Genetic variation of wild populations and cultivars of Luohanguo (Siraitia grosvenorii), a plant species endemic to southern China, was assessed using random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP) markers. Based on the results for 130 individuals from seven populations, a high level of genetic diversity of Luohanguo was observed at the species level. The percentage of polymorphic loci (P) was 89.4%, Nei’s gene diversity (H e) was 0.239, and Shannon’s information index (H o) was 0.373 based on the combined AFLP and RAPD data. There was a high degree of genetic differentiation, with 45.1% of the genetic variation attributed to differences between the populations. The genetic diversity of the Luohanguo cultivars is much lower than that of wild populations (P = 41.8%, H e = 0.141, H o = 0.211), and a distinct genetic differentiation is observed between the cultivars and wild accessions. The pool of genetic variation in the wild populations provides an excellent gene resource for Luohanguo breeding.  相似文献

Genetic diversity of seven cultivated populations of Codonopsis pilosula Nannf. from Longxi County, Gansu Province of China was estimated using randomly amplified polymorphic DNA (RAPD) markers. The 17 selected RAPD primers amplified 205 polymorphic bands out of a total of 235 (87.2%). Nei’s gene-diversity statistics and population differentiation parameters based on AMOVA analysis indicated that the cultivated C. pilosula populations remained a high level of genetic diversity with Hs = 0.299 and I = 0.450. A greater proportion of genetic diversity was found within (77%) rather than among (23%) the populations. In addition, we also detected that populations from different altitudes had a considerable genetic differentiation after 40 years of cultivation at the same site. Populations from higher altitude had lower genetic diversity than those from lower altitude. Our results suggested that irregular and sparse cultivation practices, i.e., random collecting, preserving, and planting seeds of the medicinal species without deliberate selection, might be an efficient way to conserve genetic resources of medicinal plants, in addition to their effective uses.  相似文献

The aim of the present study was to assess the genetic variation in several Israeli and Turkish populations of wild emmer wheat, Triticum turgidum ssp. dicoccoides, the progenitor of most domesticated wheat. Single spikes were collected in 2002 from 60 plants that grew in six different habitats in Ammiad, northeastern Israel (8–12 plants from each habitat), and in 1998 from 56 plants that grew in seven different habitats in Diyarbakir, southeastern Turkey (8 plants from each habitat). Seeds were planted in a nursery and DNA was extracted from every plant and analyzed by the fluorescent-based amplified fragment length polymorphism (AFLP) method. Seven primer combinations produced 788 discernible loci of which 48.6% were polymorphic in Israel and 40.5% in Turkey. The genetic diversity estimates P (frequency of polymorphic loci) and He (gene diversity) were higher in Ammiad than in Diyarbakir (means of P = 0.34 and He = 0.13 in Ammiad vs. P = 0.20 and He = 0.08 in Diyarbakir). Ammiad populations contained more unique alleles than Diyarbakir populations. The relative genetic diversity estimates (θ) values were 0.188 in Ammiad and 0.407 in Diyarbakir, suggesting better differentiation of the populations in Turkey. Genetic distance was larger between Israeli and Turkish populations than between populations of each country. The data indicate that the Israeli and Turkish populations are considerably diverged and that the Israeli populations are more polymorphic than the Turkish ones, having a larger within-populations genetic variation than among-populations one. The significance of the results in relation to the differentiation pattern of wild emmer in the Near East is discussed.  相似文献

Using the 8 specific primer pairs based on the conserved motifs of plant resistance genes, the plant disease resistance gene analog polymorphisms (RGAPs) in 15 wild emmer wheat (Triticum dicoccoides) populations from Israel had been detected. High genetic variations at the RGAP loci were observed in T. dicoccoides populations. A total of 254 discernible bands were obtained among 115 accessions, and 192 bands (75.6%) were polymorphic. Each genotype had a unique banding profile, and the genetic similarity coefficient ranged from 0.094 to 0.862. In T. dicoccoides, the proportion of polymorphic loci (P), the genetic diversity (He) and Shannon’s information index were 0.756, 0.362 and 0.541, respectively. The proportion of polymorphic loci (P) per population averaged 0.732 (range: 0.515–0.932); genetic diversity (He) averaged 0.271 (range: 0.212–0.338); and Shannon’s information index averaged 0.404 (range: 0.310–0.493). The coefficients of genetic distance (D) among populations averaged 0.107 (range: 0.043–0.178), and the results of Mantel test (r = 0.168, P = 0.091) showed that the estimates of genetic distance were geographically independent. Neighbor-joining cluster analysis suggested that the genetic relationships of T. dicoccoides populations were associated with their ecogeographic distribution. The hierarchical analysis of molecular variance (AMOVA) and the coefficient of gene differentiation (G ST ) values revealed that most of the variations were presented within populations, although significant differences among populations and regions were also detected. The values of P and Shannon’s information index were negatively correlated with the two factors: Tdd (day–night temperature difference) and Ev (mean annual evaporation), whereas they were positively correlated with one water factor: Rn (mean annual rainfall). The correlation matrix between He in the RGAPs and geographic variables contained 20 significant (P < 0.05) correlations. The present study established that T. dicoccoides in Israel had a considerable amount of genetic variations at RGAP loci at least partly correlated with ecological factors.  相似文献

Genetic diversity of 70 populations of niger (Guizotia abyssinica) representing all its growing regions in Ethiopia was investigated using random amplified polymorphic DNA (RAPD) to reveal the extent of its populations genetic diversity. Ninety-seven percent of the loci studied was revealed to be polymorphic for the whole data set. The within population diversity estimated by Shannon diversity index and Nei gene diversity estimates was revealed to be 0.395 and 0.158, respectively. The extent of genetic variation of populations from major niger producing regions was significantly lower than that of populations from other regions; however, it is distributed regardless of altitude of growth. Genetic differentiation between populations was estimated with Shannon index as G ST (0.432), Nei’s G ST (0.242) and AMOVA based F ST (0.350) and appears to be equivalent to the average values calculated from various RAPD based studies on outcrossing species. Higher proportion of the variation detected by AMOVA resided within populations (64.58%) relative to the amount of variation among populations (35.42%). UPGMA cluster analysis showed that most of the populations were clustered according to their region of origin. However, some populations were genetically distant from the majority and seem to have unique genetic properties. It is concluded that the crop has a wide genetic basis that may be used for the improvement of the species through conventional breeding and/or marker assisted selection. Collection of germplasm from areas not yet covered and/or underrepresented is the opportunity to broaden the genetic basis of genebank collection.  相似文献

In this study, the genetic diversity and differentiation of 10 natural Prunus pseudocerasus Lindl. populations were investigated using inter-simple sequence repeat (ISSR) markers. Totally, 18 selected primers generated 150 loci, with an average of 8.33 bands per primer. The results showed that the percentage of polymorphic bands (PPB) was pretty low at the population level (PPB = 1.13–32%), but relatively high at the species level (PPB = 84%). Besides, a high level of genetic differentiation among populations was detected based on the gene differentiation coefficient (G ST = 0.7118) and the hierarchical analysis of molecular variance (AMOVA) (Φ ST = 64.53%, P < 0.001), in line with the low inter-population gene flow (N m = 0.2025). Moreover, Mantel test revealed a significant correlation between genetic and geographic distances among the populations (r = 0.5272, P < 0.005). The high level of intraspecific genetic diversity was probably related with its life history traits, while its small population size and the resultant high levels of genetic drift and inbreeding might explain the low genetic diversity within populations. The relatively high inter-population genetic differentiation was largely attributed to its small population size, habitat fragmentation, the mode of pollen and seed dispersal, and geographic isolation. Based on the present study, conservation strategies were proposed to preserve this valuable natural germplasm resource.  相似文献

Genetic diversity within and among 12 populations of the dioecious tropical tree species Hagenia abyssinica (Bruce) J.F. Gmel. in Ethiopia was examined with eight inter simple sequence repeat (ISSR) primers. A total of 104 clearly scorable bands were generated, among which 84 (81%) were polymorphic. Jaccard similarity coefficient was calculated for pairwise comparisons among all 120 individuals and ranged from 0.30 to 0.88 while average within-population similarity ranged from 0.53 to 0.66. Within-population variability was estimated as percentage polymorphic loci (ranging from 52% to 87%), Shannon’s information index (0.30–0.50) and Nei’s genetic diversity (0.21–0.35). The highest variability values were obtained for one recently planted population and for one wild population growing in an undisturbed primary forest area. Significant overall differentiation among populations was detected by both Shannon’s information index (0.26) and G ST (0.25). Relatedness among samples was estimated with a principal coordinate analysis, and relatedness among populations was estimated with a cluster analysis (UPGMA). A Mantel test indicated a significant association between genetic and geographic distances, and an autocorrelation analysis showed significant evidence of gene flow over distances up to 30 km. This study is the first of its kind for H. abyssinica, which has decreased recently in Ethiopia and now must be regarded as an endangered species. Both within-population and between-population diversity estimates are typical of outcrossing, longlived and late successional species, suggesting that recent anthropogenic disturbances have not yet had much impact on population genetic parameters. DNA marker data can, however, be used to identify the most suitable sites for in situ conservation and for collection of material for establishment of genebanks and plant improvement programs.  相似文献

Genetic diversity among 115 coffee accessions from the Coffea Germplasm Collection of IAC was assessed using SSR markers. The germplasm represents 73 accessions of Coffea arabica derived from spontaneous and subspontaneous plants in Ethiopia and Eritrea, species center of origin and diversity, 13 commercial cultivars of C. arabica developed by the Breeding Program of IAC, 1 accession of C. arabica cv. ‘Geisha’, 13 accessions of C. arabica from Yemen, 5 accessions of C. eugenioides, 4 accessions of C. racemosa and 6 accessions of C. canephora. Genetic analysis was performed using average number of alleles per locus (A), proportion of polymorphic loci (P), Shannon’s genetic index (H′ and GST) and clustering analysis. All evaluated species were distinguished by a cluster analysis based on Jaccard’s coefficient. Differentiation between the cultivated plants of C. arabica and accessions derived from spontaneous and subspontaneous plants was observed. Spontaneous and subspontaneous accessions from Ethiopia were separated according to the geographical origin: east and west of the Great Rift Valley. Cultivated plants showed a low genetic diversity with a division in two groups: accessions from Yemen (H′=0,028) and Brazilian commercial cultivars (H′=0,030). The results agreed with previously reported narrow genetic basis of cultivated plants of C. arabica and supported the hypotheses about domestication of the species. This study also showed a significant genetic diversity among accessions from Ethiopia and Eritrea present in the Germplasm Collection of IAC. This diversity is specially observed in accessions from Sidamo (H′=0,143), Kaffa (H′=0,142) and Illubabor (H′=0,147) indicating their importance as source of genetic variability for coffee breeding programs.  相似文献

Tetraena mongolica Maxim, is a critically endangered and endemic species of westem Inner Mongolia in China. Genetic variability within and among eight extant populations of this species was assessed using ISSR PCR (13 primers). We expected a low genetic diversity level, but our results revealed an intermediate level of intraspecific genetic diversity, probably resulting from this species being in a refuge during the last glaciation (at population level: P=48.1%, Ae=1.305, HE=0.177 and Hpop=0.264; at species level: P=63.3%, A=1.368, HT=0.213 and Hsp=0.324). A low level of genetic differentiation among populations was detected based on Nei's genetic diversity analysis (16.91%), Shannon's diversity index (18.83%) and AMOVA (15.2%). Populations shared high levels of genetic identity (I=0.9516±0.013). The extensive gene flow was a plausible reason for the low genetic differentiation.  相似文献

Myrtus communis L. (Myrtaceae), in Tunisia, is closely associated with Quercus suber L. forest which stretched continuously from the North to parts of Cap Bon and Tunisian Dorsal. The destruction of the primary oak forests associated to an over-exploitation of Myrtus for its essential oil quality had led to discontinuous populations exhibiting various levels of degradation. Using starch gel electrophoresis, we analyzed the polymorphism of nine isozymes in order to assess genetic diversity and structuring of 17 natural populations prospected in the three geographical regions and coinciding with subhumid, humid inferior and semi-arid superior bioclimates. The analysis of the level and the distribution of the genetic diversity in this species might help in its conservation. Out of the 18 loci detected for all populations and isozymes analyzed, 12 loci were polymorphic. Allelic frequencies differed according to populations and particular alleles characterized ecological groups. A high level of genetic variation within populations was observed. The mean number of alleles per polymorphic locus was Ap = 1.67, the percentage of polymorphic loci was P = 60.3% and the observed (Ho) and the expected (He) heterozygosities were respectively 0.144 and 0.215. Populations belonging to subhumid (Cap Bon) and semi-arid superior (Tunisian Dorsal) climates, located in degraded sites exhibited the highest level of inbreeding (0.425 < FIS < 0.450). A high level of differentiation (FST = 0.396) and a low gene flow (Nm = 0.337) among populations, as a result of habitat intermediate destruction, were revealed. The differentiation of populations within the same bioclimate (or geographic) group was substantial and relatively higher for semi-arid superior populations (FST = 0.262), which were more distant. The three ecological groups exhibited a high level of structuring (FST = 0.401). These differentiations might be due to geographic distances and to the variations of ecological factors between sites, including human activities and environmental factors. Nei’s (1972) genetic distances calculated between pairs of populations were globally low (0.006 < D < 0.367) with a mean of 0.15. They indicate a high level of similarity between populations. UPGMA clustering, established through Nei’s genetic distances, showed three population aggregates according to their geographic/bioclimatic appartenances. The high differentiation between populations and the low level of their genetic divergence indicated their recent isolation under anthropic pressures. The species conservation (in situ or ex situ) strategies should take into account the genetic diversity level within populations and its variation between geographic groups.  相似文献

Genetic diversity of 69 populations of Agropyron cristatum (L.) Gaertn. originated from various regions of northern China was analyzed using 29 polymorphic microsatellite primers that were mapped on the wheat genome. The number of polymorphic bands ranged from 2 (Xgwm285, Xgwm43, Xgwm291, and Xgwm257) to 27 (Xgwm314) with an average of 10.480. The highest genetic diversity value was detected in the populations from Xinjiang Province (0.735), and the lowest was observed in populations from Qinghai Province (0.553). The proportion of diversity among and within regions was 16.9% and 83.1% of the total variation, respectively. According to the dendrogram generated by UPGMA cluster analysis based on Nei’s genetic distance matrix, all the populations of A. cristatum were distinctly clarified. At the Nei’s distance of 0.62, the populations were divided into 6 groups. The phenogram indicated that populations from similar ecogeographical regions were clustered together. The principal coordinate analysis showed that the populations from Inner Mongolia were more closely related to each other, and were less variable than the populations from Xinjiang Province.  相似文献

Juniperus procera is economically highly important but threatened tree species. It is the only species among 67 taxa in the genus Juniperus that naturally grows in Africa and south of the equator extending up to 18°S in Zimbabwe. Ethiopia is assumed to host the largest J. procera populations, which are also believed to have high genetic variation owing to their wide ecological amplitude. This study assessed genetic variation at AFLPs of J. procera populations in the Ethiopian highlands. In the study six populations, namely Chilimo, Goba, Menagesha-Suba, Wef-Washa, Yabelo and Ziquala were included. A total of 20–24 trees from each population were investigated based on 128 AFLP band positions. AMOVA revealed that most of the variation (94%) resided within populations of J. procera suggesting extensive gene flow among populations which is attributable to the outcrossing mating system and effective gene transport mechanisms of the species. However, genetic differentiation among populations was still significant (P < 0.05), and the differentiation was significantly (P < 0.05) correlated with geographic distance. All population pairs were significantly (P < 0.05) differentiated except for Menagesha-Suba and Wef-Washa. These two populations also showed the highest gene diversity (H j = 0.301 and H j = 0.297, respectively). These results are in accordance with historical records that claim the establishment of the Menagesha-Suba juniper population as plantation of seedlings from Wef-Washa back in fifteenth century.  相似文献

Sesame (Sesamum indicum L.) is a traditional oil crop cultivated throughout South East Asia. To estimate the genetic diversity of this crop in parts at the region, 22 sesame accessions collected in Vietnam and Cambodia were analyzed using 10 RAPD markers. The 10 primers generated 107 amplification products of which 88 were polymorphic fragments (83%). Genetic diversity of all populations was Ht = 0.34 when estimated by Nei’s genetic diversity and species diversity was H′sp = 0.513 when estimated by Shannon diversity index. Genetic distance ranged from 0.03 to 0.43, with a mean genetic distance of 0.23. The unweighted pair group method with arithmetic averages (UPGMA) cluster analysis for the 22 accessions divided the material in four groups. The dendrogram revealed a clear division among the sesame accessions based on their geographical region. Interestingly, some geographically distant accessions clustered in the same group, which might indicate the human factor involved in the spreading of sesame varieties. The high level of polymorphism shown suggests that RAPD techniques can also be useful for the selection of parents in sesame (Sesamum indicum L.) breeding program and for cultivar differentiation.  相似文献

To study the magnitude and nature of genetic variation in E. fibrosus, the levels and distribution of allozyme and RAPD variations were investigated in populations collected from Finland and Russia. The results obtained from the allozyme and RAPD studies were compared to each other in 10 of the populations. The allozyme analysis showed that 6 of 12 presumed loci (50%) were polymorphic within the species, while the mean number of polymorphic loci within populations was 4.8%. The mean number of allele per locus for the species was 1.5 and 1.05 across the populations. Genetic diversity at the species level was low (H es = 0.025), and the mean population genetic diversity was even lower (H ep = 0.007). Both these values were much lower than the average for other Elymus and self-fertilising species. The largest proportion of the total allozyme diversity was found among, rather than within the populations (G ST = 0.70). The allozyme genetic distances between the populations did not reflect geographic distances. Cluster and principal coordinates analyses revealed the same allozyme relationship patterns among the populations. A comparison of allozyme and RAPD variation in 10 of the populations showed differences in the amount of genetic variation. The RAPD analysis revealed higher levels of variation (A p = 1.19, P p = 20.3 and H ep = 0.09) than the allozyme one) A p = 1.06, P p = 5.8 and H ep = 0.008). For both markers, the largest proportion of the total gene diversity was found among the populations studied (G st = 0.63 for RAPDs and G st = 0.65 for allozyme). In contrast to the allozyme analysis, the RAPD based genetic distances did reflect geographic distances. The cluster and principal coordinates analyses showed different grouping of populations for each data set. There was a positive, but not significant, correlation (r = 0.41) between the genetic distance matrices resulting from these markers. Regional comparison revealed that the Finnish populations had a higher diversity than the Russian ones. Generally, this study indicates that E. fibrosus contains low genetic variation in its populations. The results are discussed in the context of conservation of the species.  相似文献

In order to give insights into the origin and historical selection process of Tunisian apricot propagated by grafting, 31 cultivars from three areas presenting contrasting ecological conditions – Kairouan, Testour and Ras Jbel were compared to cultivars from Europe, North America, North Africa, Turkey, Iran and China, using 234 AFLP markers. The phenetic analysis allowed to distinguish 5 clusters, the four previously defined groups: – ‘diversification’, ‘geographically adaptable’, ‘continental European’ and ‘Mediterranean’ – groups and the Tunisian one. The partitioning of genetic diversity within and between cultivar groups assessed according to the Bayesian approach and assuming Hardy–Weinberg equilibrium, showed a loss of 21.81–38.49% of genetic diversity in Tunisian apricot compared to Mediterranean and diversification groups, respectively. Genetic variation occurred within Tunisian subgroups rather than among (FST = 0.060) evidencing a narrow genetic pool. Mediterranean and Tunisian groups were the least differentiated. Comparing them, 24 AFLP fragments discriminated the Mediterranean group from the Tunisian group but most of them where also shared by the other groups. Strongly differentiated gene pool and low genetic diversity are probably the result of bottleneck effects linked to the occurrence of propagation by seedlings rather than by grafting during the introduction periods in the North and the Centre of Tunisia. This study points at the propagation by seedlings as an important factor which should be taken into account to understand the evolution of apricot in South Mediterranean areas.  相似文献

Eremostachys superba Royle ex Benth. (Lamiaceae) has undergone a severe decline in population size since its discovery in the North-western Himalayas in late 19th century. One hundred and seventy-two plants from six populations in the Indian states of Uttar Pradesh and Jammu & Kashmir, located between 0.45 km and 455.72 km apart from each other were evaluated for RAPD polymorphism. Sixteen random primers generated 92 bands overall, 77 of which were polymorphic. Shannon’s index of genetic diversity within populations (H o) ranged between 0.305 and 0.421; the average within-population diversity (H pop) was 0.389; and the total species diversity (H sp) was 0.478. The population from Mohand (representing the type locality) had the fewest plants, at 18, and was genetically the most depauperate. Among the other populations, ranging in size between 52 and 1,022 individuals, no relation between population size and genetic diversity was evident. It is suggested that these six populations represent relics of a larger, extended population, in which the presence of perennating rootstocks has helped preserve historic patterns of genetic diversity. AMOVA revealed that 83.01% of the variation exists within populations, which was consistent with earlier studies on the reproductive biology of E. superba, which indicated this species is predominantly allogamous. FST distances between all populations were significant, indicating geographic differentiation despite some of them being closely separated. Habitat restoration and protection from indiscriminate harvesting are proposed as primary strategies for conserving E. superba. Rejuvenation of the Mohand population through intrapopulation crossing between plants bearing diverse molecular phenotypes is also suggested.  相似文献

Levels of genetic variation and genetic structure of 15 wild populations and three domesticated populations of Capsicum annuum were studied by RAPD markers. A total of 166 bands (all of them polymorphic) and 126 bands (125 of them polymorphic) were amplified in wild and domesticated populations, respectively. Mean percentage of polymorphism was 34.2% in wild populations and 34.7% in domesticated populations. Mean and total genetic diversity were 0.069 and 0.165 for wild populations and 0.081 and 0.131 for domesticated populations. Parameters of genetic diversity estimated from 54 bands with frequencies ≥1 − (3/n) (n = sample size) showed that 56.7% of the total variation was within and 43.3% among wild populations, whereas 67.8% of the variation was within and 32.2% among domesticated populations. AMOVA indicated that total genetic diversity was equally distributed within (48.9 and 50.0%) and among (50.0 and 51.1%) populations in both wild and domesticated samples. Wild and domesticated populations were clearly resolved in a UPGMA dendrogram constructed from Jaccard’s distances (average GD = 0.197), as well as by AMOVA (17.2% of variance among populations types, p = 0.001) and by multidimensional scaling analysis. Such differentiation can be associated with domestication as well as different origin of gene pools of the wild (Northwestern Mexico) and cultivated (more probably Central Mexico) samples analyzed. The considerable genetic distances among cultivars (average GD = 0.254) as well as the high number of diagnostic bands per cultivar (33 out of 126 bands), suggest that genetic changes associated with domestication could have resulted from artificial selection intervening in different directions, but the inclusion of more domesticated samples might clarify the nature of distinctions detected here.  相似文献

To assess the genetic diversity among China’s cultivated barley, sequence tagged site (STS) marker analysis was carried out to characterize 109 morphologically distinctive accessions originating from five Chinese eco-geographical zones. Fourteen polymorphic STS markers representing at least one in each chromosome were chosen for the analysis. The 14 STS markers revealed a total of 47 alleles, with an average of 3.36 alleles per locus (range 2–8). The proportion of polymorphic loci per population averaged 0.84 (range 0.71–1.00); the mean gene diversity averaged 0.39 (range 0.28–0.49). The means of P and He were highest in the Yangtze reaches and Southern zone (P = 1.00; He = 0.46) and lowest (P = 0.71 He = 0.28) in the Yellow river reaches zone. The STS diversity in different zones is quite different from the morphology diversity. The STS variation was partitioned into 17% among the zone and 83% within the zone. Both cluster and principal coordinate analyses clearly separated the accessions into a dispersed group (mostly two-rowed barley with a lower mean GS value) and a concentrated group (mostly six-rowed barley with a higher mean GS value) according to the spike characteristic with only a few exceptions. The accessions from the Qinghai-Tibet plateau formed a distinctive subgroup and can be distinguished from the concentrated group. The role of Tibet in the origin and evolution of cultivated barley has been discussed.  相似文献

There is a need to understand whether weed genetic diversity is the same among different populations, especially between those exposed to herbicide selection and other without exposure history. Inter-simple sequence repeat (ISSR) were used to assess level and patterns of genetic diversity in wild Brassica juncea (L.) Czern. et Coss. populations. A total of 93 plants from 24 wild populations in China were analysed by eight primers resulting in 86 highly reproducible ISSR bands. The analysis of molecular variance (AMOVA) with distances among individuals corrected for the dominant nature of ISSRs showed that most of the variation (54.09%) occurred among populations, and the remaining 45.91% variance was attributed to differences among individuals within populations. The high differentiation was, perhaps, due to limited gene flow (Nm < 1.0) of this species. Though highest gene diversity was observed in resistant B. juncea population, the overall distribution of diversity across China was not geographic dependent. High F ST value (0.541) corroborated AMOVA partitioning and provided significant evidence for population differentiation in wild B. juncea. UPGMA cluster analyses, based on Nei’s genetic distance, revealed grouping pattern geographically. Based on these results, the factors affect weed population genetic diversity and implication for herbicide resistance evolution were discussed in the context of transgenic crops advent and increasing herbicide usage in China.  相似文献

豌豆蚜是我国苜蓿上危害最为严重的害虫之一。利用微卫星标记研究了我国绿色型豌豆蚜10个地理种群的遗传相似性、基因分化、遗传距离与地理距离和海拔之间的关系及其基因结构。选取15对引物扩增300个个体,共检测到41个多态性条带,多态性条带百分率(PPB)为100%。10个豌豆蚜地理种群观测等位基因数(Na)为1.592 7,有效等位基因数(Ne)为1.356 9,Nei’s基因多样性指数为0.206 6,Shannon-Wiener指数(I)为0.307 6。新疆维吾尔自治区、陕西省、河南省种群的遗传多样性较高,内蒙古自治区、山东省、青海省种群相对较低。然而,10个地理种群豌豆蚜(绿色型)的遗传分化较高(Gst=0.399 6)。种群聚类分析结果显示,全部豌豆蚜种群明显聚为两大类群,山东省、河南省种群为一大类群,其余为另一大类群。Mentel检测表明,遗传分化与地理距离、海拔无显著相关性。我国豌豆蚜种群具有非常丰富的遗传多样性,应加强豌豆蚜的监测和治理。  相似文献
