Leakage of electrolytes from leaf discs of treated Phaseolus Vulgaris L. plants was the main criterion used to study the effect of several chemicals on the permeability of leaf-cell membranes. Paraquat, diquat, dinoseb and oxyfluorfen (2-chloro-1-(3-ethoxy-4-nitrophenoxy)-4-(Trifluoromethyl) benzene) increased leaf-cell membrane permeability after exposure for 12 h or less. An‘aromatic’oil caused a large increase in permeability at 2–5 min after treatment. Increases in electrolyte release were also correlated with release of soluble amino acids from the leaf discs but the former method was the more sensitive. Increase in cell membrane permeability was always associated with injury symptoms such as appearance of necrotic areas in leaves. Chlorpropham, linuron, sodium azide, glyphosate and 2,4-D at 10?3M, as well as 1% X-77 surfactant and a non-phytotoxic isoparaffinic oil did not alter leaf-cell permeability at 12 h after treatment. Light was necessary for paraquat and oxyfluorfen to alter leaf cell permeability. Paraquat and oxyfluorfen caused a greater increase in leaf-cell permeability of a soybean mutant with yellow leaves as compared with the normal green leaves. With oxyfluorfen this difference in permeability was greater than with paraquat, and was associated with the appearance of severe necrotic injury symptoms in the yellow mutant; paraquat caused no injury symptoms.  相似文献

Summary. Studies on the physiological effects of dalapon in Cynodon dactylon (L.) Pers. were carried out. Field and glasshouse experiments showed that dalapon is freely translocated in the aerial organs and from these to the rhizomes. No transformation of the herbicide was detected during 72 h following application. In rhizomes as well as in stolons, dalapon caused growth inhibition and necrosis of buds while root growth was apparently not affected. This differential susceptibility was confirmed by culturing node explants in nutrient solution containing dalapon. Although dalapon killed aerial organs, most rhizomes were capable of recovery even when a high dose was applied. These results are discussed. An experiment was done to determine the effect of spring and autumn application of asulam and dalapon, nitrogen fertilizer and oversown white clover (Trifolium repens L.) on the productivity of subtropical pasture dominated by carpet grass (Axonopus affinis Chase). Assessments were made of pasture yield, botanical composition, weed invasion and recolonization by pasture species during the 12-month period following herbicide application. Both herbicides decreased the yield of carpet grass and increased that of the more productive paspalum (Paspalum dilatatum Poir.) except after autumn application when the yield of paspalum was greatly reduced by dalapon. White clover became established only on plots treated with herbicide and only after autumn application. Clover yields were greatest on plots treated with dalapon. Asulam was considered to be the most suitable herbicide for altering the balance between carpet grass and paspalum because dalapon initially reduced paspalum yield after autumn application and delayed recovery of total pasture. In addition, dalapon permitted greater invasion by broadleaf weeds. This work showed that herbicides have a potential role for speeding up botanical changes in unproductive carpet grass pasture and further refinement of the technique might provide a useful tool for increasing pasture productivity.  相似文献

建立了气相色谱-质谱联用(GC-MS)同时测定二甲戊灵和乙氧氟草醚在生姜、茎秆和土壤中残留量的分析方法,并采用该方法研究了两种农药在土壤中的消解动态及在土壤和生姜中的最终残留。样品前处理采用QuEChERS法,经乙腈提取,姜块和茎秆提取液分别用N-丙基乙二胺(PSA)和石墨化碳黑(GCB)净化,土壤提取液用弗罗里硅土柱净化,选择离子监测模式(SIM)扫描,外标峰面积法定量。结果表明:当添加水平为0.01~0.5 mg/kg时,二甲戊灵在生姜、茎秆和土壤中的回收率分别为91%~100%、90%~98%和86%~100%,相对标准偏差(RSD)分别为3.2%~3.7%、2.7%~4.2%和3.2%~5.1%;乙氧氟草醚在生姜、茎秆和土壤中的回收率分别为90%~95%、86%~91%和85%~95%,RSD分别为2.4%~4.4%、3.9%~5.5%和2.3%~4.9%;样品中二甲戊灵和乙氧氟草醚的定量限(LOQ)均为0.01 mg/kg。二甲戊灵在土壤中的消解半衰期在12.5~20.5 d之间,乙氧氟草醚在18.8~25.6 d之间。采用33%二甲戊灵乳油和24%乙氧氟草醚乳油混剂分别按推荐剂量(有效成分693 g/hm2和72 g/hm2)和高剂量(有效成分1 039 g/hm2和108 g/hm2)于种植后施药1次,在生姜收获期的姜块中均未检出二甲戊灵和乙氧氟草醚残留。研究结果可为二甲戊灵和乙氧氟草醚在生姜上的合理使用及其最大允许残留限量(MRL)标准制定提供参考。  相似文献

为开发新型安全高效的植物源苹果树腐烂病防治剂,采用菌丝生长速率法测定中草药地肤子乙醇提取物及各层析流分L1~L9对苹果树腐烂病菌Valsa mali的抑制作用,同时通过室内常规分析方法观测流分L7对该菌菌丝形态结构、细胞膜通透性及物质吸收和代谢的影响。结果表明,地肤子乙醇粗提物浓度为2 mg/mL时对苹果树腐烂病菌具有较强的离体抑制活性,处理后96 h的抑菌率达到92.16%。分别用石油醚、氯仿、正丁醇萃取醇提物,石油醚萃取物对苹果树腐烂病菌的抑制效果最为明显,其EC_(50)为0.07 mg/mL;在石油醚萃取物通过硅胶柱层析分离得到的9种流分中,流分L7对苹果树腐烂病菌的抑制作用显著,离体抑菌率高达96.73%;且在流分L7处理下,病菌菌丝体出现肿胀、膨大或畸形等现象,细胞膜通透性增大,可溶性蛋白、还原糖以及丙酮酸的含量随流分L7浓度的升高而持续降低,当浓度为100μg/mL时,菌丝体可溶性蛋白、还原糖及丙酮酸的含量分别降低70.78%、71.74%和78.68%。表明在离体培养条件下地肤子乙醇提取物能明显抑制苹果树腐烂病菌的生长,并对菌丝体细胞膜结构和功能稳定性产生显著影响,具有进一步开发为新型植物源苹果树腐烂病防治剂的潜力。  相似文献

Abstract

In an experiment conducted in an almost pure stand of Imperata cylindrica to compare manual and chemical methods of control, forking out of rhizomes up to a depth of 45 cm gave long-lasting control. Slashing the grass to ground level at intervals of 4–6 weeks resulted in a low degree of control in terms of visual assessment of top growth and a high degree of control in terms of suppression of rhizome development, which was comparable to the most effective chemical methods; dalapon (22.2 kg a.i./ha) and dalapon–paraquat sequential spray. The results suggest that the effectiveness of control measures can be improved by timing the dalapon spray to coincide with adequate soil moisture reserve and by regulating frequency of slashing with regard to soil moisture status and the number of previous slashings imposed. The chemical treatment and the slashings suppressed development of rhizomes but did not cause increased rhizome mortality. The stage of growth at which dalapon was sprayed in dalapon–paraquat sequential spray treatment, did not influence the degree of control achieved as assessed visually.  相似文献

Lack of control options for cool-season broadleaf weeds is a major deterrent to autumn-sown chickpea. Weed control and chickpea tolerance to PRE (pre-emergence) and POST (post-emergence) application of isoxaflutole and oxyflurofen, PRE metribuzin, POST pyridate, and flumetsulam were investigated at three locations, including Kermanshah, Kurdistan, and Hamedan provinces during 2017–2018. Untreated and weed-free checks were added for comparison. Pyridate and PRE oxyflurofen 125?g ai ha?1 caused the minor visual crop injury according to EWRS score (1–1.8), while the highest crop injury occurred with metribuzin (EWRS score 3.5–8.5) in whole locations. The most effective herbicides for weed reduction were pyridate (70–75%), PRE oxyfluorfen (69–76%), and POST oxyfluorfen (65–73%) at Kermanshah, PRE oxyfluorfen at 125 and 175?g ai ha?1 (70–78%), POST oxyfluorfen (70–76%) and pyridate (70–78%) at Kurdistan, PRE oxyfluorfen at 125 and 175?g ai ha?1 (88–96%), metribuzin (91–100%) and Pyridate (80–97%) at Hamedan. Pyridate and PRE oxyfluorfen at 125?g ai ha?1 resulted in the highest chickpea grain yield at the three locations. In general, PRE oxyfluorfen (125?g ai ha?1) was similar to pyridate in terms of efficacy in weed control and grain yield enhancement.

Summary. The doses required to give 90% control of three weed species have been estimated at different intervals after spraying for four herbicides: amitrole-T, dalapon-sodium, disodium mcthylarsonate (DMA) and paraquat. Using the point quadrat method, both first and total contact data showed Ottochloa nodosa , to be more tolerant to the herbicides than Paspalum conjugatum , which in turn was more tolerant than Axonopus compressus. Ottachloa nodosa was most susceptible to paraquat up to 6 weeks after spraying, but thereafter dalapon was more effective than either paraquat, amitrolc-T or DMA. For Paspalum conjugatum first contact data showed that paraquat was more effective than amitrole-T, particularly in the first 10–11 weeks after spraying, and that both were more effective than DMA and dalapon; total contact data showed that paraquat and DMA were the most effective chemicals for up to 6 weeks after spraying but that thereafter amitrole-T gave best results. Paraquat was the most effective herbicide against Axonopus compressus , while dalapon was more effective than amitrole-T or DMA.  相似文献

为探究番茄早疫病菌Alternaria solani对啶酰菌胺产生抗性后膜透性的变化,采用菌丝生长法、可见分光光度法、电导率法和荧光猝灭法分别测定了抗、感菌株对NaCl渗透压的敏感性、胞内可溶性蛋白和糖的含量、膜渗透率及对啶酰菌胺的吸收量。结果表明:抗性(R-Bos)菌株在2.5 g/L NaCl浓度中生长最快,敏感(S-Bos)菌株则在5 g/L NaCl浓度中生长最快;在0~10 g/L NaCl浓度下,R-Bos菌株的生长明显快于S-Bos菌株,在20~80 g/L NaCl浓度下生长无显著差异;R-Bos菌株的可溶性蛋白和可溶性糖含量显著增高,分别达61.9%~105.2%和133.1%~161.9%;用啶酰菌胺处理菌株后,所有菌株的渗透率上升,膜透性增大,但R-Bos菌株的渗透率始终低于S-Bos菌株;且R-Bos菌株对啶酰菌胺的吸收量明显减少,为48.3%~49.9%。推测番茄早疫病菌对啶酰菌胺产生抗性后,胞内保水能力提高,对渗透压表现出耐受性,同时膜透性降低使得对药剂的吸收能力下降。  相似文献

 本文以白菜幼苗为材料,研究了芸苔链格孢菌毒素(AB-毒素)对白菜叶片细胞膜透性及叶片内SOD酶、POD酶活性变化的影响。结果表明,白菜叶片用AB-毒素处理后,细胞膜透性增大,且随处理时间的延长和毒素浓度的增加而增高。不同白菜品种用毒素处理后膜透性的改变与病菌接种的发病程度呈正相关,即电导率大的白菜品种,叶片发病也重。试验结果还发现,芸苔链格孢菌及其毒素处理白菜后,体内SOD酶和POD酶活性的变化基本趋于一致,即处理后12 h内急剧下降,然后(12~48 h)活性上升。而对照处理基本变化不大。可见,芸苔链格孢菌毒素表现了与其病原菌一致的致病作用。  相似文献

为明确新型生防菌解淀粉芽胞杆菌Bacillus amyloliquefaciens HRH317菌株对病原菌串珠镰孢菌Fusarium moniliforme的抑制作用,采用牛津杯法对HRH317菌株抑菌活性进行测定,并采用扫描电子显微镜、透射电子显微镜和荧光显微镜对经HRH317菌株发酵上清液处理后的串珠镰孢菌菌丝形态及超微结构进行观察。结果显示,HRH317菌株发酵上清液对串珠镰孢菌有很好的抑菌活性,抑菌圈平均直径可达33.31 mm。扫描电镜结果显示,HRH317菌株发酵上清液处理24 h时,串珠镰孢菌菌丝体出现断裂现象;处理72 h时,串珠镰孢菌菌丝体断裂较严重,多处裂解;处理96 h时,串珠镰孢菌菌丝体彻底瓦解,且无完整菌丝体。透射电镜结果显示,HRH317菌株发酵上清液处理72 h时,串珠镰孢菌菌丝体细胞形态扭曲变形,细胞内结构紊乱,遭破坏。荧光显微镜结果显示,经PI染料染色处理12 h时,串珠镰孢菌细胞有少数细胞被染成红色,细胞膜通透性受一定程度破坏;处理16 h时,串珠镰孢菌细胞大面积被染红;处理20 h时,串珠镰孢菌细胞被染色面积增大;处理24 h时,串珠镰孢菌细胞膜受破坏程度增加,细胞内大面积被染色。表明解淀粉芽胞杆菌HRH317菌株对串珠镰孢菌菌丝形态和超微结构有破坏作用,能抑制病原菌串珠镰孢菌菌丝体生长。  相似文献

Protoplasmic viscosity, solute potential and water permeability were compared for parenchyma cells of the coleoptile in the cytoplasmic male sterile (CMS) and normal (N) variants of Zea mays L. inbred lines. Protoplasmic streaming was tested in the epidermal cells of the glume. These protoplasmic and membrane qualities of the individual variants of CMS types were distinctly different from those of the N type and were closely correlated with the susceptibility of the CMS-variants to Cochliobolus heterostrophus and with the sensitivity to T-toxins and methomyl: CMS-T type had lower protoplasmic viscosity, higher cell sap solute potential, and higher water permeability than did the N type. The CMS-C and CMS-S types were somewhat intermediate between CMS-T and N types in these parameters.Among the three parameters tested, protoplasmic viscosity was most consistent in making the distinction between the CMS variants and N cytoplasm. Higher water permeability in CMS-T, C and S types compared with N cytoplasm reflects the differences in the cell membrane structure. These correlations suggest that cytoplasmic genes are expressed in protoplasmic qualities and codetermine cell membrane structure or composition and factors controlling consistency of the cytoplasm. Protoplasmic parameters proved to be sensitive indicators for testing CMS cytoplasms.  相似文献

Treatment of human erythrocytes with 5 × 10?5M captan or captafol caused a rapid increase in the efflux of intracellular potassium. Captafol had a more pronounced effect than captan on cation permeability. Captafol also decreased anion permeability whereas captan did not affect this process. Glutathione (5 × 10?4M) had little effect in reducing potassium efflux when added to the cells after they were incubated for 1 h with captan or captafol, but it was effective in reducing the potassium loss when added to the cells prior to their treatment with the fungicides. Captafol caused an increase in osmotic fragility of the cells. Incubation of the cell membranes with captafol resulted in the liberation of a small fraction of membrane phospholipids, whereas captan produced no effect. Both the fungicides readily reacted with the sulfhydryl groups in the isolated membrane; 31.5 and 45.7% of the membrane sulfhydryl groups had disappeared following treatment with captan and captafol, respectively. It is suggested that the reaction of captan or captafol and/or their reaction products with the sulfhydryl and amino groups of the red cell membrane protein produces changes in the structure of the membrane with consequent alteration in its permeability.  相似文献

建立了气相色谱-三重四极杆串联质谱 (GC-MS/MS)检测留兰香、桂皮、薄荷和月桂叶中乙氧氟草醚、唑草酮、乙螨唑残留的分析方法。4种香辛料用超纯水饱和,乙腈提取,无水硫酸镁及氯化钠盐析,氨基/石墨化碳黑 (NH2-Carb) 固相萃取柱净化,多反应监测模式,气相色谱-串联质谱测定。结果表明:乙氧氟草醚在0.002 5~2 mg/L范围内,唑草酮和乙螨唑在0.01~2 mg/L范围内,3种农药的进样质量浓度与对应的峰面积间呈良好的线性关系,r > 0.99;乙氧氟草醚在0.025、0.5和2 mg/kg 3个添加水平下,在4种香辛料中的平均回收率在86%~112%之间,相对标准偏差 (RSD)在2.4%~9.6%之间;唑草酮在0.2、0.5和2 mg/kg 3个添加水平下的平均回收率在87%~114%之间,RSD在2.4%~11%之间;乙螨唑在0.5、2和5 mg/kg 3个添加水平下的平均回收率在86%~116%之间,RSD在3.2%~11%之间。乙氧氟草醚、唑草酮和乙螨唑在4种香辛料中的定量限 (LOQ) 分别为0.025、0.2和0.5 mg/kg。  相似文献

Summary. Trials with dalapon, paraquat and fenuron in 1963 and 1964, and with bromacil and amitrole-T in 1964, were carried out on mature Spartina sward. Dalapon, paraquat and fenuron all gave complete kill on some plots in 1963. In 1964, paraquat gave variable results and lower percentage kills than in 1963, whereas dalapon and fenuron were more consistent. Fenuron allowed a greater degree of regeneration than dalapon and was uneconomic at the high dose (60 lb/ac) required for complete kill. Dalapon gave the best results and with 100 lb/ac there was 98–100% kill 2 years after treatment. At 50 lb/ac results were initially good, but there was more regeneration in the second season after spraying. Bromacil at 5 and 20 lb/ac and a mixture of 5 lb/ac bromacil and 4 lb/ac amitrole-T maintained more than 99% kill lor 2 years after application.

The diphenyl ether herbicide oxyfluorfen (2-chloro-4-trifluoromethylphenyl 3-ethoxy-4-nitrophenyl ether) inhibits protoporphyrinogen oxidase (Protox) which catalyzes the oxidation of protoporphyrinogen IX (Protogen) to protoporphyrin IX (Proto IX), the last step of the common pathway to chlorophyll and haeme biosynthesis. We have selected an oxyfluorfen-resistant soybean cell line by stepwise selection methods, and the resistance mechanism has been investigated. No growth inhibition was observed in resistant cells at a concentration of 10(-7) M oxyfluorfen, a concentration at which normal cells did not survive. While the degree of inhibition of total extractable Protox by oxyfluorfen was the same in both cell types, the enzyme activity in the mitochondrial fraction from non-treated resistant cells was about nine-fold higher than that from normal cells. Northern analysis of mitochondrial Protox revealed that the concentration of mitochondrial Protox mRNA was much higher in resistant cells than that in normal cells. There were no differences in the absorption and metabolic breakdown of oxyfluorfen. The growth of resistant cells was also insensitive to oxadiazon [5-tert-butyl-3-(2,4-dichloro-5-isopropoxyphenyl)-1,3,4-oxadiazol-2-(3H)- one], the other chemical class of Protox inhibitor. Therefore, the resistance of the selected soybean cell line to oxyfluorfen is probably mainly due to the overproduction of mitochondrial Protox.  相似文献

Grafted watermelon is a combination of two plants, aCucurbita rootstock and a watermelon scion. Therefore, weed control for this crop faces a unique problem: the safety of the selected herbicide has to be tested for both plants that make up the grafted plant. In the current study, we evaluated the usage safety of selected herbicides forCucurbita rootstocks as well as for non-grafted and grafted watermelons, and the control ofAmaranthus retroflexus by the same herbicides. In addition, the residual effect of the herbicides was tested for seeded and transplanted melons representing the next crop following cultivation of the grafted watermelons. The herbicides ethalfluralin, pendimethalin, ethalfluralin, sulfentrazone, oxyfluorfen, chlorsulfuron and clomazone were chosen for their potential to controlA. retroflexus. Pendimethalin and trifluralin were less effective than the other herbicides in controllingA. retroflexus; sulfentrazone, chlorsulfuron and clomazone were not safe for use on the tested cucurbits and thus cannot be recommended for weed control in grafted watermelons. Therefore, by eliminating the herbicides that are toxic to cucurbits and those that are ineffective forA. retroflexus control, it was concluded that the herbicides ethalfluralin and oxyfluorfen can be considered effective and safe for weed control in grafted watermelons. We quantified the resistance levels of transgenic rice plants, expressing Myxococcus xanthus protoporphyrinogen oxidase (PROTOX) in chloroplasts and mitochondria, to PROTOX inhibitors, acifluorfen, oxyfluorfen, carfentrazone-ethyl, and oxadiazon. We also determined whether active oxygen species-scavenging enzymes are involved in the resistance mechanism of transgenic rice. The transgenic rice line M4 was about >200-fold more resistant to oxyfluorfen than the wild-type (WT). M4 was also resistant to acifluorfen, carfentrazone-ethyl, and oxadiazon, but did not show multiple resistance to imazapyr and paraquat, which have different target sites. Acifluorfen, oxyfluorfen, carfentrazone-ethyl, and oxadiazon reduced the chlorophyll content in leaves of WT, but had minimal or no effect on M4. The PROTOX inhibitors also caused significant lipid peroxidation in the treated leaves of WT rice. However, the malondialdehyde production in M4 was not affected by these herbicides. The WT rice had higher activities of superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase than M4 after treatment with PROTOX inhibitors. A similar response was observed in all cases of antioxidant isozyme profiles analyzed. However, the induction in antioxidant activity in WT was not enough to overcome the toxic effects of a PROTOX inhibitor so the plant eventually died.  相似文献

Summary. In a field trial of modified logarithmic design, NaTCA and dalapon were applied pre-sowing and pyrazon and lenacil were applied pre-emergence. There were five rates of application and each pair of herbicides (NaTCA + pyrazon, dalapon + pyrazon, NaTCA + lenacil and dalapon-|- lenacil) had thirty-six variants. NaTCA and dalapon alone were found to have some effect on the dicotyledons. Increasing rates of NaTCA and dalapon in combination with pyrazon or lenacil resulted in better weed control, but also had a slight phytotoxic effect on the beet. Because the susceptibility of beet did not increase to the same extent as that of the weeds with the combined treatments examined, the selectivity under the conditions of the experiment was at least not reduced.

Summary. Studies were conducted to determine the effects of herbicide placement at different zones of maize (Zea mays L.) and pea (Pisum sativwm L.) shoots below the soil surface after emergence. Soil was removed from around the shoots and replaced with herbicide-treated soil. A wax barrier ensured separate exposure of the zones to treated soil. EPTC, chlorpropham, propham and sulfallate did not affect pea shoot growth, but in maize the shoot zone adjacent to the crown root node was extremely sensitive. Treatment in this area markedly reduced growth and severely inhibited the crown roots. The difference in susceptibility between these species may he due to the location of the growing point relative to the treated soil. Shoots of maize and pea were sensitive to diuron. In maize the shoot adjacent to the crown root node and the tissue of the first internode were the most susceptible. In pea the- uppermost shoot (beneath the soil surface) was the most sensitive. Trifluralin did not affect growth of maize and pea when placed in the shoot zone after emergence, although the crown roots of maize were severely inhibited. Naptalam, dalapon and 2,4-D did not affect growth of maize under similar conditions, and of these only 2,4-D reduced growth of pea.
