowering during nutrient acquisition AM decreased rhizosphere respiration and number of protozoa but did not affect bacterial number suggesting top-down regulation of bacterial number by protozoan grazing. In contrast, during flowering and pod formation AM stimulated rhizosphere respiration and the negative effect on protozoa decreased. AM also affected the composition of the rhizosphere bacterial community as revealed from DNA analysis (DGGE). With or without mycorrhiza, rhizosphere respiration was P-limited on very young roots, not nutrient limited at more mature roots and C-limited at withering. This suggests changes in the rhizosphere community during plant growth also supported by changes in the bacteria (DGGE).  相似文献

The effect of the soil yeast, Rhodotorula mucilaginosa LBA, on Glomus mosseae (BEG n°12) and Gigaspora rosea (BEG n°9) was studied in vitro and in greenhouse trials. Hyphal length of G. mosseae and G. rosea spores increased significantly in the presence of R. mucilaginosa. Exudates from R. mucilaginosa stimulated hyphal growth of G. mosseae and G. rosea spores. Increase in hyphal length of G. mosseae coincided with an increase in R. mucilaginosa exudates. No stimulation of G. rosea hyphal growth was detected when 0.3 and 0.5 ml per petri dish of yeast exudates was applied. Percentage root length colonization by G. mosseae in soybean (Glycine max L. Merill) and by G. rosea in red clover (Trifolium pratense L. cv. Huia) was increased only when the soil yeast was inoculated before G. mosseae or G. rosea was introduced. Beneficial effects of R. mucilaginosa on arbuscular mycorrhizal (AM) colonization were found when the soil yeast was inoculated either as a thin agar slice or as a volume of 5 and 10 ml of an aqueous solution. R. mucilaginosa exudates (20 ml per pots) applied to soil increased significantly the percentage of AM colonization of soybean and red clover.  相似文献

The bacterium Wautersia [Ralstonia] basilensis has been shown to enhance the mycorrhizal symbiosis between Suillus granulatus and Pinus thunbergii (Japanese black pine). However, no information is available about this bacterium under field conditions. The objectives of this study were to detect W. basilensis in bulk and mycorhizosphere soils in a Japanese pine plantation in the Tottori Sand Dunes, determine the density of W. basilensis in soil, and determine the optimal cell density of W. basilensis for mycorrhizal formation in pine seedlings. We designed and validated 16S rRNA gene-targeted specific primers for detection and quantification of W. basilensis. SYBR Green I real-time PCR assay was used. A standard curve relating cultured W. basilensis cell density (103-108 cells ml−1) to amplification of DNA showed a strong linear relationship (R = 0.9968). The specificity of the reaction was confirmed by analyzing DNA melting curves and sequencing of the amplicon. The average cell density of W. basilensis was >4.8 × 107 cells g−1 of soil in the mycorrhizosphere and 7.0 × 106 cells g−1 in the bulk soil. We evaluated the W. basilensis cell density required for mycorrhizal formation using an in vitro microcosm with various inoculum densities ranging from 102 to 107 cells g−1 soil (104-109 cells ml−1). Cell densities of W. basilensis of >106 cells g−1 of soil were required to stimulate mycorrhizal formation. In vivo and in vitro experiments showed that W. basilensis was sufficiently abundant to enhance mycorrhizal formation in the mycorrhizosphere of Japanese black pine sampled from the Tottori Sand Dunes.  相似文献

 Plants colonised with the arbuscular mycorrhizal fungi (AMF) Glomus E3 and Glomus tenue were grown in microcosms that permitted separation into root:hyphae and hyphae compartments. Hydrolysed polysaccharides from the hyphae and water-soluble sugars released into the hyphosphere were assayed using chromatography. Total sugars and most monosaccharides were elevated in the hyphosphere of Glomus E3 but not in the hyphosphere of G. tenue. Differences in the levels of sugars did not depend on hyphal surface area. It is suggested the diversity in sugars produced in the hyphosphere of AMF may drive some of the spatial and temporal variation in microbial diversity and function in soils.  相似文献

The effects of bacteria associated with arbuscular mycorrhizal fungal (AMF) spores on spore germination, growth in vitro and on the pea-AMF symbiosis were evaluated. Bacterial colonies were recovered from untreated Glomus clarum NT4 spores and NT4 spores decontaminated with 5% chloramine-T for 30, 45 and 60 min on five different media. Both G+ and G− bacteria were recovered from untreated NT4 spores, whereas only G+ bacteria were isolated from decontaminated spores. An in vitro assessment of the effect of spore-associated bacteria on clean, decontaminated NT4 spores revealed that (i) most of the bacteria isolated from untreated spores generally did not significantly alter spore function, (ii) some bacteria isolated from clean, decontaminated spores inhibited or stimulated NT4 spore germination, (iii) stimulation of spore germination occurred only when bacteria were in contact with spores, and (iv) inhibition of spore germination was the result of volatile bacterial metabolites. A stimulatory bacterial isolate, Bacillus pabuli LA3, significantly (P<0.05) enhanced the shoot growth, AMF-colonization, shoot N content and P use efficiency of NT4-inoculated 6 week-old pea plants over that of plants co-inoculated with an inhibitory bacterial isolate, Bacillus chitinosporus LA6a and NT4.  相似文献

Many legume plants benefit from the tripartite symbiosis of arbuscular mycorrhizal fungi (AMF) and rhizobia. Beneficial effects for the plant have been assumed to rely on increased P supply through the mycorrhizas. Recently, we demonstrated that P does not regulate the establishment of the tripartite symbiosis. Flavonoids appear to play a role as early signals for both rhizobia and AMF. Four soybean lines known to express different concentrations of the isoflavones genistein, daidzein and glycitein in the seed were used to test three hypotheses: (i) The establishment of the tripartite symbiosis is not dependent of a nutrient mediated effect; (ii) There is a positive correlation between seed isoflavone concentrations of different soybean lines and the progress of the tripartite symbiosis; (iii) Specific flavonoids control the establishment of the tripartite symbiosis in that a change in flavonoid root accumulation resulting from the development of one microbial partner can stimulate colonization of soybean roots by the other. Disturbed versus undisturbed soil treatments were produced to vary the potential for indigenous AMF colonization of soybean. In contrast, the potential for Bradyrhizobium was kept identical in both soil disturbance treatments. The uptake of P and Zn and the concentration of flavonoids in mycorrhizal soybean roots at 10 d after emergence were analysed either separately of Bradyrhizobium or in context of the tripartite symbiosis. Zinc nutrition did not differ between AMF treatments which supports the first hypothesis. The concentration of daidzein was at least four times greater in the root than in the seed reaching 3958±249 μg g−1 dry across soybean lines. Coumestrol, which was absent in the seed, was synthesized to reach 2154±64 μg g−1 dry. Conversely, the concentration of genistein was approximately three times smaller in the root that in the seed (301±15 μg g−1 dry), while glycitein and formononetin were never detected. The establishment of the tripartite symbiosis was identical across soybean lines which does not support the second hypothesis. Concentrations of flavonoids were significantly greater in roots under disturbed soil, for which both symbioses were not as developed as in plants from undisturbed soil. This clearly supports the third hypothesis. This research provides the first data linking the function of different flavonoids to the establishment of the tripartite symbiosis, and suggests that these compounds are produced and released into the rhizosphere as a function of the colonization process.  相似文献

Hyphal growth from germinated spores of Glomus caledonium was stimulated in the presence of thiosulphate, metabisulphite, sulphite and sulphate. The largest effects were observed with sulphite and metabisulphite which gave 17- and 11-fold increases respectively at their optimum concentrations. Since no greater incorporation of radioactivity into the spores was observed from 35S-sulphite than from 35S-sulphate it was not possible to conclude how this effect was mediated.  相似文献

Burkholderia pseudomallei, the bacterial cause of the potentially fatal infection known as melioidosis, has a facultative intracellular lifestyle. The intracellular presence of B. pseudomallei in various eukaryotes including arbuscular mycorrhizal fungus (AMF) spores can be demonstrated in vitro. AMF spores were isolated from soils in a melioidosis-endemic area. B. pseudomallei and other Burkholderia spp. DNA was detected in these AMF spore samples, confirming an AMF spore-Burkholderia spp. association in soils which did not yield Burkholderia spp. by culture. This association may explain the environmental persistence, difficulty of recovery and dispersal of Burkholderia spp. in specific environments.  相似文献

The objective of this study was to determine if root-feeding by insect larvae affects subsequent colonization of roots by arbuscular mycorrhizal fungi. We investigated grazing by larvae of Tipula paludosa on colonization of Agrostis capillaris by two species of fungi, Glomus mosseae and G. intraradices. Host plants were subjected to 7 days of grazing only, continuous grazing for 42 days, or no herbivory. Those plants with no herbivory had significantly lower levels of colonization by arbuscules and hyphae compared to plants which were grazed for 7 or 42 days. The effect only occurred in the upper parts of the root system, where larvae were active. We suggest that this effect was most likely mediated by a change in quantity and composition of root exudates.  相似文献

The symbioses between Trifolium subterraneum, mycorrhizal fungi and Rhizohium are affected by (NH4)2SO4 and by the nitrification inhibitors 2-chloro-6 (trichloromethyl) pyridine (N-Serve) and 2-trichloromethyl pyridine (2TMP). At 50 μg · g?1 soil N-Serve and 2TMP had toxic effects on plant growth, measured as leaf expansion, root length and dry weight. Lower concentrations of N-Serve also produced some toxic symptoms. The addition of (NH4)2SO4 to the soil at 2 and 6 m-equiv NH+4 per pot, resulted in reduced root length and nodulation. Shoot dry weight was reduced at 6 m-equiv NH+4 per pot. In the presence of (NH4)2SO4 the toxic effects of the nitrification inhibitors on plant growth were less.Both nitrification inhibitors reduced development of mycorrhizal entry-points and extent of root colonization (% infection). Percentage infection of the root system was also reduced by (NH4)2SO4. Development of nodules on the lateral roots was increased in the presence of N-Serve at 5 and 15 μ · g?1. This effect, however, was accompanied by a marked reduction in N2ase activity. Smaller increases in nodulation were apparent with 2TMP and were associated with variable N2ase activity.  相似文献

Development of seabird translocation techniques is required to meet species recovery objectives, to improve conservation status, and to restore ecological processes. During 1997-2008 we undertook translocation trials on eight petrel species of four genera within the New Zealand region: common diving petrel (Pelecanoides urinatrix), fairy prion (Pachyptila turtur), grey-faced petrel (Pterodroma macroptera gouldi), Pycroft’s petrel (Pterodroma pycrofti), Chatham petrel (Pterodroma axillaris), Chatham Island taiko (Magenta petrel; Pterodroma magentae), fluttering shearwater (Puffinus gavia), and Hutton’s shearwater (Puffinus huttoni). A total of 1791 chicks within 5 weeks of fledging were moved up to 240 km, placed in artificial burrows and hand-fed until they fledged. Of these, 1546 fledged, and so far at least 68 have returned as adults to the translocation sites. Most birds were crop-fed a puree based on tinned sardines and fresh water. This diet worked well for all species regardless of their typical natural diet (planktonic crustaceans, squid, or fish) with all species fledging above or close to mean natural fledging weights.  相似文献

Earthworms and arbuscular mycorrhizal fungi (AMF) might interactively impact plant productivity; however, previous studies reported inconsistent results. We set up a three-factorial greenhouse experiment to study the effects of earthworms (Aporrectodea caliginosa Savigny and Lumbricus terrestris L.) and AMF (Glomus intraradices N.C. Schenck & G.S. Sm.) on the performance (productivity and shoot nutrient content) of plant species (Lolium perenne L., Trifolium pratense L. and Plantago lanceolata L.) belonging to the three functional groups grasses, legumes and herbs, respectively. Further, we investigated earthworm performance and plant root mycorrhization as affected by the treatments. Our results accentuate the importance of root derived resources for earthworm performance since earthworm weight (A. caliginosa and L. terrestris) and survival (L. terrestris) were significantly lower in microcosms containing P. lanceolata than in those containing T. pratense. However, earthworm performance was not affected by AMF, and plant root mycorrhization was not modified by earthworms. Although AMF effectively competed with T. pratense for soil N (as indicated by δ15N analysis), AMF enhanced the productivity of T. pratense considerably by improving P availability. Remarkably, we found no evidence for interactive effects of earthworms and AMF on the performance of the plant species studied. This suggests that interactions between earthworms and AMF likely are of minor importance.  相似文献

Five ectomycorrhizal fungi were found to have an optimum growth in Mt. Burr sand at total soil water potentials of ?300 to ?600 kPa but could grow or survive at a total water potential of ?4000 kPa when glucose was supplied exogenously. Different fungi responded differently to reduced soil water potential induced by the addition of KCl solutions to soil.Growth characteristics of mycorrhizal fungi in the rhizosphere were poorly related to growth in soil with added glucose and yeast extract. Colonisation of Pinus radiata roots by Rhizopogon luteolus was greatest at soil water contents of 13% (?120 kPa osmotic potential) and was significantly depressed at 6.5%, (?510 kPa) and at 27% (?40 kPa). Cencoccum graniforme colonised roots most at 20%, (?60 kPa). Colonisation by this fungus was depressed at soil water contents of 13 to 6.5% and at 27%.Mycorrhizal infection was greatest at soil water contents of 13% (?120 kPa osmotic potential) to 20% (?60 kPa) and was significantly depressed at 27%, (?40 kPa). The depression of fungal growth in the rhizosphere and infection at soil water contents approaching saturation is discussed in terms of soil void space and aeration problems.  相似文献

Interactions between the arbuscular mycorrhizal (AM) fungus Glomus intraradices and bacteria from the genus Paenibacillus (P. macerans and P. polymyxa) were examined in a greenhouse pot experiment with Cucumis sativus with and without organic matter amendment (wheat bran). P. polymyxa markedly suppressed AM fungus root colonization irrespective of wheat bran amendment, whereas P. macerans only suppressed AM fungus root colonization in combination with wheat bran amendment. Dual inoculation with P. macerans and G. intraradices in combination with wheat bran amendment also caused severe plant growth suppression. Inoculation with G. intraradices was associated with increased levels of dehydrogenase activity and available P in the growth substrate suggesting that mycorrhiza formation accelerated the decomposition of organic matter resulting in mobilization of phosphorus. Inoculation with both Paenibacillus species increased all measured microbial fatty acid biomarkers in the cucumber rhizosphere, except for the AM fungus biomarker 16:1ω5, which was reduced, though not significantly. Similarly, inoculation with G. intraradices increased all measured microbial fatty acid biomarkers in the cucumber rhizosphere, except for the Gram-positive bacteria biomarker 15:0 anteiso, which was overall decreased by G. intraradices inoculation. In combination with wheat bran amendment G. intraradices inoculation caused a 39% reduction in the amount of 15:0 anteiso in the treatment with P. polymyxa, suggesting that G. intraradices suppressed P. polymyxa in this treatment. In conclusion, plant growth promoting species of Paenibacillus may have suppressive effects of AM fungi and plant growth, especially in combination with organic matter amendment. The use of an inert plant growth media in the present study allowed us to study rhizosphere microbial interactions in a relative simple substrate with limited interference from other soil biota. However, the results obtained in the present work mainly show potential interactions and should not be directly extrapolated to a soil situation.  相似文献

为了研究紫茎泽兰(Ageratina adenophora)入侵对土壤菌根真菌(mycorrhizal fungi, MF)群落的影响,采用嵌套PCR 技术分析了外来植物紫茎泽兰入侵生境内土著植物群落、土著植物与紫茎泽兰混生群落、紫茎泽兰单优群落中, 侵染紫茎泽兰及土著植物的MF 群落结构, 及紫茎泽兰与土著植物根围土壤中MF 群落结构。结果表明, 紫茎泽兰不同入侵进程MF 群落结构存在差异, 其中, 从土著植物群落的植物根内检测到内养球囊霉(Glomus intraradices)型克隆; 从土著植物与紫茎泽兰混生群落的紫茎泽兰根内也检测到内养球囊霉型克隆, 而在土著植物根内检测到1 个球囊霉属(Glomus sp 2)型克隆; 从紫茎泽兰单优群落的紫茎泽兰根内未检测到MF, 但从其根围土壤中检测到2 个球囊霉属(Glomus sp 1 和Glomus sp 2)型克隆。在土著植物与紫茎泽兰混生群落中, 从紫茎泽兰根围土壤中检测到4 个克隆型, 分别为毛舌菌阔孢(Trichoglossum hirsutum)、皂味口磨(Tricholoma saponaceum)、亚盖趋本菌(Xylobolus subpileatus)和翘鳞肉齿菌(Sarcodon imbricatus), 从土著植物根围土壤中也检测到4 个克隆型, 分别为小皮伞(Camarophyllopsis hymenocephala)、肉色香蘑(Lepista irina)、皂味口磨及亚侧耳(Panellus serotinus)型克隆; 在土著植物群落中, 从根围土壤只检测到皂味口磨型克隆。紫茎泽兰入侵改变了土著MF 群落结构, 其中在土著植物占据的土壤中以外生菌根真菌为主, 而外来植物紫茎泽兰则更多地积累了丛枝菌根真菌。文中讨论了紫茎泽兰改变入侵地土壤菌根菌群落及其可能对紫茎泽兰入侵的反馈。  相似文献

Plant root exudates induce the transition from asymbiosis to presymbiosis in arbuscular mycorrhizal fungi. In order to get an insight into this developmental switch, two libraries of Gigaspora rosea and one library of Gigaspora gigantea were screened for fragments of genes that show enhanced RNA accumulation 1 h after addition of a semi-purified exudate fraction of carrot roots. Among 150 clones, 40 seemed to contain inserts of root exudate-induced genes. One of the genes, GrosRbp1, putatively encoding an RNA binding protein involved in developmental control showed RNA accumulation which correlates to the extent of stimulation of presymbiotic hyphal branching.  相似文献

Plants inoculated with arbuscular mycorrhizal (AM) fungi utilize more soluble phosphorus from soil mineral phosphate than non-inoculated plants. However, there is no information on the response of soil microflora to mineral phosphate weathering by AM fungi and, in particular, on the catabolic diversity of soil microbial communities.The AM fungus, Glomus intraradices was examined for (i) its effect on the growth of Acacia holosericea, (ii) plant-available phosphate and (iii) soil microbial activity with and without added rock phosphate.After 4-months culture, AM fungal inoculation significantly increased the plant biomasses (by 1.78× and 2.23× for shoot and root biomasses, respectively), while mineral phosphate amendment had no effect in a sterilized soil. After 12-months culture, the biomasses of A. holosericea plants growing in a non-sterilized soil amended with mineral phosphate were significantly higher than those recorded in the control treatment (by 2.5× and 5× for shoot and root biomasses, respectively). The fungal inoculation also significantly stimulated plant growth, which was significantly higher than that measured in the mineral phosphate treatment. When G. intraradices and mineral phosphate were added together to the soil, shoot growth were significantly stimulated over the single treatments (inoculation or amendment) (1.45×). The P leaf mineral content was also higher in the G. intraradices+mineral phosphate treatment than in G. intraradices or rock phosphate amendment. Moreover, the number of fluorescent pseudomonads has been significantly increased when G. intraradices and/or mineral phosphate were added to the soil. By using a specific type of multivariate analysis (co-inertia analysis), it has been shown that plant growth was positively correlated to the metabolization of ketoglutaric acid, and negatively linked to the metabolisation of phenylalanine and other substrates, which shows that microbial activity is also affected.G. intraradices inoculation is highly beneficial to the growth of A. holosericea plants in controlled conditions. This AM symbiosis optimises the P solubilization from the mineral phosphate and affects microbial activity in the hyphosphere of A. holosericea plants.  相似文献
