Enterococcus cecorum is the most frequently occurring enterococcal species in the intestine of chickens of over 12 weeks of age, and there are few reports on its isolation from the skeleton of broiler parent chicks. In the present study, observations on vertebral osteomyelitis and spondylolisthesis ('kinky back syndrome') showing high incidence in 8 broiler parent flocks in different parts of Hungary are summarised. Clinical signs were seen only in roosters between 5 and 13 weeks of age. Diseased birds were alert and remained sitting on their hocks with their feet slightly raised off the ground. Incidence of the disease among male birds ranged from 8% to 30% depending on flocks. Enlargement and distortion of the body of the 6th vertebra were seen as the main pathological lesions. The cavity of the spinal canal was constricted by the distorted vertebral bodies. Resorption of bone tissue and sequestrum formation, signs of increased osteoclast activity, proliferation of fibrotic tissues, infiltration with heterophils and formation of sclerotic layers were detected in the vertebral bodies. From all 24 samples collected from the vertebral lesions, Enterococcus cecorum was isolated and identified using metabolic fingerprinting as well as 16S rRNA gene sequencing. Demonstration of E. cecorum from the vertebral lesions in all examined broiler breeder roosters showing the same clinical and pathological findings in different flocks suggested the pathogenic role of this microorganism for the first time in Hungary.  相似文献

Reoviruses are an important cause of suboptimum performance in commercial broilers worldwide. Integrators use the enzyme-linked immunosorbent assay against the S1133 antigen for monitoring serum of breeders for indicating pullet vaccine success. However, without correlating serology to reovirus challenge, it is difficult to determine whether titers reflect protective immunity. We developed a broiler challenge test against 2 common reovirus isolates (2408 and S1133) to evaluate the efficacy of reovirus pullet vaccine programs. Two reovirus serologic and challenge studies were undertaken using chicks from broiler integrators from the southeastern United States. Breeder flocks, from which the chicks were obtained, received at least 1 live and 2 inactivated reovirus vaccines during their pullet phase. One-day-old progeny were collected from 6 breeder flocks. At 1 d of age, 20 chicks from each broiler flock were bled, and serum was analyzed for antibodies. At 3 to 4 d of age, 20 progeny per flock were challenged with the 2408 reovirus by intratracheal route. At 10 to 14 d of age, another 20 birds per flock were challenged with the S1133 reovirus by footpad. Twenty birds per flock were used as nonchallenged controls. At 3 wk of age, all birds were killed and weighed. Percentage of protection was calculated for each flock based on the absence of gross lesions. Flocks with at least 50% protection were considered well protected. Most flocks were well protected against both viruses. The percentage of protection correlated with day-old enzyme-linked immunosorbent assay titers. Chicks from younger hens had higher titers and the best protection against challenge. Producers, whose hen flocks were monitored herein, were doing a good job of immunizing pullets against reovirus. They are now using reovirus progeny challenge studies along with breeder antibody titers to determine vaccination success of their pullets.  相似文献

This study evaluated bacterial skeletal disease in conjunction with the major histocompatibility complex (MHC) in a genetically pure line of broiler breeder chickens. Chickens from six broiler breeder flocks were examined for skeletal lesions, bacterial pathogens, and MHC genotype. During a 10-week period, eighty-eight, 9- to 21-week-old lame chickens and 34 normal, age-matched controls were selected. Tenosynovitis, arthritis, and femoral or tibiotarsal (or both) osteomyelitis occurred in 86 of 88 (97.7%) lame chickens. Ninety-five bacterial isolates were obtained from 83 of 88 (94.3%) lame birds and 4 of 34 (11.8%) controls. Staphylococcus spp. was isolated from 72.6% of the skeletal lesions, predominantly Staphylococcus aureus (38.9%). MHC B complex genotypes were determined by hemagglutination for 88 lame birds, 34 controls, and 200 randomly selected birds from each of the six flocks (1,200 total). Combined chi-square analysis revealed that the homozygous MHC genotypes B(A4/A4) (chi(2) = 14.54, P = 0.0063) and B(A12/A12) (chi(2) = 42.77, P = 0.0001) were overrepresented in the sample of symptomatic birds compared with random samples from the same flocks. The homozygous A4 and A12 MHC genotypes influenced flock chi-square values more than the corresponding heterozygotes. An MHC B complex influence on bacterial skeletal disease was apparent in this line of broiler breeders.  相似文献

1. Clinical and morphological studies were performed in 8 broiler breeder flocks (one hybrid) originating from two different farms in Bulgaria, with an incidence of lameness in female birds between 3 and 4% on one farm and 7 and 8% on the other. Clinical and morphological studies were performed. 2. Ruptured gastrocnemius tendons were observed in 78 broiler breeders, 24 to 31 weeks old with signs of lameness, selected for pathoanatomical study from different flocks. Unilateral ruptures predominated, over 80%. 3. A histological study of 20 samples from one farm and 8 samples from the other revealed that lesions varied between birds. In about 50% of them, haemorrhages, dystrophic and necrobiotic changes in tendons were present. In the other 50% of cases, the resolving haematoma and ruptured tendon were surrounded by fibrous tissue growth. No inflammatory changes were detected.  相似文献

A serological survey for antibody to Chicken Anaemia Virus (CAV) was performed on broiler breeders as well as layer breeding birds in Sweden at the end of their rearing period. Grandparents (GP) of both types leaving quarantine were in 21 out of 26 cases free from antibody to CAV, but often became infected soon thereafter. A total of 10 outbreaks of blue wing disease (BWD) in 3 series were recorded in the broiler and layer parent generation, all of which were progeny of 3 late seroconverting GP-flocks. All but one of 22 layer parent flocks had been infected and had seroconverted during the rearing period. Subsequently BWD was not recorded from commercial layers. Broiler parent flocks were more protected from CAV infection during rearing. Eighteen out of 94 broiler parent flocks had not developed antibody to CAV before coming into lay. Outbreaks of BWD were reported in progeny flocks from all these broiler breeders, with the exception of those that had been vaccinated. Good hygienic routines along with isolation of the birds delayed the seroconversion to CAV in broiler breeders and vaccination of these breeders protected their progeny from outbreaks of BWD. Broiler flocks in houses where BWD had occurred recently had always antibodies to CAV at slaughter. It was possible to eradicate the infection from the house and prevent the infection between flocks by proper cleaning and disinfection of the broiler houses.  相似文献

A field study was designed to determine the prevalence of subclinical infectious bursal disease (IBD) in broiler chickens from a commercial poultry company. Bursae of Fabricius (BF) from two vaccinated and three nonvaccinated broiler flocks were evaluated histologically, and antibody profiles of these broiler and matched parent breeder flocks were established. Lesions of IBD, including lymphoid necrosis, stromal edema, and infiltrates of heterophils and macrophages, were first detected in BF at 24 days of age in both vaccinated and nonvaccinated chickens. At 41 days, all BF had lesions characteristic of IBD, including severe lymphoid depletion, proliferation of epithelial cells, and mild fibroplasia. Although mean maternal antibody levels (measured by enzyme-linked immunosorbent assay) in broilers were apparently protective through day 12, IBD antibodies decreased to nonprotective levels (below 1,000) by day 16 or 20. Titers began to increase by day 28 or 32 because of field exposure. Sentinel birds, placed with broiler flocks, also developed IBD antibody titers. Broiler breeders had low and nonuniform antibody titers. Prevalence of field IBD exposure was high, and existing vaccination programs were not effective.  相似文献

In this investigation, we determined the prevalence of the Ornithobacerium rhinotracheale (ORT) infection in broilers and broiler breeders in southern Brazil. We also correlated the presence of antibodies in broilers with performance. Sera from 1550 broilers from 50 flocks were collected during the slaughter time in nine companies with federal veterinary inspection of the state of Rio Grande do Sul. Sera from 480 meat-type breeders of 40 flocks from 14 companies in southern Brazil were also analyzed by enzyme-linked immunosorbent assay, and the prevalence of antibodies was determined. The prevalence of ORT antibodies in broiler flocks was 63.83%, but in each individual flock only 6.52% of the birds were positive. The prevalence in broiler breeder flocks was 100.00%, and in each individual flock 94.62% of the birds were positive. There was a positive correlation between the presence of antibodies to ORT and decreased body weight in broilers. There was no significant correlation between presence of antibodies to ORT and age, lineage, efficiency index, feed conversion, and mortality. There was a positive correlation between the presence of respiratory signs and antibodies to ORT, although the reverse correlation was not significant. These results confirm that ORT is present and widespread in broilers and broiler breeders in southern Brazil.  相似文献

A broiler breeder operation experienced a sudden outbreak of high mortality in nine different houses of breeders at eight different locations. In the first house to be affected, the losses were mainly in the females, but later flocks had losses up to 6.4%, mainly in the males. Affected chickens ranged in age from 7 to 22.5 weeks. Multifocal areas of necrosis and vasculitis were found in skin, muscle, and internal organs. No involvement of the brain was identified, but many of the birds had uveitis. The outbreak lasted 3 weeks and ended without treatment. Neither infectious agents nor mycotoxins were identified.  相似文献

To evaluate the economic effects of subclinical chicken anemia agent (CAA) infection on broiler performance, clinically normal broiler flocks were grouped into two categories: A) flocks in which none of 10 birds sampled at slaughter had antibody to CAA, and B) flocks in which six or more of 10 similarly sampled birds had CAA antibody. Production and performance parameters of 25 flocks in each category were compared. No statistically significant differences were found between category A and category B flocks in major production parameters such as sex, feed manufacturer, type of litter, stocking density, and age at slaughter. However, category A flocks achieved 13% (P less than 0.05) greater net income per 1000 birds, 2.0% better feed-conversion ratio (P less than 0.05), and 2.5% (P less than 0.05) greater average weight per bird than category B flocks. No significant differences in hockburn bonus per 1000 birds (a bonus payable to growers whose broilers have the lowest prevalence of contact dermatitis lesions on the hocks) and mortality were found between category A and category B flocks. These results show that subclinical CAA infection has a substantial, statistically significant effect on commercial broiler performance and profitability.  相似文献

The prevalence of footpad dermatitis (FPD) in broiler chickens in Japan was investigated. In the first examination at slaughterhouses, lesions were commonly observed on the footpads of a total of 8,985 broiler chickens from 45 flocks on 36 farms. In 3 flocks, all the birds examined had lesions. In the other 42 flocks, the incidence of FPD ranged from 31.9% (81/254) to 99.5% (1/222). The footpad lesions were classified into 4 categories according to the severity of dermatitis as follows; score 0, 1,181 birds (13.1%); score 1, 2,992 birds (33.3%); score 2, 3,000 birds (33.4%); and score 3, 1,812 birds (20.2%). The mean scores of the flocks varied widely from 0.31 to 2.69. Males had higher mean scores than females. No significant differences were found between the mean scores of birds reared in windowless houses and those in open-sided houses. On 4 farms, the investigation was carried out in different seasons, and 3 of them showed higher mean scores in winter than in summer. Next, observation of 2,255 birds from 15 flocks on 5 farms during a 4-week rearing period revealed that FPD was already produced at 7 days of age and worsened with age.  相似文献

Swollen Head Syndrome (SHS) affects both broiler and broiler breeders and has become a problem in many countries. Clinical signs similar to those described in SHS conditions have been observed in northern Germany with the disease observed mostly in chicken broilers between 4 and 5 weeks of age and in broiler breeders between 24 and 36 weeks of age. The mortality rate in broilers has been variable from negligible to 5%. In broiler breeders, the symptoms were accompanied with a drop in egg production reaching 2-3% after 1 to 2 weeks. 422 Serum samples from 21 broiler flocks were examined for the presence of antibodies to TRT-virus using indirect ELISA tests. Antibodies were detected in 2 flocks which suffered from SHS, but not in 2 flocks showing respiratory manifestations nor in 17 apparently healthy flocks. The results obtained from testing 178 serum samples from 10 breeder flocks revealed that antibodies to TRT-virus were present in one flock with SHS signs as well as in 6 apparently healthy flocks. In addition, no antibodies were found in sera collected from 3 flocks--one with SHS and respiratory disease history; one recovered from respiratory infection and one without disease history. Examination of paired serum samples from 3 breeder flocks that suffered from typical SHS-signs collected at the onset of the disease and during the convalescent stage, as well as sera from one broiler and one breeder flocks that showed respiratory manifestations, showed that there are significant increases in the number of positive sera in the flocks with SHS symptoms. During a field study in 2010 the daily growth, feed conversion, first-week mortality, broiler loss due to mortality and slaughterhouse condemnation, and production index were monitored in 100 broiler flocks derived from four breeder farms vaccinated with Nobilis OR inac and four Ornithobacterium rhinotracheale-unvaccinated breeder farms of the same organization in Belgium. Other parameters related to the broiler flocks, such as flock size, season, age of the breeders, and corresponding breeder farms, were also noted. All gathered data were examined with ANOVA, linear correlation, and linear regression analyses. Results demonstrated a significant 22.3% lower broiler loss and a significant 3.9% higher production index in the broiler flocks derived from breeders vaccinated with Nobilis OR inac. These results confirm field observations obtained in 1999, thereby providing further evidence for an effect of O. rhinotracheale vaccination in breeders with regard to the improved performance of broilers.  相似文献

Ornithobacteriosis is an infectious disease of avian species that has been reported in almost all countries around the world, except Thailand. The objectives of this study were to determine the seroprevalence of Ornithobacterium rhinotracheale (ORT) and to isolate and identify ORT in broilers and broiler breeders in Thailand. Chicken antibodies had been randomly checked from 17 farms (19 flocks) of broilers and 23 farms (28 flocks) of broiler breeders. The seropositive flocks were 63% and 100% in broilers and broiler breeders, respectively. The sera analysis showed that the individual 280 broiler sera antibody responses were 67.5% negative, 12.9% suspect, and 19.6% positive. The individual antibody responses of 510 broiler breeder sera revealed 12.2% negative, 38.0% suspect, and 49.8% positive samples. The bacteria were isolated and identified by polymerase chain reaction (PCR). Bacterial isolation and identification revealed that nine isolates of the 12 PCR analysis samples showed positive results to PCR analysis. All the positive PCR samples were collected from the broiler breeder farms.  相似文献

Two broiler flocks contained 0.5 and 3% small chickens. The small chickens were approximately one-third the size of their penmates, were very active, and retained much of their chick down. They had distended abdomens with full intestinal tracts, often containing undigested feed. In many, the pancreases were thin, white, and firm because of loss of exocrine tissue and replacement by fibrous tissue. Many had skeletal changes suggestive of rickets. In a survey of 48 broiler flocks for small birds and pancreatic lesions, five flocks had a noticeable incidence of small chickens, varying from less than 1% to a maximum of 2%. In 33 of the flocks surveyed, a low incidence of pancreatic lesions was found in birds at the processing plant. No correlation was found between a noticeable number of small birds at the farm and pancreatic lesions found at processing.  相似文献

The present study examines the responses of broiler chickens to oral administration of Clostridium perfringens freshly isolated from field cases of necrotic enteritis (NE). The challenge studies included long-term exposure and short-term exposure, factored in with dietary and management variables including high levels of dietary components such as fish meal, meat meal, abrupt change of feed, and fasting. In the long-term exposure trials, the birds were orally inoculated daily, with 1 ml (1.0 or 2 x 10(8) CFU/ml) of an overnight culture of C. perfringens for 7 days. Short-term exposure trials involved challenge with 1 ml (3 x 10(10) CFU/ml) administered as a single dose. The responses of broilers to orally administered C. perfingens under laboratory controlled conditions are presented and discussed in the context of authentic field cases of necrotic enteritis. None of the challenge trials produced overt clinical signs of NE and there were no mortalities associated with oral exposure to high doses of C. perfringens. However, many of the challenged birds showed distinctly pronounced pathological changes in the intestinal tissue. On gross examination the responses in birds challenged orally with C. perfringens could be placed into two categories: (1) no apparent pathological changes in the intestinal tissue and (2) sub-clinical inflammatory responses with focal, multi-focal, locally extensive, or disseminated distribution throughout various sections of duodenum, jejunum, ileum, and ceca. In birds that responded with intestinal lesions, hyperemia and occasional hemorrhages were the main gross changes. In some birds, the mucosa was covered with a brownish material, but typically, the mucosa was lined by yellow or greenish, loosely adherent material. Mild gross changes were seen in some control birds, but both qualitatively and quantitatively, the lesions were distinctly more pronounced in the challenged birds. Upon histological examination, none of the experimentally exposed birds showed overt mucosal necrosis typical of field cases of NE, but typically the lamina propria was hyperemic and infiltrated with numerous inflammatory cells. Most significant changes were seen at the interface of the basal domain of enterocytes and lamina propria. Multifocally, these areas were extensively edematous, allowing for the substantial disturbance of the structural integrity between the lamina propria and the enterocytes. The lesions observed in the present study were consistently reproduced in all of our challenge trials, hence these responses may signify newly emerging patterns of sub-clinical enteric disorders in contemporary strains of poultry. The pathological changes observed in broilers challenged orally with C. perfringens in the present study, differ significantly from those reported previously, and must be clearly differentiated from those described in cases of NE or ulcerative enteritis. Although no overt necrosis of the intestinal mucosa typical of field cases of NE were observed in the present study, the birds challenged with C. perfringens showed strong inflammatory reaction to the introduced pathogens. The distinct features of the microscopic lesions were changes involving apparently normal enterocytes at the interface of the basal domain of villar epithelia and lamina propria. Although the pathological changes in the intestinal tissues observed in our trials appear to be rather subtle when compared to field cases of NE, the nature of these lesions suggest a significant negative effect on the digestive physiology of intestinal mucosa.  相似文献

A live attenuated Mycoplasma gallisepticum vaccine, ts-11, has been used for control of M gallisepticum in several countries. The rapid serum agglutination test is usually used as an indicator of flock response to vaccination; however, in some flocks, the detected response may be weak or absent. We investigated whether the low level, or lack, of systemic antibodies in ts-11-vaccinated flocks is correlated with susceptibility to infection after challenge with a virulent M. gallisepticum strain. Birds from 2 separate ts-11-vaccinated commercial flocks with no, or weak, rapid serum agglutination responses (at 11 or 14 wk postvaccination) were randomly selected and subjected to aerosol challenge with either M gallisepticum strain Ap3AS or sterile mycoplasma broth. A group of nonvaccinated specific-pathogen-free chickens at similar age were also exposed to aerosolization with M. gallisepticum strain Ap3AS and used as positive controls. Postmortem examination of the birds, performed 2 wk after challenge, revealed no significant difference in microscopic tracheal lesions or mucosal thicknesses between the ts-11-vaccinated field birds irrespective of their aerosolization treatment. However, both microscopic tracheal lesions and tracheal mucosal thicknesses of nonvaccinated challenged birds were significantly greater than those of ts-11 vaccinates. Hence, broiler breeders vaccinated in the field showed significant protection against virulent M. gallisepticum challenge even when no serum antibody was detected by rapid serum agglutination test. These results reveal that seroconversion detected by rapid serum agglutination test after ts-11 vaccination is not a reliable predictor of protection against M. gallisepticum infection. The possible significance of local antibody response and cell-mediated immunity against M. gallisepticum infection is discussed.  相似文献

An outbreak of anemia dermatitis syndrome caused by chicken anemia agent (CAA) occurred in 15 broiler flocks. An average of 29% of chickens in these flocks were derived from a common breeder flock. The breeder flock had no antibody to CAA at 20 weeks of age but had seroconverted by 31 weeks. Diseased broiler flocks were derived from eggs laid by the breeder flock between 25 and 30 weeks of age. CAA infection in the breeder flock was subclinical, with no apparent effects on mortality or performance. A strategic program of therapeutic and/or prophylactic antibiotic therapy was begun in affected broiler flocks as soon as the disease was diagnosed. Nevertheless, when the cost of therapy was taken into account, affected broiler flocks had a net income 17.3% to 19.6% lower than normal flocks. Average bird weights were 3.3% to 3.5% lower in affected flocks than in unaffected flocks, and affected flocks had a significantly greater proportion of lighter birds. Average mortality in affected flocks was 2.0% to 2.3% higher than in normal flocks, with peak mortality occurring in the third week of life. There was no apparent effect on feed-conversion ratio.  相似文献

Inclusion body hepatitis (IBH) was diagnosed in 15 broiler flocks supplied by one breeder in the South Island of New Zealand. The affected flocks suffered mortality up to 30%. Malaise and slightly increased mortality were noticed by growers from about day 12 post-hatch; mortality peaked in the fourth week, and, in most flocks, declined to normally accepted levels from day 33 on. Gross signs seen at necropsy usually included bone-marrow aplasia, atrophy of the bursa of Fabricius and the thymus, and swollen hemorrhagic livers with focal necrosis. Jaundice was seen in many surviving birds. In some flocks, there was also proventricular hemorrhage, mild tracheitis, and airsacculitis. Downgrading and condemnation rates were increased in all flocks. Eosinophilic intranuclear inclusion bodies were seen in hepatocytes of some affected birds. An adenovirus was isolated from a number of cases investigated. The disease in broilers was preceded by production drops associated with feed refusal and increased mortality in the breeder stock.  相似文献

Since a new envelope subgroup (J) of the avian leukosis-sarcomatosis-complex was isolated for the first time from broiler breeders in the United Kingdom in 1989 and was characterized and associated with myeloid leukosis (syn. myelocytomatosis) the emergence of this subgroup was reported from all over the world. Thus the first known case of subgroup J avian leukosis in Switzerland in four imported broiler breeder flocks will be described. A total of 53 broiler breeder birds from four flocks showing reduced performance and increased mortality were submitted for postmortem examination. Approximately 20 blood samples from each flock were monitored serologically for antibodies against avian leukosis virus subgroup J (ALV-J). On necropsy myeloid leukosis (ML) was diagnosed in all four flocks. Furthermore the blood samples of three flocks showed significant ELISA-titres for ALV-J.  相似文献
