采取10头健康乳牛的咽后淋巴结,按常规制备石蜡切片,用兔抗牛S-100多克隆抗体进行免疫组织化学染色,观察了健康乳牛咽后淋巴结内树突状细胞(DC)的分布。结果显示,在咽后淋巴结中,S-100^+DCs不仅分布在皮质,而且在髓质中也有大量分布;在被膜下窦内也有S-100^+DCs,在皮质的最外层有较多的S-100^+DCs;在淋巴滤泡中,滤泡树突状细胞(FDCs)呈S-100阳性标记,而在淋巴滤泡之间则有少量的S-100^+DCs;髓质中有大量的S-100^+DCs,多数呈贴壁分布,少数散布在窭腔内。表明,在正常乳牛咽后淋巴结内就有树突状细胞分布,一群是皮质树突状细胞,另一群是髓质树突状细胞。  相似文献

Canine dendritic cells were prepared from peripheral blood or lymph nodes using a series of steps including fractionation on bovine plasma albumin (BPA), irradiation with 4000 R, incubation for 16–18 hours, and refractionation on BPA. Dendritic cells were recovered in the low density (LD) fraction containing approximately 0.6% of the unfractionated cells. Measured by the incorporation of 3H-thymidine, the response of the high density (HD) cells to neuraminidase-galactose oxidase (NGO) was lower than that of the unfractionated lymph node cells (LNC) but increased in a concentration dependent manner after the addition of a population of cells enriched for dendritic cells (30–70% by morphologic criteria). Cooperation between HD- and LD- cells was not restricted to identity of the major histocompatibility complex. Canine dendritic cells also displayed stimulatory activity higher than unfractionated peripheral blood mononuclear cells (PBMC) in a one way mixed leukocyte culture (MLC). Canine dendritic cells were nonadherent to plastic, were of low density, and remained viable and functional after irradiation. For the first time, canine dendritic cells have been identified in peripheral blood and lymph nodes and have been shown to act as accessory cells in the response of lymphocytes to NGO and as stimulator cells in a MLC.  相似文献

To investigate the contribution of different bovine cell subpopulations in the development of in vitro induced responses by Ostertagia ostertagi third larval antigen extract (L3), bovine abomasal lymph node cell suspensions were depleted of specific cell populations. The depleted cell suspensions were subsequently assayed for their proliferative responses to O. ostertagi L3 antigen extract. Proliferative responses to O. ostertagi L3 antigen extract were restricted to a CD2+ CD4- CD8- cell population and MHC II+ cells different from B-cells were of major importance. Depletion of CD4, CD8, CD4CD8, IgM or CD21 positive cells did not decrease proliferation to L3 antigen extract. Depletion of gammadelta T-cells, which also comprise a subpopulation of CD2+ CD4- CD8- cells, reduced proliferation to L3 antigen extract only in one animal. The results suggest that either gammadelta T-cells could be involved in the proliferation or that another as yet unidentified population is important for proliferation. The precise role of these populations during infection with O. ostertagi and the mechanism by which these cells may influence the host immune response are important issues that remain to be elucidated.  相似文献

Subpleural lymph nodes were found in more than 4% of 3245 goat lungs during a slaughterhouse survey of respiratory disease. Their frequency varied between flocks from 2% to as much as 27%. They were more common in feral goats than in crossbreds or Angoras. Their numbers were positively associated with carcass weight and the number of nodular muellerius lesions on the lung surface. Their distribution and architecture were different from the pulmonary lymphoid nodules described in cattle with dictyocaulus re-infection syndrome.  相似文献

The jejunal and medial iliac lymph nodes of 53 clinically normal dogs between the age of 4 and 6 weeks were examined ultrasonographically. At least two jejunal and both left and right medial iliac lymph nodes were seen in all dogs. One hundred forty‐five jejunal, 53 right medial iliac and 53 left medial iliac lymph nodes in six litters of dogs, for a total of 251 lymph nodes, were measured for cross sectional maximum diameters. Mean jejunal lymph node length was 16.4 mm (range 6.4–34.9 mm) and mean width was 6.0 mm (range 2.3–15.7 mm). The mean medial iliac lymph node length was 13.6 mm (range 7.2–27.8 mm) and mean width was 4.4 mm (range 1.9–8.2 mm). Significant differences of lymph node size, noted between and within breeds, may not be of clinical significance. The mean size of the combined left and right medial iliac nodes was within previously published ranges for normal adult dogs. Lymph nodes were described in four litters of dogs (162 lymph nodes). Lymph nodes were either uniformly hypoechoic (108/163, 66%) or centrally hyperechoic with a hypoechoic rim (55/163, 34%). Although most (60%) lymph nodes were oval, a variety of shapes were seen, including vermiform and complex branching shapes. We concluded that in 4‐ to 6‐week‐old dogs, medial iliac lymph nodes are similar in size to adult dogs and jejunal lymph nodes are multiple, routinely seen, are larger than in adults and often have unconventional shapes.  相似文献

Pig lymph nodes have an unusual arrangement of cortical and medullary tissue, and lack a hilus. The aim of the present study was to describe the pathways by which blood reaches these lymph nodes. Natural or synthetic latex casts were made of blood vessels of superficial inguinal, popliteal and jejunal lymph nodes. Major nodal arteries approach these lymph nodes and give rise to about five branches, most of which remain on the node surface. These branches often wrap around part or all of the node in a claw-like manner and they divide to form a network on its surface. Arteries from this network penetrate the capsule directly to supply the lymphoid parenchyma. Anastomoses are common at all levels between the arteries supplying the nodes and arteriovenous anastomoses occur within the nodes. These may help to maintain and regulate blood flow within the lymph node under different conditions of stimulation.  相似文献

Immunoglobulin (Ig)-containing cells were investigated immunohistochemically in the stomach, intestine and associated lymph nodes of clinically normal calves, cows and fetuses to examine mucosal immune responses in the forestomach to rumen microbial flora. IgG-containing cells were observed in the mucosal propria of the forestomach of a 32 day-old, a 37 day-old, two 90 day-old calves, and of all 5 cases of cows. However, no IgA- and IgM-containing cells were observed. Further, no Ig-containing cells were detected in the associated lymph nodes of all calves and cows. Various numbers of Ig-containing cells, predominantly IgG, were observed in the mucosal propria of the abomasum, small intestine and cecum, and in the mesenteric lymph nodes of aged calves and all cows. No Ig-containing cells were observed in any tissues of fetuses. The results suggest that the development of mucosal immune responses in the forestomach may be incomplete as compared with that of the intestine.  相似文献
